Abstract: Aims. This article presents very-high-energy (VHE; E > 100?GeV) data from the ?-ray binary PSR?B1259-63 as taken during the years 2005, 2006 and before as well as shortly after the 2007 periastron passage. These data extend the knowledge of the lightcurve of this object to all phases of the 3.4?year binary orbit. The lightcurve constrains physical mechanisms present in this TeV source.Methods. Observations of VHE ?-rays with the HESS telescope array using the Imaging Atmospheric Cherenkov Technique were performed. The HESS?instrument features an angular resolution of < 0.1? and an energy resolution of < 20%. Gamma-ray events in an energy range of 0.5-70?TeV were recorded. From these data, energy spectra and lightcurve with a monthly time sampling were extracted.Results. VHE ?-ray emission from PSR?B1259-63?was detected with an overall significance of 9.5?standard deviations using 55h of exposure, obtained from April to August 2007. The monthly flux of ?-rays during the observation period was measured, yielding VHE lightcurve data for the early pre-periastron phase of the system for the first time. No spectral variability was found on timescales of months. The spectrum is described by a power law with a photon index of ? = 2.8 ?0.2stat?0.2sysand flux normalisation ?0= (1.1 ?0.1stat?0.2sys) ? 10-12?TeV-1?cm-2?s-1. PSR?B1259-63?was also monitored in 2005 and 2006, far from periastron passage, comprising 8.9?h and 7.5?h of exposure, respectively. No significant excess of ?-rays is seen in those observations.Conclusions. PSR B1259-63? has been re-confirmed as a variable TeV ?-ray emitter. The firm detection of VHE photons emitted at a true anomaly ??-0.35 of the pulsar orbit, i.e.?already ~50?days prior to the periastron passage, disfavors the stellar disc target scenario as a primary emission mechanism, based on current knowledge about the companion star's disc inclination, extension, and density profile.

Abstract: Aims. A detailed study of the spectrum and variability of the source HESS J1745-290 in the Galactic Center (GC) region using new data from the H.E.S.S. array of Cherenkov telescopes is presented. Flaring activity and quasi periodic oscillations (QPO) of HESS J1745-290 are investigated.Methods. The image analysis is performed with a combination of a semi-analytical shower model and the statistical moment-based Hillas technique. The spectrum and lightcurves of HESS J1745-290 are derived with a likelihood method based on a spectral shape hypothesis. Rayleigh tests and Fourier analysis of the H.E.S.S. GC signal are used to study the periodicity of the source.Results. With a three-fold increase in statistics compared to previous work, a deviation from a simple power law spectrum is detected for the first time. The measured energy spectrum over the three years 2004, 2005 and 2006 of data taking is compatible with both a power law spectrum with an exponential cut-off and a broken power law spectrum. The curvature of the energy spectrum is likely to be intrinsic to the photon source, as opposed to effects of interstellar absorption. The power law spectrum with an exponential cut-off is characterized by a photon index of 2.10 ? 0.04stat? 0.10systand a cut-off energy at 15.7 ? 3.4stat? 2.5systTeV. The broken power law spectrum exhibits spectral indices of 2.02 ? 0.08stat? 0.10systand 2.63 ? 0.14stat? 0.10systwith a break energy at 2.57 ? 0.19stat? 0.44systTeV. No significant flux variation is found. Increases in the ?-ray flux of HESS J1745-290 by at least a factor of two would be required for a 3? detection of a flare with time scales of an hour. Investigation of possible QPO activity at periods claimed to be detected in X-rays does not show any periodicities in the H.E.S.S. signal.

103. Effect of Lactobacillus acidophilusSupplementation of Milk Replacer on Preweaning Performance of Calves

Abstract: Forty Holstein-Friesian calves were used to evaluate the effect on young calves of daily dietary supplementation with Lactobacillus acidophilus. Calves were randomly assigned at 2 d of age to one of two treatments; 1) milk replacer with no additives or 2) milk replacer supplemented with 1ml (5 × l07) of viable L. acidophilusbacteria at each of two feedings per day. Milk replacer was reconstituted to 12% DM and fed at 10% of BW/d for the duration of the 6-wk trial. A commercial starter pellet was offered for ad libitum intake from 7 d of age. Treatment had no effect on actual BW at any stage or on total BW gain; however, average daily gain during wk 2 was affected by L. acidophilussupplementation. Calves receiving L. acidophilusmaintained initial BW, and the control calves lost BW until 2 wk of age, at an average rate of 112 g/d. Starter intake, total DMI, feed efficiency, and occurrence of diarrhea were unaffected by treatment. Therefore, L. acidophilussupplementation for calves fed milk replacer may be beneficial during the first 2 wk of life.

