This article presents the results of an ongoing inventory of Ascomycota in Yunnan, China, carried out as part of the research project series "Exploring ascomycete diversity in Yunnan". From over 100 samples collected from diverse host substrates, microfungi have been isolated, identified and are currently being documented. The primary objective of this research is to promote the discovery of novel taxa and explore the ascomycete diversity in the region, utilising a morphology-phylogeny approach. This article represents the second series of species descriptions for the project and introduces three undocumented species found in the families Bambusicolaceae, Dictyosporiaceae and Periconiaceae, belonging to the suborder Massarineae (Pleosporales, Dothideomycetes). These novel taxa exhibit typical morphological characteristics of Bambusicola, Periconia and Trichobotrys, leading to their designation as Bambusicola hongheensis, Periconia kunmingensis and Trichobotrys sinensis. Comprehensive multigene phylogenetic analyses were conducted to validate the novelty of these species. The results revealed well-defined clades that are clearly distinct from other related species, providing robust support for their placement within their respective families. Notably, this study unveils the phylogenetic affinity of Trichobotrys within Dictyosporiaceae for the first time. Additionally, the synanamorphism for the genus Trichobotrys is also reported for the first time. Detailed descriptions, illustrations and updated phylogenies of the novel species are provided, and thus presenting a valuable resource for researchers and mycologists interested in the diversity of ascomycetes in Yunnan. Endophytic microorganisms are those that inhabit the interior of plant tissues and organs without causing damage to the plant, triggering a mutualistic interaction. These endophytes are known to produce compounds with various applications in the areas of biotechnology, pharmaceutical industry and agriculture; therefore, molecular methods are used to investigate the phylogeny of these organisms. The Brazilian Cerrado is the second largest biome in Brazil and is considered a hotspot, with a high diversity of endemic fauna and flora. The objective of this work was to analyze, through in silico analysis, the phylogeny of cultivable endophytic fungi isolated from plants found in the Brazilian Cerrado biome. A search was carried out for articles that worked with endophytes in the period between 2012 and 2022. The dendrogram was generated using the Neighbor-joining method based on the ITS1-5.8S-ITS2 conserved region obtained by GenBank codes of isolated endophytes. The genera Xylaria, Diaporthe, and Colletotrichum were isolated in more than three plants in the seven articles found. Most of the articles found related to the biodiversity of endophytic fungi from the Brazilian Cerrado aim at the bioprospecting of bioactive compounds, through culture-dependent methods; as such, a part of endophytic diversity may be lost due to the inability of certain endophytes to grow in artificial media. In silico analysis can assist in the investigation of phylogenetic relationships between endophytic fungi and has the potential to guide future work aimed at prospecting for bioactive compounds, phylogenetic identification, or biodiversity of this group of endophytes. The Mediterranean Sea stands out as a hotspot of biodiversity, whose fungal composition remains underexplored. Marine sediments represent the most diverse substrate; however, the challenge of recovering fungi in culture hinders the precise identification of this diversity. Concentration techniques like skimmed milk flocculation (SMF) could represent a suitable solution. Here, we compare the effectiveness in recovering filamentous ascomycetes of direct plating and SMF in combination with three culture media and two incubation temperatures, and we describe the fungal diversity detected in marine sediments. Sediments were collected at different depths on two beaches (Miracle and Arrabassada) on the Spanish western Mediterranean coast between 2021 and 2022. We recovered 362 strains, and after a morphological selection, 188 were identified primarily with the LSU and ITS barcodes, representing 54 genera and 94 species. Aspergillus, Penicillium, and Scedosporium were the most common genera, with different percentages of abundance between both beaches. Arrabassada Beach was more heterogeneous, with 42 genera representing 60 species (Miracle Beach, 28 genera and 54 species). Although most species were recovered with direct plating (70 species), 20 species were exclusively obtained using SMF as a sample pre-treatment, improving our ability to detect fungi in culture. In addition, we propose three new species in the genera Exophiala, Nigrocephalum, and Queenslandipenidiella, and a fourth representing the novel genus Schizochlamydosporiella. We concluded that SMF is a useful technique that, in combination with direct plating, including different culture media and incubation temperatures, improves the chance of recovering marine fungal communities in culture-dependent studies. The checklist serves as an informative method for evaluating the diversity, geography, and ecology of established and reproducing macrofungi. Additionally, considering macrofungi as bioindicator species, their census should be incorporated into efforts to monitor the state of health of ecosystems and directly applied to conservation policies. Between 2019 and 2023, a census of macrofungal species was conducted in Taburno-Camposauro Regional Park (Campania, Italy) across nine distinct habitats. A total of 453 fungal taxa were identified, including several new records for the Campania region. The fungal diversity exhibited significant variations based on the dominant plant species in each habitat. Fagacean tree species and Carpinus spp. shared similar fungal communities. Equally, coniferous tree species displayed a comparable fungal composition. In Abies alba and mixed broad-leaved forests, low levels of ectomycorrhizal taxa were observed alongside a concurrent increase in saprotrophs, indicating a disturbed habitat and a reduction in the Gadgil effect. Notably, lower fungal diversity was documented in the grassland habitat, suggesting the potential implications of wildlife imbalance and excessive grazing. Ramalina farinacea is a widely distributed epiphytic lichen from the Macaronesian archipelagos to Mediterranean and Boreal Europe. Previous studies have indicated a specific association between R. farinacea and Trebouxia microalgae species. Here, we examined the symbiotic interactions in this lichen and its closest allies (the so-called "R. farinacea group") across ten biogeographic subregions, spanning diverse macroclimates, analyzing the climatic niche of the primary phycobionts, and discussing the specificity of these associations across the studied area. The most common phycobionts in the "R. farinacea group" were T. jamesii and T. lynnae, which showed a preference for continentality and insularity, respectively. The Canarian endemic R. alisiosae associated exclusively with T. lynnae, while the other Ramalina mycobionts interacted with both microalgae. The two phycobionts exhibited extensive niche overlap in an area encompassing Mediterranean, temperate Europe, and Macaronesian localities. However, T. jamesii occurred in more diverse climate types, whereas T. lynnae preferred warmer and more humid climates, often close to the sea, which could be related to its tolerance to salinity. With the geographical perspective gained in this study, it was possible to show how the association with different phycobionts may shape the ecological adaptation of lichen symbioses. Rock-inhabiting fungi (RIF) are usually extremely tolerant or extremophilic, as they can survive on natural and artificial rocks despite being exposed to stressful conditions. RIF have serious negative effects on the appearance and cohesion of rocky substrates, causing the alteration and decomposition of building materials, but also on human and animal health, as they can act as opportunistic pathogens. Their identification is therefore of great importance, especially in urban areas. In the present study, culturing techniques for isolating fungi, and a polyphasic taxonomic approach to their identification, were used to assess the diversity of micromycetes that darken the surfaces of buildings in various villages and cities in Tarragona Province (Spain). Sixty-four species of RIF belonging to forty-one genera were identified, including a new genus (Coccodomyces) and the following six new fungal species: Coccodomyces pleiosporus, Exophiala caementiphila, Exophiala multiformis, Neocatenulostroma spinulosum, Neodevriesia longicatenispora, and Paradevriesia holothallica. Xylaria is a widely distributed genus in the Ascomycota phylum that can decompose wood. It is an essential decomposer in ecosystems and a source of bioactive secondary metabolites. Based on morphological characteristics and molecular evidence, this article thoroughly describes two new species discovered on the fallen leaves in Hainan Tropical Rainforest National Park, along with illustrations and comparisons with similar species. Xylaria diaoluoshanensis is characterized by filamentous stromata with long infertile apexes, ascospores sometimes with non-cellular appendages. Xylaria fulvotomentosa differentiates itself from other Xylaria species that grow on fallen leaves by its stroma surface, being yellow tomentose. These two new species of the genus Xylaria were found by phylogenetic analysis using the ITS-β-tubulin-RPB2 sequence dataset. Furthermore, a species first discovered in China, X. petchii, is described. Finally, a search table for 44 species related to fallen leaves and petioles in the world is established. During a survey of fungi on Wurfbainia villosa in Guangdong Province, China, three novel species, Periconia endophytica, P. yangjiangensis, and P. wurfbainiae, belonging to Periconiaceae in Pleosporales, Dothideomycetes are proposed based on morphological and phylogenetic evidence. Periconia endophytica was isolated from the healthy leaves of W. villosa, while P. yangjiangensis and P. wurfbainiae were obtained from the dead stems of the same host. Notably, holomorphs were observed in P. wurfbainiae. The morphological characteristics of the novel taxa are compared with closely related species within Periconia. Illustrations, morphological descriptions, and phylogenetic analyses are provided for the novel taxa. Multilocus phylogeny of the combined internal transcribed spacer (ITS), large subunit nuclear rDNA (LSU), small subunit nuclear ribosomal rDNA (SSU), and partial translation elongation factor 1–α (tef1-α) regions supported the establishment of three new species. Furthermore, the taxa clustering in Periconia, Flavomyces fulophazii, and Sporidesmium tengii, are discussed for further investigation of their taxonomic placements. [ABSTRACT FROM AUTHOR]

