Abstract

This paper proposes a novel hierarchical architecture and resource-management protocol for the delegation of work within a ubiquitous and heterogeneous environment. The protocol is based on serving an SVP place, where a component of work is delegated together with the responsibility for meeting any non-functional computational requirements such as deadline or throughput constraints. The protocol is based on a market where SANE processors bid for jobs to execute and selection is based on a cost model that reflects the energy required to meet the jobs requirements.

Abstract

The Enhancing NeuroImaging Genetics through Meta-Analysis (ENIGMA) Consortium is a collaborative network of researchers working together on a range of large-scale studies that integrate data from 70 institutions worldwide. Organized into Working Groups that tackle questions in neuroscience, genetics, and medicine, ENIGMA studies have analyzed neuroimaging data from over 12,826 subjects. In addition, data from 12,171 individuals were provided by the CHARGE consortium for replication of findings, in a total of 24,997 subjects. By meta-analyzing results from many sites, ENIGMA has detected factors that affect the brain that no individual site could detect on its own, and that require larger numbers of subjects than any individual neuroimaging study has currently collected. ENIGMA's first project was a genome-wide association study identifying common variants in the genome associated with hippocampal volume or intracranial volume. Continuing work is exploring genetic associations with subcortical volumes (ENIGMA2) and white matter microstructure (ENIGMA-DTI). Working groups also focus on understanding how schizophrenia, bipolar illness, major depression and attention deficit/hyperactivity disorder (ADHD) affect the brain. We review the current progress of the ENIGMA Consortium, along with challenges and unexpected discoveries made on the way.

Abstract

The Enhancing NeuroImaging Genetics through Meta-Analysis (ENIGMA) Consortium is a collaborative network of researchers working together on a range of large-scale studies that integrate data from 70 institutions worldwide. Organized into Working Groups that tackle questions in neuroscience, genetics, and medicine, ENIGMA studies have analyzed neuroimaging data from over 12,826 subjects. In addition, data from 12,171 individuals were provided by the CHARGE consortium for replication of findings, in a total of 24,997 subjects. By meta-analyzing results from many sites, ENIGMA has detected factors that affect the brain that no individual site could detect on its own, and that require larger numbers of subjects than any individual neuroimaging study has currently collected. ENIGMA's first project was a genome-wide association study identifying common variants in the genome associated with hippocampal volume or intracranial volume. Continuing work is exploring genetic associations with subcortical volumes (ENIGMA2) and white matter microstructure (ENIGMA-DTI). Working groups also focus on understanding how schizophrenia, bipolar illness, major depression and attention deficit/hyperactivity disorder (ADHD) affect the brain. We review the current progress of the ENIGMA Consortium, along with challenges and unexpected discoveries made on the way. © 2014 The Author(s).

Abstract

Identifying genetic variants influencing human brain structures may reveal new biological mechanisms underlying cognition and neuropsychiatric illness. The volume of the hippocampus is a biomarker of incipient Alzheimer's disease and is reduced in schizophrenia, major depression and mesial temporal lobe epilepsy. Whereas many brain imaging phenotypes are highly heritable, identifying and replicating genetic influences has been difficult, as small effects and the high costs of magnetic resonance imaging (MRI) have led to underpowered studies. Here we report genome-wide association meta-analyses and replication for mean bilateral hippocampal, total brain and intracranial volumes from a large multinational consortium. The intergenic variant rs7294919 was associated with hippocampal volume (12q24.22; N = 21,151; P = 6.70 × 10 -16) and the expression levels of the positional candidate gene TESC in brain tissue. Additionally, rs10784502, located within HMGA2, was associated with intracranial volume (12q14.3; N = 15,782; P = 1.12 × 10 -12). We also identified a suggestive association with total brain volume at rs10494373 within DDR2 (1q23.3; N = 6,500; P = 5.81 × 10 -7). © 2012 Nature America, Inc. All rights reserved.

Abstract

With a multistage laboratory protocol immunodeficiencies can be efficiently identified. The article presents a diagnostic protocol that consists of three schemes. Scheme 1 describes the diagnostic protocol for the large group of patients with recurrent pulmonary and ENT-infections, where an antibody deficiency can occasionally be found. Scheme 2 presents the diagnostic protocol for the much smaller group of patients with opportunistic infections, wasting or failure to thrive. Several of these patients suffer from a severe T-lymphocyte disorder. Early diagnosis and treatment is important for the prognosis in these patients. Scheme 3 shows the diagnostic protocol for patients with recurrent infections of surface areas and deeper organs; these patients may suffer from a phagocyte disorder

Abstract

A multistage laboratory protocol for the diagnosis of immunodeficiency is useful for the efficient identification of immunodeficient patients. The protocol presented in this article starts with the patient's clinical presentation. In the initial stages a low threshold for the performance of simple screening is applied, thus allowing early exclusion of potential immunodeficiencies, as well as identification of patients before serious infections have compromised their general condition. In the later stages, more elaborate tests leading to diagnosis and definitive classification are reserved for those few patients in whom the presence of an immunodeficiency is more probable. This definitive classification is important for the identification of carriers and for the genetic counselling of the family. The protocol described has been developed in cooperation with the Dutch national working parties of clinical immunologists (paediatric as well as internal medicine) and laboratory immunologists who are involved in diagnosing or treating patients with immunodeficiency

