Within its historic urban areas, the city has implemented innovative practices and strategies to revitalize its tangible and intangible heritage. This paper delves into the crucial issue of heritage building reuse, aiming to strike a balance between their "specimen" and "living" values. To define the success of reuse projects, this study compares evaluation criteria and parameters from both domestic and international contexts. The paper then focuses on examining the exemplary reuse practices in Suzhou, leading to the identification and summarization of the "Suzhou reuse pathway." Specifically, value construction serves as a case study to assess the impact of heritage reuse through the analysis of public feedback. This examination underscores the effectiveness of Suzhou's pathway, positioning it as a positive reference in the global discourse on architectural heritage conservation and sustainable development in World Heritage Cities. In the context of burgeoning interest in entrepreneurship among recent Chinese college graduates, the disappointingly low success rate of startups has prompted a critical inquiry. This study endeavors to delve into the factors shaping entrepreneurial success, aiming to illuminate pathways for addressing this pervasive issue. Employing the csQCA method, we analyze 23 cases of successful entrepreneurship, revealing a nuanced and intricate interplay of factors that challenges simplistic narratives in prior research. The analysis unveils five distinct pathways to success: “Environmental Incentive,” “Motivation,” “Platform Assistance,” “Policy Support,” and “Demand.” Notably, the first two pathways significantly shape pre-graduation success, while the latter three become prominent in the post-graduation phase. We also find that entrepreneurship education and self-fulfillment emerge as key elements leading to success before graduation, while social relationships and self-fulfillment play pivotal roles after graduation. Furthermore, technology emerges as the predominant field drawing the interest of startups, followed by culture and academia. Inflammation is a normal immune response in organisms, but it often triggers chronic diseases such as colitis and arthritis. Currently, the most widely used antiinflammatory drugs are non-steroidal anti-inflammatory drugs, albeit they are accompanied by various adverse effects such as hypertension and renal dysfunction. Bioactive peptides (BAPs) provide therapeutic benefits for inflammation and mitigate side effects. Herein, this review focuses on the therapeutic effects of various BAPs on inflammation in different body parts. Emphasis is placed on the immunomodulatory mechanisms of BAPs in treating inflammation, such as regulating the release of inflammatory mediators, modulating MAPK and NF-κB signaling pathways, and reducing oxidative stress reactions for immunomodulation. Simple Summary: Ulva prolifera is an important species with a large market value. This species thrives at 10–20 °C but struggles at lower temperatures. Research has demonstrated that environmental stresses, such as temperature and light intensity, have a substantial impact on its growth through changing pathways and gene expression. Our present study examines the processes by which U. prolifera responds to combined high light intensity/temperature challenges, and the results point to the involvement of several genes and pathways in these intricate adaptive mechanisms. Gaining an understanding of these systems is essential to enhancing the efficiency and sustainability of macroalgae farming, particularly in light of climate change. Our research focused on understanding the genetic mechanisms that contribute to the tolerance of Ulva prolifera (Chlorophyta), a marine macroalgae, to the combined stress of high temperature and high light intensity. At the mRNA level, the up-regulated DEGs showed enrichment in pathways related to ribosomes, proteasomes, and peroxisomes. The spliceosome pathway genes were found to be vital for U. prolifera's ability to adapt to various challenging situations in all the comparison groups. In response to elevated temperature and light intensity stress, there was a significant increase in genes and pathways related to ribosomes, proteasomes, and peroxisomes, whereas autophagy showed an increase in response to stress after 24 h, but not after 48 h. Stenotrophomonas maltophilia (S. maltophilia) is a spoilage microorganism widely distributed in vegetables, dairy products, and various environments and can cause serious foodborne illnesses. Renowned for its environmentally friendly, safe, and non-toxic properties, rose essential oil (REO) is widely recognized as an exceptional natural antibacterial agent. In the present study, we conducted a comprehensive analysis to elucidate the inhibition mechanism of REO against S. maltophilia at both cellular and metabolic levels. The results demonstrated that REO treatment of cells resulted in the disruption of cell structure and leakage of cellular contents. Remarkable changes in the morphology of bacterial cells were observed under both scanning electron microscopy and transmission electron microscopy. At the metabolic level, multiple metabolic pathways were disrupted after REO treatment, including amino acid metabolism, nucleotide metabolism, and aminoacyl tRNA biosynthesis. These results shed new light to the inhibition mechanism of REO against S. maltophilia and provide a theoretical basis for the development of effective essential oil preservatives. Cardiovascular diseases (CVD) represent a significant global health challenge resulting from a complex interplay of genetic, environmental, and lifestyle factors. However, the molecular pathways and genetic factors involved in the onset and progression of CVDs remain incompletely understood. Here, we performed an integrative bioinformatic analysis to highlight specific genes and signaling pathways implicated in the pathogenesis of 80 CVDs. Differentially expressed genes (DEGs) were identified through the integrated analysis of microarray and GWAS datasets. Then, hub genes were identified after gene ontology functional annotation analysis and protein-protein internet (PPI) analysis. In addition, pathways were identified through KEGG and gene ontology enrichment analyses. A total of 821 hub genes related to 80 CVDs were identified, including 135 common and frequent CVD-associated genes. TNF, IL6, VEGFA, and TGFB.1 genes were the central core genes expressed in 50% or more of CVDs, confirming that the inflammation is a key pathological feature of CVDs. Analysis of hub genes by KEGG enrichment revealed predominant enrichment in 201 KEGG pathways, of which the AGE-RAGE signaling pathway in diabetic complications was identified as the common key KEGG implicated in 62 CVDs. In addition, the outcomes showed an overrepresentation in pathways categorized under human diseases, particularly in the subcategories of infectious diseases and cancers, which may be common risk factors for CVDs. In conclusion, this powerful approach for in silico fine-mapping of genes and pathways allowed the identification of determinant hubs genes and pathways implicated in the pathogenesis of CVDs which could be employed in developing more targeted and effective interventions for preventing, diagnosing, and treating CVDs. Metabolism is a network of chemical reactions that sustain cellular life. Parts of this metabolic network are defined as metabolic pathways containing specific biochemical reactions. Products and reactants of these reactions are called metabolites, which are associated with certain human-defined metabolic pathways. Metabolic knowledgebases, such as the Kyoto Encyclopedia of Gene and Genomes (KEGG) contain metabolites, reactions, and pathway annotations; however, such resources are incomplete due to current limits of metabolic knowledge. To fill in missing metabolite pathway annotations, past machine learning models showed some success at predicting the KEGG Level 2 pathway category involvement of metabolites based on their chemical structure. Here, we present the first machine learning model to predict metabolite association to more granular KEGG Level 3 metabolic pathways. We used a feature and dataset engineering approach to generate over one million metabolite-pathway entries in the dataset used to train a single binary classifier. This approach produced a mean Matthews correlation coefficient (MCC) of 0.806 ± 0.017 SD across 100 cross-validation iterations. The 172 Level 3 pathways were predicted with an overall MCC of 0.726. Moreover, metabolite association with the 12 Level 2 pathway categories was predicted with an overall MCC of 0.891, representing significant transfer learning from the Level 3 pathway entries. Women face numerous gender-based barriers that hinder their access to resources, nutritious foods, nutrition services, and maternity entitlements. Evidence shows that certain types of women’s groups can improve women’s access to resources and social capital and in some approaches also improve health and nutrition outcomes. Women’s rights-based organizations in South Asia have a longstanding tradition of collective action toward gender equality. Women’s rights-based organizations work in areas such as microfinance, livelihoods, women’s rights, health, and combating violence against women. In this perspective article, we explore how women’s groups and rights-based organizations can leverage their collective strength to advance nutrition outcomes for women and children. We identify seven pathways implemented through women’s groups toward improving nutrition outcomes. These pathways include (i) income generation, (ii) agriculture, (iii) health and nutrition behavior change communication and participatory learning and action, (iv) advocating for rights to better health and social services, (v) food access, (vi) cash transfers, and (vii) strengthening service delivery and fostering convergence with health systems. We also note that women’s groups have the potential to implement integrated interventions through combined food-systems-rights pathways. Investing in this area can support transforming nutrition policy from a service delivery model to a rights-based approach.

