Abstract

Introduction Extra virgin olive oil (EVOO) is the main lipid component of Mediterranean Diet (MD). Many studies, including the Seven Country Study, demonstrated that MD is able to reduce the incidence of chronic degenerative diseases, and that integration of MD with Extra Virgin Olive Oil (EVOO) decreases the risk of major cardiovascular events. The Food and Drug Administration had recognized EVOO (23 g/day) as a qualified health claim to decrease the risk of Cardio-Vascular Diseases (CDVs). EVOO supplementation brings several benefits, due to its antioxidant, anti-inflammatory, vasodilatory, and antiplatelet aggregation properties. The main EVOO cardio-protective effect is attributable to the presence of its phenolic compounds, of which hydroxytyrosol (HT) is the main component. The European Food Safety Authority has set out the health claims related to specific EVOO phenolic compounds: "A daily intake of 5mg of HT and its derivatives protect blood lipids from oxidative stress". Based on this assumption, we hypothesized that 3 weeks administration of 15 mg/day of HT, in a gastroresistant capsules new formulation, could lead to a change in nutritional status. Methods The randomized double-blind, placebo-controlled crossover trial was conducted on 28 volunteers. Evaluation was carried out on: nutritional status, serum metabolites, biomarkers of oxidative stress and related to oxidative stress, inflammation and CVD 9 genes expression. The study consisted of 2 cycles of 3- weeks chronic treatment with HT or placebo separated by 1 week washout period. At the beginning and at the end of each treatment, body composition evaluations, gene expression, and blood analysis were performed. Results Oxidation biomarkers, total antioxidant status, SOD 1, and plasma HT concentration were significantly increased, while nitrite, nitrate and malondialdehyde, as well as percentage of fat mass, sovrailiac fold and weight were drastically reduced after treatment. Conclusions This pilot study shows that the regular intake of 15 mg/day of HT modified the parameters of body composition and modulated the antioxidant profile and inflammatory and oxidative genes expression. The results, despite the limit of the reduced number of subjects enrolled, suggest the opportunity of a personalization of the therapy. The study, repeated on a larger scale, could acquire statistical significance.

132. Phytochemical characterization and biological properties of two standardized extracts from a non‐psychotropic Cannabis sativa L. cannabidiol (CBD)‐chemotype.

Abstract

The aim of study was to evaluate and compare the phytochemical profile, the antioxidant and antimicrobial properties of two standardized extracts from non‐psychotropic (Δ9‐tetrahydrocannabinol ≤0.2%) Cannabis sativa L. var. fibrante rich in cannabidiol (CBD). The two extracts, namely Cannabis Fibrante Hexane Extract 1 (CFHE1) and Cannabis Fibrante Hexane Extract 2 (CFHE2), were obtained by extraction with acidified hexane from dried flowering tops as such and after hydrodistillation of the essential oil, respectively. Gas chromatographic analysis showed that cannabinoids remained the predominant class of compounds in both extracts (82.56% and 86.38%, respectively), whereas a marked depletion of the terpenes occurred. Moreover, liquid chromatographic analysis highlighted a high titer of cannabidiol acid (CBDA) and CBD in CFHE1 and CFHE2, respectively. Both extracts showed a strong and concentration‐dependent antioxidant activity and a potent antimicrobial activity against both Staphylococcus aureus ATCC 6538 (MIC and MBC of 4.88 μg/ml for CFHE1, and 4.88 and 19.53 μg/ml, respectively, for CFHE2) and methicillin resistant clinical strains (MIC values between 1.22 and 9.77 μg/ml and MBC values between 4.88 and 78.13 μg/ml). Considering this, the obtained results suggest that standardized extracts of C. sativa var. fibrante could find promising applications as novel antimicrobial agents. [ABSTRACT FROM AUTHOR]

Abstract

itrus fruit and juices represent one of the main sources of compounds with a high potential for health promoting properties. Among these compounds, flavanones (such as hesperetin, naringenin, eriodictyol, isosakuranetin, and their respective glycosides), which occur in quantities ranging from ∼180 to 740 mg/L (depending on the Citrus species and cultivar) are responsible for many biological activities. These compounds support and enhance the body's defenses against oxidative stress and help the organism in the prevention of cardiovascular diseases, atherosclerosis, and cancer. Moreover, among other properties, they also show anti-inflammatory, antiviral, and antimicrobial activities. This review analyzes the biochemistry, pharmacology, and biology of Citrus flavanones, emphasizing the occurrence in Citrus fruits and juices and their bioavailability, structure-function correlations and ability to modulate signal cascades both in vitro and in vivo. © 2017 International Union of Biochemistry and Molecular Biology.

Abstract

The ubiquitin-proteasome pathway (UPP) is the main non-lysosomal proteolytic system responsible for degradation of most intracellular proteins, specifically damaged and regulatory proteins. The UPP is implicated in all aspects of the cellular metabolic networks including physiological or pathological conditions. Alterations in the components of the UPP can lead to stabilization of oncoproteins or augmented degradation of tumour suppressor favouring cancer appearance and progression. Polyphenols are natural compounds that can modulate proteasome activity or the expression of proteasome subunits. All together and due to the pleiotropic functions of UPP, there is a great interest in this proteasome system as a promising therapeutic target for the development of novel anti-cancer drugs. In the present review, the main features of the UPP and its implication in cancer development and progression are described, highlighting the importance of bioactive polyphenols that target the UPP as potential anti-cancer agents.
