Summary: Purpose: Long-term valproate (VPA) treatment has been associated with hyperandrogenism and polycystic ovaries in women with epilepsy. The exact mechanisms of action of the drug on sex steroid hormone function are still unsettled. The aim of the present study was to investigate the action of VPA on basal and gonadotropin-stimulated steroid secretion in porcine ovarian follicular cells and to measure the conversion of testosterone to estradiol. Second, the action of VPA on proliferation and apoptosis of follicular cells was investigated. Methods: Small and medium follicles were obtained from pig ovaries on days 8‐10 and 14‐16 of the estrus cycle. Both follicular compartments, theca and granulosa cells, were cultured as a coculture resembling follicles in vivo. VPA in concentrations of 100 and 250 µg/ml was added to the control or gonadotropinstimulated cultures. Results: VPA caused a significant increase in basal and luteinizing hormone (LH)-stimulated testosterone secretion from small follicles, whereas in medium follicles, an increased basal but decreased LH-stimulated testosterone secretion was found. VPA caused decreased basal and follicle-stimulating hormone (FSH)-stimulated estradiol secretion by small follicles, whereas only the higher concentration decreased estradiol secretion in medium follicles. The conversion of testosterone to estradiol by small follicles was decreased under the influence of VPA in testosterone-alone and in testosterone-plus-FSH‐stimulated cultures, whereas this was seen at only the higher VPA concentration in medium follicles. VPA had no effect on cell proliferation and viability, whereas in a dose-dependent manner, VPA increased caspase-3 activity. Conclusions: VPA affected steroidogenesis in both unstimulated and gonadotropin-stimulated porcine ovarian follicular cells and inhibited the conversion of testosterone to estradiol. In addition, VPA may act as an apoptotic agent in both small and medium-sized follicles. Key Words: Valproate—Testosterone—Estrogen—LH—FSH—Polycystic ovaries—Apoptosis—Caspase-3.

High concentrations of polychlorinated biphenyls (PCBs) in polar bears from Svalbard have increased concern for that population's reproductive health. We examined whether there were associations between the plasma concentrations of PCBs and reproductive hormones [progesterone (P4)] and 17 beta-estradiol (E2)] in free-living female polar bears from Svalbard. Concentrations of P4 depended on reproductive status, and concentrations were lowest in females with offspring--females with cubs and females with yearlings. In these females, the P4 concentrations were positively correlated with plasma sigma PCBs (sum of all analyzed polychlorinated biphenyl congeners) concentrations. The sigma PCBs concentrations explained 27% of the variation in the P4 concentrations. There were no correlations between sigma PCBs and E2 and cortisol in any of the groups of polar bears, or between sigma PCBs and P4 in single polar bears. Although the sigma PCBs-P4 relationship in female polar bears with offspring is not evidence per se of a direct cause-effect association, the results indicate that PCBs may affect levels of P4 in polar bear females. There is a clear need to further assess the hormone balance and population health of polar bears at Svalbard.

Normal sexual development and subsequent reproductive function are dependent on appropriate testosterone production and action. The regulation of steroid hormones, including androgens, can be influenced by both biological and environmental factors, including environmental chemicals. Concentrations of organochlorines are considerably greater in Svalbard polar bears than in polar bears from other regions. Between 1995 and 1998, samples were collected from 121 male polar bears (Ursus maritimus) from the Svalbard area. In this study, testosterone concentration variations were described for male polar bears during different seasons and for all age groups. To study possible relationships between plasma testosterone concentrations and biological factors, such as age, axial girth, and extractable plasma fat, and organochlorine contaminants including hexachlorocyclohexanes, hexachlorobenzene, chlordanes, p,p'-DDE, and 16 individual polychlorinated biphenyl (PCB) congeners, identical statistical analyses were performed on the total population and a subsample of reproductively active adults. Of the biological factors, axial girth showed a significant positive relationship and percentage extractable fat and a significant negative relationship with the testosterone concentrations. Both the epsilon pesticides and epsilon PCBs made significant negative contributions to the variation of the plasma testosterone concentration. The continuous presence of high concentrations of organochlorines in male polar bears throughout their life could possibly aggravate any reproductive toxicity that may have occurred during fetal and early postnatal development.

Ecological risk assessment of persistent organic chemicals in marine mammals in the Arctic would comprise assessments of both exposure and effect and risk characterization. Results from a comprehensive collaborative study between Norwegian and Canadian researchers, comprising epizoological (ecological) studies (reproductive rate, offspring survival), experimental studies on the immune system function, monitoring studies (e.g. physiological assays of thyroid hormones, retinol, IgG and testosterone) and registration of biological data (e.g. sex, age, reproductive status, nutritional status) were coupled with PCB exposure data. Assessment of the results indicates that population status and health of polar bears with very high PCB levels may be at risk.

Summary 1 It is well known that gastrointestinal nematodes can have a significant impact on the growth of farmed ruminants. The clinical signs of these parasites are often subtle, with production losses mainly due to reductions in host appetite and gut function. However, little is known about the impact of this widespread group of parasites on wild ruminants. We use experiments and cross-sectional data to investigate the effects of gastrointestinal nematodes on a wild host, the Svalbard reindeer. 2 Individually marked reindeer were treated for parasites. Their body mass, back fat depth and fecundity were compared with the control group one year later. The effect of treatment on worm burdens was investigated in a subsample of animals that were culled 3 and 6 months after treatment. Also, the relationship between the intensity of infection and body mass, back fat depth and host pregnancy rates was investigated in cross-sectional data from culled reindeer. 3 The anthelmintic treatment caused an increase in the body mass, back fat depth and fecundity of the reindeer. Treatment depressed the abundance of adult parasites of Ostertagia gruehneri for at least 6 months, but had no significant effect on the abundance of adults of the other dominant parasite species, Marshallagi marshalli, or the abundance of larval stages of either species. 4 In the cross-sectional study, the probability of a reindeer being pregnant in late winter was negatively related to the abundance of adult O. gruehneri when controlling for host body mass. However, no clear evidence were found for an effect of parasitism on host condition in the cross-sectional study. 5 Our experimental results show for the first time in a natural ruminant host population that gastrointestinal nematodes can have a significant effect on host condition and fecundity. However, effects of infection on host condition was not detectable in the cross-sectional study. Also, we show that the experimental effects on host condition and fecundity is most likely to be due to a negative effect of O. gruehneri, while the experimental design did not allow detection of potential negative effects of M. marshalli.

Relationships among milk progesterone, concentrate allocation, energy balance (EB), milk yield and conception rate were studied in 146 lactations in 94 moderate yielding cows. All animals were of the dual purpose breed Norwegian cattle, and were monitored through their first and second lactations. The cows were assigned three different concentrate allocations and had free access to grass silage. Energy balance was estimated by subtracting energy required for maintenance and lactation from energy intake. Milk progesterone concentration was determined three times weekly from calving until pregnancy. The cumulative progesterone concentration was calculated as area under the progesterone curve for the first three luteal phases postpartum. The conception rate increased linearly by rising milk progesterone for values of cumulative progesterone in the lowest third of the range, whereas the likelihood of conception did not differ between milk progesterone concentrations within the upper two-thirds. This implies that the progesterone values were below a threshold value for optimal reproductive success in one-third of the services performed in this study. Milk progesterone concentrations during the third luteal phase postpartum were low when the high-energy diet was fed. Negative EB was associated with reduced values for milk progesterone during the third luteal phase in second parity cows. Likewise, milk yield was inversely related to progesterone levels during both the first and third luteal phases postpartum in second parity cows. Energy balance was higher and milk yield lower during peak lactation among second parity cows that conceived compared to cows that remained open after the first artificial insemination. The present study have demonstrated an association between likelihood of conception and the energy coverage in Norwegian cattle. This relationship is possibly mediated through progesterone deficiency.