Sarcoscypha (Sarcoscyphaceae, Pezizales) is a saprobic fungus characterized by the cup or disc-shaped blight red apothecium and oblong to ellipsoid ascospores. The 18 species of Sarcoscypha were known to occur in Europe, North America, and tropical Asia. However, up to date, only two Sarcoscypha species have been reported in Korea. In this study, novel Sarcoscypha specimens were collected from Juwangsan, Odaesan, and Taebaeksan National Parks from September to October in Korea. This species is well distinguished from other Sarcoscypha species according to the molecular and phylogenetic analysis based on internal transcribed spacer (ITS) region. Even though fungi are ubiquitous in the biosphere, the ecological knowledge of marine fungi remains rather rudimentary. Also, little is known about their tolerance to salinity and how it influences their activities. Extracellular enzymatic activities (EEAs) are widely used to determine heterotrophic microbes' enzymatic capabilities and substrate preferences. Five marine fungal species belonging to the most abundant pelagic phyla (Ascomycota and Basidiomycota) were grown under non-saline and saline conditions (0 g/L and 35 g/L, respectively). Due to their sensitivity and specificity, fluorogenic substrate analogues were used to determine hydrolytic activity on carbohydrates (β-glucosidase, β-xylosidase, and N-acetyl-β-D-glucosaminidase); peptides (leucine aminopeptidase and trypsin); lipids (lipase); organic phosphorus (alkaline phosphatase), and sulfur compounds (sulfatase). Afterwards, kinetic parameters such as maximum velocity (Vmax) and half-saturation constant (Km) were calculated. All fungal species investigated cleaved these substrates, but some species were more efficient than others. Moreover, most enzymatic activities were reduced in the saline medium, with some exceptions like sulfatase. In non-saline conditions, the average Vmax ranged between 208.5 to 0.02 μmol/g biomass/h, and in saline conditions, 88.4 to 0.02 μmol/g biomass/h. The average Km ranged between 1553.2 and 0.02 μM with no clear influence of salinity. Taken together, our results highlight a potential tolerance of marine fungi to freshwater conditions and indicate that changes in salinity (due to freshwater input or evaporation) might impact their enzymatic activities spectrum and, therefore, their contribution to the oceanic elemental cycles. The fungal genus Nigrospora is known to be a plant pathogen, endophyte, and saprobe, and it is usually isolated from various substrates like soil and air. During the surveys of soil fungi in Hebei Province of China, two isolates of Nigrospora were obtained. A multi-locus phylogeny of combined loci of the 5.8S nuclear ribosomal gene with the two flanking transcribed spacers (ITS), part of the translation elongation factor 1-alpha (tef1), and the beta-tubulin (tub2) loci, in conjunction with morphological characters were used to identify the newly collected isolates. Nigrospora humicola sp. Nov. is described and proposed herein, which differs from its phylogenetically close species N. chinensis and N. globosa by the sequences of ITS, tef1, and tub2. Bromo Tengger Semeru National Park (TNBTS) is a conservation area in East Java with an abundance of biodiversity, including macrofungi. This study aims to identify the macrofungal species existing in Poncokusumo district, TNBTS area, and their habitat characteristics for a further sustainable study of fungi. This study used a purposive sampling method by opportunistic exploration. Identification of macrofungi is conducted by morphological analysis and habitat characterization. The study identified 15 macrofungal species categorized as Ascomycota and Basidiomycota, that were distinct in their habitat characteristics. The 15 macrofungal species inhabited leaf litter, wood litter, soil, and bamboo.

Abstract Orchids comprise one of the largest, most diverse, and most broadly distributed families of flowering plants and contribute significantly to habitat biodiversity. One key aspect of orchid growth and development is the formation of mycorrhizal symbioses with compatible endophytic fungi, which are maintained throughout the life of the plant. Substantial efforts to identify the fungi that form mycorrhizal symbioses across a range of orchid species have often also uncovered numerous nonmycorrhizal, endophytic fungi. These fungi could also have significant effects on orchid growth and development and are beginning to be analyzed more closely, particularly in wild species. The role of endophytic fungi in the production, distribution, and continued growth by the hobbyist of orchids is not known. As an initial step toward characterizing nonmycorrhizal endophytic fungi associated with cultivated orchids, we undertook a survey of fungi residing within roots of Phalaenopsis plants growing in home environments. Sequence analysis of ITS regions amplified from total DNA isolated from roots allowed rapid identification of endophytic fungi to the class level and may offer a useful initial screening method for beneficial species, for example, in horticultural settings. ITS‐PCR sequences subsequently obtained from individual fungi cultured from surface‐sterilized orchid roots corroborated the findings of the initial screen, while also providing a more complete characterization of the array of fungal taxa that were present. Although lower in diversity than has been reported for orchids growing in the wild, these endophytes have the potential to substantially enhance the growth and disease resistance of horticultural orchids.

