Abstract

An investigation of the iron (Fe) perturbations needed to stimulate diatom production requires an assessment of the lowest Fe concentration whereby uptake still allows maximum cellular growth rates and of what Fe flux to surface waters is then required to sustain high diatom productivity once the concentration threshold is exceeded. Research in this area has concluded that climate forcing has definitely had an impact on productivity in the eastern equatorial Pacific. Furthermore, equatorial sediment records give an insight into these impacts. However, some element of this cycling may have been fuelled by localized tectonic processes in the Papua New Guinea island-arc area.

116. Apparatus for Sampling Surface Contamination

Abstract

An apparatus denoted a swab device has been developed as a convenient means of acquiring samples of contaminants from surfaces and suspending the samples in liquids. (Thereafter, the liquids can be dispensed, in controlled volumes, into scientific instruments for analysis of the contaminants.) The swab device is designed so as not to introduce additional contamination and to facilitate, simplify, and systematize the dispensing of controlled volumes of liquid into analytical instruments. The swab device is a single apparatus into which are combined all the equipment and materials needed for sampling surface contamination. The swab device contains disposable components stacked together on a nondisposable dispensing head. One of the disposable components is a supply cartridge holding a sufficient volume of liquid for one complete set of samples. (The liquid could be clean water or another suitable solvent, depending on the application.) This supply of liquid is sealed by Luer valves. At the beginning of a sampling process, the user tears open a sealed bag containing the supply cartridge. A tip on the nondisposable dispensing head is engaged with a Luer valve on one end of the supply cartridge and rotated, locking the supply cartridge on the dispensing head and opening the valve. The swab tip includes a fabric swab that is wiped across the surface of interest to acquire a sample. A sealed bag containing a disposable dispensing tip is then opened, and the swab tip is pushed into the dispensing tip until seated. The dispensing head contains a piston that passes through a spring-loaded lip seal. The air volume displaced by this piston forces the liquid out of the supply cartridge, over the swab, and into the dispensing tip. The piston is manually cycled to enforce oscillation of the air volume and thereby to cause water to flow to wash contaminants from the swab and cause the resulting liquid suspension of contaminants to flow into the dispensing tip. After several cycles to ensure adequate mixing, liquid containing the suspended contaminant sample is dispensed. The disposable components are then removed from the dispensing head, which may then be reused with a fresh set of disposable components.

Abstract

The continual and long-term habitation of enclosed environments, such as Antarctic stations, nuclear submarines and space stations, raises unique engineering, medical and operational challenges. There is no easy way out and no easy way to get supplies in. This situation elevates the importance of monitoring technology that can rapidly detect events within the habitat that affect crew safety such as fire, release of toxic chemicals and hazardous microorganisms. Traditional methods to monitor microorganisms on the International Space Station (ISS) have consisted of culturing samples for 3-5 days and eventual sample return to Earth. To augment these culture methods with new, rapid molecular techniques, we developed the Lab-on-a-Chip Application Development - Portable Test System (LOCAD-PTS). The system consists of a hand-held spectrophotometer, a series of interchangeable cartridges and a surface sampling/dilution kit that enables crew to collect samples and detect a range of biological molecules, all within 15 minutes. LOCAD-PTS was launched to the ISS aboard Space Shuttle Discovery in December 2006, where it was operated for the first time during March-May 2007. The surfaces of five separate sites in the US Lab and Node 1 of ISS were analyzed for endotoxin, using cartridges that employ the Limulus Amebocyte Lysate (LAL) assay; results of these tests will be presented. LOCAD-PTS will remain permanently onboard ISS with new cartridges scheduled for launch in February and October of 2008 for the detection of fungi (Beta-glucan) and Gram-positive bacteria (lipoteichoic acid), respectively.