108. Barbiturate ingestion in three adult captive tigers (Panthera tigris) and concomitant fatal botulism of one

Abstract: Zoo animals, including tigers, have been reported to suffer from barbiturate intoxication, with pentabarbitone being most commonly recorded. Clinical signs range from mild ataxia to general anaesthesia with recovery over hours to days with several factors affecting hepatic barbiturate metabolism and tissue partitioning. Botulism is an often fatal intoxication in man, animals, birds and certain fish. The occurrence in carnivores is uncommon to rare, with only 2 reports found of botulism in felids. This report relates to 3 adult captive cohabiting tigers that simultaneously developed signs of abdominal discomfort, progressive ataxia, recumbency and comatose sleep resembling stage 2 anaesthesia, alternating with periods of distracted wakefulness and ataxic movements. These signs occurred 4 days after being fed the carcass of a horse that had ostensibly died of colic and not been euthanased. The male tiger that was the dominant animal in the feeding hierarchy was worst affected and had to be given intravenous fluids. The female that was lowest in hierarchy was unaffected. After 48-72 hours of treatment at the Onderstepoort Veterinary Academic Hospital the females could eat and made an uneventful recovery. The male tiger showed partial recovery but died during the night a few hours after drinking water on his return to the owner. Necropsy revealed severe oesophageal dilation and impaction with decaying grass; some of this material and water were present in the pharynx and trachea, and had been aspirated causing acute widespread bronchopneumonia. Colon content tested negative for common pesticides but, together with liver, tested positive for barbiturate. Serum taken on the day of admission had tested negative for barbiturate and the residual serum from the 3 animals later tested negative for botulinum toxin. Colon and oesophageal content from the male at necropsy were positive for Clostridium botulinum toxin type C by the mouse bioassay neutralisation test, confirming that this male had had concomitant barbiturate toxicity and botulism, and had succumbed to aspiration bronchopneumonia secondary to pharyngeal, laryngeal and oesophageal paralysis and oesophageal impaction.

Abstract: Ground-based gamma-ray astronomy has had a major breakthrough with the impressive results obtained using systems of imaging atmospheric Cherenkov telescopes. Ground-based gamma-ray astronomy has a huge potential in astrophysics, particle physics and cosmology. CTA is an international initiative to build the next generation instrument, with a factor of 5-10 improvement in sensitivity in the 100 GeV to 10 TeV range and the extension to energies well below 100 GeV and above 100 TeV. CTA will consist of two arrays (one in the north, one in the south) for full sky coverage and will be operated as open observatory. The design of CTA is based on currently available technology. Abstract: The Building Connections in Aged Care research was conducted in Tasmania the island state of Australia between 2003 and 2006, and funded by Australian Government Department of Health and Ageing and six participating residential aged care facilities (RACFs). It involved 61 nursing students who undertook a three week clinical placements in the facilities and 37 preceptors (mentors) who supported them during their placement. The project was undertaken over three stages: (1) scoping issues that impacted on teaching and learning in the six RACFs and their capacity to support nursing students (n=20) on a three week clinical placement; (2) a period of reporting, consultation and strategy development prior to implementing interventions to build capacity within the six RACFs to facilitate quality clinical placements for the next cohort of students (n=20), while concurrently mapping who students worked with and what they did during placements, and; (3) assessment of the sustainability of gains achieved in stage 2 with a third cohort of students (n=21). Stage 1 findings highlighted the limited capacity within aged care facilities to effectively support students. Of note, the study highlighted students lack of preparation for practice in a RACF, their poor experience of teaching and learning and the inadequacy of orientation programs, all of which had an extremely negative impact on their placement experience. On entry, 50% indicated a possible/definite willingness to work in the sector following graduation while on exit this figure did not improve. Stage 2 findings revealed a great improvement in student-stated willingness to work in an RACF following graduation (an increase from 50% on entry to 90% at completion), together with a significant improvement in facility orientation and preceptor capacity to support effective teaching and learning. Abstract: Consistent with reccomendations from the 'Building Connections in Aged Care Final Report (2005), the Australian Government Department of Health and Ageing funded the conduct of of a follow-up evaluation. The Building Connections in Aged Care Follow-up Evaluation, investigated two further cohorts of students on placements in the aged care facilities involved in the Building Connections project (intervention group), at 6 and 12 months post completion of Stage 3. The evaluation also included two cohorts of students on placement in aged care facilities, which had no prior involvement in the Building Connections project (control group). The findings highlighted a statistically significant improvement in the intervention group students attitude to working in aged care following graduation. No such improvement was evident in students on placement in the control facilities. These findings highlighted the sustainability of the improvement achieved as a result of the interventions implemented in the aged care facilities in earlier stages of the Building Connections project.

