Abstract Genome-wide association studies have succeeded in identifying genetic variants associated with complex diseases, but the findings have not been well interpreted biologically. Although it is widely accepted that epistatic interactions of high-order single nucleotide polymorphisms (SNPs) [(1) Single nucleotide polymorphisms (SNP) are mainly deoxyribonucleic acid (DNA) sequence polymorphisms caused by variants at a single nucleotide at the genome level. They are the most common type of heritable variation in humans.] are important causes of complex diseases, the combinatorial explosion of millions of SNPs and multiple tests impose a large computational burden. Moreover, it is extremely challenging to correctly distinguish high-order SNP epistatic interactions from other high-order SNP combinations due to small sample sizes. In this study, a multitasking harmony search algorithm (MTHSA-DHEI) is proposed for detecting high-order epistatic interactions [(2) In classical genetics, if genes X1 and X2 are mutated and each mutation by itself produces a unique disease status (phenotype) but the mutations together cause the same disease status as the gene X1 mutation, gene X1 is epistatic and gene X2 is hypostatic, and gene X1 has an epistatic effect (main effect) on disease status. In this work, a high-order epistatic interaction occurs when two or more SNP loci have a joint influence on disease status.], with the goal of simultaneously detecting multiple types of high-order (k 1-order, k 2-order, …, k n -order) SNP epistatic interactions. Unified coding is adopted for multiple tasks, and four complementary association evaluation functions are employed to improve the capability of discriminating the high-order SNP epistatic interactions. We compare the proposed MTHSA-DHEI method with four excellent methods for detecting high-order SNP interactions for 8 high-order epistatic interaction models with no marginal effect (EINMEs) and 12 epistatic interaction models with marginal effects (EIMEs) (*) and implement the MTHSA-DHEI algorithm with a real dataset: age-related macular degeneration (AMD). The experimental results indicate that MTHSA-DHEI has power and an F1-score exceeding 90% for all EIMEs and five EINMEs and reduces the computational time by more than 90%. It can efficiently perform multiple high-order detection tasks for high-order epistatic interactions and improve the discrimination ability for diverse epistasis models.

In order to make full use of the spatial detail information of remote sensing images, a remote sensing image fusion method based on convolution sparse representation and non-subsampled contourlet transform (NSCT) was proposed. First, the convolution sparse representation is used to establish a model to complete the super-resolution of image and achieve the purpose of detail enhancement. Then, the two images are fused, and the super-resolution image and panchromatic image are subjected to NSCT transformation to obtain their respective high-resolution sub-band images and low-resolution sub-band images. Appropriate methods are adopted according to the characteristics of different sub-bands. The new sub-band information is obtained by the fusion rules, and finally the NSCT inverse transform is performed to obtain fusion result. Experiments proved that the fusion image obtained by this method is superior to these of other methods in both visual effects and objective indicators.

In this study, we used the modified CRISPR/Cas9 system to produce targeted point mutations in cauliflower. Acetolactate synthase (ALS) and Centromere-specific histone H3 variant (CENH3) genes were selected as the base-editing targets and hypocotyls of cauliflower were used as explants. For ALS gene, a C-to-T conversion in the Pro182 codon (CCT) can alter the encoded amino acid, likely resulting in herbicide resistance, and a C-to-T mutation in the Leu133 codon (CTT) in the CENH3 gene may produce a haploid inducer. Results indicated that the transformation efficiency was 1.8%–4.5% and the mutation efficiencies for the ALS and CENH3 genes were approximately 22% and 87%, respectively. The ALS mutant cauliflower showed strong herbicide resistance, with possible immediate implications for broadleaf weed control in cauliflower fields.

