108. Phthalonitrile resins based on biomass vanillin with high storage modulus and good processing window.

Abstract

Bio-based phthalonitrile resins were synthesized using a two-step process. Initially, vanillin underwent a nucleophilic reaction with nitro and potassium carbonate to form intermediates, which were then dissolved in N, N-dimethylformamide. The intermediate was divided into two portions, with 4,4’-oxydianiline and 4,4’-methylenedianiline added to each portion, respectively, to produce two different vanillin-based phthalonitrile monomers: OVA monomer and DVA monomer. The successful synthesis of these two bio-based phthalonitrile monomers was confirmed using Fourier transform infrared spectroscopy and proton nuclear magnetic resonance spectroscopy. Differential scanning calorimetry was employed to study the processability of the monomers, revealing that OVA monomer and DVA monomer had a wide processing window of 110°C and 69°C, respectively, which were advantageous for resin processing. Dynamic mechanical analysis showed that the storage modulus of the OVA and DVA resins reached 4818 MPa and 3526 MPa, respectively, with a 24-h water absorption rate of only 1.03% and 0.90%. This study presents a viable method for synthesizing bio-based phthalonitrile resins, which can be widely utilized as functional and structural materials in daily life due to their renewability, degradability, and minimal environmental impact. Abstract

Although nano‐porous materials are widely used, they are still difficult to prepare in large size and expand flexibly. In this work, a new method for the synthesis of porous materials is proposed. First, the block polymers were prepared by atom transfer radical polymerization. Through electrostatic spinning, the fiber films were fabricated, then the fibers were modified by surface post‐treatment and related Friedel–Crafts alkylation reaction. The pore and CO2 adsorption properties of the porous fiber films are characterized by nitrogen adsorption–desorption isotherm. In order to improve the pore properties and CO2 adsorption properties of thin films, the additives and pretreatment process are optimized. The maximum specific surface area of the porous fiber membrane is determined to be 640 m2/g, and the CO2 adsorption capacity is high up to 36.3 wt.% (8.68 mmol/g). Moreover, the porous fiber membrane derived from this strategy is superior in the aspects of large size, convenient adjustment and simple processing, which makes up for the drawbacks of traditional porous materials. The fiber membrane obtained by this strategy has high productivity, and the fiber membrane is easy to be postprocessed, which can be integrated into functional devices, and is widely used. Abstract

Tumors may develop a variety of immune evasion mechanisms during the progression of colorectal cancer (CRC). Here, we intended to explore the mechanism of histone methyltransferase SETDB1 in immune evasion in CRC. The expression of SETDB1, microRNA-22 (miR-22), BATF3, PD-L1, and FOSB in CRC tissues and cells was determined with their interactions analyzed also. Gain-of-function and loss-of-function approaches were employed to evaluate the effects of the SETDB1/FOSB/miR-22/BATF3/PD-L1 axis on T cell function, immune cell infiltration, and tumorigenesis. Aberrant high SETDB1 expression in CRC was positively associated with PD-L1 expression. SETDB1 negatively regulated miR-22 expression by downregulating FOSB expression, while miR-22 downregulated PD-L1 expression via targeting BATF3. Furthermore, SETDB1 silencing promoted the T cell-mediated cytotoxicity to tumor cells via the FOSB/miR-22/BATF3/PD-L1 axis and hindered CRC tumor growth in mice while leading to decreased immune cell infiltration. Taken together, SETDB1 could activate the BATF3/PD-L1 axis by inhibiting FOSB-mediated miR-22 and promote immune evasion in CRC, which provides a better understanding of the mechanisms underlying immune evasion in CRC. Abstract

Converting peanut protein biomass waste into environmentally friendly meat substitutes by a high-moisture extrusion process can help solve both resource and waste problems and be "double green". A multiscale method combined with some emerging techniques such as atomic force microscopy-based infrared spectroscopy and X-ray microscopy was used to make the whole extrusion process visible to show the process of forming a meat-like fibrous structure using two-dimensional and three-dimensional perspectives. The results showed that the protein molecules underwent dramatic structural changes and unfolded in the extruder barrel, which created favorable conditions for molecular rearrangement in the subsequent zones. It was confirmed that the meat-like fibrous structure started to form at the junction of the die and the cooling zone and that this structure was caused by the phase separation and rearrangement of protein molecules in the cooling zone. Moreover, the interactions between hydrogen bonds and disulfide bonds formed in the cooling zone maintained the meat-like fibrous structure with an α-helix > β-sheet > β-turn > random coil. Of the two main peanut proteins, arachin played a greater role in forming the fibrous structure than conarachin, especially those subunits of arachin with a molecular weight of 42, 39, and 22 kDa.

