Urine can accumulate systemic changes with no mechanism to be stable, which may reflect early changes associated with physiological or pathophysiological processes. To explore the potential value of the urine proteome, two rat models were established by intrahepatic injection of two different hepatoma cell lines, CBRH-7919 and RH-35. Urine samples were collected and analyzed. Compared with controls, the two models exhibited different numbers and types of differentially expressed urinary proteins despite having similar histological results. The results were compared with the urine proteome of a Walker 256 (W-256) liver tumor model. The differentially expressed urinary protein patterns in the three models were different. These findings demonstrate that changes in the urine proteomes of the two models can be detected at early stages and that the patterns of differentially expressed urinary proteins can differ even when the histological results are similar. Urinary proteins have potential utility for distinguishing among different tumor cells grown in the same organ.

Growth hormone (GH) is a polypeptide hormone that could reduce the mitochondria-mediated oxidative stress and improve the mitochondrial function. However, the mechanisms of GH on granulosa cell apoptosis and mitochondrial function is still unclear. The aim of this study is to determine the effects of GH on granulosa cells apoptosis and the underlying mechanisms. In this study, we exposed the ovarian granulosa cell line (KGN cell) with cisplatin to establish an ovarian granulosa cell apoptosis and mitochondrial dysfunction model in vitro. To examine the benefit of GH in restoration of granulosa cell, we determined cell proliferation, cell apoptosis, reactive oxygen species (ROS) level, the expression of antioxidant components Sod2, Sirt3, as well as the mitochondrial membrane potential and mitochondrial DNA (mtDNA) copy number after GH treatment. We found that the cisplatin exposure significantly inhibited cell proliferation and elevated the apoptotic rate by student's t-test (p < 0.05). Whereas, the GH treatment could rescue the cell proliferation and decrease the apoptotic rate, as well as reduce the Bax/Bcl-2 ratio (p < 0.05). Additionally, GH significantly reduced abnormal ROS levels and increased the level of Sirt3 and Sod2 thus alleviating the oxidative stress. We also found that GH facilitated the recovery of mitochondrial membrane potential and mitochondrial DNA (mtDNA) copy number in granulosa cells. Our results indicated that GH exerted protective effects in cisplatin-induced ovarian granulosa cell apoptosis by alleviating oxidative stress and enhancing mitochondrial function via Sirt3-Sod2 pathway.

Recent studies show that intracellular accumulation of cholesterol leads to acquired resistance to gefitinib in non-small cell lung cancer (NSCLC) cells. In this study we investigated how to regulate the cholesterol levels in gefitinib-resistant NSCLC cells. We showed that intracellular cholesterol levels in gefitinib-resistant cell lines (PC-9/GR, H1975, H1650, and A549) were significantly higher than that in gefitinib-sensitive cell line (PC-9). Treatment with gefitinib (5 μM) significantly increased intracellular cholesterol levels in PC-9/GR, H1975, and H1650 cells. Gefitinib treatment downregulated the expression of PPARα, LXRα, and ABCA1, leading to dysregulation of cholesterol efflux pathway. We found that a lipid-lowering drug fenofibrate (20, 40 μM) dose-dependently increased the expression of PPARα, LXRα, and ABCA1, decreased the intracellular cholesterol levels, and enhanced the antiproliferative effects of gefitinib in PC-9/GR, H1975, and H1650 cells. We revealed that fenofibrate increased the gefitinib-induced apoptosis via regulating the key proteins involved in the intrinsic apoptosis pathway. In PC-9/GR, H1975 and H1650 cells, fenofibrate dose-dependently increased the expression of AMPK, FoxO1, and decreased the expression of AKT, which were remarkably weakened by knockdown of PPARα. In PC-9/GR cell xenograft mice, combined administration of gefitinib (25 mg · kg(−1) · d(−1)) and fenofibrate (100 mg · kg(−1) · d(−1)) caused remarkable inhibition on tumor growth as compared to treatment with either drug alone. All the results suggest that fenofibrate relieves acquired resistance to gefitinib in NSCLC by promoting apoptosis via regulating PPARα/AMPK/AKT/FoxO1 pathway. We propose that combination of gefitinib and fenofibrate is a potential strategy for overcoming the gefitinib resistance in NSCLC.

