The accumulation of dysfunctional mitochondria has been implicated in aging, but a deeper understanding of mitochondrial dynamics and mitophagy during aging is missing. Here, we show that upregulating Drp1-a Dynamin-related protein that promotes mitochondrial fission-in midlife, prolongs Drosophila lifespan and healthspan. We find that short-term induction of Drp1, in midlife, is sufficient to improve organismal health and prolong lifespan, and observe a midlife shift toward a more elongated mitochondrial morphology, which is linked to the accumulation of dysfunctional mitochondria in aged flight muscle. Promoting Drp1-mediated mitochondrial fission, in midlife, facilitates mitophagy and improves both mitochondrial respiratory function and proteostasis in aged flies. Finally, we show that autophagy is required for the anti-aging effects of midlife Drp1-mediated mitochondrial fission. Our findings indicate that interventions that promote mitochondrial fission could delay the onset of pathology and mortality in mammals when applied in midlife.Mitochondrial fission and fusion are important mechanisms to maintain mitochondrial function. Here, the authors report that middle-aged flies have more elongated, or 'hyper-fused' mitochondria, and show that induction of mitochondrial fission in midlife, but not in early life, extends the health and life of flies.

Conventional methods to engineer electroconductive hydrogels (ECHs) through the incorporation of conductive nanomaterials and polymers exhibit major technical limitations. These are mainly associated with the cytotoxicity, as well as poor solubility, processability, and biodegradability of their components. Here, we describe the engineering of a new class of ECHs through the functionalization of non-conductive polymers with a conductive choline-based bio-ionic liquid (Bio-IL). Bio-IL conjugated hydrogels exhibited a wide range of highly tunable physical properties, remarkable in vitro and in vivo biocompatibility, and high electrical conductivity without the need for additional conductive components. The engineered hydrogels could support the growth and function of primary cardiomyocytes in both two dimentinal (2D) and three dimensional (3D) cultures in vitro. Furthermore, they were shown to be efficiently biodegraded and possess low immunogenicity when implanted subcutaneously in rats. Taken together, our results suggest that Bio-IL conjugated hydrogels could be implemented and readily tailored to different biomedical and tissue engineering applications.

Ageing results in loss of tissue homeostasis across taxa. In the intestine of Drosophila melanogaster, ageing is correlated with an increase in intestinal stem cell (ISC) proliferation, a block in terminal differentiation of progenitor cells, activation of inflammatory pathways, and increased intestinal permeability. However, causal relationships between these phenotypes remain unclear. Here, we demonstrate that ageing results in altered localization and expression of septate junction proteins in the posterior midgut, which is quite pronounced in differentiated enterocytes (ECs) at tricellular junctions (TCJs). Acute loss of the TCJ protein Gliotactin (Gli) in ECs results in increased ISC proliferation and a block in differentiation in intestines from young flies, demonstrating that compromised TCJ function is sufficient to alter ISC behaviour in a non-autonomous manner. Blocking the Jun N-terminal kinase signalling pathway is sufficient to suppress changes in ISC behaviour, but has no effect on loss of intestinal barrier function, as a consequence of Gli depletion. Our work demonstrates a pivotal link between TCJs, stem cell behaviour, and intestinal homeostasis and provides insights into causes of age-onset and gastrointestinal diseases.

Working as an administrator in higher education often requires a graduate degree in order to qualify for entry-level positions. Yet, graduate programmes in higher education are often criticised for not adequately training students for their first administrative responsibilities. In this qualitative study, researchers asked a wide range of higher education administrators to identify specific skills and tasks they observed as challenging to new professionals that needed to be addressed as part of their training. Findings suggest that new professionals may need guidance on navigating and understanding institutional culture; performing required tasks and processes; adjusting their own behaviour, expectations and performance based on feedback and experiences; managing and supervising personnel; and making sound decisions without direct guidance. Implications for preparation programmes and administrative practice are shared.

Context: Mentorship has been identified as a primary organizational socialization tactic and has been directly associated with transition to practice. Objective: Understand how the mentoring relationship develops for the newly credentialed athletic trainer during the first year of clinical practice. Design: Grounded theory. Setting: Athletic training practice settings. Patients or Other Participants: Thirteen athletic trainers, who graduated from a professional master's program, certified between February and July of 2016, and obtained employment between July to August of 2016, participated in this study (6 female, 7 male, 26 6 [plus or minus] 3 years; work settings included professional sports, college, secondary and middle school, and clinic). Data saturation was met. Main Outcome Measure(s): Semistructured phone interviews were conducted with all participants during 3 specific time points (3, 8, and 12 months posthire). All interviews were transcribed verbatim and coded following the steps of a grounded theory study. Credibility strategies included researcher triangulation and peer review. Results: All 13 participants recognized a mentor, someone who they believed serve in a mentoring capacity during their transition into clinical practice as a newly credentialed athletic trainer. The development of the mentoring relationship for the newly credentialed athletic trainer appeared to be characterized by (1) identification of a mentor who had experience and knowledge, (2) a recognition of the need to have support and continued avenues for growth as a health care professional, (3) an informal initiation of the relationship through a professional relationship by the mentee, and (4) communication that overtime was reduced in frequency. Conclusions: Mentors provide career support and professional growth. Newly credentialed athletic trainers should seek out mentors who can support their continued development. The informal relationship appears to be of the greatest importance during the first few months of practice, and then once comfort and self-confidence improves, the frequency of communication is reduced.

