Two acylphosphatase isoenzymes have been purified from Lamna nasus muscle, and their complete amino acid sequences have been determined. The former (E1) consists of 99 amino acid residues, while the latter (E2) consists of 102 residues. Both are acetylated at their N termini. E1 has the FFRK active site motif characteristic of all common-type acylphosphatase isoenzymes, whereas E2 contains the CFRM active site motif characteristic of all muscle-type acylphosphatase isoenzymes. They have quite similar kinetic properties. The comparison of sequences of fish E1 and E2 isoenzymes with other known mammalian and bird acylphosphatases reveals that the E2 isoenzyme has an N terminus tail, four residues long, similar to those previously found in all known bird species muscle-type isoenzymes. Among organ-common-type acylphosphatases about 50% of residues are completely conserved, whereas about 60% of muscle-type acylphosphatase residues are completely conserved, indicating that the latter type of isoenzyme has a slower evolutionary rate than the former. The sequences of E1 and E2 acylphosphatases from L. nasus represent the first primary structures of this kind of enzyme determined among fish species.

Alzheimer’s disease (AD) is a degenerative disorder of the central nervous system which causes progressive cognitive decline during mid to late adult life. The primary causes of AD have not yet been identified. Mutated genes have been located on chromosomes 21,14, and 1, leading to the early onset familial form of the disease (EOFAD) (Shellenberg, 1995; Sorbi, 1993). The first presenile AD gene encoding for β-amyloid precursor protein (APP) was identified in 1991 (Goate et al., 1991). The most frequent pathogenic mutation in exon 17, a Val-Ile substitution at codon 717, has been described in more than 10 families (Levy-Lahad et al., 1996). The largest portion of early onset FAD cases have been associated with mutations in Presenilin 1 (PS1) gene and Presenilin 2 (PS2) gene (Van Broeckhoven, 1995; Rogaev et al., 1995; Levy-Lahad et al., 1995). Fibroblasts from patients and pre-symptomatic carriers of mutations in PS1 and PS2 contain increased amounts of Aβ 1-42 peptide (Scheuner et al., 1996). Moreover, the allele E4 of APOE gene has been associated with increased risk for late-onset familial form of AD (Saunders et al., 1993). Apolipoprotein E is the first identified genetic susceptibility factor for sporadic AD. The familial AD form account for 10% of all AD cases (Van Broeckhoven, 1995).

The structural and catalytic importance of the 15-21 and 42-45 loop residues of the acylphosphatase muscular isoenzyme has been investigated by oligonucleotide-directed mutagenesis. Seven mutants involving conserved residues of the two loops have been prepared and characterized for structural, kinetic, and stability features by using different spectroscopic techniques and compared to the wild-type enzyme. The results are discussed in light of the crystal structure of the highly homologous common type acylphosphatase [Thunnissen et al. (1997) Structure 5, 69-79]. A differential role of the two loops has emerged: the 15-21 and the 42-45 loops appear mainly involved in active site formation and enzyme structural stabilization, respectively. These conclusions are supported by a strong impairment of the catalytic efficiency, in terms of enzymatic activity and substrate binding capability, for most of the 15-21 loop mutants. In particular, the Gly15Ala mutant is completely inactive and displays a native-like overall fold, indicating that the correct geometry of the 15-21 loop is an essential requisite for optimal enzymatic catalysis. Instead, the Gly45Ala mutant, though revealing unchanged catalytic properties, shows a considerably reduced conformational stability, as judged by circular dichroism and 1H NMR spectroscopy. This finding confirms previous results relative to Thr42 and Thr46 residues [Taddei et al. (1996) Biochemistry 35, 7077-7083] underlining the structural importance of the 42-45 loop as a linker for the two beta alpha beta units constituting the overall enzyme structure.