Abstract: The report addresses the findings of Stage one of the Building Connections in Aged Care project. In Stage one the primary intent was to scope the issues, which impact on teaching and learning and the capacity of the involved Residential Aged Care Facilities (RACFs) to support an educative agenda. Because the researchers had an ongoing and intense involvement with both the second year nursing students (N=20) participating 3 week clinical placements in six RACFs and nurses who worked with them as preceptors, many issues were revealed in the research discussions. Furthermore, the collaborative interactive methodology used to facilitate the project, supported the participants sense of ownership and their desire to flesh out and address the issues raised. Consequently, the findings presented in this report provide a unique insight into the operation of residential care facilities not previously documented in the literature, not least a lack of capacity within RACFs to effectively support students on placement.

Abstract: A key focus of the Building Connections in Aged Care project was to facilitate the development of quality clinical placements in aged care as a strategy to positively influence student attitudes to working in the sector. Consistent with the recommendations the outlined in the Stage 1 project report, Stage 2 of the project implemented a number of strategies to: address problems with the orientation of students to the RACFs; develop resources to support students in practice, and; promote a greater degree of continuity between students and their preceptors. The implementation of these strategies and associated changes had a major impact, which was no more obvious than the positive change in student attitude to working in the sector. The proejct findings documented in the Stage 2 report demonstrate that the implementation of the Stage 1 recommendations had a significant positive impact on students (n=20) stated career intentions. On commencement 55% of students indicated they would possibly not/definitely not have an interest in working in aged care following graduation. However, at the end of their first week in practice, following orientation to the facilities, 80% of students indicated a possible/definite interest in working in aged care following graduation. Moreover, at the completion of the practicum 90% of students indicated a possible/definite interest in working in aged care following graduation. The evaluation also demonstrated a threefold decrease in the number of students who held definite views that they would not work in aged care following graduation. The findings also demonstrated that in the context of a supportive environment, the presence of students provides a significant impetus for aged care nurses involvement in professional development activities.

Abstract: That the issue of ageing is going to be high on the agenda of Australian governments and community interests for at least the next four decades is clear and well understood. Despite successive waves of government-driven reforms in financing, planning, provision and standards, extending now over two decades, we still have serious problems of staff recruitment and retention in this area, problems which, on present indications, are likely to peak at about the same time in the not-too-distant future as the demand pressures reach a new level of intensity. There is still a measure of uncertainty about some dimensions of the aged care workforce despite recent scholarly attempts to clarify the situation (Richardson & Martin, 2004). However it can be agreed that rates of RN retirement and withdrawal for other reasons will exceed RN recruitment rates and thereby maintain pressure on the labour force supply side. That there are several causes of this declining trajectory is clear; that a widespread rejection of the actual or anticipated aged care clinical experience by student nurses is one of those several causes is equally certain.

Abstract: A key focus of the Building Connections in Aged Care project was to facilitate the development of quality clinical placements in aged care as a strategy to positively influence student attitudes to working in the sector. Consistent with the recommendations the outlined in the Stage 1 project report, Stage 2 of the project implemented a number of strategies to: address problems with the orientation of students to the RACFs; develop resources to support students in practice, and; promote a greater degree of continuity between students and their preceptors. The implementation of these strategies and associated changes had a major impact, which was no more obvious than the positive change in student attitude to working in the sector. The proejct findings documented in the Stage 2 report demonstrate that the implementation of the Stage 1 recommendations had a significant positive impact on students (n=20) stated career intentions. On commencement 55% of students indicated they would possibly not/definitely not have an interest in working in aged care following graduation. However, at the end of their first week in practice, following orientation to the facilities, 80% of students indicated a possible/definite interest in working in aged care following graduation. Moreover, at the completion of the practicum 90% of students indicated a possible/definite interest in working in aged care following graduation. The evaluation also demonstrated a threefold decrease in the number of students who held definite views that they would not work in aged care following graduation. The findings also demonstrated that in the context of a supportive environment, the presence of students provides a significant impetus for aged care nurses involvement in professional development activities.

