Heat shock protein (Hsp) 60 plays a key role in the translocation of proteins and cytoprotection, is primarily localized inside mitochondrial, and its levels increase in skeletal muscle upon exercise (Folkesson et al., 2013). The aim of this study was to examine muscle fiber specificity of HSP60 at rest and after an endurance training program of 45 days. Forth-eight male young (7-weeks old) healthy mice (BALB/c) were subdivided into six groups (8 mice per group). Three groups were trained on a rota-rod, at a gradually increasing duration and speed; while the other three groups did not perform any type of regular physical activity. One group of each condition was sacrificed after 15, 30 and 45 days. Forth-eight hours after the last exercise session all mice were sacrificed by cervical dislocation and posterior muscles group of hindlimb (gastrocnemius, soleus and plantaris) were dissected, weighed and embedded into paraffin or frozen in liquid nitrogen. Immunohystochemistry and immunofluorescence analysis showed that skeletal muscle type I fiber expressed high levels of Hsp60. The western blotting analyses of the entire posterior muscle group did not show any difference in the protein levels after endurance training, while the analysis of soleus muscle (reach in type I fibers) showed an over expression of Hsp60 after 30 and 45 days of endurance training. These data indicated that Hsp60 is muscle fibre type–specific. This may be due to the differences in mitochondrial content between slow and fast fibres. Anyway Hsp60 may be localized also in the cytoplasm, in the outer membrane, in the interstitium and in the blood stream, hence the role of this protein in endurance training need be elucidated., Italian Journal of Anatomy and Embryology, Vol 118, No 2 (Supplement) 2013

There is growing evidence that molecular chaperones/heat shock proteins are involved in the pathogenesis of a number of human diseases, known as chaperonopathies. A better molecular understanding of the pathogenetic mechanisms is essential for addressing new strategies in diagnostics, therapeutics and clinical management of chaperonopathies, including those in which Hsp10 is involved. This chaperonin has been studied for a long time as a member of the mitochondrial protein-folding machine. However, although in normal cells Hsp10 is mainly localized in the mitochondrial matrix, it has also been found during and after stress in other subcellular compartments, such as cytosol, vesicles and secretory granules, alone or in combination with other proteins. In these extramitochondrial locales, Hsp10 plays an active role in cell signalling. For example, cancer cells often show altered levels of Hsp10, compared to normal cells. Hsp10 may also be found in the extracellular space and in the bloodstream, with a possible immunomodulatory activity. This minireview focuses on some studies to date on the involvement of Hsp10 in human disease pathogenesis.

Apoptosis is considered an important mechanism of selective deletion that occurs during hematopoiesis. Myelolipoma is a rare benign tumor composed of adipose tissue and hematopoietic cells. The pathogenesis of this benign tumor is still unclear. Analysing the structural levels and apoptosis of normal human bone marrow (NHBM) and human myelolipoma (HM), the apoptotic events resulted abundantly present in NHBM compared to HM, which showed a small number of apoptotic cells. By contrast, Fas expression was strongly present both in NHBM and HM. These findings suggest that an altered function of Fas in myelolipoma is not able to trigger the apoptotic machinery. In conclusion, we hypothesize that drastic reduction of apoptosis in myelolipoma can be considered one of the growth regulatory mechanisms.

This review addresses the significance of the expression of proliferating cell nuclear antigen (PCNA), p53 and some heat shock proteins (Hsps) in prostate carcinoma (PC). In fact, PCNA and p53 are two widely discussed tools in PC diagno- sis, mainly because of the controversy regarding the signifi- cance of their expression during prostate cancer development and progression. At the same time, only few studies have shown the potential role of Hsps in carcinogenesis and their overexpression in pre-neoplastic and neoplastic lesions of the prostate. We briefly describe the physiological roles of Hsps in normal cells, and the significance of their immunohistochem- ical detection in PC as well as in pre-cancerous lesions of the prostate. We will also discuss the possible functional interac- tions of these molecules in both dysplastic and neoplastic cells

