Ips sexdentatus (six-spined engraver beetle) from Austria and Poland were dissected and examined for the presence of pathogens. Specimens collected in Austria were found to contain the ascomycetous fungus Metschnikowia cf. typographi. Infection rates ranged from 3.6% to 26.8% at different collection sites. M. cf. typographi infected midguts were investigated by histological, ultrastructural and molecular techniques. Extraordinary ultrastructural details are shown, such as ascospores with bilateral flattened flanks resembling alar rims at both sides of their attenuating tube-like ends. These have not yet been described in other yeast species. Molecular investigations showed a close phylogenetic relationship to the fungi Metschnikowia agaves and Candida wancherniae. Presence of the entomopathogenic fungus Beauveria bassiana found in Austria was confirmed both morphologically and molecularly. The eugregarine Gregarina typographi was diagnosed most frequently. Infection rates of all I. sexdentatus specimens ranged from 21.4% to 71.9% in Austria and 54.1% to 68.8% in Poland. Other entomopathogenic protists, bacteria, or viruses were not detected.

Polyporoid and corticioid fungi are among the most important wood-decay fungi. Not only do they contribute to nutrient cycling by decomposing wood debris, but they are also valuable sources for natural products. Polyporoid and corticioid wood-inhabiting fungi were investigated in Odaesan National Park. Fruit bodies were collected and identified based on morphological and molecular analyses using 28S and internal transcribed spacer regions of DNA sequences. As a result, a total of 149 species, 69 genera, 22 families, and 11 orders were recognized. Half (74 species) of the species were polypores, and the other half (75 species) were corticioid fungi. Most of the species belonged to Polyporales (92 species) followed by Hymenochaetales (33 species) and Russulales (11 species). At the genus level, a high number of species was observed from Steccherinum, Hyphodontia, Phanerochaete, Postia, and Trametes. Concerning distribution, almost all the species could be found below 1,000 m, and only 20% of the species were observed from above 1,000 m. Stereum subtomentosum, Trametes versicolor, T. hirsuta, T. pubescens, Bjerkandera adusta, and Ganoderma applanatum had wide distribution areas. Deciduous wood was the preferred substrate for the collected species. Sixty-three species were new to this region, and 21 species were new to Korea, of which 17 species were described and illustrated.

Aspergillus (Trichocomaceae, Eurotiales, and Ascomycota) is a genus of well-defined asexual spore-forming fungi that produce valuable compounds such as secondary metabolites and enzymes; however, some species are also responsible for diseases in plants and animals, including humans. To date, 26 Aspergillus species have been reported in Korea, with most species located in terrestrial environments. In our study, Aspergillus species were isolated from mudflats and sea sand along the western and southern coasts of Korea. A total of 84 strains were isolated and identified as 17 Aspergillus species in 11 sections on the basis of both morphological characteristics and sequence analysis of the calmodulin gene (CaM) locus. Commonly isolated species were A. fumigatus (26 strains), A. sydowii (14 strains), and A. terreus (10 strains). The diversity of Aspergillus species isolated from mudflats (13 species) was higher than the diversity of those from sea sand (five species). Four identified species-A. caesiellus, A. montenegroi, A. rhizopodus, and A. tabacinus-are in the first records in Korea. Here, we provide detailed descriptions of the morphological characteristics of these four species.

Members of the genus Penicillium are commonly isolated from various terrestrial and marine environments, and play an important ecological role as a decomposer. To gain insight into the ecological role of Penicillium in intertidal zones, we investigated the Penicillium diversity and community structure using a culture-dependent technique and a culture independent metagenomic approach using ITS (ITS-NGS) and partial β-tubulin (BenA-NGS) as targets. The obtained isolates were tested for halotolerance, enzyme activity, and polycyclic aromatic hydrocarbons (PAHs) degradation. A total of 96 Penicillium species were identified from the investigated intertidal zones. Although the BenA-NGS method was efficient for detecting Penicillium, some species were only detected using conventional isolation and/or the ITS-NGS method. The Penicillium community displayed a significant degree of variation relative to season (summer and winter) and seaside (western and southern coast). Many Penicillium species isolated in this study exhibited cellulase and protease activity, and/or degradation of PAHs. These findings support the important role of Penicillium in the intertidal zone for nutrient recycling and pollutant degradation.

Pine mushroom (Tricholoma matsutake) is one of the most valued ectomycorrhizal fungi in Asia because of its unique pine-like aroma, however, due to exceptionally slow growth of its mycelia in artificial conditions, its cultivation has been largely deemed as not possible. Previous studies have shown that some bacteria and a few Trichoderma species associated with pine mushroom promoted the growth of T. matsutake isolate, but this effect is relatively unexplored. In this study, we investigated the diversity of microfungi in the fairy ring of T. matsutake and their effect on the growth of T. matsutake isolate. From 184 fungal isolates, 28 species were identified based on suitable molecular markers. Penicillium was most frequently observed (16 species), followed by Trichoderma (4 species). Five Zygomycota species showed a high promoting effect on the growth of T. matsutake while the effects of ascomycetes were mixed. The microfungi that promote the growth of T. matsutake can be useful for forest nursery and artificial cultivation of T. matsutake.

