103. Triiodothyronine (T3) promotes browning of white adipose through inhibition of the PI3K/AKT signalling pathway

Abstract Obesity arises from an imbalance between energy consumption and energy expenditure, and thyroid hormone levels serve as a determinant of energy expenditure. We conducted experiments at the animal and cellular levels and combined those findings with clinical data to elucidate the role of triiodothyronine (T3) in facilitating the browning of white adipose tissue (WAT) and its underlying mechanism. The results showed (i) the impaired metabolic function of local WAT and the compensatory elevation of systemic thermogenesis in obesity; (ii) T3 treatment of white adipocytes in vitro and local WAT in vivo induced a shift towards a morphologically “brown” phenotype, accompanied by upregulation of mRNA and protein expression of browning-related and mitochondrial function markers, which suggest that T3 intervention promotes the browning of WAT; and (iii) the aforementioned processes could be modulated through inhibition of the PI3K/AKT signalling pathway; however, whether T3 affects the PI3K/AKT signalling pathway by affecting insulin signalling remains to be studied and clarified. The results of our study indicate that T3 treatment promotes browning of WAT through inhibition of the PI3K/AKT signalling pathway; these findings offer novel perspectives regarding the potential of localised therapies for addressing WAT volume in individuals with obesity.

Abstract Chronic obstructive pulmonary disease(COPD) is an inflammatory disease characterized by the progressive and irreversible structural and functional damage of lung tissue. Although COPD is a significant global disease burden, the available treatments only ameliorate the symptoms, but cannot reverse lung damage. Researchers in regenerative medicine have examined the use of stem cell transplantation for treatment of COPD and other diseases because these cells have the potential for unlimited self-renewal and the ability to undergo directed differentiation. Stem cells are typically classified as embryonic stem cells, induced pluripotent stem cells, and adult stem cells (which includes mesenchymal stem cells [MSCs]), each with its own advantages and disadvantages regarding applications in regenerative medicine. Although the heterogeneity and susceptibility to senescence of MSCs make them require careful consideration for clinical applications. However, the low tumourigenicity and minimal ethical concerns of MSCs make them appear to be excellent candidates. This review summarizes the characteristics of various stem cell types and describes their therapeutic potential in the treatment of COPD, with a particular emphasis on MSCs. We aim to facilitate subsequent in-depth research and preclinical applications of MSCs by providing a comprehensive overview.

105. Global, regional, and national burden of hearing loss in children and adolescents, 1990–2021: a systematic analysis from the Global Burden of Disease Study 2021

Abstract Background Hearing loss in children and adolescents is attracting increasing attention as a growing public health problem. This study aimed to analyze the burden of hearing loss in children and adolescents from 1990 to 2021 to provide a new basis for further epidemiological research, disease prevention, and public health policy development. Methods The prevalence and years lived with disability (YLDs) of hearing loss in children and adolescents from 1990 to 2021 at the global, regional, and national levels were extracted from the Global Burden of Disease 2021 database. The estimated annual percentage change (EAPC) was used to assess trends in prevalence and YLDs. Pearson correlation analysis was used to assess the relationships between sociodemographic index (SDI) and prevalence and YLDs. Results In 2021, approximately 97.83 million children and adolescents under the age of 20 years were affected by hearing loss globally, resulting in approximately 3.91 million YLDs. From 1990 to 2021, the prevalence rate increased from 3537 per 100,000 to 3711 per 100,000, with an EAPC of 0.15% (95% CI: 0.12–0.17). The burden of hearing loss was greater in low-middle-SDI region and low-SDI region. Mild hearing loss accounted for 62.1% of the total cases and was the predominant type of hearing loss. Otitis media was the leading preventable cause of hearing loss, with 46.9% of hearing loss attributed to otitis media. Moreover, in children under 5 years of age, 68.7% of hearing loss was attributed to otitis media. Background: This study determined the knowledge and attitudes regarding COVID-19 and assessed the acceptance of the COVID-19 vaccine among the Chinese population. Methods: An online and offline cross-sectional study was conducted from 1 to 18 June 2021 among the Chinese population. Demographic characteristics, attitudes, knowledge, values, impact, and autonomy regarding the COVID-19 vaccine were collected using questionnaire. The variables in our study were analyzed by Mann-Whitney test and chi-square test. Results: A total of 93.8% participants were willing to be vaccinated, 2.7% refused, and 3.5% hesitated. In regards to knowledge about the COVID-19 vaccine, 94.3% citizens surveyed knew about the spread of droplets and 65% had knowledge about surfaces touched by an infected person. In addition, 93.8% of participants had knowledge of the common symptoms related to COVID-19, such as fever and cough (93.8%), shortness of breath/anorexia/fatigue/nausea/vomiting/diarrhea (80.2%), and panic and chest tightness (69.4%). Most participants had a strong self-prevention awareness, such as washing hands regularly (92.1%) and wearing a facemask (94.1%). Besides, over ninety percent of respondents would report exposure to SARS-CoV-2 (96.6%) and exposure to symptoms possibility related to COVID-19 (92.9%). If necessary, most respondents would agree to isolate at home (93.5%) or an isolation in hospital (96.3%). Knowledge of COVID-19, including transmission, symptoms, protective measures, and vaccines itself, is associated with vaccination attitude. Values, perceived impacts, knowledge, and autonomy, assessed by the scale of DrVac-COVID19S, have also been revealed as important determinants to vaccine acceptance. Conclusions: Almost 93% of Chinese people surveyed in this study showed a willing attitude toward COVID-19 vaccination. Based on the above results, government and social workers can take measures from these perspectives to improve the vaccination attitude, so as to increase vaccine immunization rates.

