Abstract

A 40‐day feeding trial was conducted to investigate the effect of dietary microalgae Oedocladium sp. or Tribonema ultriculosum supplementation on growth performance, fillet fatty acid composition, skin pigmentation, and immune response of yellow catfish Pelteobagrus fulvidraco. Two hundred and seventy experimental fish (51.19 ± 1.17 g) were distributed among nine net cages (2 × 2 × 2 m3) in a randomized design and were fed diets containing 0% (control), 4% Oedocladium meal (Oedocladium), and 5% T. utriculosum meal (Tribonema) to apparent satiation twice a day. After the feeding trial, a hypoxic stress test was conducted to evaluate the effect of dietary microalgal supplementation on fish immune response. Compared to the control, microalgal supplementation did not affect fish growth performance. Dietary supplementation of T. utriculosum significantly increased fillet contents of palmitoleic acid by 100.44% and eicosapentaenoic acid by 12.51%, while the supplementation of Oedocladium meal in diet significantly increased the fish skin ventral lutein content by 61.6%. In addition, dietary Oedocladium or T. utriculosum supplementation significantly increased the plasma total superoxide dismutase activity (before hypoxic stress) and complement component 3 contents (after hypoxic stress). The addition of T. utriculosum meal also significantly increased the plasma immunoglobulin M content regardless of hypoxic stress. These results demonstrated Oedocladium sp. and T. utriculosum can be used to enhance fillet nutritional quality, skin pigmentation, and fish immune response. [ABSTRACT FROM AUTHOR]

Abstract

Previous nutritional studies have shown that insulin regulation is different between DT and A strains of gibel carp. As leptin plays a pivotal role in the effects of insulin, we hypothesised that leptin regulation of glucose and lipid metabolism would differ between the two strains. To test our hypothesis, recombinant human leptin was injected into two strains. The results showed that leptin activated the phosphatidylinositol 3-kinase (PI3K)–protein kinase B (AKT), AMP-activated protein kinase–acetyl coenzyme A carboxylase and Janus kinase 2 (JAK2)–signal transducer and activator of transcription (STAT) signalling pathways in both strains. Hypoglycaemia induced by leptin might be due to higher glucose uptake by the liver and muscles together with enhanced glycolytic potential and reduced gluconeogenic potential. Decreased lipogenesis and up-regulated fatty acid oxidation were induced by leptin. In terms of genotype, the PI3K–AKT signalling pathway was more strongly activated by leptin in the muscle tissue of the A strain, as reflected by the heightened phosphorylation of AKT. Furthermore, glycogen content, glycolytic enzyme activity and gluconeogenic capability were higher in the A strain than the DT strain. Strain A had higher levels of fatty acid synthesis and lipolytic capacity in the liver than the DT strain, but the opposite was true in white muscle. Regarding leptin–genotype interactions, the DT strain displayed stronger regulation of glucose metabolism in the liver by leptin as compared with the A strain. Moreover, a more active JAK2–STAT signalling pathway accompanied by enhanced inhibition of fatty acid synthesis by leptin was observed in the DT strain. Abstract

An 8‐week feeding experiment was performed to evaluate the effects of dietary arachidonic acid (ARA) on the reproductive performance, tissue fatty acid profile and sex hormone secretion in female Pelteobagrus fulvidraco during the breeding season. Three diets with different ARA levels (0.64%, 4.67% and 10.07% of total fatty acids) were formulated, a control diet (C) and two diets with low ARA (ARA‐L) or high ARA (ARA‐H). The results showed that the highest gonadosomatic index (GSI) and condition factor (CF) were observed in the ARA‐H group. The diets with supplementation of ARA resulted in a significantly higher fertilization rate, more normal larvae and greater survival rate of 7 days posthatching (7DPH) larvae. The ARA contents in liver and 3DPH larvae were improved with increasing dietary ARA concentrations. The lowest EPA/ARA ratios were observed in liver and eggs of fish fed with the ARA‐L diet. Significantly reduced serum estradiol (E2), 11‐keto‐testosterone (11‐KT) and testosterone (T) levels were found in the ARA‐L group females compared with the other treatments. The combined results indicated that a higher dietary ARA level and ARA/EPA ratio mainly influence spawning performance and egg quality of yellow catfish and that ARA regulates sex steroid hormone synthesis in broodstock. Abstract

Improvement in fish feed conversion efficiency (FCE) is beneficial for sustaining global food fish supplies. Here, we show that a set of polymorphisms at locus of the corticotropin releasing hormone receptor 2(crhr2), which is involved in hypothalamus-pituitary-interrenal (HPI) axis signaling, is associated with improved FCE in farmed allogynogenetic gibel carp strain CAS III compared with that in the wild gibel carp strain Dongting (DT). This set of polymorphisms downregulates the expression levels of crhr2mRNA in the brain and pituitary tissues in gibel carp strain CAS III compared with those in strain DT. Furthermore, compromised HPI axis signaling is observed in gibel carp strain CAS III, such as decreased α-melanocyte stimulating hormone protein levels, plasma cortisol content, and stress responses. Moreover, enhanced activation of protein kinase B/mammalian target of rapamycin complex 1 signaling observed in the muscle tissue of strain CAS III in comparison to that in strain DT indicated elevated anabolic metabolism in strain CAS III. Thus, these studies demonstrate that the genetic markers associated with compromised HPI axis signaling, such as crhr2, are potentially useful for genetic selection toward improvement in farmed fish growth and FCE, which would reduce fishmeal consumption and thereby indirectly facilitate sustainable fisheries.

