PurposeThis paper presents M-PoP, a method to model large, complex, and dynamic business processes. These processes have sometimes resulted from alliances of organizations (i.e. joint ventures and mergers and acquisitions) and are referred to as Processes-of-Business Processes (PoP). Due to the difficulty of modeling these dynamic processes, alliances of organizations have often lost opportunities, competitiveness, and profitability, so requiring suitable modeling methods.Design/methodology/approachThe authors proposed M-PoP that can model PoP through three views in different abstraction levels and using well-known techniques in industry and academia, mainly those from Business Process Model and Notation (BPMN). For this, M-PoP presents three main steps: identification of PoP elements, modeling of PoP, and verification of PoP models. To evaluate M-PoP, we applied it in a real-world business process in the health domain.FindingsThe evaluation results point out the capability and viability of M-PoP to deal with dynamic business processes.Research limitations/implicationsM-PoP still needs to be applied in various real-world scenarios to gather evidence of its productivity, efficiency, and scalability.Practical implicationsThis novel method could change the way organizations model their business processes and, as a consequence, it could leverage strategic business opportunities.Originality/valueM-PoP is the first method that makes it possible to model large and complex business processes and, most importantly, dynamic processes.

Mitochondria–ER contacts (MERCs), tightly regulated by numerous tethering proteins that act as molecular and functional connections between the two organelles, are essential to maintain a variety of cellular functions. Such contacts are often compromised in the early stages of many neurodegenerative disorders, including amyotrophic lateral sclerosis (ALS). TDP-43, a nuclear protein mainly involved in RNA metabolism, has been repeatedly associated with ALS pathogenesis and other neurodegenerative diseases. Although TDP-43 neuropathological mechanisms are still unclear, the accumulation of the protein in cytoplasmic inclusions may underlie a protein loss-of-function effect. Accordingly, we investigated the impact of siRNA-mediated TDP-43 silencing on MERCs and the related cellular parameters in HeLa cells using GFP-based probes for MERCs quantification and aequorin-based probes for local Ca2+ measurements, combined with targeted protein and mRNA profiling. Our results demonstrated that TDP-43 down-regulation decreases MERCs density, thereby remarkably reducing mitochondria Ca2+ uptake after ER Ca2+ release. Thorough mRNA and protein analyses did not highlight altered expression of proteins involved in MERCs assembly or Ca2+-mediated ER–mitochondria cross-talk, nor alterations of mitochondrial density and morphology were observed by confocal microscopy. Further mechanistic inspections, however, suggested that the observed cellular alterations are correlated to increased expression/activity of GSK3β, previously associated with MERCs disruption.