Pulmonary manifestations of systemic lupus erythematosus (SLE) are appearing in 4-5% of patients involving lung in almost half of the cases during the disease course.We compared the autoimmune pulmonary inflammation in the lung tissue of mice to determine the association between decreased expression levels of Forkhead Box J1 (FOXJ1) and the activation of the NF-κB pathway in autoimmune pulmonary inflammation of MRL/Lpr mice.The female BALB/c mice (n=6) and MRL/Lpr mice (n=30) were divided into 5 groups including a control group (BALB/c), and five MRL/Lpr mice groups (8W, 12W, 16W, 24W, and 32W). The infiltration of the inflammatory cells was determined in lung tissue by performing histological analysis. The western blotting was used to examine the expression levels of the age-related FOXJ1, and p50 and p65 proteins in the lungs of MRL/Lpr mice. The expression levels of MMP2 and MMP9 were determined via immunohistochemistry and immunofluorescence.There were severe infiltrates of lung cells with high levels of tracheal damage, perivascular injury and interstitial inflammatory cell infiltration when the MRL/Lpr mice from 16w to 32w comparing to the 8w old healthy MRL/Lpr mice in the control group (p0.05). Moreover, the reduced expression levels of FOXJ1 were associated with the activation of the NF-κB pathway in interstitial lung disease of MRL/Lpr mice via the modulation of p50 and p65. In addition, the expression levels of MMP2 and MMP9 pro-inflammation factors increased in the lungs of the MRL/Lpr mice from 16w to 32w.The expression level of FOXJ1 might be an indicator of the degree of lung disease in lupus-prone mice.

AIM Sleep disturbances are common in primary Sjogren's syndrome (pSS) patients and may lead to disease aggravation and decreased health-related quality of life (HRQoL). There are currently no known reported studies related to the prevalence, correlates, and impact of sleep disturbance in pSS patients from China. Therefore, this study aims to assess the sleep quality in Chinese pSS patients and evaluate its relationship with the disease activity, quality of life and mood disorders. METHODS A self-report survey was administered to 221 pSS patients and 198 healthy individuals using the Pittsburgh Sleep Quality Index (PSQI) for sleep quality. Disease activity and damage were evaluated with the European League Against Rheumatism Sjogren's syndrome disease activity index (ESSDAI). Independent samples t tests, Chi-square analysis, logistic regression were used to analyze these data. RESULTS Our results found that the prevalence of poor sleep (PSQI ≥ 6) was 57.5% and the mean global score of PSQI was 6.57 (SD 3.19) in patients, which were significantly higher than the controls (32.3% and 4.93 [SD 2.86], respectively). When trying to fall asleep, patients with pSS had some sleep disturbances, reduced sleep efficiency, increased number of awakenings than controls. There were significant correlations among dryness, ocular surface disease, HRQoL, pain, disease activity, anxiety/depression and sleep quality in pSS patients. Meanwhile, logistic regression models identified depression and Short Form-36 mental composite score as predictors of poor sleep quality. CONCLUSIONS Sleep disturbances are commonly reported in pSS patients and sleep quality is lower in pSS patients than in healthy controls. The data suggested the need for holistic assessment and management of pSS patients.

The main purpose of this study was to investigate the current situation of primary Sjögren's syndrome (pSS) patients with interstitial lung disease (ILD) in China. The relationships between social demography, disease activity, psychological status, clinical variables, and ILD were analyzed. Besides that, the quality of life (QoL) in pSS with ILD was also analyzed.In the cross-sectional study, 101 pSS patients participated in this study. Under the guidance of the researchers, the European League Against Rheumatism Sjögren's Syndrome Disease Activity Index (ESSDAI), EULAR Sjögren's Syndrome Patient-Reported Index (ESSPRI), the Short-Form 36 health survey (SF-36), Hospital Anxiety and Depression Scale (HADS), Pittsburgh sleep quality index (PSQI), Summated Xerostomia Inventory (SXI), and other related questionnaires were completed. Independent sample t tests, Mann-Whitney U test, chi-square test, and correlational analysis were used.The data showed that 28 (30.1%) pSS people with ILD. The occurrence and development of ILD were associated with disease duration, fatigue, alanine aminotransferase (ALT), neutrophils, albumin, and use of hormone. The survey also found that the QoL of pSS-ILD was significantly lower than non-ILD, especially the score in the dimension of role physical function.ILD was very common in pSS. ILD had a significant negative impact on the QoL of patients. Therefore, it is of great significance to strengthen the early intervention and drug treatment of pSS patients to prevent ILD and improve their QoL.Key Points• This cross-sectional study finds that the incidence of ILD in patients with pSS is 30.1%.• The occurrence and development of ILD is associated with disease duration, fatigue, alanine aminotransferase (ALT), neutrophils, albumin, and use of hormone.•The study also finds that the QoL of pSS-ILD is significantly lower than in patients with non-ILD, especially the score in the dimension of role physical function.

Objective To evaluate the prevalence and severity of periodontal disease in Chinese rheumatoid arthritis patients. Subjects and methods This cross-sectional study included 128 RA and 109 healthy controls. Two dentists conducted periodontal status including Plaque index (PI), Gingival index (GI), pocket probing depths (PPDs), Clinical attachment level (CAL) and Bleeding on probing (BOP) independently. Sociodemographic, lifestyle, clinical parameters and use of medication were assessed. Data were analyzed by Student's t test, χ2 test, Wilcoxin-Mann- Whitney's test, Correlational Analysis, univariate or multivariate logistic regression. Results The periodontal status was significantly worse in RA, especially the condition of dental and gingival status. RA had 4.68-fold. After adjusted potential risk factors, RA had 10.26-fold. The independent variable related to GI was DAS28 (p = .05) negatively, to the contrary, ESR (p = .013) was positively associated; the independent variable positively and related to periodontitis was educational level (p = .021) and anti-CCP positivity (p = .002). Through multivariate logistic regression, age and swollen joint were the independent variable related to periodontitis of RA (OR 1.087, p = .044) and (OR 1.560, p = .008) respectively. Conclusions Chinese RA patients show higher odds of PD. It is important to take early interventions in combination with medical therapy.