The subcellular distribution of both muscle and common type acylphosphatases was studied during apoptosis. Both isoenzymes were previously shown to have in vitro nucleolytic activity having a pH optimum at 6.8. In this paper we demonstrate a nuclear migration of the muscle type isoenzyme in response to various apoptotic stimuli either in K562 or in Jurkat cells, while the common type acylphosphatase isoform remains cytoplasmatic under the same conditions. Furthermore, we present evidences of a direct in vivo interaction between muscle acylphosphatase and two other DNAses of about 60 and 80 kDa in size. Our results are consistent with an in vivo involvement of the muscle isoform in apoptosis, possibly as a part of a multimeric protein complex that binds and hydrolyses DNA during this process.

Acylphosphatase is one of the smallest enzymes known (about 98 amino acid residues). It is present in organs and tissues of vertebrate species as two isoenzymes sharing over 55% of sequence homology; these appear highly conserved in differing species. The two isoenzymes can be involved in a number of physiological processes, though their effective biological function is not still certain. The solution and crystal structures of different isoenzymes are known, revealing a close packed protein with a fold similar to that shown by other phosphate-bind ing proteins. The structural data, together with an extended site-directed mutagenesis investigation, led to the identification of the residues involved in enzyme catalysis. However, it appears unlikely that these residues are able to perform the full catalytic cycle: a substrate-assisted catalytic mechanism has therefore been proposed, in which the phosphate moiety of the substrate could act as a nucleophile activating the catalytic water molecule.

Levels of free intracellular calcium have been measured on two cell lines of cultured human fibroblasts carrying the genetic lesions occurring in Duchenne and Becker dystrophies. SUMMARY A novel enzymatic activity on nucleic acids was discovered in both muscle type (MT) and erythrocyte or common type (CT) isoforms of acylphosphatase, an enzyme that was previously known as a hydrolase (E.C.3.6.1.7). Both' deoxyribonucleic and ribonucleic hydrolitic activity were assayed on a variety of substrates. Our results demonstrate that acylphosphatase possesses both Mg ++ dependent deoxyribonuclease and ribonuclease acivities, at pH ranging from 5.0 to 6.8. Furthermore, we present evidences, for both isoenzymatic forms, of the coexistence of exonucleolytic and endonucleolytic activities on DNA.

Acylphosphatase (AcPase), an enzyme that modulates the activity of Ca(2+)-ATPase by hydrolysing its phosphorylated moiety, has been found to be significantly higher in cultured skin fibroblasts from donors affected by early onset familial Alzheimer's disease (EOFAD) with PS-1 and PS-2 gene mutations. Of the two known isoenzymes of acylphosphatase, only the erythrocyte one accounts for the total increase in activity. No relevant alteration was observed in phosphotyrosine phosphatase activity (PTPase), in Ca(2+)-ATPase and Na+, K(+)-ATPase activities of the same cells as compared to age-matched controls. This finding could suggest a possible explanation for the calcium-dependent biochemical alterations previously described in Alzheimer's disease fibroblasts.

Asn41, Thr42, and Thr46 are invariant residues in both muscle and erythrocyte acylphosphatases isolated so far. Horse muscle acylphosphatase solution structure suggests their close spatial relationship to Arg23, the main substrate binding site. The catalytic and structural role of such residues, as well as their influence on muscle acylphosphatase stability, was investigated by preparing several gene mutants (Thr42Ala, Thr46Ala, Asn41Ala, Asn41Ser, and Asn41Gln) by oligonucleotide-directed mutagenesis. The mutated genes were cloned and expressed in Escherichia coli, and the mutant enzymes were purified by affinity chromatography and investigated as compared to the wild-type enzyme. The specific activity and substrate affinity of Thr42 and Thr46 mutants were not significantly affected. On the contrary, Asn41 mutants showed a residual negligible activity (about 0.05-0.15% as compared to wild-type enzyme), though maintaining an unchanged binding capability of both substrate and inorganic phosphate, an enzyme competitive inhibitor. According to the 1H nuclear magnetic resonance spectroscopy and circular dichroism results, all mutants elicited well-constrained native-like secondary and tertiary structures. Thermodynamic parameters, as calculated from circular dichroism data, demonstrated a significantly decreased stability of the Thr42 mutant under increasing temperatures and urea concentrations. The reported results strongly support a direct participation of Asn41 to the enzyme catalytic mechanism, indicating that Asn41 mutants may well represent a useful tool for the investigation of the enzyme physiological function by the negative dominant approach.