Se encontraron árboles enfermos en plantaciones de Tectona grandis en la provincia de Guanacaste, Costa Rica, que mostraban síntomas de pudrición del cuello y la raíz. A nivel morfológico y molecular, se identificó la especie Kretzschmaria zonata como el agente causal de la enfermedad. El micelio del patógeno creció de forma circuncéntrica en un medio PDA, con coloración gris en el centro y blanca en los bordes. Se registraron ascosporas de color café oscuro, cuyo tamaño era (21.6-) 27.4 – 32.1 (-33.5) x (6.2-) 8.7 – 10.9 (-11.8) µm. Las plantas inoculadas con el patógeno presentaron lesiones, con diferencias altamente significativas respecto a las plantas testigo. Se realizaron reaislamientos y se recuperó el hongo K. zonata, cumpliendo con los postulados de Koch. Es la primera vez que se reporta este patógeno causando enfermedad en plantaciones forestales de teca en Costa Rica, lo que constituye un avance para el estudio de estrategias de manejo de la enfermedad.

The interaction between stingless bees and fungi has garnered recent interest due to potential mutual benefits. In Brazil, Melipona scutellaris stands out ecologically and economically. We investigated its microbiota, focusing on yeast species in pot-honey, pot-pollen, and nest surfaces. Samples from the Atlantic Forest (Pernambuco state, Brazil) were analyzed through morphological and physiological methods and D1/D2 LSU rDNA sequence analysis. We identified 20 yeast species, comprising 15 ascomycetes and five basidiomycetes, representing 11 and five genera, respectively. All yeast species, except Blastobotrys meliponae, were reported for the first time in association with M. scutellaris. Honey exhibited the highest species richness. In this study, the species Blastobotrys meliponae, Meyerozyma caribbica, Wickerhamomyces anomalus, and Moniliella carnis were exclusively isolated from honey. The understanding of the ecological association between stingless bees and yeasts, as revealed in our research, can significantly aid in insect conservation programs. We present the first report of yeasts associated with M. scutellaris substrates living in the Brazilian Atlantic Forest.

Muscadinia rotundifolia cv. Trayshed is a valuable source of resistance to grape powdery mildew. It carries 2 powdery mildew resistance-associated genetic loci, Run1.2 on chromosome 12 and Run2.2 on chromosome 18. The purpose of this study was to identify candidate resistance genes associated with each haplotype of the 2 loci. Both haplotypes of each resistance-associated locus were identified, phased, and reconstructed. Haplotype phasing allowed the identification of several structural variation events between haplotypes of both loci. Combined with a manual refinement of the gene models, we found that the heterozygous structural variants affected the gene content, with some resulting in duplicated or hemizygous nucleotide-binding leucine-rich repeat genes. Heterozygous structural variations were also found to impact the domain composition of some nucleotide-binding leucine-rich repeat proteins. By comparing the nucleotide-binding leucine-rich repeat proteins at Run1.2 and Run2.2 loci, we discovered that the 2 loci include different numbers and classes of nucleotide-binding leucine-rich repeat genes. To identify powdery mildew resistance-associated genes, we performed a gene expression profiling of the nucleotide-binding leucine-rich repeat genes at Run1.2b and Run2.2 loci with or without powdery mildew present. Several nucleotide-binding leucine-rich repeat genes were constitutively expressed, suggesting a role in powdery mildew resistance. These first complete, haplotype-resolved resistance-associated loci and the candidate nucleotide-binding leucine-rich repeat genes identified by this study are new resources that can aid the development of powdery mildew-resistant grape cultivars.

Members of the fungal genus Phyllosticta can colonize a variety of plant hosts, including several Citrus species such as Citrus sinensis (orange), Citrus limon (lemon), and Citrus maxima (pomelo). Some Phyllosticta species have the capacity to cause disease, such as Citrus Black Spot, while others have only been observed as endophytes. Thus far, genomic differences underlying lifestyle adaptations of Phyllosticta species have not yet been studied. Furthermore, the lifestyle of Phyllosticta citrichinaensis is ambiguous, as it has been described as a weak pathogen but Koch's postulates may not have been established and the presence of this species was never reported to cause any crop or economic losses. Here, we examined the genomic differences between pathogenic and endophytic Phyllosticta spp. colonizing Citrus and specifically aimed to elucidate the lifestyle of Phyllosticta citrichinaensis. We found several genomic differences between species of different lifestyles, including groups of genes that were only present in pathogens or endophytes. We also observed that species, based on their carbohydrate active enzymes, group independent of their phylogenetic association, and this clustering correlated with trophy prediction. Phyllosticta citrichinaensis shows an intermediate lifestyle, sharing genomic and phenotypic attributes of both pathogens and endophytes. We thus present the first genomic comparison of multiple citrus-colonizing pathogens and endophytes of the genus Phyllosticta, and therefore provide the basis for further comparative studies into the lifestyle adaptations within this genus.

Profiling the host-mycobiota interactions in healthy vs. diseased forest ecosystems helps understand the dynamics of understudied yet increasingly important threats to forest health that are emerging due to climate change. We analyzed the structural and functional changes of the mycobiota and the responses of Pinus contorta in the Lophodermella needle cast pathosystem through metabarcoding and metatranscriptomics. When needles transitioned from asymptomatic to symptomatic, dysbiosis of the mycobiota occurred, but with an enrichment of Lophodermella pathogens. Many pathogenicity-related genes were highly expressed by the mycobiota at the necrotrophic phase, showing an active pathogen response that are absent in asymptomatic needles. This study also revealed that Lophodermella spp. are members of a healthy needle mycobiota that have latent lifestyles suggesting that other pine needle pathogens may have similar biology. Interestingly, Pinus contorta upregulated defense genes in healthy needles, indicating response to fungal recognition, while a variety of biotic and abiotic stresses genes were activated in diseased needles. Further investigation to elucidate the possible antagonistic interplay of other biotic members leading to disease progression and/or suppression is warranted. This study provides insights into microbial interactions in non-model pathosystems and contributes to the development of new forest management strategies against emerging latent pathogens.