Abstract: Consistent with reccomendations from the 'Building Connections in Aged Care Final Report (2005), the Australian Government Department of Health and Ageing funded the conduct of of a follow-up evaluation. The Building Connections in Aged Care Follow-up Evaluation, investigated two further cohorts of students on placements in the aged care facilities involved in the Building Connections project (intervention group), at 6 and 12 months post completion of Stage 3. The evaluation also included two cohorts of students on placement in aged care facilities, which had no prior involvement in the Building Connections project (control group). The findings highlighted a statistically significant improvement in the intervention group students attitude to working in aged care following graduation. No such improvement was evident in students on placement in the control facilities. These findings highlighted the sustainability of the improvement achieved as a result of the interventions implemented in the aged care facilities in earlier stages of the Building Connections project.

Abstract: The report addresses the findings of Stage one of the Building Connections in Aged Care project. In Stage one the primary intent was to scope the issues, which impact on teaching and learning and the capacity of the involved Residential Aged Care Facilities (RACFs) to support an educative agenda. Because the researchers had an ongoing and intense involvement with both the second year nursing students (N=20) participating 3 week clinical placements in six RACFs and nurses who worked with them as preceptors, many issues were revealed in the research discussions. Furthermore, the collaborative interactive methodology used to facilitate the project, supported the participants sense of ownership and their desire to flesh out and address the issues raised. Consequently, the findings presented in this report provide a unique insight into the operation of residential care facilities not previously documented in the literature, not least a lack of capacity within RACFs to effectively support students on placement.

Abstract: Aims. Recent theoretical predictions of the lowest very high energy (VHE) luminosity of SN 1006 are only a factor 5 below the previously published HESS upper limit, thus motivating further in-depth observations of this source.Methods. Deep observations at VHE energies (above 100 GeV) were carried out with the high energy stereoscopic system (HESS) of Cherenkov Telescopes from 2003 to 2008. More than 100 h of data have been collected and subjected to an improved analysis procedure.Results. Observations resulted in the detection of VHE γ-rays from SN 1006. The measured γ-ray spectrum is compatible with a power-law, the flux is of the order of 1% of that detected from the Crab Nebula, and is thus consistent with the previously established HESS upper limit. The source exhibits a bipolar morphology, which is strongly correlated with non-thermal X-rays.Conclusions. Because the thickness of the VHE-shell is compatible with emission from a thin rim, particle acceleration in shock waves is likely to be the origin of the γ-ray signal. The measured flux level can be accounted for by inverse Compton emission, but a mixed scenario that includes leptonic and hadronic components and takes into account the ambient matter density inferred from observations also leads to a satisfactory description of the multi-wavelength spectrum.

Abstract: Aims. Our aim is to study the very high energy (VHE; E>100GeV) ?-ray emission from BL?Lac objects and the evolution in time of their broad-band spectral energy distribution (SED).Methods. VHE observations of the high-frequency peaked BL?Lac object PKS?2005-489 were made with the High Energy Stereoscopic System (HESS) from 2004 through 2007. Three simultaneous multi-wavelength campaigns at lower energies were performed during the HESS data taking, consisting of several individual pointings with the XMM-Newtonand RXTE satellites.Results. A strong VHE signal, ~17stotal, from PKS?2005-489 was detected during the four years of HESS observations (90.3 h live time). The integral flux above the average analysis threshold of 400 GeV is ~3% of the flux observed from the Crab Nebula and varies weakly on time scales from days to years. The average VHE spectrum measured from ~300 GeV to ~5 TeV is characterized by a power law with a photon index, $\Gamma = 3.20\pm 0.16_{\rm stat}\pm 0.10_{\rm syst}$. At X-ray energies the flux is observed to vary by more than an order of magnitude between 2004 and 2005. Strong changes in the X-ray spectrum ($\Delta\Gamma_{\rm X} \approx 0.7$) are also observed, which appear to be mirrored in the VHE band.Conclusions. The SED of PKS?2005-489, constructed for the first time with contemporaneous data on both humps, shows significant evolution. The large flux variations in the X-ray band, coupled with weak or no variations in the VHE band and a similar spectral behavior, suggest the emergence of a new, separate, harder emission component in September 2005.