Abstract

The bioactive peptide has wide functions, such as lowering blood glucose levels and reducing inflammation. Meanwhile, computational methods such as machine learning are becoming more and more important for peptide functions prediction. Most of the previous studies concentrate on the single-functional bioactive peptides prediction. However, the number of multi-functional peptides is on the increase; therefore, novel computational methods are needed. In this study, we develop a method MLBP (Multi-Label deep learning approach for determining the multi-functionalities of Bioactive Peptides), which can predict multiple functions including anti-cancer, anti-diabetic, anti-hypertensive, anti-inflammatory and anti-microbial simultaneously. MLBP model takes the peptide sequence vector as input to replace the biological and physiochemical features used in other peptides predictors. Using the embedding layer, the dense continuous feature vector is learnt from the sequence vector. Then, we extract convolution features from the feature vector through the convolutional neural network layer and combine with the bidirectional gated recurrent unit layer to improve the prediction performance. The 5-fold cross-validation experiments are conducted on the training dataset, and the results show that Accuracy and Absolute true are 0.695 and 0.685, respectively. On the test dataset, Accuracy and Absolute true of MLBP are 0.709 and 0.697, with 5.0 and 4.7% higher than those of the suboptimum method, respectively. The results indicate MLBP has superior prediction performance on the multi-functional peptides identification. Abstract

During the makeup and breakout operation of iron roughneck, the performance of the clamp tooth holding the pipe string directly affects the drilling efficiency and the service life of the pipe string. In this paper, a cylindrical non-clamp-tooth Clamping body with 20CrMnTi material is designed to replace the upper shackle clamp tooth. In the up-shackle operation of the torque wrench, the clamping body contacts the outer wall of the string in the form of two parallel axis cylinders, the relationship between each influencing factor and contact stress is established, and the contact stress and deformation of the string are simulated by finite element method. This non-clamp-tooth clamping body can reduce the damage degree of the string, make its elastic deformation not exceed 1mm, and improve the service life of the clamping body.

Abstract

ABSTRACTNeuropeptide Y (NPY) is a highly conserved endogenous peptide in the central and peripheral nervous systems, which has been implicated in nociceptive signaling in neuropathic pain. However, downstream mechanistic actions remain uncharacterized. In this study, we sought to investigate the mechanism of NPY and its receptor NPY2R in the amygdala in rats with neuropathic pain-like behaviors induced by chronic constriction injury (CCI) of the sciatic nerve. The expression of NPY and NPY2R was found to be aberrantly up-regulated in neuropathic pain-related microarray dataset. Further, NPY was found to act on NPY2R in the basolateral amygdala (BLA). As reflected by the decrease in mechanical withdrawal threshold (MWT) and thermal withdrawal latency (TWL) as well as the increase of NPY expression in the amygdala of rats with neuropathic pain-like behaviors, NPY was closely related to the effect of amygdala nerve activity in neuropathic pain. Subsequently, mechanistic investigations indicated that NPY2R activated the MAPK signaling pathway in the amygdala. NPY2R-induced decrease of MWT and TWL were also restored in the presence of MAPK signaling pathway antagonist. Moreover, it was revealed that NPY2R overexpression promoted the viability while inhibiting the apoptosis of microglia. Taken together, NPY in the amygdala interacts with NPY2R to activate the MAPK signaling pathway, thereby promoting the occurrence of neuropathic pain.

Abstract

At present, the rod production system is difficult to meet the requirements of directional wells and cluster wells with large slant, the rod and pipe of these pumping wells suffer from serious wear, high energy consumption and low system efficiency. Downhole drive reciprocating pump production system, cancelling the sucker rob for transferring forces and motion, utilizing downhole submersible motor drives the reciprocating pump through the downhole drive mechanism, which can avoid the wear of the rod and pipe and improve the system’s efficiency. The composition and working principle of downhole drive reciprocating pump production system are introduced. The scheme of downhole drive reciprocating pump production system based on rack and pinion drive is analyzed in detail. The dynamic model of rack and pinion is established. By the dynamic simulation of the downhole transmission mechanism, the vibration loads of the mechanism with start up different accelerations are analyzed, and it is concluded that it is better to start with the variable acceleration. The scheme can reduce the vibration load caused by the motion inertia to the mechanism and improve the service life of the whole oil production system.

Abstract

Noise pollution has been given more attention due to its negative impacts on human health and disease. The portable low-frequency noise reduction device we developed in this research can provide an effective way for solving low-frequency noise pollution problem in the small space. This work describes the design principle and the prototype structures for two versions of V1.5 and V2.0 and builds the noise test systems for small spaces, respectively. These devices, installed on the outer surface of the small spaces, can automatically identify the noise spectrum and implement noise reduction by means of the active noise control (ANC) technology. The testing results indicate that the noise can be reduced 12 dB in the range of 250 Hz~400 Hz for the small closed space while, for the small open space, the best effect of 5.88 dB occurs in the optimal frequency of 450 Hz. These effects will be weakened with the increasing distance away from the source and show the obvious axisymmetric distribution in the inverted cone space.
