Background: Cardiorespiratory fitness (CRF) supports cognition, though it is unclear what mechanisms underly this relationship. Insulin resistance adversely affects cognition but can be reduced with habitual exercise. Objective: We investigated whether insulin resistance statistically mediates the relationship between CRF and cognition. Methods: In our observational study, we included n = 1,131 cognitively unimpaired, nondiabetic older adults from a cohort characterized by elevated Alzheimer’s disease (AD) risk. We estimated CRF (eCRF) using a validated equation that takes age, sex, body mass index, resting heart rate, and habitual physical activity as inputs. The Homeostatic Model Assessment for Insulin Resistance (HOMA-IR) quantified insulin resistance. Standardized cognitive factor scores for cognitive speed/flexibility, working memory, verbal learning/memory, and immediate memory were calculated from a battery of neuropsychological tests. Linear regression models and bootstrapped estimates of indirect effects were used to determine whether HOMA-IR mediated significant relationships between eCRF and cognition. Results: eCRF was positively associated with cognitive speed/flexibility (p = 0.034). When controlling for HOMA-IR, eCRF was no longer associated with cognitive speed/flexibility (p = 0.383). HOMA-IR had a significant indirect effect on the eCRF-cognition relationship (B = 0.025, CI = [0.003,0.051]). eCRF was not associated with working memory (p = 0.236), immediate memory (p = 0.345), or verbal learning/memory (p = 0.650). Conclusion: Among older adults at risk for AD, peripheral insulin resistance mediates the relationship between CRF and cognitive speed.

Background: Prior research suggests a link between menopausal hormone therapy (MHT) use, memory function, and diabetes risk. The menopausal transition is a modifiable period to enhance long-term health and cognitive outcomes, although studies have been limited by short follow-up periods precluding a solid understanding of the lasting effects of MHT use on cognition. Objective: We examined the effects of midlife MHT use on subsequent diabetes incidence and late life memory performance in a large, same-aged, population-based cohort. We hypothesized that the beneficial effects of MHT use on late life cognition would be partially mediated by reduced diabetes risk. Methods: 1,792 women from the Wisconsin Longitudinal Study (WLS) were included in analysis. We employed hierarchical linear regression, Cox regression, and causal mediation models to test the associations between MHT history, diabetes incidence, and late life cognitive performance. Results: 1,088/1,792 women (60.7%) reported a history of midlife MHT use and 220/1,792 (12.3%) reported a history of diabetes. MHT use history was associated with better late life immediate recall (but not delayed recall), as well as a reduced risk of diabetes with protracted time to onset. Causal mediation models suggest that the beneficial effect of midlife MHT use on late life immediate recall were at least partially mediated by diabetes risk. Conclusion: Our data support a beneficial effect of MHT use on late life immediate recall (learning) that was partially mediated by protection against diabetes risk, supporting MHT use in midlife as protective against late life cognitive decline and adverse health outcomes.