Abstract

An oral starch administration trial was used to evaluate glucose homoeostasis in grass carp (Ctenopharyngodon idella) and Chinese longsnout catfish (Leiocassis longirostris Günther). Fish were administered with 3 g of a water and starch mixture (with 3:2 ratio) per 100 g body weight after fasting for 48 h. Fish were sampled at 0, 1, 3, 6, 12, 24 and 48 h after oral starch administration. In grass carp, plasma levels of glucose peaked at 3 h but returned to baseline at 6 h. However, in Chinese longsnout catfish, plasma glucose levels peaked at 6 h and returned to baseline at 48 h. The activity of intestinal amylase was increased in grass carp at 1 and 3 h, but no significant change in Chinese longsnout catfish was observed. The activity of hepatic glucose-6-phosphatase fell significantly in grass carp but change was not evident in Chinese longsnout catfish. The expression levels and enzymic activity of hepatic pyruvate kinase increased in grass carp, but no significant changes were observed in the Chinese longsnout catfish. Glycogen synthase (gys) and glycogen phosphorylase (gp) were induced in grass carp. However, there was no significant change in gys and a clear down-regulation of gp in Chinese longsnout catfish. In brief, compared with Chinese longsnout catfish, grass carp exhibited a rapid increase and faster clearance rate of plasma glucose. This effect was closely related to significantly enhanced levels of digestion, glycolysis, glycogen metabolism and glucose-induced lipogenesis in grass carp, as well as the inhibition of gluconeogenesis. Abstract

Proliferation of cyanobacteria is a global problem in eutrophic freshwater ecosystems,and the associated cyanotoxins are of growing concern with respect to human health and environmental problem.Most of the studies on microcystins(MCs) have only focused on terrestrial mammals,few have been conducted on the effects of MCs on fishes.Actually,many fishes are at the top of the trophic pyramids in aquatic ecosystems.MCs,which are the secondary metabolites of cyanobacteria,may accumulate in fish via feeding or bioaccumulation,and finally harm human health through the food chain.On the other hand,the harmful effects of MCs on fishes were mainly limited to acute toxic experiments based on oral gavaging,intraperitoneal injection,or administration via the dorsal aorta of the toxins,which can′t reflect the uptake route under natural environments.In fish,cyanobacterial toxins may ingestion via different routes: direct feeding on toxic cyanobacterial cells,uptake of dissolved toxins via the gill epithelium,or exposure via the foodweb.More importantly,cyanobacteria are an important dietary component for many fishes.Therefore,cyanobacteria is a kind of nutrient.On the other hand,it also has potential poisonousness on fish.Fortunately,the nutri-toxicological effects of microcystins on fish have been recently drawn to scientists′s much attention.In rencent years,mass cyanobacterial blooms occurred frequently in many freshwater lakes in China,e.g.,Lake Dianechi,Lake Taihu,Lake Caohu.In the project of the control of cyanobacteria in Chinese lakes,gathering the cyanobacteria using machine has been used in a large scale.Thus,it′s a big amount of biomass for the gathered cyanobacteria,and it′s an important issue on the comprehensive utilization of the gathered cyanobacteria using machines.Protein is the most expensive component in fish feeds.Due to the scarcity and cost of fish meal in the world market,it is imperative to reduce feed cost by exploring cheaper alternative protein in aquaculture feeds.In addition,cyanobacteria have been reported to contain more than 50% crude protein.Based on cost effectiveness,availability,and crude protein content,cyanobacteria seem to have considerable potential in fish feeds.Considering their contributions to both the basic research and the practice,it is necessary to explore the nutri-toxicological effects of cyanobacteria on fish.In the context of researching alternative protein sources for aquatic animals,growth(weight gain or specific growth rate),feed utilization(feeding rate,feed conversion efficiency,apparent digestibility coefficients of dry,protein and energy),and nutrient retention(protein retention efficiency and energy retention efficiency) are the main parameters for evaluation.More importantly,dietary cyanobacteria may severely influence fish′s health and food safety,which would emerge from toxicological indices,such as mortality,antioxidant responses,histological and cytopathology alterations,and accumulation of toxin in tissues.In the present paper,the nutri-toxicological effects of cyanobacterial meal or cyanobacterial cells on fish were summaried,and it focused on both nutrtional and toxicological effects of cyanobacteria via oral administration on fish.The present review will help to gain a new insight in the toxic effects of cyanobacteria on fish and provide new solutions for comprehensive utilization of the cyanobacteria.
