As a transcription factor first found in plants, WRKY plays an important role in plant growth and development. However, it has not been studied in Physcomitrella patens. By using the WRKY gene family data(accession number is PF03106) in Pfam database, this paper studied the basic information of WRKY in P. patens, which included physicochemical properties, protein secondary structure prediction, chromosome localization, exon and intron distribution and phylogenetic relationship. The results were as follows:(1)The WRKY gene family in P. patens consisted of 38 members which were divided into two major categories ─Ⅰ and Ⅱ, and the conserved domains of some WRKY genes had mutated.(2)The amino acid length of WRKY protein was 216-775 aa and the relative molecular mass was 24.5-82.8 kDa; Subcellular location showed that the proteins of WRKY gene family were distributed in the nucleus.(3)The secondary structure of WRKY protein was composed of four constituent elements: α-helix, extended strand, β-turn, and random coil; Except for PpWRKY11(α-helix dominated), the proportion of random coil was up to 70%.(4)The phylogenetic relationship with Arabidopsis showed that the number of the WRKY family members and the way of their revolution had changed in the process of plants' evolution, the number of exons of WRKY gene family members were 3-7.(5)The members of WRKY gene were randomly dispersed on 21 chromosomes without forming a gene cluster. This study analyzed the basic structure and properties of the WRKY gene family, and it was found that the WRKY gene family of Physcomitrella patens has evolutionary diversity and unique variability in conserved domains, laying the foundation for subsequent research. On the occasion of celebrating the 40th anniversary of Guihaia's launch, we retrospected the journal development, which could be divided into four stages: initially developed from an in-house journal; published at home and abroad, and journal influence surged; seized the opportunity to realize journal leapfrog development; created journal brand characteristics and promoted the overall development of the journal with featured columns. And we analyzed the academic influence of the journal from the aspects of published information, objects of reports(authors), sources and characteristics of authors, funding projects, citations by domestic journals, downloads of published articles and so on, based on the data of CNKI, CNKI CItation Database and related evaluation institutions through citation analysis method. We also summarized the achievements, such as finding and nurturing talents and providing good growth platfrom for them; improving the evaluation indexes and expanding the influence of the journal; focusing on the national strategy and major social needs to create disciplinary characteristics of the column; the author group and the reader group enlargement and the editorial board construction and so on. Looking ahead, we need to face great challenges. In this regard, we put forward the following ideas: First, center on the task of building an innovative country and a scientific and technological power, enhance academic leading ability and brand influence; Second, focus on the major strategic needs of the country, create special columns on major themes, and serve the main battlefield of economic and social development; Third, follow the trend of media convergence, ad apt to the direction of mobile and intelligent development, explore new publishing models such as all-media publishing, and speed up the development of journal convergence; Fourth, appropriately increase the proportion of international editors, provide long English abstracts of papers, strengthen the construction of bilingual academic websites, and improve the level of open running journal and international communication ability, and strive to build a first-class domestic academic journal with international influence. BACKGROUND: Classically activated and alternative activated macrophages have different phenotypes and functions, and there are few studies to explore the effect of different induction methods on the biological functions of bone marrow-derived macrophages. OBJECTIVE: To explore the effect of different induced polarization ways on biological behaviors of bone marrow-derived macrophages from rats. METHODS: Rat bone marrow-derived macrophages were isolated and cultured in vitro, and identified by morphological observation, Wright Giemsa staining and flow cytometry. After being induced and activated by interferon-γ, lipopolysaccharide and interleukin-4 for 24 hours, the morphology of bone marrow-derived macrophages was observed by inverted microscope. The expression levels of cell surface markers Nos2 and Arg1 were detected by real time-PCR. CCK8 assay was performed to determine the proliferation ability of bone marrow-derived macrophages. Flow cytometry was performed to detect the apoptosis, and neutral red staining was performed to identify the phagocytic function of bone marrow-derived macrophages. RESULTS AND CONCLUSION: Bone marrow-derived macrophages that stimulated by interferon-γ and lipopolysaccharide were highly expressing the cell surface marker Nos2 of M1, and apoptosis of bone marrow-derived macrophages can be slightly promoted as well as the phagocytic function of bone marrow-derived macrophages is reduced; while bone marrow-derived macrophages stimulated by interleukin-4 are highly expressing the cell surface marker Arg1 of M2. Cell proliferation and phagocytosis of bone marrow-derived macrophages are promoted, and apoptosis of bone marrow-derived macrophages is inhibited. Objective·To explore the therapeutic frequency and effect of phage treatment on pan-drug resistant Klebsiella pneumoniae by aerosolized phage. Methods·The patient's sputum was taken 1 h before the start of treatment and every hour during the treatment of pulmonary infection using nebulized inhaled phage. The sputum samples were subjected to bacterial isolation, purification and mass spectrometry identification. The lytic spectrum of different bacterial isolations was compared. And blood samples for bacteria were taken 1 h before the start of the treatment and 2 h and 4 h after the first treatment for bacterial culture and phage volume determination. Results·The phage continued to proliferate in vivo for 2 h, and decreased after 3 h. When the second phage treatment was performed after 4 h, the pattern of phage proliferation in vivo was the same as the first time. During the third treatment, the phage was proliferated slightly in vivo. After phage treatment, Klebsiella pneumoniae was still isolated in the patient's