Anti-tubulin antibody indirect immunofluorescence microscopy is used in this report. Difference of intracellular microtubule fluorescent staining patterns between normal and malignant human esophageal cells has been observed. Primarily cultured human esophageal epithelial cells display an elaborate array of fluorescent cytoplasmic microtubule filaments, the cytoplasmic microtubule complex (CMTC), in interphase cells and mitotic spindle microtubules in mitotic cells. Malignant esophageal cells from human esophageal carcinoma, ECa109 cells, contain very few cytoplasmic microtubules in interphase cells, but there is no difference of mitotic spindle microtubule fluorescence between normal and malignant cells. The results presented in this report suggest that the diminution of interphase cytoplasmic microtubules in tumor cells is probably due to the deficiency of microtubule organizing mechanism in interphase tumor cells.

The distribution and lateral mobility of Con A receptors in the course of the cell cycle has been studied on human gastric low-differentiated mucous adenocarcinoma cells (MGC 80-3). Synchronized MGC 80-3 cells in monolayer were labeled with F-ConA in the different phases of cell cycle. The discontinuous pattern of surface labelling was present during G1, S and G2, while there were marked differences among them. The cells in mitosis displayed uniform distribution of Con A receptors complex with strong fluorescence. The fluorescence photobleaching recovery method has been used to determine the lateral mobilities of Con A receptors complex. There was marked difference among various phases for mobility pattern and diffusion coefficient. The pattern of mobility of Con A receptor complex was appeared diffusion mobility in M and G1 phases, while flow pattern occurred in S and G2 phases. The diffusion coefficient (D) G1 greater than M, the rate of flow S greater than G2, but mobile fraction in G2 phase is higher than other phases. These results showed that dynamic properties of Con A receptor are apparently different which were regulated by the cell cycle.

This article presents a pedigree of 80 members of which there are 13 patients (13 males) with peroneal muscular atrophy. The analysis of this pedigree shows that an affected male married to a normal female will have normal son(s) and carrier-daughter(s). The normal son(s) will have normal offspring, but half the sons of the carrier-daughter(s) will be affected. The pathogenic genes of affected males are transmitted to their daughters, but not to their sons. So this inheritance is of X-linked recessive type. This is the first report of X-linked recessively inherited peroneal muscular atrophy in china.

Essential and non-essential metal ions were compared on the basis of their growth-inhibitory potency and their mediation of metallothionein induction in a Chinese hamster ovary cell line resistant to cadmium. Cadmium-resistant cells were found to be 20-fold and 6-fold more resistant than wild-type Chinese hamster ovary cells to the non-essential metals CdCl2 and HgCl2, respectively. In contrast, cadmium-resistant cells showed 2-fold or less resistance to growth inhibition due to the metals with known or possible biological essentiality, ZnCl2, CuSO4, CoCl2, and NiCl2. Resistance to either cadmium or mercury was not due to decreased uptake as measured isotopically or by X-ray fluorescence. At concentrations near the threshold of growth inhibition, CdCl2 and ZnCl2 induced metallothionein 8- to 10-fold above background levels in cadmium-resistant cells within 8-10 hr. A 2- to 3-fold induction of this protein was produced in resistant cells by levels of HgCl2, CuSO4, and CoCl2 near the threshold of growth inhibition whereas NiCl2 produced no measurable elevations of metallothionein at concentrations below, near, and above those that inhibit cell growth. Induction of metallothionein was measured by a modified 203Hg binding assay and by [35S]cysteine incorporation. No measurable induction of metallothionein was evident in wild-type cells with any metal treatment using a reasonable quantity of cells consistent with our assay. These results in cadmium-resistant cells demonstrate selective induction of metallothionein by various metals and suggest that induction of this protein alone is not solely responsible for differences in the growth-inhibitory potential of these elements.

A strain of Gram negative bacteria was isolated from the surface soil of Wuying Hill at Jinan, Shandong province with Gause's medium in 1973. It is a strain of antagonistic bacteria for hysterocervicoma, hepatoma and melanoma of mice screened from 2100 strains of bacteria. It is also antagonistic to Staphylococcus aureus, Bacillus subtilis and Micrococcus. It is a Gram negative bacterium with lophotrichous polar flagella. Straight rods in shape or with a little slightly curved rods, 0.5-0.6 X 1-2 microns, randomly arranged, poly-beta-hydroxybutyrate granules are accumulated in cells after 2-5 days cultivation. Water green soluble pigment and green fluorescent pigment are produced. Respiratory metabolism, chemoorganotroph, many carbon-containing organic compounds can be used as carbon sources, such as glucose, trehalose, ethanol, cellulobiose, fucose, arginine and betaine, but propionic acid or tartaric acid is not utilized. Inorganic nitrogen containing compounds can be used ae the sole source of nitrogen. No growth factor is necessary for growth. Gelatin is hydrolyzed. Starch and cellulose are not hydrolyzed. Nitrate is not reduced. Arginine dihydrolase is produced. Levan is produced from sucrose. Growth occurs from 7 degrees C to 37 degrees C and from pH 5.65-8.40. No growth occurs at 40 degrees C and at pH value below 4.86. It can not grow autotrophically with hydrogen. Its G + C contents in DNA is 58.1 mol%. DNA-DNA hybridization experiments reveals a relatedness value of 58.6% between this strain and Ps. fluorescens. The above evidence shows that this strain differs from all species known in Pseudomonas, such as Pseudomonas fluorescens group. Pseudomonas caryophylli, Pseudomonas cepacia, Pseudomonas marginata, Pseudomonas acidovorans, Pseudomonas testosteroni and Pseudomonas delafieldii.(ABSTRACT TRUNCATED AT 250 WORDS)

A model for Raman scattering by a molecule adsorbed at the surface of a spherical particle is articulated by treating the molecule as a classical electric dipole. This follows Moskovits's suggestion [J. Chem. Phys. 69, 4159 (1978)] and the experiments by Creighton et al. [J. Chem. Soc. Faraday Trans. II, 75, 790 (1979)] that such a system may exhibit SERS similar to that at roughened electrode surfaces. The molecule is stimulated by a primary field comprised of the incident and near-scattered fields. Emission consists of the dipole field plus a scattered field, each at the shifted frequency. Addition of feedback terms between the dipole and the particle makes only a negligible contribution to the fields. For pyridine adsorbed at the surface of a silver sphere, the 1010-cm(-1) band is enhanced by ~10(6) if the radius is much less than the wavelengths and the excitation wavelength is ~382 nm, a wavelength for which the relative refractive index of silver is close to m = radical2i. Detailed results are given for the effect upon the angular distribution and the polarization of the Raman emission of particle size, distance from the surface, excitation wavelength, and location of the molecule upon the surface. These results simulate those observed at roughened silver electrodes and suggest that the mechanism of SERS at those electrodes may resemble the electromagnetic mechanism elucidated here. We predict that comparable effects should be observed for fluorescent scattering.