This paper offers a feminist reflection on the findings of an ongoing study of health and wellbeing associated with urban food security in Nairobi and Cape Town. It offers five pathways through which a strengthened informal sector can contribute to transforming urban food systems: identifying stronger entry points for institutionalized collaboration between local governments and community-based organizations; enhancing government capacities to collaborate with grassroots actors; the potential, and challenges, of controlled-environment agriculture; rethinking and shifting the regulatory environment surrounding the informal economy; and responding to detailed new data on the state of food security in South African informal settlements. Conducted in partnerships between the Urban Food Futures program and people who were permanently displaced during the colonial era, the study is informed by an emerging body of analysis that responds to the complex trauma of undernutrition, and approaches health as something far beyond an individual’s own somatic/bodily state of being. Participation in the study enabled communities living in a near-permanent state of precarity and food insecurity, in the absence of culturally appropriate and readily-available supports to mental health, to move beyond an isolated focus on food to explore feelings of psychic safety and security, and also environmental wellbeing, including access to clean air and water, decent and affordable housing, safe and dignified work, and freedom from violence of all forms.

Academic research plays a pivotal role in creating and sustaining pathways for clinical impact. Over nine years, a series of collaborative research projects informed the translation pathway for structured journal clubs within a tertiary health service. This pathway included conceptualisation, implementation, sustainability and spread of research evidence into practice within the innovative journal club initiative. This brief identifies four key stages of the pathway, summarising the contribution of research to support ongoing practice transformation.

Africa has recently experienced adverse climate changes and has recognized tremendous advancement of numerous Climate Smart Agriculture (CSA) technologies with ability to promote resilience and productivity. We have created several high protein (HPBL) and low protein breeding lines (LPBL) by crossing between NC-Raleigh and PI407228. The NC-Raleigh is a low protein cultivar widely grown in North Carolina with excellent yield potential and a high protein wild soybean (Glycine soja) with low yield. We analyzed the F6 generation of this cross for protein and oil compositions using near-infrared spectroscopy (NIR). Two soybean lines HPBL and LPBL were selected for biochemical analysis using mass spectrometry-based proteomic and metabolomic techniques. We identified 4538 proteins and 267 metabolites in these lines. The HPBL showed the enrichment of proteins associated with protein synthesis, processing, and storage in addition to higher content of primary and secondary metabolites. The mapping of metabolites on the global metabolic pathways revealed the increased protein content in the HPBL that may have resulted from increased arginine content routed from citrulline-argininosuccinic acid. This information will be useful for breeders and biotechnologists to produce a value-added soybean trait.

Introduction: Taraxasterol is a pentacyclic triterpenoid which was first discovered from the roots of Taraxacum officinale. However, it is generally found in the Taraxacum genus (Pugongying in Chinese). It has been reported to possess several medicinal properties, and modulate several biochemical and metabolic signaling pathways as a means of exerting its pharmacological effects. Even though some biological effects have been ascribed to taraxasterol, it is still being explored for more therapeutic benefits. This review aimed to highlight the relevance of taraxasterol as an antidiabetic, anti-cancer, anti-inflammatory and an antioxidant agent. Methods: Articles for this review were searched on Scopus, Nature, SpringerLink and PubMed databases. The keywords used for the search were “taraxasterol pharmacology”, “taraxasterol antioxidant”, “taraxasterol cancer”, “taraxasterol biosynthesis” and “taraxasterol antidiabetic”. Relevance and report of distinct mechanisms of action were considered in the selection. Results: This review underlines the therapeutic potential of taraxasterol in cancer and diabetes conditions, while also highlighting its anti-inflammatory and antioxidant effects. Some specific biochemical pathways that are modulated due to the pharmacological effect of taraxasterol were diagrammatically represented. Conclusions: It is hoped that this review would stimulate interest in taraxasterol by focusing on its pharmacological relevance, specific mechanisms of action and pharmacokinetics. This might facilitate its eventual development into drug and adoption into treatment regimen of various diseases.