111. Dynamic Electromagnetic Behaviors of High-Temperature Superconductor Levitation System Based on YBCO Material from 3D Perspective

In this paper, a 3D numerical model, which was theoretically established and experimentally validated, was utilized to investigate the dynamic electromagnetic behaviors of a high-temperature superconductor (HTS) levitation system consisting of a cylindrical permanent magnet (PM) and an HTS fabricated by the YBCO material. The effects of critical current, cooling temperature, multiseeded material, and thickness of the HTS on electromagnetic behaviors were discussed from a 3D perspective. The dynamic electromagnetic behaviors indicate the induced current is mainly distributed near the top surface and edges of the HTS. As the PM approaches the HTS, the induced current density increases gradually. As the PM withdraws, it reduces initially and is then induced in the opposite direction. The levitation force increases with critical current but exhibits saturation. The lower the temperature, the larger the levitation force and the smaller the hysteresis. The induced current of the multiseeded HTS is characterized by a symmetrical distribution with the seed domain boundary or central axis. Its maximum value increases with the number of seeds but the levitation force significantly reduces. The induced current on the cross section is symmetrically distributed. As the thickness of the HTS decreases, the levitation force reduces drastically but the maximum value of the induced current increases. The reason was explained. Finally, the dynamic electromagnetic behaviors of a radial-type HTS bearing were investigated from a 3D perspective.

Substantial discoveries suggested that exosomes released from multiple sources of stem cells can affect the biological functions of target cells. In present period, the immunosuppressive properties of exosomes derived from bone marrow mesenchymal stem cells (BMMSCs-E) have been extensively recognized, but few studies have been reported about exosomes secreted from dental pulp stem cells (DPSCs-E) in the field of medical immunity. Hence, the aim of this study is to compare the immunomodulatory capacity of BMMSCs-E and DPSCs-E. Peripheral blood mononuclear cells (PBMCs) were co-cultured with them respectively and the proportion of regulatory T cells (Treg) was detected to increase. Subsequently, we stimulated CD4+T cells with BMMSCs-E and DPSCs-E to observe their effects on the polarizations, chemokines secretion, apoptosis, and proliferation of CD4+T cells. We found that DPSCs-E inhibited the differentiation of CD4+T cells into T helper 17 cells (Th17) and reduced the secretions of pro-inflammatory factors IL-17 and TNF-α, while promoted the polarization of CD4+T cells into Treg and increased the release of anti-inflammatory factors IL-10 and TGF-β. What's more, these capabilities of DPSCs-E were stronger than those of BMMSCs-E. In addition, DPSCs-E were more effective in inducing apoptosis of CD4+T cells compared with BMMSCs-E, and DPSCs-E inhibited the proliferation of CD4+T cells, which is similar to BMMSCs-E. We draw a conclusion that DPSCs-E have stronger immune-modulating activities than BMMSCs-E, and may be a new therapeutic tool for the treatment of immunological diseases.

Sjogren's syndrome (SS) often leads to disease-related body defects and functional impairments, which may result in the body image disturbances (BID) of patients. The aim of this study was to investigate the severity and predictors of BID among SS patients. Two hundred and thirty-one SS patients [mean (IQR) age: 51 (42-58); females: 94.4%] and 224 age and sex-matched healthy controls were included in this cross-sectional study. Questionnaires were applied: body image disturbance questionnaire (BIDQ), the hospital anxiety and depression scale (HADS), fatigue severity scale (FSS), the 10-cm pain Visual Analog Scale (VAS), the oral health impact profile-14 (OHIP-14), the ocular surface disease index (OSDI), the social support rate scale (SSRS). Independent sample t-test, Mann-Whitney U-test, Chi-square test, spearman rank correlation, and stepwise linear regression were performed by SPSS version 20.0 to analyze these data. In 231 SS patients, the mean of the overall BIDQ score was 1.80 ± 1.21, and SS patients had significantly higher scores in each domain of BIDQ compared with healthy controls (p < 0.05). The stepwise multiple linear regression analysis revealed that high BIDQ score was predicted by severe anxiety (β = 0.081; p < 0.001), high disease activity (β = 0.038; p < 0.001) and poor oral health (β = 0.017; p = 0.007) in SS patients. Patients with SS suffer from severe BID and it is necessary for rheumatologists to pay more attention to SS patients' body image disturbance, especially those with high disease activity, severe anxiety, and poor oral health to improve patients' quality of life.

To understand the effects of fish density on water quality and physiological changes of golden pompano (Trachinotus ovatus) fingerlings during transportation, fish were stocked at three densities (27 kg/m3, 54 kg/m3 and 81 kg/m3) for 8 h using the method of anesthesia hypothermia in sealed containers. The fish reared in the recycling system were used as the control. The pH of water at three fish densities was significantly lower than the initial value and decreased gradually with the increase of fish density. The lowest pH and ammonia nitrogen were found in the 81 kg group, reaching 6.24 and 0.08 mg/L, respectively. The liver catalase activity increased gradually but was still significantly lower than that in the control. The liver peroxidase activity fell gradually corresponding to the increase of fish density but was higher than that in the control. Fish density during transportation did not affect the lactate dehydrogenase activity in the liver. After 8-h transportation, fish in all three groups had a lower level of glucose and creatinine in the serum than in the control fish. The content of serum cortisol soared after transportation, but no significant difference was found among three fish densities. Glutamate pyruvate transaminase and glutamic oxaloacetic transaminase activities in the serum had a similar pattern to those of the fish in the control and showed a gradual reduction with increasing fish density. Fish density during transport significantly affected water quality (e.g., pH and NH3-N) and liver function. Our results indicate that the fish density of 81 kg/m3 is safe to transport golden pompano fingerlings for 8 h.

