Local and privileged expression of dendritic proteins allows segregation of distinct functions in a single neuron but may represent one of the underlying mechanisms for early and insidious presentation of sensory neuropathy. Tangible characteristics of early hearing loss (HL) are defined in correlation with nascent hidden hearing loss (HHL) in humans and animal models. Despite the plethora of causes of HL, only two prevailing mechanisms for HHL have been identified, and in both cases, common structural deficits are implicated in inner hair cell synapses, and demyelination of the auditory nerve (AN). We uncovered that Na+-activated K+ (KNa) mRNA and channel proteins are distinctly and locally expressed in dendritic projections of primary ANs and genetic deletion of KNa channels (Kcnt1 and Kcnt2) results in the loss of proper AN synaptic function, characterized as HHL, without structural synaptic alterations. We further demonstrate that the local functional synaptic alterations transition from HHL to increased hearing-threshold, which entails changes in global Ca2+ homeostasis, activation of caspases 3/9, impaired regulation of inositol triphosphate receptor 1 (IP3R1), and apoptosis-mediated neurodegeneration. Thus, the present study demonstrates how local synaptic dysfunction results in an apparent latent pathological phenotype (HHL) and, if undetected, can lead to overt HL. It also highlights, for the first time, that HHL can precede structural synaptic dysfunction and AN demyelination. The stepwise cellular mechanisms from HHL to canonical HL are revealed, providing a platform for intervention to prevent lasting and irreversible age-related hearing loss (ARHL).

Effective selective recycling of spent lithium‐ion batteries (S‐LIBs) and giving recycled products a "second life" are crucial for advancing energy supply circularity, environmental and economic sustainability development. However, separating metal compounds with similar charge differences requires substantial energy, water, and chemical inputs. Herein, an innovative strategy is present for selective recycling S‐LIBs by photoexcitation inspired by the Hard Soft Acid Base (HSAB) principle. Theoretical calculations and experimental results show that photoexcitation drives charge transfer and modulates subtle charge density differences among metal components, thereby enhancing their solubility disparity and facilitating metal separation. Remarkably, the photoexcitation‐induced metal separation factor reaches 46900 and the metal recovery efficiency approaches 100%, representing a significant improvement over non‐photoexcitation separation with a separation factor of non‐photoexcitation of merely 2.7. Through techno‐economic analysis, the viability of photoexcitation selective recycling technology has been confirmed as an eco‐friendly and economical approach for battery recycling. Furthermore, high‐value reuse of recovered Mn components is implemented. The Recycled Mn components are treated by calcination to obtain porous, defect‐rich Mn2O3, which showed a specific capacity of 613 mAh g−1 at 0.1 A g−1) in aqueous Zn‐ion batteries (AZIBs). In order to identify the pathogen and its characteristics in diseased Cuora flavomarginata in Hangzhou, Zhejiang, in June 2020, a dominant strain HZ202006 was isolated from the liver of the diseased turtles. Morphological observations, 16S rRNA sequence analysis, whole-genome sequencing, artificial infection experiments, and antimicrobial sensitivity test were performed on this strain. The results confirmed that the strain is Providencia rettgeri, a short rod-shaped, Gram-negative bacterium. Whole-genome sequencing revealed that the complete genome sequence of this strain consists of a 4.8 Mbp double-stranded DNA and two plasmids. Annotation of the sequenced genes using the Comprehensive Antibiotic Resistance Database (CARD) and the Virulence Factors Database (VFDB) identified 20 resistance genes and four virulence factors in the bacterium. Antimicrobial sensitivity test demonstrated that HZ202006 was moderately sensitive only to amikacin, and resistant to 16 common antibiotics, including penicillin, cephalosporins, and aminoglycosides. This study isolated and identified a strain of P. rettgeri causing disease in C. flavomarginata, providing insights for disease control in aquaculture animals affected by this bacterium. To improve the handling and stability of four-wheel independent drive electric vehicles (FWID EVs), this paper introduces a hierarchical architecture lateral stability control system. The upper-level controller is responsible for generating the additional yaw moment required by the vehicle. This includes a control strategy based on feedforward control and a Linear Quadratic Regulator (LQR) for handling assistance control, an LQR-based stability control, a PID controller-based speed-following control, and a stability assessment method. The lower-level controller uses Quadratic Programming (QP) to optimally distribute the additional yaw moment to the four wheels. A "normalized" method was proposed to determine vehicle stability. After comparing it with the existing double-line method, diamond method, and curved boundary method through the open-loop Sine with Dwell test and the closed-loop Double Lane Change (DLC)test simulation, the