During the 2014 survey of the mushroom flora of Gwangneung forest in South Korea, we collected two specimens of boletoid mushroom growing on a felled tree of Pinus koraiensis. These specimens were characterized by a light brown to reddish-brown pileus with appressed tomentum, pore surface bluing instantly when bruised, golden-yellow mycelium at the base of stipe, and lignicolous habitat. Both specimens were identified as Buchwaldoboletus lignicola, a rare basidiomycete, based on morphological characteristics and sequences of internal transcribed spacer (ITS; fungal barcode). Here, we describe these specimens and provide the first report of this genus in South Korea.

Bacterial nanocellulose (BNC) which is generally synthesized by several species of bacteria has a wide variety of industrial uses, particularly in the food and material industries. However, the low levels of BNC production during the fermentation process should be overcome to reduce its production cost. Therefore, in this study, we screened and identified a new cellulose-producing bacterium, optimized production of the cellulose, and investigated the morphological properties of the cellulosic materials. Out of 147 bacterial isolates from ripened fruits and traditional vinegars, strain SFCB22-18 showed the highest capacity for BNC production and was identified as

Agarum clathratum, a brown macroalgae species, has recently become a serious environmental problem on the coasts of Korea. In an effort to solve this problem, fungal diversity associated with decaying A. clathratum was investigated and related β-glucosidase and endoglucanase activities were described. A total of 233 fungal strains were isolated from A. clathratum at 15 sites and identified 89 species based on morphology and a multigene analysis using the internal transcribed spacer region (ITS) and protein-coding genes including actin (act), β-tubulin (benA), calmodulin (CaM), and translation elongation factor (tef1). Acremonium, Corollospora, and Penicillium were the dominant genera, and Acremonium fuci and Corollospora gracilis were the dominant species. Fifty-one species exhibited cellulase activity, with A. fuci, Alfaria terrestris, Hypoxylon perforatum, P. madriti, and Pleosporales sp. Five showing the highest enzyme activities. Further enzyme quantification confirmed that these species had higher cellulase activity than P. crysogenum, a fungal species described in previous studies. This study lays the groundwork for bioremediation using fungi to remove decaying seaweed from populated areas and provides important background for potential industrial applications of environmentally friendly processes.

The production of rechargeable batteries is rapidly expanding, and there are going to be new challenges in the near future about how the potential environmental impact caused by the disposal of the large volume of the used batteries can be minimized. Herein, a novel strategy is proposed to address these concerns by applying biodegradable device technology. An eco-friendly and biodegradable sodium-ion secondary battery (SIB) is developed through extensive material screening followed by the synthesis of biodegradable electrodes and their seamless assembly with an unconventional biodegradable separator, electrolyte, and package. Each battery component decomposes in nature into non-toxic compounds or elements via hydrolysis and/or fungal degradation, with all of the biodegradation products naturally abundant and eco-friendly. Detailed biodegradation mechanisms and toxicity influence of each component on living organisms are determined. In addition, this new SIB delivers performance comparable to that of conventional non-degradable SIBs. The strategy and findings suggest a novel eco-friendly biodegradable paradigm for large-scale rechargeable battery systems.

A total of 28 strains of 19 Penicillium species were isolated in a survey of extracellular enzyme-producing fungi from macroalgae along the coast of Jeju Island of Korea. Penicillium species were identified based on morphological and β-tubulin sequence analyses. In addition, the halo-tolerance and enzyme activity of all strains were evaluated. The diversity of Penicillium strains isolated from brown algae was higher than the diversity of strains isolated from green and red algae. The commonly isolated species were Penicillium antarcticum, P. bialowiezense, P. brevicompactum, P. crustosum, P. oxalicum, P. rubens, P. sumatrense, and P. terrigenum. While many strains showed endoglucanase, β-glucosidase, and protease activity, no alginase activity was detected. There was a positive correlation between halo-tolerance and endoglucanase activity within Penicillium species. Among 19 Penicillium species, three species-P. kongii, P. olsonii, and P. viticola-have not been previously recorded in Korea.

Most known species in the Strophariaceae are decomposers and grow on various kind of organic matter. Approximately 18 genera and 1,316 species in the Strophariaceae have been reported worldwide. Through an ongoing survey of indigenous fungi in Korea, 29 specimens belonging to the Strophariaceae were collected from 2012 to 2016. These specimens were identified based on morphological characteristics and molecular analysis of internal transcribed spacer sequences. Fifteen taxa were confirmed, with eight species matching those previously recorded. Seven species in five genera were shown to be new records in Korea: Galerina marginata, Gymnopilus crociphyllus, Gymnopilus picreus, Hebeloma birrus, Hebeloma cavipes, Pholiota multicingulata, and Psilocybe thaizapoteca. In this study, we provide detailed morphological descriptions of these species and investigate their evolutionary relationships by constructing phylogenetic trees.