The Fundão dam failure in 2015 severely impaired the economy, the lives of riverine communities, and the aquatic ecosystems of the Rio Doce basin in southeast Brazil. Several contaminants, including polycyclic aromatic hydrocarbons (PAHs), were transported downstream, deposited in the estuary, and released into the Atlantic Ocean. The high concentration of PAHs in estuarine sediments may pose ecological risks and deleterious effects to benthic organisms, so here we aimed at determining the source and fate of these compounds before and after the tailings' arrival. The mean concentration of the analyzed Σ16PAHs increased from 34.05 µg kg−1 in the prefailure period to 751.77 µg kg−1 one year after the arrival of the tailing. The classification of the sediment quality changed from low to moderate contamination. Our results suggest that there was PAHs remobilization by mine tailings along the Rio Doce basin. The target analytes exhibited mostly a pyrolytic profile from fossil fuel and biomass burning. In addition to other contaminants deposited in the estuary after the arrival of the tailings, this study revealed that the profile change of PAHs in the estuary region is a consequence of the mud's erosive power. The coast of Espírito Santo state (Southeast Brazil) is recognized for its environmental arsenic (As) enrichment and, over the years, mining operations have potentialized it. We aimed to evaluate the effect of Rio Doce discharge on As inputs and the role of iron ore tailings from the Fundão dam disaster in enhancing As contamination in the marine sediment. Two scenarios were evaluated: Predisaster and Postdisaster; dry and wet conditions were considered in each period. High As concentrations were found in the Predisaster (28.44 ± 13.53 µg g−1), but a significant increase in As was remarkable during the Postdisaster in the wet season, one year after the disaster (maximum of 58.39 µg g−1; geoaccumulation index (Igeo) Class 3, moderately severe pollution). On that occasion, iron (Fe) oxy‐hydroxides from tailings were remobilized from the Rio Doce channel and deposited on the continental shelf bottom. Therefore, chemical interactions among Fe, As, and carbonates were enhanced, resulting in As and Fe coprecipitation and the trapping by carbonate adsorption. Rio Doce discharge seems to be the main factor in As inputs to the inner continental shelf when flooding do not occur previously in samplings, which allows further dispersion of contaminants, although this hypothesis should be tested further. Toxocara cati is a common roundworm of cats and wild felids and, together with T. canis, it is the main causative agent of human toxocariasis. Humans may become infected by ingestion of embryonated eggs via contaminated soil, food, or water, or by ingestion of raw or undercooked meat of paratenic hosts that are infected by Toxocara larvae. In this study, we report the detection of T. cati larvae from meat samples of ostriches and wild boars. These samples were inspected by enzymatic digestion, as part of the trichinellosis surveillance. As ostrich meat is intended for “carpaccio” preparation, a traditional Italian raw meat preparation, there is the need to make the consumption of this meat safe. For this purpose, it is recommended to freeze the meat before preparation. Our findings confirmed that T. cati larvae can contaminate muscle tissues of paratenic hosts, increasing the risk of infection due to the consumption of raw or undercooked meat.

Amyotrophic lateral sclerosis (ALS) is a motor neuron (MN) disease associated with progressive muscle atrophy, paralysis, and eventually death. Growing evidence demonstrates that the pathological process leading to ALS is the result of multiple altered mechanisms occurring not only in MNs but also in other cell types inside and outside the central nervous system. In this context, the involvement of skeletal muscle has been the subject of a few studies on patients and ALS animal models. In this work, by using primary myocytes derived from the ALS transgenic hSOD1(G93A) mouse model, we observed that the myogenic capability of such cells was defective compared to cells derived from control mice expressing the nonpathogenic hSOD1(WT) isoform. The correct in vitro myogenesis of hSOD1(G93A) primary skeletal muscle cells was rescued by the addition of a conditioned medium from healthy hSOD1(WT) myocytes, suggesting the existence of an in trans activity of secreted factors. To define a dataset of molecules participating in such safeguard action, we conducted comparative metabolomic profiling of a culture medium collected from hSOD1(G93A) and hSOD1(WT) primary myocytes and report here an altered secretion of amino acids and lipid-based signaling molecules. These findings support the urgency of better understanding the role of the skeletal muscle secretome in the regulation of the myogenic program and mechanisms of ALS pathogenesis and progression. Resumo Neste artigo é analisado o surgimento da População em Situação de Rua (PSR) no século XIX, a partir das investigações históricas de Karl Marx e Friedrich Engels. Em seguida, examina-se a relação da PSR com as crises econômicas do modo de produção capitalista, desde o século XIX até hoje, com base nos estudos de István Mészáros sobre a tendência incontrolável de autorreprodução destrutiva do capital. Por fim, apresenta-se um breve retrato histórico da origem da PSR no contexto brasileiro por meio de compilação de estudos críticos do campo das Ciências Humanas, sobretudo do Serviço Social. Este artigo se desdobra de duas pesquisas em andamento, e consiste em uma investigação qualitativa de cunho teórico realizada por meio de revisão bibliográfica sistemática nos autores clássicos e contemporâneos do marxismo. Como resultado desta pesquisa, concluiu-se que a PSR é um problema estrutural do capital, cuja efetiva superação depende do desenvolvimento de medidas anticapitalistas. Abstract This article analyzes the emergence of the homeless in the 19th century, based on the historical research of Karl Marx and Friedrich Engels. It then examines the relation of homeless to the economic crises of the capitalist mode of production, from the 19th century until today, based on István Mészáros studies on capital's uncontrollable destructive self-reproducing tendency. Finally, it brings a brief historical portrait of the origin of homeless in the Brazilian context by compiling critical studies from the field of Human Sciences, especially Social Service. This article is an offshoot of two ongoing research projects, and consists of a qualitative investigation of a theoretical nature carried out through a systematic bibliographic review of classical and contemporary Marxist authors. As a result of this research, it was concluded that homeless is a structural problem of capital, whose effective overcoming depends on the development of anti-capitalist measures.