Fragile X-associated Tremor/Ataxia Syndrome (FXTAS) is a neurodegenerative disorder associated with the FMR1 premutation. Currently, it is not possible to determine when and if individual premutation carriers will develop FXTAS. Thus, with the aim to identify biomarkers for early diagnosis, development, and progression of FXTAS, along with associated dysregulated pathways, we performed blood proteomic profiling of premutation carriers (PM) who, as part of an ongoing longitudinal study, emerged into two distinct groups: those who developed symptoms of FXTAS (converters, CON) over time (at subsequent visits) and those who did not (non-converters, NCON). We compared these groups to age-matched healthy controls (HC). We assessed CGG repeat allele size by Southern blot and PCR analysis. The proteomic profile was obtained by liquid chromatography mass spectrometry (LC-MS/MS). We identified several significantly differentiated proteins between HC and the PM groups at Visit 1 (V1), Visit 2 (V2), and between the visits. We further reported the dysregulated protein pathways, including sphingolipid and amino acid metabolism. Our findings are in agreement with previous studies showing that pathways involved in mitochondrial bioenergetics, as observed in other neurodegenerative disorders, are significantly altered and appear to contribute to the development of FXTAS. Lastly, we compared the blood proteome of the PM who developed FXTAS over time with the CSF proteome of the FXTAS patients recently reported and found eight significantly differentially expressed proteins in common. To our knowledge, this is the first report of longitudinal proteomic profiling and the identification of unique biomarkers and dysregulated protein pathways in FXTAS.

Abstract Using a longitudinal national educational dataset, data science methods were applied to explain students’ educational trajectories and determine the most predictive variables in STEM degree attainment. Challenging the notion of the STEM pipeline, an Alternative Pathways to STEM (APS) model was proposed. Using a foundation of Social Cognitive Career Theory, academic, demographic, and economic variables were selected for analysis and Imbalanced dataset and random forest data analytic techniques were implemented. Results supported the consideration of the Alternate Pathways to STEM model given the varied educational pathways identified throughout STEM matriculation. Additionally, multiple academic variables, except for career interest, were found to be most predictive of STEM graduation, while demographic and economic variables were not significant. These findings have important implications for recruiting STEM students and increasing STEM graduation rates. Additional research is recommended using these data extraction and analysis methodologies on other datasets to further test the efficacy of the Alternative Pathways to STEM model.

Abstract Background We have previously discovered clusters of sequentially negative and positive modulators of acute inflammation during cytokine stimulation in epithelial cells and identified potential targets for therapy within these clusters. MAP3K8 is a druggable kinase that we found to be a hub of a principal interaction network. We describe here the results of MAP3K8 knockdown in the A549 lung cancer cell line, the BEAS-2B epithelial cell line and normal human bronchial epithelial (NHBE) cells following IL-1β stimulation. We analysed signalling transduction and global gene expression after IL-1β stimulation with and without MAP3K8 knockdown, quantifying levels of the inflammatory cytokines IL-6, IL-8 and RANTES levels by qPCRs and/or by ELISAs. We also examined potential small molecule inhibitors for MAP3K8 in the same models. Results IL-1β significantly and consistently increased MAP3K8 expression after 2 h in A549, BEAS-2B and NHBE cells. Phosphorylation of MAP3K8 occurred at 20 min after IL-1β stimulation and MAP3K8 protein was degraded at 30 min. MAP3K8 knockdown significantly reduced IL-6, IL-8 levels after IL-1β stimulation and yielded a 10-fold enhancement of the anti-inflammatory effects of dexamethasone. Phosphorylation of ERK1/2 (P-ERK1/2) and phosphorylation of SAPK/JNK (P-SAPK/JNK) decreased at 30 min after IL-1β stimulation with MAP3K8 knockdown. The combination of dexamethasone and MAP3K8 knockdown resulted in greater inhibition of phosphorylated ERK1/2 and SAPK/JNK. Abstract Introduction Contraceptive use is known to have a positive impact on maternal and child health outcomes; however, its use is still low in low-income countries, especially among people in humanitarian situations. This study explored decision-making processes towards the use of contraceptives by people in humanitarian situations to inform program design and uptake. Methods A qualitative exploratory study was conducted among women of reproductive age (15–49 years) and men (15–60 years) living in three refugee settlements of Pagirinya, Nyumanzi, and Mirieyi and the surrounding host communities in Adjumani district, Uganda. Data were collected using 49 in-depth interviews (IDIs), 11 Key Informant Interviews (KIIs,) and 20 Focus Group Discussions (FGDs). Inductive thematic analysis was done with the aid of Atlas ti. Version 14. Results We found that the decision-making processes entailed linear and nonlinear internalized cognitive and contextual processes involving four dynamic pathways. In the linear pathway, participants reported starting with 1) idea inception, 2) followed by cognitive processing, 3) consultation, and 4) decision-making for contraceptive use. The complex linear pathway happened when participants did not go through consultation but went straight to decision-making. However, participants who followed the non-linear pathway repeatedly went back to cognitive processing. Some women after consultation, or those already using and those not using contraceptives, decided to go back to cognitive processing to reconsider their current positions. This study found that some women who were not using contraceptives ended up using, while some who were using contraception ended up dropping out. Conclusions This study showed dynamic decision-making processes involving both internal and external environments as triggers to decision-making for contraceptive use. Interventions to increase contraceptive use should target both users and significant others who influence the decision to use particularly among refugees. African swine fever virus (ASFV), a highly contagious pathogen characterized by a complex structure and a variety of immunosuppression proteins, causes hemorrhagic, acute, and aggressive infectious disease that severely injures the pork products and industry. However, there is no effective vaccine or treatment. The main reasons are not only the complex mechanisms that lead to immunosuppression but also the unknown functions of various proteins. This review summarizes the interaction between ASFV and the host immune system, along with the involvement of virulence-related genes and proteins, as well as the corresponding molecular mechanism of immunosuppression of ASFV, encompassing pathways such as cGAS-STING, nuclear factor kappa–light-chain-enhancer of activated B cells (NF-κB), Janus Kinase (JAK) and JAK Signal Transducers and Activators of Transcription (STAT), apoptosis, and other modulation. The aim is to summarize the dynamic process during ASFV infection and entry into the host cell, provide a rational insight into development of a vaccine, and provide a better clear knowledge of how ASFV impacts the host.

Australia's higher education sector has seen substantial developments in equity and inclusivity as a result of initiatives which work to improve the numbers and experiences of students from a wide diversity of backgrounds. This practice report shares learnings from a recent national project about the impacts of a wide range of access and equity programs and services, shedding light on effective strategies throughout the student life cycle. The study, known as the Critical Interventions Framework Part 3 (CIF 3), analysed published studies which provide details about the impact of initiatives, highlighting the mechanisms which have enabled students to gain access to and succeed in higher education. The focus here is on a key outcome of the CIF 3, the updated Equity Initiatives Framework (EIF), designed for utility as a quick reference guide to aid equity practitioners and others working to support students in their educational endeavours.