The aim of this meta-analysis was to compare sleep outcomes of AS patients and controls. Six electronic databases were searched: PubMed, Embase, IndMed, Cochrane Library, CNKI and Web of Science. Statistical analyses were executed using Revman 5.3 software. Nine studies and a total of 3169 participants were included in the meta-analysis. When sleep was assessed using PSQI, significant differences were observed in subjective sleep quality, sleep latency, sleep efficiency, sleep disorders, sleep medication use and total PSQI between patients with AS and controls, and subjective sleep quality was most severely affected. In PSG, eight variables of sleep quality (stage I sleep, stage II sleep, slow wave sleep, rapid eye movement, arousal index, periodic leg movement index, sleep latency, sleep efficiency) were lower in AS patients than in controls. Three variables of sleep quality assessed by USI (estimated sleep time, sleep sufficiency index, number of awakenings per night) and two variables of sleep quality assessed by MOS sleep scale (sleep disturbance scale, sleep problem index II) were lower in AS patients than in controls. It indicated that AS patients experience more serious sleep disorders. Early recognition and appropriate interventions are essential to improve patients' sleep quality.

A study on Chlamys nobilis sperm cryopreservation by a programmable freezing method was conducted under laboratory condition. Four cryoprotectant agents (dimethyl sulfoxide [DMSO], methanol [MET], propanediol[PG] and ethylene glycol [EG]) and four concentrations (5%, 10%, 20% and 30%) were evaluated for their ability to retain sperm motility, movement characteristics and fertility. Results showed that cryopreserved sperm total motility produced by DMSO and MET at 5%, 10% and 20% were higher than other cryoprotectant treatment groups (CPA groups), as well as rapid sperm percentage. The curvilinear (VCL) and straight line (VSL) velocity produced by DMSO at 5% significantly higher than other CPA groups (p 0.05), as well as the beat‐cross frequency (BCF). A significant higher fertilization rate was produced in DMSO than that in MET at same concentration (p 0.05). Overall, 5%‐20% DMSO was more suitable for Chlamys nobilis sperm programmable cryopreservation when the calcium‐free Hanks’ balanced salt solution was used as the extender, and 10°C/min from 0°C to −80°C was used as freezing rate. The findings presented in this study will benefit conservation programs for Chlamys nobilis.

To identify the impact of systemic lupus erythematosus (SLE) on health-related quality of life (HRQoL) domains compared with the results of healthy controls by systematically reviewing the literature. A systematic literature search was performed on MEDLINE, Web of Science and EMBASE until April, 2018 to obtain eligible studies reporting mean and standard deviation scores for each domain of the 36-item Short-Form Health Survey (SF-36) in SLE patients and healthy controls. Random effect model was performed to summarize the scores of each domain. The forest plot was used to compare the scores of SLE patients with healthy controls. Review Manager (version 5.3) was adopted in the meta-analysis. In all, 13 studies were included in the work, including 1279 SLE patients and 1466 healthy controls. Compared with controls, patients with SLE had lower scores in all SF-36 dimensions (physical function, physical role function, body pain, general health, vitality, social function, emotional role function, mental health), especially in the physical role function. SLE does impair HRQoL to varying degrees. It is indispensable to measure and assess HRQoL of SLE patients regularly, which contributes to formulate targeted interventions appropriately and provide effective ways of management of the disease positively.

The color of aquatic animals plays an important role in consumer acceptability and perceived quality. In a previous study, a red claw crayfish Cherax quadricarinatus strain with steady and hereditary shell color of blue was selected. To evaluate the potential value of this strain in crayfish culture, the growth performance, carotenoid content, and temperature tolerance of two crayfish strains (shell color; blue and blue-green) were conducted. After the 56-day rearing period, blue crayfish showed higher feed intake and growth rate, but lower survival than blue-green crayfish. Astaxanthin was the main pigment observed in the carapace of both strains, and the content of astaxanthin in blue crayfish was 18.62 mg/kg of wet weight, which was significantly lower than that of astaxanthin in blue-green crayfish (83.23 mg/kg of wet weight). The 7-day survival of two crayfish strains less than 34°C was 100%, and blue-green crayfish showed stronger tolerance to high water temperature exposure (35°C–38°C) than blue crayfish. Furthermore, after high temperature stress, lower activities of superoxide dismutase and catalase, and higher malondialdehyde in the hepatopancreases of blue crayfish were observed than those in blue-green crayfish. This study provides important information for future breeding programs for the red claw crayfish.