Recent studies have demonstrated that acquisition of cancer stem-like properties plays an essential role in promoting epidermal growth factor receptor-tyrosine kinase inhibitors (EGFR-TKIs) resistance in non-small cell lung cancer (NSCLC); however, how to regulate cancer stem-like properties and EGFR-TKI resistance is largely unclear. In this study, we discovered that increased iroquois-class homeodomain protein 4 (IRX4) was related to gefitinib resistance in NSCLC cells. Knockdown of IRX4 inhibited cell proliferation, sphere formation, and the expression of CD133, ALDH1A1, NANOG, Sox2 and Notch1, and the transcriptional activity of NANOG promoter. IRX4 overexpression increased the protein level of NANOG and CD133 in PC-9 cells. Combination of knocking-down IRX4 with gefitinib increased cell apoptosis and decreased cell viability and the expression of p-EGFR and NANOG in PC-9/GR cells. IRX4 knockdown in a PC-9/GR xenograft tumor model inhibited tumor progression and the expression of NANOG and CD133 more effectively than single treatment alone. Knockdown of NANOG inhibited the expression of CD133 and restored gefitinib cytotoxicity, and NANOG overexpression-induced cancer stem-like properties and gefitinib resistance could be obviously reversed by knocking-down IRX4. Further, we found that 1,25-dihydroxyvitamin D3 (1,25(OH)2D3) reduced obviously the expression of IRX4 and NANOG by inhibiting the activation of TGF-β1/Smad3 signaling pathway; moreover, combination of 1,25(OH)2D3 and gefitinib decreased cell viability and proliferation or tumor progression and the expression of IRX4 and NANOG compared with single treatment alone both in PC-9/GR cells and in a PC-9/GR xenograft tumor model. These results reveal that inhibition of IRX4-mediated cancer stem-like properties by regulating 1,25(OH)2D3 signaling may increase gefitinib cytotoxicity. Combination therapy of gefitinib and 1,25(OH)2D3 by targeting IRX4 and NANOG, could provide a promising strategy to improve gefitinib cytotoxicity.

Indirect pulp capping, pulpotomy, and apexification are three common endodontic treatments for immature traumatized incisors. They all affect tooth root development to some extent. The aim of this retrospective study was to compare the influence of these treatments on root development of immature permanent incisors following dental trauma.Twenty-one indirect pulp capping, 48 pulpotomy, and 58 apexification cases with a mean age of 8.4 ± 1.0 years and median follow up of 12 months were included. NIH ImageJ with TurboReg plug-in was used to correct angular differences between the pre-operative and recall periapical radiographs, and to calculate variations of root length, dentin wall thickness, and apical closure. Kruskal-Wallis ANOVA followed by pairwise comparisons was applied to compare the radiographic variations. The type of apical closure was assessed qualitatively and analyzed using Fisher's exact test.The apexification group had a lower trend toward apical closure than the other two groups (P .05). It also showed thinner dentin wall thickness compared with the pulpotomy group (P = .001). There was no significant difference between pulpotomy and indirect pulp capping in the trend to apical closure (P .05) or dentin wall thickness (P = .775). There was no significant difference in the variation of root length among the three groups (P = .06). There was a moderate correlation between the treatment and the type of apical closure (Cramer's V Coefficient = .375). Pulpotomy tended to form a normal apical constriction rather than a calcific barrier while apexification showed the opposite inclination. Indirect pulp capping had no specific inclination toward any type of apical closure.Apexification resulted in an abnormal root development mostly by affecting the dentin wall thickness and apical closure. Pulpotomy was beneficial for normal root development of immature traumatized teeth.

In order to reduce the vibration of the stiffened plate, this paper presents a stiffened plate structure with a local resonance unit of the spring oscillator arranged periodically on the side of the stiffened plate without the stiffened bar, and studies the energy band structure and transmission characteristics of the structure. The band structure analysis shows that two band gaps are formed in the structure. The frequency range of the first band gap is 1165.23~1300.45Hz, and the frequency range of the second band gap is 198.97~243.88Hz. Furthermore, the influences of lattice constant, matrix mass ratio of the stiffened plate to the vibrator mass ratio, and the resonator spring stiffness on the band gap range are further analyzed. The transmission characteristics of local resonance stiffened plate structure with finite period are analyzed. These research structures provide important theoretical basis and technical support for vibration reduction design of stiffened plate structure.

