Abstract Current methods for the early detection and minimal residual disease (MRD) monitoring of urothelial carcinoma (UC) are invasive and/or possess suboptimal sensitivity. We developed an efficient workflow named urine tumor DNA multidimensional bioinformatic predictor (utLIFE). Using UC-specific mutations and large copy number variations, the utLIFE-UC model was developed on a bladder cancer cohort (n = 150) and validated in The Cancer Genome Atlas (TCGA) bladder cancer cohort (n = 674) and an upper tract urothelial carcinoma (UTUC) cohort (n = 22). The utLIFE-UC model could discriminate 92.8% of UCs with 96.0% specificity and was robustly validated in the BLCA_TCGA and UTUC cohorts. Abstract Autism spectrum disorder (ASD) affects 1–2% of all children and poses a great social and economic challenge for the globe. As a highly heterogeneous neurodevelopmental disorder, the development of its treatment is extremely challenging. Multiple pathways have been linked to the pathogenesis of ASD, including signaling involved in synaptic function, oxytocinergic activities, immune homeostasis, chromatin modifications, and mitochondrial functions. Here, we identify secretagogin (SCGN), a regulator of synaptic transmission, as a new risk gene for ASD. Two heterozygous loss-of-function mutations in SCGN are presented in ASD probands. Deletion of Scgn in zebrafish or mice leads to autism-like behaviors and impairs brain development. Mechanistically, Scgn deficiency disrupts the oxytocin signaling and abnormally activates inflammation in both animal models. Both ASD probands carrying Scgn mutations also show reduced oxytocin levels. Importantly, we demonstrate that the administration of oxytocin and anti-inflammatory drugs can attenuate ASD-associated defects caused by SCGN deficiency. Altogether, we identify a convergence between a potential autism genetic risk factor SCGN, and the pathological deregulation in oxytocinergic signaling and immune responses, providing potential treatment for ASD patients suffering from SCGN deficiency. Our study also indicates that it is critical to identify and stratify ASD patient populations based on their disease mechanisms, which could greatly enhance therapeutic success.

Alzheimer’s disease (AD) is a chronic and progressive neurodegenerative disease and clinically manifests with cognitive decline and behavioral disabilities. Over the past years, mounting studies have demonstrated that the inflammatory response plays a key role in the onset and development of AD, and neuroinflammation has been proposed as the third major pathological driving factor of AD, ranking after the two well-known core pathologies, amyloid β (Aβ) deposits and neurofibrillary tangles (NFTs). Epigenetic mechanisms, referring to heritable changes in gene expression independent of DNA sequence alterations, are crucial regulators of neuroinflammation which have emerged as potential therapeutic targets for AD. Upon regulation of transcriptional repression or activation, epigenetic modification profiles are closely involved in inflammatory gene expression and signaling pathways of neuronal differentiation and cognitive function in central nervous system disorders. In this review, we summarize the current knowledge about epigenetic control mechanisms with a focus on DNA and histone modifications involved in the regulation of inflammatory genes and signaling pathways in AD, and the inhibitors under clinical assessment are also discussed.

Introduction: Phellodendri Chinensis Cortex is a necessary part of healthcare for its significant clinical efficacy. Raw and processed Phellodendri Chinensis Cortex is both documented in the Chinese Pharmacopoeia (2015). After processing, the therapeutic effects are believed to differ according to traditional Chinese medicine theories. However, the chemical mechanism responsible for this processing, according to traditional Chinese medicine theories, is still not clear.Methods: In this study, the therapeutic effects of various ions were examined based on traditional Chinese medicine theories by ultra-high performance liquid chromatography-hybrid quadrupole-Orbitrap mass spectrometry (UHPLC-Q-Orbitrap MS) coupled with multivariate statistical analysis, such as principal component analysis (PCA) and orthogonal partial least squares discriminant analysis (OPLS-DA), to comprehensively compare the differences between raw and processed Phellodendri Chinensis Cortex for the first time.Results: A total of 48 compounds were screened, out and 10 of them simultaneously transformed with significant variation in processed products compared with raw materials. It was illustrated that the contents of berberine, palmatine, jatrorrhizine, magnoflorine, menisperine, phellodendrine, tetrahydrojatrorrhizine, and tetrahydropalmatine decreased, while the compounds of berberrubine and fernloylquinic acid methyl ester newly appeared in processed herbs. This is likely to be related to the conversion of ingredients during processing.Discussion: Altogether, the fact that quality markers have been successfully identified to differentiate processed Phellodendri Chinensis Cortex from raw materials suggests that this approach could be used for the investigation of chemical transformation mechanisms involved in the processing of herbal medicine.