While chemicals are vital to modern society through materials, agriculture, textiles, new technology, medicines, and consumer goods, their use is not without risks. Unfortunately, our resources seem inadequate to address the breadth of chemical challenges to the environment and human health. Therefore, it is important we use our intelligence and knowledge wisely to prepare for what lies ahead. The present study used a Delphi-style approach to horizon-scan future chemical threats that need to be considered in the setting of chemicals and environmental policy, which involved a multidisciplinary, multisectoral, and multinational panel of 25 scientists and practitioners (mainly from the United Kingdom, Europe, and other industrialized nations) in a three-stage process. Fifteen issues were shortlisted (from a nominated list of 48), considered by the panel to hold global relevance. The issues span from the need for new chemical manufacturing (including transitioning to non-fossil-fuel feedstocks); challenges from novel materials, food imports, landfills, and tire wear; and opportunities from artificial intelligence, greater data transparency, and the weight-of-evidence approach. The 15 issues can be divided into three classes: new perspectives on historic but insufficiently appreciated chemicals/issues, new or relatively new products and their associated industries, and thinking through approaches we can use to meet these challenges. Chemicals are one threat among many that influence the environment and human health, and interlinkages with wider issues such as climate change and how we mitigate these were clear in this exercise. The horizon scan highlights the value of thinking broadly and consulting widely, considering systems approaches to ensure that interventions appreciate synergies and avoid harmful trade-offs in other areas. We recommend further collaboration between researchers, industry, regulators, and policymakers to perform horizon scanning to inform policymaking, to develop our ability to meet these challenges, and especially to extend the approach to consider also concerns from countries with developing economies. The ocean, with its low albedo and vast thermal inertia, plays key roles in the climate system, including absorbing massive amounts of heat as atmospheric greenhouse gas concentrations rise. While the Argo array of profiling floats has vastly improved sampling of ocean temperature in the upper half of the global ocean volume since the mid-2000s, they are not sufficient in number to resolve eddy scales in the oceans. However, satellite sea surface temperature (SST) and sea surface height (SSH) measurements do resolve these scales. Here we use random forest regressions to map ocean heat content anomalies (OHCA) using in situ training data from Argo and other sources on a 7-day × 1/4° × 1/4° grid with latitude, longitude, time, SSH, and SST as predictors. The maps display substantial patterns on eddy scales, resolving variations of ocean currents and fronts. During the well-sampled Argo period, global integrals of these maps reduce noise relative to estimates based on objective mapping of in situ data alone by roughly a factor of 3 when compared to time series of CERES (satellite data) top-of-the-atmosphere energy flux measurements and improve correlations of anomalies with CERES on annual time scales. Prior to and early on in the Argo period, when in situ data were sparser, global integrals of these maps retain low variance, and do not relax back to a climatological mean, avoiding potential deficiencies of various methods for infilling data-sparse regions with objective maps by exploiting temporal and spatial patterns of OHCA and its correlations with SST and SSH. Significance Statement We use a simple machine learning technique to improve maps of subsurface ocean warming by exploiting the relationships between subsurface ocean temperature both sea surface temperature and sea level. Mapping ocean warming is important because it contributes to sea level rise through thermal expansion; impacts marine life through marine heatwaves and changes in mixing, oxygen, and carbon dioxide levels; increases energy available to tropical cyclones; and stores most of the energy building up in Earth’s climate system owing to the accumulation of anthropogenic greenhouse gases in the atmosphere. Our new estimates generally have lower noise energy and higher correlations than other products when compared with global energy fluxes at the top of the atmosphere measured by satellite.

Objective: The preeminent in-vivo cerebrospinal fluid (CSF) biomarkers of Alzheimer’s disease (AD) are amyloid β 1-42 (Aβ42), p-tau and t-tau. The goal of this study was to examine how well traditional (total and delayed recall) and process-based (recency ratio; Rr) measures derived from Rey’s AVLT were associated with these biomarkers.\ud Method: Data from 235 participants (mean age = 65.5, SD = 6.9), who ranged from cognitively unimpaired to mild cognitive impairment, and for whom CSF values were available, were extracted from the Wisconsin Registry for Alzheimer’s Prevention. Bayesian regression analyses were carried out using CSF scores as outcomes, AVLT scores as predictors, and controlling for demographic data and diagnosis.\ud Results: We found moderate evidence that Rr was associated with both CSF p-tau (BFm = 5.55;) and t-tau (BFm = 7.28), above and beyond the control variables, while it did not correlate with CSF Aβ42 levels. In contrast, total and delayed recall scores were not linked with any of the AD biomarkers, in separate analyses. When comparing all memory predictors in a single regression, Rr remained the strongest predictor of CSF t-tau levels (BFm = 3.57).\ud Conclusions: Our findings suggest that Rr may be a better cognitive measure than commonly used AVLT scores to assess CSF levels of p-tau and t-tau in non-demented individuals.

