RIVER sediments ,ECOSYSTEM health ,DRUG resistance in bacteria ,MICROBIAL communities ,BACTERIAL communities ,NUTRIENT cycles - Abstract
Introduction: The Brahmaputra, a major transboundary river of the Himalayas flowing predominantly through Northeast India, particularly Assam, is increasingly endangered by contamination due to rapid urbanization and anthropogenic pressures. These environmental changes pose significant risks at the microbial level, affecting nutrient cycling and productivity, and thereby impacting river ecosystem health. The next-generation sequencing technology using a metagenomics approach has revolutionized our understanding of the microbiome and its critical role in various aquatic environments. Methods: The present study aimed to investigate the structure of the bacterial community and its functional potentials within the sediments of the Brahmaputra River, India, using high-throughput shotgun metagenomics. Additionally, this study sought to explore the presence of antimicrobial resistance genes in the river's sediment. Results and discussion: Shotgun metagenomics revealed a diverse bacterial community comprising 31 phyla, 52 classes, 291 families, 1,016 genera, and 3,630 species. Dominant phyla included Pseudomonadota (62.47–83.48%), Actinobacteria (11.10–24.89%), Bacteroidetes (0.97–3.82%), Firmicutes (0.54–3.94%), Cyanobacteria (0.14–1.70%), and Planctomycetes (0.30–0.78%). Functional profiling highlighted significant involvement in energy metabolism, amino acid and central carbon metabolism, stress response, and degradation pathways, emphasizing the microbial community's role in ecosystem functioning and resilience. Notably, 50 types of antibiotic resistance genes (ARGs) were detected, with resistance profiles spanning multidrug, aminoglycoside, β -lactam, fluoroquinolone, rifampicin, sulfonamide, and tetracycline classes. Network analysis underscored the intricate relationships among ARG subtypes, suggesting potential mechanisms of resistance propagation. Furthermore, plasmid-related genes and 185 virulence factor genes (VFGs) were identified, indicating additional layers of microbial adaptation and potential pathogenicity within the river sediments. This comprehensive microbial and functional profiling of the Brahmaputra's sediment metagenome provides crucial insights into microbial diversity, resistance potential, and ecological functions, offering a foundation for informed management and mitigation strategies to preserve river health and mitigate pollution impacts. Papillary muscle rupture is commonly linked to ischemic events, occurring typically postmyocardial infarction. However, iatrogenic and nonischemic etiologies can play a role in papillary muscle rupture, including infective endocarditis. The case presented herein is an 85-year-old female with a history of heart failure with preserved ejection fraction (HFpEF), who presented with progressive dyspnea despite furosemide therapy, and was discovered to have a flail posterior mitral valve leaflet secondary to papillary muscle rupture associated with Streptococcus lutetiensis endocarditis. Transthoracic echocardiography revealed flail posterior mitral valve leaflet with newly appreciated severe wide-open mitral regurgitation. Emergent mitral valve repair was performed with a successful MitraClip placement. Patient had an atypical presentation without ischemic symptomology or electrocardiogram (EKG) changes indicative of ischemia. Despite transesophageal echocardiography, not revealing vegetations, infective endocarditis was presumed due to presence of Streptococcus lutetiensis bacteremia, osteomyelitis, elevated inflammatory markers, and clinical presentation. Papillary muscle rupture results in severe mitral regurgitation that is a medical emergency that requires prompt diagnosis and management from a multidisciplinary team. Therefore, it is imperative to ensure clinical insight into the multiple etiologies of papillary muscle rupture to provide timely treatment. [ABSTRACT FROM AUTHOR]

Disease diagnosis is most challenging task today. Different datasets are available in web source that contains important features to diagnose the diseases. This paper explores different classification algorithms on medical liver bench mark datasets like BUPA and Indian Liver patient dataset (ILPD). The ILPD is best fit for the model and also gives high classifier accuracy. In proposed model the following classifiers like Naive Bayes (NB), support vector machine (SVM), K-nearest neighbor (KNN), decision tree (DT), and random forest (RF) classification, multi-layer perceptron (MLP), artificial neural network (ANN), deep belief network (DBN) and probabilistic neural network (PNN) are used. The results shown that ILPD is best dataset for all classifiers and RF classification in particular is best classifier. [ABSTRACT FROM AUTHOR]

A new series of 1,2,3-triazole skeleton incorporated pyrrole derivatives (11a–j) were developed and their chemical structures were confirmed by analytical data. Further, the anticancer profile of these newly derived compounds 11a–j was assessed against four types of human cancer cell lines such as human breast cancer (MCF-7), lung cancer (A549), colon cancer (Colo-205) and ovarian cancer (A2780) by employing of the MTT method and was compared with etoposide used as a positive control. Most of the examined derivatives displayed moderate to good activity compared with the positive control. Among them, five compounds 11a, 11b, 11c, 11d, and 11e showed more potent activity. Particularly, compound 11a showed superior activity. [ABSTRACT FROM AUTHOR]