Ser-Ala and Ser-Ala-Ser-Ala C-terminus elongated (delta+2 and delta+4, respectively) and two C-terminus deleted (delta-2 and delta-3) muscle acylphosphatase mutants were investigated to assess the catalytic and structural roles of the C-terminal region. The kinetic analysis of these mutants shows that the removal of two or three C-terminal residues reduces the catalytic activity to 7% and 4% of the value measured for the wild-type enzyme, respectively; instead, the elongation of the C-terminus does not significantly change the enzyme behaviour. 1H Nuclear magnetic resonance spectroscopy indicates that all mutants display a native-like fold though they appear less stable, particularly delta-2 and delta-3 mutants, as compared to the wild-type enzyme. Such destabilisation of the C-terminal modified mutants is further confirmed by urea inactivation experiments. The results here presented account for an involvement of the C-terminal region in the stabilisation of the three-dimensional structure of acylphosphatase, particularly at the active-site level. Moreover, a participation of the C-terminal carboxyl group to the catalytic mechanism can be excluded.

Acylphosphatase purified from heart muscle actively hydrolyzes the phosphoenzyme intermediate of cardiac sarcoplasmic reticulum Ca(2+)-ATPase. This effect was evident with acylphosphatase concentrations (up to 100 units/mg sarcoplasmic reticulum protein) that fall within the physiological range, and the low value of the apparent Km, on the order of 10(-7)M, suggests a high affinity towards this special substrate. Moreover, acylphosphatase addition to sarcoplasmic reticulum vesicles significantly enhanced the rate of Ca(2+)-dependent ATP hydrolysis. Maximal stimulation, observed with 100 units/mg vesicular protein, resulted in an ATPase activity which was about two folds over basal value. The same acylphosphatase concentration increased at a similar extent the rate of ATP driven Ca2+ influx into sarcoplasmic reticulum vesicles. Taken together these findings lead to suppose that acylphosphatase, owing to its hydrolytic activity, induces an accelerated turnover of the phosphoenzyme intermediate, whence an overall stimulation of heart sarcoplasmic reticulum Ca2+ pump, affecting both ATP hydrolysis and Ca2+ influx.

Three independent cDNAs coding for the erythrocyte isoform of human acylphosphatase were isolated and characterized. All the clones were incomplete at the 5′ end, but Northern blot analysis using the cDNA as a probe showed the presence of an unusually long mRNA 5′-untranslated region. The transcript was present in a variety of human cell lines of different origins, although at different levels. Southern blot analysis on DNA from different individuals revealed a simple hybridization pattern. Large amounts of pure enzyme with kinetic characteristics very similar to those of the native protein were expressed in E. coli.

Acylphosphatase, purified from cardiac muscle, catalyzes the hydrolysis of the phosphorylated intermediate of heart sarcolemmal Na+,K(+)-ATPase. This effect was remarkable even using acylphosphatase amounts (100-300 units/mg of membrane protein) near the lower limit of the physiological range; besides the low value of the apparent Km, on the order of 10(-7) M, indicates that the enzyme has a high affinity for this special substrate. The results of a dot-immunobinding assay suggest the possibility of an interaction between acylphosphatase and native, undenaturated Na+,K(+)-ATPase. Moreover, when added to sarcolemmal vesicles, acylphosphatase was found to affect the functional properties of the Na+,K+ pump with regard to the rate of both ATP hydrolysis and cation transport. However, while ATPase activity and Na+ uptake were stimulated, the last at a greater extent, the active K+ transport was inhibited, so that the Na+/K+ ratio, which was calculated as 1.50 without acylphosphatase, rose to 6.68 in the presence of 300 units/mg of vesicle protein of this enzyme. Taken together, the reported results indicate that acylphosphatase, because of its hydrolytic activity on the phosphoenzyme intermediate, induces a sort of "uncoupling" effect on the heart sarcolemmal membrane Na+,K+ pump. Possible mechanisms for such an effect, which suggests a potential role of acylphosphatase in the control of this active transport system, are discussed.