Evidence that some of the fungal metabolites present in food and feed may act as potential endocrine disruptors is increasing. Enniatin B (ENN B) is among the emerging Fusarium mycotoxins known to contaminate cereals. In this study, the H295R and neonatal porcine Leydig cell (LC) models, and reporter gene assays (RGAs) have been used to investigate the endocrine disrupting activity of ENN B. Aspects of cell viability, cell cycle distribution, hormone production as well as the expression of key steroidogenic genes were assessed using the H295R cell model. Cell viability and hormone production levels were determined in the LC model, while cell viability and steroid hormone nuclear receptor transcriptional activity were measured using the RGAs. ENN B (0.01-100μM) was cytotoxic in the H295R and LC models used; following 48h incubation with 100μM. Flow cytometry analysis showed that ENN B exposure (0.1-25μM) led to an increased proportion of cells in the S phase at higher ENN B doses (10μM) while cells at G0/G1 phase were reduced. At the receptor level, ENN B (0.00156-15.6μM) did not appear to induce any specific (ant) agonistic responses in reporter gene assays (RGAs), however cell viability was affected at 15.6μM. Measurement of hormone levels in H295R cells revealed that the production of progesterone, testosterone and cortisol in exposed cells were reduced, but the level of estradiol was not significantly affected. There was a general reduction of estradiol and testosterone levels in exposed LC. Only the highest dose (100μM) used had a significant effect, suggesting the observed inhibitory effect is more likely associated with the cytotoxic effect observed at this dose. Gene transcription analysis in H295R cells showed that twelve of the sixteen genes were significantly modulated (p0.05) by ENN B (10μM) compared to the control. Genes HMGR, StAR, CYP11A, 3βHSD2 and CYP17 were downregulated, whereas the expression of CYP1A1, NR0B1, MC2R, CYP21, CYP11B1, CYP11B2 and CYP19 were upregulated. The reduction of hormones and modulation of genes at the lower dose (10μM) in the H295R cells suggests that adrenal endocrine toxicity is an important potential hazard.

The mycotoxin alternariol (AOH) is an important contaminant of fruits and cereal products. The current study sought to address the effect of a non-toxic AOH concentration on the proteome of the steroidogenic H295R cell model. Quantitative proteomics based on stable isotope labeling by amino acids in cell culture (SILAC) coupled to 1D-SDS-PAGE-LC-MS/MS was applied to subcellular-enriched protein samples. Gene ontology (GO) and ingenuity pathway analysis (IPA) were further carried out for functional annotation and identification of protein interaction networks. Furthermore, the effect of AOH on apoptosis and cell cycle distribution was also determined by the use of flow cytometry analysis. This work identified 22 proteins that were regulated significantly. The regulated proteins are those involved in early stages of steroid biosynthesis (SOAT1, NPC1, and ACBD5) and C21-steroid hormone metabolism (CYP21A2 and HSD3B1). In addition, several proteins known to play a role in cellular assembly, organization, protein synthesis, and cell cycle were regulated. These findings provide a new framework for studying the mechanisms by which AOH modulates steroidogenesis in H295R cell model.

The objective of the study was to compare two different flow cytometers to reveal if there are differences between them and to find the most suitable protocol for analysis of spermatozoa. These two flow cytometers; Cell Lab Quanta™ and Coulter Epics XL, have different principles to calculate cell size, electric volume, and forward scatter (FS), respectively. Flow cytometry is a valuable tool to assess various spermatozoa quality traits simultaneously, such as plasma membrane and acrosome integrity. A double- and triple-stain combination was performed to compare evaluation of these two parameters by both flow cytometers and to assess the need of a fluorescent probe to identify the spermatozoa. Propidium iodide was used to assess the proportion of dead spermatozoa, whereas Alexa Fluor(®) 488 conjugated peanut agglutinin (PNA- Alexa 488) was used to evaluate the percentage of acrosome intact and acrosome-reacted cells or degenerated cells. In the triple-stain protocol, MitoTracker(®) Orange (MO) was included to test the capacity of this probe to discriminate spermatozoa from egg yolk and debris particles present in the semen sample. Cryopreserved semen from 13 Norwegian Red bulls was included in the study and the semen was evaluated immediately after thawing and after 3 hr incubation at 37°C. The results show that there is good agreement between the instruments. Nevertheless, a significant difference was found in percentages of acrosome intact live spermatozoa (% AIL) when including MO as a spermatozoa identification probe, compared to assessment without MO, with the Coulter Epics XL, while no significant difference was found when including the probe with the Cell Lab Quanta. In conclusion, the results show that cell size measurement based on electronic volume used by the Cell Lab Quanta flow cytometer is more accurate than FS used by the Coulter Epics XL flow cytometer in identification of spermatozoa.

The relationship between milk fat progesterone concentration at first artificial insemination (AI) and reproductive performance of Norwegian Red Cattle dairy cows was investigated in a field study. Fifteen AI technicians collected milk samples from 2250 cows from 458 herds and progesterone was analysed in the milk fat portion of the samples. Logistic regression revealed decreased probability of non-return, and increased likelihood of repeat-breeding, with increasing progesterone concentrations at insemination. The odds ratio for non-return and for repeat-breeding between the minimum and maximum value for milk fat concentrations between 4 and 32.2ng/ml was 4.5 (P0.01) and 3.2 (P = 0.01), respectively. The variable 'Technician' did not significantly influence non-return rate or repeat-breeding. Progesterone concentration at AI was positively related to calving to last insemination interval, and to number of inseminations per cow (P0.05). The association between the progesterone concentration at insemination and calving interval was only marginally significant. Technician was significantly (P0.01) associated with interval from calving to first AI, interval from calving to last AI (P = 0.05), and number of AI per cow (P = 0.01). Technician was not significantly related to the calving interval. We conclude that increased suprabasal progesterone concentrations at the time of first insemination, and higher return rate at AI may, at least partly, be due to endocrinological asynchrony at AI, a condition which may lead to decreased fertility.

The aim of the present study was to investigate whether long-term exposure to valproate (VPA) alters follicular steroidogenesis and whether or not this effect is dependent on the degree of follicular development. Small- and medium-sized follicles were obtained from pig ovaries collected, respectively, at days 8–10 and 14–16 of oestrus cycle. Theca interna and granulosa cells were isolated from follicles and placed in the same well in the ratio 1 : 3 with or without the VPA in doses of 100, 300 and 500 μ g ml−1. The culture medium was changed after 2, 4, 6 and 8 days. In both types of follicles, VPA caused a significant and dose-dependent reduction in both testosterone and estradiol secretion from follicular cells. In small-sized follicles, the testosterone to oestrogen ratio increased at all doses used and after all lengths of time in culture. In medium-sized follicles, a significant increase in the testosterone to oestrogen ratio was only observed at the highest dose level. All doses of VPA caused a marked inhibition of progesterone secretion after 48 hours while during long-term VPA exposure progesterone gradually increased demonstrating luteinization of cells. In conclusion, the present study demonstrates a direct effect of VPA on steroidogenesis. The effect seems to differ to some extent depending on the follicular stage of development. The elevated ratio of testosterone to estradiol suggests that VPA inhibits the conversion of testosterone to estradiol.

Reindeer are either wild or kept under very extensive farming systems. They are seasonal breeders, with mating coinciding with the decreasing photoperiod in the autumn, and with calving in the spring. Little is known regarding the factors that influence reproduction in reindeer or of their reproductive physiology. Studies carried out to date have mainly focused on issues related to the population dynamics of wild populations and semi-domestic herds, and to a limited extent on the reproductive physiology of the female. Nor is much known about reproductive disorders and their medical treatment, or of the possibilities to manipulate or control reproduction by the use of hormones. Modern reproductive techniques such as artificial insemination and in vitro fertilisation, maturation and transfer of embryos have so far received scant attention. In the future, it is possible that reindeer under certain conditions might be kept in more intensive production systems. Limited access to high-quality winter pastures and increased demands for productivity have resulted in artificial feeding becoming a common practice in various reindeer herding areas in Scandinavia. In efforts to enhance the productivity of reindeer herds, attention has been focused on factors affecting reproduction in the female and survival of the offspring. Further knowledge on these issues seems necessary when developing strategies for optimalization of meat production in domestic herds and the harvesting of wild populations. This paper puts a broad focus on various aspects of reproduction, including factors influencing the fecundity of reproductively active females. In order to understand these effects it is important also to have a basic understanding of the reproductive physiology of these animals.