The utilization of electrode materials with high-performance and low-cost is crucial for the development of electrochemical energy storage devices. Waste engine oil is a toxic and hazardous waste; it is estimated that about 45 million tons of waste engine oil are generated per year worldwide. Herein, waste engine oil was first recycled as a raw material to fabricate hierarchical porous carbon nanosheets. The obtained material exhibited high specific surface area (up to 2276 m2 g−1), hierarchical porous structure, and excellent capacitance performance. Its specific capacitance reached 352 F g−1, and more than 87.7% of initial capacitance was retained when the current density was increased from 0.5 to 20 A g−1. In addition, no apparent capacitance drop was observed after 5000 charge and discharge cycles. The superior capacitance performance than that of the commercial activated carbon and most of the waste-derived porous carbons, cheap and readily available raw materials, and environmental benefit due to proper recycling of the hazardous waste make the reported hierarchical porous carbon nanosheet a promising energy storage material.

The dramatically increased use of electricity is raising major concerns as to the consequences of the interaction between electromagnetic field (EMF) and neurobiology. The aim of this study is to investigate the effects of magnetic field on working memory in the hippocampal region by analyzing local field potentials (LFPs) and spikes pattern in vivo. In present study, mice were exposed to EMF (50Hz, 1mT), static magnetic field (SMF, 1mT), or placed in the exposure tube but without EMF exposure (SHAM), respectively. During the exposure for 7 consecutive days, mice were subjected to perform working memory (WM) tasks in Y-maze, and multichannel electrophysiology signals from hippocampus of mice were recorded during the test, from which LFPs, spike firing rates, band power at different frequencies, and theta-gamma modulation index (MI) were analyzed in details. From our results, correct choice rate during WM task was found significantly decreased in EMF group after 3-day exposure, which was consistent with noticeable decline in firing rate. Starting from Day 3 after EMF exposure, the power of theta (4-12Hz) and gamma (LG, 30-60Hz) before reference point (RP) in Y-maze were also found to be descending, together with decrease of oscillatory activities of theta and gamma frequencies. The results indicated that MI between theta and gamma could play a significant role in modulating the spikes discharge and encoding WM. Therefore, the analysis of theta-gamma coupling and its oscillation strength may provide a new perspective for mechanistic investigation of EMF-induced WM deficits.

Cranial capacity is one of the most important features used in hominin taxonomic and morphological analyses. For complete or nearly complete modern human crania, the traditional methods of estimating cranial capacity include filling the vault with seeds, the water displacement method, and the use of regression formulae based on craniometrics. For incomplete human fossils, cranial capacities are estimated by reconstructing endocasts manually or virtually or by using existing modern human skull regression formulae; however, the accuracies of these methods are usually dubious. To find a more accurate way of estimating cranial capacity of partial skulls, seven different estimation methods are compared, including the manual reconstruction of the endocast, models built on skulls and models built on endocasts. We then estimated the cranial capacity of a fragmentary Late Pleistocene cranium, Jingchuan 1. The models are tested on 30 modern human skulls, three Homo erectus fossils and one Late Pleistocene Homo sapiens fossil. In terms of estimating the cranial capacity of the fossil humans, our results indicate that the cranial capacity estimates based on endocasts are more precise than those from exterior skull dimensions, that multivariate models are better than univariate ones, and that the new models using PCR and PLSR have the smallest errors (

The electrochemical aptamer-based (E-AB) sensing platform appears to be a convenient (rapid, single-step, and calibration-free) and modular approach to measure concentrations of specific molecules (irrespective of their chemical reactivity) directly in blood and even in situ in the living body. Given these attributes, the platform may thus provide significant opportunities to render therapeutic drug monitoring (the clinical practice in which dosing is adjusted in response to plasma drug measurements) as frequent and convenient as the measurement of blood sugar has become for diabetics. The ability to measure arbitrary molecules in the body in real time could even enable closed-loop feedback control over plasma drug levels in a manner analogous to the recently commercialized controlled blood sugar systems. As initial exploration of this, we describe here the selection of an aptamer against vancomycin, a narrow therapeutic window antibiotic for which therapeutic monitoring is a critical part of the standard of care, and its adaptation into an electrochemical aptamer-based (E-AB) sensor. Using this sensor, we then demonstrate: (i) rapid (seconds) and convenient (single-step and calibration-free) measurement of plasma vancomycin in finger-prick-scale samples of whole blood, (ii) high-precision measurement of subject-specific vancomycin pharmacokinetics (in a rat animal model), and (iii) high-precision, closed-loop feedback control over plasma levels of the drug (in a rat animal model). The ability to not only track (with continuous-glucose-monitor-like measurement frequency and convenience) but also actively control plasma drug levels provides an unprecedented route toward improving therapeutic drug monitoring and, more generally, the personalized, high-precision delivery of pharmacological interventions.