Enzymatic activity and structure of N-terminus truncated and C-terminus substituted muscle acylphosphatase mutants were investigated by kinetic studies under different conditions and 1H NMR spectroscopy, respectively. The N-terminus truncated mutant lacked the first six residues (delta 6), whereas arginine 97 and tyrosine 98 were replaced by glutamine giving two C-terminus substituted mutants (R97Q and Y98Q, respectively). All acylphosphatase forms were obtained by modifications of a synthetic gene coding for the human muscle enzyme which was expressed in E. coli. The delta 6 deletion mutant elicited a reduced specific activity and a native-like structure. The kinetic and structural properties of R97Q and Y98Q mutants indicate a possible role of Arg-97 in the stabilisation of the active site correct conformation, most likely via back-bone and side chain interactions with Arg-23, the residue involved in phosphate binding by the enzyme. This study also suggests a possible involvement of Tyr-98 in the stabilisation of the acylphosphatase overall structure.

A new aromatic acyl phosphate, 2-methoxybenzoyl phosphate, has been synthesized. The compound shows an intrinsic fluorescence; it displays an intense emission band at 390 nm upon excitation in the near UV region. This band practically disappears after hydrolysis of the product. On the other hand, the product displays differences in the near UV absorption spectra measured before and after hydrolysis. The delta epsilon at 301 nm is 2720 M-1 cm-1, a value that is 4.3-fold higher than that of benzoyl phosphate (the usual substrate for acylphosphatase assay) at 283 nm. The main kinetic parameters of three different acylphosphatase molecular forms (the muscular isoenzyme and two subtypes of the organ common isoenzyme) were determined using both benzoyl phosphate and 2-methoxybenzoyl phosphate as substrates, and then compared. These kinetic data and the UV absorption and fluorescence properties of 2-methoxybenzoyl phosphate suggest that this compound has better substrate features than benzoyl phosphate, and can be used for both high sensitivity continuous fluorimetric and UV absorption spectrophotometric assays of acylphosphatase.

Maturation of prokaryotic hydrogenase involves several protein factors, among which is the accessory protein HypF, which hosts the consensus sequence of acylphosphatases and a sequence motif common to proteins catalyzing O-carbamoylations. The specific functions of HypF are largely unknown, although it has been observed that CN(-) and CO ligands at the hydrogenase Ni,Fe active centre originate from carbamoylphosphate. The HypF N-terminal domain (91 residues, acylphosphatase-like domain) has been crystallized in two different crystal forms belonging to the orthorhombic P2(1)2(1)2(1) space group (unit-cell parameters a = 35.5, b = 59.8, c = 87.6 A) and to the rhombohedral space group R32 (unit-cell parameters a = b = 58.1, c = 155.6 A in the hexagonal setting).