Objective: The rotational speed of the roller of the rice mill was optimized to reduce the crushing rate of rice grains in the process of whitening. Methods: The extrusion filling model was established by using SolidWorks software, and the collision contact model of single rice grain and two rice grains was established by using the particle bond model in EDEM software. The collision process of rice grains with different moisture content at different falling speeds was simulated, and the collision results of corresponding rice grains were observed. Results: There were three critical states of rice grains in the process of collision, which were intact rice grains, slightly broken rice grains and broken rice grains. Under the set moisture content range, the crushing rate of rice grains increased with the increase of moisture content and collision velocity, and the critical crushing rates of intact rice grains in the collision model of single grain and two grains were similar, which were 23.5 m/s and 22.0 m/s, respectively. Conclusion: The speed and moisture content have a significant effect on the crushing morphology of rice grains, and when the moisture content is 10.6%, the maximum speed of non-crushing of rice grains is 22.0 m/s, at this time, the crushing rate of rice grains is small and meets the demand of milling.

Abstract Human sideroflexin 2 (SFXN2) belongs to the SFXN protein family, which is a mitochondrial outer membrane protein involved in mitochondrial iron metabolism. Mitochondria are indispensable for cellular energy production and iron metabolism. However, it remains elusive how SFXN2 modulates mitochondrial homeostasis and cellular iron metabolism in multiple myeloma (MM). In this study, we first found that SFXN2 was significantly elevated and correlated to poor outcomes in MM patients from clinical datasets. SFXN2 overexpression promoted MM cell proliferation and suppressed starvation-induced autophagy/mitophagy, while SFXN2 knockdown aggravated mitochondria damage and autophagic processes in ARP1 and H929 MM cell lines. Furthermore, inhibition of SFXN2 exerted effectively anti-myeloma activity in vivo by using myeloma xenograft model. Mechanism studies indicated that heme oxygenase 1 (HO1) with anti-oxidant function contributed to the process of autophagy suppression and cellular proliferation mediated by SFXN2. Our study revealed the critical role of SFXN2 in regulating mitochondrial bioenergetics, mitophagy, cellular iron metabolism, and redox homeostasis in interconnected and intricate way. Collectively, these findings not only provide insights into the metabolic reprogramming of tumor cells, but also highlight the therapeutic potential of SFXN2 in combination with iron metabolism as target for prognosis and treatment in MM patients.

Abstract Classical Monte Carlo simulation of the Heisenberg model poorly describes many thermodynamic phenomena due to its neglect of the quantum nature of spins. Alternatively, we discuss how to semiclassically approach the quantum problem and demonstrate a simple method for introducing a locally approximate form of spin quantization. While the procedure underestimates magnetic short-range order, our results suggest a simple correction for recovering realistic spin–spin correlations above the critical temperature. Moreover, ensemble fluctuations are found to provide reasonably accurate thermodynamics, largely reproducing quantum mechanically calculated heat capacities and experimental magnetometry for ferromagnetic Fe and antiferromagnetic RbMnF3. Extensions of the method are proposed to address remaining inaccuracies.