Aim. The results of the inventory of species composition of epiphytic lichens and aphyllophoroid fungi on two 1 ha sample plots in lowland floodplain forests are presented. Such integrated studies of these groups of organisms are poorly known. Fungi, including lichenized fungi, are essential components of forest ecosystems. Data on their diversity and substrate distribution can provide important information on the condition and biological value of the forests studied. Material and Methods. The main method of the fieldwork was the "1‐ha method". For the alpha‐diversity study, two 1‐ha sample plots were established in well‐preserved forest areas: the first one in a site of mixed broad‐leaved forest with lianas, and the second one in communities dominated by Carpinus betulus and Quercus robur. The specimens were collected from all variety of woody substrate within plots in spring and autumn 2023. Results. We have revealed 89 species of lichens and 60 species of aphyllophoroid fungi. The lichen families Arthoniaceae, Lecanoraceae, Physciaceae, Ramalinaceae and Roccellaceae contain 47.2 % of all species. High proportion of lichens with Trentepohlia photobiont (29.2 %), significant number of Arthoniomycetes (Arthoniaceae, Lecanographaceae, Roccellaceae) and crustose lichens (75.3 %) have been noted. Among the aphyllophoroid fungi identified in the sample plots, 11 species are new to Dagestan, including three species (Antrodia leucaena, Coronicium gemmiferum, Steccherinum litschaueri) recorded for the Caucasus for the first time. Fourteen species are new to the Samursky National Park. Conclusion. The study of alpha diversity and substrate preferences of xylobionts revealed the richest and most specific phorophytes, as well as some ecological features of the surveyed plots. Most of the species were found on the bark and wood of Carpinus betulus, Populus alba and Quercus robur. The majority of species reported for the first time for the region have been recorded on these tree species. We expand the number of known species on Populus alba, on which only few species were previously known. High proportion of lichens with the Trentepohlia photobiont and the predominance of crustose species were revealed. In terms of basidiomata morphology, corticioid fungi dominated over polypores and clavarioids in general. The leading ecological and trophic group of aphyllophoroid fungi are saprotrophs. Revealed features indicate a significant contribution of the wood of the main forest‐forming tree species to the preservation of the species richness of myco ‐and lichen biota.

جنس Myxotrichum از جمله قارچ‌های آسکومیست در راسته Helotiales و رده Leotiomycetes است که زیستگاه معمول گونه‌های آن، لایه‌های هوموس در جنگل‌ها و تعدادی نیز از خاک، چوب، مواد سلولزی و سایر مواد پوسیده گزارش شده‌اند. بیشتر گونه‌های این جنس در دمای پایین رشد کرده و به ‌صورت پوده‌رست در تجزیه مواد گیاهی نقش مؤثری ایفا می‌کنند. تاکنون 20 گونه معتبر از این جنس در جهان گزارش و تایید شده ولی در ایران تا کنون مطالعه‌ای در مورد جنس مذکور صورت نگرفته و گزارشی از این جنس و گونه‌های آن وجود ندارد. طی بازدیدهای دوره‌ای از جنگل‌های بلوط در استان کرمانشاه، برگ‌های پوسیده درختان جمع‌‌آوری و با کشت آن‌ها یک آرایه قارچی با مشخصات زیر به دست آمد. پرگنه روی محیط کشت عصاره سیب زمینی-دکستروز-آگار رشد بسیار کمی داشت و پس از چهار هفته در دمای ۲۵ درجه سلسیوس به ۲۰ میلی‌متر رسید. پرگنه‌ در ابتدا سفید رنگ و با گذشت زمان به قهوه‌ای متمایل به قرمز تغییر یافت. رنگ پشت پرگنه قرمز شرابی بود. رنگدانه‌های صورتی تا قرمز نیز در محیط کشت تولید شد. آسکوکارپ‌ها کروی و با زواید قهوه‌ای تیره و منشعب پوشیده شده بودند. انشعابات زواید راست و تعدادی نیز برگشته و وارونه و انتهای آن‌ها تا حدودی شفاف بودند. آسک‌ها شفاف، دارای هشت آسکوسپور، کروی تا نیمه‌کروی و به ابعاد 15/12-7/2 × 12/8-10/7 میکرومتر بودند. آسکوسپورها شفاف، بیضوی و به ابعاد 4/4-3/3 × 3-2 میکرومتر بودند. مرحله میتوسپوری قارچ در محیط کشت مشاهده نشد. توصیف این جنس با توصیف سایر محققان در مورد جنس Myxotrichum و گونه M. deflexum Berk منطبق بود. به علاوه، هویت گونه مذکور، با استفاده از توالی‌یابی ناحیه نسخه‌برداری شده داخلی دی.ان.ای. ریبوزومی و واکاوی فیلوژنتیکی انجام شد.

The new combination Thalloidima squamatum is proposed for a morphologically and phylogenetically distinct species, until now included within the variation of T. sedifolium . Compared to T. sedifolium , the species is characterized by its larger squamules and apothecia, shorter ascospores, the pale color of the excipulum, and its habitat growing on moss cushions over calcareous rock or directly on rock rather than on soil. Most of the material studied originates from Norway and Sweden, but specimens are also reported from Austria, Croatia, Germany, Greenland, Italy, Poland, Russia, Scotland and Switzerland.

The current list of fungi from Colombia updated in the present review contains a total of 7619 species. The Ascomycota appears as the most diverse group, with 4818 species, followed by the Basidiomycota, with 2555 species. Despite this, we presume that the actual fungal diversity in Colombia could amount to between 105,600 and 300,000 species. Fungi represent an underestimated resource, indispensable for human well-being. Even though the current knowledge on potential applications of Colombian fungi is still limited, the number of studies on areas such as natural products discovery, biological control, and food and beverages, among other biotechnological applications, are increasing. With the current review, we aim to present a comprehensive update on the fungal diversity in Colombia and its potential applications. Colombia’s native fungal biodiversity holds much potential within the country’s current social-economical context, and the future must ensure efforts to preserve both the biodiversity and the untapped resources of the fungi in Colombia, which in alignment with the Sustainable Development Goals (SDGs) might result in new bioeconomy avenues for the country.

AbstractNigrospora (Xylariales, Apiosporaceae) consists of species of terrestrial plant endophytes and pathogens. Nigrospora has also been reported in marine environments such as mangroves, sea fans, and macroalgae. However, limited research has been conducted on Nigrospora associated with macroalgae. Here, we isolated Nigrospora species from three types of algae (brown, green, and red algae) from Korean islands (Chuja, Jeju, and Ulleung) based on phylogenetic analyses of multigenetic markers: the internal transcribed spacers (ITS), beta-tubulin (BenA), and translation elongation factor 1 (TEF1-α). A total of 17 Nigrospora strains were isolated from macroalgae and identified as nine distinct species. The majority of Nigrospora species (seven) were found on brown algae, followed by red algae (three), and then green algae (two). To our understanding, this study represents the first account of N. cooperae, N. covidalis, N. guilinensis, N. lacticolonia, N. osmanthi, N. pyriformis, and N. rubi occurring in marine environments. Additionally, this study provides the first report of the occurrence of N. cooperae, N. covidalis, N. guilinensis, N. lacticolonia, and N. osmanthi in South Korea. This study will provide valuable insights for future research exploring the functions of fungi in macroalgal communities.