Rock damage from underground nuclear explosions (UNEs) has a strong influence on sub-surface gas movement and on seismic waveform characteristics, both of which are used to detect UNEs. Although advanced numerical simulation capabilities exist to predict rock damage patterns and corresponding detection signals, those predictions are dependent on (generally) unknown properties of the host rock. For example, the effects of in-situ mechanical heterogeneities on the explosively generated damage/fractures that provide gas flow pathways to the surface are not well understood, due largely to the difficulty in accessing and characterizing the near-source region. In this paper we demonstrate the emerging use of electrical resistivity tomography (ERT) for imaging rock damage and gas flow patterns resulting from two relatively small-scale underground chemical explosions. Pre-explosion ERT and crosshole seismic imaging revealed a natural fracture zone within the test bed. Post-explosion imaging revealed that the damage zone was non-symmetric and was focused primarily within the pre-existing fracture zone, located 10 m above the first explosion and 5 m above the second explosion. Time-lapse ERT imaging of heated air injected into the detonation borehole revealed the primary gas flow paths to be within the upper margin of the same primary damage zone. These results point to the utility of ERT imaging for understanding rock damage and gas flow patterns under experimental conditions, and to the importance of understanding the effects of geologic heterogeneity on UNE detection signals, particularly gas surface breakthrough times.

Color centers in diamond are promising platforms for quantum technologies. Most color centers in diamond discovered thus far emit in the visible or near-infrared wavelength range, which are incompatible with long-distance fiber communication and unfavorable for imaging in biological tissues. Here, we report the experimental observation of a new color center that emits in the telecom O-band, which we observe in silicon-doped bulk single crystal diamonds and microdiamonds. Combining absorption and photoluminescence measurements, we identify a zero-phonon line at 1221 nm and phonon replicas separated by 42 meV. Using transient absorption spectroscopy, we measure an excited state lifetime of around 270 ps and observe a long-lived baseline that may arise from intersystem crossing to another spin manifold.

Tiger Grouper (Mycteroperca tigris) form fish spawning aggregations (FSAs) around the winter full moons (typically January through April) in the Caribbean. Males defend territories to attract mates in a lek-like reproductive strategy. Prior studies have documented rapid declines in populations with FSA-associated fisheries. This study examines the migratory behavior of adult male Tiger Grouper in Little Cayman, Cayman Islands, to better understand the impacts of aggregation fishing. As part of the Grouper Moon Project, we acoustically tagged ten spawning male Tiger Grouper at the western end of Little Cayman in February 2015. Using a hydrophone array surrounding the island, we tracked the movements of the tagged fish for 13 months. We observed 3 migratory strategies: resident fish (n = 2) that live at the FSA site, neighboring fish (n = 5) that live within 4 km of the site, and commuter fish (n = 3) that travel over 4 km for spawning. Fish began aggregating 2 days before the full moon and left 10–12 days after the full moon, from January to May. Regardless of migratory strategy, all tagged fish that aggregated after February 2015 returned to the west end FSA. However, in January 2016, one fish appeared to attend a different FSA closer to its presumed home territory. Tiger Grouper may establish multiple FSAs around Little Cayman, and males appear to attend FSAs near their home territories. Protracted spawning seasons, FSA site infidelity, and putative FSA catchments should all be considered to ensure sustainable fisheries management for this important species.

Specific ion effects govern myriad biological phenomena, including protein-ligand interactions and enzyme activity. Despite recent advances, detailed understanding of the role of ion hydrophobicity in specific ion effects, and the intersection with hydrotropic effects, remains elusive. Short chain fatty acid sodium salts are simple amphiphiles which play an integral role in our gastrointestinal health. We hypothesise that increasing a fatty acid's hydrophobicity will manifest stronger salting-out behaviour.Here we study the effect of these amphiphiles on an exemplar thermoresponsive polymer brush system, conserving the carboxylate anion identity while varying anion hydrophobicity via the carbon chain length. Ellipsometry and quartz crystal microbalance with dissipation monitoring were used to characterise the thermoresponse and viscoelasticity of the brush, respectively, whilst neutron reflectometry was used to reveal the internal structure of the brush. Diffusion-ordered nuclear magnetic resonance spectroscopy and computational investigations provide insight into polymer-ion interactions.Surface sensitive techniques unveiled a non-monotonic trend in salting-out ability with increasing anion hydrophobicity, revealing the bundle-like morphology of the ion-collapsed system. An intersection between ion-specific and hydrotropic effects was observed both experimentally and computationally; trending from good anti-hydrotrope towards hydrotropic behaviour with increasing anion hydrophobicity, accompanying a change in hydrophobic hydration.