Succession is defined as variation in ecological communities caused by environmental changes. Environmental succession can be caused by rapid environmental changes, but in many cases, it is slowly caused by climate change or constant low-intensity disturbances. Odaesan National Park is a well-preserved forest located in the Taebaek mountain range in South Korea. The forest in this national park is progressing from a mixed-wood forest to a broad-leaved forest. In this study, microbial community composition was investigated using 454 sequencing of soil samples collected from 13 different locations in Odaesan National Park. We assessed whether microbial communities are affected by changes in environmental factors such as water content (WC), nutrient availability (total carbon (TC) and total nitrogen (TN)) and pH caused by forest succession. WC, TC, TN and pH significantly differed between the successional stages of the forest. The WC, TC and TN of the forest soils tended to increase as succession progressed, while pH tended to decrease. In both successional stages, the bacterial genus Pseudolabrys was the most abundant, followed by Afipia and Bradyrhizobium. In addition, the fungal genus Saitozyma showed the highest abundance in the forest soils. Microbial community composition changed according to forest successional stage and soil properties (WC, TC, TN, and pH). Furthermore, network analysis of both bacterial and fungal taxa revealed strong relationships of the microbial community depending on the soil properties affected by forest succession.

Microorganisms are regarded as a sustainable source of biologically active molecules. Among them, Trichoderma spp. have been an attractive source of biological compounds. However, the study of marine-derived Trichoderma has developed slowly because of the difficulty in isolating the fungi. In our study, 30 strains of marine-derived Trichoderma were identified through the translation elongation factor 1-alpha (EF1&alpha, ) sequences, and their biological activities, such as antioxidant activity by ABTS and DPPH assays, antifungal activity against Asteromyces cruciatus and Lindra thalassiae, and tyrosinase inhibition activity, were investigated. As a result, the 30 marine Trichoderma species were classified into 21 taxa, including three new species candidates. Three strains of T. asperellum showed the highest ABTS radical scavenging activity and antifungal activity. T. bissettii SFC20170821-M05 and T. guizhouense SFC20180619-M23 showed notable DPPH radical scavenging activity and tyrosinase inhibition activity, respectively. This study showed the potential of marine-derived Trichoderma as a source of bioactive compounds.

Wood-decay fungi (WDF) play a significant role in recycling nutrients, using enzymatic and mechanical processes to degrade wood. Designated as a biodiversity hot spot, Central Siberia is a geographically important region for understanding the spatial distribution and the evolutionary processes shaping biodiversity. There have been several studies of WDF diversity in Central Siberia, but identification of species was based on morphological characteristics, lacking detailed descriptions and molecular data. Thus, the aim of this study was to identify WDF in Central Siberia, regarding the degradation of host trees based on both morphological and molecular analyses. We collected 106 WDF samples from Krasnoyarsk and the Republic of Khakassia in 2014 and 2017, and identified a total of 52 fungal species from six main host tree genera. In order to assess the host preference of the WDF, we examined previous literature, and data from this study. We confirmed a division in host preference of WDF between gymnosperms and angiosperms. DNA-based identification and host preference assessment of the WDF provide preliminary data on WDF diversity and their role in nutrient cycles in the ecosystem of Central Siberia. To fully understand WDF diversity in Central Siberia, continuous long-term surveys, including DNA sequence data, are needed.

Tricholoma matsutake (pine mushroom, PM) is a prized mushroom in Asia due to its unique flavor and pine aroma. The fruiting body of PM forms only in its natural habitat (pine forest), and little is known regarding the natural conditions required for successful generation of the fruiting bodies in this species. Recent studies suggest that microbial interactions may be associated with the growth of PM; however, there have been few studies of the bacterial effects on PM growth. In this study, we surveyed which bacteria can directly and indirectly promote the growth of PM by using co-cultures with PM and molds associated with the fruiting body. Among 16 bacterial species isolated from the fruiting body, some species significantly influenced the mycelial growth of PM and molds. Most bacteria negatively affected PM growth and exhibited various enzyme activities, which suggests that they use the fruiting body as nutrient source. However, growth-promoting bacteria belonging to the Dietzia, Ewingella, Pseudomonas, Paenibacillus, and Rodococcus were also found. In addition, many bacteria suppressed molds, which suggests an indirect positive effect on PM as a biocontrol agent. Our results provide important insights toward a better understanding of the microbial interactions in the fruiting body of PM, and indicate that growth-promoting bacteria may be an important component in successful cultivation of PM.

Depending on the mode of nutrition exploitation, major fungal guilds are distinguished as ectomycorrhizal and saprotrophic fungi. It is generally known that diverse environmental factors influence fungal communities; however, it is unclear how fungal communities respond differently to environment factors depend on fungal guilds. In this study, we investigated basidiomycetes communities associated with Quercus mongolica using 454 pyrosequencing. We attempted to detect guild pattern (ectomycorrhizal or saprotrophic fungal communities) by comparing the influence of geography and source (root and surrounding soil). A total of 515 mOTUs were detected from root (321) and soil (394) of Q. mongolica at three sites of Mt. Jeombong in Inje County. We found that patterns of diversity and community structure were different depending on the guilds. In terms of alpha diversity, only ectomycorrhizal fungi showed significant differences between sources. In terms of community structure, however, geography significantly influenced the ectomycorrhizal community, while source appeared to have a greater influence on the saprotrophic community. Therefore, a guild-based view will help to elucidates novel features of the relationship between environmental factors and fungal communities.

