Abstract Noninflammatory clearance of dying cells by professional phagocytes, termed efferocytosis, is fundamental in both homeostasis and inflammatory fibrosis disease but has not been confirmed to occur in chronic pancreatitis (CP). Here, we investigated whether efferocytosis constitutes a novel regulatory target in CP and its mechanisms. PRSS1 transgenic (PRSS1 Tg ) mice were treated with caerulein to mimic CP development. Phospholipid metabolite profiling and epigenetic assays were performed with PRSS1 Tg CP models. The potential functions of Atp8b1 in CP model were clarified using Atp8b1-overexpressing adeno-associated virus, immunofluorescence, enzyme-linked immunosorbent assay(ELISA), and lipid metabolomic approaches. ATAC-seq combined with RNA-seq was then used to identify transcription factors binding to the Atp8b1 promoter, and ChIP-qPCR and luciferase assays were used to confirm that the identified transcription factor bound to the Atp8b1 promoter, and to identify the specific binding site. Flow cytometry was performed to analyze the proportion of pancreatic macrophages. Decreased efferocytosis with aggravated inflammation was identified in CP. The lysophosphatidylcholine (LPC) pathway was the most obviously dysregulated phospholipid pathway, and LPC and Atp8b1 expression gradually decreased during CP development. H3K27me3 ChIP-seq showed that increased Atp8b1 promoter methylation led to transcriptional inhibition. Atp8b1 complementation substantially increased the LPC concentration and improved CP outcomes. Bhlha15 was identified as a transcription factor that binds to the Atp8b1 promoter and regulates phospholipid metabolism. Our study indicates that the acinar Atp8b1/LPC pathway acts as an important “find-me” signal for macrophages and plays a protective role in CP, with Atp8b1 transcription promoted by the acinar cell-specific transcription factor Bhlha15. Bhlha15, Atp8b1, and LPC could be clinically translated into valuable therapeutic targets to overcome the limitations of current CP therapies.

Objective To evaluate the protective effect and inflammation inhibition mechanism of icariin (ICA) on lung injury in rat model with severe acute pancreatitis (SAP). Methods The rats were randomly divided into sham operation group (sham group), model group (SAP group, 5% sodium taurocholate was retrograde pumped into bile duct according to 0.1 mL/kg through micropump) and experimental group (ICA group, icariin 80 mg/kg intervention was given 2 hours before modeling). There were 6 rats in each group. Twenty-four hours after the establishment of the model, the inferior vena cava blood, pancreatic tissue and lung tissue were collected. The pathological injury of pancreas and lung was observed by HE staining; The activity of amylase, serum IL-6, IL-1β and TNF-α were measured by ELISA; The expression of inflammatory factors IL-6, IL-1β and TNF-α in lung tissue was determined by immunohistochemical staining; And the ratio of phosphorylated JNK/JNK to phosphorylated NF-κB/NF-κB in lung tissue was measured by Western blot. Results The pathological scores of pancreas and lung in SAP group were significantly higher than those in sham group; And the pathological scores of pancreas and lung in ICA group were significantly lower than those in SAP group. Serum amylase activity, serum IL-6, IL-1β and TNF-α content and the expression of IL-6, IL-1β and TNF-α in lung tissue in SAP group were significantly higher than those in sham group; While serum amylase activity, serum IL-6, IL-1β and TNF-α content and the expression of IL-6, IL-1β and TNF-α in lung tissue in ICA group were significantly lower than those in SAP group. The level of phosphorylated JNK and phosphorylated NF-κB in lung tissue of SAP group were significantly higher than those of sham group; While icariin might significantly reduce the level of phosphorylated JNK and phosphorylated NF-κB in lung tissue. Conclusions ICA ameliorates lung injury in SAP rat model by inhibiting acute inflammation response,which is potentially mediated by JNK/NF-κB signal pathway.

The genus Leptobrachella is a species-rich genus of megophrid frog. Rapid discovery of many new species within this genus emphasizes the importance of regional research. In this study, we describe a new species of Leptobrachella, Leptobrachella verrucosa sp. nov., from northwestern Guangdong Province, China, based on a combination of molecular and morphological data. A key including congeners from the same province, namely L. laui, L. liui, L. mangshanensis, L. shimentaina, and L. yunkaiensis, is provided.