A comparative cohort study of reproductive performance in organic and conventional dairy husbandry was conducted using longitudinal data from the Norwegian National Board of Animal Production Recording from January 1, 1994 to December 31, 1996. The present study is the first study comparing reproductive efficiency in organic and conventional husbandry in which characteristics such as herd size, breeding season, milk yield, parity, breed, use of artificial insemination (AI), and geographical distribution were taken into account. The organically managed cohort comprised 998 lactation periods, and the conventionally managed cohort comprised 3016 lactation periods. Both groups were similar in herd size and geographical distribution. The following reproduction variables were studied: days open, calving interval, calving to first AI interval, calving to last AI interval, and AI per cow. No consistent difference in reproductive performance was found between the cohorts before adjustments were made for milk yield, breeding season, service, and parity. After inclusion of these independent variables in the repeated measures, mixed-model analyses, reproductive efficiency of organically managed dairy cows was impaired compared with those under conventional management. In organic dairy farming, breeding efficiency was difficult to maintain in cows bred during winter. Organic husbandry proved more efficient than did conventional husbandry in converting roughage into milk. Furthermore, the average multiparity percentage was higher in organically managed cows.

Dairy farms in southern Norway were surveyed to obtain information regarding reproduction management in tied herds. A total of 1613 farms were included in the analyses. Reproductive performance during the main breeding period of the year (November 1 to February 28) was measured using the following dependent variables: calving to first service and last service interval, number of artificial inseminations per cow, non-return rate at 60 days, and calving interval. Culling for failure to conceive was found to be associated with longer calving to first service interval, more inseminations per cow and lower non-return rate. More inseminations per cow and lower non-return rate were also recorded in herds where breeding was close to calving. Oestrous checks late in the evening and frequent observations were associated with shorter calving to last service interval and shorter calving interval. Calving to last service interval was prolonged if the farmers were occupied with routine work while conducting oestrous checks. Manual rectal pregnancy testing was of little importance for reproductive efficiency in dairy herds with good breeding performance. More inseminations per cow occurred in herds where oestrous checks were conducted systematically 3 and 6 weeks after service. Calving to last service interval and calving interval were shorter when only one person was responsible for the herd breeding management.

The study investigated the effect of the place of storage of milk in the mammary gland on progesterone concentrations in whole milk, skim milk and milk fat. Skim milk, milk fat and whole milk progesterone concentrations were lower (P0.05) in milk fractions obtained from the cisternal part of the mammary gland compared to those in the milk fractions from the alveoli. Mean milk fat concentrations did not mirror the changes in the mean skim milk, milk fat and whole milk progesterone concentrations. After administration of oxytocin, milk fat concentrations rose significantly (P0.01). At the same time, skim milk and milk fat progesterone concentrations remained unchanged (P0.05), compared to those in the milk fractions of alveolar origin, obtained before oxytocin administration. Skim milk and whole milk progesterone concentrations were higher (P0.01) in composite milk and in milk samples collected 1 h after milking, compared to concentrations in the milk samples collected before morning milking and at 3, 5, 7 and 9 h after milking. The results suggest that defatted milk, milk fat and whole milk progesterone concentrations were affected by the place of storage of the milk in the mammary gland, and that this effect is independent of milk fat content. Time of milk sampling, not the milk fat concentration, in relation to time of milking, was a critical factor in determining skim milk, milk fat and whole milk progesterone. The study also revealed that the concentrations of the other milk components, somatic cell count, lactose and protein were affected by the place of storage of milk in the mammary gland.

To investigate the causes and mechanisms of foetal loss in Norwegian dairy goats, blood parameters in 40 goats that lost foetuses were compared with those in 40 goats that experienced a normal pregnancy. High mean levels of 15-ketodihydro-PGF2alpha, and low mean levels of oestrone sulphate throughout pregnancy were associated with foetal loss. The mean oestrone sulphate level was low before abortion, and the distinct peak that occurred at parturition in the control goats was not observed in connection with abortion. Association of other blood parameters with foetal loss was not detected. Infectious agents and toxins did not appear to be major causes of foetal loss in this study. The normal level of progesterone and cortisol in goats with foetal loss indicated that the function of the corpus luteum and adrenal glands, respectively, were not disturbed. The rapid decline in progesterone level associated with foetal loss may therefore be a result, rather than the cause, of foetal death. The lowered level of oestrone sulphate and elevated level of 15-ketodihydro-PGF2alpha in goats with foetal loss clearly indicated that the endocrine foetal-placental function was disturbed.

The response to Claviceps purpurea sclerotia administration in pregnant goats was examined in terms of changes in the levels of plasma hormones, the development of pregnancy and kid production. Six treated goats were each given 15 mg milled sclerotia (i.e. 0.105 mg ergotamine) per kilogram live weight twice daily via a stomach tube from days 98 +/- 2 to 107 +/- 2 of gestation. Seven control goats were given water twice daily via a stomach tube during the same period of gestation. The goats were observed for clinical signs of disease, rectal temperatures and live weights were recorded and the condition of the foetuses was monitored by real-time ultrasonography. All control goats delivered live kids. In the treated group two goats aborted 33 and 47 days, respectively, after the start of the administration period, two goats each delivered one normal and one weak kid, and the two remaining goats delivered apparently normal kids. All six treated goats became depressed and had poor appetite during the period of sclerotia administration. Rectal temperatures were significantly increased and live weight changes significantly decreased in the animals in the treated group compared to the control group during the period of C. purpurea administration. Ultrasound examination revealed that foetal deaths occurred between 1 and 42 days before abortion or birth. The appearance of the aborted foetuses varied from fresh to mummified, depending on the number of days between foetal death and expulsion. Microbiological and serological investigations revealed no infectious causes of reproductive failure. The level of 15-ketodihydro-PGF2 alpha was high in goats that aborted following administration of C. purpurea compared with the level in control goats. The oestrone sulphate level did not increase before abortion in the treated goats as in the controls before parturition. There were also changes in these hormones in the four treated goats that delivered live kids, but the changes were considerably smaller. These findings indicate that the endocrine foetal-placental function was disturbed, probably due to injury caused by the C. purpurea toxin ergotamine in the placenta and foetus.

The nature of hormonal involvement in pubertal brain development has attracted wide interest. Structural changes within the brain that occur during pubertal development appear mainly in regions closely linked with emotion, motivation and cognitive functions. Using a sheep model, we have previously shown that peri-pubertal pharmacological blockade of gonadotropin releasing hormone (GnRH) receptors, results in exaggerated sex-differences in cognitive executive function and emotional control, as well as sex and hemisphere specific patterns of expression of hippocampal genes associated with synaptic plasticity and endocrine signaling. In this study, we explored effects of this treatment regime on the gene expression profile of the ovine amygdala. The study was conducted with 30 same-sex twin lambs (14 female and 16 male), half of which were treated with the GnRH agonist (GnRHa) goserelin acetate every 4th week, beginning before puberty, until approximately 50 weeks of age. Gene expression profiles of the left and right amygdala were measured using 8×15 K Agilent ovine microarrays. Differential expression of selected genes was confirmed by qRT-PCR (Quantitative real time PCR). Networking analyses and Gene Ontology (GO) Term analyses were performed with Ingenuity Pathway Analysis (IPA), version 7.5 and DAVID (Database for Annotation, Visualization and integrated Discovery) version 6.7 software packages, respectively. GnRHa treatment was associated with significant sex- and hemisphere-specific differential patterns of gene expression. The current study presents a novel objective measure for characterizing sexually active groups (SAG 3-5) and relates this measure to other behaviors of lactating Holstein-Friesian cows. Cows in SAG 3-5 were required to participate in a minimum of 1 estrus behavior per 5 min while staying within 3 m (2 cow lengths) of its partner(s) for a minimum of 5 mm. Twenty Holstein-Friesian cows were video-monitored continuously through 1 complete estrous cycle. (22 d). Standing behavior, SAG 3-5, secondary estrus signs (SEC), and other social and agonistic behaviors were recorded continuously. The period of mounting estrus (MTE) was divided into the 3 parts: prestand, standing estrus (STE), and poststand. The mean durations of MTE, prestand, STE, and poststand period were 12.9 +/- 1.84, 4.0 +/- 1.93, 7.1 +/- 1.44, and 1.8 +/- 0.57 h (n = 13). The fractions of time spent in SAG 3-5 during MTE, prestand, STE, and poststand period were 13, 8, 19, and 1% (n = 11). During MTE, cows participated, on average, in 5.8 +/- 1.24 SAG 3-5 and initiated 9.5 +/- 2.99 mounts, with mean durations of 0.25 +/- 0.03 h and 4.00 +/- 0.36 s, respectively. The novel measure, SAG 3-5 was a sign of long duration not confined only to groups of STE cows. On one day when no cows were in estrus and during the periods 4 to 24 h before and after MTE, no SAG 3-5 behaviors were observed. Luteal-phase cows participated in SAG 3-5 only when the partner was a single cow in estrus. The time spent in SAG 3-5 increased between 1 and 3 h before MTE and the prestand period (3 vs. 8%) and reached a peak level during STE. From STE to poststand, time spent in SAG 3-5 decreased considerably (19 vs. 1%). The observed decrease in nonmutual agonistic behaviors 4 to 24 h before MTE is suggested as an early sign of pre-estrus. Changes in SAG 3-5, agonistic behaviors, and SEC are suggested as indicators of the specific stages of MTE. Increased SEC initiated and SAG 3-5 were indicators of late pre-estrus and early estrus (prestand). Peak levels of SAG 3-5, SEC, and social agonistic behaviors were indicators of STE. A sudden decrease in behaviors, preceded by frequent interactions, was indicative of late estrus (poststand). On the basis of the findings reported here, we propose that SAG 3-5, as well as proceptive and receptive patterns of SEC and agonistic behaviors, be included in estrus detection protocols. Updated knowledge of these behavioral interactions may assist when determining the stage of estrus and the optimal time to breed dairy cows.