Background Because acute ethanol (EtOH) administration is known to influence cognitive processes by impairing hippocampal function, electrophysiological responses of the hippocampus following EtOH exposure warrant investigation. To mimic in vivo conditions, we recorded and analyzed critical firing characteristics of the neuronal population dynamically, particularly in the hippocampal region, before and after acute EtOH administration. Methods Microelectrodes were inserted in the hippocampus CA1 region of 21 Institute of Cancer Research mice. The mice were divided into 3 groups, including an EtOH injection group (1.5 g/kg), a saline injection group (1.5 g/kg), and a negative control group that received no injection. A data acquisition system was employed to detect the local field potentials (LFPs) and spike potentials following acute EtOH administration. Various multichannel electrophysiological signals were collected in vivo in each group at 60 minutes, from which the firing rate and wavelet entropy (WE) were analyzed further. Results Firing rates began to decline at 20 minutes postinjection and then gradually recovered from 40 to 60 minutes. Wireless power transfer (WPT) devices have been widely accepted by more and more industrial producers and consumers due to their safe, convenient, and high-efficient features. Square coils with ferrite cores are usually utilized as magnetic coupling devices by WPT systems for electric vehicles (EVs). However, the research on coupling coils often depends on three-dimensional finite element analysis (3-D FEA) tools such as ANSYS Maxwell or HFSS, which are troublesome and time-consuming. This paper explores a calculation model of mutual coupling between two square coils with ferrite cores based on magnetic mirror method. With this model, accurate results and fast operation speed can be obtained, and 3-D FEA tool is used to compare and verify the performance of this work.

Urine, as a potential biomarker source among the body fluids, can accumulate many changes in the body due to the lack of a mechanism to maintain a homeostatic state. Previous studies have demonstrated that proteomic technology can find many potential biomarkers to reflect different diseases in the urine. This study aims to detect early changes in the urinary proteome in a rat liver tumor model. The tumor model was established with the Walker-256 carcinosarcoma cell line (W256). Compared to before the injection, ninety-five differential proteins were significantly changed in the experimental rats. At day 3, twelve proteins were identified in the absence of pathological changes, and four of them were altered at all four time-points (B2MG, VCAM1, HA11, and LG3BP). Seven had previously been associated with liver cancer. At day 5, fifty-two differential proteins were identified. At day 7 and day 11, there was a significant decrease in the body weight of the rats, and tumor tissue was observed in the liver. Fifty-two and forty differential proteins were changed significantly at day 7 and day 11, respectively. Of the proteins that were identified at these three time-points, and twenty-four were reported to be associated with liver cancer. Comparing the differential urinary proteins and biological processes of liver tumor model with those in different models of W256 grown in other organs, specific differential protein patterns were found among the four models, which indicates that the differential urinary proteins can reflect the differences when the same tumor cell grown in different organs.SignificanceThis study demonstrated that (1) the rat liver tumor model caused early changes in urinary proteins may give new insight into the early diagnosis of liver cancer; (2) the same tumor cell grown in different organs can be reflected in differential urinary proteins.

Urine accumulates systemic changes in the body without homeostatic control; thus, it has the potential for early detection of cancer. In this chapter, five tumor-bearing models were established by injection of Walker 256 tumor cells into the subcutaneous, lung, brain, liver, or bone cavity of rats. Urine samples were collected at multiple time points after tumor cell inoculation. Dynamic urine proteomes were analyzed using label-free relative quantification. We found the urinary protein patterns changed significantly with cancer development, and some urinary proteins even changed at an early onset of tumor growth. Moreover, urine proteomics could differentiate the same cancer cells grown at different organs. We think that urine is a noninvasive and promising source in cancer biomarker discovery especially in the early phase.