The polypore genus Abundisporus Ryvarden is characterized by resupinate to pileate fruitbodies with a purplish brown hymenophore, slightly thick-walled, pale yellowish and non-dextrinoid basidiospores, and causing white rot. A purple color hymenophore, an easily observable and striking character, was considered the main distinctive feature at the generic level within polypores. However, due to highly similar basidiocarp features, species identification within these purple polypores is particularly difficult. Three species of purple colored polypores have been reported in Korea (Abundisporus fuscopurpureus, A. pubertatis, and Fomitopsis rosea). Based on morphological re-examination, ecological information, and sequence analysis of the internal transcribed spacer, we showed that previous classification was incorrect and there is only one species (A. pubertatis) in Korea. We provide a detailed description of A. pubertatis in Korea, as well as a taxonomic key to distinguish wood rot fungi with a purple hymenophore.

In this study, three different methods (fruiting body collection, mycelial isolation, and 454 sequencing) were implemented to determine the diversity of wood-inhabiting basidiomycetes from dead Manchurian fir (Abies holophylla). The three methods recovered similar species richness (26 species from fruiting bodies, 32 species from mycelia, and 32 species from 454 sequencing), but Fisher’s alpha, Shannon-Wiener, Simpson’s diversity indices of fungal communities indicated fruiting body collection and mycelial isolation displayed higher diversity compared with 454 sequencing. In total, 75 wood-inhabiting basidiomycetes were detected. The most frequently observed species were Heterobasidion orientale (fruiting body collection), Bjerkandera adusta (mycelial isolation), and Trichaptum fusco-violaceum (454 sequencing). Only two species, Hymenochaete yasudae and Hypochnicium karstenii, were detected by all three methods. This result indicated that Manchurian fir harbors a diverse basidiomycetous fungal community and for complete estimation of fungal diversity, multiple methods should be used. Further studies are required to understand their ecology in the context of forest ecosystems.

The veiled polypore, Cryptoporus volvatus, is distributed widely in North America and East Asia and is believed to have a mutualistic relationship with coleopteran species—the fungus providing food and shelter in basidiocarps and beetle helping disperse spores. Seventy fresh basidiocarps of C. volvatus were collected from the Japanese red pine (Pinus densiflora) in the spring season of 2013 from two sites in Korea. A total of 251 insects (101 adult and 150 larvae) were collected from the inside of basidiocarps and identified using morphology and mitochondrial cytochrome c oxidase subunit I (COI) sequences. Six species belonging to five coleopteran families were identified. The number of spores attached to the bodies of adult insects was counted and average spore counts for each of the six species ranged between 1.0 × 104 and 5.2 × 105 spores/individual. Across localities, three species were shared (Aethina suturalis, Trogossita japonica and Parabolitophagus felix) and carried spores at high densities on their bodies, making them more likely to aid in spore dispersal.

In South Korea, three species of Heterobasidion, H. annosum, H. araucariae and H. insulare have been recorded. However, these species have not been found in the neighboring countries such as China and Japan. For resolving this matter, a total of 11 materials of Heterobasidion spp. from South Korea were gathered and morphologically examined. Species descriptions from the previous studies were referred if the materials were not available. Phylogenetic analyses using ITS and TEF gene sequences were also performed to infer the species relationships of the Korean Heterobasidion spp with their allied species. Through the morphological investigations and phylogenetic analyses we found that specimens recorded as H. annosum and H. araucariae are actually H. ecrustosum, and the ones recorded as H. insulare are actually H. orientale. The two recognized species are described and illustrated.

White-rot fungi of the genus Bjerkandera are cosmopolitan and have shown potential for industrial application and bioremediation. When distinguishing morphological characters are no longer present (e.g., cultures or dried specimen fragments), characterizing true sequences of Bjerkandera is crucial for accurate identification and application of the species. To build a framework for molecular identification of Bjerkandera, we carefully identified specimens of B. adusta and B. fumosa from Korea based on morphological characters, followed by sequencing the internal transcribed spacer region and 28S nuclear ribosomal large subunit. The phylogenetic analysis of Korean Bjerkandera specimens showed clear genetic differentiation between the two species. Using this phylogeny as a framework, we examined the identification accuracy of sequences available in GenBank. Analyses revealed that many Bjerkandera sequences in the database are either misidentified or unidentified. This study provides robust reference sequences for sequence-based identification of Bjerkandera, and further demonstrates the presence and dangers of incorrect sequences in GenBank.