Background: Thermophilic enzymes are often less active than their mesophilic homologues at low temperatures. One hypothesis to explain this observation is that the extra stabilizing interactions increase the rigidity of thermophilic enzymes and hence reduce their activity. Here we employed a thermophilic acylphosphatase from Pyrococcus horikoshii and its homologous mesophilic acylphosphatase from human as a model to study how local rigidity of an active-site residue affects the enzymatic activity. Methods and Findings: Acylphosphatases have a unique structural feature that its conserved active-site arginine residue forms a salt-bridge with the C-terminal carboxyl group only in thermophilic acylphosphatases, but not in mesophilic acylphosphatases. We perturbed the local rigidity of this active-site residue by removing the salt-bridge in the thermophilic acylphosphatase and by introducing the salt-bridge in the mesophilic homologue. The mutagenesis design was confirmed by x-ray crystallography. Removing the salt-bridge in the thermophilic enzyme lowered the activation energy that decreased the activation enthalpy and entropy. Conversely, the introduction of the salt-bridge to the mesophilic homologue increased the activation energy and resulted in increases in both activation enthalpy and entropy. Revealed by molecular dynamics simulations, the unrestrained arginine residue can populate more rotamer conformations, and the loss of this conformational freedom upon the formation of transition state justified the observed reduction in activation entropy. Conclusions: Our results support the conclusion that restricting the active-site flexibility entropically favors the enzymatic activity at high temperatures. However, the accompanying enthalpy-entropy compensation leads to a stronger temperature-dependency of the enzymatic activity, which explains the less active nature of the thermophilic enzymes at low temperatures. © 2011 Lam et al., published_or_final_version

The oxidized form of the mercuric ion binding protein MerP has been studied by two-dimensional NMR. MerP, which is a periplasmic water-soluble protein with 72 amino acids, is involved in the detoxification of mercuric ions in bacteria with resistance against mercury. The mercuric ions in the periplasmic space are first scavenged by the MerP protein, then transported into the cytoplasm by the membrane-bound transport protein MerT, and finally reduced to elementary (nontoxic) mercury by the enzyme mercuric reductase. In this work, the 1H NMR spectrum of oxidized MerP (closed disulfide bridge) has been assigned by using homonuclear 2D NMR techniques. The secondary structure and global fold have been inferred from the nuclear Overhauser effect (NOE) data. The secondary structure comprises four beta-strands and two alpha-helices, in the order beta 1 alpha 1 beta 2 beta 3 alpha 2 beta 4. The protein folds into an antiparallel beta-sheet, beta 2 beta 3 beta 1 beta 4, with the two antiparallel helices on one side of the sheet. The folding topology is similar to that of acylphosphatase, the activation domain of porcine pancreatic procarboxypeptidase B, the DNA-binding domain of bovine papillomavirus-1 E2 and the RNA-binding domains of the U1 snRNP A and hnRNP C proteins. However, there is no structural similarity between MerP and other bacterial periplasmic binding proteins.

Xenopus laevis oocytes are physiologically arrested in the G2/M phase border of the first meiotic division. A number of different stimuli can trigger off the re-entry into the cell cycle as a consequence of activation of either membrane-dependent or -independent intracellular signals. This system has been widely used to study signal transduction mechanisms induced by hormones. Among those more intensively researched, special attention has been devoted to elucidate the mechanism of activation induced by progesterone. However, despite intense efforts to understand the intracellular signalling mechanism of progesterone, a clear notion of the most relevant events involved in this process has not yet been elucidated. We provide evidence that acylphosphatase, an enzyme responsible for the regulation of membrane pumps in eukaryotic cells, synergizes with progesterone for induction of oocyte maturation. We deduced that this synergism may be related to the regulation of intracellular Ca2+ levels for several reasons: (1) maturation of oocytes by extracellular Ca2+ is blocked by acylphosphatase; (2) both progesterone and acylphosphatase drastically reduced Ca2+ uptake; (3) progesterone-induced maturation does not depend on a rise in intracellular Ca2+, since microinjection of EGTA, a calcium chelator, does not affect maturation induced by progesterone.

Acylphosphatase is a widespread cytosolic enzyme that catalyzes the hydrolysis of carboxylphosphate bond compounds. Among natural acylphosphates hydrolyzed by the enzyme are 3-phosphoglyceroylphosphate (1) carbamoylphosphate (2), succinoylphosphate (3).