results demonstrate that this method is more sensitive and accurate in determining vehicle stability, significantly enhancing vehicle handling and stability. To solve the oxygen dependence problem of photodynamic therapy (PDT), it is critical to explore photosensitizers that do not rely on O2 molecule to generate reactive oxygen species (ROS). Herein, a stable cationic metal‐organic cage [Pd6(RuLoz3)8](BF4)28 (MOC‐88) that possesses high +28 charges is designed. The cage‐confined positive microenvironment enables efficient generation of hydroxyl radicals and improved yield of the singlet oxygen under one‐/two‐photon excitation, showing excellent performance to concurrently enhance Type‐II and O2‐independent‐Type‐I PDT. Moreover, the effective ROS production and robust lipid peroxidation trigger a series of signaling pathways (inflammasome, cyclic guanosine monophosphate–adenosine monophosphate synthase stimulator of interferon genes, and NF‐κB) to evoke PANoptosis and ferroptosis in tumor cells, enabling MOC‐88 to simultaneously cause the loss of cell membrane integrity, release a series of inflammatory cytokines and damage‐associated molecular patterns, stimulate the maturation and antigen presentation ability of dendritic cells, and ultimately activate T‐cell‐dependent adaptive immunity in vivo to inhibit tumor growth. This study focuses on developing an advanced anti-lock braking system (ABS) for motorcycles, specifically targeting the challenges associated with cornering. Significant roll angles during motorcycle turns can often lead to slipping and the loss of control, increasing the risk of accidents. Existing ABSs primarily address longitudinal dynamics and fail to provide optimal braking control during cornering. To address this gap, this study utilizes BikeSim and MATLAB/Simulink for simulations and experiments to design an ABS that adapts to varying roll angles by analyzing motorcycle dynamics during cornering. A tire model is constructed using the Magic Formula to examine both longitudinal and lateral characteristics under different conditions, which helps determine the current tire slip set-point. The controller, designed with a finite-state machine combined with bang-off-bang control, uses tire slip as the control variable. It adjusts the slip set-point based on changes in roll angle and sends control signals to the hydraulic actuator to regulate braking pressure, ensuring optimal braking performance without the loss of control. Finally, hardware-in-the-loop experiments are conducted, with real-time control commands sent to the hardware platform's actuator via BikeSim RT. Alginate oligosaccharides (AOs), derived from alginate degradation, exhibit diverse biological activities and hold significant promise in various fields. The enzymatic preparation of AOs relies on alginate lyases, which offers distinct advantages. In contrast to the conventional use of sodium alginate derived from brown algae as the substrate for the enzymatic preparation of AOs, AO preparation directly from brown algae is more appealing due to its time and energy efficiency. Thus, the identification of potent alginate lyases and cost-effective brown algae substrates is crucial for optimizing AO production. Herein, we identified and characterized an alginate lyase, AlyP18, capable of efficiently decomposing algae, from a marine bacterium Pseudoalteromonas agarivorans A3 based on secretome analysis. AlyP18 is a mesothermal, endo-type and bifunctional alginate lyase with high enzymatic activity. Two brown algae substrates, Laminaria japonica roots and Macrocystis pyrifera, were used for the AO preparation by AlyP18. Upon optimization of AlyP18 hydrolysis parameters, the substrate degradation efficiency and AO production reached 53% and ~32% for L. japonica roots, respectively, and 77% and ~46.5% for M. pyrifera. The generated AOs primarily consisted of dimers to pentamers, with trimers and tetramers being dominant. Small-conductance Ca2+-activated K+ (SK) channels regulate the excitability of cardiomyocytes by integrating intracellular Ca2+ and membrane potentials on a beat-to-beat basis. The inextricable interplay between activation of SK channels and Ca2+ dynamics suggests the pathology of one begets another. Yet, the exact mechanistic underpinning for the activation of cardiac SK channels remains unaddressed. Here, we investigated the intracellular Ca2+ microdomains necessary for SK channel activation. SK currents coupled with Ca2+ influx via L-type Ca2+ channels (LTCCs) continued to be elicited after application of caffeine, ryanodine or thapsigargin to deplete SR Ca2+ store, suggesting that LTCCs provide the immediate Ca2+ microdomain for the activation of SK channels in cardiomyocytes. Super-resolution imaging of SK2, Cav1.2 Ca2+ channel, and ryanodine receptor 2 (RyR2) was performed to quantify the nearest neighbor distances (NND) and localized the three molecules within hundreds of nanometers. The distribution of NND between SK2 and RyR2 as well as SK2 and Cav1.2 was bimodal, suggesting a spatial relationship between the channels. The activation mechanism revealed by our study paved the way for the understanding of the roles of SK channels on the feedback mechanism to regulate the activities of LTCCs and RyR2 to influence local and global Ca2+ signaling.