- Abstract
Background: Delirium is thought to be common across various settings of care; however, still little research has been conducted in rehabilitation. Aim: We investigated the prevalence of delirium, its features and motor subtypes in older patients admitted to rehabilitation facilities during the three editions of the “Delirium Day project”. Methods: We conducted a cross-sectional study in which 1237 older patients (age ≥ 65 years old) admitted to 50 Italian rehabilitation wards during the three editions of the “Delirium Day project” (2015 to 2017) were included. Delirium was evaluated through the 4AT and its motor subtype with the Delirium Motor Subtype Scale. Results: Delirium was detected in 226 patients (18%), and the most recurrent motor subtype was mixed (37%), followed by hypoactive (26%), hyperactive (21%) and non-motor one (16%). In a multivariate Poisson regression model with robust variance, factors associated with delirium were: disability in basic (PR 1.48, 95%CI: 1.17–1.9, p value 0.001) and instrumental activities of daily living (PR 1.58, 95%CI: 1.08–2.32, p value 0.018), dementia (PR 2.10, 95%CI: 1.62–2.73, p value < 0.0001), typical antipsychotics (PR 1.47, 95%CI: 1.10–1.95, p value 0.008), antidepressants other than selective serotonin reuptake inhibitors (PR 1.3, 95%CI: 1.02–1.66, p value 0.035), and physical restraints (PR 2.37, 95%CI: 1.68–3.36, p value < 0.0001). Conclusion: This multicenter study reports that 2 out 10 patients admitted to rehabilitations had delirium on the index day. Mixed delirium was the most prevalent subtype. Delirium was associated with unmodifiable (dementia, disability) and modifiable (physical restraints, medications) factors. Identification of these factors should prompt specific interventions aimed to prevent or mitigate delirium.

Sulfur is an essential plant macronutrient, and its adequate supply allows an efficient root storage and sugar extractability in sugar beets (Beta vulgaris L.). In this study, we investigated the effect of changes in sulfur availability on the endophytic community structure of sugar beets. Plants were hydroponically grown in a complete nutrient solution (S-supplied), a nutrient solution without MgSO4 (S-deprived), and a nutrient solution without MgSO4 for six days and resupplied with 100 μM MgSO4 for 48 h (S-resupplied). The sulfur status was monitored by inductively coupled plasma ICP–OES, and combustion analysis together with the evaluation of microRNA395 as a biomarker for sulfate status. Metabarcoding of the bacterial 16S rRNA gene was carried out in order to determine leaf endophytic community structure. The Shannon diversity index significantly differed (p < 0.05) between sulfate-supplied and sulfate-deprived seedlings. Validation by Real-Time PCR showed a significant increase (p < 0.05) of Burkholderia spp. in sulfate-deprived plants as compared to sulfate-supplied ones. The study sheds new light on the effects of nutrient deficiency on the microbiome of sugar beet plants.