Alkali-treated titanium and its alloys (with or without heat treatment) have been paid more attention in the biomaterial field for the formation of a nanoporous layer that can induce apatite deposition and improve bone-implant integration. In this work, a hierarchical micro-nano structure was fabricated by alkali treatment and subsequent induction heating at different temperatures. The surface morphology, phase composition, microstructure, and elemental binding state were investigated using scanning electron microscopy (SEM), X-ray diffractometer (XRD), transmission electron microscopy (TEM), and X-ray photoelectron spectroscopy (XPS), respectively. As the induction heating temperature increases, the porous structure of the outmost sodium titanate layer is maintained with increased anatase TiO2 and decreased wetting angle from 21.8° to 11.8°. At a relatively high temperature (700 °C), the composite layer is composed of a porous sodium titanate layer with a thickness of around 1 μm and a titanium oxide layer of about 2 μm thickness with nanopores. Importantly, the amorphous structure with the oxygen vacancy that exists on the sample surface is conducive to apatite deposition. The assessment of the in-vitro bioactivity performed in 1.5 simulated body fluid shows that the ability to induce apatite deposition increases with the increasing induction heating temperature from 500 to 700 °C. These results are helpful to understand the microstructure of the alkali and induction heating treated titanium that may be responsible for its bioactivity.

The Dalong tin deposit, located in the Luziyuan polymetallic ore field of the southern Baoshan block, Southwest China, is composed of lenticular and bedded orebodies hosted in a Cambrian slate-marble sequence. As the only tin deposit in the orefield, the genesis of the Dalong deposit is of great significance to the understanding of regional metallogenic regularity. Based on a systematic field survey and petrographic observation, EPMA and LA-ICP-MS major/trace element analyses of cassiterite were carried out in this study. The enrichment of Fe, W, and Ti in cassiterite, as well as the depletion of Nb, Ta, and Mn, reveal cassiterite’s features in granitic magmatic-hydrothermal deposits. The Dalong deposit can be categorized as a magmatic-hydrothermal vein-type deposit because of its sulfur isotopic properties, and the main sources of ore-forming fluid and minerals are thought to be buried intermediate-acid intrusions in the mining area’s depths.

Abstract Superconductivity remains one of most fascinating quantum phenomena existing on a macroscopic scale. Its rich phenomenology is usually described by the Ginzburg–Landau (GL) theory in terms of the order parameter, representing the macroscopic wave function of the superconducting condensate. The GL theory addresses one of the prime superconducting properties, screening of the electromagnetic field because it becomes massive within a superconductor, the famous Anderson–Higgs mechanism. Here the authors describe another widely‐spread type of superconductivity where the Anderson–Higgs mechanism does not work and must be replaced by the Deser–Jackiw–Templeton topological mass generation and, correspondingly, the GL effective field theory must be replaced by an effective topological gauge theory. These superconductors are inherently inhomogeneous granular superconductors, where electronic granularity is either fundamental or emerging. It is shown that the corresponding superconducting transition is a 3D generalization of the 2D Berezinskii–Kosterlitz–Thouless vortex binding–unbinding transition. The binding–unbinding of the line‐like vortices in 3D results in the Vogel‐Fulcher‐Tamman scaling of the resistance near the superconducting transition. The authors report experimental data fully confirming the VFT behavior of the resistance.

ObjectiveThis study aimed to investigate the feasibility of Transcranial Doppler Ultrasonography (TCD) in evaluating neonatal hypoxic-ischemic encephalopathy (NHIE) modeling through monitoring the alteration of cerebrovascular flow in neonatal hypoxic-ischemic (HI) rats.MethodsPostnatal 7-day-old Sprague Dawley (SD) rats were divided into the control group, HI group, and hypoxia (H) group. TCD was applied to assess the changes of cerebral blood vessels, cerebrovascular flow velocity, and heart rate (HR) in sagittal and coronal sections at 1, 2, 3, and 7 days after the operation. For accuracy, cerebral infarct of rats was examined by 2,3,5-Triphenyl tetrazolium chloride (TTC) staining and Nissl staining to simultaneously verify the establishment of NHIE modeling.ResultsCoronal and sagittal TCD scans revealed obvious alteration of cerebrovascular flow in main cerebral vessels. Obvious cerebrovascular back-flow was observed in anterior cerebral artery (ACA), basilar artery (BA), middle cerebral artery (MCA) of HI rats, along with accelerated cerebrovascular flows in the left internal carotid artery (ICA-L) and BA, decreased flows in right internal carotid artery (ICA-R) relative to those in the H and control groups. The alterations of cerebral blood flows in neonatal HI rats indicated successful ligation of right common carotid artery. Besides, TTC staining further validated the cerebral infarct was indeed caused due to ligation-induced insufficient blood supply. Damage to nervous tissues was also revealed by Nissl staining.ConclusionCerebral blood flow assessment by TCD in neonatal HI rats contributed to cerebrovascular abnormalities observed in a real-time and non-invasive way. The present study elicits the potentials to utilize TCD as an effective means for monitoring the progression of injury as well as NHIE modeling. The abnormal appearance of cerebral blood flow is also beneficial to the early warning and effective detection in clinical practice.