Co3O4 spinel oxides have manifested promising activity toward the oxygen evolution reaction (OER) through effective modifications. For them to become top electrocatalysts, however, accurate accounts of the catalytic kinetics are essential to gain a deep understanding of the activity promotion mechanisms. Herein, we use a newly proposed kinetic model based on energetic span as the rate-determining term for the electrocatalytic reaction to throw light on the promotion mechanism of Co3O4 interfaced with nickel hydroxides (NiOxHy) for the OER. We find that depending on the electrode potential, the OER kinetics at the designed interface between Co3O4 and NiOxHy are boosted in entirely different ways. As a result, the OER can occur at a lower onset potential as well as a low Tafel slope. This work emphasizes the benefit of using rational theoretical models for electrocatalyst design.

Background: The functional integrity of the meniscus continually decreases with age, leading to meniscal degeneration and gradually developing into osteoarthritis (OA). In this study, we identified diagnostic markers and potential mechanisms of action in aging-related meniscal degeneration through bioinformatics and experimental verification.Methods: Based on the GSE98918 dataset, common differentially expressed genes (co-DEGs) were screened using differential expression analysis and the WGCNA algorithm, and enrichment analyses based on Gene Ontology (GO) terms and Kyoto Encyclopedia of Genes and Genomes (KEGG) pathways were further performed. Next, the co-DEGs were imported into the STRING database and Cytoscape to construct a protein‒protein interaction (PPI) network and further validated by three algorithms in cytoHubba, receiver operating characteristic (ROC) curve analysis and the external GSE45233 dataset. Moreover, the diagnostic marker lactotransferrin (LTF) was verified in rat models of senescence and replicative cellular senescence via RT‒qPCR, WB, immunohistochemistry and immunofluorescence, and then the potential molecular mechanism was explored by loss of function and overexpression of LTF.Results: According to the analysis of the GSE98918 dataset, we identified 52 co-DEGs (42 upregulated genes and 10 downregulated genes) in the OA meniscus. LTF, screened out by Cytoscape, ROC curve analysis in the GSE98918 dataset and another external GSE45233 dataset, might have good predictive power in meniscal degeneration. Our experimental results showed that LTF expression was statistically increased in the meniscal tissue of aged rats (24 months) and senescent passage 5th (P5) meniscal cells. In P5 meniscal cells, LTF knockdown inhibited the NF-κB signaling pathway and alleviated senescence. LTF overexpression in passage 0 (P0) meniscal cells increased the expression of senescence-associated secretory phenotype (SASP) and induced senescence by activating the NF-κB signaling pathway. However, the senescence phenomenon caused by LTF overexpression could be reversed by the NF-κB inhibitor pyrrolidine dithiocarbamate (PDTC).Conclusion: For the first time, we found that increased expression of LTF was observed in the aging meniscus and could induce meniscal senescence and degeneration by activating the NF-κB signaling pathway. These results revealed that LTF could be a potential diagnostic marker and therapeutic target for age-related meniscal degeneration.

Product reviews provide crucial information for both consumers and businesses, offering insights needed before purchasing a product or service. However, existing sentiment analysis methods, especially for Chinese language, struggle to effectively capture contextual information due to the complex semantics, multiple sentiment polarities, and long-term dependencies between words. In this paper, we propose a sentiment classification method based on the BiLSTM algorithm to address these challenges in natural language processing. Self-Attention-CNN BiLSTM (SAC-BiLSTM) leverages dual channels to extract features from both character-level embeddings and word-level embeddings. It combines BiLSTM and Self-Attention mechanisms for feature extraction and weight allocation, aiming to overcome the limitations in mining contextual information. Experiments were conducted on the onlineshopping10cats dataset, which is a standard corpus of e-commerce shopping reviews available in the ChineseNlpCorpus 2018. The experimental results demonstrate the effectiveness of our proposed algorithm, with Recall, Precision, and F1 scores reaching 0.9409, 0.9369, and 0.9404, respectively.