An acylphosphatase has been purified from turkey muscle in a rapid and high-yield way. The enzyme has been characterized for structural, kinetic, and immunological parameters, as well as with regard to its stability to thermal, urea, and phenylglyoxal inactivation. The enzyme is quite different from the turkey muscular isoenzyme, and shows structural and kinetic properties that are very similar to those previously reported for the erythrocyte isoenzyme from human erythrocytes and from chicken muscle. From the data reported it appears that this enzyme corresponds to the acylphosphatase erythrocyte isoenzyme. Unlike the erythrocyte isoenzymes studied so far, this enzyme is able to cross-react with antibodies that are raised against the muscular isoenzyme.

Distributions of acylphosphatase isozymes among organs of several animal species were investigated. Organ extracts of pig and chicken were treated with isozyme-specific antibodies, subjected to electrophoresis on a polyacrylamide gel, then the gel was stained for acylphosphatase activity. Both animals showed three activity bands; one band was named common type isozyme because of its wide distribution in testis, muscle, brain, heart, spleen, kidney, liver, and erythrocyte, and the other two bands were named muscle type isozymes because of their localization in skeletal muscle. This classification was supported by selective and quantitative reactions of the isozymes to the isozyme-specific antibodies. Because the two bands of the muscle type have the same amino acid sequence and differ only in modifications on an SH group, it is suggested that pig and chicken have only the two major types of acylphosphatase. This conclusion was supported by similar experiments on dog, human, rabbit, and pigeon.

We studied the effect of human acylphosphatase on the activity of human erythrocyte membrane Ca2(+)-ATPase. Both the acylphosphatase that is contained in hemolysate and the purified enzyme isolated from red blood cells were able to stimulate Ca2(+)-ATPase activity in erythrocyte membranes. Given the same acylphosphatase activity, however, the hemolysate showed higher stimulatory effect than the purified enzyme. Acylphosphatase stimulation was additive to that induced by calmodulin, thus indicating that acylphosphatase acts in a calmodulin-independent manner. Trifluoperazine, a calmodulin antagonist, did not inhibit acylphosphatase-induced stimulation of Ca2(+)-ATPase activity. Acylphosphatase significantly decreased the rate of Ca2+ influx into inside-out erythrocyte membrane vescicles, thus acting as Ca2+ pump inhibitor. Taken together these findings indicate that acylphosphatase is a soluble, non-calmodulin activator of erythrocyte membrane Ca2(+)-ATPase and might be involved in the control of calcium transport across the plasma membrane.

Eukaryotic nuclear RNA binding proteins share a common sequence motif thought to be implicated in RNA binding. One of the two domains present in A1 hnRNP protein, has been modelled by homology in order to make a prediction of the main features of the RNA binding site. Acyl-phosphatase (EC 3.6.1.7) was selected as template for the modeling experiment. The predicted RNA binding site is a β-sheet containing the two RNP consensus sequences as well as lysines and arginines conserved among the family.

To explore a possible role of acylphosphatase in the regulation of energy metabolism, we measured this enzyme's activity and content in skeletal muscle, liver and erythrocytes of normal and tri-iodothyronine treated rabbits. Besides acylphosphatase we assayed (Na+ + K+)-ATPase, Ca2(+)-ATPase and several enzymes of carbohydrate metabolism. Acylphosphatase activity in erythrocytes rose steadily during treatment with triiodothyronine (25 micrograms/Kg per day for 5 weeks), and its increase occurred earlier and was much more pronounced than that of other soluble enzymes. In erythrocytes of treated animals (Na+ + K+)-ATPase declined whereas Ca2(+)-ATPase activity increased, in agreement with previously reported findings. In muscle and liver of the treated animals acylphosphatase activity was about twice as high as in the controls; in these tissues we found also increased activities for (Na+ + K+)-ATPase, fructose-1,6-bisphosphatase and glucose-6-phosphatase. In any case, among the enzymes we examined, acylphosphatase was one of the most strongly and regularly stimulated by the treatment. Furthermore we observed, through an immunochemical procedure, that there was a congruence between increases in acylphosphatase activity and content. On the basis of these results we conclude that the rise in acylphosphatase levels in treated animals is probably due to its increased biosynthesis. The possible significance of these findings in the metabolic modifications associated with hyperthyroidism are discussed.

