Healing of large calvarial bone defects remains challenge but may be improved by stimulating bone regeneration of implanted cells. The aim of this study is to specially co-activate transforming growth factor β1 (TGF-β1) and vascular endothelial growth factor (VEGF-A) genes expressions in pre-osteoblast MC3T3-E1 cells through the non-viral CRISPR activation (CRISPRa) system to promote osteogenesis. A cationic copolymer carrying nucleus localizing peptides and proton sponge groups dimethyl-histidine was synthesized to deliver CRISPRa system into MC3T3-E1 cells with high cellular uptake, lysosomal escape, and nuclear translocation, which activated VEGF-A and TGF-β1 genes expressions and thereby additively or synergistically induced several osteogenic genes expressions. A tunable dual-crosslinked hydrogel was developed to implant the above engineered cells into mice calvaria bone defect site to promote bone healing in vivo. The combination of multi-genes activation through non-viral CRISPRa system and tunable dual-crosslinked hydrogel provides a versatile strategy for promoting bone healing with synergistic effect.

A typical polyanionic based material Na3V2(PO4)2O2F (Na3VPO2F) attracts much interest as a cathode for large‐scale sodium‐ion batteries in consideration of its stable structure and remarkable energy density. Nevertheless, the large coulombic attraction and repulsion suffered by the mobile Na+ from structural anions and surrounding Na+, respectively, result in a torpid reaction kinetics and inferior rate capability. Herein, Br−‐doped and Na+ vacancy preinstalled Na3−yVPO2−xBrxF is prepared to dilute the charges on and inside the Na+ transportation tunnel. In virtue of density functional theory analysis, Na3−yVPO2−xBrxF reveals a reduction in the bandgap and an increase in electronic conductivity. Meanwhile, the almost electrostatically shielded tunnel in Na3−yVPO2−xBrxF alleviates the coulombic hindrance imposed on Na+ during its (de)intercalation, which demonstrates a Na+ diffusivity about five times higher than that of Na3VPO2F. Consequently, the Na3−yVPO2−xBrxF cathode shows a superior rate capacity of 77.7 mAh g−1 under 50 C and great cycling property corresponding to a high capacity retention of 94.4% over 800 cycles at 10 C. The assembled Na3−yVPO2−xBrxF//hard‐carbon sodium‐ion full‐cell presents excellent specific energy/power (226 Wh kg−1@15424.2 W kg−1) as well as outstanding long‐term cyclic stability over 1000 cycles at 5 C. In this article, we perform a numerical investigation on the polarization of a terahertz wave transmitting through a dust acoustic wave. Assuming a triangular waveform of the particle number density in the dust acoustic wave, the transmission characteristics are calculated using the finite-difference time-domain method. It is shown that dust acoustic waves can function as a polarizer to control the polarization states of the terahertz wave, and the conversion efficiency depends on the parameters of the dust acoustic wave, such as the particle number density and the thickness of particle cloud. Abstract In this study, we use nitrogen-doped to improving the gas-sensing properties of reduced graphene oxide. Graphene oxide was prepared according to a modified Hummers’ method and then nitrogen-doped reduced graphene oxide (N-rGO) was synthesized by a hydrothermal method using graphene oxide and NH4OH as precursors. The rGO is flat and smooth with a sheet-like morphology while the N-rGO exhibits folded morphology. This type of folding of the surface morphology can increase the gas sensitivity. The N-rGO and the rGO sensors showed n-type and p-type semiconducting behaviors in ambient conditions, respectively, and were responsive to low concentrations of NO gases (

Gastric cancer (GC) is one of the leading causes of cancer-related death worldwide. MicroRNAs (miRNAs) are known to be important regulators of GC. This study aims to investigate the role of miRNA (miR)-497 in GC. We demonstrated that the expression of miR-497 was downregulated in human GC tissues. After N-methyl-N-nitrosourea treatment, the incidence of GC in miR-497 knockout mice was significantly higher than that in wild-type mice. miR-497 overexpression suppressed GC cell proliferation, cell-cycle progression, colony formation, anti-apoptosis ability, and cell migration and invasion capacity. Additionally, miR-497 overexpression decreased the expression levels of cell division cycle 42 (CDC42) and integrin β1 (ITGB1) and inhibited the phosphorylation of focal adhesion kinase (FAK), paxillin (PXN), and serine-threonine protein kinase (AKT). Furthermore, overexpression of miR-497 inhibited the metastasis of GC cells in vivo, which could be counteracted by CDC42 restoration. Furthermore, the focal adhesion of GC cells was found to be regulated by miR-497/CDC42 axis via ITGB1/FAK/PXN/AKT signaling. Collectively, it is concluded that miR-497 plays an important role in the repression of GC tumorigenesis and progression, partly via the CDC42/ITGB1/FAK/PXN/AKT pathway.