Managing acute innominate artery (IA) dissection associated with severe stenosis is challenging due to its rarity, possible complex dissection patterns, and compromised blood flow to the brain and upper extremities. This report describes our treatment strategy for this challenging disease using the kissing stent technique. A 61-year-old man had worsening of an acute IA dissection secondary to an extension of a treated aortic dissection. Four possible treatment strategies for kissing stent placement were proposed based on different approaches (open surgical or endovascular) and accesses (trans-femoral, trans-brachial, or trans-carotid access). We chose to place two stents simultaneously via a percutaneous retrograde endovascular approach through the right brachial artery and a combined open surgical distal clamping of the common carotid artery with a retrograde endovascular approach through the carotid artery. This hybrid approach strategy highlights the three key points for maintaining safety and efficacy: (1) good guiding catheter support is obtainable through retrograde, rather than antegrade, access to the lesion, (2) concomitant cerebral and upper extremity reperfusion is guaranteed by placing kissing stents into the IA, and (3) peri-procedural cerebral emboli are prevented by surgical exposure of the common carotid artery with distal clamping.

This study aimed to clarify the effects of two processed forms of American ginseng (Panax quinquefolius L.) on immunosuppression caused by cyclophosphamide (CTX) in mice. In the CTX-induced immunosuppressive model, mice were given either steamed American ginseng (American ginseng red, AGR) or raw American ginseng (American ginseng soft branch, AGS) by intragastric administration. Serum and spleen tissues were collected, and the pathological changes in mice spleens were observed by conventional HE staining. The expression levels of cytokines were detected by ELISA, and the apoptosis of splenic cells was determined by western blotting. The results showed that AGR and AGS could relieve CTX-induced immunosuppression through the enhanced immune organ index, improved cell-mediated immune response, increased serum levels of cytokines (TNF-α, IFN-γ, and IL-2) and immunoglobulins (IgG, IgA, and IgM), as well as macrophage activities including carbon clearance and phagocytic index. AGR and AGS downregulated the expression of BAX and elevated the expression of Bcl-2, p-P38, p-JNK, and p-ERK in the spleens of CTX-injected animals. Compared to AGS, AGR significantly improved the number of CD4+CD8-T lymphocytes, the spleen index, and serum levels of IgA, IgG, TNF-α, and IFN-γ. The expression of the ERK/MAPK pathway was markedly increased. These findings support the hypothesis that AGR and AGS are effective immunomodulatory agents capable of preventing immune system hypofunction. Future research may investigate the exact mechanism to rule out any unforeseen effects of AGR and AGS.

Fingerprint orientation field (OF) estimation is important for basic fingerprint image processing and impacts the accuracy of fingerprint image enhancements, such as Gabor filters. In this article, we introduce an OF estimation algorithm based on differential values of grayscale intensity and examine the accuracy and reliability of the proposed algorithm by applying it to fingerprint images processed using Gaussian blurring and the Gaussian white noise process. The experimental results indicate that the OF estimation reliability of the proposed algorithm is higher than the gradient-based method and the power spectral density (PSD) based method in low quality fingerprints. The proposed algorithm is especially useful in noisy fingerprint images, where the OF estimation reliability of the algorithm is 6.46% and 32.93% higher than the gradient-based method and the PSD-based method, respectively.

Abstract The characteristics of global prevalence and high recurrence of bladder cancer has led numerous efforts to develop new treatments. The spontaneous voiding and degradation of the chemodrug hamper the efficacy and effectiveness of intravesical chemotherapy following tumor resection. Herein, the externally thiolated hollow mesoporous silica nanoparticles (MSN‐SH(E)) is fabricated to serve as a platform for improved bladder intravesical therapy. Enhanced mucoadhesive effect of the thiolated nanovector is confirmed with porcine bladder. The permeation‐enhancing effect is also verified, and a fragmented distribution pattern of a tight junction protein, claudin‐4, indicates the opening of tight junction. Moreover, MSN‐SH(E)‐associated reprogramming of M2 macrophages to M1‐like phenotype is observed in vitro. The antitumor activity of the mitomycin C (MMC)‐loaded nanovector (MMC@MSN‐SH(E)) is more effective than that of MMC alone in both in vitro and in vivo. In addition, IHC staining is used to analyze IFN‐γ, TGF‐β1, and TNF‐α. These observations substantiated the significance of MMC@MSN‐SH(E) in promoting anticancer activity, holding the great potential for being used in intravesical therapy for non‐muscle invasive bladder cancer (NMIBC) due to its mucoadhesivity, enhanced permeation, immunomodulation, and prolonged and very efficient drug exposure.