In order to increase knowledge on the morphology and structure of the articular disc of the temporomandibular joint (TMJ) for a better understanding of the functional role of the same, it proceeded with an investigation on histological samples in the block of TMJ and periarticular tissues of adult rabits and human fatustes at different stage of development. The limited and inadequate availability of organs from human donors has resulted in the utilisation of xenografts as an alternative tool. Nevertheless, hyperacute rejection (HAR) following xenograft determines the loss of the transplanted organ. The “primum movens” is the activation of the complement pathway mediated by the binding of natural xenogenic antibodies to the endothelium of the graft, followed by the lysis of the endothelial cells with subsequent oedema, thrombosis and necrosis of the transplanted organ. In this work we describe morphological and biomolecular observations of isolated human-decay accelerating factor (h-DAF, CD55) transgenic pig hearts, after perfusion for four hours with human blood. H-DAF is a membrane glycoprotein inhibiting the complement activation in humans. We describe considerably reduced damages in transgenic hearts, compared to controls. The cardiac function resulted preserved. Our data are in agreement with what was already shown by other groups using different experimental models. In conclusion, we encourage the use of new sources of transgenic animals, pointing out the importance of morphological analysis in evaluation of xenograft.

Probiotics (PB) are living microorganisms that act as a commensal population in normal intestines and confer numerous beneficial effects on the host. The introduction of probiotics in the treatment of inflammatory bowel disease (IBD) prolongs remission. The aim of this study was to investigate the intestinal and hepatic effects of PB supplementation in an experimental IBD model in mice induced by 2,4,6-trinitrobenzene sulfonic acid (TNBS). In the first step of the experimental procedure, CD-1 male mice, 5 to 6 weeks old, were randomly divided into 3 groups and inoculated intrarectally with, respectively, saline, alcohol, or TNBS to assess the experimental IBD model. In the second step, mice treated, or not, with TNBS inoculation, were treated with PB (Lactobacillus Casei, Bifidobacterum Lactis) for 1, 2 or 3 weeks, on a daily basis. Large bowel (colon and rectum) and liver were processed for histological alterations, according to a scoring system. Large bowel was also assessed for apoptosis by TUNEL assay. TNBS induced, as expected, severe damage and inflammation in the large bowel, including nuclear alterations and apoptosis, and, to a lesser extent, to the liver. Administration of PB determined significant reduction of both histological alterations and apoptosis. PB administration in advance protects from inflammation. In conclusion, supplementation with Lactobacillus casei, Bifidobacterum lactis PB is able to ameliorate the colitis by reversing the histological changes caused by TNBS in mice. Experimentation in human subjects in needed to prove their efficacy in reducing histological alterations that may be present in subjects with IBD.

Background: The study of the expression of different biological markers in non-neoplastic, pre-neoplastic and neoplastic lesions of prostate could help to better understand their role in carcinogenesis and to find new diagnostic and prognostic tools. Materials and Methods: In the present work we evaluated, by immunohistochemistry, the presence and the expression of PCNA, p53, HSP60, HSP10 and MUC-2 in a series of nodular hyperplasia, low- and high-grade prostatic intraepithelial lesions and adenocarcinomas. Results: Our data confirmed that: 1) PCNA expression could be related to the grade of progression of cancer; and that 2) p53 mutation could be a late event in prostate carcinogenesis. Moreover, we reported that: 1) HPS60 and HPS10 were overexpressed early in prostate carcinogenesis; and that 2) MUC-2 is absent in both tumoral and non-tumoral prostatic tissue. Conclusion: We suggest the further examination, by molecular and genetic studies, of the role of HSP60 and HSP10 during carcinogenesis of the prostate as well as of other organs.

