Abstract

Wound dressing, which can release anti‐infectives in a controlled way, is taking an important role in the treatment and recovery of the open wound. An adequate release of antibiotics can prevent infections from microorganisms effectively. Among the new candidates of fabricating base materials for wound dressing, electrospinning fiber mats are attracting numerous attentions for their excellent performance in controlled drug delivery. The drug release behavior of electrospinning fiber mats can be tuned by changing the chemical components and the geometric structures of the mats. In this study, fiber mats with different geometric structures, which composed of poly‐ε‐caprolactone (PCL), polyethylene glycol (PEG), and ciprofloxacin (Cip) with different blending ratios, were successfully fabricated by direct‐writing melt electrospinning, and the release behavior of Cip were subsequently investigated in vitro. The results showed that the addition of PEG improved the hydrophilicity of the mats, which in turn affected the manner of drug release. The presence of PEG changed the releasing mechanism from a non‐Fickian diffusion into Fickian diffusion, which indicated that the diffusion of Cip from the composite fiber mats became the main factor of drug release instead of polymer degradation. Besides, with the same composition but different geometric structures, the drug release behavior is of significant difference. Therefore, all the Cip‐loaded composite fiber mats showed antibacterial activities but with different efficiency. Abstract

Protein crystallization is a process that is very sensitive to the physical environment. Audible sound is an environmental characteristic that can significantly affect the crystallization process. Previously, it was found that the crystallization result is frequency dependent, that is, the crystallization of protein under different sound frequencies yields different results. Here, we further investigate the effect of varying frequency (or a frequency program) on protein crystallization. Twelve different frequency programs and six proteins were used to test the effect of varying sound frequency on protein crystallization. The results showed that varying the audible sound frequency from high to low exhibited the most significant improvement in protein crystallization. Varying frequency linearly from 15 000 to 100 Hz in 12 h best promoted crystallization, with the average number of crystallization hits 36.5% higher than in the control. Crystal quality was improved with sound irradiation using STW2 program. Our study showed that varying the sound frequency has positive effects on facilitating protein crystallization and purposely optimizing sound frequency programs may be helpful for obtaining protein crystals.

Abstract

Heat shock protein 90 (Hsp90) is a ubiquitously expressed ATP-dependent molecular chaperone across all species that helps to the correct the folding of many proteins related to important signaling pathways. Tumor cells expressing Hsp90 have more ATP-binding affinity than normal cells. Many correlative inhibitors have been developed to promising anti-tumor strategies and have been evaluated in clinical trials. However, the effect of Hsp90 inhibitors on immunocytes cannot be ignored. Natural killer (NK) cells are key components of the innate immune system that play a pivotal role in tumor surveillance. The present study has investigated the potential effect of four Hsp90 inhibitors (NVP-AUY922, BIIB021, 17-DMAG, and SNX-2112) on human primary NK cells. The viability, cytotoxicity, apoptosis, phenotype, and cytokine secretion of NK cells after inhibitor treatment were assessed. The results of this study demonstrated that the inhibitors had negative effects on NK cell activity in a dose-dependent manner. The four inhibitors significantly reduced the cytotoxicity of the NK cells by decreasing viability, inducing apoptosis and down-regulating the expression of cytokines and functional receptors. These findings suggest that more attention should be given to the effect of Hsp90 inhibitors on NK cell function during clinical trials and also represent a potential immunosuppressant strategy.