Abstract

Subduction associated with mesoscale eddies is an important but difficult to observe process that can efficiently export carbon and oxygen to the mesopelagic zone (100-1000 db). Using a novel BGC-Argo dataset covering the western North Pacific (20-50° N, 120-180° E), we identified imprints of episodic subduction using anomalies in dissolved oxygen and spicity, a water mass marker. These subduction patches were present in 4.0 % (288) of the total profiles (7,120) between 2008 and 2019, situated mainly in the Kuroshio Extension region between March and August (70.6 %). Unlike eddy subduction processes observed at higher latitudes, roughly half (52 %) of these episodic events injected carbon- and oxygen-enriched waters below the annual permanent thermocline depth (450 db), with > 20 % occurring deeper than 600 db. Export rates within these subductions are estimated to be on the order of 85-159 mg C m-2 day-1 and 175 to 417 mg O2 m-2 day-1. These mesoscale events would markedly increase carbon removal above that due to biological gravitational settling as well as oxygen ventilation in the region, both helping to support the nutritional and metabolic demands of mesopelagic organisms. Climate-driven patterns of increasing eddy kinetic energies in this region imply that the magnitude of these processes will grow in the future, meaning that these unexpectedly effective small-scale subduction processes need to be better constrained in global climate and biogeochemical models. Abstract

Global demand for macroalgal and microalgal foods is growing, and algae are increasingly being consumed for functional benefits beyond the traditional considerations of nutrition and health. There is substantial evidence for the health benefits of algal-derived food products, but there remain considerable challenges in quantifying these benefits, as well as possible adverse effects. First, there is a limited understanding of nutritional composition across algal species, geographical regions, and seasons, all of which can substantially affect their dietary value. The second issue is quantifying which fractions of algal foods are bioavailable to humans, and which factors influence how food constituents are released, ranging from food preparation through genetic differentiation in the gut microbiome. Third is understanding how algal nutritional and functional constituents interact in human metabolism. Superimposed considerations are the effects of harvesting, storage, and food processing techniques that can dramatically influence the potential nutritive value of algal-derived foods. We highlight this rapidly advancing area of algal science with a particular focus on the key research required to assess better the health benefits of an alga or algal product. There are rich opportunities for phycologists in this emerging field, requiring exciting new experimental and collaborative approaches.

Abstract

Investigators worldwide are preparing experiments to test the influence of low gravity found in space on the growth of many crystalline materials. However, power limitations prevent existing space crystal growth furnaces from being able to process samples any larger than about 2 cm, and in addition, the background microgravity levels found on the Space Shuttle are not low enough to significantly benefit samples much larger than 2 cm. This paper describes a novel concept of a free-flying platform utilizing well-established solar furnace technology to enable materials processing in space experiments on large-diameter crystals. The conceptual design of this Solar Furnace Satellite is described along with its operational scenario and the anticipated g levels.

Abstract

With the nexus of climate change, globalization, and dwindling marine fishes, more attention is being paid to the increasing global burden of ciguatera fish poisoning (CFP) and the development of appropriate strategies for intervention, especially in endemic areas such as the South Pacific Islands and the Caribbean Sea. CFP is a non-bacterial food poisoning event with outbreaks around the world in which fish concentrate a potent neurotoxin (ciguatoxin) in their organs and tissues from inadvertent feeding on toxic benthic algal species. Outbreaks are frequent in endemic areas in the tropics and semitropics with developed corals and benthic fish fisheries. Occurrences are also present in non-endemic areas where contaminated fish are exported for consumption. Local populations on tropical or semitropical islands are particularly vulnerable to this poisoning through the consumption of ciguatoxincontaminated fish. Here, we consider the pecuniary costs of population exposure to CFP using two published studies as a template for developing a case study of the eco-socio-economic cost model in island nations. Abstract

Typhoons are assumed to stimulate ocean primary production through the upward mixing of nutrients into the surface ocean, based largely on observations of increased surface chlorophyll concentrations following the passage of typhoons. This surface chlorophyll enhancement, seen on occasion by satellites, more often is undetected due to intense cloud coverage. Daily data from a BGC-Argo profiling float revealed the upper-ocean response to Typhoon Trami in the Northwest Pacific Ocean. Temperature and chlorophyll changed rapidly, with a significant drop in sea surface temperature and surge in surface chlorophyll associated with strong vertical mixing, which was only partially captured by satellite observations. However, no net increase in vertically integrated chlorophyll was observed during Typhoon Trami or in its wake. Contrary to the prevailing dogma, the results show that typhoons likely have limited effect on net ocean primary production. Observed surface chlorophyll enhancements during and immediately following typhoons in tropical and subtropical waters are more likely associated with surface entrainment of deep chlorophyll maxima. Moreover, the findings demonstrate that remote sensing data alone can overestimate the impact of storms on primary production in all oceans. Full understanding of the impact of storms on upper ocean productivity can only be achieved with ocean observing robots dedicated to high-resolution temporal sampling in the path of storms.