This study investigates the impact of various zinc supplementation methods on anemia in rats induced by phenylhydrazine (PHZ) and in 5/6-nephrectomized anemic rats. We compare oral zinc sulfate (ZnSO4) supplementation, oyster Crassostrea gigas supplementation, and hard clam Meretrix lusoria supplementation on red blood cell (RBC) levels. Oral zinc-rich oyster supplementation (2.70 mg Zn (30 g oyster)/day/rat) effectively corrects anemia in both experimental groups. Rats orally fed oysters for four days exhibit similar effectiveness as those receiving a single ZnSO4 injection (0.95 mg Zn (4.18 mg ZnSO4⋅7H2O)/rat). In contrast, oral ZnSO4 supplementation (2.70 mg Zn (11.88 mg ZnSO4⋅7H2O)/day/rat) does not significantly increase RBC levels, suggesting better zinc absorption from oysters. A placebo group of anemic rats supplemented with hard clams, similar in composition to oysters but much lower in zinc, did not change RBC counts. This supports oysters’ high zinc content as the key to correcting anemia. Oysters also contain high iron levels, offering a potential solution for iron-deficiency anemia while supporting bone marrow erythropoiesis. In summary, oral oyster supplementation emerges as an effective strategy to correct anemia in rats with added zinc and iron support for erythropoiesis.

There are multiple routes to prepare semi-solid slurries with a globular microstructure for semi-solid forming. The variations in the microstructure of semi-solid slurries prepared using different routes may lead to significant differences in the flow behavior and mechanical properties of rheo-diecasting parts. Therefore, it is crucial to have a comprehensive understanding of the microstructure evolution associated with different slurry preparation routes and their resulting effects. In this study, the gas-induced semi-solid process (GISS) and the swirl enthalpy equilibrium device (SEED) routes were employed to prepare semi-solid Al-Si-Mg slurries for their simplicity and productivity in potential industrial applications. The prepared slurries were then injected into the shoot sleeves of a high-pressure die casting (HPDC) machine to produce tensile test bars. Subsequently, the bars underwent T6 treatment to enhance their mechanical properties. The microstructure, segregation, and mechanical properties of the samples were investigated and compared with those of conventional HPDC. The results indicated that the GISS and SEED can produce semi-solid slurries containing a spherical α-Al primary phase, as opposed to the dendritic structure commonly found in conventional castings. The liquid fraction had a significant effect on the flow behavior, resulting in variations in liquid segregation and mechanical properties. It was observed that a higher solid fraction (>75%) had a suppressing effect on surface liquid segregation. In addition, the tendency for liquid segregation gradually increased along the filling direction due to the special flow behavior of the semi-solid slurry with a low solid fraction. Furthermore, under the same die-casting process parameters, the conventional HPDC samples exhibit higher yield stress (139 ± 3 MPa) compared to SEED-HPDC and GISS-HPDC samples, which may be attributed to the small grain size and the distribution of eutectic phases. After undergoing the T6 treatment, both SEED-HPDC and GISS-HPDC samples showed a significant improvement in yield and tensile strength. These improvements are a result of solution and precipitation strengthening effects as well as the spheroidization of the eutectic Si phase. Moreover, the heat-treated SEED-HPDC samples demonstrate higher ultimate strength (336 ± 5 MPa) and elongation (13.7 ± 0.3%) in comparison to the GISS-HPDC samples (307 ± 4 MPa, 8.8 ± 0.2%) after heat treatment, mainly due to their low porosity density. These findings suggest that both GISS-HPDC and SEED-HPDC processes can be utilized to produce parts with favorable mechanical properties by implementing appropriate heat treatments. However, further investigation is required to control the porosities of GISS-HPDC samples during heat treatment.

Morphometric disparities were employed to characterize the geographical variations in and elucidate the phylogeography of Portunus sanguinolentus in East Asia. In this study, a comprehensive assessment was undertaken based on measurements from 833 individual crabs. These crabs were sampled from eight distinct locations, namely Kyushu (KS), Fuzhou (FZ), Yilan (YL), Xiamen (XM), Tainan (TN), Hong Kong (HK), Vietnam (VN), and Singapore (SG). Twelve morphometric measurements were standardized based on size using the allometric method. A cluster analysis and a canonical variate analysis (CVA) were then conducted for a data analysis. Randomization tests were subsequently performed to validate the morphometric differences observed among the groups. The analytical results classified the eight sampled populations into at least two distinct clusters. The first cluster included three samples from China (specifically FZ, XM, and HK), while the second cluster consisted of samples from East Asia (including KS, YL, TN, VN, and SG). The CVA revealed a tripartite division into three groups: the mainland China group (FZ, XM, and HK), the Kuroshio group (KS, YL, and TN), and the Southeast Asia group (VN and SG). The observed morphometric variations between these broad groups were statistically significant for both genders. These differences may be attributed to evolutionary origins, geographical events, or environmental adaptations. This study highlights the effectiveness of morphometric techniques in providing comprehensive insights into species delimitation and philogeographic relationships.