Amyotrophic lateral sclerosis (ALS) is a progressive neurodegenerative disease whose pathophysiology is largely unknown. Despite the fact that motor neuron (MN) death is recognized as the key event in ALS, astrocytes dysfunctionalities and neuroinflammation were demonstrated to accompany and probably even drive MN loss. Nevertheless, the mechanisms priming astrocyte failure and hyperactivation are still obscure. In this work, altered pathways and molecules in ALS astrocytes were unveiled by investigating the proteomic profile and the secreted metabolome of primary spinal cord astrocytes derived from transgenic ALS mouse model overexpressing the human (h)SOD1(G93A) protein in comparison with the transgenic counterpart expressing hSOD1(WT) protein. Here we show that ALS primary astrocytes are depleted of proteins—and of secreted metabolites—involved in glutathione metabolism and signaling. The observed increased activation of Nf-kB, Ebf1, and Plag1 transcription factors may account for the augmented expression of proteins involved in the proteolytic routes mediated by proteasome or endosome–lysosome systems. Moreover, hSOD1(G93A) primary astrocytes also display altered lipid metabolism. Our results provide novel insights into the altered molecular pathways that may underlie astrocyte dysfunctionalities and altered astrocyte–MN crosstalk in ALS, representing potential therapeutic targets to abrogate or slow down MN demise in disease pathogenesis.

Parvovirus B19 (B19V) infection in pregnancy is mostly asymptomatic, but can cause complications including abortion and fetal hydrops. Although its infection is ubiquitous, seroprevalence among pregnant women varies according to different geographical areas. Since seroprevalence data in Italy are limited, the prevalence of antibodies and DNA in pregnant women was evaluated retrospectively, correlating the clinical situation of mothers and newborns. One thousand eight hundred and ninety-three sequential sera were examined from pregnant women (60.8% in the first trimester, 16.6% in the second one, and 22.6% in the third one, respectively) for anti-B19V IgG and IgM (confirmed by immunoblot); 1402 (74.1%) were of Italian origin and 491 (25.9%) non-Italian women. Molecular tests were used to search for viral genome. One thousand three hundred and fifteen (69.5%) samples were IgG-positive, 21 (1.1%) IgM-positive, and 578 (30.5%) nonimmune. The difference in IgG seroprevalence between Italian (71.1%) and non-Italian women (64.8%) was statistically significant. Purpose To date, there is no consensus on how to semi-quantitatively assess brain amyloid PET. Some approaches use late acquisition alone (e.g., ELBA, based on radiomic features), others integrate the early scan (e.g., TDr, which targets the area of maximum perfusion) and structural imaging (e.g., WMR, that compares kinetic behaviour of white and grey matter, or SI based on the kinetic characteristics of the grey matter alone). In this study SUVr, ELBA, TDr, WMR, and SI were compared. The latter — the most complete one — provided the reference measure for amyloid burden allowing to assess the efficacy and feasibility in clinical setting of the other approaches. Methods We used data from 85 patients (aged 44–87) who underwent dual time-point PET/MRI acquisitions. The correlations with SI were computed and the methods compared with the visual assessment. Assuming SUVr, ELBA, TDr, and WMR to be independent measures, we linearly combined them to obtain more robust indices. Finally, we investigated possible associations between each quantifier and age in amyloid-negative patients. Results Each quantifier exhibited excellent agreement with visual assessment and strong correlation with SI (average AUC = 0.99, ρ = 0.91). Exceptions to this were observed for subcortical regions with ELBA and WMR (ρELBA = 0.44, ρWMR = 0.70). The linear combinations showed better performances than the individual methods. Significant associations were observed between TDr, WMR, SI, and age in amyloid-negative patients (p Conclusion Among the other methods, TDr came closest to the reference with less implementation complexity. Moreover, this study suggests that combining independent approaches gives better results than the individual procedure, so efforts should focus on multi-classifier systems for amyloid PET. Finally, the ability of techniques integrating blood perfusion to depict age-related variations in amyloid load in amyloid-negative subjects demonstrates the goodness of the estimate.