Heat shock proteins (HSPs) constitute a heterogeneous family of proteins involved in cell homeostasis. During cell life they are involved in harmful insults, as well as in immune and inflammatory reactions. It is known that they regulate gene expression, and cell proliferation, differentiation and death. HSP60 is a mitochondrial chaperonin, highly preserved during evolution, responsible of protein folding. Its function is strictly dependent on HSP10 in both prokaryotic and eukaryotic elements. We investigated the presence and the expression of HSP60 and HSP10 in a series of 20 normal human bone marrow specimens (NHBM) by the means of immunohistochemistry. NHBM showed no expression of HSP60, probably due to its being below the detectable threshold, as already demonstrated in other normal human tissues. By contrast, HSP10 showed a selective positivity for myeloid and megakaryocytic lineages. The positivity was restricted to precursor cells, while mature elements were constantly negative. We postulate that HSP10 plays a role in bone marrow cell differentiation other than being a mitochondrial co-chaperonin. The present data emphasize the role of HSP10 during cellular homeostasis and encourage further investigations in this field.

Hashimoto’s thyroiditis (HT) is a chronic autoimmune disease resulting from Fas-mediated thyrocyte destruction. Although autocrine/paracrine Fas-Fas ligand (FasL) interaction is responsible for thyrocyte cell death during the active phases of HT, the role of infiltrating T lymphocytes (ITL) in this process is still unknown. Therefore, we investigated the expression and function of Fas and FasL in ITL. All ITL expressed high levels of Fas and CD69, an early marker of T cell activation associated with functional Fas expression in T cells in vivo. In contrast to thyrocytes that were found to produce high levels of FasL, ITL did not express significant amounts of FasL, suggesting that ITL are not directly involved in thyrocyte destruction. The analysis of ITL purified from HT thyroids showed that ITL were massively killed by Fas crosslinking and that a considerable number (24–36%) underwent spontaneous apoptosis within 36 h of culture. Accordingly, in situ TUNEL (terminal deoxynucleotidyl transferase dUTP nick end labeling) staining revealed that a significant number (10–15%) of ITL in proximity to FasL-producing thyroid follicles were apoptotic. Moreover, virtually all ITL in proximity to thyroid follicles were preapoptotic, as they expressed high levels of GD3 ganglioside, a killer glycolipid responsible for the generation of irreversible apoptotic signals that accumulate in hematopoietic cells shortly after Fas crosslinking. These data demonstrate that ITL are not directly involved in thyrocyte cell death during HT, suggesting that autocrine/paracrine Fas-FasL interaction is a major mechanism in autoimmune thyrocyte destruction.

During embryonic development, a pool of cells may become a reserve of undifferentiated cells, the embryo-stolen adult stem cells (ESASC). ESASC may be responsible for adult tissue homeostasis, as well as disease development. Transdifferentiation is a sort of reprogramming of ESASC from one germ layer-derived tissue towards another. Transdifferentiation has been described to take place from mesoderm to ectodermal- or endodermal-derived tissues and viceversa but not from ectodermal- to endodermal-derived tissues. We hypothesise that two different populations of ESASC could exist, the first ecto/mesoblast-committed and the second endo/mesoblast-committed. If confirmed, this hypothesis could lead to new studies on the molecular mechanisms of cell differentiation and to a better understanding of the pathogenesis of a number of diseases.

The interaction of Fas with FasL has been demonstrated to be implicated in the pathogenesis of several autoimmune and liver diseases. Recently, attention has been focused on the hypothesis that thyrocytes and beta cells undergo massive Fas/FasL-mediated apoptosis during autoimmune response. Similarly, hepatocyte cell death occurring following copper accumulation points towards the same mechanism.