Purpose Few studies have assessed fatigue of meningioma patients. The purpose of this study was to explore the prevalence, severity, and risk factors of multidimensional fatigue, as well as the impact on health-related quality of life (HRQoL) in Chinese meningioma patients newly diagnosed. Methods This cross-sectional study included 120 Chinese meningioma patients recruited from Affiliated Hospital of Nantong University from January 2020 to February 2021. Data were collected before surgery, including demographic, clinical and psychological characteristics, as well as fatigue scores, based on completion of the Multidimensional Fatigue Inventory (MFI-20). Spearman correlation and multiple linear regression were used to analyze the data. Results The results demonstrated that a high prevalence of severe fatigue for each dimension: general fatigue (33.3%), physical fatigue (27.5%), reduced activity (28.3%), reduced motivation (12.5%), mental fatigue (11.7%), and total fatigue (23.3%). Headache and anxiety were predictors of general fatigue. Depression was an independent predictor of physical fatigue. Karnofsky Performance Status (KPS) score and depression independently predicted reduced activity. Depression and Epworth Sleepiness Scale (ESS) score were risk factors predicting reduced motivation, while KPS score and anxiety predicted mental fatigue. Importantly, comorbidity, KPS score, headache, depression, sleep disturbances, and ESS score could predict total fatigue. Furthermore, MFI-20 scores in all dimensions were negatively correlated with Short Form 36 Health Survey (SF-36) scores in all dimensions. Conclusion Our findings indicate that meningioma patients newly diagnosed are frequently affected by fatigue, potentially contributing to impair HRQoL. For patients with risk factors of fatigue, targeted interventions are advised in order to decrease fatigue and improve HRQoL.

Therefore, the purpose of this study was to investigate the relationship between dietary diversity score (DDS) and anxiety and depression among centenarians and their offspring and spouses. METHODS Evidence suggests that poor mental health (MH) is a risk factor for the health of older adults. Dietary diversity is considered to be related to healthy aging. However, the relationship between diet and MH is still unclear. Therefore, the purpose of this study was to investigate the relationship between dietary diversity score (DDS) and anxiety and depression among centenarians and their offspring and spouses. RESULTS Among the 288 older adults, 12.8% reported symptoms of depression, and 8.7% reported anxiety. People with a lower dietary diversity had higher rates of anxiety and depression. After controlling for age, place of residence, economic status, alcohol drinking, and physical activity, a lower DDS was found to be a risk factor for depressive symptoms [odds ratio (OR): 2.237; 95% confidence interval (CI): 1.009-4.959; P=0.048]. Purpose Sleep disturbance is common in meningioma patients and may lead to disease aggravation and decreases health-related quality of life (HRQoL). However, the sleep quality of meningioma patients newly diagnosed and ready for surgery has not been well clarified in China. This study aims to evaluate the prevalence, correlates, and impact of sleep disturbance among Chinese meningioma patients. Methods In this cross-sectional study, meningioma patients were recruited from the Affiliated Hospital of Nantong University from January 2020 to November 2020. A series of questionnaires were applied: the 0–10 Numerical Rating Scale (NRS), the Hospital Anxiety and Depression Scale (HADS), the Multidimensional Fatigue Inventory (MFI-20), the Epworth Sleepiness Scale (ESS), the Short-Form 36 (SF-36), the Pittsburgh Sleep Quality Index (PSQI). Independent samples t test, Mann-Whitney U test, chi-square analysis, Pearson/Spearman correlation, and binary logistic regression were used to analyze the data. Results 100 meningioma patients completed the questionnaires. Sleep disturbance affected 43% of the meningioma patients and was linked to many concomitant symptoms, such as headache, fatigue, anxiety and depression. Binary logistic regression indicated that fatigue and headache were predictors of sleep disturbance in meningioma patients. Meanwhile, severe sleep disturbance led to lower quality of life. Conclusions These findings demonstrated that a considerable number of meningioma patients newly diagnosed and ready for surgery suffered from sleep disturbance, potentially contributing to impair HRQoL. Medical personnel should pay more attention to meningioma patients with sleep disturbance and take effective measures to improve sleep quality, with the ultimate goal to improve their HRQoL.

The effects of high hydrostatic pressure (HHP) processing at 200 MPa, 400 MPa, and 600 MPa on the proximate composition, inactivation of bacteria, and color of the red claw crayfish Cherax quadricarinatus were evaluated. High hydrostatic pressure processing resulted in complete shelling and a significant decrease in the moisture content (P < 0.05), whereas no significant differences were observed in the content of ash, crude protein, and crude lipid, except for the crude lipid content between 200 MPa and 600 MPa groups (P < 0.05). After pressurization, the Escherichia coli, Vibrio vulnificus, and Vibrio parahaemolyticus counts decreased significantly with the increase of pressure, even to an undetected level for V. vulnificus and V. parahaemolyticus at 400 and 600 MPa (P < 0.05), respectively. During 30-d storage at 4°C and –20°C, the counts of these three bacterial species increased significantly with increased storage time and temperature. After HHP processing, a* value changed substantially, whereas L* and b* values had little change for meat. The total color difference (ΔE*) suggested that the HHP-treated red claw crayfish exhibited very distinctive or substantial differences compared with fresh individuals.

Although therapies of cancer are advancing, it remains challenging for therapeutics to reach the sites of metastasis, which accounts for majority of cancer associated death. In this study, we have developed a strategy that guides an anti-programmed cell death-ligand 1 (aPDL1) antibody to accumulate in metastatic lesions to promote anti-tumour immune responses. Briefly, we have developed a combination in which Vadimezan disrupts tumour blood vessels of tumour metastases and facilitates the recruitment and activation of adoptively transferred aPDL1-conjugated platelets. In situ activated platelets generate aPDL1-decorated platelet-derived microparticles (PMP) that diffuse within the tumour and elicit immune responses. The proposed combination increases 10-fold aPDL1 antibody accumulation in lung metastases as compared to the intravenous administration of the antibody and enhances the magnitude of immune responses leading to improved antitumour effects., Cancer metastasis is the leading cause of death in patients, here, the authors show disrupting tumor vasculature could recruit and activate anti-PD-L1 engineered platelet at metastatic tumor sites to block the PD-1/PD-L1 crosstalk and enhance the anticancer immunotherapy.