Cladosporium fulvum is a fungal pathogen that causes leaf mould of tomato. The reference genome of this pathogen was released in 2012 but its high repetitive DNA content prevented a contiguous assembly and further prohibited the analysis of its genome architecture. In this study, we combined third generation sequencing technology with the Hi-C chromatin conformation capture technique, to produce a high-quality and near complete genome assembly and gene annotation of a Race 5 isolate of C. fulvum. The resulting genome assembly contained 67.17 Mb organized into 14 chromosomes (Chr1-to-Chr14), all of which were assembled telomere-to-telomere. The smallest of the chromosomes, Chr14, is only 460 kb in size and contains 25 genes that all encode hypothetical proteins. Notably, PCR assays revealed that Chr14 was absent in 19 out of 24 isolates of a world-wide collection of C. fulvum, indicating that Chr14 is dispensable. Thus, C. fulvum is currently the second species of Capnodiales shown to harbour dispensable chromosomes. The genome of C. fulvum Race 5 is 49.7 % repetitive and contains 14 690 predicted genes with an estimated completeness of 98.9%, currently one of the highest among the Capnodiales. Genome structure analysis revealed a compartmentalized architecture composed of gene-dense and repeat-poor regions interspersed with gene-sparse and repeat-rich regions. Nearly 39.2 % of the C. fulvum Race 5 genome is affected by Repeat-Induced Point (RIP) mutations and evidence of RIP leakage toward non-repetitive regions was observed in all chromosomes, indicating the RIP plays an important role in the evolution of this pathogen. Finally, 345 genes encoding candidate effectors were identified in C. fulvum Race 5, with a significant enrichment of their location in gene-sparse regions, in accordance with the 'two-speed genome' model of evolution. Overall, the new reference genome of C. fulvum presents several notable features and is a valuable resource for studies in plant pathogens.

Although secondary metabolites are typically associated with competitive or pathogenic interactions, the high bioactivity of endophytic fungi in the Xylariales, coupled with their abundance and broad host ranges spanning all lineages of land plants and lichens, suggests that enhanced secondary metabolism might facilitate symbioses with phylogenetically diverse hosts. Here, we examined secondary metabolite gene clusters (SMGCs) across 96 Xylariales genomes in two clades (Xylariaceae s.l. and Hypoxylaceae), including 88 newly sequenced genomes of endophytes and closely related saprotrophs and pathogens. We paired genomic data with extensive metadata on endophyte hosts and substrates, enabling us to examine genomic factors related to the breadth of symbiotic interactions and ecological roles. All genomes contain hyperabundant SMGCs; however, Xylariaceae have increased numbers of gene duplications, horizontal gene transfers (HGTs) and SMGCs. Enhanced metabolic diversity of endophytes is associated with a greater diversity of hosts and increased capacity for lignocellulose decomposition. Our results suggest that, as host and substrate generalists, Xylariaceae endophytes experience greater selection to diversify SMGCs compared with more ecologically specialised Hypoxylaceae species. Overall, our results provide new evidence that SMGCs may facilitate symbiosis with phylogenetically diverse hosts, highlighting the importance of microbial symbioses to drive fungal metabolic diversity.

We studied the occurrence of endophytic fungi and bacteria in the roots of F. gigantea, a woodland perennial grass common throughout Europe and temperate Asia. The taxonomic assignment was carried out according to the isolate’s colony and cytological species morphotype characteristics and confirmed by the assessment of the standard DNA sequences, ITS, RPB2, SSU, and TEF1-a for fungi and 16S rDNA for bacteria. Our study has shown that F. gigantea roots are the habitat to a wide range of fungi and bacteria. The occurrence of fungal structures was determined in ~40% of the roots examined by Trypan Blue staining. In a surface-sterile root-cutting culture on PDA medium, we obtained isolates of six endophytic fungi species: four members of Ascomycota—Alternaria alternata, Cadophora fastigiata, Chaetomium funicola, and Microdochium bolleyi—and two of Basidiomycota—Coprinellus sp. and Sistotrema brinkmannii. In addition, we report bacteria co-occurring endophytically in the roots of this grass. The Firmicutes group was the most prevalent, consisting of four Gram-positive, endospore-forming bacteria taxa. The isolates were identified as Bacillus pumilus, Bacillus sp., Lysinibacillus sp., and Priestia aryabhattai. Moreover, two Gram-negative bacteria were detected—Kosakonia sp. (Proteobacteria) and Pedobacter sp. (Bacteroidetes). Thus, applying the isolate-culture approach, we identified a set of microorganisms in the roots of a typical grass native to the deciduous forest floor. The functional roles of these endophytes are diverse, and many of them, saprotrophs and decomposers of wood and plant debris, are linked to the decomposition of organic matter. This is the first detailed report on fungal and bacterial endophytes inhabiting the roots of F. gigantea. This study fills in a research gap on endophytes associated with the below-ground parts of Festuca spp., hitherto extensively studied for Epichloë/Neotyphodium associations in their foliar parts.

Disease results from interactions among the host, pathogen, and environment. Inoculation trials can quantify interactions among these players and explain aspects of disease ecology to inform management in variable and dynamic natural environments. White-nose Syndrome, a disease caused by the fungal pathogen, Pseudogymnoascus destructans (Pd), has caused severe population declines of several bat species in North America. We conducted the first experimental infection trial on the tri-colored bat, Perimyotis subflavus, to test the effect of temperature and humidity on disease severity. We also tested the effects of temperature and humidity on fungal growth and persistence on substrates. Unexpectedly, only 37% (35/95) of bats experimentally inoculated with Pd at the start of the experiment showed any infection response or disease symptoms after 83 days of captive hibernation. There was no evidence that temperature or humidity influenced infection response. Temperature had a strong effect on fungal growth on media plates, but the influence of humidity was more variable and uncertain. Designing laboratory studies to maximize research outcomes would be beneficial given the high costs of such efforts and potential for unexpected outcomes. Understanding the influence of microclimates on host-pathogen interactions remains an important consideration for managing wildlife diseases, particularly in variable environments.

Verticillium dahliae is a widespread soilborne fungus that causes Verticillium wilt on numerous economically important plant species. In tomato, until now, three races have been characterized based on the response of differential cultivars to V. dahliae, but the genetic basis of race divergence in V. dahliae remains undetermined. To investigate the genetic basis of race divergence, we sequenced the genomes of two race 2 strains and four race 3 strains for comparative analyses with two known race 1 genomes. The genetic basis of race divergence was described by the pathogenicity-related genes among the three races, orthologue analyses, and genomic structural variations. Global comparative genomics showed that chromosomal rearrangements are not the only source of race divergence and that race 3 should be split into two genotypes based on orthologue clustering. Lineage-specific regions (LSRs), frequently observed between genomes of the three races, encode several predicted secreted proteins that potentially function as suppressors of immunity triggered by known effectors. These likely contribute to the virulence of the three races. Two genes in particular that can act as markers for race 2 and race 3 (VdR2e and VdR3e, respectively) contribute to virulence on tomato, and the latter acts as an avirulence factor of race 3. We elucidated the genetic basis of race divergence through global comparative genomics and identified secreted proteins in LSRs that could potentially play critical roles in the differential virulence among the races in V. dahliae. IMPORTANCE Deciphering the gene-for-gene relationships during host-pathogen interactions is the basis of modern plant resistance breeding. In the Verticillium dahliae-tomato pathosystem, two races (races 1 and 2) and their corresponding avirulence (Avr) genes have been identified, but strains that lack these two Avr genes exist in nature. In this system, race 3 has been described, but the corresponding Avr gene has not been identified. We de novo-sequenced genomes of six strains and identified secreted proteins within the lineage-specific regions (LSRs) distributed among the genomes of the three races that could potentially function as manipulators of host immunity. One of the LSR genes, VdR3e, was confirmed as the Avr gene for race 3. The results indicate that differences in transcriptional regulation may contribute to race differentiation. This is the first study to describe these differences and elucidate roles of secreted proteins in LSRs that play roles in race differentiation.