In the present study, the DNA barcoding of the nematode parasite infecting Ompok bimaculatus and Nemacheilus anguilla fish species was carried out in Barvi Reservoir, Maharashtra. To ascertain the taxonomic status of these nematode parasites, an 18S gene marker was used. Accurate identification of fish parasites is essential to formulate preventive strategies and to study host–environment relations. The present study did barcoding of the nematode parasites of the fishes caught from the Barvi Reservoir using the nuclear 18S rDNA (SSU) sequence. The nuclear 18S rDNA (SSU) was amplified into two overlapping amplicons and sequenced to identify the species based on the sequence similarity with the NCBI GenBank database. The present study sequences (both fragments) showed 98% similarity with the species of Eustrongylides. The average genetic distance value between the present study sample and species of Eustrongylides was 0.003. In the phylogenetic tree also, the sequence was clustered with the species of Eustrongylides with significant bootstrap values. The present study identified the nematode parasite of the fish caught from the Barvi Reservoir, as species of Eustrongylides. The species‐level identification could not be possible due to the insufficient/lack of reference sequences in the database. It indicates the knowledge gap concerning the species‐specific molecular markers for nematode parasites of the fish. [ABSTRACT FROM AUTHOR]

Background: Traditionally, Usnea genus has significant uses in the treatment of swelling and tumors in Africa and Asia. The aim of the present study was to investigate the chemical constituents present in the acetone extract (AE) of Usnea subfloridana Stirton and also to evaluate their anti-inflammatory and anti-gout effects. Methods: Isolation and characterization of secondary metabolites from AE were evaluated by chromatography and spectral studies. Anti-inflammatory activities were assessed through cyclooxygenase (COX1 and COX2) and 5-lipooxygenase (5-LOX) enzyme inhibition assays, while anti-gout effects were evaluated by xanthine oxidase (XO) inhibition assay. Results: The existence of five known depsidones, identified as galbinic acid (1), conprotocetraricacid (2), constictic acid (3), salazinic acid (4), and lobaric acid (5), were exposed by chemical investigation of AE and confirmed by spectral data. Using in vitro enzyme inhibition assays, it was noticed that all the isolates showed dose-dependent activity against all the tested enzymes. Mainly, compounds 2 and 5 showed better inhibition efficiency on COX2 enzyme with the IC50of 7.17±1.07 and 7.01±0.94 nM, respectively, than the reference drug indomethacin (7.3±0.65nM). Furthermore, all isolates exhibited potent inhibition effects on the XO enzyme. Conclusion: The results indicated that U. subfloridana can be a favorable natural source for thetreatment of inflammation and gout. Compounds 2 and 5 were responsible for these biologicalactions by regulating pro-inflammatory enzymes, namely COXs, 5-LOX, and XO.

Abstract Enterobacter cloacae complex (Ecc) species are widely distributed opportunistic pathogens mainly associated with humans and plants. In this study, the genomes of clinical isolates including E. hormaechei, E. kobei, and E. ludwigii and non-clinical isolate including E. nimipressuralis were analysed in combination with the genome of E. asburiae by using the reference strain E. cloacae subsp. cloacae ATCC 13047; the Ecc strains were tested on artificial sputum media (ASM), which mimics the host, to evaluate T6SS genes as a case study. All five Ecc strains were sequenced in our lab. Comparative genome analysis of the Ecc strains revealed that genes associated with the survival of Ecc strains, including genes of metal-requiring proteins, defence-associated genes and genes associated with general physiology, were highly conserved in the genomes. However, the genes involved in virulence and drug resistance, specifically those involved in bacterial secretion, host determination and colonization of different strains, were present in different genomic regions. For example, T6SS accessory and core components, T4SS, and multidrug resistance genes/efflux system genes seemed vital for the survival of Ecc strains in various environmental niches, such as humans and plants. Moreover, the ASM host-mimicking growth medium revealed significantly high expression of T6SS genes, including PrpC, which is a regulatory gene of the T6SS, in all tested Ecc strains compared to the control medium. The variations in T6SS gene expression in ASM vs. control showed that the ASM system represents a simple, reproducible and economical alternative to animal models for studies such as those aimed at understanding the divergence of Ecc populations. In summary, genome sequencing of clinical and environmental Ecc genomes will assist in understanding the epidemiology of Ecc strains, including the isolation, virulence characteristics, prevention and treatment of infectious disease caused by these broad-host-range niche-associated species.