Sleep disturbance is common in meningioma patients and may lead to disease aggravation and decreases health-related quality of life (HRQoL). However, the sleep quality of meningioma patients newly diagnosed and ready for surgery has not been well clarified in China. This study aims to evaluate the prevalence, correlates, and impact of sleep disturbance among Chinese meningioma patients. In this cross-sectional study, meningioma patients were recruited from the Affiliated Hospital of Nantong University from January 2020 to November 2020. A series of questionnaires were applied: the 0–10 Numerical Rating Scale (NRS), the Hospital Anxiety and Depression Scale (HADS), the Multidimensional Fatigue Inventory (MFI-20), the Epworth Sleepiness Scale (ESS), the Short-Form 36 (SF-36), the Pittsburgh Sleep Quality Index (PSQI). Independent samples t test, Mann–Whitney U test, chi-square analysis, Pearson/Spearman correlation, and binary logistic regression were used to analyze the data. One hundred meningioma patients completed the questionnaires. Sleep disturbance affected 43% of the meningioma patients and was linked to many concomitant symptoms, such as headache, fatigue, anxiety, and depression. Binary logistic regression indicated that fatigue and headache were independently associated with sleep disturbance of meningioma patients. Meanwhile, severe sleep disturbance led to lower quality of life. These findings demonstrated that a considerable number of meningioma patients newly diagnosed and ready for surgery suffered from sleep disturbance, potentially contributing to impair HRQoL. Medical personnel should pay more attention to meningioma patients with sleep disturbance and take effective measures to improve sleep quality, with the ultimate goal to improve their HRQoL.

Background Intestinal flora disorder and immune abnormalities have been reported in systemic lupus erythematosus (SLE) patients. Few researches indicated the intestinal status in Chinese SLE patients. Berberine (BBR) showed significant effects on regulating the intestinal flora, repairing gut barriers and regulating immune cells. This study mainly explored intestinal flora and metabolites in local Chinese SLE patients and the influence of BBR to MRL/Lpr mice. Methods 16S high-throughput sequence and gas chromatographic technique were used to analyzed the intestinal flora and metabolites in SLE patients. MRL/Lpr mice were oral treated with BBR in low, medium and high dosages for 6 weeks. Urine protein was monitored. After the procedure, gintestinal status of MRL/Lpr mice were analyzed like human. A wide range of autoantibodies were tested. Kidney tissue was analyzed for C3, IgG and IgM expressions and colon tissue was analyzed for gut barrier markers. Flow cytometry determined the immune cells. Results Dysbacteriosis and abnormal metabolism influenced the Chinese lupus pathogenesis. BBR treatment reduced the urine protein, inhibited the auto-antibodies and ameliorated lupus nephritis (LN) in MRL/Lpr mice. In addition, BBR altered the relative abundance of Bacteroides and Verrucomicrobia and the butyric acid content in colon of MRL/Lpr mice. The increase of tight junction protein also indicated that the gut barrier was repaired by BBR. Treg and Tfr cells in spleen and mesenteric lymph node (MLN) were increased. Conclusions These results revealed a therapeutic effect of berberine on SLE from gut microbiota to immune status.

OBJECTIVES: This study aims to explore body image disturbance (BID) in Chinese patients with rheumatoid arthritis (RA). PATIENTS AND METHODS: A total of 211 RA patients (30 males, 181 females; mean age 50.4±12.4 years; range, 20 to 81 years) and 203 age-matched healthy subjects (26 males, 177 females; mean age 49.6±5.6 years; range, 34 to 64 years) were included in this study. Patients were assessed by Body Image Disturbance Questionnaire (BIDQ), Visual Analog Scale, Health Assessment Questionnaire-Disability Index, disease activity score in 28 joints, Short Form 36 health survey, Hospital Anxiety and Depression Scale, Rosenberg Self-Esteem Scale and erythrocyte sedimentation rate. Rosuvastatin is a synthetic statin of 3-hydroxy-methyl-3-glutamyl coenzyme A reductase inhibitor. It has pleiotropic characteristics including hepatic selectivity, minimal metabolism, inhibition of inflammation, and induction of osteoblast differentiation. In this study, dental pulp stem cells (DPSCs) were treated with lipopolysaccharide alone or with rosuvastatin. Then, we examined the accelerative effects of rosuvastatin on odontoblast differentiation and mineralized nodule formation by real-time polymerase chain reaction (RT-PCR), western blot, alizarin red S staining, and alkaline phosphatase staining. The extent of anti-inflammation was determined by RT-PCR and analysis of the expression of tumor necrosis factor α, interleukin 1β (IL-1β), and IL-6. Furthermore, the activation of nuclear factor kappa B (NF-κB) was determined by western blot. This study demonstrates that rosuvastatin may speed up odontoblast differentiation and rescue inflammatory reaction by suppressing the NF-κB signaling pathway. It is believed that our findings provide novel perceptions on odontogenic differentiation of DPSCs.