This is a continuity of a series of taxonomic and phylogenetic papers on the fungi where materials were collected from many countries, examined and described. In addition to extensive morphological descriptions and appropriate asexual and sexual connections, DNA sequence data are also analysed from concatenated datasets to infer phylogenetic relationships and substantiate systematic positions of taxa within appropriate ranks. Wherever new species or combinations are proposed, we apply an integrative approach using morphological and molecular data as well as ecological features wherever applicable. Notes on 112 fungal taxa are compiled in this paper including Biatriosporaceae and Roussoellaceae, Didysimulans gen. nov., 81 new species, 18 new host records and new country records, five reference specimens, two new combinations, and three sexual and asexual morph reports. The new host records and country records are Ascochyta medicaginicola, Boletellus emodensis, Cyptotrama asprata, Cytospora ceratosperma, Favolaschia auriscalpium, F. manipularis, Hysterobrevium mori, Lentinus sajor-caju, L. squarrosulus, L. velutinus, Leucocoprinus cretaceus, Lophiotrema vagabundum, Nothophoma quercina, Platystomum rosae, Pseudodidymosphaeria phlei, Tremella fuciformis, Truncatella spartii and Vaginatispora appendiculata and three sexual and asexual morphs are Aposphaeria corallinolutea, Dothiora buxi and Hypocrella calendulina.

The fungal genus Suillus Micheli ex S. F. Gray plays important roles in the survival and growth of plant seedlings. Humans have utilized these ectomycorrhizal fungi to enhance the nutrient uptake and defense systems of plants, particularly in the reforestation of coniferous forests. The genus Suillus is easily distinguishable by its distinctive morphological features, although the morphology of the fruiting body does not facilitate reliable interspecies discrimination. On the basis of micro-morphological features and internal transcribed spacer sequence analysis, we found that 51 of 117 Korean Suillus specimens had initially been misidentified. The list of the 12 Suillus species previously recorded in Korea was re-evaluated and revised to only eight distinct species: S. americanus, S. bovinus, S. granulatus, S. grevillei, S. luteus, S. pictus, S. placidus, and S. viscidus. We provide taxonomical descriptions for six of these species from the sample specimens.

Distinguishing individual Russula species can be very difficult due to extensive phenotypic plasticity and obscure morphological and anatomical discontinuities. In this study, we use the internal transcribed spacer (ITS) and 28S nuclear ribosomal large subunit (LSU) markers to identify and study the genetic diversity of species in the Russula subgenus Compacta in Korea. We focus on two morphologically similar species that are often misidentified for each other: R. nigricans and R. subnigricans. Based on molecular phylogenetic analyses, we identify three subgroups of R. nigricans, with two from Asia and one from Europe/North America. Surprisingly, we find Korean R. subnigricans are more closely related to R. eccentrica from North America than the type specimen of R. subnigricans from Japan. These molecular data, along with habitat data, reveal that Korean R. subnigricans had previously been misclassified and should now be recognized as R. eccentrica. Both ITS and LSU exhibit high interspecific and low intraspecific variation for R. eccentrica, R. nigricans, and R. subnigricans. These markers provide enough resolutional power to differentiate these species and uncover phylogeographic structure, and will be powerful tools for future ecological studies of Russula.

The wood decay fungi Antrodia P. Karst. play important ecological roles and have significant industrial and economic impacts as both wood degraders and sources of pharmaceutical and biotechnological products. Although each Antrodia species has distinct morphological characteristics, the misidentification rate is especially high due to their simple morphological characters. A combination of morphological and internal transcribed spacer region sequence analyses revealed that 27 of 89 specimens previously identified by morphology alone were correct, whereas 35 of these specimens were misidentified as other Antrodia species. We report here that seven Antrodia species exist in Korea (A. albida, A. heteromorpha, A. malicola, A. serialis, A. sinuosa, A. sitchensis, and A. xantha) and based on these specimens, we provide taxonomic descriptions of these species, except for A. serialis, which was only confirmed by isolate.

In Seoul, a majority of plant communities have undergone significant changes over the last few decades; however, how wood decay fungi have responded and adapted to the changes in vegetation remains unknown. Through an ongoing investigation of Korean indigenous fungi, ca. 300 specimens with poroid basidiocarp were collected in Seoul during 2008~2012. Morphological examination and molecular analysis using the internal transcribed spacer and nuclear large subunit ribosomal DNA region sequences helped identify 38 species belonging to 28 genera, 10 families, and 5 orders in this area. Among them, three polypores, Abundisporus pubertatis, Coriolopsis strumosa, and Perenniporia maackiae were found to be new to South Korea.

Fungal spores are widespread and common in the atmosphere. In this study, we use a metagenomic approach to study the fungal diversity in six total air samples collected from April to May 2012 in Seoul, Korea. This springtime period is important in Korea because of the peak in fungal spore concentration and Asian dust storms, although the year of this study (2012) was unique in that were no major Asian dust events. Clustering sequences for operational taxonomic unit (OTU) identification recovered 1,266 unique OTUs in the combined dataset, with between 223᾿96 OTUs present in individual samples. OTUs from three fungal phyla were identified. For Ascomycota, Davidiella (anamorph: Cladosporium) was the most common genus in all samples, often accounting for more than 50% of all sequences in a sample. Other common Ascomycota genera identified were Alternaria, Didymella, Khuskia, Geosmitha, Penicillium, and Aspergillus. While several Basidiomycota genera were observed, Chytridiomycota OTUs were only present in one sample. Consistency was observed within sampling days, but there was a large shift in species composition from Ascomycota dominant to Basidiomycota dominant in the middle of the sampling period. This marked change may have been caused by meteorological events. A potential set of 40 allergy-inducing genera were identified, accounting for a large proportion of the diversity present (22.5᾿7.2%). Our study identifies high fungal diversity and potentially high levels of fungal allergens in springtime air of Korea, and provides a good baseline for future comparisons with Asian dust storms.