Youth homelessness presents a complex and persistent challenge worldwide, particularly affecting young adults between 16 and 24 years of age in the US and Canada. This population faces elevated risks of exploitation, victimization, and various health issues upon detachment from familial support structures. Understanding the multi-faceted nature of youth homelessness requires the consideration of individual, structural, and systemic factors within the socio-ecological model. Historically, when examining youth homelessness, traditional methods have concentrated either on individual factors contributing to homelessness or on broader structural issues within society. The emergence of the new orthodoxy attempted to bridge the apparent gap between individual- and structural-level factors by considering both to be equally significant, but it faced skepticism for its theoretical framework. In response, the “pathways” approach gained traction, emphasizing the subjective experiences and agency of youth experiencing homelessness. Departing from conventional epidemiological models, the pathways approach views homelessness as a dynamic process intertwined with individual life contexts. This paper navigates the scholarly discourse on youth homelessness and examines the distinct characteristics of the pathways approach. By exploring its implications for research and policy, this study contributes to a nuanced understanding of youth homelessness and informs future prevention-focused interventions.

Background: Identifying molecular mechanisms responsible for the response to heat stress is essential to increase production, reproduction, health, and welfare. This study aimed to identify early biological responses and potential biomarkers involved in the response to heat stress and animal's recovery in tropically adapted beef cattle through proteomic analysis of blood plasma. Methods: Blood samples were collected from 14 Caracu males during the heat stress peak (HSP) and 16 h after it (heat stress recovery--HSR) assessed based on wet bulb globe temperature index and rectal temperature. Proteome was investigated by liquid chromatography-tandem mass spectrometry from plasma samples, and the differentially regulated proteins were evaluated by functional enrichment analysis using DAVID tool. The protein-protein interaction network was evaluated by STRING tool. Results: A total of 1,550 proteins were detected in both time points, of which 84 and 65 were downregulated and upregulated during HSR, respectively. Among the differentially regulated proteins with the highest absolute log-fold change values, those encoded by the GABBR1, EPHA2, DUSP5, MUC2, DGCR8, MAP2K7, ADRA1A, CXADR, TOPBP1, and NEB genes were highlighted as potential biomarkers because of their roles in response to heat stress. The functional enrichment analysis revealed that 65 Gene Ontology terms and 34 pathways were significant (P < 0.05). We highlighted those that could be associated with the response to heat stress, such as those related to the immune system, complement system, hemostasis, calcium, ECM-receptor interaction, and PI3K-Akt and MAPK signaling pathways. In addition, the protein-protein interaction network analysis revealed several complement and coagulation proteins and acute-phase proteins as important nodes based on their centrality and edges. Conclusion: Identifying differentially regulated proteins and their relationship, as well as their roles in key pathways contribute to improve the knowledge of the mechanisms behind the response to heat stress in naturally adapted cattle breeds. International shipping is a major contributor to climate change. The International Maritime Organization (IMO) has jurisdiction over the sector's efforts to reduce greenhouse gas emissions, and in July 2023 agreed a revised climate change strategy, setting tightened targets for emissions reductions by 2030, 2040 and 2050. In this perspective article, we analyse the remaining carbon budgets available to the international shipping sector, to assess whether these new targets constitute a sufficient contribution to meeting the overarching Paris Agreement goal to limit global heating to 1.5°C. The new strategy sets both 'indicative checkpoints' and more ambitious 'strive' targets. Both represent a major advance over the previous strategy, however, only the 'strive' targets are compatible with the 1.5°C limit. The first 'strive' target is for 30% reductions by 2030, just seven years away. To meet this goal, it is imperative that the IMO, nation states and the shipping industry act immediately to accelerate deployment of known technologies and operational practices that improve energy efficiency and cut CO2 emissions in the existing fleet. The new IMO strategy represents a major improvement on the targets in the previous 2018 strategy. The 'indicative checkpoint' targets for 2030 and 2040 are not sufficient for a fair contribution from international shipping to meeting the Paris 1.5°C limit. Under generous assumptions, the 'strive' targets of 30% reductions by 2030 and 80% by 2040 are compatible with the 1.5°C limit. These should be considered a minimum level of ambition for the sector. Further delay would push compatibility with 1.5°C out of reach. The frequency of people presented in emergency departments (EDs) after self‐harming events is increasing. Previous studies have shown that the complexity of the disorders of patients admitted to the ED after self‐harming events can be overwhelming for ED healthcare professionals (HCPs) to handle. The objective of this study was to observe and investigate the pathways for patients admitted to the ED after self‐harming events to either transition or discharge. Participant observation and interviews were selected as the methods to generate insight into the pathways of patients admitted to the ED after self‐harming events. The data were analysed using interpretative phenomenological analysis. A sample size of 20 patients was analysed, and a total of 213 h of observation took place during the data collection. Three main themes appeared: (1) patients' mental stress versus high expectations, (2) uncertainty about how to address the self‐harming event and (3) a system of chaos. Patients admitted to the ED after self‐harming events struggle with difficult mental stress. Despite this, they face high expectations that they will fit in and cooperate in the ED. The healthcare system is organised with unclear responsibilities and without systematic ways to care for self‐harm patients and so provides chaotic patient pathways. There is a need for improved cross‐sectional competencies, mutual agreements and systematic communication for discharge, transitions and follow‐up care between those involved in the patient's pathway and care. Sixty‐two species of ants (Hymenoptera: Formicidae) were intercepted at South Korean points of entry on plants for planting imported from tropical and subtropical Asia from 1996 to 2023. Information on the number of interceptions, host plants, consignment origin and their years of interception is provided. Data on intercepted ant species were analysed to determine potential invasive species of ants that could cause damage not only to agricultural, forestry, and natural ecosystems but also to industry and that could endanger human health. Introduction: Contraceptive use is known to have a positive impact on maternal and child health outcomes; however, its use is still low in low-income countries, especially among people in humanitarian situations. This study explored decision-making processes towards the use of contraceptives by people in humanitarian