Background While the NOD-Like Receptor Protein-3 (NLRP3) inflammasome is involved in a variety of nervous system diseases, its role in epilepsy still needs to be further investigated. Methods The expressions of NLRP3 inflammasome and apoptosis related proteins were examined by Western blot. MTT was used to assess cell viability. The role of NLRP3 inflammasome in epileptic neuronal apoptosis was further validated in NLRP3 knockout (KO) mice by Nissl staining. Results Exposure of SH-SY5Y cells to free-Mg2+ solutions increased the expression of NLRP3 inflammasome with a concomitant increase in neuronal apoptosis. This effect was inhibited in cells treated with MCC950 as a common NLRP3 inhibitor, thereby implicating the role of NLRP3 inflammasome in epileptic neuronal apoptosis. In vivo relevance of this finding was further corroborated in the NLRP3 KO mice. Compared with the wild type mice, neuronal loss induced by pentylenetetrazole was significantly inhibited in the NLRP3 KO mice. Conclusion The study presented herein demonstrates the interaction between NLRP3 inflammasome and epilepsy progression. In addition, MCC950 might represent an important therapeutic drug for the treatment of NLRP3 inflammasome driven epileptogenic activity.

Objective The purpose of this meta-analysis was to systematically review the literature to evaluate the impact of systemic sclerosis (SSc) on the health-related quality of life (HRQoL) assessed by the Medical Outcomes Short-Form-36 questionnaire (SF-36). Methods A systematic literature search was performed on PubMed, Web of Science and Embase until April 2017 to obtain eligible studies. Random effect model was performed to summarize the scores of each domain. Scores from the SF-36 questionnaire were used as the outcome measurements, and mean differences with 95% confidence intervals were calculated. Results In total, seven studies were eligible for inclusion criteria, comprising 795 SSc patients and 1154 healthy controls. The SF-36 questionnaire score of each domain (physical function, role physical function, emotional role function, vitality, mental health, social function, body pain, general health) was lower in SSc patients than in healthy controls, meanwhile, physical component scale (PCS) and mental component scale (MCS) scores were all lower in patients with SSc than in healthy controls. Likewise, pooled mean scores of PCS and MCS ranged from 31.20 to 52.80, 37.40 to 68.30, respectively. Additionally, the score of PCS was lower than that of MCS in SSc patients. Conclusions This meta-analysis showed that SSc patients had lower HRQoL than healthy controls, and SSc had negative influence on the HRQoL of patients. This indicates that clinical workers should pay more attention to SSc patients' HRQoL, so as to improve global health of patients with SSc.

The forkhead box C1 (Foxc1) protein, a member of the forkhead/winged helix transcription factor family, is required in stem cell developmental processes. Recently, multiple studies have indicated the crucial role of Foxc1 in mesenchymal stem cell differentiation, but the precise effects and mechanisms on dental pulp stem cells (DPSCs) remain unclear. In this study, we evaluate the role of Foxc1 on the odontogenic differentiation and proliferation of DPSCs. Our results show that Foxc1 decreases time dependently in odontogenic differentiation of DPSCs. Meanwhile, overexpression of Foxc1 could significantly inhibit the mineralization of DPSCs and the expression of odontogenic-related genes, such as runt-related transcription factor 2 (Runx2), dentin sialophosphoprote (DSPP), and dentin matrix acidic phosphoprotein 1 (DMP-1). Foxc1 overexpression does not significantly alter the proliferation of DPSCs. In addition, Foxc1 reduces the expression of p-Smad1/5, an important modulator of bone morphogenetic protein (BMP)/Smad signaling pathway, inhibiting BMP/Smad signaling pathway. In conclusion, our data demonstrated that Foxc1 inhibits odontogenic differentiation of DPSCs and odontogenic-related gene expression through the BMP/Smad signaling pathway which may be useful for the dental regeneration and repair.

Increasing evidence shows miR-155 plays an important role in regulating inflammatory processes in systemic lupus erythematosus (SLE), especially in lupus nephritis (LN). Because the chemokine CXCL13 is implicated in the pathogenesis of LN, here we examined whether miR-155 can modulate the activity of CXCL13 or its receptor CXCR5. We determined the expression of CXCL13 in normal and MRL/lpr mice and found elevated levels of CXCL13 in the kidneys of MRL/lpr mice compared with normal kidneys. Besides, CXCL13 expression was mainly detected in the glomerulus, specifically to mesangial areas. We then transfected a miR-155 mimic in human renal mesangial cells (HRMCs) to overexpress miR-155 and detected decreased protein levels of CXCR5 by western blot analysis. Transfection of the miR-155 mimic into CXCL13-treated HRMCs resulted in a significantly reduced proliferation rate of HRMCs as measured by the cell-counting assay and flow cytometry. Moreover, increased intracellular miR-155 also led to decreased phosphorylation of ERK and TGF-β1 production. Together, these results revealed that miR-155 may play a role in the pathogenesis of LN.

Relationships between shell morphological traits and byssus dimensions in adult winged pearl oysters Pteria penguin are described to include a more detailed description of byssus growth in juveniles under typical culture conditions. Byssus growth, byssus diameter (BD) relations between the byssus and shell traits [shell height; total wet weight (TW)], byssus occurrence, and the size distributions in samples collected monthly or bimonthly over a 1-y duration were recorded. Pearl oysters exhibited continuous shell growth and weight increase over the experimental period, but byssal measurements exhibited a different pattern. The increase in BD decreased as oysters aged and no further increase was observed after day 267. Moreover, BD/TW ratio exhibited a steadily diminishing trend as P. penguin aged. The relationships between the BD and the shell traits in 68-day-old oysters proved to be insignificant, probably because the oysters were neither heavy nor large enough to exhibit the relationships, but later, when oysters increased in size and weight, the relationships became significant. Byssus diameter was determined to be slightly more closely related to the weight of the animal than the size of the shell, and oysters at day 183 exhibited comparatively higher r2 values than oysters at day 366 of cultivation. The occurrence of oysters without byssus decreased gradually during cultivation from 30% initially to 0 after a year of cultivation.