Chronic atrophic gastritis (CAG) is a common and possibly precancerous digestive tract disease. Development of drugs with effect of preventing precancerous lesions draws the eyes of global researchers. Qilianshupi decoction (QLSP) is a Traditional Chinese Medicine (TCM) that is commonly used to treat CAG, but few studies have explored the mechanism of QLSP on treating CAG. This study investigated the molecular targets of the component herbs of QLSP in preventing precancerous lesions based on network pharmacology. Network pharmacology analysis revealed that the 6 herbs regulated multiple CAG-related genes, among which the most important were cancer-related pathway (apoptosis, p53, and VEGF) and epithelial cell signaling in Helicobacter pylori infection. Further animal experiments showed that the expression of survivin and p53 in precancerous lesions of CAG rats was significantly increased which was suppressed by QLSP. Moreover, telomerase activity was inhibited in precancerous lesions of CAG rats, and telomere length of gastric mucosa was increased, which was reversed by QLSP. Our results suggest that the components of QLSP prevents gastric precancerous lesions through decreasing the expression of survivin and p53 and regulating telomerase activity and telomere length in CAG.

Background Ethanol is associated with various medical comorbidities affecting the brain and central nervous system. Objectives The use of puerarin in treating alcohol-induced memory disorders is systematically evaluated in this study based upon an analysis of the firing rate and oxygen saturation (SO2). Material and methods A multi-channel data acquisition system and near-infrared spectroscopy (NIRS) were combined to obtain the electroneurophysiological signals and SO2 in the hippocampus of mice from the Institute of Cancer Research (ICR) after ethanol injection. A T-maze test was performed to study the alteration of spatial memory function. Ethanol was administered intraperitoneally (i.p.) in 2 dosages (1.5 g/kg and 0.5 g/kg). To investigate the effects of puerarin against acute ethanol-induced memory impairment, the same parameters corresponding to electroneurophysiological signals, SO2 and behavior performance in mice were also recorded upon the supplementation of puerarin in 2 dosages (25 mg/kg and 50 mg/kg). Results An inhibited firing rate and decreased SO2 were found in mice treated with an ethanol dose of 1.5 g/kg, which resulted in a low correct choice rate in the T-maze test, while an opposite trend appeared in mice which had been administered a moderate ethanol dose (0.5 g/kg). The trend of SO2 was positively correlated with that of the firing rate. A decreased firing rate and SO2 were accompanied by a decrease in the correct choice rate. With the supplementation of puerarin, a significant increase in the mean firing rate and SO2, as well as an improved correct choice rate, can be found in mice injected with excessive ethanol. Conclusions The electroneurophysiological signals and NIRS were combined for the first time to prove that an excessive intake of ethanol can inhibit the spatial learning and memory function of mice. The supplementation of puerarin can suppress these adverse effects induced by ethanol at a high dosage, as evidenced by the increased firing rate and SO2.

A set of intracranial pressure and intracranial temperature monitor was developed. Moreover, it was verified to be effective in the monitoring of intracranial parameters by designed experiments.The intracranial pressure and intracranial temperature monitor was tested in the water bath comparing with the Codman intracranial pressure monitor and mercury thermometers. As well, the monitor was applied in the monitoring of rat brain edema in vivo.The maximum error is less than 266.64 Pa in the intracranial pressure measurement compared to the Codman intracranial pressure monitor, and the maximum error is less than 0.3 oC in the temperature measurement according to mercury thermometers. Furthermore, the monitor could real-time obtain the intracranial pressure and intracranial temperature in the brain edema in vivo.The intracranial pressure and intracranial temperature monitor realizes the real-time in vivo monitoring of intracranial pressure and intracranial temperature. The measurement accuracy meets the acquirement of doctors. The instrument has potential for clinical use.