Powdery mildews comprise a large group of economically important phytopathogenic fungi. However, limited information exists on their mitochondrial genomes. Here, we assembled and compared the mitochondrial genomes of the powdery mildew pathogens Blumeria graminis f. sp. tritici, Erysiphe pisi, and Golovinomyces cichoracearum. Included in the comparative analysis was also the mitochondrial genome of Erysiphe necator that was previously analysed. The mitochondrial genomes of the four Erysiphales exhibit a similar gene content and organization but a large variation in size, with sizes ranging from 109800 bp in B. graminis f. sp. tritici to 332165 bp in G. cichoracearum, which is the largest mitochondrial genome of a fungal pathogen reported to date. Further comparative analysis revealed an unusual bimodal GC distribution in the mitochondrial genomes of B. graminis f. sp. tritici and G. cichoracearum that was not previously observed in fungi. The cytochrome b (cob) genes of E. necator, E. pisi, and G. cichoracearum were also exceptionally rich in introns, which in turn harboured rare open reading frames encoding reverse transcriptases that were likely acquired horizontally. Golovinomyces cichoracearum had also the longest cob gene (45 kb) among 703 fungal cob genes analysed. Collectively, these results provide novel insights into the organization of mitochondrial genomes of powdery mildew pathogens and represent valuable resources for population genetics and evolutionary studies.

BackgroundPlant pathogens and their hosts undergo adaptive changes in managed agricultural ecosystems, by overcoming host resistance, but the underlying genetic adaptations are difficult to determine in natural settings. Verticillium dahliae is a fungal pathogen that causes Verticillium wilt on many economically important crops including lettuce. We assessed the dynamics of changes in the V. dahliae genome under selection in a long-term field experiment.ResultsIn this study, a field was fumigated before the Verticillium dahliae race 1 strain (VdLs.16) was introduced. A derivative 145-strain population was collected over a 6-year period from this field in which a seggregating population of lettuce derived from Vr1/vr1 parents were evaluated. We de novo sequenced the parental genome of VdLs.16 strain and resequenced the derivative strains to analyze the genetic variations that accumulate over time in the field cropped with lettuce. Population genomics analyses identified 2769 single-nucleotide polymorphisms (SNPs) and 750 insertion/deletions (In-Dels) in the 145 isolates compared with the parental genome. Sequence divergence was identified in the coding sequence regions of 378 genes and in the putative promoter regions of 604 genes. Five-hundred and nine SNPs/In-Dels were identified as fixed. The SNPs and In-Dels were significantly enriched in the transposon-rich, gene-sparse regions, and in those genes with functional roles in signaling and transcriptional regulation.ConclusionsUnder the managed ecosystem continuously cropped to lettuce, the local adaptation of V. dahliae evolves at a whole genome scale to accumulate SNPs/In-Dels nonrandomly in hypervariable regions that encode components of signal transduction and transcriptional regulation.

Structural biology has the potential to illuminate the evolution of pathogen effectors and their commonalities that cannot be readily detected at the primary sequence level. Recent breakthroughs in protein structure modeling have demonstrated the feasibility to predict the protein folds without depending on homologous templates. These advances enabled a genome-wide computational structural biology approach to help understand proteins based on their predicted folds. In this study, we employed structure prediction methods on the secretome of the destructive fungal pathogen Magnaporthe oryzae. Out of 1,854 secreted proteins, we predicted the folds of 1,295 proteins (70%). We showed that template-free modeling by TrRosetta captured 514 folds missed by homology modeling, including many known effectors and virulence factors, and that TrRosetta generally produced higher quality models for secreted proteins. Along with sensitive homology search, we employed structure-based clustering, defining not only homologous groups with divergent members but also sequence-unrelated structurally analogous groups. We demonstrate that this approach can reveal new putative members of structurally similar MAX effectors and novel analogous effector families present in M. oryzae and possibly in other phytopathogens. We also investigated the evolution of expanded putative ADP-ribose transferases with predicted structures. We suggest that the loss of catalytic activities of the enzymes might have led them to new evolutionary trajectories to be specialized as protein binders. Collectively, we propose that computational structural genomics approaches can be an integral part of studying effector biology and provide valuable resources that were inaccessible before the advent of machine learning-based structure prediction.[Formula: see text] Copyright © 2021 The Author(s). Ectomycorrhizal (ECM) fungi associated with plants constitute one of the most successful symbiotic interactions in forest ecosystems. ECM support trophic exchanges with host plants and are important factors for the survival and stress resilience of trees. However, ECM clades often harbour morpho-species and cryptic lineages, with weak morphological differentiation. How this relates to intraspecific genome variability and ecological functioning is poorly known. Here, we analysed 16 European isolates of the ascomycete Cenococcum geophilum, an extremely ubiquitous forest symbiotic fungus with no known sexual or asexual spore-forming structures but with a massively enlarged genome. We carried out whole-genome sequencing to identify single-nucleotide polymorphisms. We found no geographic structure at the European scale but divergent lineages within sampling sites. Evidence for recombination was restricted to specific cryptic lineages. Lineage differentiation was supported by extensive copy-number variation. Finally, we confirmed heterothallism with a single MAT1 idiomorph per genome. Synteny analyses of the MAT1 locus revealed substantial rearrangements and a pseudogene of the opposite MAT1 idiomorph. Our study provides the first evidence for substantial genome-wide structural variation, lineage-specific recombination and low continent-wide genetic differentiation in C. geophilum. Our study provides a foundation for targeted analyses of intra-specific functional variation in this major symbiosis.

Abstract Background The genus Camillea was created in 1849 from collections made in French Guiana with eight species included. Numerous species assigned to Camillea were subsequently discovered, especially in the forests of the Amazon basin, but new discoveries have not been reported from French Guiana since 1849. Recent fieldwork in French Guiana has begun to fill this gap by identifying five new species, most of which were collected in the vicinity of Saül village. Results Based on macro- and micromorphological study of their stromata, including SEM images of ascospore wall ornamentation, five new species were recognized, including C. cribellum, C. heterostomoides, C. nitida, C. rogersii and C. saulensis. Cultures could be obtained for C. heterostomoides and C. rogersii, and ITS and LSU sequences were obtained for all of the five new species. Camillea heterostoma and its variety microspora were shown to be conspecific. Provisional molecular phylogenetic analyses support the possible reinstatement of Hypoxylon melanaspis, currently regarded as merely an applanate form of C. leprieurii. Conclusion The current study is based on a relatively limited fieldwork in its duration and sampling area but was able to substantially increase the number of Camillea species known from French Guiana. This augurs an exceptional and still unknown diversity of the genus in this area and by extension in the adjacent neotropical forests.