Deep infection is a debilitating complication after shoulder arthroplasty. The authors hypothesized that an intra-articular, intraoperative injection of antibiotics would result in a lower infection rate compared with intravenous antibiotics alone. Before 2007, 164 patients (group A) did not receive intra-articular antibiotics. From 2007 to 2018, 1324 patients (group B) received intra-articular antibiotics. Patients received intra-articular gentamicin at the end of surgery with the addition of 1 g of cefazolin in January 2014. Records were retrospectively reviewed for comorbidities, type of surgery, and infection. The cohort that received intra-articular antibiotics was compared with the cohort that did not to determine the effect of prophylactic intra-articular antibiotic administration in preventing infection. There was 1 deep infection in the antibiotic group compared with 5 in the non-antibiotic group ( P P =.62). One previous study evaluated intra-articular injection of antibiotics for shoulder arthroplasty and found significantly lower rates of infection with the injection of intra-articular gentamicin. In this retrospective follow-up study, the injection of intra-articular gentamicin or gentamicin and cefazolin effectively decreased rates of postoperative infection. At mean follow-up of 399 days, intra-articular antibiotics at the time of surgery resulted in significantly fewer deep infections. Given the minimal risk of adverse events and minimal cost, this is a valid method of reducing infections in total shoulder arthroplasty. [ Orthopedics . 202x;4x(x):xx–xx.]

Studying coupling between different galactic components is a challenging problem in galactic dynamics. Using basis function expansions (BFEs) and multichannel singular spectrum analysis (mSSA) as a means of dynamical data mining, we discover evidence for two multi-component disc-halo dipole modes in a Milky-Way-like simulated galaxy. One of the modes grows throughout the simulation, while the other decays throughout the simulation. The multi-component disc-halo modes are driven primarily by the halo, and have implications for the structural evolution of galaxies, including observations of lopsidedness and other non-axisymmetric structure. In our simulation, the modes create surface density features up to 10 per cent relative to the equilibrium model stellar disc. While the simulated galaxy was constructed to be in equilibrium, BFE+mSSA also uncovered evidence of persistent periodic signals incited by aphysical initial conditions disequilibrium, including rings and weak two-armed spirals, both at the 1 per cent level. The method is sensitive to distinct evolutionary features at and even below the 1 per cent level of surface density variation. The use of mSSA produced clean signals for both modes and disequilibrium, efficiently removing variance owing to estimator noise from the input BFE time series. The Indian Ocean Sanctuary was established in 1979 in an effort to allow exploited stocks of whales to recover from whaling and to facilitate benign research. Further information is required to establish comprehensive management and conservation measures for species within these waters. The current study took place in the offshore waters of Sri Lanka in early 2003. During three research cruises conducted between 29 March and 17 June 2003 the R/V Odyssey covered a total track line of 4,480km around the island resulting in 52 confirmed group sightings of 11 species from three cetacean families. As the tracklines were designed to locate sperm whales (Physeter macrocephalus) for tissue sampling, they accounted for the greatest number of sightings. Only two species of balaenopterids, the blue whale (Balaenoptera musculus) and the Bryde’s whale (Balaenoptera edeni), were recorded with the blue whale being the most frequently sighted species. Spinner dolphins (Stenella longirostris) were the most dominant species in terms of numbers. Some small odonotocetes such as the common bottlenose dolphin (Tursiops truncatus), striped dolphin (Stenella coeruleoalba) and Fraser’s dolphin (Lagenodelphis hosei) were observed in mixed-species groups, while one group of melon-headed whales (Peponocephala electra) was seen associating with a group of sperm whales. Risso’s dolphins (Grampus griseus) were frequently sighted throughout the research cruise, with one unusual record of a large mating group. Many sightings were made in the vicinity of the numerous submarine canyons around Sri Lanka’s coastline highlighting their potential role in enhancing productivity in the offshore waters. It is concluded that Sri Lankan offshore waters hold a rich, but little surveyed cetacean fauna that warrants further studies and implementation of conservation measures to protect these populations.