Disease activity was assessed using the European League Against Rheumatism Sjögren’s Syndrome Disease Activity Index. We examined these data using independent samples t-tests, Mann–Whitney U test, chi squared analysis, and linear regression.Results: The result for each domain in Short Form 36 health survey was lower in pSS patients than in healthy controls, especially the score in the dimension of role physical function. In the bivariate analysis, age, pain, fatigue, disease activity, disease complication, anxiety/depression, oral disorders, and impaired swallowing correlated with QoL. Also, in the linear regression model, pain, fatigue, disease activity, impaired swallowing, and anxiety/depression remained the main predictors of QoL.Conclusion: pSS patients had a considerably impaired QoL compared to the controls, and pSS could negatively affect the QoL of patients. Measuring QoL should be considered as a vital part of the comprehensive evaluation of the health status of pSS patients, which could contribute some valuable clues in improving the management of disease and treatment decisions. Nonvolatile taste active compounds, including free amino acids (FAAs), 5′-nucleotides, betaine, soluble sugars, trimethylamine oxide and organic acids of wild and cultured mud crab Scylla paramamosain were examined. The main taste in mud crab (MC) is derived from the FAAs, 5′-nucleotides, betaine, and organic acids, based on the fact that these compounds have a taste activity value (TAV) > 1. Wild MCs had a higher content of total FAAs, 5′-nucleotides, betaine, and organic acids in meat compared with cultured animals. However, no differences in the taste active compound levels were observed in the gonads of wild and cultured MCs, with betaine being the exception (wild crab > cultured crab). The total concentrations of FAAs in MC ranged from 12.71 to 36.94 mg/g. Monosodium glutamate (MSG)-like FAAs were primarily contributed by glutamic acid, and ranged from 0.71 to 0.93 mg/g. The concentrations of 5′-nucleotides were 1.10–2.24 mg/g, in which 5′-inosine monophosphate (IMP) and 5′-adenosine monophosphate (AMP) were the predominant TAVs (TAV > 1). The total concentrations of organic acids in MC were 2.41–11.10 mg/g, in which lactic and succinic acid in meat, and citric acid and succinic acid in female gonads were the main organic acids. The gonads were observed to have higher concentrations of 5′-nucleotides and organic acids compared with crab meat, with the equivalent umami concentration in gonads being higher than that in crab meat. In conclusion, wild MCs contained more active-taste nonvolatile flavor components relative to those detected in cultured MCs.

A number of studies have reported the suicidal ideation (SI) or suicide attempts (SA) of patients with rheumatic diseases. However, the estimated prevalence of those disorders varies substantially between studies. This systematic review aimed to describe the prevalence of SI and SA in rheumatic diseases. Literature search was done using Web of Science, Embase, the Cochrane database library, PubMed and CNKI database through June 2017. Studies were screened according to inclusion and exclusion criteria and the qualities of included studies were evaluated. The data was analyzed using STATA version 12.0. A random-effect meta-analysis was conducted on all eligible data. A total of 17 identified studies matched the inclusion criteria, involving 5174 participants with systemic lupus erythematosus (SLE), osteoarthritis (OA), rheumatoid arthritis (RA) and fibromyalgia (FM). Meta-analysis showed that rheumatic diseases patients have high prevalence of SI (26%, 95% CI: 19%-32%, I²=96.2%) and SA (12%, 95% CI: 3%-21%, I²=96.6%). We also found the prevalence of SI and SA in females may be higher than in males. All of these indicated that rheumatologists should screen for SI and SA in their patients. Early appropriate intervention is therefore essential to promote the patients' good mental health.

The diagnoses of bacterial infections remain a challenge in environment safety and human health area. Nanomaterial-based ‘enzyme nose’ sensor is a useful analytical technique for the detection of toxicologically important targets in biological samples. In this paper, we used three quaternized magnetic nanoparticles–urease sensors to detect bacteria with excellent sensitivity for several gram-positive or gram-negative bacteria. The response intensity of the nanoarray sensor is dependent on the level of displacement determined by three quaternized magnetic nanoparticles–urease binding strength and pathogenic cells–nanoparticles interaction. Based on the ability of urease to hydrolyze urea and increase the pH value of the system, the detection of pathogen is translated into a pH increase, which can be readily detected using a litmus dye. These characteristic responses of color show repeatable pathogenic cells and can be differentiated by principle component analysis (PCA). Our approach has been used to measure bacteria with an accuracy of 90.7% for 102 cfu mL−1 within 30 min. The measurement system has a potential for further applications and provides a facile and simple method for the rapid analysis of pathogens.

Abnormal odontoblast differentiation of dental pulp stem cells (DPSCs) caused by inflammation is closely related to the development of dental caries. Neuropilin-1 (NRP1) is one of the members of neuropilin family. It can combine with disparate ligands involved in regulating cell differentiation. FYN belongs to the protein-tyrosine kinase family, which has been implicated in the control of cell growth, and the effect can be further strengthened by inflammatory factors. In our studies, we verified that NRP1 can form complexes with FYN and have the correlation changes in odontoblast differentiation of DPSCs. Therefore, we surmise that in the progress of dental caries, NRP1 interacts with FYN, by expanding inflammation and inhibition of odontoblast differentiation of DPSCs through nuclear factor kappa B (NF-κB) signaling pathway. In this subject, we first investigated the expression and interaction of NRP1 and FYN in DPSCs. And then, we researched the effect of this complex controlling downstream signal pathway in normal or inflammation stimulated DPSCs. Finally, we analyzed the relationship between this role and odontoblast differentiation of DPSCs. This research will provide the molecular mechanism of inflammation factors of dental caries through activating NF-κB signal regulating odontoblast differentiation in DPSCs for finding new potential drug targets for the clinical treatment of dental caries.