situations to inform program design and uptake. Methods: A qualitative exploratory study was conducted among women of reproductive age (15–49 years) and men (15–60 years) living in three refugee settlements of Pagirinya, Nyumanzi, and Mirieyi and the surrounding host communities in Adjumani district, Uganda. Data were collected using 49 in-depth interviews (IDIs), 11 Key Informant Interviews (KIIs,) and 20 Focus Group Discussions (FGDs). Inductive thematic analysis was done with the aid of Atlas ti. Version 14. Results: We found that the decision-making processes entailed linear and nonlinear internalized cognitive and contextual processes involving four dynamic pathways. In the linear pathway, participants reported starting with 1) idea inception, 2) followed by cognitive processing, 3) consultation, and 4) decision-making for contraceptive use. The complex linear pathway happened when participants did not go through consultation but went straight to decision-making. However, participants who followed the non-linear pathway repeatedly went back to cognitive processing. Some women after consultation, or those already using and those not using contraceptives, decided to go back to cognitive processing to reconsider their current positions. This study found that some women who were not using contraceptives ended up using, while some who were using contraception ended up dropping out. Conclusions: This study showed dynamic decision-making processes involving both internal and external environments as triggers to decision-making for contraceptive use. Interventions to increase contraceptive use should target both users and significant others who influence the decision to use particularly among refugees. We focus on the use of flourishing as a new measure in studies of pathways in STEM (science, technology, engineering, and mathematical) fields. While the concept of flourishing is promising, the concept may need careful interrogation to ensure it takes structural and personal (cultural, religious, socioeconomic, and racial) differences into account. Our longitudinal study explores emerging adult's educational and career pathways with careful attention to structural inequities, enabling us to productively explore the concept of flourishing in a larger systemic context. Drawing from a set of qualitative interviews with our participants, we explore the ways that our sample of emerging adults (N = 30), predominantly people of color, define and discuss flourishing. The concept resonated with our diverse participants, and a substantial number did report flourishing. But despite the regularity with which the participants described experiencing racism and microaggressions, they did not often mention those harmful experiences when discussing flourishing. We caution that flourishing data on its own may provide an overly rosy image of the pathways and development, especially of young people of color. Our data suggest that it may be especially important to examine flourishing in context with other measures that can flesh out a fuller picture of well-being, especially in relation to race, racism, sexism, or any other experiences related to personal identities. Background: We have previously discovered clusters of sequentially negative and positive modulators of acute inflammation during cytokine stimulation in epithelial cells and identified potential targets for therapy within these clusters. MAP3K8 is a druggable kinase that we found to be a hub of a principal interaction network. We describe here the results of MAP3K8 knockdown in the A549 lung cancer cell line, the BEAS-2B epithelial cell line and normal human bronchial epithelial (NHBE) cells following IL-1β stimulation. We analysed signalling transduction and global gene expression after IL-1β stimulation with and without MAP3K8 knockdown, quantifying levels of the inflammatory cytokines IL-6, IL-8 and RANTES levels by qPCRs and/or by ELISAs. We also examined potential small molecule inhibitors for MAP3K8 in the same models. Results: IL-1β significantly and consistently increased MAP3K8 expression after 2 h in A549, BEAS-2B and NHBE cells. Phosphorylation of MAP3K8 occurred at 20 min after IL-1β stimulation and MAP3K8 protein was degraded at 30 min. MAP3K8 knockdown significantly reduced IL-6, IL-8 levels after IL-1β stimulation and yielded a 10-fold enhancement of the anti-inflammatory effects of dexamethasone. Phosphorylation of ERK1/2 (P-ERK1/2) and phosphorylation of SAPK/JNK (P-SAPK/JNK) decreased at 30 min after IL-1β stimulation with MAP3K8 knockdown. The combination of dexamethasone and MAP3K8 knockdown resulted in greater inhibition of phosphorylated ERK1/2 and SAPK/JNK. Nineteen genes including MMP1, MMP3, MMP10, ITGB8, LAMC2 and PLAT (P corrected < 0.01 respectively) demonstrated a distinct altered temporal response to IL-1β following suppression of MAP3K8. However, putative MAP3K8 inhibitors including Tpl2-1, Tpl2-2 and GSK2222867A only showed inhibition of IL-6 and IL-8 production at a high dose. Conclusions: These results confirm that MAP3K8 is a key mediator of the early inflammatory response and that it is a potential target in inflammatory diseases. Introduction: Glioma, a prevalent and deadly brain tumor, is marked by significant cellular heterogeneity and metabolic alterations. However, the comprehensive cell-of-origin and metabolic landscape in high-grade (Glioblastoma Multiforme, WHO grade IV) and low-grade (Oligoastrocytoma, WHO grade II) gliomas remains elusive. Methods: In this study, we undertook single-cell transcriptome sequencing of these glioma grades to elucidate their cellular and metabolic distinctions. Following the identification of cell types, we compared metabolic pathway activities and gene expressions between high-grade and low-grade gliomas. Results: Notably, astrocytes and oligodendrocyte progenitor cells (OPCs) exhibited the most substantial differences in both metabolic pathways and gene expression, indicative of their distinct origins. The comprehensive analysis identified the most altered metabolic pathways (MCPs) and genes across all cell types, which were further validated against TCGA and CGGA datasets for clinical relevance. Discussion: Crucially, the metabolic enzyme phosphodiesterase 8B (PDE8B) was found to be exclusively expressed and progressively downregulated in astrocytes and OPCs in higher-grade gliomas. This decreased expression identifies PDE8B as a metabolism-related oncogene in IDH-mutant glioma, marking its dual role as both a protective marker for glioma grading and prognosis and as a facilitator in glioma progression. The ever‐increasing and expanding globalisation of trade and transport underpins the escalating global problem of biological invasions. Developing biosecurity infrastructures is crucial to anticipate and prevent the transport and introduction of invasive alien species. Still, robust and defensible forecasts of potential invaders are rare, especially for species without known invasion history. Here, we aim to support decision‐making by developing a quantitative invasion risk assessment tool based on invasion syndromes (i.e., generalising typical attributes of invasive alien species). We implemented a workflow based on 'Multiple Imputation with Chain Equation' to estimate invasion syndromes from imputed datasets of species' life‐history and ecological traits and macroecological patterns. Importantly, our models disentangle the factors explaining (i) transport and introduction and (ii) establishment. We showcase our tool by modelling the invasion syndromes of 466 amphibians and reptile species with invasion history. Then, we