Color is the prime feature directly associated with the consumer’s attraction and choice of their food. The flavor, safety, and nutritional value of any food product are directly associated with the food color. Natural and synthetic colorants (dyes and pigments) have diversified applications in various sectors such as food, feed, pharmaceutical, textiles, cosmetics, and others. Concerning the food industry, different types of natural and synthetic colorants are available in the market. Synthetic food colorants have gained popularity as they are highly stable and cheaply available. Consumers worldwide prefer delightful foodstuffs but are more concerned about the safety of the food. After its disposal, the colloidal particles present in the synthetic colorants do not allow sunlight to penetrate aquatic bodies. This causes a foul smell and turbidity formation and gives a bad appearance. Furthermore, different studies carried out previously have presented the toxicological, carcinogenic effects, hypersensitivity reactions, and behavioral changes linked to the usage of synthetic colorants. Natural food colorings, however, have nutraceutical qualities that are valuable to human health such as curcumin extracted from turmeric and beta-carotene extracted from carrots. In addition, natural colorants have beneficial properties such as excellent antioxidant properties, antimutagenic, anti-inflammatory, antineoplastic, and antiarthritic effects. This review summarizes the sources of natural and synthetic colorants, their production rate, demand, extraction, and characterization of food colorants, their industrial applications, environmental impact, challenges in the sustainable utilization of natural colorants, and their prospects.

- Abstract
Background Patients with out‐of‐hospital cardiac arrest (OHCA) in rural areas experience longer emergency response times and have lower survival rates compared with patients in urban areas. Volunteer responders might improve care and outcomes for patients with OHCA specifically in rural areas. Therefore, we investigated volunteer responder interventions based on the degree of urbanization. Methods and Results We included 1310 OHCAs from 3 different regions in Denmark where volunteer responders had arrived at the OHCA location. The location was classified as urban, suburban, or rural according to the Eurostat Degree of Urbanization Tool. A logistic regression model was used to examine associations between the degree of urbanization and volunteer responder arrival before emergency medical services, cardiopulmonary resuscitation, or defibrillation. We found the odds for volunteer responder arrival before emergency medical services more than doubled in rural areas (odds ratio [OR], 2.60 [95% CI, 1.91–3.53]) and suburban areas (OR, 2.05 [95% CI, 1.56–2.69]) compared with urban areas. In OHCA cases where volunteer responders arrived first, odds for bystander cardiopulmonary resuscitation was tripled in rural areas (OR, 3.83 [95% CI, 1.64–8.93]) and doubled in suburban areas (OR, 2.27 [95% CI, 1.17–4.41]) compared with urban areas. Bystander defibrillation was more common in suburban areas (OR, 1.53 [95% CI, 1.02–2.31]), where almost 1 out of 4 patients received bystander defibrillation, compared with urban areas. Conclusions Volunteer responders are significantly more likely to arrive before emergency medical services in rural and suburban areas than in urban areas. Patients with OHCA received more cardiopulmonary resuscitation in rural and suburban areas and more defibrillation in suburban areas than in urban areas., BACKGROUND: Patients with out-of-hospital cardiac arrest (OHCA) in rural areas experience longer emergency response times and have lower survival rates compared with patients in urban areas. Condensed Matter - Materials Science - Abstract
We report real-time average stress measurements on composite silicon electrodes made with two different binders [Carboxymethyl cellulose (CMC), and polyvinylidene fluoride (PVDF)] during electrochemical lithiation and delithiation. During galvanostatic lithiation at very slow rates, the stress in a CMC-based electrode becomes compressive and increases to 70 MPa, where it reaches a plateau and increases slowly thereafter with capacity. The PVDF-based electrode exhibits similar behavior, although with lower peak compressive stress of about 12 MPa. These initial experiments indicate that the stress evolution in a Si composite electrode depends strongly on the mechanical properties of the binder. Stress data obtained from a series of lithiation/delithiation cycles suggests plasticity induced irreversible shape changes in contacting Si particles, and as a result, the stress response of the system during any given lithiation/delithiation cycle depends on the cycling history of the electrode. While these results constitute the first in-situ stress measurements on composite Si electrodes during electrochemical cycling, the diagnostic technique described herein can be used to assess the mechanical response of a composite electrode made with other active material/binder combinations., Comment: 22 pages, 8 figures

Deep learning-based classification and detection algorithms have emerged as a powerful tool for vehicle detection in intelligent transportation systems. The limitations of the number of high-quality labeled training samples makes the single vehicle detection methods incapable of accomplishing acceptable accuracy in road vehicle detection. This paper presents detection and classification of vehicles on publicly available datasets by utilizing the YOLO-v5 architecture. This paper’s findings utilize the concept of transfer learning through fine tuning the weights of the pre-trained YOLO-v5 architecture. To employ the concept of transfer learning, extensive data sets of images and videos of the congested traffic patterns were collected by the authors. These datasets were made more comprehensive by pointing various attributes, for instance high- and low-density traffic patterns, occlusions, and different weather circumstances. All of these gathered datasets were manually annotated. Ultimately, the improved YOLO-v5 structure becomes accustomed to any difficult traffic patterns. By fine-tuning the pre-trained network through our datasets, our proposed YOLO-v5 has exceeded several other traditional vehicle detection methods in terms of detection accuracy and execution time. Detailed simulations performed on the PKU, COCO, and DAWN datasets demonstrate the effectiveness of the proposed method in various challenging situations.