The “common sense” intervention by toxicology journal editors regarding proposed European Union endocrine disrupter regulations ignores scientific evidence and well-established principles of chemical risk assessment. In this commentary, endocrine disrupter experts express their concerns about a recently published, and is in our considered opinion inaccurate and factually incorrect, editorial that has appeared in several journals in toxicology. Some of the shortcomings of the editorial are discussed in detail. We call for a better founded scientific debate which may help to overcome a polarisation of views detrimental to reaching a consensus about scientific foundations for endocrine disrupter regulation in the EU., Environmental Health, 12 (1), ISSN:1476-069X

The effect of Toxoplasma gondii inoculation on pregnancy and on endocrine foetal-placental function in pregnant goats was studied. Five susceptible goats were inoculated subcutaneously with T. gondii bradyzoites at 71 +/- 2 days of gestation. Another five goats were used as controls. Plasma was analysed for progesterone, oestrone sulphate and 15-ketodihydro-PGF2 alpha. The condition of the foetuses was monitored by real-time ultrasonography. All inoculated goats aborted or delivered stillborn or weak kids 54-73 days after inoculation. None of the goats showed signs of general disease. In cases of foetal death, the ultrasound examination revealed that death occurred between day 1 and 12 before abortion or birth. The appearance of the foetuses varied from fresh to mummified, depending on the number of days between foetal death and expulsion. All five goats became serologically positive to T. gondii after inoculation. None of the goats used as controls aborted, but one goat delivered one mummified and one weak kid for unknown reasons. In inoculated animals an increase in 15-ketodihydro-PGF2 alpha levels in plasma and a subsequent tendency to a decrease in oestrone sulphate levels were observed from about day 40 after inoculation and until abortion or birth. High levels of 15-ketodihydro-PGF2 alpha were seen after foetal death. High levels of 15-ketodihydro-PGF2 alpha were not always followed by a drop in progesterone levels. The mean level of progesterone was slightly decreased after inoculation and onwards. The pattern of progesterone levels around abortion in the inoculated goats was very similar to the pattern around parturition in the control goats. However, 15-ketodihydro-PGF2 alpha levels were higher both before and after abortion in inoculated goats than in control goats. The level of oestrone sulphate did not increase in the inoculated group before abortion in contrast to the level in goats which delivered healthy kids. The patterns of changes in levels of 15-ketodihydro-PGF2 alpha and oestrone sulphate in inoculated animals indicate that the endocrine foetal-placental function was disturbed in most of the inoculated goats, probably due to the injury caused by the establishment and development of T. gondii infection in the placenta and foetus.

Norwegian dairy goats (n = 188) were fed either barn-dried hay, wilted silage of good quality or poor silage ad libitum plus 400 g concentrate/kg−1 milk for the first 90 days of pregnancy or to the end of lactation. From Day 91 to Day 120 of gestation they received either roughage ad libitum plus 100 g concentrate per goat daily or were fed only roughage to 70% of the maintenance energy requirement. From Day 121 of pregnancy until kidding all the goats were given 200 g barn-dried hay, good silage ad libitum and 900 g concentrate (12.5% DCP) per goat daily. Plasma glucose level did not differ between roughage groups whereas plasma acetoacetate (ACAC) and non-esterified fatty acids (NEFA) concentration increased by 33% and 20%, respectively, (P < 0.05) in goats fed poor quality silage compared with those fed hay or good silage. Plasma glucose concentration was lower and plasma NEFA and ACAC concentrations were higher (P < 0.05) in goats fed low energy between 91 and 120 days of gestation than in those receiving high energy during this period. Plasma glucose was lower (P < 0.01) whereas plasma NEFA and ACAC were higher in aborting goats compared with normal goats.

In many species sexual dimorphisms in brain structures and functions have been documented. In ovine model, we have previously demonstrated that peri-pubertal pharmacological blockade of gonadotropin releasing hormone (GnRH) action increased sex-differences of executive emotional behavior. The structural substrate of this behavioral alteration however is unknown. In this magnetic resonance image (MRI) study on the same animals, we investigated the effects of GnRH agonist (GnRHa) treatment on the volume of total brain, hippocampus and amygdala. In total 41 brains (17 treated; 10 females and 7 males, and 24 controls; 11 females and 13 males) were included in the MRI study. Image acquisition was performed with 3-T MRI scanner. Segmentation of the amygdala and the hippocampus was done by manual tracing and total gray and white matter volumes were estimated by means of automated brain volume segmentation of the individual T2-weighted MRI volumes. Statistical comparisons were performed with general linear models. Highly significant GnRHa treatment effects were found on the volume of left and right amygdala, indicating larger amygdalae in treated animals. Significant sex differences were found for total gray matter and right amygdala, indicating larger volumes in male compared to female animals. Additionally, we observed a significant interaction between sex and treatment on left amygdala volume, indicating stronger effects of treatment in female compared to male animals. The effects of GnRHa treatment on amygdala volumes indicate that increasing GnRH concentration during puberty may have an important impact on normal brain development in mammals. These novel findings substantiate the need for further studies investigating potential neurobiological side effects of GnRHa treatment on the brains of young animals and humans.

Summary Prenatal exposure to androgens has been shown to modulate brain development, resulting in changed behavioral attitudes, sexual orientation and cognitive functions, including processing of spatial information. Whether later changes in gonadotropic hormones during puberty induce further organizational effects within the brain is still insufficiently understood. The purpose of this study was to assess development of spatial orientation before and after the time of normal pubertal development, in an ovine model where half of the animals did not undergo typical reproductive maturation due to the pharmacological blockade of gonadotropin releasing hormone receptor (GnRHR) signaling. The study formed part of a larger trial and utilized 46 pairs of same sex Scottish Mule Texel Cross twins (22 female and 24 male). One twin remained untreated throughout (control) while the other received a subcutaneous GnRH agonist (GnRHa: Goserelin-Acetate) implant every fourth week. GnRHa treatment began at eight and 28 weeks of age, in males and females respectively, because the timing of the pubertal transition is sexually differentiated in sheep as it is in humans. Spatial orientation was assessed at three different time points: eight weeks of age, before puberty and treatment in both sexes; 28 weeks of age, after 20 weeks GnRHa treatment in males and before puberty and GnRHa treatment in females; and at 48 weeks of age, which is after the normal time of the pubertal transition in both sexes. Spatial orientation was tested in a spatial maze with traverse time as the main outcome measure. GnRHa treatment did not affect spatial maze performance as no significant differences in traverse time between treated and untreated animals were observed at any time-point. Adolescent females (48 weeks of age) traversed the maze significantly faster than adolescent males, whereas no sex differences in traverse time were seen at earlier developmental stages (eight and 28 weeks). Development of sex differences in spatial orientation was independent of exposure to pubertal hormones since puberty-blocked and control animals both showed the same pattern of spatial maze performance. This result demonstrates the prenatal nature of spatial orientation development. Furthermore, the unexpected finding that female animals outperformed males in the spatial orientation task, underscores the importance of the testing context in spatial orientation experiments.