The Channichthyidae or ''icefish'' represent an intriguing example of extreme adaptation to the stable low temperature and high oxygen content of the Antarctic waters. The lack of respiratory pigments (hemoglobin and myoglobin) in these teleosts is associated with relatively low oxygen consumption and relevant. cardio-circulatory adjustments which include large blood volume, increased relative heart weight (cardiomegaly), and very high cardiac output. The heart has the ability to displace large systolic volumes at a low rate and relatively low pressure, with large ventricular fillings (high ventricular compliance), whereas it is incapable effacing increased afterloads. These functional aspects of mechanical flexibility and restrictions of the cardiac pump have been tentatively related to some constructional aspects of the icefish cardiomegaly, particularly, at the whole ventricular level, to the trabeculate type of myo-architecture, and, at the subcellular level, to the conflict in space economy between the exceptionally high mitochondrial densities and the consequent severe reduction in myofibrillar volume. On the basis of this morphodynamic approach, we suggest that the icefish may illustrate how a certain feature (i.e., an architectural cardiac design) common to the suborder and to most teleosts, and apparently with ''irrelevant'' properties, can become useful for a specialized purpose (i.e., volume pump design); and how, in contrast, the internal machinery construction. because of structural and ultrastructural constraints, may pre;ent these stenothermal sedentary teleosts from conquering niches requiring more active locomotory habits.

This study focuses the aging-related modification of skeletal fiber types in two skeletal muscles of different-age swine (6 and 18 month). Rectus abdominis and vastus medialis were employed. It was performed an immunohistochemical staining for slow fibers and it was made a quantitative evaluation, using an automatic interactive image analysis system. The percentage of slow fibers decreased in adult swine. Moreover, slow fibers in rectus abdominis were less numerous than in vastus medialis. Aging and muscle function are two important factors able to modify fiber types. Morphometric analyses can ascertain this modification for diagnostic or nourishmental purposes.

The purpose of this study was to verify the difference between diameters of abdominal aorta obtained on corpses and on living bodies, using C.T. The Authors focused the attention on abdominal aorta, beneath kidney veins. Moreover, a linear relationship was found between height and diameter of abdominal aorta. A careful examination of the literature has shown discordance among the values of the abdominal aorta. The Authors conclude that improved techniques of radiological anatomy may offer an important support to obtain important data for clinical practice.

In the last few years, a major goal of cardiac research has been to drive stem cell differentiation to replace damaged myocardium. Several research groups have attempted to differentiate potential cardiac stem cells (CSCs) using bi- or three-dimensional systems supplemented with growth factors or molecules acting as differentiating substances. We hypothesize that these systems failed to induce a complete differentiation because they lacked an architectural space. In the present study, we isolated a pool of small proliferating and fibroblast-like cells from adult rat myocardium. The phenotype of these cells was assessed and the characterized cells were cultured in a collagen I/OPLA scaffold with horse serum to obtain fine myocardial differentiation. C-Kit(POS)/Sca-1(POS) CSCs fully differentiated in vitro when an environment more similar to the CSC niche was created. These experiments demonstrated an important model for the study of the biology of CSCs and the biochemical pathways that lead to myocardial differentiation. The results pave the way for a new surgical approach.

This study aimed to analyze the two-phase heat transfer in a vertical upward flow configuration under different gravity conditions. The facility is a new version of MicroBo (Microgravity Boiling), the working fluid used is perfluorohexane C6F14 and a 4.0 mm aluminum channel is arranged vertically to perform ebullition process. The platform used to get data in microgravity was the parabolic flight, which represents one of the most widely used platforms despite the period of time available to collect data is very short. The analysis of the results has been carried out following two main approaches: the first, ground-flight data comparison, for the study of variable gravity data collected during the 67th ESA campaign of parabolic flight held in November 2017, while the second, ground data analysis, to analyze the data collected on the ground in the ENEA laboratory with the same facility in December 2018. In particular ground – flight data have been analyzed comparing boiling curves at different flow rates, from a minimum of 5.3 l/h up to 17 l/h, in micro, hyper and normal gravity. The operating inlet pressure during experiments has been fixed at 1.6 bar with a heat flux range of 5.4 - 85.6 kW/m2. For what regard ground data boiling curves, with a flow rate of 10 l/h, are shown with a direct flow pattern visualization. The operating pressure has been fixed at 1.3 bar and 1.8 bar with a heat flux range of 2.5 – 94.6 kW/m2. For the latter analysis, it also has been carried out a qualitative study for the validation of the void fraction model, starting from the visualization of the images of flow patterns recorded during the acquisitions.
