117. Haplotype-resolved and telomere-to-telomere reference genome of diploid banana Musa acuminata

Abstract

Here, we reported a haplotype-resolved and telomere-to-telomere genome assembly of M. acuminata using PacBio HiFi reads, Nanopore ultra-long reads and high-covered Hi-C data. This assembly represented a significantly high-quality reference assessed by contiguity (contig N50: 16.53Mb and 18.58Mb; LAI: 20.18 and 19.48), completeness (BUSCOs: 98.57% and 98.57%), and correctness (QV: 45.97 and 46.12). All centromeres and 36 of 44 telomeres had been captured. The mapping rates of HiFi and Illumina reads both exceeded 99.80%. In total, 40,889 and 38,269 protein-coding genes were annotated in the two haploid assemblies, respectively, of which 9.56% and 3.37% were newly annotated compared with previous databases. Gene BUSCOs reached up to 89.41% and 90.27%, respectively. Through comparing our assembly and the previously published genome of M. acuminata, we identified two large translocation regions with length of 3 Mb and 10 Mb, which were further confirmed by Hi-C heatmap and targeted-PCR gel electrophoresis. This haplotype-resolved and telomere-to-telomere reference genome of M. acuminata provides an excellent resource for further understanding the mechanisms of subgenome evolution, domestication history, and genetic breeding of banana.

119. Telomere-to-telomere haplotype-resolved reference genome reveals subgenome divergence and disease resistance in triploid Cavendish banana

Abstract

Banana is among the most popular fruits worldwide. Cavendish-type bananas, which have a monospecific Musa acuminata origin (AAA), account for around half of the global banana production, thereby are of great significance for human societies. However, until now, the high-quality haplotype-resolved reference genome is still undecoded for banana cultivars. Here, we reported the telomere-to-telomere (T2T) and haplotype-resolved reference genome of ‘Baxijiao’ (Cavendish) consisting of three haploid assemblies. The sizes of the three haploid assemblies were estimated to be 477.16 Mb, 477.18 Mb and 469.57 Mb, respectively. Although with monospecific origins, the three haploid assemblies showed great differences with low levels of sequence collinearity. Several large reciprocal translocations were identified among chromosomes 1, 4 and 7. An expansion of gene families that might affect fruit quality and aroma was detected, such as those belonging to sucrose/disaccharide/oligosaccharide catabolic processes, sucrose metabolic process, starch metabolic process, and aromatic compound biosynthetic process. Besides, an expansion of gene families associated with anther and pollen development was observed, which could be related to parthenocarpy and sterility of the Cavendish cultivar. Finally, loss of resistance genes in the ‘Baxijiao’ genome was observed, particularly in the gene clusters in chromosomes 3 and 10, providing potential targets to investigate for molecular dissection of disease resistance in banana. This T2T haplotype-resolved reference genome will thus be a valuable genetic resource for biological studies, molecular breeding and genetic improvement of banana.

This assembly represented a significant high-quality on contiguity (contig N50: 16.53Mb and 18.58Mb; LAI: 20.18 and 19.48), completeness (BUSCOs: 98.60% and 98.60%), correctness (QV: 45.97 and 46.12). All centromeres and 36 of 44 telomeres have been captured. Both HiFi and Illumina reads mapping rates exceeded 99.8% while RNA-seq reads mapping rates ranged from 92.44% to 97.34%. Two haploid genome annotated 40,889 and 38,269 protein-coding genes, respectively, of which 9.56% and 3.37% were newly annotated comparing with the previous database. Gene BUSCOs were up to 89.40% and 90.30%, respectively. Through comparing haplotype-resolved genome and previous T2T genome, we identified two large translocation regions with length of ~3Mb and ~10Mb, which were further confirmed by HiC heatmap and targeted-PCR gel electrophoresis. This haplotype-resolved and telomere-to-telomere genome of M. acuminata ssp. malaccensis was an excellent resource for further understanding mechanisms of subgenome evolution, domestication history and genetic breeding.