project these models to amphibians and reptiles worldwide (16,236 species [c.76% global coverage]) to identify species with a risk of being unintentionally transported and introduced, and risk of establishing alien populations. Our invasion syndrome models showed high predictive accuracy with a good balance between specificity and generality. Unintentionally transported and introduced species tend to be common and thrive well in human‐disturbed habitats. In contrast, those with established alien populations tend to be large‐sized, are habitat generalists, thrive well in human‐disturbed habitats, and have large native geographic ranges. We forecast that 160 amphibians and reptiles without known invasion history could be unintentionally transported and introduced in the future. Among them, 57 species have a high risk of establishing alien populations. Our reliable, reproducible, transferable, statistically robust and scientifically defensible quantitative invasion risk assessment tool is a significant new addition to the suite of decision‐support tools needed for developing a future‐proof preventative biosecurity globally. The current study investigated whether hope and its two components (agency and pathways) weakened the relations between perceived burdensomeness, thwarted belongingness, and fearlessness about death and suicidal ideation among older adults living in rural Nepal. A community sample of 300 people aged from 60 to 90 years (Mage = 67.07, SDage = 6.23) who resided in the rural mid-hills of Nepal completed standardized measures. Hope and agency moderated the relationship between perceived burdensomeness and suicidal ideation and pathways moderated the relationship between thwarted belongingness and suicidal ideation. Hope, and particularly agency, may play a protective role in reducing suicidal ideation among older Nepali adults living in rural areas who experience perceived burdensomeness. In contrast, pathways may be an additional risk factor for older adults experiencing unmet belongingness needs. Research is required to understand fearlessness about death and suicidality among older Nepali adults. Increasing hope and particularly agency may be associated with a reduction in suicidal ideation among older Nepali adults who experience feelings of being a burden. However, increasing pathways may be associated with an increase in suicidal ideation among those who experience unmet belongingness needs. The current study investigated whether hope and its two components (agency and pathways) acted as protective factors by weakening the relations between perceived burdensomeness, thwarted belongingness, and acquired capability for suicide and suicide risk among older adults. A community sample of 594 older Australians aged from 60 to 95 years (Mage = 68.72, SDage = 6.67) completed the Suicidal Behaviors Questionnaire-Revised, Interpersonal Needs Questionnaire-Revised, Acquired Capability for Suicide Scale-Fearlessness about Death Scale, Adult Hope Scale, and Geriatric Depression Scale-short form. After controlling for depressive symptoms and sociodemographic variables, hope and agency moderated the thwarted belongingness-suicide risk relation, and hope, agency, and pathways moderated the acquired capability for suicide-risk relation. Hope may play an important role in reducing suicide risk among older adults experiencing thwarted belongingness and who have acquired the capability for suicide. Research is required to identify ways of reducing suicide risk among older adults who experience perceived burdensomeness. Head and neck cancer ranks as the sixth-most common malignancy worldwide, characterized by high mortality and recurrence rates. Research studies indicate that molecular diagnostics play a crucial role in the early detection and prognostic evaluation of these diseases. This study aimed to identify potential biomarkers for head and neck cancer and elucidate their interactions with miRNAs and possible therapeutic drugs. Four drivers, namely, FN1, IL1A, COL1A1, and MMP9, were identified using network biology and machine learning approaches. Gene set variation analysis (GSVA) showed that these genes were significantly involved in different biological processes and pathways, including coagulation, UV-response-down, apoptosis, NOTCH signaling, Wnt-beta catenin, and other signal pathways. The diagnostic value of these hub genes was validated using receiver operating characteristic (ROC) curves. The top interactive miRNAs, including miR-128-3p, miR-218-5p, miR-214-3p, miR-124-3p, miR-129-2-3p, and miR-1-3p, targeted the key genes. Furthermore, the interaction between the key genes and drugs was also identified. In summary, the key genes and miRNAs or drugs reported in this study might provide valuable information for potential biomarkers to increase the prognosis and diagnosis of head and neck cancer. Glycans participate in a vast number of recognition systems in diverse organisms in health and in disease. However, glycans cannot be sequenced because there is no sequencer technology that can fully characterize them. There is no "template" for replicating glycans as there are for amino acids and nucleic acids. Instead, glycans are synthesized by a complicated orchestration of multitudes of glycosyltransferases and glycosidases. Thus glycans can vary greatly in structure, but they are not genetically reproducible and are usually isolated in minute amounts. To characterize (sequence) the glycome (defined as the glycans in a particular organism, tissue, cell, or protein), glycosylation pathway prediction using in silico methods based on glycogene expression data, and glycosylation simulations have been attempted. Since many of the mammalian glycogenes have been identified and cloned, it has become possible to predict the glycan biosynthesis pathway in these systems. By then incorporating systems biology and bioprocessing technologies to these pathway models, given the right enzymatic parameters including enzyme and substrate concentrations and kinetic reaction parameters, it is possible to predict the potentially synthesized glycans in the pathway. This review presents information on the data resources that are currently available to enable in silico simulations of glycosylation and related pathways. Then some of the software tools that have been developed in the past to simulate and analyze glycosylation pathways will be described, followed by a summary and vision for the future developments and research directions in this area. To develop harmful sexual behavior (HSB) early intervention strategies it is essential to understand how children come to engage in HSB. Through the lens of a pathways theory, this paper reports a scoping review of the literature in answer to the research question: What are the pathways to HSB onset for children and young people? Eight bibliographic databases were searched over the time period January 2000 to May 2022 using the following terms (and derivatives): harmful sexual behavior AND children and young people AND pathways. Forty-three papers were included and 10 pathways to onset identified through synthesis: Child sexual abuse victimization; Physical and emotional abuse; Living with domestic and family violence; Disrupted attachments; Sexual arousal; "Antisociality;" Pornography use; Inadequate sexual boundaries; Sexual attraction to children; and Hypermasculinity. In this context, the development of high-quality green buildings requires a focus on improving carbon reduction effects throughout their entire lifespan. Based on a systematic review of the achievements and existing problems in the development of