BIOCHEMISTRY In fully folded proteins, hydrophobic amino acids are hidden in the protein core. But when a protein is unfolded or partially folded, these residues render it prone to aggregation. Chiti et al. now provide much-needed insights into the molecular basis of aggregate formation. They introduced point mutations in the protein acylphosphatase (AcP) and determined the relative aggregation rates of the mutants. Mutations in two particular regions of the protein, which possess high hydrophobicity and propensity to form β-sheet structure, led to increased rates of aggregate formation. Mutational “hot spots” in protein misfolding diseases may thus result from changes in the aggregation behavior. The regions determining the aggregation rate are distinct from those that determine protein folding. Only a small number of residues is involved in setting the scene for aggregation, raising the hope that aggregation is governed by relatively simple principles. — JU Nature Struct. Sequence-specific assignment of 1 H nuclear magnetic resonance spectra of acylphosphatase (EC 3.6.1.7) isolated from rabbit skeletal muscle have made it possible to identify short distance constraints from nuclear Overhauser enhancement spectra, to evaluate spin-spin coupling constants of many backbone amide hydrogens and to assess their slow exchange with deuterons in 2 H 2 O solution. Analysis of these data show that the major regular secondary structure of the enzyme consists of five extended β-strands, four of which are arranged in an antiparallel β-sheet, while the fifth is attached parallel. A helix consisting of 11 residues has also been identified. Consideration of additional distance constraints between sequentially remote residues has allowed us to give an outline of the overall fold of the protein.

Acylphosphatase (acylphosphate phosphohydrolase, EC 3.6.1.7) was purified from guinea-pig muscle by a procedure involving immuno-affinity chromatography and a subsequent ion-exchange chromatography. This purification technique gave an overall yield of about 60% and permitted the isolation of three molecular forms with acylphosphatase activity, with a distribution greatly resembling those found in horse and turkey muscle. The main form appears to be very similar to the corresponding form in horse and turkey muscle, as indicated by amino acid composition, end-group analysis, the presence of glutathione as a mixed disulphide in almost the same stoichiometric ratio and kinetic analysis. From turnover data, the main form of acylphosphatase in guinea-pig muscle exhibits a degradation constant of 0.10 day-1, corresponding to a half-life of 6.8 days. These values are very close to those found for muscle total soluble proteins.

Carbamoyl phosphate, an intermediate metabolite of ammonia, was found to be markedly hydrolyzed in the rat brain tissues. This activity was demonstrated with light and electron microscopy certainly being indicated as enzymatic nature, by capturing released phosphate through the activity with lead ions present in the incubating medium. The reaction products were localized mostly in the endoplasmic reticulum and nuclear envelope of the nerve and glial cells. Inner cisternae of the Golgi apparatus were also occasionally stained. The antigenic properties of acylphosphatase are compared with its various sequential characteristics (hydrophobicity, chemical shift of the main-chain 1H-NMR resonances, numbers and intensities of the nuclear Overhauser enhancements, hydrogen-deuterium exchange and sequential arrangement of the secondary structure units). The discussion is based on the complete sequential assignment of the 1H-NMR spectrum and the knowledge of the three-dimensional fold of the protein obtained by NMR spectroscopy from distance geometry calculations. Regions with very different degrees of mobility can be distinguished. It is found that all major antigenic sites are located in the most mobile surface loops.

Horse muscle acylphosphatase (EC 3.6.1.7) was found to hydrolyze the labeled phosphorylated intermediate of (Ca2+ + Mg2+)-ATPase from rabbit muscle. In addition, the phosphorylated peptides obtained by pepsin digestion of the labeled phosphorylated microsomes were completely hydrolyzed by acylphosphatase. These findings suggest a possible regulatory role of this enzyme in vivo on the calcium transport process by sarcoplasmic reticulum.
