1. Comparisons of the effects of 4 and 16 weeks of exercise were made on; cardiac output, stroke volume, heart rate, left intraventricular systolic and diastolic pressures, dP/dt, and heart calcium in the Bio 14.6 cardiomyopathic and F1 B hamsters. 2. In the cardiomyopathic hamster the cardiac output, stroke volume, left intraventricular systolic pressure and dP/dt, which were all depressed in the age related sedentary animals, were increased by both periods of exercise. The left intraventricular diastolic pressure which was elevated was likewise decreased by both exercise periods. Only the 16 week exercise period decreased the resting heart rate. 3. In the normal F1 B hamster, both periods of exercise increased the cardiac output and stroke volume while the left intraventricular systolic pressure was decreased. Only the 16 week exercise decreased the resting heart rate and left intraventricular diastolic pressure and increased the left ventricular dP/dt. 4. Both periods of exercise increased the total heart calcium in the Bio 14.6 hamster while the heart calcium in the F1 B was increased only by the 16 week exercise period.

138. Significance of blood taurine levels in patients with first time acute ischaemic cardiac pain.

Abstract

We studied blood taurine levels of 91 consecutive patients admitted with first time cardiac pain suggestive of myocardial ischaemia. Blood taurine levels of patients with coronary arterial disease, but without a recent myocardial infarction (n = 36), at rest and after a maximal treadmill stress testing were also determined. The blood taurine level at the time of admission was significantly elevated (P less than 0.001) in patients with an acute myocardial infarction (n = 63) (271 +/- 98 mumol/l) and those with unstable angina (n = 22) [214 +/- 81 mumol/l] compared to that of normal subjects (n = 75) at rest (140 +/- 40 mumol/l). Patients with a myocardial infarction had a higher level than those with unstable angina (P less than 0.01) and non-ischaemic chest pain (n = 6) [P less than 0.05]. The levels peaked after 12-48 hours only in patients with infarction [367 +/- 140 mumol/l] (P less than 0.001) and unstable angina (273 +/- 82 mumol/l) (P less than 0.02). The levels of creatine kinase within the serum at the time of admission did not correlate well with those of blood taurine, but the peak levels of the former did correlate with the latter (P less than 0.02). Patients with known coronary arterial disease had a higher resting [236 +/- 69 mumol/l] level of blood taurine than normal subjects (P less than 0.001), which was further elevated [269 +/- 80 mumol/l] following exercise (P less than 0.001). Thus, an elevated level of taurine in whole blood at the time of admission of patients with an acute cardiac pain suggested the diagnosis of either a myocardial infarction or unstable angina. The level of taurine may be utilised to differentiate the two conditions.

Comparisons of left intraventricular end diastolic and systolic pressures, cardiac output, dP/dt, stroke volume and heart rate were made between the Bio 14.6 cardiomyopathic and F1B normal hamster at 45, 80, 150 and 240 days of age. 2. Comparisons of the ventricular calcium and taurine contents were made between the two strains of hamsters at similar ages. 3. Interstrain comparisons of the 240 day Bio 14.6 with age matched F1B hamsters and intrastrain comparisons with 45 day Bio 14.6 hamsters showed a decreased stroke volume, cardiac output and dP/dt with an increased left intraventricular end diastolic pressure, ventricular weight, ventricular weight/body weight ratio, heart calcium and taurine. 4. Despite the decreased left ventricular systolic pressure and cardiac output in the 80 day and older groups of Bio 14.6 hamsters, no compensatory increase in heart rate was observed.

Abstract

A method was developed for determining caffeine in decaffeinated coffee, tea, and beverage products by high performance liquid chromatography (HPLC). The HPLC system consisted of a Bio-Sil ODS-5S C18 column, methanol-water (25 + 75) mobile phase at 1 mL/min, and a UV detector. The method is simple and specific. Caffeine recoveries were 93.8-98.3% and coefficients of variation were 0.90-2.25%.

Abstract

Administration of either taurine or verapamil raised heart taurine and reduced heart calcium levels in BIO 14.6 hamsters. Combined administration of the two agents magnified both effects. It is suggested that the marked reduction of heart calcium produced by verapamil may be partially mediated through the membrane-stabilizing, calcium-modulating actions of taurine at the sarcolemma.