Androstenone and testosterone levels in Duroc boars with an estimated breeding value for androstenone (EBV androstenone ) were followed in the period from birth to sexual maturity. The breeding value for androstenone had been estimated based on androstenone levels in 1202 Duroc boars at an age of 24 weeks. Testosterone and androstenone levels in plasma were recorded in 19 boars at 1 week of age and in their 15 respective litter-siblings at 3 weeks of age. Between 12 and 27 weeks of age, plasma levels were recorded weekly in a third set of 16 litter-siblings. In the last group, histomorphology was performed at 12, 16, 20, and 27 weeks of age to determine sexual maturity status. The EBV androstenone was positively related to plasma androstenone in animals 12 to 27 weeks of age and to plasma testosterone levels in 1- and 3-week-old animals. The EBV androstenone was not related to testis morphology. The concentration of fat androstenone was positively correlated to the percentage of immature seminiferous tubules and negatively correlated to the percentage of mature seminiferous tubules at 20 weeks. Testosterone in plasma showed no relationship with testis morphology. Most individuals reached puberty at 20 weeks of age, which indicates that Duroc mature later than crossbred boars. The results indicated that breeding value based on the single trait boar taint parameter EBV androstenone was not related to testicular development.

Oil pollution from various sources, including exploration, production and transportation, is a growing global concern. Of particular concern is the environmental impact of produced water (PW), the main waste discharge from oil and gas platforms. In this study, we have investigated the potential of polar hydrocarbon pollutants to disrupt or modulate steroidogenesis in vitro , using a human adrenocortical carcinoma cell line, the H295R assay. Effects of two of the major groups of compounds found in the polar fraction of crude oil and PW; alkylphenols (C 2 - and C 3 -AP) and naphthenic acids (NAs), as well as the polar fraction of PW as a whole has been assessed. Endpoints include hormone (cortisol, estradiol, progesterone, testosterone) production at the functional level and key genes for steroidogenesis (17β-HSD1, 17β-HSD4, 3β-HSD2, ACTHR, CYP11A1, CYP11B1, CYP11B2, CYP17, CYP19, CYP21, DAX1, EPHX, HMGR, SF1, STAR) and metabolism (CYP1A) at the molecular level. All compounds induced the production of both estradiol and progesterone in exposed H295R cells, while the C 3 -AP and NAs decreased the production of testosterone. Exposure to C 2 -AP caused an up-regulation of DAX1 and EPHX, while exposure to NAs caused an up-regulation of ACTHR. All compounds caused an up-regulation of CYP1A1. The results indicated that these hydrocarbon pollutants, including PW, have the potential to disrupt the vitally important process of steroidogenesis.

3-Methylsulfonyl-DDE (MeSO₂-DDE) is a potent adrenal toxicant formed from the persistent insecticide DDT. MeSO₂-DDE is widely present in human plasma, milk and fat, and in tissues of marine mammals. In the present study, we investigated endocrine-disrupting properties of MeSO₂-DDE in primary neonatal porcine Leydig cells. Unstimulated and LH-stimulated cells were exposed to MeSO₂-DDE at concentrations ranging from 0.6 to 20 μM for 48 h. Cell viability, hormone secretion and expression of steroidogenesis related genes were recorded. Secretion of testosterone and estradiol was increased in a concentration-dependent fashion in unstimulated Leydig cells, while in LH-stimulated cells, secretion of testosterone, estradiol and progesterone was decreased. The expression of important steroidogenic genes was down-regulated both in unstimulated and LH-stimulated cells. Notably, no significant impairment of cell viability occurred at any exposure except the highest concentration (20 μM) in LH-stimulated cells. This indicated that the effects on hormone secretion and gene expression were not caused by cytotoxicity. We conclude that the adrenal toxicant MeSO₂-DDE disrupts hormone secretion in a complex fashion in neonatal porcine Leydig cells. The different endocrine responses in unstimulated and LH-stimulated cells imply that the endocrine disruptive activity of MeSO₂-DDE is determined by the physiological status of the Leydig cells.

Ochratoxin A (OTA) is a mycotoxin and extrolite of fungi which has been reported in a range of foods. This study uses mammalian reporter gene assays (RGAs) with natural steroid receptors and the H295R steroidogenesis assay to assess the endocrine disrupting activity of OTA. At the receptor level, OTA (within a concentration range of 0.25–2500 ng/ml) did not induce an agonistic response in an oestrogen, androgen, progestagen or glucocorticoid RGA. An antagonistic effect was observed in all of the RGAs at the highest concentration tested (2500 ng/ml). However, while there was no significant cytotoxic effect observed in the MTT (thiazolyl blue tetrazolium bromide) cell viability assay at this concentration, there was a corresponding change in cell morphology which may be related to the resulting antagonistic effect. At the hormone production level, H295R cells were used as a steroidogenesis model and exposed to OTA (within a concentration range of 0.1–1000 ng/ml). Treatment of the cells with 1000 ng/ml OTA increased the production of estradiol (117 ± 14 ng/ml) over 3 times that of the solvent control (36 ± 9 pg/ml). Western blotting confirmed an increase in aromatase protein. Overall the results indicate that OTA does not appear to interact with steroid receptors but has the potential to cause endocrine disruption by interfering with steroidogenesis. This is the first study identifying the effect OTA may have on production of the steroid hormone estradiol.

Trichothecenes are a large family of chemically related mycotoxins. Deoxynivalenol (DON), T-2 and HT-2 toxins belong to this family and are produced by various species of Fusarium. The H295R steroidogenesis assay, regulation of steroidogenic gene expression and reporter gene assays (RGAs) for the detection of androgen, estrogen, progestagen and glucocorticoid (ant)agonist responses, have been used to assess the endocrine disrupting activity of DON, T-2 and HT-2 toxins. H295R cells were used as a model for steroidogenesis and gene expression studies and exposed with either DON (0.1-1000ng/ml), T-2 toxin (0.0005-5ng/ml) or HT-2 toxin (0.005-50ng/ml) for 48h. We observed a reduction in hormone levels in media of exposed cells following radioimmunoassay. Cell viability was determined by four colorimetric assays and we observed reduced cell viability with increasing toxin concentrations partly explaining the significant reduction in hormone levels at the highest toxin concentration of all three trichothecenes. This study reports the effects of peripubertal GnRH receptor inactivation on development of psychophysiological motoric reactivity (PMR; sometimes also called emotional reactivity), plasma cortisol concentrations and the relationship between plasma cortisol and PMR in male and female sheep. The study formed part of a larger trial and utilised 46 same sex twins. One twin remained untreated (control) while the other received a subcutaneous GnRH agonist (GnRHa Goserelin-Acetate) implant every 4th week, beginning at 8 and 28 weeks of age, in males and females, respectively (different, due to sex specific age of puberty). PMR, a measure of an animals' response to social isolation, was measured over a two minute period at 8, 28 and 48 weeks of age, using a three axis accelerometer. During the test period vocalisation rate was recorded. Cortisol was assayed in blood samples collected on a single day when animals were 40 weeks of age. PMR and vocalisation rate were significantly higher in females than males at all ages tested. At 28 weeks of age (20 weeks treatment) PMR was increased in treated males to the level seen in control females, by 48 weeks of age treated males' PMR was significantly less than controls. In females, 20 weeks of GnRHa treatment (28-48 weeks of age) was not associated with differences in PMR. Cortisol concentrations were significantly higher in females than males but were not affected by treatment. Plasma cortisol concentrations were positively correlated with PMR; this relationship being driven by the treated animals in both sexes. The results demonstrate that PMR is sexually dimorphic and cortisol dependent in sheep from at least 8 weeks of age. Importantly, they also demonstrate that long-term treatment of males with a GnRH agonist results in changes in age-dependent development of PMR.

Mycotoxins are secondary fungal metabolites that can cause pathological, physiological, and/or biochemical alterations in other species, including higher vertebrates. Zearalenone (ZEN) is produced by the genus Fusarium and after ingestion via contaminated cereals, may lead to animal fertility problems and reproductive pathologies. The compound has endocrine disrupting effects with a strong estrogenic action. Current data indicates that pig (hyperestrogenism) is amongst the most susceptible to ZEN, whilst in humans, this mycotoxin is suspected to be a triggering factor for central precocious puberty development in girls. Biotransformation of ZEN forms the metabolites α-zearalenol (α-ZOL) and β-zearalenol (β-ZOL), which are also produced by Fusarium in cultures, but in lower concentrations than ZEN.