This study aims to develop a refractive-index sensor operating in the visible region using an all-dielectric metasurface, which was chosen for its advantages of low optical loss and narrow spectral bandwidth, compared to those of conventional metallic metasurfaces. COMSOL software was utilized as a calculation tool to simulate the resonant properties of an all-dielectric metasurface composed of a circular nanohole-structured titanium oxide (TiO2) thin film, with the aim of enhancing the sensitivity of the refractive index for sensing targets. The simulation focused on finding the best geometrical conditions for the all-dielectric metasurface to achieve high sensitivity. Two resonance modes observed in this metasurface were considered: the quasi-bound-state-in-the-continuum (qBIC) mode and the perfect-reflection (PR) mode. The simulated results demonstrated that high sensitivities of 257 nm/RIU at the PR mode and 94 nm/RIU at the qBIC mode in the visible spectral range could be obtained by periodically constructing the metasurface with a unit cell having a lattice constant of 350 nm, a nanohole radius of 160 nm, and a nanohole depth of 250 nm. Furthermore, the study showed that the resonance mode that enabled high sensitivity was the PR mode, with a sensitivity nearly three times larger than that of the qBIC mode and the ability to reach the highest reflectance at the resonance wavelength. The optimized feature had the highest reflectance at a resonant wavelength of 570.19 nm, and although the quality factor was 25.50, these designed parameters were considered sufficient for developing a refractive index biosensor with high sensitivity and optical efficiency when operating in the visible spectral range.

This study evaluated and compared the functional recovery and histopathological outcomes of treatment involving low-intensity pulsed ultrasound (LIPUS) and methylcobalamin (B12) on brachial plexus injury (BPI) in an experimental rat model. Three days after BPI, the rats were assigned to receive either LIPUS or methylcobalamin alone or in combination consecutively for 12 days. Serial changes in sensory and motor behavioral responses, as well as morphological and immunohistochemical changes for substance P (SP), ionized calcium-binding adapter molecule 1 (iba1), brain-derived neurotrophic factor (BDNF), and S100 were examined 28 days after BPI as the outcome measurements. Early intervention of LIPUS and methylcobalamin, whether alone or in combination, augmented the sensory and motor behavioral recovery as well as modulated SP and iba1 expression in spinal dorsal horns, BDNF, and S100 in the injured nerve. Moreover, the combined therapy with its synergistic effect gave the most beneficial effect in accelerating functional recovery. In view of the effective initiation of early recovery of sensory and motor functions, treatment with LIPUS and methylcobalamin in combination has a potential role in the clinical management of early-phase BPI.

Zinc (Zn) and its alloys are considered as promising biodegradable metals due to its suitable degradability and important physiological functions. However, in the process of degradation, excessive release of Zn ions (Zn2+) will significantly affect the cytocompatibility and antibacterial properties. Surface modification technology can not only control the corrosion behavior of biodegradable metals but also improve its biocompatibility and antibacterial properties according to clinical requirements. In this study, an in-situ hopeite (HP, Zn3(PO4)2·4H2O) coating was prepared on Zn alloy by phosphate chemical conversion (PCC) method using Zn2+ provided by self-corrosion of the substrates. And the effect of phosphating time on the microstructure, phase composition, corrosion resistance, and wettability of HP coatings was investigated. The results showed that a complete HP coating was formed on the Zn alloy after reaction for 1 min, even if Zn2+ were supplied by self-corrosion without an additional Zn source. It also revealed that the reaction time had a marked impact on the morphologies and microstructures of the HP coating. The electrochemical test results proved that the prepared HP coatings had obvious protection on the Zn alloy and could regulate the corrosion rate of the substrate. Meanwhile, the Zn alloy modified by the PCC method also possessed excellent hydrophilicity. These primary findings might support new opportunities in the exploration of controllable coatings on biomedical Zn alloys.