Abstract

Construction of polymer‐based artificial solid‐electrolyte interphase films on Zn metal anode holds great potential in the suppression of both dendrite growth and side reaction in rechargeable aqueous Zn‐ion batteries. However, the traditional polymer films suffer from the critical issues of sluggish Zn2+ transport kinetics and rigid interface. Herein, zinc alginate (ZA) hydrogel is designed and prepared as a dynamic interface and Zn2+ redistributor on Zn anode via in situ cross‐linking reaction. The zincophilic and negatively charged carboxyl groups of ZA promote the transport of Zn2+ ions along a "Z‐type" pathway, the repulsion of free SO42‐ anions, and the desolvation of Zn2+ ions, consequently leading to the homogeneous deposition of Zn and the effective suppression of side reaction. Additionally, the dynamic flexibility of ZA hydrogel endows the Zn anode with self‐adapting interface to accommodate the volume variation and repair the possible ruptures, thereby guaranteeing the long‐term cycling stability. Assisted by the ZA layer, the Zn anode achieves a prolonged lifespan over 2200 h without the formation of Zn dendrites and by‐products. Outstanding cycling stability is also demonstrated for the Zn anode when coupled with MnO2 cathode, further demonstrating its prospects for practical application. [ABSTRACT FROM AUTHOR]

Abstract

Inspired by carbon monoxide dehydrogenase (CODH), mimicking its inner and outer spheres is a promising strategy in CO2 reduction catalyst design. However, artificial CODH‐like catalysts are generally limited to the inner sphere effect and only applicable in organic solvents or for electrocatalysis. Herein, an aqueous CODH mimic with both inner and outer spheres for photocatalysis is reported. In this polymeric unimolecular catalyst, the inner sphere is composed of cobalt porphyrin with four appended amido groups and the outer sphere consists of four poly(2‐(dimethylamino)ethyl methacrylate) (PDMAEMA) arms. Upon visible light irradiation (λ >420 nm), the as‐prepared catalyst exhibits a turnover number (TONCO) of 1731.2 in the reduction of CO2 into CO, which is comparable to most reported molecular catalysts in aqueous solution. The mechanism studies indicate that, in this water‐dispersible and structurally well‐defined CODH mimic, the cobalt porphyrin core serves as the catalysis center and the amido groups function as hydrogen‐bonding pillars helping to stabilize the CO2 adduct intermediate, whereas the PDMAEMA shell renders both water solubility and a CO2 reservoir through reversibly capturing of CO2. 141. High-loading single cobalt atoms on ultrathin MOF nanosheets for efficient photocatalytic CO2 reduction.

Abstract

Based on suitable scaffolds, constructing high-loading single-atom catalysts is a promising strategy to achieve highly efficient catalysis. Herein, using ultrathin metal-organic framework (MOF) nanosheets (2.4±0.5 nm) as the support, single-atom catalysts with high cobalt loading (6.0 wt%) were constructed (denoted as Co-MNSs) by a simple bottom-up synthetic strategy. The catalytic system of Co-MNSs exhibited an outstanding photocatalytic CO2-to-CO evolution rate of 7,041 µmol g−1 h−1 and a selectivity of 86% in aqueous media under visible-light irradiation, which has reached the top level of the reported MOF-based photocatalysts. The control experiments and theoretical calculation revealed that the Co—N4 moiety in the MOF nanosheets acted as the active site for the photocatalytic CO2-to-CO conversion. The boosted photocatalytic performance could be ascribed to the high aspect-ratio of layered Co-MNSs providing abundant accessible active sites on their surfaces, which reduced the energy barrier, improved the charge separation efficiency, and also facilitated the adsorption of CO2 to form the reactive radicals of *COOH. Our study provides an appealing strategy for constructing high-loading single-atom catalysts and demonstrates the significance of 2D ultrathin MOF nanosheets as the support in boosting CO2 photoreduction efficiency. Abstract

Lithium/thionyl chloride (Li/SOCl2) primary batteries are appealing power solutions because of their remarkable electrochemical performances. However, their mass applications are hindered by the challenges in sustainability, cost and safety concerns owing to the employed Li chemistry. Here, magnesium (Mg) chemistry is shown as a promising alternative through synergistic optimization of electrolyte solvation and electrode reaction kinetics. The first Mg/SOCl2 primary battery yields surprisingly high specific capacities up to ≈14 000 mAh g−1 at a decent discharge voltage of ≈1.67 V, which outperforms the state‐of‐the‐art Mg‐based primary batteries. In addition, it retains almost 100% of the original capacity after 20‐day reservation. The impressive battery performances are originated from the stabilized MgCl2 formation on high‐surface‐area carbon cathode and suppressed Mg anode corrosion via the Mg‐induced solvation effect. Mg/SOCl2 primary batteries are promising candidates for low‐cost and recyclable power supplies, and they thus open new avenues for the development of sustainable battery chemistries.