green buildings in China over the past thirty years, this article benchmarks the new trends in green building standards in developed countries abroad, dissects the new requirements for the development of green buildings in China under the new situation, and proposes a high-quality carbon reduction pathways from four aspects: theoretical research, policy systems, technical systems, and software platforms. Finally, it proposes implementation suggestions for collaborative development from four dimensions: theoretical methods for calculating carbon emissions, green finance policies, a full-process carbon reduction technology system, and carbon emission accounting tools. Urologic oncology is a significant public health concern on a global scale. Recent research indicates that long chain non-coding RNAs (lncRNAs) and autophagy play crucial roles in various cancers, including urologic malignancies. This article provides a summary of the latest research findings, suggesting that lncRNA-mediated autophagy could either suppress or promote tumors in prostate, kidney, and bladder cancers. The intricate network involving different lncRNAs, target genes, and mediated signaling pathways plays a crucial role in urological malignancies by modulating the autophagic process. Dysregulated expression of lncRNAs can disrupt autophagy, leading to tumorigenesis, progression, and enhanced resistance to therapy. Consequently, targeting particular lncRNAs that control autophagy could serve as a dependable diagnostic tool and a promising prognostic biomarker in urologic oncology, while also holding potential as an effective therapeutic approach. Herein, four different metal-free nitrogen containing Covalent Triazine Frameworks (CTFs) based on their applied monomers (DCP, DCBP, m DCB and p DCB) are synthesized via a classical ionothermal synthesis route (ZnCl 2 , 400/600 °C). These materials are fully characterized and their electrochemical activities for ORR are tested and compared to each other including Carbon Super P and Pt black as standards in 0.1 M KOH. While DCP provides similar catalytic activity to Carbon Super P showing mostly a 2eˉ process (n=2.95) with high H 2 O 2 formation of 52.6 %, the other three CTFs (DCBP, m DCB and p DCB) possess higher ORR activities, surprisingly even much higher limiting current densities than Pt black, proving that O 2 is mainly reduced via direct 4eˉ pathway since n values are in the range of 3.52 to 3.62 and the detected H 2 O 2 values are in the range of 19–23.9 %. Among the studied CTFs, m DCB reaches a limiting current density of −5.61 mA cm-2 (1.21 mA cm-2 larger than that for Pt black, −4.40 mA cm-2) with 0.11 V larger onset potential compared to Pt black. The significant electrochemical performances of the CTF materials in ORR via a 4eˉ process are correlated to the high specific surface areas (up to 2500 m2 g-1), large pore volumes (up to 2.05 cm3 g-1) and the largest total N-graphitic/quaternary contents as well as micro-mesoporous structure properties. Background and objectives: Major Depressive Disorder (MDD) is one of the most prevalent and debilitating health conditions worldwide. Previous studies have reported a link between metabolic dysregulation and MDD. However, evidence for a causal relationship between blood metabolites and MDD is lacking. Methods: Using a two-sample bidirectional Mendelian randomization analysis (MR), we assessed the causal relationship between 1,400 serum metabolites and Major Depressive Disorder (MDD). The Inverse Variance Weighted method (IVW) was employed to estimate the causal association between exposures and outcomes. Additionally, MR-Egger regression, weighted median, simple mode, and weighted mode methods were used as supplementary approaches for a comprehensive appraisal of the causality between blood metabolites and MDD. Pleiotropy and heterogeneity tests were also conducted. Lastly, the relevant metabolites were subjected to metabolite function analysis, and a reverse MR was implemented to explore the potential influence of MDD on these metabolites. Results: After rigorous screening, we identified 34 known metabolites, 13 unknown metabolites, and 18 metabolite ratios associated with Major Depressive Disorder (MDD). Among all metabolites, 33 were found to have positive associations, and 32 had negative associations. The top five metabolites that increased the risk of MDD were the Arachidonate (20:4n6) to linoleate (18:2n6) ratio, LysoPE(18:0/0:0), N-acetyl-beta-alanine levels, Arachidonate (20:4n6) to oleate to vaccenate (18:1) ratio, Glutaminylglutamine, and Threonine to pyruvate ratio. Conversely, the top five metabolites that decreased the risk of MDD were N6-Acetyl-L-lysine, Oleoyl-linoleoyl-glycerol (18:1 to 18:2) [2] to linoleoyl-arachidonoyl-glycerol (18:2 to 20:4) [2] ratio, Methionine to phosphate ratio, Pregnanediol 3-O-glucuronide, and 6-Oxopiperidine-2-carboxylic acid. Metabolite function enrichment was primarily concentrated in pathways such as Bile Acid Biosynthesis (FDR=0.177), Glutathione Metabolism (FDR=0.177), Threonine, and 2-Oxobutanoate Degradation (FDR=0.177). In addition, enrichment was noted in pathways like Valine, Leucine, and Isoleucine Biosynthesis (p=0.04), as well as Ascorbate and Aldarate Metabolism (p=0.04). Discussion: Within a pool of 1,400 blood metabolites, we identified 34 known metabolites and 13 unknown metabolites, as well as 18 metabolite ratios associated with Major Depressive Disorder (MDD). Additionally, three functionally enriched groups and two metabolic pathways were selected. It is adaptive to restrict eating under uncertainty, such as during habituation to novel foods and unfamiliar environments. However, sustained restrictive eating can become maladaptive. Currently, the neural substrates of restrictive eating are poorly understood. Using a model of feeding avoidance under novelty, our recent study identified forebrain activation patterns and found evidence that the central nucleus of the amygdala (CEA) is a core integrating node. The current study analyzed the activity of CEA inputs in male and female rats to determine if specific pathways are recruited during feeding under novelty. Recruitment of direct inputs from the paraventricular nucleus of the thalamus (PVT), the infralimbic cortex (ILA), the agranular insular cortex (AI), the hippocampal ventral field CA1, and the bed nucleus of the stria terminals (BST) was assessed with combined retrograde tract tracing and Fos induction analysis. The study found that during consumption of a novel food in a novel environment, larger number of neurons within the PVTp and the CA1 that send monosynaptic inputs to the CEA were recruited compared to controls that consumed familiar food in a familiar environment. The ILA, AI, and BST inputs to the CEA were similarly recruited across conditions. There were no sex differences in activation of any of the pathways analyzed. These results suggest that the PVTp-CEA and CA1-CEA pathways underlie feeding inhibition during novelty and could be potential sites of malfunction in excessive food avoidance. Direct oral anticoagulants (DOACs) are the standard treatment for thromboembolic protection in atrial fibrillation (AF) patients. Epigenetic modifications, such as DNA methylation and microRNAs, have emerged as potential biomarkers of AF. The epigenetics of DOACs is still an understudied field. It is largely unknown whether epigenetic modifications interfere with DOAC response or whether DOAC treatment induces epigenetic modifications. To fill this gap, we started the miR-CRAFT (Circulating microRNAs and DNA methylation as regulators of Direct Oral