Abstract: Zoo animals, including tigers, have been reported to suffer from barbiturate intoxication, with pentabarbitone being most commonly recorded. Clinical signs range from mild ataxia to general anaesthesia with recovery over hours to days with several factors affecting hepatic barbiturate metabolism and tissue partitioning. Botulism is an often fatal intoxication in man, animals, birds and certain fish. The occurrence in carnivores is uncommon to rare, with only 2 reports found of botulism in felids. This report relates to 3 adult captive cohabiting tigers that simultaneously developed signs of abdominal discomfort, progressive ataxia, recumbency and comatose sleep resembling stage 2 anaesthesia, alternating with periods of distracted wakefulness and ataxic movements. These signs occurred 4 days after being fed the carcass of a horse that had ostensibly died of colic and not been euthanased. The male tiger that was the dominant animal in the feeding hierarchy was worst affected and had to be given intravenous fluids. The female that was lowest in hierarchy was unaffected. After 48-72 hours of treatment at the Onderstepoort Veterinary Academic Hospital the females could eat and made an uneventful recovery. The male tiger showed partial recovery but died during the night a few hours after drinking water on his return to the owner. Necropsy revealed severe oesophageal dilation and impaction with decaying grass; some of this material and water were present in the pharynx and trachea, and had been aspirated causing acute widespread bronchopneumonia. Colon content tested negative for common pesticides but, together with liver, tested positive for barbiturate. Serum taken on the day of admission had tested negative for barbiturate and the residual serum from the 3 animals later tested negative for botulinum toxin. Colon and oesophageal content from the male at necropsy were positive for Clostridium botulinum toxin type C by the mouse bioassay neutralisation test, confirming that this male had had concomitant barbiturate toxicity and botulism, and had succumbed to aspiration bronchopneumonia secondary to pharyngeal, laryngeal and oesophageal paralysis and oesophageal impaction. Abstract: Background: Roflumilast, an oral, once-daily phosphodiesterase 4 inhibitor, is currently in clinical development for the treatment of asthma. Objectives: This pilot study examined the effect of roflumilast on allergen-induced airway hyperresponsiveness (AHR) to histamine challenge and asthmatic response to allergen challenge. Methods: In a randomized, double-blind, 2-period, crossover trial, 13 patients with mild allergic asthma [mean forced expiratory volume in 1 s (FEV1) % predicted = 86%] received a single dose of oral roflumilast 1,000 μg or placebo. Patients were administered roflumilast 60 min before allergen challenge, and asthmatic responses were assessed via change in FEV1 ≤9 h after allergen challenge. AHR to histamine provocation was measured before and repeated 24 h after allergen provocation. Patients inhaled histamine in doubling concentrations until attaining a decrease in FEV1 of ≤20% (PC20FEV1). Results: Roflumilast had no detectable bronchodilator activity 60 min after administration. Roflumilast significantly attenuated AHR compared with placebo, with a mean change in pre- to postallergen challenge PC20FEV1 ratio of 1.23 ± 2.75 and 2.51 ± 2.95 for roflumilast and placebo, respectively (p = 0.002). During the late asthmatic response, roflumilast reduced the mean maximum decrease in FEV1 from 2 to 9 h after allergen challenge compared with placebo (p = 0.005). Additionally, FEV1 at 9 h after challenge was significantly higher in patients treated with roflumilast (p = 0.03). Early asthmatic responses to allergen challenge were not significantly reduced by the single dose of roflumilast. Conclusions: Roflumilast attenuated allergen-induced AHR in patients with mild asthma. These results support further investigation of roflumilast as an anti-inflammatory treatment of asthma. Abstract: Anaesthesia was required in an African male lion (Panthera leo) from the Pretoria Zoo for dental surgery. For immobilisation and transportation a combination of zolazepam and tiletamine were used at a dose of 3 mg/kg. Medetomidine was administered to improve relaxation. At the veterinary hospital the trachea was intubated, and halothane in oxygen administered for maintenance of anaesthesia during surgery. The mean (+/- SD) heart rate during anaesthesia was 58 (3) beats/min, arterial blood pressure 17.6 (1.7) kPa, and ventilation controlled at 10 breaths/min. Abstract: MCom (Computer Science and Information Systems), North-West University, Potchefstroom Campus, 2016 In this study, we are investigating how systems development methodologies and virtual software development teams work together to produce a better end product. Systems development methodologies have been defined as an approach that helps organisations to build their systems in a well-standardised and comprehensive manner in order to achieve common goals. This includes all the procedures or steps to be followed depending on the specific approach used by the individual organisation. A virtual team has been defined as a group of people, separated locally or internationally, working to achieve a common goal. Achieving a desired goal involve use of various technologies such as telephones, e-mails, video-conferencing, and any other effective communication modes. Human resource could include the most qualified candidates from inside or outside their companies who always form or reform a specific task team continuously. Organisations would also be able to deliver quality products and increase client’s satisfaction and build strong reporting relationships as team members report to various authorities. I followed a positivistic approach which entails a survey as a research method using a questionnaire for data collection from a large group of people. Statistical analysis was applied for data analysis. Research findings were very reliable and significant based on the test for validity of data. Techniques that were used to analyse data were descriptive analysis, factor analysis, reliability analysis, correlation analysis, cross-tabulations and regression analysis. Specific objectives of this study included identifying challenges and benefits faced by virtual software development teams. The results showed that even though there are still challenges; there are more benefits in virtualisation. For instance, challenges such as technology, culture, customs, personal conflict and distance between the teams occurred. Another objective was to evaluate the use (if any) and the suitability of current systems development methodologies for use by virtual software development teams. Descriptive measures using frequency and correlations and cross-tabulations were applied to the data obtained through questionnaire surveys. The results showed that virtual software teams are using a combination of traditional and agile systems development methodologies. The other objective was to define how many are using SDM; what SDM they are using and if none; and what are the reasons for not using them. The results showed that 35% of virtual teams adapted systems development methodology (SDM) on a project-to-project basis while 33% of participants used a general guideline for all projects, and 32% of participants used a standard procedure which is followed thoroughly for all projects. Another objective was to determine the success of projects developed by virtual software development teams. The results showed that the participants strongly felt that their projects were very successful in terms of customer satisfaction, product, competencies, process, communication and leadership. The last objective was to investigate the role (if any) of systems development methodologies in the success of those projects. The stepwise regression analysis was used to determine factors that influence the overall success of recent projects developed. The results for this study showed that support provided by SDM as production technology, support provided by SDM as control technology, gender_m (males) and team performance have a large influence on the success of projects developed by virtual software development teams. 147. A frog in hot water: the effect of temperature elevation on the adrenal stress response of an African amphibian.