In recent years sheep (Oviesaries) and goats (Capra aegagrushircus) have been increasingly used as model animals in toxicology and a plethora of effects due to exposure with polyhalogenated aromatic hydrocarbons such as polychlorinated biphenyls (PCBs) has been described in these species (1,7–10,12,13,16,17). The effects reported range from the molecular, to the organ and to the whole organism level including reproduction (12,13), behavior (8), stress adaptation (16,17), and bone tissue (7). PCBs have recently even been tested in vitro on their potential to induce changes in the protein pattern in exposed cells and a range of significant changes within the proteome of exposed cells has been described (11). To the best of our knowledge no toxicoproteomic studies have been performed on sheep or goat, despite the proven usefulness of the toxicoproteomic approach for toxicological research in other animals (2–5,14).

Assessing the role of weather in the dynamics of wildlife populations is a pressing task in the face of rapid environmental change. Rodents and ruminants are abundant herbivore species in most Arctic ecosystems, many of which are experiencing particularly rapid climate change. Their different life-history characteristics, with the exception of their trophic position, suggest that they should show different responses to environmental variation. Here we show that the only mammalian herbivores on the Arctic islands of Svalbard, reindeer ( Rangifer tarandus ) and sibling voles ( Microtus levis ), exhibit strong synchrony in population parameters. This synchrony is due to rain-on-snow events that cause ground ice and demonstrates that climate impacts can be similarly integrated and expressed in species with highly contrasting life histories. The finding suggests that responses of wildlife populations to climate variability and change might be more consistent in Polar regions than elsewhere owing to the strength of the climate impact and the simplicity of the ecosystem.

Persistent organic pollutants are widely distributed in the environment and lots of toxicological data are available. However, little is known on the intracellular fate of such compounds. Here a method applying secondary ion mass spectrometry is described that can be used to visualize cellular localization of halogenated compounds and to semi-quantitatively calculate concentrations of such compounds. Of the model compounds tested, TBBPA was homogenously distributed in the cell membrane of the H295R cells while PFOS accumulated in very distinct locations in the cell membrane. Relative intracellular concentrations of 4-OH-BDE69 and 4-OH-BDE121 in GH3.TRE were 61 % and 18 %, respectively, compared to the parent compounds. These differences may partly explain that observed effect concentrations for 4-OH-BDEs in in vitro experiments are usually lower than what would be expected based on receptor binding studies. NanoSIMS50 proved to be a powerful tool to describe the cellular distribution of halogenated compounds. The semi-quantitative data that can be obtained may help to further explain results from in vitro or in vivo experiments.

Reindeer oocytes were submitted to in vitro maturation, fertilization and culture (IVM,IVF,IVC) using the established procedures for bovine in vitro embryo production. The study was conducted outside the main breeding season. Semen was collected from epididymides immediately after slaughter, and was diluted in Tris-fructose-citric acid extender containing 6% glycerol and 20% egg-yolk and then frozen in liquid nitrogen. Following 24 h of maturation, cumulus expansion was complete, and 71% of the oocytes reached Metaphase II (MII), with extrusion of the first polar body. Of the remaining oocytes, 22% were at the germinal vesicle stage (GV), 2% at diakinesis and 5% at Metaphase I (MI). The percentages of fertilization and cleavage were 36.0 and 31.8%, respectively. Two of the fertilized oocytes developed to the morula stage after 7 d of culture.

The mycotoxin zearalenone (ZEN) is a secondary metabolite of fungi which is produced by certain species of the genus Fusarium and can occur in cereals and other plant products. Reporter gene assays incorporating natural steroid receptors and the H295R steroidogenesis assay have been implemented to assess the endocrine disrupting activity of ZEN and its metabolites α-zearalenol (α-ZOL) and β-zearalenol (β-ZOL). α-ZOL exhibited the strongest estrogenic potency (EC(50) 0.022±0.001 nM), slightly less potent than 17-β estradiol (EC(50) 0.015±0.002 nM). ZEN was ~70 times less potent than α-ZOL and twice as potent as β-ZOL. Binding of progesterone to the progestagen receptor was shown to be synergistically increased in the presence of ZEN, α-ZOL or β-ZOL. ZEN, α-ZOL or β-ZOL increased production of progesterone, estradiol, testosterone and cortisol hormones in the H295R steroidogenesis assay, with peak productions at 10 μM. At 100 μM, cell viability decreased and levels of hormones were significantly reduced except for progesterone. β-ZOL increased estradiol concentrations more than α-ZOL or ZEN, with a maximum effect at 10 μM, with β-ZOL (562±59 pg/ml)>α-ZOL (494±60 pg/ml)>ZEN (375±43 pg/ml). The results indicate that ZEN and its metabolites can act as potential endocrine disruptors at the level of nuclear receptor signalling and by altering hormone production.

International audience; In mammals, sex specialization is reflected by differences in brain anatomy and function. Measurable differences are documented in reproductive behavior, cognition, and emotion. We hypothesized that gonadotropin-releasing hormone (GnRH) plays a crucial role in controlling the extent of the brain's sex specificity and that changes in GnRH action during critical periods of brain development, such as puberty, will result in altered sex-specific behavioral and physiological patterns. We blocked puberty in half of the 48 same-sex Scottish mule Texel cross sheep twins with GnRH analog (GnRHa) goserelin acetate every 3 weeks, beginning just before puberty. To determine the effects of GnRHa treatment on sex-specific behavior and emotion regulation in different social contexts, we employed the food acquisition task (FAT) and measurement of heart rate variability (HRV). ANOVA revealed significant sex and sex x treatment interaction effects, suggesting that treated males were more likely to leave their companions to acquire food than untreated, while the opposite effect was observed in females. Concordant results were seen in HRV; treated males displayed higher HRV than untreated, while the reverse pattern was found in females, as shown by significant sex and sex x treatment interaction effects. We conclude that long-term prepubertal GnRHa treatment significantly affected sex-specific brain development, which impacted emotion and behavior regulation in sheep. These results suggest that GnRH is a modulator of cognitive function in the developing brain and that the sexes are differentially affected by GnRH modulation.

This study investigated the effects of two mixtures of persistent organic pollutants (POPs) on steroidogenesis in the H295R cell line. The two mixtures were obtained from the livers of burbot ( Lota lota ) caught in two Norwegian lakes (Mjosa and Losna) with different contaminant profiles. Steroid hormone levels in the cell culture medium and mRNA levels of 16 genes involved in steroidogenesis were investigated. The crude Lake Mjosa extract had to be diluted ten times more than the Lake Losna extract in order to prevent cytotoxicity. The ten times diluted Lake Mjosa mixture had higher levels of DDT and derivates (∑ DDTs, 1.7 times) and brominated flame retardants (∑ BDEs and HBCD, 15–25 times) than the Lake Losna mixture, which, on the other hand, had higher concentrations of ∑ PCBs (1.5 times higher) and also of HCB, ∑ HCH isomers and ∑ chlordane isomers (5–20 times higher). In the cell culture media, only cortisol levels were increased at the highest exposure concentration to the Lake Mjosa mixture, while both cortisol and estradiol levels were increased following exposure to the two highest Lake Losna mixture exposure concentrations. Testosterone levels decreased only at the highest exposure concentration of the Lake Losna mixture. Multivariate models suggested that ∑ PCBs, and to a lesser extent ∑ DDTs, were responsible for the cortisol responses, while estradiol and testosterone alterations were best explained by HCB and ∑ PCBs, respectively. Exposure to the mixtures generally increased mRNA levels, with smaller effects exerted by the Lake Mjosa mixture than the Lake Losna mixture. It was concluded that both mixtures affected steroidogenesis in the H295R cells. Small differences in mixture composition, rather than the high content of brominated flame retardants in the Lake Mjosa mixture, were suggested to be the most probable reason for the apparent differences in potencies of the two mixtures.

Crude cod liver oil and liver oil supplements are consumed as a source of vitamin A, D and polyunsaturated fatty acids; during winter and early pregnancy. Crude cod liver oil however constitutes a considerable source of persistent organic pollutants (POPs). This paper aimed at characterizing and quantifying the influence of POP mixtures extracted from three different steps in the cod liver oil industrial process on hormone production and the expression of steroidogenesis-related genes in H295R cells. Exposure to extracts from crude cod liver oil and from its industrial waste increased progesterone (P4), cortisol (Cort), testosterone (T) and estradiol (E2) production; and among others, the expression of MC2R, CYP11B1 and HSD3B2 genes. Observed effects after exposure to pharmaceutical cod liver oil extract were considerably lower. The type of effects on gene expression and hormone production were similar to those induced by forskolin and PCBs, the latter being the major contaminants within the extracts. Additional research is required to further unveil the mechanisms behind the observed steroidogenic effects and to assess whether the potential risk might outweigh the potential benefits of crude and processed cod liver oil consumption.