Abstract

Comparisons of calcium levels in heart, skeletal muscle, brain, liver and plasma were made throughout the life spans of normal and genetically myopathic hamsters. A large calcium accumulation occurred in, and was restricted to, myopathic tissues (heart and skeletal muscle). Calcium elevation in myopathic tissues corresponded to documented lesion appearance, and persisted throughout the remainder of the life span. Calcium buildup in heart and skeletal muscle, as a central feature of the myopathic process, does not reflect a general failure of calcium homeostasis; nonmyopathic tissues are unaffected. Calcium increased moderately in some normal tissues late in life.

Abstract

Ascorbic acid has been determined quantitatively in a variety of fresh and frozen fruits and vegetables, fresh and fortified canned juices, and powdered drinks by liquid chromatography (LC). The method consists of blending the sample in a solution of 0.05% EDTA in 0.2N H2SO4, centrifuging or filtering the mixture, then injecting a portion into the LC system. An Aminex HPX-87 LC column was used with 0.009N H2SO4 as solvent with a flow rate of 0.5 mL/min. Detection was at 245 nm. The method is simple and sensitive, and yields a high recovery. It compares favorably with the AOAC method, and has the advantage of being more accurate for samples with interfering pigments.

Abstract

Acute electrolyte depletion markedly reduced the elevated heart calcium in 60-day-old BIO 14.6 hamsters. This exchangeability may explain the susceptibility of calcium to therapeutic intervention at 60 days. At 120 days, the procedure did not alter heart calcium concentration.

Abstract

Heart calcium was measured in hamsters and rats following 30 days of drinking (i) tap water, (ii) taurine solution (T) or (iii) a taurine uptake inhibitor (GES). The regimes were duplicated in animals receiving verapamil (V). Heart calcium was reduced in both species by T + V; a comparable effect was obtained with GES alone; T alone had no effect. In the hamster, the GES effect was reversed by GES + V; V alone had no effect. In rats, the separate and combined effects of GES and V on heart calcium were identical. Possible mechanisms and underlying species differences are discussed.

Abstract

Epileptic foci are associated with locally reduced taurine (2-aminoethanesulfonic acid) concentration and Na+,K+-ATPase (EC 3.6.1.3) specific activity. Topically applied and intraperitoneally administered taurine can prevent the development and/or spread of foci in many animal models. Taurine has been implicated as a possible cytosolic modulator of monovalent ion distribution, cytosolic "free" calcium activity, and neuronal excitability. Taurine may act in part by modulating Na+,K+-ATPase activity of neuronal and glial cells. We characterized the requirements for in vitro modulation of Na+,K+-ATPase by taurine. Normal whole brain homogenate Na+,K+-ATPase activity is 5.1 +/- 0.4 (4) mumol Pi X h-1 X mg-1 Lowry protein. Partial purification of the plasma membrane fraction to remove cytosolic proteins and extrinsic proteins and to uncouple cholinergic receptors yields a membrane-bound Na+,K+-ATPase activity of 204.6 +/- 5.8 (4) mol Pi X h-1 X mg-1 Lowry protein. Taurine activates the Na+,K+-ATPase at all levels of purification. The concentration dependence of activation follows normal saturation kinetics (K1/2 = 39 mM taurine, activation maximum = +87%). The activation exhibits chemical specificity among the taurine analogues and metabolites: taurine = isethionic acid greater than hypotaurine greater than no activation = beta-alanine = methionine = choline = leucine. Taurine can act as an endogenous activator/modulator of Na+,K+-ATPase. Its action is mediated by a membrane-bound protein.

Abstract

Taurine concentrations were determined in hearts and whole blood of female Syrian golden hamsters and Wistar rats following 30 days of drinking: (i) tap water, (ii) taurine solution or (iii) a taurine uptake inhibitor (GES). The groups were duplicated in animals receiving daily verapamil injections during the treatment period. Heart taurine increased with (ii) and decreased with (iii) in both species; blood taurine changed similarly in rats only. Verapamil enhanced taurine uptake and offset the GES inhibition in the hamster heart, but had no such effect in rats. Possible implications of the species differences are elaborated upon.