Abstract: Amphibians, with their unique physiology and habitat requirements, are especially vulnerable to changes in environmental temperatures. While the activation of the physiological stress response can help to mitigate the impact of such habitat alteration, chronic production of elevated glucocorticoid levels can be deleterious in nature. There is no empirical evidence indicating the physiological response of African amphibians to temperature changes, where individuals are unable to emigrate away from potential stressors. To rectify this, we used the edible bullfrog ( Pyxicephalus edulis ) as a model species to determine the effect of elevated temperature on the adrenocortical response of the species using a recently established matrix. While a control group was kept at a constant temperature (25 °C) throughout the study period, an experimental group was exposed to control (25 °C) and elevated temperatures (30 °C). Mucous swabs were collected throughout the study period to determine dermal glucocorticoid (dGC) concentrations, as a proxy for physiological stress. In addition to this, individual body mass measurements were collected. The results showed that individuals within the experimental group who experienced increased temperatures had significantly elevated dGC levels compared to the control animals. Furthermore, there was a significant difference in the percentage mass change between experimental and control animals . These findings indicate the physiological sensitivity of the edible bullfrog to a thermal stressor in captivity. While this study shows the importance of proper amphibian management within the captive environment, it also highlights the coming danger of global climate change to this and similar amphibian species., Competing Interests: The authors declare there are no competing interests., (©2024 Scheun et al.)

148. Interactive effects of elevated temperature and Photobacterium swingsii infection on the survival and immune response of marine mussels (Perna canaliculus): A summer mortality scenario.

