This study evaluated the effects of single-bottle, multipurpose, universal adhesives on the bond strength of resin cement to zirconia ceramic. Polished zirconia ceramic (Cercon base) discs were randomly divided into four groups (n=40) according to the applied surface-conditioning agent: Single Bond 2, Single Bond Universal, All-Bond Universal, and Alloy Primer. Cured composite cylinders (Ø 0.8 mm x 1 mm) were cemented to the conditioned zirconia specimens with resin cement (RelyX ARC). The bonded specimens were subjected to a microshear bond-strength test after 24 hours of water storage and after 10,000 cycles of thermocycling. The surface-conditioning agent significantly influenced the bond strength (p<0.05). Single Bond Universal showed the highest initial bond strength (37.7 ± 5.1 MPa), followed by All-Bond Universal (31.3 ± 5.6 MPa), Alloy Primer (26.9 ± 5.1 MPa), and Single Bond 2 (8.5 ± 4.6 MPa). Artificial aging significantly reduced the bond strengths of all the test groups (p<0.05). After 10,000 cycles of thermocycling, All-Bond Universal showed the highest bond-strength value (26.9 ± 6.4 MPa). Regardless of artificial aging, Single Bond Universal and All-Bond Universal showed significantly higher bond strengths than Alloy Primer, a conventional metal primer. [ABSTRACT FROM AUTHOR]

Replacing glazing with polishing is still controversial in terms of the surface roughness of dental porcelains. This study investigated the polishing performance of a ceramic-polishing rubber wheel (CP-RW), which contains large uniform and rounded silicon carbide particles and small diamond particles, in improving the surface roughness of two feldspathic porcelains for sintering and CAD/CAM milling. Using a confocal laser scanning microscopy, the changes in the surface roughness parameters were evaluated before and after polishing or glazing for three surface treatment groups: SofLex polishing, CP-RW polishing, and Glazing. Regardless of the parameters, all treatments reduced roughness values (repeated measures ANOVA, p<0.05). The roughness values obtained after CP-RW polishing were lower than those obtained after SofLex polishing and glazing (2-way ANOVA, p<0.05). Polishing both ceramics with CP-RW made the surfaces smooth with the lowest roughness values in all parameters. The effect was dependent on the materials used. [ABSTRACT FROM AUTHOR]

Decreased neuronal dendrite branching and plasticity of the hippocampus, a limbic structure implicated in mood disorders, is thought to contribute to the symptoms of depression. However, the mechanisms underlying this effect, as well as the actions of antidepressant treatment, remain poorly characterized. Here, we show that hippocampal expression of neuritin, an activity-dependent gene that regulates neuronal plasticity, is decreased by chronic unpredictable stress (CUS) and that antidepressant treatment reverses this effect. We also show that viral-mediated expression of neuritin in the hippocampus produces antidepressant actions and prevents the atrophy of dendrites and spines, as well as depressive and anxiety behaviors caused by CUS. Conversely, neuritin knockdown produces depressive-like behaviors, similar to CUS exposure. The ability of neuritin to increase neuroplasticity is confirmed in models of learning and memory. Our results reveal a unique action of neuritin in models of stress and depression, and demonstrate a role for neuroplasticity in antidepressant treatment response and related behaviors. The biocompatibility of fluoridated apatite (FHA) was significantly improved by the addition of a resorbable phosphate-based glass. With the addition of small amounts of the glass (up to 5%) to the FHA, a completely densified body was obtained after sintering at 1250°C. The resultant crystalline phase was composed of tricalcium phosphate (TCP) and FHA. The addition of the glass to the FHA significantly improved the viability and functional activity of osteoblastic cells, while these were only slightly improved when the glass was added to pure hydroxyapatite, demonstrating that the fluoridation and glass addition had a synergistic stimulatory effect. FJH-KO obtained from Antarctic krill, especially Euphausia superba, has been reported to contain high amounts of omega-3 polyunsaturated fatty acids (n-3 PUFA) and to exhibit anticancer and anti-inflammatory properties. However, its antithrombotic effects have not yet been reported. This study aimed to investigate the antithrombotic effects of FJH-KO in carrageenan-induced thrombosis mouse models and human endothelial cells. Thrombosis was induced by carrageenan injection, whereas the mice received FJH-KO pretreatment. FJH-KO attenuated carrageenan-induced thrombus formation in mouse tissue vessels and prolonged tail bleeding. The inhibitory effect of FJH-KO was associated with decreased plasma levels of thromboxane B2, P-selectin, endothelin-1, β-thromboglobulin, platelet factor 4, serotonin, TNF-α, IL-1β, and IL-6. Meanwhile, FJH-KO induced plasma levels of prostacyclin I2 and plasminogen. In vitro, FJH-KO decreased the adhesion of THP-1 monocytes to human endothelial cells stimulated by TNF-α via eNOS activation and NO production. Furthermore, FJH-KO inhibited the expression of TNF-α-induced adhesion molecules such as ICAM-1 and VCAM-1 by suppressing the NF-κB signaling pathway. Taken together, our study demonstrates that FJH-KO protects against carrageenan-induced thrombosis by regulating endothelial cell activation and has potential as an antithrombotic agent.

Summary: Neuroblastoma is a solid, heterogeneous pediatric tumor. Chemotherapy is widely used to treat neuroblastoma. However, dose-dependent responses and chemoresistance mechanisms of neuroblastoma cells to anticancer drugs remain challenging. Here, we investigated the dose-dependent effects of topotecan on human neuroblastoma cells (SK-N-SH, SH-SY5Y, and SK-N-BE) under various nutrient supply conditions. Serum-starved human neuroblastoma cells showed reduced toxicity. Their survival rate increased upon treatment with a high concentration (1 μM) of topotecan. Quantitative profiling of global and phosphoproteome identified 12,959 proteins and 48,812 phosphosites, respectively, from SK-N-SH cells. Network analysis revealed that topotecan upregulated DNA repair and cholesterol-mediated topotecan efflux, resulting in topotecan resistance. Results of DNA damage assay, cell cycle, and quantitative analyses of membrane cholesterol supported the validity of these resistance factors and their applicability to all neuroblastoma cells. Our results provide a model for high dose-dependent chemoresistance in neuroblastoma cells that could enable a patient-dependent chemotherapy screening strategy.