Endocrine disruptive effects have been frequently observed in patients using antiepileptic drugs (AEDs). Two different AEDs, valproate (VPA) and levetiracetam (LEV), were tested in forskolin-stimulated human adrenal carcinoma (H295R) cells to explore their effect on steroidogenesis. VPA has a long history as an anticonvulsant and is linked with many of the endocrine disorders associated with AED use. LEV is a newer AED, and no endocrine disruptive effects have been reported in humans to date.H295R cells, which are capable of full steroidogenesis, were stimulated with forskolin and exposed to either VPA or LEV for 48 h. Medium was collected and analyzed for hormone production. For the VPA-exposed cells, steroidogenic gene expression analysis was also conducted.VPA exposure resulted in a significant reduction in progesterone and estradiol (E2) production, whereas testosterone (T) levels remained unchanged. There were also significant alterations in expression level for most genes analyzed. LEV exposure resulted in a minor, but statistically significant, reduction in T and E2 production.Exposure of forskolin-stimulated H295R cells to VPA led to an increased T/E2 ratio through a significant decrease in estradiol production. Gene analysis suggested that VPA affects NR0B1 expression. NR0B1 inhibits promoters of other genes involved in steroidogenesis, and the altered expression of NR0B1 might explain the observed down-regulation in hormone production. The effects of LEV exposure on hormone secretion were not considered to be biologically significant.

The effects of in utero and lactational exposure to two structurally different polychlorinated biphenyl (PCB) congeners on follicular dynamics and the pituitary-gonadal axis in female lambs were investigated. Pregnant ewes received corn oil, PCB 118, or PCB 153, and offspring was maintained until 60 days postpartum. Ovarian follicles were quantified using stereology. Plasma luteinizing hormone (LH) and follicle-stimulating hormone (FSH) were measured using radioimmunoassay before and after administration of a gonadotropin releasing hormone (GnRH) analog. PCB 118 exposure increased numbers of transitional, secondary, and the sum of secondary, early antral, and antral (Σsecondary-antral) follicles, PCB 153 exposure only increased the number of primary follicles. GnRH-induced LH levels were significantly elevated in the PCB 153 exposure group. We conclude that PCB 153 and PCB 118 alter follicular dynamics in lambs and modulate the responsiveness of the pituitary gland to GnRH. © 2011 Wiley Periodicals, Inc. Environ Toxicol, 2011.

This study investigated the effects of exposure to the ubiquitous contaminants polychlorinated biphenyls (PCBs) on the fetal adrenal cortex and on plasma cortisol using the domestic sheep (Ovis aries) as a model. Pregnant ewes were intendedly subjected to oral treatment with PCB 153 (98 μg/kg bw/day), PCB 118 (49 μg/kg bw/day) or the vehicle corn oil from mating until euthanasia on gestation day 134 (±0.25 SE). However, because of accidental cross-contamination occurring twice causing a mixed exposure scenario in all three groups, the focus of this paper is to compare three distinct groups of fetuses with different adipose tissue PCB levels (PCB 153high, PCB 118high and low, combined groups) rather than comparing animals exposed to single PCB congeners to those of a control group. When comparing endocrine and anatomical parameters from fetuses in the PCB 153high (n = 13) or PCB 118high (n = 14) groups with the low, combined group (n = 14), there was a significant decrease in fetal body weight (P < 0.05), plasma cortisol concentration (P < 0.001) and adrenal cortex thickness (P < 0.001). Furthermore, adrenal weight was decreased and plasma ACTH was increased only in the PCB 118high group. Expression of several genes encoding enzymes and receptors related to steroid hormone synthesis was also affected and mostly down-regulated in fetuses with high PCB tissue levels. In conclusion, we suggest that mono-and di-ortho PCBs were able to interfere with growth, adrenal development and cortisol production in the fetal sheep model. © 2011 Wiley Periodicals, Inc. The objectives of the present study were to describe, in detail, behavior associated with standing estrus (STE) in lactating dairy cows and behavioral changes during complete estrous cycles. Estrus signs were monitored by continuous video recording of 20 Holstein-Friesian (HF) cows housed on an outdoor wood-chip pad during 1 estrous cycle (22 d). Other social behavior was recorded during STE and, for comparison, during 1 selected day when none of the cows were in estrus. Standing stationary when mounted was defined as the primary estrus sign. Anogenital sniff, chin rest, attempt to mount, and mount were defined as secondary estrus signs. Ovarian cyclicity was onfirmed by progesterone measurements. This study reports short mean duration of STE (7.1 ± 1.44 h) and estrus (mount period; 12.9 ± 1.84 h) of the 13 cows expressing these signs. All mounting activities involved at least one cow in, or within 4 h of, STE. The most frequent sign during STE was anogenital sniff initiated, followed by chin rest received, chin rest initiated, chase up initiated, anogenital sniff received, mount initiated, head butt, mount received, attempt to mount initiated, push away received, play rub, attempt to mount received, follow initiated, threat received, flehmen, avoid, bellow, and social lick received. Standing and mounting activity in HF cows was inconsistent during estrus, indicating that other signs could be of greater use. The frequency of secondary estrus signs initiated and received increased gradually during the last 12 h before STE, revealing significant differences between periods from 4 to 6 and 1 to 3 h before STE. A considerable increase in receptive behavior (secondary estrus signs received) was identified between 1 to 3 h prior to STE and STE. Both frequent initiated and received behaviors were associated with STE. A significant decrease in the frequency of secondary estrus signs initiated and received occurred 3 h after STE. Cows in STE simultaneously predominantly chose the other standing cow as mate and expressed secondary estrus signs more frequently. Based on the results of this study, we suggest that chase up could be regarded as a reliable indicator of estrus and that the changes in proceptive (initiated) and receptive (received) behavior could be used as predictors of different stages in estrus. Knowledge of these behavioral signs may improve heat detection rates and the ability to predict the optimum breeding time for dairy cows.

Polychlorinated biphenyls (PCB) are ubiquitous environmental pollutants that have been linked to adverse health effects including endocrine disruption. This study compared the mono-ortho-substituted PCB 118 and di-ortho-substituted PCB 153 with the non-ortho-substituted PCB 126, for possible effects on steroid hormone production and on the expression of 10 genes encoding proteins involved in steroidogenesis. The H295R human adenocarcinoma cell line was used as an in vitro model. Cells were exposed for 48 h to solvent control (dimethyl sulfoxide, DMSO) or 6 different concentrations ranging from 40 pM to 4 muM of one of the three test compounds. All three congeners significantly increased the production of estradiol-17beta. PCB 118 produced a rise in progesterone and cortisol in a concentration-dependent manner, similar to PCB 126. Testosterone was significantly reduced in response to PCB 153 but not PCB 118 or PCB 126. All three congeners elevated aldosterone at the highest concentration tested. A significant increase was observed in CYP11B2 mRNA levels in cells exposed to the three congeners. In addition, PCB 126 upregulated CYP19, 3beta-HSD2, StAR, and HMGR mRNA levels at the highest concentration tested, and downregulated CYP21 at 40 nM. In conclusion, all three PCB congeners are capable of modulating steroidogenesis in H295R in a concentration-dependent manner, whereby the hormone profile following PCB 118 exposure resembles that of PCB 126. Where changes in gene expression profile are concerned, exposure to PCB 126 showed the greatest effects.

The study-objective was to measure the effect of weight and growth related parameters on the risk of development of Canine Hip Dysplasia (CHD). The hypothesis was that heavy and fast growing dogs of large sized breeds were at increased risk of development of CHD compared to lighter and slower growing dogs. A prospective cohort study was conducted among dogs of four large breeds: Newfoundland (NF), Leonberger (LEO), Labrador retriever (LR), and Irish wolfhound (IW). The dogs were privately owned with individualized nutrition and environment, and they were followed from birth and throughout the growth period until the official screening for CHD was performed. The study sample consisted of 501 dogs from 103 litters, with the breed distribution 125 NF, 180 LEO, 133 LR, and 63 IW. Because the dogs were clustered in litters a multivariable random effects logistic regression model was used to assess statistically significant growth-related risk factors for CHD. The estimated incidence risk of CHD was 36% in NF, 25% in LEO, 20% in LR, and 10% in IW. Based upon the final multilevel model it appears that the odds of CHD among both LR and IW (odds ratio (OR) 0.22) are about one-fifth of the odds for NF. The odds for LEO (OR 0.60) are not significantly different from NF. There appeared to be an inverse relationship between body weight at 3 months of age and odds of CHD, with an OR of 0.89 (P = 0.044). The degree of clustering at the litter-level was high (22.6%) and highly significant (P < 0.001). Findings failed to support the hypothesis that heavy and fast growing dogs from four large sized breeds were at increased risk for development of CHD. There might be other unmeasured environmental risk factors for CHD in this cohort of dogs, although the contribution of the genetic variance to the litter-level clustering also needs further investigation.