Menopause is associated with adverse changes in vascular health coinciding with an increased risk of stroke and vascular cognitive impairment. However, there is significant variation in the age at menopause. The present study examined how the age at natural menopause impacts cerebrovascular reactivity and structural biomarkers of brain aging. Thirty-five healthy postmenopausal women were classified as early-onset menopause (Early; n = 19, age at menopause: 47 ± 2 yr) or later-onset menopause (Late; n = 16, age at menopause: 55 ± 2 yr). Middle cerebral artery blood velocity (MCAv), mean arterial blood pressure (MAP), and end-tidal carbon dioxide (ETCO2) were recorded during a stepped hypercapnia protocol. Reactivity was calculated as the slope of the relationship between ETCO2 and each variable of interest. Brain volumes and white matter hyperintensities (WMHs) were obtained with 3T MRI. Resting MAP was greater in the Early group (99 ± 9 mmHg) compared with the Late group (90 ± 12 mmHg; P = 0.02). Cerebrovascular reactivity, assessed using MCAv, was blunted in the Early group (1.87 ± 0.92 cm/s/mmHg) compared with the Late group (2.37 ± 0.75 cm/s/mmHg; P = 0.02). Total brain volume did not differ between groups (Early: 1.08 ± 0.07 L vs. Late: 1.07 ± 0.06 L; P = 0.66), but the Early group demonstrated greater WMH fraction compared with the Late group (Early: 0.36 ± 0.14% vs. Late: 0.25 ± 0.14%; P = 0.02). These results suggest that age at natural menopause impacts cerebrovascular function and WMH burden in healthy postmenopausal women.

Salvage logging is a controversial tool for post-wildfire management that removes fire-killed trees. We use a generalized randomized experimental design to fulfill two main objectives: (1) quantify the immediate (1-year post-harvest) effects of salvage logging on stand structure, fine and coarse woody fuel loadings; and (2) use pre- and post-empirical field data and the Fire and Fuels Extension to the Forest Vegetation Simulator (Reinhardt and Crookston 2003) to simulate post-wildfire dead woody fuel succession and snag dynamics. We compared the effects on woody fuel loadings of two salvage logging prescriptions: (1) seed tree harvest, thin to 3.4 m2·ha−1; and (2) full salvage of all merchantable timber, relative to unlogged controls. There was substantial block-level variability in the implementation of the treatments and in their immediate effects on fine fuel loading, complicating comparison of the two prescriptions. Overall, salvage logging did reduce snag basal area and, relative to unlogged controls, significantly increased measured fine woody fuel loading (10 and 100 h). Simulated snag fall was rapid, with a mean predicted snag basal area loss of 61% within 10 years. Future long-term monitoring of permanent field plots will supplement model predictions and provide valuable data to inform post-wildfire management decisions.

Background: Preeclampsia increases the incidence of maternal stroke, a devastating condition that is on the rise. We investigated stroke outcome in a model of experimental preeclampsia with and without treatment with clinically relevant doses of magnesium sulfate (experimental preeclampsia+MgSO 4 ) compared to normal late-pregnant and nonpregnant rats. Methods: Transient middle cerebral artery occlusion was used to induce focal stroke for either 1.5 or 3 hours. Infarct volume and hemorrhagic transformation were determined as measures of stroke outcome. Changes in core middle cerebral artery and collateral flow were measured by dual laser Doppler. The relationship between middle cerebral artery perfusion deficit and infarction was used as a measure of ischemic tolerance. Oxidative stress and endothelial dysfunction were measured by 3-nitrotyrosine and 8-isoprostane, in brain and serum, respectively. Results: Late-pregnant animals had robust collateral flow and greater ischemic tolerance of brain tissue, whereas experimental preeclampsia had greater infarction that was related to poor collateral flow, endothelial dysfunction, and oxidative stress. Importantly, pregnancy appeared preventative of hemorrhagic transformation as it occurred only in nonpregnant animals. MgSO 4 did not provide benefit to experimental preeclampsia animals for infarction. Conclusions: Stroke outcome was worse in a model of preeclampsia. As preeclampsia increases the risk of future stroke and cardiovascular disease, it is worth understanding the influence of preeclampsia on the material brain and factors that might potentiate injury both during the index pregnancy and years postpartum.