Abstract: The New Zealand Greenshell™ mussel (Perna canaliculus) is an economically important aquaculture species. Prolonged increases in seawater temperature above mussel thermotolerance ranges pose a significant threat to mussel survival and health, potentially increasing susceptibility to bacterial infections. Using challenge experiments, this study examined the combined effects of increased seawater temperature and bacterial (Photobacterium swingsii) infection on animal survival, haemocyte and biochemical responses of adult mussels. Mussels maintained at three temperatures (16, 20 and 24 °C) for seven days were either not injected (control), injected with sterile marine broth (injection control) or P. swingsii (challenged with medium and high doses) and monitored daily for five days. Haemolymph and tissue samples were collected at 24, 48, 72, 96, 120 h post-challenge and analysed to quantify bacterial colonies, haemocyte responses and biochemical responses. Mussels infected with P. swingsii exhibited mortalities at 20 and 24 °C, likely due to a compromised immune system, but no mortalities were observed when temperature was the only stressor. Bacterial colony counts in haemolymph decreased over time, suggesting bacterial clearance followed by the activation of immune signalling pathways. Total haemocyte counts and viability data supports haemocyte defence functions being stimulated in the presence of high pathogen loads at 24 °C. In the gill tissue, oxidative stress responses, measured as total antioxidant capacity and malondialdehyde (MDA) levels, were higher in infected mussels (compared to the controls) after 24h and 120h post-challenge at the lowest (16 °C) and highest temperatures (24 °C), indicating the presence of oxidative stress due to temperature and pathogen stressors. Overall, this work confirms that Photobacterium swingsii is pathogenic to P. canaliculus and indicates that mussels may be more vulnerable to bacterial pathogens under conditions of elevated temperature, such as those predicted under future climate change scenarios., Competing Interests: Declaration of competing interest The authors declare no conflict of interest., (Copyright © 2024 The Authors. 149. Quantification of Photobacterium swingsii and characterisation of disease progression in the New Zealand Greenshell™ mussel, Perna canaliculus.

Abstract: Greenshell™ mussels (Perna canaliculus) are endemic to New Zealand and support the largest aquaculture industry in the country. Photobacterium swingsii was isolated and identified from moribund P. canaliculus mussels following a mass mortality event. In this study, a challenge experiment was used to characterise, detect, and quantify P. swingsii in adult P. canaliculus following pathogen exposure via injection into the adductor muscle. A positive control (heat-killed P. swingsii injection) was included to account for the effects of injection and inactive bacterial exposure. Survival of control and infected mussels remained 100% during 72-hour monitoring period. Haemolymph was sampled for bacterial colony counts and haemocyte flow cytometry analyses; histology sections were obtained and processed for histopathological assessments; and adductor muscle, gill, digestive gland were sampled for quantitative polymerase chain reaction (PCR) analyses, all conducted at 12, 24, 48 h post-challenge (hpc). The most profound effects of bacterial injection on mussels were seen at 48 hpc, where mussel mortality, haemocyte counts and haemolymph bacterial colony forming were the highest. The quantification of P. swingsii via qPCR showed highest levels of bacterial DNA at 12 hpc in the adductor muscle, gill, and digestive gland. Histopathological observations suggested a non-specific inflammatory response in all mussels associated with a general stress response. This study highlights the physiological effects of P. swingsii infection in P. canaliculus mussels and provides histopathological insight into the tissue injury caused by the action of injection into the adductor muscle. 150. Histopathological investigation of four populations of abalone (Haliotis iris) exhibiting divergent growth performance.

Abstract: The black-foot abalone (pāua), Haliotis iris, is a unique and valuable species to New Zealand with cultural importance for Māori. Abalone are marine gastropods that can display a high level of phenotypic variation, including slow-growing or 'stunted' variants. This investigation focused on identifying factors that are associated with growth performance, with particular interest in the slow-growing variants. Tissue alterations in H. iris were examined using histopathological techniques, in relation to growth performance, contrasting populations classified by commercial harvesters as 'stunted' (i.e., slow-growing) and 'non-stunted' (i.e., fast-growing) from four sites around the Chatham Islands (New Zealand). Ten adults and 10 sub-adults were collected from each of the four sites and prepared for histological assessment of condition, tissue alterations, presence of food and presence of parasites. The gut epithelium connective tissue, digestive gland, gill lamellae and right kidney tissues all displayed signs of structural differences between the slow-growing and fast-growing populations. Overall, several factors appear to be correlated to growth performance. The individuals from slow-growing populations were observed to have more degraded macroalgal fragments in the midgut, increased numbers of ceroid granules in multiple tissues, as well as increased prevalence of birefringent mineral crystals and haplosporidian-like parasites in the right kidney. The histopathological approaches presented here complement anecdotal field observations of reduced seaweed availability and increased sand incursion at slow-growing sites, while providing an insight into the health of individual abalone and sub-populations.
