154. Analysis of plasmid DNA from Pasteurella haemolytica and construction of cloning vectors

Abstract

Pasteurella haemolytica, a Gram-negative bacterium, is the principal causative agent of pneumonic pasteurellosis in cattle and sheep. The presence of plasmids in various isolates of this species and their relationship to antibiotic resistance and to the health status of the source animal were investigated. Plasmids conferring resistance to ampicillin were analyzed and the plasmid-encoded p-lactamase enzymes characterized. A suitable plasmid was identified for the construction of shuttle cloning vectors for P. haemolytica. Thirty five typable and untypable isolates of P. haemolytica from cattle or sheep were screened for the presence of plasmids and for resistance to a range of antibiotics. Eight strains (four of serotype Al, three of serotype A2 and one untypable) contained plasmid DNA. Isolates of the same serotype had similar plasmid profiles, which were different from those of the other serotypes. All but one of the plasmid-bearing strains were isolated from pneumonic, or from animals in contact with pneumonic, cattle or sheep. In A2 and untypable strains, there was no obvious correlation between antibiotic resistance and the presence of a specific plasmid. In contrast, all plasmid-bearing Al strains exhibited ampicillin resistance, which was shown by transfer studies (transformation and conjugation) to be plasmid-mediated. Plasmid DNA prepared from ampicillin-resistant E. coli transformants was not routinely detected on ethidium-bromide-stained agarose gels, but could be amplified to detectable levels by treatment of cultures with chloramphenicol or by modifying the growth conditions. Terrific broth or broth supplemented with 1% yeast nitrogen base was found to be the best growth medium for plasmid amplification. All Apft plasmids had a similar size (~4. 3 kb), and one of them (pPH843) was highly amplifiable and more stable in E. coll compared to the other plasmids (pPH2, pPH33 and pPH821). An improved method was developed for the large-scale purification of covalently closed circular plasmid DNA molecules over a wide size range. The protocol used an alkaline-lysis procedure followed by acid-phenol extraction and included several modifications to previously reported methods. Two principal modifications were the replacement of NaCl by MgCl in the extraction buffer and vortexing instead of shaking the crude DNA suspensions to more efficiently remove chromosomal and other non-CCC plasmid DNA and to improve yield of purified CCC DNA forms. The Apft plasmids from P, haemolytica were identical by restriction enzyme analysis. Restriction analysis and hybridization data indicated that these plasmids were closely related to the prototype ROB-1 p-lactamase-encoding plasmid, originally isolated from Haemophilus influenzae, but a part of the DNA sequence < ~0. 7 kb) from the Haemophilus plasmid was not present in the Pasteurella plasmid. This suggested that one plasmid had been derived from the other during the course of evolution. From substrate profiles and isoelectric focusing data, the p-lactamases encoded by the P. haemolytica plasmids were found to be indistinguishable from the ROB-1 0- lactamase. A restriction map of plasmid pPH843 was constructed and the putative Apft, oriV, and mob regions of the plasmid were located by deletion and fusion experiments. The plasmid was converted into a series of possible cloning vectors (pAKAlb, pAKA15-l, pAKA16, pAKA16-l and pAKA17) by insertion at different points in the plasmid of the lacZ alpha-peptide-coding gene and a multiple cloning site (for insertional inactivation of beta-galactosidase activity) from the E. coli vector pIC20H and an IncP mobilization function from plasmid RK2. Transconjugants obtained by transfer of the constructs from E, coli to P. haemolytlca were found to be unstable on subculture, although they were stable upon transfer to another E, coli strain. This indicated that P. haemolytlca might have a restriction/modification system which affected plasmid DNA previously propagated in E. coli.

Abstract

Alginate is a natural biopolymer widely studied for pharmaceutical applications due to its biocompatibility, low toxicity, and mild gelation abilities. This review summarizes recent advances in alginate-based encapsulation systems for targeted drug delivery. Alginate formulations like microparticles, nanoparticles, microgels, and composites fabricated by methods including ionic gelation, emulsification, spray drying, and freeze drying enable tailored drug loading, enhanced stability, and sustained release kinetics. Alginate microspheres prepared by spray drying or ionic gelation provide gastric protection and colon-targeted release of orally delivered drugs. Alginate nanoparticles exhibit enhanced cellular uptake and tumor-targeting capabilities through the enhanced permeation and retention effect. Crosslinked alginate microgels allow high drug loading and controlled release profiles. Composite alginate gels with cellulose, chitosan, or inorganic nanomaterials display improved mechanical properties, mucoadhesion, and tunable release kinetics. Alginate-based wound dressings containing antimicrobial nanoparticles promote healing of burns and chronic wounds through sustained topical delivery. Although alginate is well-established as a pharmaceutical excipient, more extensive in vivo testing is needed to assess clinical safety and efficacy of emerging formulations prior to human trials. Future opportunities include engineered systems combining stimuli-responsiveness, active targeting, and diagnostic capabilities. In summary, this review discusses recent advances in alginate encapsulation techniques for oral, transdermal, and intravenous delivery, with an emphasis on approaches enabling targeted and sustained drug release for enhanced therapeutic outcomes. Abstract

In diesel engines, emission formation inside the combustion chamber is a complex phenomenon. The combustion events inside the chamber occur in microseconds, affecting the overall engine performance and emissions characteristics. This study opted for using computational fluid dynamics (CFD) to investigate the combustion patterns and how these events affect nitrogen oxide (NOx) emissions. In this study, a diesel engine model with a flat combustion chamber (FCC) was developed for the simulation. The simulation result of the heat release rate (HRR) and cylinder pressure was validated with the experimental test data (the engine test was conducted at 1500 rpm at full load conditions). The validated model and its respective boundary conditions were used to investigate the effect of modified combustion chamber profiles on NOx emissions. Modified chambers, such as a bathtub combustion chamber (BTCC) and a shallow depth chamber (SCC), were developed, and their combustion events were analysed with respect to the FCC. This study revealed that combustion events such as fuel distribution, unburnt mass fractions, temperature and turbulent zones directly impact NOx emissions. The modified chambers controlled the spread of combustion and provided better fuel distribution, improving engine performance and combustion rates. The SCC (63.2 bar) showed peak pressure rates compared to the FCC (63.02 bar) and BTCC (62.72 bar). This study concluded that the SCC showed better results than other chambers. This study further recommends conducting lean fuel mixture combustion with chamber modifications and optimising fuel spray, such as by adjusting the fuel injection profile, spray angle and injection timing, which has a better tendency to create complete combustion. 170. Uncommon CD markers in acute myeloid leukemia

Abstract

This study was done to assess the unusual CD expression in 100 cases of acute myeloid leukemia from October 2016 to April 2018. The age limit was from 3 to 50 years. Four color flow cytometry was used to diagnose the fresh aspirated bone marrow or peripheral blood sample of acute leukemia. The unusual lymphoid CD expression on myeloblasts was analyzed. Among the cases, 44% were acute myeloid leukemia, 52% of patients were of acute lymphoblastic leukemia and mixed phenotype acute leukemia was 4%. Aberrant CD expression was observed in 58% acute myeloid leukemia patients. Both aberrant CD5 and CD7 lymphoid markers expressed in acute myeloid leukemia patients were 4.2%. Aberrant CD7, CD5, cCD79a and cCD3 were in 45.8%, 33.3%, 8.3%, 8.3% of acute myeloid leukemia patients respectively. In acute myeloid leukemia, the frequency of aberrant CD expression was compared with recent international data.