Abstract To inhibit the agglomeration of nanotitanium dioxide, a poly (vinyl chloride) (PVC) composite film doped with folic acid-modified titanium dioxide was synthesized and characterized using X-ray powder diffraction, X-ray photoelectron spectroscopy and thermogravimetric analysis coupled with Fourier transform infrared spectroscopy. The average grain size of the folic acid-modified titanium dioxide was found to decrease by 1.3 nm, indicating that the cohesiveness of the nanoparticles is decreased. The lowest temperature for 1.0% thermal decomposition of PVC was determined to be 230.0 °C. The decomposition rate at the peak temperature is found to be 39.6% lower than that of a control sample. The stability of the PVC is improved due to a lower number of surface chlorine atoms as well intermolecular attraction. A mechanism for folic acid modification of titanium dioxide-doped PVC is proposed. After doping, the ester groups in the plasticizer show a significant decrease in the vibration peak intensities observed at 1264 cm−1, 1736 cm−1 and 1106 cm−1. The doped PVC film suppresses the release of CO2, and the strongest vibration peak at 1264 cm−1 is found to be 17.2% lower than that for the blank sample, indicating that doping is beneficial for plasticizer recovery.

Core-shell microspheres have attracted widespread attention in the field of drug delivery due to their special morphology and excellent surface properties. However, the preparation of the core–shell microspheres composed of hydroxyapatite (HA) have been rarely reported. In this paper, porous HA core–shell microspheres were prepared through a one-step hydrothermal method by using sodium trimetaphosphate (STMP) as the phosphorus source. SEM and TEM observations showed that the prepared porous HA core–shell microspheres were composed of a porous shell and a solid core, with a gap between the core and the shell. These microspheres bear uniform size and identical structure. With the increase of the hydrothermal time, the solid microspheres transformed to porous core–shell microspheres with the simultaneous transformation of their phase from amorphous calcium phosphate (ACP) to HA. The possible formation mechanism is proposed, providing inspiration for the design of porous HA core–shell microspheres and their potential biological applications.

Byzantine agreement, the underlying core of blockchain, aims to make every node in a decentralized network reach consensus. Classical Byzantine agreements unavoidably face two major problems. One is 1/3 fault-tolerance bound, which means that the system to tolerate f malicious players requires at least 3f + 1 players. The other is the security loopholes from its classical cryptography methods. Here, we propose a Byzantine agreement framework with unconditional security to break this bound with nearly 1/2 fault tolerance due to multiparty correlation provided by quantum digital signatures. It is intriguing that quantum entanglement is not necessary to break the 1/3 fault-tolerance bound, and we show that weaker correlation, such as asymmetric relationship of quantum digital signature, can also work. Our work strictly obeys two Byzantine conditions and can be extended to any number of players without requirements for multiparticle entanglement. We experimentally demonstrate three-party and five-party consensus for a digital ledger. Our work indicates the quantum advantage in terms of consensus problems and suggests an important avenue for quantum blockchain and quantum consensus networks.

Roseomonas mucosa (R. mucosa) is a pink-pigmented, aerobic, nonfermentative, slow-growing Gram-negative coccus typically isolated from the natural environment, human skin, and hospital environment. This pathogen, in most circumstances, leads to infections in immunocompromised hosts, but it may sometimes invade immunocompetent individuals. Bacteraemia is the most common form of infection caused by R. mucosa. In contrast, only two case reports have described R. mucosa-related epidural abscess formation and infective spondylitis. In this case report, we shared the history and treatment experience of a 76-year-old female who was diagnosed with infective spondylitis and epidural abscess caused by R. mucosa. She received a local transdermal injection into the lower back to relieve her back pain two months before symptom onset, which was considered to be associated with this infection episode. After admission to the hospital, neurosurgeons performed emergent decompression and debridement. She was treated with intravenous ceftriaxone for four weeks, followed by oral ciprofloxacin for another eight weeks. The patient recovered well without any sequelae and had no relapse of infection at least six months after the end of treatment. In addition to the case report, we reviewed the literature for reported cases caused by R. mucosa. Our experience suggests that clinicians should include R. mucosa as one of the possible healthcare-associated pathogens among individuals who have undergone transdermal procedures. We believe that this article will help clinicians better recognize R. mucosa infection.