The fourth generation of the Sloan Digital Sky Survey (SDSS-IV) began observations in 2014 July. It pursues three core programs: the Apache Point Observatory Galactic Evolution Experiment 2 (APOGEE-2), Mapping Nearby Galaxies at APO (MaNGA), and the Extended Baryon Oscillation Spectroscopic Survey (eBOSS). As well as its core program, eBOSS contains two major subprograms: the Time Domain Spectroscopic Survey (TDSS) and the SPectroscopic IDentification of ERosita Sources (SPIDERS). This paper describes the first data release from SDSS-IV, Data Release 13 (DR13). DR13 makes publicly available the first 1390 spatially resolved integral field unit observations of nearby galaxies from MaNGA. It includes new observations from eBOSS, completing the Sloan Extended QUasar, Emission-line galaxy, Luminous red galaxy Survey (SEQUELS), which also targeted variability-selected objects and X-ray-selected objects. DR13 includes new reductions of the SDSS-III BOSS data, improving the spectrophotometric calibration and redshift classification, and new reductions of the SDSS-III APOGEE-1 data, improving stellar parameters for dwarf stars and cooler stars. DR13 provides more robust and precise photometric calibrations. Value-added target catalogs relevant for eBOSS, TDSS, and SPIDERS and an updated red-clump catalog for APOGEE are also available. This paper describes the location and format of the data and provides references to important technical papers. The SDSS web site, http://www.sdss.org, provides links to the data, tutorials, examples of data access, and extensive documentation of the reduction and analysis procedures. DR13 is the first of a scheduled set that will contain new data and analyses from the planned ∼6 yr operations of SDSS-IV.

Abstract The clustered regularly interspaced short palindromic repeats (CRISPR)/associated protein 9 (CRISPR/Cas9) gene editing technology, as a revolutionary breakthrough in genetic engineering, offers a promising platform to improve the treatment of various genetic and infectious diseases because of its simple design and powerful ability to edit different loci simultaneously. However, failure to conduct precise gene editing in specific tissues or cells within a certain time may result in undesirable consequences, such as serious off-target effects, representing a critical challenge for the clinical translation of the technology. Recently, some emerging strategies using genetic regulation, chemical and physical strategies to regulate the activity of CRISPR/Cas9 have shown promising results in the improvement of spatiotemporal controllability. Herein, in this review, we first summarize the latest progress of these advanced strategies involving cell-specific promoters, small-molecule activation and inhibition, bioresponsive delivery carriers, and optical/thermal/ultrasonic/magnetic activation. Next, we highlight the advantages and disadvantages of various strategies and discuss their obstacles and limitations in clinical translation. Finally, we propose viewpoints on directions that can be explored to further improve the spatiotemporal operability of CRISPR/Cas9.

The shear stress relaxation of the anchor-soil interface is the key factor causing the prestress loss of anchor rod (cable). Firstly, a device for testing the shear stress relaxation of the anchor-soil interface was developed. Secondly, a constant interface shear displacement was applied in stages to the red clay anchored element sample, and the whole process of shear stress relaxation curve of anchor-soil interface was obtained, which can be transformed to the relaxation curve at each specific loading level by using coordinate translation method. Then, the theory of fractional calculus was introduced to improve the viscous pot element, and established the red clay-anchor grout interface shear fractional M||N (composed of Maxwell body and Newton body in parallel) relaxation model. The model parameters were yielded by regression analysis of relaxation test curves under partial shear displacements, and the relationship between the model parameters and the shear displacements was also obtained by fitting. Finally, the established fractional M||N relaxation model was applied to predict another part of the relaxation curve under shear displacement level. By comparing the integer-order M||N model, the Burgers model and the five-element model (H||M||M), the results indicate that the proposed fractional M||N relaxation model not only has the advantages of simple structure and fewer parameters, but also has higher fitting and prediction accuracy.

This study focuses on modifying the surface of an AA 5052-H32 aluminum alloy using plasma treatment. Discharge power, exposure time, and working gas were adjusted as process parameters to improve the adhesion between the aluminum alloy and glass fiber-reinforced thermoplastic (GFRTP) polycarbonate composite. The surface composition and morphology of the aluminum alloy sheet were analyzed by X-ray photoelectron spectroscopy and scanning electron microscopy, and surface roughness and wettability were evaluated using a surface roughness-measuring instrument and contact angle goniometry, respectively. The bonding performance of GFRTP/aluminum alloy was also assessed. The surface of the aluminum alloy was subjected to chemical treatment prior to plasma treatment. The results revealed that nitrogen plasma treatment led to a substantial increase (25%) in bonding strength due to the synergistic effect of rough surface mechanical bonding and chemical bonding through functional groups between the aluminum alloy and GFRTP. However, the improvement in surface wettability by plasma treatment is time dependent and may gradually diminish over time due to the re-adsorption of hydrocarbon contamination from the surrounding air.