The Black Pine, Pinus thunbergii, is widely distributed along the eastern coast of Korea and its importance as a shelterbelt was highlighted after tsunamis in Indonesia and Japan. The root endophytic diversity of P. thunbergii was investigated in three coastal regions; Goseong, Uljin, and Busan. Fungi were isolated from the root tips, and growth rates of pure cultures were measured and compared between PDA with and without 3% NaCl to determine their saline resistance. A total of 259 isolates were divided into 136 morphotypes, of which internal transcribed spacer region sequences identified 58 species. Representatives of each major fungi phylum were present: 44 Ascomycota, 8 Zygomycota, and 6 Basidiomycota. Eighteen species exhibited saline resistance, many of which were Penicillium and Trichoderma species. Shoreline habitats harbored higher saline-tolerant endophytic diversity compared with inland sites. This investigation indicates that endophytes of P. thunbergii living closer to the coast may have higher resistance to salinity and potentially have specific relationships with P. thunbergii.

Pine mushroom (PM, Tricholoma matsutake) is an important ectomycorrhizal fungus in Asia primarily due to its value as a food delicacy. Recent studies have shown that fairy rings of PM have distinctive fungal communities, which suggests that other fungi influence the growth of PM. Trichoderma is a well-known saprotrophic fungus commonly found in pine roots within PM fairy rings; however, little is known about the diversity of Trichoderma associated with PM and how these species influence PM growth. This study focused on diversity of Trichoderma isolated from pine roots within PM fairy rings and how these species affect the growth of PM isolate. Based on tef1a phylogenetic analyses, nine Trichoderma species (261 isolates) were identified. Trichoderma songyi and T. spirale were the dominant species, and Trichoderma community varied geographically. Growth experiments indicated that metabolites from five Trichoderma species had a significant influence on the growth of PM isolates. Metabolites of two Trichoderma species increased PM growth, while those of three Trichoderma species suppressed the growth. Within the fairy rings, Trichoderma that had a positive or neutral effect comprised the majority of Trichoderma communities. The results of this study suggest that various Trichoderma species co-exist within PM fairy rings and that these species influence PM growth.

The wood decay fungi are a diverse taxonomic group that plays a pivotal role in forest carbon cycling. Wood decay fungi use various enzymatic pathways to digest dead or living wood in order to obtain carbon and other nutrients and these enzymatic systems have been exploited for both industrial and medical applications. Over 600 wood decay fungi species have been described in Korea; however, the recent application of molecular markers has dramatically altered the taxonomy of many of these wood decay fungi at both the genus and species levels. By combining molecular methods, specifically sequences of the internal transcribed spacer region, with traditional morphological characters, this study identified five new species records for Korea in five genera: Aurantiporus, Favolus, Neofavolus, Loweomyces, and Hymenochaetopsis. Three of these genera (Aurantiporus, Favolus, and Loweomyces) were previously unknown in Korea. The relatively simple morphology of the wood decay fungi often leads to ambiguous taxonomic assignment. Therefore, molecular markers are a necessary component of any taxonomic or evolutionary study of wood decay fungi. Our study highlights the need for a more robust and multifaceted approach in investigating new wood decay fungi in Korea.

Agarum clathratum, brown algae, play important ecological roles in marine ecosystem, but can cause secondary environment pollution when they pile up on the beach. In order to resolve the environment problem by A. clathratum, we focus to isolate and identify Penicillium because many species are well known to produce extracellular enzymes. A total of 32 Penicillium strains were isolated from A. clathratum samples that collected from 13 sites along the mid-east coast of Korea in summer. They were identified based on morphological characters and phylogenetic analysis using β-tubulin DNA sequences as well as a combined dataset of β-tubulin and calmodulin. A total of 32 strains were isolated and they were identified to 13 Penicillium species. The commonly isolated species were Penicillium citrinum, P. roseomaculatum, and Penicillium sp. Among 13 Penicillium species, four species - P. bilaiae, P. cremeogriseum, P. madriti, and P. roseomaculatum - have not been previously recorded in Korea. For these four new species records to Korea, we provide morphological characteristics of each strain.

Tricholoma matsutake, the pine mushroom, is a valuable forest product with high economic value in Asia, and plays an important ecological role as an ectomycorrhizal fungus. Around the host tree, T. matsutake hyphae generate a distinctive soil aggregating environment called a fairy ring, where fruiting bodies form. Because T. matsutake hyphae dominate the soil near the fairy ring, this species has the potential to influence the microbial community. To explore the influence of T. matsutake on the microbial communities, we compared the microbial community and predicted bacterial function between two different soil types D T. matsutake dominant and T. matsutake minor. DNA sequence analyses showed that fungal and bacterial diversity were lower in the T. matsutake dominant soil compared to T. matsutake minor soil. Some microbial taxa were significantly more common in the T. matsutake dominant soil across geographic locations, many of which were previously identified as mycophillic or mycorrhiza helper bacteria. Between the two soil types, the predicted bacterial functional profiles (using PICRUSt) had significantly distinct KEGG modules. Modules for amino acid uptake, carbohydrate metabolism, and the type III secretion system were higher in the T. matsutake dominant soil than in the T. matsutake minor soil. Overall, similar microbial diversity, community structure, and bacterial functional profiles of the T. matsutake dominant soil across geographic locations suggest that T. matsutake may generate a dominance effect.