Anticoagulant Response in Atrial Fibrillation) research study. In miR-CRAFT, we follow, over time, changes in DNA methylation and microRNAs expression in naïve AF patients starting DOAC treatment. The ultimate goal of miR-CRAFT is to identify the molecular pathways epigenetically affected by DOACs, beyond the coagulation cascade, that are potentially mediating DOAC pleiotropic actions and to propose specific microRNAs as novel circulating biomarkers for DOAC therapy monitoring. Introduction: Patients in the community with suspected Chronic limb‐threatening ischaemia (CLTI) should be urgently referred to vascular services for investigation and management. The Theoretical Domains Framework (TDF) allows identification of influences on health professional behaviour in order to inform future interventions. Here, the TDF is used to explore primary care clinicians' behaviours with regards to recognition and referral of CLTI. Methods: Semi‐structured interviews were conducted with 20 podiatrists, nurses and general practitioners in primary care. Directed content analysis was performed according to the framework method. Utterances were coded to TDF domains, and belief statements were defined by grouping similar utterances. Relevance of domains was confirmed according to belief frequency, presence of conflicting beliefs and the content of the beliefs indicating relevance. Results: Nine TDF domains were identified as relevant to primary care clinicians: Knowledge, Environmental context and resources, Memory, Decision and attention processes, Beliefs about capabilities, Skills, Emotions, Reinforcement and Behavioural regulation. Relationships across domains were identified, including how primary care clinician confidence and working in a highly pressurized environment can affect behaviour. Conclusion: We have identified key barriers and enablers to timely recognition and referral behaviour. These beliefs identify targets for theory‐driven behaviour change interventions to reduce delays in CLTI pathways. [ABSTRACT FROM AUTHOR]

The West Antarctic Ice Sheet is experiencing rapid thinning of its floating ice shelves, largely attributed to oceanic basal melt. Numerical models suggest that the Bellingshausen Sea has a key role in setting water properties in the Amundsen Sea and further downstream. Yet, observations confirming these pathways of volume and tracer exchange between coast and shelf break and their impact on inter‐sea exchange remain sparse. Here we analyze the circulation and distribution of glacial meltwater at the boundary between the Bellingshausen Sea and the Amundsen Sea using a combination of glider observations from January 2020 and hydrographic data from instrumented seals. Meltwater distributions over previously unmapped western regions of the continental shelf and slope reveal two distinct meltwater cores with different optical backscatter properties. At Belgica Trough, a subsurface meltwater peak is linked with hydrographic properties from Venable Ice Shelf. West of Belgica Trough, the vertical structure of meltwater concentration changes, with peak values occurring at greater depths and denser isopycnals. Hydrographic analysis suggests that the western (deep) meltwater core is supplied from the eastern part of Abbot Ice Shelf, and is exported to the shelf break via a previously‐overlooked bathymetric trough (here named Seal Trough). Hydrographic sections constructed from seal data reveal that the Antarctic Coastal Current extends west past Belgica Trough, delivering meltwater to the Amundsen Sea. Each of these circulation elements has distinct dynamical implications for the evolution of ice shelves and water masses both locally and downstream, in the Amundsen Sea and beyond. Plain Language Summary: Floating ice shelves in West Antarctica are thinning, which is largely due to melting of the ice shelf base by the ocean. Here, measurements of ocean temperature, salinity, and dissolved oxygen, collected by a remotely‐controlled underwater vehicle (a glider), are used to estimate the amount of ice shelf meltwater released in the Bellingshausen Sea. Distinct cores of meltwater can be distinguished by the amount of suspended material that is present in the water, which we attribute to meltwater following different circulation pathways after entering the ocean. Historical data from seals equipped with temperature and salinity sensors provide additional information about how the meltwater circulates in the region. The seal data show the presence of a narrow coastal current that brings meltwater from the Bellingshausen Sea into the Amundsen Sea. The pathways of meltwater revealed in this study suggest an important influence of the Bellingshausen Sea on ice shelves throughout West Antarctica. Youth homelessness presents a complex and persistent challenge worldwide, particularly affecting young adults between 16 and 24 years of age in the US and Canada. This population faces elevated risks of exploitation, victimization, and various health issues upon detachment from familial support structures. Understanding the multi-faceted nature of youth homelessness requires the consideration of individual, structural, and systemic factors within the socio-ecological model. Historically, when examining youth homelessness, traditional methods have concentrated either on individual factors contributing to homelessness or on broader structural issues within society. The emergence of the new orthodoxy attempted to bridge the apparent gap between individual- and structural-level factors by considering both to be equally significant, but it faced skepticism for its theoretical framework. In response, the "pathways" approach gained traction, emphasizing the subjective experiences and agency of youth experiencing homelessness. Departing from conventional epidemiological models, the pathways approach views homelessness as a dynamic process intertwined with individual life contexts. This paper navigates the scholarly discourse on youth homelessness and examines the distinct characteristics of the pathways approach. By exploring its implications for research and policy, this study contributes to a nuanced understanding of youth homelessness and informs future prevention-focused interventions. Pedestrian pathways should be designed to meet the needs of regular pedestrians and accommodate individuals with disabilities. Presently, a substantial number of pedestrian routes across Indonesia remain inaccessible to those reliant on wheelchairs and people with disabilities. This study aimed to evaluate and design disability-friendly pedestrian pathways for optimal accessibility and safety in Indonesia. A pedestrian pathways model was derived using a triangulation of data methodology (a review of literature, observation and simulation, and a focus group discussion convening stakeholders). Based on research results, none of the pedestrian pathways meet the criteria for individuals with disabilities. The Customer Satisfaction Index (CSI) score on pedestrian pathways indicates the unsatisfied category. Pedestrians with physical limitations argue that pedestrian pathways cannot provide comfort to support their movement. The evaluation results show that people with disabilities face difficulty accessing pedestrian pathways. This research produces pedestrian pathway designs based on three universal principles: adequate maneuvering space, clear signage and information, and appropriate surface materials. These universal design principles refer to efforts to create accessible, safe, and comfortable environments for everyone, regardless of age and physical ability. [ABSTRACT FROM AUTHOR]