Foliar fungal endophytes are known to alter plant physiology but the mechanisms by which they do so remain poorly understood. We focused on how plant gene expression was altered by six fungal strains that generated “water-saver” and “water-spender” drought physiologies in a C4 grass, Panicum hallii. Water-saver physiologies have lower plant water loss, improved wilt resistance, and higher survival compared with water-spender strategies. We expected that fungi within each functional group would have similar effects on P. hallii, and this was largely true for plant physiology but not for plant gene expression. When we focused only on genes that were differentially expressed relative to fungus-free controls, we found surprisingly little overlap in plant differentially expressed genes or gene regulatory pathways across the fungal treatments, including within and between the water-saver and water-spender strategies. Nevertheless, using lasso regression, we identified a small subset of genes that predicted 39 and 53% of the variation in plant wilt resistance and water loss, respectively. These results suggest that fungal effects on plant transcription may identify how they extend the plant phenotype, and the comparison across multiple fungi allows us to differentiate broadly fungal-responsive plant genes versus those plant genes that respond only to single fungal taxa. The genes identified here could be targeted for future study to understand their function and, ultimately, represent candidates for precision breeding efforts to increase plant drought tolerance. Prescribed fire frequencies have been widely used to reduce the risk of severe wildfire occurrences. In addition, several studies have been conducted to assess the impact of fire frequencies on vegetation, vertebrate, and invertebrate species, as well as soil physical and chemical properties. However, there is a lack of empirically based knowledge concerning the impact of fire frequency on soil microorganisms. This study assessed the effect of different fire frequencies on the diversity and composition of soil fungal communities in a semi-arid savanna rangeland. Soil samples were collected from an ongoing long-term trial at the University of Fort Hare (South Africa) on the following treatments: (i) no burning; (ii) annual burning (burned once every year); (iii) biennial burning (burned once every 2 years); (iv) triennial burning (burned once every 3 years); (v) quadrennial burning (burned once every 4 years); and (vi) sexennial burning (burned once every 6 years). Fungi were identified using high-throughput sequencing, with Shannon-Wiener and Inverse Simpson diversity indexes being used for diversity and network analysis. Principal coordinate analysis was used for Bray-Curtis distance matrices to visualise the relationships between treatments. The highest diversity was found in biennial burning, which was significantly different (p < 0.05) from the sexennial, quadrennial, and no burning treatments but was not different from the triennial and annual burning treatments. Regarding the taxa, Ascomycota and Basidiomycota were the phyla with the highest relative abundance, followed by Mortierellomycota, Chytridiomycota, and Rozellomycota. The different fire frequencies had an influence on soil fungi diversity and taxonomic composition in semi-arid savanna rangelands.

Seventeen mushroom species from fourteen genera were collected and identified from various locations, such as: Bardarash, Barzan, Erbil city center, Hiran, Mergasor, Shaqlawa, Khalefan, Tawska and Taqtaq. The following genus and species have been identified: Agaricus bisporus, A. campestris, A. incultorum, Coprinus comatus, Lycoperdon umbrinum, Amanita virosa, Panaeolus semiovatus, Pleurotus ostreatus, Omphalina pyxidate, O. rosella, Phallus impudicus, Fomitopsis betulina, Ganoderma applanatum, Daedaleopsis confragosa, lactarius vellereus, Helvella leucopus and Bisporella citrina. Seven genera (8 species) of mushrooms were discovered for the first timein kurdistan region-Iraq, such as: Lycoperdon umbrinum, Amanita virosa, Omphalina pyxidate, O. rosella, Phallus impudicus, Daedaleopsis confragosa, Helvella leucopus and Bisporella citrina. The goal of this research is to investigate and identify wild mushrooms that naturally occur in the Erbil Governorate, Kurdistan Region of Iraq, in various areas and during various seasons.

Two novel Comoclathris species were identified from dicotyledonous plants (Clematis sp. and Xanthoceras sorbifolium) in China. The results were supported by morphological characters and Maximum Likelihood (ML) and Bayesian Inference (BI) analyses. Multi-gene phylogenetic analyses of the ITS, LSU, SSU and rpb2 sequences revealed two new species Comoclathris clematidis and C. xanthoceratis, which are phylogenetically distinct. The new species are phylogenetically closely related to C. arrhenatheri. However, they are distinguishable from C. arrhenatheri by having comparatively larger asci and ascospores. This study improves our knowledge of Comoclathris as no species has been previously described from China. This suggests such taxa may be rare and it is likely that new taxa will be discovered from hosts and environments that have not yet been extensively investigated. [ABSTRACT FROM AUTHOR]

Here we report morphological observations and phylogenetic analysis of Skyttella mulleri, a lichenicolous fungus newly recorded from Japan. It forms white to pale yellow apothecia abundantly on the thallus of Peltigera sp. The apothecia are formed on the upper cortex of the host thallus, become translucent when wet, and lack an exciple. Phylogenetic analysis of the large subunit of the nuclear ribosomal RNA gene suggested that this fungus belongs to Cordieritidaceae {Helotiales). [ABSTRACT FROM AUTHOR]