Background Reproduction is the single greatest factor limiting beef cattle production. Previous research on beef suckler luteal activity has largely focused on the mechanisms, and duration, of postpartum anoestrus. However, the temporal pattern of luteal activity after resumption of post-partum ovarian activity, and the impact of pattern type on days open (DO) in purebred beef suckler cows, are unknown. Methods Progesterone concentration was measured in milk samples taken thrice weekly from 120 lactations, in 87 animals, on 3 farms, over two years. Onset of luteal activity (OLA) was defined as the first day milk progesterone concentration exceeded 3 ng/ml for two successive measurements, or exceeded 5 ng/ml once. It was defined as delayed if it occurred more than 61 days postpartum. A short initial luteal phase consisted of progesterone concentrations which exceeded 3 ng/ml for fewer than 4 sequential measurements. Temporal progesterone patterns were classified as: 1) Normal cyclicity; 2) Cessation of luteal activity; 3) Prolonged luteal activity; 4) Erratic phase: failure to conform to 1, 2 or 3. Data concerning parity, previous calving interval, breeding values, calf birth and 200-d weight were obtained from the Norwegian Beef Cattle Recording System database. Results The mean (SD) OLA was 41 d (20). Parity and calf birth weight were inversely correlated with OLA. Delayed OLA occurred in 14.4% of lactations. A short first luteal phase occurred in 61.5% of lactations, but this was unrelated to irregular luteal phase occurrence, pregnancy or DO. Irregular luteal phases occurred in 22% of lactations. The irregularities were: prolonged luteal phase (11%); cessation of luteal activity (5%); erratic luteal activity (6%). Early OLA was associated with prolonged luteal phases. DO was positively correlated with irregular luteal phases and negatively correlated with calf 200-d weight. Conclusions This study demonstrates that irregular luteal phases negatively affect reproductive performance in purebred beef suckler cattle. A moderate incidence of irregular luteal phases was seen in the study population. Whilst a positive relationship was seen between OLA and DO, unfavourable associations between early OLA and incidence of irregular luteal phases should be considered when developing breeding programmes.

The main objective of the study was to follow reproductive performance and blood biochemical parameters associated with fat and carbohydrate metabolism in a herd of free-ranging, semi-domesticated reindeer (Rangifer tarandus tarandus) over a two-year period, with extreme between-year variation in forage availability. The effects of climatic factors on reindeer reproductive performance were investigated by analysing whether time of onset of luteal function in autumn and calf survival were associated with changes in body mass and weather conditions, such as snow depth, precipitation, and temperature. Considerable between-year variation in the onset of luteal activity was found. In 1997, 4.2% of the female reindeer were either cycling or pregnant in the second week of October, whilst in 1998, in the same week, 100% were cycling or pregnant. Although energy balance was important for timing of the onset of luteal activity, delayed conception had no apparent effect on calf survival. The results indicated that maternal body mass (BM) in spring was of primary importance for calf survival, and the productivity of the herd. Since climatic factors influence the availability of forage, and hence female BM, it also has an indirect impact on calf survival. Females with low BM demonstrated greater seasonal variation in BM than heavier females. Plasma concentrations of free fatty acids and β-hydroxy butyric acid responded to changes in forage availability, but the initial condition of the reindeer and their fat reserves also seemed to have a major influence on these parameters.Abstract in Norwegian / Sammendrag:Sammenheng mellom års- og sesongvariasjon i kroppsvekt og henholdsvis reproduksjonssuksess og biokjemiske blodparametre hos tamreinHovedformålet med studien var å følge reproduksjonen og blodparametre knyttet til fett og karbohydratmetabolismen hos en tamreinflokk (Rangifer tarandus tarandus) gjennom en toårs periode med stor årlig variasjon i næringstilgang. Det ble funnet en betydelig forskjell mellom år når det gjaldt igangsetting av lutealfunksjon. I oktober 1997, var kun 4,2% av simlene drektige eller i lutealfasen av brunstsyklusen mens i den samme uken i 1998 var alle dyrene drektige eller i brunstens lutealfase. Selv om energibalanse var viktig for igangsetting av lutealfunksjonen, hadde forsinkelse i drektighetene ingen innflytelse på kalveoverlevelsen. Resultatene våre indikerte at simlenes vårvekt var av størst betydning for kalveoverlevelsen, og dermed også for produktiviteten til flokken. Siden de til enhver tid gjeldene værforhold påvirket tilgangen av fôr, påvirket dette også simlenes vekt med en indirekte effekt på kalveoverlevelsen. Simler med lav levendevekt viste større sesongvariasjon i vekt enn tyngre simler. Plasmakonsentrasjonen av frie fettsyrer, β-hydroxy-smørsyre forandret seg i takt med beitegrunnlaget, men den initiale kondisjonen og fettreservene hos simlene syntes også å påvirke disse parametrene.

Objectives - To better understand the interaction of antiepileptic drugs and production of sex hormones, possible effects of valproate (VPA), levetiracetam (LEV) and carbamazepine (CBZ) on steroidogenesis were investigated in the human adrenal carcinoma cell line H295R. Materials and methods - H295R cells were exposed to different concentrations of VPA, LEV or CBZ for 48 h. Sex hormone concentrations and mRNA expression levels were analyzed via radioimmunoassay and quantitative real time (RT)-PCR, respectively. Results - In VPA-exposed cells estradiol levels decreased in a dose-dependent manner, while testosterone and progesterone levels were unaffected. Expression of 3-hydroxy-3-methyl-glutaryl-CoA reductase (HMGR), steroidogenic acute regulatory protein (StAR), CYP11a, CYP17, CYP21, 3βHSD2, 17βHSD1 was downregulated and expression of CYP11β2 was upregulated. No effect on sex hormone production was observed under influence of LEV or CBZ. Expression of StAR, CYP17, CYP19 and 3βHSD2 was downregulated in LEVexposed cells, and expression of HMGR, CYP11β2 and CYP17 was downregulated in CBZ-exposed cells. Conclusions - VPA exposure resulted in a decrease in estradiol levels and a general downregulation of expression of genes encoding for enzymes early in steroidogenesis. No consistent changes were seen with LEV or CBZ exposure.

Relationships among commencement of luteal activity (C-LA), milk yield, and energy balance (EB) were investigated in 3 selection lines of Norwegian Red cows at the Norwegian University of Life Sciences from 1994 through 2001. The cows were selected for low genetic merit for milk yield (LMP), high genetic merit for milk yield (HMP), and a combination of high indices for milk yield and fertility (HI). Breeding values for fertility were based on 56-d nonreturn rate. The material included 268 lactations from 147 cows. Milk samples for progesterone analysis were drawn 3 times weekly from 1994 through 1998, and 2 times weekly from 1999 to 2001. Commencement of luteal activity was defined as the first 2 consecutive measurements of progesterone concentration >3 ng/mL not earlier than 10 d after calving. Selection line was significantly related to C-LA, so that the least squares mean days from calving to C-LA were 22.5, 30.4, and 27.2 d for LMP, HMP, and HI cows, respectively. The HMP cows produced more milk than the LMP cows. The average milk yield in the sixth week of lactation was 24.0, 27.1, and 25.3 kg for LMP, HMP, and HI cows, respectively. The interval to C-LA decreased for the HMP and HI cows after phenotypic adjustment for EB in the model. Least squares means for the interval to C-LA were 23.2, 29.7, and 25.6 d for the LMP, HMP, and HI cows, respectively, in a model that included parity, selection lines, and EB as covariates. Cumulated EB during the first 4 wk of lactation, which itself differed between selection lines, did not fully account for differences in interval to C-LA between selection lines. Thus, the results of the present investigation indicate that selection for milk yield negatively affects C-LA over and above the effects caused by concurrent changes in EB. The increase in days to C-LA caused by selection for high yields can be reduced if selection for milk yield is combined with fertility in the breeding program.
