152. Nuclear translocation of PKM2 mediates keratinocyte metabolic reprogramming in psoriasis.

Abstract

PKM2 mediates the Warburg effects and is crucial for tumorigenesis, but its role in hyperplastic skin disorders remains elusive. In this study, we investigated the function of PKM2 in psoriatic keratinocytes. We found that PKM2 expression and its nuclear translocation were induced in the epidermis of psoriasis patients, contributing to aerobic glycolysis and cell growth. Moreover, mass spectrometry combined with immunoprecipitation analysis revealed that PKM2 could interact with TRIM33, an E3 ubiquitin ligase in the nucleus, and this interaction is critical for the nuclear retention of PKM2. As a result of TRIM33‐mediated ubiquitination, PKM2 nuclear protein kinase function is promoted, thus leading to the phosphorylation of STAT3. In addition, blocking PKM2 nuclear translocation abrogated TRIM33‐triggered glycolysis and cell proliferation in keratinocytes. Taken together, our experiments demonstrate that ubiquitination regulates the nuclear retention of PKM2 in keratinocytes. Abstract

BackgroundThere is growing interest in the joint effects of hazardous trace elements (HTEs) on lung function deficits, but the data are limited. This is a critical research gap given increased global industrialisation.MethodsA national cross-sectional study including spirometry was performed among 2112 adults across 11 provinces in China between 2020 and 2021. A total of 27 HTEs were quantified from urine samples. Generalised linear models and quantile-based g-computation were used to explore the individual and joint effects of urinary HTEs on lung function, respectively.ResultsOverall, there were negative associations between forced expiratory volume in 1 s (FEV1) and urinary arsenic (As) (z-score coefficient, −0.150; 95% CI, −0.262 to –0.038 per 1 ln-unit increase), barium (Ba) (−0.148, 95% CI: −0.258 to –0.039), cadmium (Cd) (−0.132, 95% CI: −0.236 to –0.028), thallium (Tl) (−0.137, 95% CI: −0.257 to –0.018), strontium (Sr) (−0.147, 95% CI: −0.273 to –0.022) and lead (Pb) (−0.121, 95% CI: −0.219 to –0.023). Similar results were observed for forced vital capacity (FVC) with urinary As, Ba and Pb and FEV1/FVC with titanium (Ti), As, Sr, Cd, Tl and Pb. We found borderline associations between the ln-quartile of joint HTEs and decreased FEV1(−20 mL, 95% CI: −48 to +8) and FVC (−14 mL, 95% CI: −49 to+2). Ba and Ti were assigned the largest negative weights for FEV1and FVC within the model, respectively.ConclusionOur study investigating a wide range of HTEs in a highly polluted setting suggests that higher urinary HTE concentrations are associated with lower lung function, especially for emerging Ti and Ba, which need to be monitored or regulated to improve lung health.

192. Effect of Tungsten Doping on Performance of MAO Coatings on Ti6Al4V Drill Pipe

Abstract

A tungsten doped micro-arc oxidation (MAO) coating on TC4 Titanium Alloy Drill Pipe was prepared by adding sodium tungstate in electrolytes. The effects of different adding concentrations on the electrical conductivity of the electrolyte, performance and structure of MAO coatings on titanium alloy drill pipe were studied. The electrical conductivity of the solution was tested using Ohm's law. The surface morphologies of the coatings were observed by SEM. The element contents of the MAO coatings were analyzed by EDS. The hardness of the coatings was measured by microhardness tester. The phase composition was analyzed by XRD and the corrosion resistance was evaluated by polarization curves. The results show that WO42−begins to form WO3when the temperature of micro-arc discharge area is above 1314 K. With increasing sodium tungstate doping concentration, the conductivity of the solution increases first and then decreases, and the hardness and corrosion resistance of the coatings also show the identical trend. Tungsten doping slightly affects the phase composition of the MAO coating and it is helpful to improve the hardness and corrosion resistance of the coatings. The optimum doping concentration is 3 g L−1in this work.

Abstract

Pore structures are critical for most properties of cementitious materials. To describe the initial pore structure of fresh cement paste, which is the foundation of the evolution of a pore network induced by hydration of cement particles in hardened concrete, statistical modelling of pore structure is carried out based on a concentric-shell sphere model, by taking cement paste as a random two-phase media. Calculations by the dispersion models agree well with the pore size distributions tested by the low-field H1 nuclear magnetic resonance (NMR) technique. For cement paste that disperses completely with the help of a dispersant, a hard sphere model is practical. However, in most real conditions, an agglomeration of cement particles will occur both before and after mixing with water, which can be characterized by the redistribution of pore size determined by the critical grain size and the hardness parameter in the concentric-shell model, respectively. Calculation results suggest that agglomeration of particles leads to coarser pores. This work will promote the understanding of the formation mechanisms of pore networks and further offers a foundation for modelling the evolution of pore structures in cementitious materials. Abstract

Although TiAl3 is widely used in various high-temperature fields, the adjustment of the balance between the strength and ductility of the tetragonal TiAl3 remains a challenge. To solve the key problem, here, we apply the first-principles method to study the influence of four transition metals (TM = V, Cr and Mo) on the mechanical properties of the tetragonal TiAl3. The structural stability of the TM-alloyed TiAl3 is examined by the impurity formation energy and phonon dispersion. The calculated results show that the TM-doped TiAl3 is stable at the ground state. In particular, the Mo-alloyed TiAl3 has better thermodynamic stability than the other TM-alloyed TiAl3. Importantly, the alloyed elements enhance the bulk modulus of TiAl3 because the alloyed elements improve the electronic hybridisation between the Ti atom and the Al atom. Here, the Mo-alloyed TiAl3 has the highest bulk modulus among the four TM-alloyed TiAl3. Furthermore, it is found that the alloyed elements improve the ductility of TiAl3. [ABSTRACT FROM AUTHOR]