Abstract

An increasing amount of datasets that can be combined with other datasets is emerging. Particularly the SemanticWeb allows creating links between datasets, which increase the value of the dataset that is being linked. Finding links between datasets, as well as capturing the data inside another dataset that is of interest is a process for which tools have yet to be perfected. Often the process of enriching data involves understanding how to access the data, and finding clues how to use auxiliary data to add value to the original dataset. This Thesis document proposes a framework for visually specifying the enrichment of RDF data. The framework is called RDF Gears, and allows incremental enrichment of RDF datasets. The platform provides functionality for composing enrichment flows from individual enrichment steps. The enrichment flow describes the order in which RDF data will be gathered from remote RDF datasources. After specifying the flow, the enrichments can be applied to the RDF dataset. RDF Gears also provides functionality for creating and reusing enrichment steps and (nested) flows. The approach discussed in this document shows the potential of specifying RDF enrichments incrementally. Also the possibility for abstracting Semantic Web technologies using the visual programming paradigm is shown. Abstract

Wij hebben ons bachelorproject uitgevoerd bij software- en consultancy bedrijf TOPdesk. Zij hadden ons een interessante opdracht te bieden waarmee wij 8 april 2008 begonnen zijn. De opdracht hield in dat er een systeem gemaakt moest worden waarmee het toevoegen van softwaremodules (Eclipse-projecten) aan de Eclipse workspace vereenvoudigd wordt, en het aanpassen van de instellingen van een project vanuit een centraal systeem kan worden doorgevoerd. Dit systeem is ontwikkeld onder de naam TOPspace. Het project begon in april met het houden van interviews met ir. T.J. Ouwens en ing. R. Spilker, waardoor een lijst met requirements opgesteld kon worden. Deze lijst is uitgewerkt in Use Cases en scenario’s, waardoor een beter inzicht werd verkregen voor de ontwerpfase, deze fase sloten we eind april af. Voordat we in mei aan het ontwerp konden beginnen is er een klein onderzoek gedaan. Tijdens dit onderzoek zijn de verschillende implementatievormen, het gebruik van Subversion en het hergebruik van Eclipse-functionaliteiten onderzocht. In de ontwerpfase is de afweging gemaakt tussen de verschillende implementatievormen die we voor het ontwerpen onderzocht hebben en is besloten dat een Eclipse-plugin de beste oplossing bood. Bij het ontwerp is rekening gehouden met de modulariteit van het systeem en de samenwerking tussen Eclipse en de gemaakte Eclipse-plugin. De modulariteit is gerealiseerd door gebruik te maken van Java als programmeertaal en door reflection toe te passen. Tot slot zijn we begonnen met de implementatiefase, waarin het implementeren en testen in parallel uitgevoerd is. Voor het testen is gebruik gemaakt van JUnit en JMock. Tijdens deze fase zijn er ook gebruikerstests uitgevoerd. Uit deze tests kwamen een aantal extra functionele wensen, alsmede een aantal bugs. Wij denken dat voldaan is aan alle requirements die opgesteld zijn in ons requirements document. Hiertoe behoren de twee verschillende delen van de opdracht, namelijk het implementeren van het stap, Software Engineering, Electrical Engineering, Mathematics and Computer Science

121. Identification of an unusual Fc gamma receptor IIIa (CD16) on natural killer cells in a patient with recurrent infections

Abstract

We found an unusual fc gamma receptor IIIa (CD16) phenotype on the natural killer (NK) cells of a 3-year-old boy, who suffered from recurrent viral respiratory tract infections since birth. He also had severe clinical problems after Bacille Calmette-Geérin (BCG) vaccination and following Epstein-Barr virus and Varicella Zoster virus infections. His peripheral blood lymphocytes contained a normal percentage and absolute number of CD3-CD7+ cells, which were positively stained with the CD16 monoclonal antibodies (MoAbs) 3G8 and CLBFcRgran1, but did marginally stain with the CD16 MoAb Lau11c/B73.1. Fc gamma RillIb expression on granulocytes appeared to be normal. NK cell function, analyzed in vitro by direct cytotoxicity on K562 target cells and ADCC-activity on P815 target cells, was normal compared with an age-matched healthy control. Sequence analysis of the Fc gamma RIIIA gene, encoding CD16 on NK cells and macrophages, showed a T to A nucleotide substitution at position 230 on both alleles, predicting a leucine (L) to histidine (H) amino acid change position 48 in the first extracellular lg-like domain of Fc gamma RIIIa, which contains the Leu11c/B73.1 epitope. The combined use of CD16 and CD56 MoAbs labeled with the same fluorescent dye, as often applied in routine immunophenotyping procedures, will leave these homozygotes undiagnosed. The pattern of infections in this patient is in agreement with the postulated function of NK cells in the immunological defense against viruses and other intracellular microorganisms. Further analysis of the NK cell function in vitro and follow-up of the clinical course of Fc gamma RIIIA-48H/H homozygotes is required to ascertain whether this genotype is causally related to an NK cell immunodeficiency

Abstract

We present data from a 10-year follow-up study of 11 children who had been infected in the neonatal period by small aliquots of plasma from a single donation. Three of the children died within the first 2.5 years of life, 5 other children died between 6.2 and 11 years after infection and 3 are alive at present. The latter children are classified as P1B (asymptomatic), P2A (non-specific findings) and P2B (neurological changes). All infected children showed progressive decline of cellular immunity. Immunoglobulin levels in serum were increased in the majority of children for prolonged periods and homogeneous immunoglobulin components were present. The severity of the disease was related neither to the clinical condition of the infants in the neonatal period nor to the volume of transfused plasma, the interval between freezing and thawing of the plasma, gestational age at birth and age at transfusion. Coinciding infections with other viruses had no impact on disease progression during the follow-up period of 10 years