Biomarker is the change associated with the disease. Without homeostatic control, urine can accumulate early changes in the body. We expect that urinary proteome can also reflect early changes in the nervous system and urine is a better biomarker source for nervous system diseases. Multiple sclerosis is a chronic autoimmune demyelinating disease of the central nervous system and is difficult to diagnose in early stages. In this study, a tandem mass labeling approach coupled with high-resolution mass spectrometry was used to analyze seven-day urinary proteome changes in a rat model of experimental autoimmune encephalomyelitis when the clinical scores in the EAE group were "0" and no obvious histological changes were observed. Thirty-one urinary proteins were altered, based on Ingenuity Pathway Analysis, seventeen of these proteins were associated with neurological functions. The top canonical pathways represented by these dysregulated proteins included the acute phase response and metabolic processes. The acute phase response was characterized by an increase in inflammatory factors that are known to cause multiple sclerosis. Additionally, lipid or glucose metabolic alterations may provide clues for future mechanistic studies on multiple sclerosis. Fourteen proteins were identified to have catalytic activities that may contribute to neuronal damage. Furthermore, among the seven proteins that were most affected, six were reported to be expressed in the serum/cerebrospinal fluid/brain tissue of multiple sclerosis patients, thereby indicating that urine can be a good source of biomarkers for the early detection of multiple sclerosis.

Large micropore with a pore size of 1–2 nm can allow high charge storage capability while ensure fast ions transport, which serves to improve the energy density of supercapacitors without sacrificing high power density. Herein, we report a large micropore dominant microporous carbon derived from a biomass waste, flaxseed residue from the edible oil industry, for high-performance supercapacitors. The reported material exhibits a large specific surface area of up to 3230 m2 g−1, and more than 70.1% of micropore volume is contributed by large micropores. The specific capacitance of the obtained material reaches up to 369 and 398 F g−1 in KOH and H2SO4 electrolyte, respectively. Meanwhile, the assembled supercapacitor device based on the obtained material exhibits excellent rate and cycle performance. Over 92.7% of the initial capacitance is retained even under a large current density of 20 A g−1 and the capacitance retention is more than 98.1% after 10000 times cycle in KOH electrolyte. The energy density of the assembled supercapacitor device reaches 61.2 Wh kg−1 at a power density of 468.8 W kg−1, and a high energy density of 43.5 Wh kg−1 is retained at a large power density of 13.3 kW kg−1 in ionic liquid EMIMBF4 electrolyte.

Absrtact Carbon nanotubes has attracted much attentions due to its huge application potential in various high-tech fields. Exploition of the simple and cost-efficient synthesis method for carbon nanotubes is crucial for its practical application. Herein, multi-wall carbon nanotubes (MCNTs) was successfully synthesized by one-step pyrolysis of a mixture of benzene and ferrocene in a sealed autoclave under autogenetic-pressure. A pyrolysis temperature of 800 °C, pyrolysis time for 30 min and a ferrocene/benzene molar ratio of 1:2 is found to be the optimal conditions for synthesizing the MCNTs via the mentioned autogenetic-pressure method. The synthesized MCNTs exhibits a “bamboo joint”-like multi-wall structure with about 12 layers of highly graphited tube wall and uniform external diameter of ca 20 nm. The specific capacitance of the synthesized MCNTs reaches 64 F g-1, which value is about 3.0 times higher than that of the typical commercial MCNTs. The synthesized MCNTs also exhibits very excellent superior rate and cycle performance. More than 95.5% initial specific capacitance is retained even under a large current density of 20 A g-1, and no apparent specific capacitance decrement is observed after 10,000 times charge-discharge cycles. The energy density of the synthesized MCNTs-based supercapacitor arrives 2.27 Wh kg-1 at a power density of 157 W kg-1, and 2.07 Wh kg-1 of energy density is still retained at a power density of 5.3 kW kg -1, while an energy density of 0.72 Wh kg-1 is arrived at a power density of 2.6 kW kg-1 for the typical commercial MCNTs-based supercapacitor. The superior capacitance performance of the synthesized MCNTs than that of the typical commercial MCNTs is ascribed to the higher conductivity induced by the higher graphitization degree than that of the typical commercial MCNTs, good surface wettability due to the high content of oxygen-contained functional groups on surface, and the supplementary contribution of pseudo-capacitance.

The notions of explanation-radius and the local information based on the data distribution are proposed. The former measures the injective degree and the latter depicts the difference between the original data and the reduction data. Thereafter, through the experiments, the linear and nonlinear dimension-reduction is analyzed, included PCA (Principal Component Analysis), PP (Projection Pursuit) and LLE (Locally Linear Embedding), Laplacion Eigenmap. The experiments show the effectiveness and advantages of the researches.