sputum. However, the phage cleavage spectrum of the isolations was different, and the patient's clinical symptoms improved. Conclusion·Phages can effectively target infectious bacteria and kill them. The clinical symptoms of patient improve. Twice aerosolized phage treatment for the patient with pulmonary infection is enough to achieve the best effect. Objective To investigate the influencing factors for the clinical effect of emergency endoscopic therapy in the treatment of patients with acute - on - chronic liver failure and gastroesophageal variceal bleeding. Methods A total of 51 patients with acute - on - chronic liver failure and gastroesophageal variceal bleeding who underwent emergency endoscopic therapy in Beijing Shijitan Hospital and The Fifth Medical Center of Chinese PLA General Hospital from January 2016 to December 2018 were enrolled, among whom 26 had successful hemostasis and 25 had failed hemostasis. The two groups were compared in terms of general information, varices grade and bleeding manifestations under endoscope, blood biochemical parameters, ultrasound findings, Child - Pugh class, and Model for End - Stage Liver Disease ( MELD) score, and the influencing factors for the outcome of hemostasis were analyzed. The t - test or the Mann - Whitney U test was used for comparison of continuous data between two groups, and the chi - square test was used for comparison of categorical data between two groups ; the logistic regression model was used to perform the multivariate analysis. Results Of all patients, 26 achieved successful hemostasis, with a success rate of hemostasis of 51 % . There were no significant differences between the two groups in sex, age, etiology of liver cirrhosis, presence or absence of liver cancer, presence or absence of portal vein thrombosis, bleeding for the first time or not, white blood cell count, hemoglobin, platelet count, prothrombin time activity, alanine aminotransferase, total bilirubin, albumin, cholinesterase, MELD score, and bleeding site and bleeding manifestations under gastroscope (all P >0. 05 ) . Compared with the failed hemostasis group, the successful hemostasis group had a significantly longer course of disease ( t = 2. 760, P = 0. 008) and significantly larger portal vein diameter and diameter of varicose veins under endoscope ( t = - 4. 847, x2 = -6. 590, both P <0. 05), and the failed hemostasis group had a significantly higher proportion of patients with Child - Pugh class C disease than the successful hemostasis group (x² = 5. 684, P = 0. 017 ). Course of liveicirrhos is (odds ratio [ OR J = 0. 913, 95 % confidence interval [ CI ] : 0. 838 - 0. 994, P < 0. 05 ), portal vein diameter ( OR = 1. 925,95 % CI: 1. 516 - 2. 443, P < 0. 05), and diameter of varicose veins (OR = 23. 254, 95 % CI: 2. 250 - 240. 352, P < 0. 05) were independent influencing factors for the clinical effect of endoscopic hemostasis. Conclusion There is a relatively low success rate of emergency endoscopic hemostasis in patients with acute - on - chronic liver failure, and course of liver cirrhosis, portal vein diameter, and diameter of varicose veins are independent influencing factors for the clinical effect of endoscopic hemostasis. The planting area of pitaya (Hylocereus polyrhizus) is expanding continuously in China in recent years, due mainly to the high economic and ecological value of this tropical fruit . However, drought has become one of the key factors restricting the growth and yield of H. polyrhizus in arid regions. This study aims to explore the characteristics of water consumption subjected to transpiration, and the influencing factors of H. polyrhizus. A Dynamax sap flow measuring system was employed to measure and monitor the sap flow velocity in seven consecutive days per month from September 2019 to August 2020. The planting field was located in the digital agricultural experimental base of Guizhou Academy of Agriculture Sciences in western China. Meanwhile, the related environmental data was simultaneously measured. The correlation of sap flow features with various factors was analyzed to quantify the water demand threshold of H. polyrhizusand water-saving irrigation. The results showed that the single-day sap flow of H. polyrhizus was dominated by a single peak curve. There was an increasing trend of sap flow after sunset, whereas, the sap flow rate varied gently from 00:00 to sunrise. Diurnal sap flow accounted for 49.60%-71.51% of the whole day, whereas, the night sap flow was relatively low. The total sap flow of H. polyrhizus sequenced as spring>winter >autumn >summer. The new shoots emerged sharply in spring, where the peak value of sap flow and average daily flow rate reached the maximum, 2.68 and 3.08 times those in the slow-growing period, while 1.65 and 1.52 times those in the flowering and fruiting period, indicating a leading role of the growth period. However, the proportion of nocturnal sap flow in spring (the average sap flow ratio was 31.05%) was significantly lower than that in other seasons (41.01%-43.65%). The proportion of sap flow velocity at night was ranked in order: summer> winter> autumn > spring. There was a significant correlation between the total night and day sap flow, where the correlation coefficient was 0.917 2. The peaks and valleys of sap flow were scattered with time. Specifically, the peaks appeared more frequently during 10:00-13:00, whereas, the valleys were more concentrated in 17:00-20:00. The sap flow rate had a very significant positive correlation with the total solar radiation (TSR), photosynthetically active radiation (PAR), whereas, extremely negatively correlated with the soil temperature (Ts), soil moisture (Ms), relative humidity (RH), and vapor pressure deficit (VPD). Additionally, the sap flow rate was negatively correlated with the TSR and PAR under a high light intensity (TSR>550 W/m²). Furthermore, the responses of sap flow velocity to each environmental factor were ranked in order: TSR >PAR >Ms>RH >Ts>VPD >Ta. There was a significant positive correlation between the total sap flow during the day and night, but the sap flow rate responded differently to temperature and relative air humidity. During the day, Ta had a weaker effect on the sap flow, while at night a significant negative effect. The RH had a negative effect on the liquid flow during the day, whereas, a positive effect at night. Consequently, there were some effects of seasonal factors on the sap flow of H. polyrhizus at night. Adequate irrigation was therefore very important in the spring for the water and fertilizer management of H. polyrhizus planted in the greenhouses.