During the investigation of Korean indigenous fungi from Seoul, three genera-Fuscoporia, Porostereum, and Trametopsis, and four species-Fuscoporia senex, Phlebia acerina, Porostereum spadiceum, and Trametopsis cervina were found. Their morphological characteristics were examined and their identification was confirmed by molecular analysis based on internal transcribed spacer (ITS) and nuclear large subunit ribosomal DNA region sequences. These fungi are new to Korea and registered here with descriptions.

Marine environments are unique habitats for microorganisms and represent uninvestigated areas that possess valuable resources. Fungi may also be important because they contribute to marine ecosystems as decomposers. The aims of this study were to investigate the halo-tolerance of marine-derived fungi and their enzymatic properties. Eighteen fungal strains representing 11 different species were used, which included 17 ascomycetes and a zygomycete. The majority were not affected by salinity and showed endo-glucanase (EG) and β-glucosidase (BGL) activities. Interestingly, the cellulolytic enzyme activity derived from Penicillium chrysogenum FU42 increased with salinity. To investigate whether this increase was due to an adaptation or an innate ability of the species, P. chrysogenum KCTC6933, which originated from a terrestrial environment, was used as a control, and its enzymatic properties were compared. Consequently, P. chrysogenum FU42, which was derived from the ocean, showed unique enzymatic properties that might enable the fungus to live in the ocean and contribute to the nutrient cycle in marine ecosystems.

When lodgepole pines ( Pinus contorta Douglas ex Louden var. latifolia Engelm. ex S. Watson) that are killed by the mountain pine beetle ( Dendroctonus ponderosae ) and its fungal associates are not harvested, fungal decay can affect wood and fibre properties. Ophiostomatoids stain sapwood but do not affect the structural properties of wood. In contrast, white or brown decay basidiomycetes degrade wood. We isolated both staining and decay fungi from 300 lodgepole pine trees killed by mountain pine beetle at green, red, and grey stages at 10 sites across British Columbia. We retained 224 basidiomycete isolates that we classified into 34 species using morphological and physiological characteristics and rDNA large subunit sequences. The number of basidiomycete species varied from 4 to 14 species per site. We assessed the ability of these fungi to degrade both pine sapwood and heartwood using the soil jar decay test. The highest wood mass losses for both sapwood and heartwood were measured for the brown rot species Fomitopsis pinicola and the white rot Metulodontia and Ganoderma species. The sap rot species Trichaptum abietinum was more damaging for sapwood than for heartwood. A number of species caused more than 50% wood mass losses after 12 weeks at room temperature, suggesting that beetle-killed trees can rapidly lose market value due to degradation of wood structural components.

Seventy-nine white rot strains were screened to determine if they had the potential for use in the degradation of oligocyclic aromates (PAHs) by measuring their dye-decoloration rate. Fourteen strains that were selected based on their dye-decoloration rate were then evaluated for the ability to tolerate various levels of PAHs spiked in agar medium. The ability of white rot fungi to degrade 3- or 4-ring PAHs (anthracene, phenanthrene, fluoranthene, pyrene) was determined. Two strains of Phanerochaete sordida (KUC8369, KUC8370) were possible PAHs degraders, degrading a significantly greater amount of phenanthrene and fluoranthene than the culture collection strain P. chrysosporium (a known PAHs degrader). The production of manganese peroxidase, the only extracellular ligninolytic enzyme detected during the cultivation, was evaluated.

This study was conducted to generate information regarding the diversity of fungi inhabiting creosote-treated wood in a storage yard for crosstie wastes in Gwangmyeong, Korea. Additionally, the resistance to polycyclic aromatic hydrocarbons (PAHs) of indigenous fungi that mainly occupy creosote-treated wood was evaluated. We isolated fungi from the surface and inner area of crosstie wastes and identified them using a combination of traditional methods and molecular techniques. Overall, 179 isolates including 47 different species were isolated from 240 sampling sites. The identified fungal species included 23 ascomycetes, 19 basidiomycetes, and 5 zygomycetes. Three species, Alternaria alternata, Irpex lacteus, and Rhizomucor variabilis, were the most frequently isolated ascomycetes, basidiomycetes, and zygomycetes, respectively. The results of this study showed that there was a large difference in the fungal diversity between the surface and the inner area. Additionally, zygomycetes and ascomycetes were found to have a greater tolerance to PAHs than basidiomycetes. However, two basidiomycetes, Heterobasidion annosum and Schizophyllum commune, showed very high resistance to PAHs, even in response to the highest concentration (1,000 ppm), which indicates that these species may play a role in the degradation of PAHs.