New records of bryoparasitic fungi from Styria are discussed and partly illustrated by macro- and microscopic photographs, and their host bryophytes are given. The following species are reported: Arrhenia lobata, Bryocentria brongniartii, B. metzgeriae, Bryoscyphus rhytidiadelphi, Lamprospora benkertii, L. dicranellae, L. esterlechnerae, L. retispora, L. sylvatica, L. tortulae-ruralis, L. tuberculatella, Mniaecia albida, M. jungermanniae, Neottiella ricciae, Octospora affinis, O. bicarpa, O. erzbergeri, O. guestfaliensis, O. gyalectoides agg., O. hygrohypnophila, O. leucoloma var. leucoloma, O. lilacina, O. mnii, O. musci-muralis var. neglecta, O. phagospora, Octosporella erythrostigma, O. jungermanniarum and O. ornithocephala. Three new host bryophytes are given: Lewinskya speciosa for O. affinis, Schistidium trichodon for O. musci-muralis var. neglecta and Syntrichia norvegica for L. retispora. 15 taxa are reported for the first time for Austria and two taxa are reported for the first time for Styria. The prominent problem of continuous cropping obstacle has been frustrating the morel farming. To deepen the understanding on morel continuous cropping obstacle, the allelopathic effects of phenolic acid extracts from morel continuous cropping soils on growth and development of Morchella sextelata, M. eximia, M. importuna, pathogenic fungus Fusarium sp. and soil-dominant fungus Chaetomium sp. were investigated. These effects were expressed as response index (RI). Under actual content of phenolic acids (6.150 μg/g fresh mixed continuous cropping soil), the mycelial growth of all tested morel strains was inhibited (RI < 0), while the allelopathic effect of control phenolic acids (4.252 μg/g fresh mixed control soil) was between promotion and inhibition, which suggested that the phenolic acid extracts from morel continuous cropping soils may exhibit certain extent of autotoxicity for the existence of morel-specific allelochemicals. In addition, the aggravated pigmentation and reduced occurrence of sclerotium in three Morchella fungi at growth inhibitory concentrations of phenolic acids indicated the induction of morel strain aging. Meanwhile, most concentrations of phenolic acids showed stimulatory effects on sporulation of Fusarium sp. and Chaetomium sp. (RI > 0), manifesting the enrichment of soil-borne pathogenic fungi and dominance of certain fungal population in soil ecosystem. Collectively, the allelopathic effects of phenolic acid extracts play an instrumental role in morel continuous cropping obstacle. Apiospora is widely distributed throughout the world, and most of its hosts are Poaceae. In this study, Arthrinium-like strains were isolated from non-Poaceae in the Hainan and Fujian provinces of China. Based on the combined DNA sequence data of the internal transcriptional spacer (ITS), partial large subunit nuclear rDNA (LSU), translation extension factor 1-α gene (TEF1-α) and β-tubulin (TUB2), the collected Apiospora specimens were compared with known species, and three new species were identified. This study investigates the phylogeny and taxonomy of Anthostomella-like fungi (Xylariales, Sordariomycetes) found in association with bamboo in Japan. Four new genera, Amphigermslita (including three new species, i.e., A. deformis, A. fusiformis, and A. pseudofusiformis), monotypic Crassipseudostroma (C. phyllostachydis) and Minuticlypeus (M. discosporus), and Pallidoperidium (two new species, P. exasperatum and P. paraexasperatum), and one known genus, Nigropunctata (one new species, N. complanata) are recognized and described. These five genera were found to constitute a distinct monophyletic lineage based on molecular phylogenetic analyses utilizing sequences of ITS and LSU nrDNA, rpb2, and tef1-a sequences. A new family, Pallidoperidiaceae, is proposed to accommodate these bambusicolous Anthostomella-like fungi. The identification of this lineage contributes to our understanding of the evolutionary relationships and classification of these bambusicolous fungi. Seventeen mushroom species from fourteen genera were collected and identified from various locations, such as: Bardarash, Barzan, Erbil city center, Hiran, Mergasor, Shaqlawa, Khalefan, Tawska and Taqtaq. The following genus and species have been identified: Agaricus bisporus, A. campestris, A. incultorum, Coprinus comatus, Lycoperdon umbrinum, Amanita virosa, Panaeolus semiovatus, Pleurotus ostreatus, Omphalina pyxidate, O. rosella, Phallus impudicus, Fomitopsis betulina, Ganoderma applanatum, Daedaleopsis confragosa, lactarius vellereus, Helvella leucopus and Bisporella citrina. Seven genera (8 species) of mushrooms were discovered for the first timein kurdistan region-Iraq, such as: Lycoperdon umbrinum, Amanita virosa, Omphalina pyxidate, O. rosella, Phallus impudicus, Daedaleopsis confragosa, Helvella leucopus and Bisporella citrina. As the continuation of Fungal Diversity Notes series, the current paper is the 16th contribution to this series. A total of 103 taxa from seven classes in Ascomycota and Basidiomycota are included here. Of these 101 taxa, four new genera, 89 new species, one new combination, one new name and six new records are described in detail along with information of hosts and geographic distributions. Corresponding to morphological characters, phylogenetic evidence is also utilized to place the above-mentioned taxa in appropriate taxonomic positions. The current morphological and phylogenetic data is helpful for further clarification of species diversity and exploration of evolutionary relationships in the related fungal groups. Hypoxylaceous fungi are abundant in China, but their discovery and report are uneven in various provinces, with more fungi in Yunnan and Hainan and fewer fungi in Tibet. During the investigation of macro-fungi in Motuo county, Tibet Autonomous Region, we collected a number of xylarialean specimens. Six hypoxylaceous specimens growing on dead angiosperm were collected from the forests of Motuo county, and they were described and illustrated as two new species in Hypoxylon based on a combination of morphological characters and molecular evidence. Hypoxylon diperithecium was characterized by its bistratal perithecia, purple-brown stromatal granules, citrine to rust KOH-extractable pigments, and light brown to brown ascospores ellipsoid-inequilateral with conspicuous coil-like ornamentation. Hypoxylon tibeticum was distinct from other species by having pulvinate and applanate stromata, surface vinaceous, with orange granules, orange KOH-extractable pigments, and brown ascospores with inconspicuous ornamentation. The multi-gene phylogenetic analyses (ITS-LSU-RPB2-TUB) supported the two new taxa as separate lineages in the genus Hypoxylon. A key to all known Hypoxylon taxa from China is provided. Macrofungi play a diverse role in ecosystems, serving as decomposers, sources of food, and providers of medicinal benefits to humans for millennia. However, the status of macrofungi diversity in Perlis State Park is poorly understood. This study aims to identify the diversity of macrofungi species in Perlis State Park. Sampling was conducted using both opportunistic and selective approaches along a transect line comprising 10 trails during the rainy season between 2019 and 2023. Identification was based on the morphological characteristics of macrofungi. A total of 69 species of macrofungi were discovered, including 59 species from Basidiomycota and 10 species from Ascomycota. These species can be categorized into eight groups, namely Cup (2), Gill (20), Shelf and Bracket (16), Bolete (7), Coral (6), Jelly (6), Earthball and Puffball (4), and Tooth (4) fungi. Of the identified macrofungi species, 33 are confirmed to be edible. Additionally, three species from the Cordycipitaceae and Ophiocordycipitaceae families within the suborders of Ascomycota are entomopathogenic. Some microfungi fall under the category of 'Least Concern (LC)' according to the IUCN Red List. Plectania rhytidia is an uncommon discomycete now detected in Brazil. It is characterised by having a black ascomata, and transversally ribbed on one side ascospores measuring 19.6–27.4 (–29.4) × (6.9–) 7.8–12.3 μm. This species has been briefly referred to from Rio Grande do Sul without a detailed description and discussion. Studies on the genus Plectania in this country are scarce, and only P. chilensis, P.camplyspora, and P. milleri were cited. Previously, members of the genus have been mostly reported from temperate regions of the northern hemisphere, and in Australia and New Zealand. In this report, an interesting species of Plectania, found for the first time in the State of Paraiba, Brazil, is presented and comprehensively described. Nigrospora (Xylariales, Apiosporaceae) consists of species of terrestrial plant endophytes and pathogens. Nigrospora has also been reported in marine environments such as mangroves, sea fans, and macroalgae. However, limited research has been conducted on Nigrospora associated with macroalgae. Here, we isolated Nigrospora species from three types of algae (brown, green, and red algae) from Korean islands (Chuja, Jeju, and Ulleung) based on phylogenetic analyses of multigenetic markers: the internal transcribed spacers (ITS), beta-tubulin (BenA), and translation elongation factor 1 (TEF1-α). A total of 17 Nigrospora strains were isolated from macroalgae and identified as nine distinct species. The majority of Nigrospora species (seven) were found on brown algae, followed by red algae (three), and then green algae (two). To our understanding, this study represents the first account of N. cooperae, N. covidalis, N. guilinensis, N. lacticolonia, N. osmanthi, N. pyriformis, and N. rubi occurring in marine environments. Additionally, this study provides the first report of the occurrence of N. cooperae, N. covidalis, N. guilinensis, N. lacticolonia, and N. osmanthi in South Korea.