ABSTRACT The cytotoxic granules of human NK and CD8 T cells contain the effector molecule granulysin. Although in vitro studies indicate that granulysin is bactericidal to Mycobacterium tuberculosis and human CD8 T cells restrict intracellular M. tuberculosis by granule exocytosis, the role of granulysin in cell-mediated immunity against infection is incompletely understood, in part because a granulysin gene ortholog is absent in mice. Transgenic mice that express human granulysin (GNLY-Tg) under the control of human regulatory DNA sequences permit the study of granulysin in vivo. We assessed whether granulysin expression by murine CD8 T cells enhances their control of M. tuberculosis infection. GNLY-Tg mice did not control pulmonary M. tuberculosis infection better than non-Tg control mice, and purified GNLY-Tg and non-Tg CD8 T cells had a similar ability to transfer protection to T cell deficient mice. Lung CD8 T cells from infected control and GNLY-transgenic mice similarly controlled intracellular M. tuberculosis growth in macrophages in vitro. Importantly, after M. tuberculosis infection of GNLY-Tg mice, granulysin was detected in NK cells but not in CD8 T cells. Only after prolonged in vitro stimulation could granulysin expression be detected in antigen-specific CD8 T cells. GNLY-Tg mice are an imperfect model to determine whether granulysin expression by CD8 T cells enhances immunity against M. tuberculosis. Better models expressing granulysin are needed to explore the role of this antimicrobial effector molecule in vivo. IMPORTANCE Human CD8 T cells express the antimicrobial peptide granulysin in their cytotoxic granules, and in vitro analysis suggest that it restricts growth of Mycobacterium tuberculosis and other intracellular pathogens. The murine model of tuberculosis cannot assess granulysin’s role in vivo, as rodents lack the granulysin gene. A long-held hypothesis is that murine CD8 T cells inefficiently control M. tuberculosis infection because they lack granulysin. We used human granulysin transgenic (GNLY-Tg) mice to test this hypothesis. GNLY-Tg mice did not differ in their susceptibility to tuberculosis. However, granulysin expression by pulmonary CD8 T cells could not be detected after M. tuberculosis infection. As the pattern of granulysin expression in human CD8 T cells and GNLY-Tg mice seem to differ, GNLY-Tg mice are an imperfect model to study the role of granulysin. An improved model is needed to answer the importance of granulysin expression by CD8 T cells in different diseases.

Objective: Decision aids have been developed to help prospective parents make informed, shared decisions about medical tests, but these options are rapidly changing. This study aimed to identify and evaluate publicly available decision aids written in English for prospective parents seeking prenatal test information. Methods: A systematic review process was followed using 3 sources: known decision aid repositories, fetal medicine organisations and Google. The search, screening process, quality assessment, and data extraction was performed by two independent researchers. The quality assessment of the decision aids was based on the International Patient Decision Aids Standards (IPDAS v.4.0). Results: We identified 13 decision aids, which varied in the screening and diagnostic tests that they discussed. No decision aid met all the IPDAS v.4.0. criteria and no decision aid reported updated risk of miscarriage for amniocentesis and chorionic villus sampling (CVS). There was a lack of decision aids for some common decisions in the prenatal context. Conclusion: We identified outdated content in current prenatal decision aids. The findings will inform healthcare professionals of the quality of current prenatal decision aids, which may facilitate their patients’ informed decision-making about prenatal tests. Innovation: Considerations for improving future decision aids are outlined.

Background: Maternal Toxoplasma gondii (T. gondii) infection during pregnancy has been associated with various mental illnesses in the offspring. Ginsenoside Rh2 (GRh2) is a major bioactive compound obtained from ginseng that has an anti-T. gondii effect and attenuates microglial activation through toll-like receptor 4 (TLR4)/nuclear factor-kappa B (NF-κB) signaling pathway. GRh2 also alleviated tumor-associated or lipopolysaccharide-induced depression. However, the effects and potential mechanisms of GRh2 on depression-like behavior in mouse offspring caused by maternal T. gondii infection during pregnancy have not been investigated. Methods: We examined GRh2 effects on the depression-like behavior in mouse offspring, caused by maternal T. gondii infection during pregnancy, by measuring depression-like behaviors and assaying parameters at the neuronal and molecular level. Results: We showed that GRh2 significantly improved behavioral measures: sucrose consumption, forced swim time and tail suspended immobility time of their offspring. These corresponded with increased tissue concentrations of 5-hydroxytryptamine and dopamine, and attenuated indoleamine 2,3-dioxygenase or enhanced tyrosine hydroxylase expression in the prefrontal cortex. GRh2 ameliorated neuronal damage in the prefrontal cortex. Molecular docking results revealed that GRh2 binds strongly to both TLR4 and high mobility group box 1 (HMGB1). Conclusion: This study demonstrated that GRh2 ameliorated the depression-like behavior in mouse offspring of maternal T. gondii infection during pregnancy by attenuating the excessive activation of microglia and neuroinflammation through the HMGB1/TLR4/NF-κB signaling pathway. It suggests that GRh2 could be considered a potential therapy in preventing and treating psychiatric disorders in the offspring mice of mothers with prenatal exposure to T. gondii infection.