Ceratocystiopsis minuta (Siemaszko) H.P. UpadhyayW.B. Kendr., originally isolated in Poland, is the type species of genus Ceratocystiopsis H.P. UpadhyayW.B. Kendr. Species in this genus are characterized by dark perithecia with short conical beaks, usually with convergent ostiolar hyphae and dark ascocarps, and by falcate or lunate ascospores. Work within the genus is complicated by historical inconsistencies, errors in strain identification and the absence of a holotype specimen. We used sequence data from the beta-tubulin gene, internal transcribed spacer and large subunit regions of ribosomal DNA to phylogenetically characterize 23 putative strains of Cop. minuta from Europe, Japan and North America, as well as strains from other species in genus Ceratocystiopsis. Our results show that Cop. minuta strains from Europe and Japan are monophyletic, whereas those from North American are polyphyletic and likely misidentified. This suggests that prior research groups have used misidentified strains of Cop. minuta or fungal strains that were only distantly related to the Cop. minuta strain originally described from Poland. Further our multigene phylogenetic analysis also shows that Cop. minuta strains from Europe and Japan can be segregated into three clades. This suggests the presence of several phylogenetic species that are morphologically similar to Cop. minuta, and we anticipate that this species complex will challenge researchers until such relationships are resolved.

The successional diversity of basidiomycetous fungi was studied on Korean pine (Pinus koraiensis) and pitch pine (Pinus rigida) logs that were left exposed on the forest floor based on fungal surveys conducted after 18, 42, and 54 months of exposure. A total of 131 basidiomycetous isolates were recovered from the logs and grouped by their cultural morphology. Fungal identification was achieved by a BLAST search of partial nuclear large subunit ribosomal DNA sequences in GenBank. The results of the 18 month survey revealed that the isolation frequency and fungal diversity were higher for pitch pine than Korean pine. The dominant species found on Korean and pitch pine logs during the first survey were Hypochnicium karstenii and Phlebiopsis gigantea, respectively. In the 42 and 54 month surveys, the primary species were replaced by Hypochnicium eichleri, Phanerochaete velutina, Phlebia radiata, Rhizochaete sp., and Trametes versicolor. These results showed that decay fungi have host preference for woody materials that vary according to the species of tree being colonized.

We tested the effect of leaching on the concentration of western red cedar (WRC; Thuja plicata Donn ex D. Don) heartwood extractives that are known to exhibit antimicrobial activity and correlated this with fungal growth and decay. We assessed the extractive tolerance of the following fungal species: Acanthophysium lividocaeruleum, Coniophora puteana, Heterobasidion annosum, Pachnocybe ferruginea, Phellinus sulphurascens, and Phellinus weirii by measuring their growth rate (mm/day) on media with or without WRC leachate. These data were correlated with the ability of the fungal species to grow on and decay leached versus nonleached WRC. We used an ergosterol assay to estimate growth and a standard soil-block test to assess decay. We estimated that leaching reduced the concentration of 5 major extractives: (-)-plicatic acid, beta-thujaplicin, gamma-thujaplicin, beta-thujaplicinol, and thujic acid by approximately 80%. Phellinus sulphurascens exhibited the lowest extractive-tolerance in vitro, grew poorly on and caused minimal decay in nonleached WRC, but it grew well on and decayed pine and leached WRC. Coniophora puteana, H. annosum, and P. weirii displayed moderate to high tolerance to leachate, grew on and caused decay in nonleached as well as leached WRC, but their growth and decay were always greatest on leached WRC and pine, suggesting that leaching enhances decay by these fungi. Acanthophysium lividocaeruleum and Pachnocybe ferruginea exhibited high extractive-tolerance. Whereas A. lividocaeruleum clearly caused decay on all types of wood, no decay was observed with Pachnocybe ferruginea, which grew very slowly in the different wood species, and it may or may not be able to decay wood.

The entomopathogenic fungal genusLecanicilliumGams and Zare includes species that are highly pathogenic to many genera of insects. Three species,Lecanicillium longisporum(Petch) Zare and W. Gams,L. muscarium(Petch) Zare and W. Gams, andL. pissodisKope and Leal, were found to be entomopathogens of adult white pine weevils,Pissodes strobi(Peck), in coastal British Columbia. Morphological characteristics were used to identify these species, but variation in conidial shape and size made it difficult to classify some of the isolates into the correct species ofLecanicillium. To confirm the identity of theseLecanicilliumspecies, we used molecular tools such as polymerase chain reaction – restriction fragment length polymorphism and DNA sequencing.

Characterization of the virulence of bark beetle-vectored fungi is important for assessing potential impacts of beetle outbreaks. Massive inoculation of trees with a cork borer appears to give the most accurate estimate of fungal virulence, but cork borer inoculation is time and labor intensive. In October 2003, 18 Pinus contorta var. latifolia were inoculated with a beetle-associated fungus, Grosmannia clavigera (Robinson-Jeffrey and Davidson) Zipfel et al., at densities of 200 and 800 cork borer holes m−2. In July 2004 nine trees were inoculated using bark flap inoculations. The fungal-induced moisture content reduction, sapwood occlusion area and needle discoloration were similar for the 800 cork borer holes m−2 density and for bark flap inoculations, while pathogenicity symptoms induced by the 200 cork borer holes m−2 were less intense.