Objective: Human immunodeficiency virus (HIV)-associated neurocognitive disorder (HAND) prevalence is expected to increase in East Africa as treatment coverage increases, survival improves, and this population ages. This study aimed to better understand the current cognitive phenotype of this newly emergent population of older combination antiretroviral therapy (cART)-treated people living with HIV (PLWH), in which current screening measures lack accuracy. This will facilitate the refinement of HAND cognitive screening tools for this setting. Method: This is a secondary analysis of 253 PLWH aged ≥50 years receiving standard government HIV clinic follow-up in Kilimanjaro, Tanzania. They were evaluated with a detailed locally normed low-literacy neuropsychological battery annually on three occasions and a consensus panel diagnosis of HAND by Frascati criteria based on clinical evaluation and collateral history. Results: Tests of verbal learning and memory, categorical verbal fluency, visual memory, and visuoconstruction had an area under the receiver operating characteristic curve >0.7 for symptomatic HAND (s-HAND) (0.70–0.72; p < 0.001 for all tests). Tests of visual memory, verbal learning with delayed recall and recognition memory, psychomotor speed, language comprehension, and categorical verbal fluency were independently associated with s-HAND in a logistic mixed effects model (p < 0.01 for all). Neuropsychological impairments varied by educational background. Conclusions: A broad range of cognitive domains are affected in older, well-controlled, East African PLWH, including those not captured in widely used screening measures. It is possible that educational background affects the observed cognitive impairments in this setting. Future screening measures for similar populations should consider assessment of visual memory, verbal learning, language comprehension, and executive and motor function. [ABSTRACT FROM AUTHOR]

Mountain regions harbor unique and rich biodiversity, forming an important part of our global life support system. This rich biodiversity underpins the ecological intactness and functioning of mountain ecosystems, which are imperative for the provision of key ecosystem services. A considerable amount of data are required to assess ecological intactness and ecosystem functioning and, given the profound anthropogenic pressures many mountain regions are being subjected to, are urgently needed. However, data on mountain biodiversity remain lacking. The essential biodiversity variables (EBVs) framework can help focus efforts related to detecting, investigating, predicting, and managing global biodiversity change, but has not yet been considered in the context of mountains. Here, we review key biological processes and physical phenomena that strongly influence mountain biodiversity and ecosystems and elucidate their associations with potential mountain EBVs. We identify seven EBVs of highest relevance for tracking and understanding the most critical drivers and responses of mountain biodiversity change. If they are implemented, the selected EBVs will contribute useful information to inform management and policy interventions seeking to halt mountain biodiversity loss and maintain functional mountain ecosystems. This paper discusses modelling and numerical issues in the simulation of the Landau-de Gennes (LdG) model of nematic liquid crystals (LCs) with cholesteric effects. We propose a fully implicit, (weighted) L2 gradient flow for computing energy minimizers of the LdG model, and note a time-step restriction for the flow to be energy decreasing. Furthermore, we give a mesh size restriction, for finite-element discretizations, that is critical to avoid spurious numerical artifacts in discrete minimizers, particularly when simulating cholesteric LCs that exhibit 'twist.' Furthermore, we perform a computational exploration of the model and present several numerical simulations in three dimensions, on both slab geometries and spherical shells, with our finite-element method. The simulations are consistent with experiments, illustrate the richness of the cholesteric model, and demonstrate the importance of the mesh size restriction. [ABSTRACT FROM AUTHOR]

Transcriptome and genome data from twenty stony coral species and a selection of reference bilaterians were studied to elucidate coral evolutionary history. We identified genes that encode the proteins responsible for the precipitation and aggregation of the aragonite skeleton on which the organisms live, and revealed a network of environmental sensors that coordinate responses of the host animals to temperature, light, and pH. Furthermore, we describe a variety of stress-related pathways, including apoptotic pathways that allow the host animals to detoxify reactive oxygen and nitrogen species that are generated by their intracellular photosynthetic symbionts, and determine the fate of corals under environmental stress. Some of these genes arose through horizontal gene transfer and comprise at least 0.2% of the animal gene inventory. Our analysis elucidates the evolutionary strategies that have allowed symbiotic corals to adapt and thrive for hundreds of millions of years.

This article evaluates two alternative standards for resetting property assessments on title transfer in property tax systems like Oregon’s and California’s that are explicitly designed to protect property owners against rapid, unanticipated increases in their tax bills, California’s reset to market and one in which the property is given a new assessed value based on its market value multiplied by a ratio that reflects the average relationship between assessed value and market value within a jurisdiction. We find that the latter promotes assessment quality better than the former and probably mitigates lock-in.