Abstract Background Spinal dural arteriovenous fistula (SDAVF) usually occurs during the 4th to 6th decades of life, and adolescent SDAVF is rarely reported. SDAVF arising around a tumor is also rare, and reported tumors are mostly schwannoma and lipoma. Case presentation We reported a 16-year-old male presented with progressive weakness and numbness of lower limbs for 3 months. A SDAVF was found, which was fed by right radicular arteries from segmental artery at L2 level and drained retrogradely into perimedullary veins. A concomitant spinal extradural nodular fasciitis at right L1/L2 intervertebral foramen was also noted. The SDAVF was completely obliterated by endovascular treatment and the tumor was debulked. The patient recovered well after the procedures. Conclusions Our case report suggests SDAVF can occur in adolescent. The concomitant presence with a nodular fasciitis indicates that although it usually arises in subcutaneous tissue but can rarely form on the dura of spine.

Myeloperoxidase is an important inflammatory factor in the myeloid system, primarily expressed in neutrophils and microglia. Myeloperoxidase and its active products participate in the occurrence and development of hemorrhagic and ischemic stroke, including damage to the blood-brain barrier and brain. As a specific inflammatory marker, myeloperoxidase can be used in the evaluation of vascular disease occurrence and development in stroke, and a large amount of experimental and clinical data has indicated that the inhibition or lack of myeloperoxidase has positive impacts on stroke prognosis. Many studies have also shown that there is a correlation between the overexpression of myeloperoxidase and the risk of stroke. The occurrence of stroke not only refers to the first occurrence but also includes recurrence. Therefore, myeloperoxidase is significant for the clinical evaluation and prognosis of stroke. This paper reviews the potential role played by myeloperoxidase in the development of vascular injury and secondary brain injury after stroke and explores the effects of inhibiting myeloperoxidase on stroke prognosis. This paper also analyzes the significance of myeloperoxidase etiology in the occurrence and development of stroke and discusses whether myeloperoxidase can be used as a target for the treatment and prediction of stroke.

Graphene and graphene-based materials have the ability to induce stem cells to differentiate into neurons, which is necessary to overcome the current problems faced in the clinical treatment of spinal cord injury. This review summarizes the advantages of graphene and graphene-based materials (in particular, composite materials) in axonal repair after spinal cord injury. These materials have good histocompatibility, and mechanical and adsorption properties that can be targeted to improve the environment of axonal regeneration. They also have good conductivity, which allows them to make full use of electrical nerve signal stimulation in spinal cord tissue to promote axonal regeneration. Furthermore, they can be used as carriers of seed cells, trophic factors, and drugs in nerve tissue engineering scaffolds to provide a basis for constructing a local microenvironment after spinal cord injury. However, to achieve clinical adoption of graphene and graphene-based materials for the repair of spinal cord injury, further research is needed to reduce their toxicity.

Abstract To investigate the value of the star-VIBE sequence in dynamic contrast-enhanced magnetic resonance imaging of esophageal carcinoma under free breathing conditions. From February 2019 to June 2020, 60 patients with esophageal carcinoma were prospectively enrolled to undergo dynamic contrast-enhanced magnetic resonance imaging (DCE-MRI) with the K-space golden-angle radial stack-of-star acquisition scheme (star-VIBE) sequence (Group A) or conventional 3D volumetric-interpolated breath-hold examination (3D-VIBE) sequence (Group B), completely randomized grouping. The image quality of DCE-MRI was subjectively evaluated at five levels and objectively evaluated according to the image signal-to-noise ratio (SNR) and contrast-noise ratio (CNR). The DCE-MRI parameters of volume transfer constant (Ktrans), rate constant (Kep) and vascular extracellular volume fraction (Ve) were calculated using the standard Tofts double-compartment model in the post-perfusion treatment software TISSUE 4D (Siemens). Each group included 30 randomly selected cases. There was a significant difference in subjective classification between the groups (35.90 vs 25.10, p = 0.009). The study showed that both the SNR and CNR of group A were significantly higher than those of group B (p = 0.004 and 0.05). The star-VIBE sequence can be applied in DCE-MRI examination of esophageal carcinoma, which can provide higher image quality than the conventional 3D-VIBE sequence in the free breathing state.

This study aimed to investigate whether the significant policies and new auditing standards have substantial effects on audit effort and audit risk, and if such effects are reflected in audit fees. The observations from 2008 to 2021 were divided into three periods including "filing deadline reduction", "implementation of new auditing standards", and "border controls due to Coronavirus disease 2019 (COVID-19 pandemic)". The empirical results indicate that both mandatory policies and the adoption of new auditing standards, as well as the significant changes brought about by the COVID-19 pandemic, have a substantial impact on auditors' audit work and audit risk. As a response to these varying audit risks, auditors adjust their audit fees accordingly. This study validated that even in countries where audit fees are generally at lower level, auditors still reflect audit risk in audit fees. Therefore, audit fees can indeed serve as one of the indicators for assessing audit risk and a firm's risk. [ABSTRACT FROM AUTHOR]