Abstract Background In this study, it was aimed to evaluate the feasibility and effectiveness of full-size three-dimensional individual printed model (3D-IPM) based on computerized tomography (CT) reconstruction combined with 3D individual digital models (3D-IDMs) for improving the patient’s and their families’ comprehension levels of robotic-assisted laparoscopic partial nephrectomy (RALPN) preoperatively. Methods Between January 2020 and January 2021, 37 patients underwent RALPN in our institution. 3D individual digital models (3D-IDMs) were reconstructed based on the data of computerized tomography (CT) scanning and full-size 3D-IPMs were fabricated correspondingly. For each patient and his/her closest accompanying immediate family member (CAIFM) (spouse or son/daughter), two semi-structured conversations were held by using CT films (1st conversation) and 3D-IPM combined with 3D-IDM demonstration (2nd one) respectively. The preoperative levels of comprehension were evaluated quantitatively by using a self-made preoperative comprehending score (PCS) in the patients and CAIFMs. Results All the fabrications of full-size 3D-IPMs and all the operations were technically successful. The total PCS elevated significantly by presenting 3D-IPM combined with 3D-IDM demonstration compared with CT films (42.5 vs 35.5 in patients, P < 0.001; 42.9 vs 35.8 in CAIFMs, P < 0.001). Sub-PCSs in the evaluating aspects of renal anatomy, mass characteristics, the upcoming RALPN procedure, potential complication risks, and prognosis also showed a uniformed climbing pattern with the assistance of 3D-IPM+3D-IDM. Conclusion The application of 3D-IPM presentation combined with 3D-IDM demonstration can improve the preoperative comprehension of patient and CAIFM to RALPN with more direct-viewing and verisimilar presentation, and can be used in RALPN patient education for increasing patients’ and their families’ cognitive empowerment.

As formation mechanisms of plugging zone and criteria for fracture plugging remain unclear, plugging experiments and methods testing granular material mechanical properties are used to study the formation process of the plugging zone in fractured formations, analyze composition and ratios of different sizes of particles in the plugging zone, and reveal the essence and driving energy of the formation and damage of the plugging zone. New criteria for selecting lost circulation materials are proposed. The research results show that the formation of the plugging zone has undergone a process from inertial flow, elastic flow, to quasi-static flow. The plugging zone is composed of fracture mouth plugging particles, bridging particles and filling particles, and the proportion of the three types of particles is an important basis for designing drilling fluid loss control formula. The essence of the construction of the plugging zone is non-equilibrium Jamming phase transition. The response of the plugging zone particle system to pressure is driven by entropy force; the greater the entropy, the more stable the plugging zone. Lost circulation control formula optimized according to the new criteria has better plugging effect than the formula made according to conventional plugging rules and effectively improves the pressure-bearing capacity of the plugging zone. The research results provide a theoretical and technical basis for the lost circulation control of fractured formations.

Abstract Background This study was set to investigate the correlation between square dance and musculoskeletal system of early postmenopausal Chinese women. Methods Chinese postmenopausal women, who had been without menstruation for 1–10 years from the onset of menopause were recruited from community centers for this study. A standardized structured face-to-face interview was performed to collect demographic information, life styles, personal medical history, diet and menstrual status. Subjects who had been practicing regular square dance without participated in other sports activities for more than 2 years and over 4 h per week (usually more than 45 min per time and more than 5 times per week) were assigned to square dance group. Those postmenopausal women who had not participated in regular exercises (no more than 0.5 h per week) were recruited as the sedentary control group. Bone mineral density (BMD) of spine, total hip and femoral neck was measured by using dual-energy X-ray absorptiometry. Lower limb muscle strength was measured for the non-dominant leg, body flexibility was measured by a simple trunk bend-and-reach test, and body balance was evaluated using a single-stance test for the non-dominant leg. Independent two-tailed Student’s t-test was used for data analysis. Results 152 subjects from community centers were selected for this study and divided into square dance group (n = 74) and control group (n = 78). The square dance subjects had higher lumbar spine BMD (p = 0.01) and total hip BMD (p = 0.02) than control subjects, but there was no significant difference of femoral neck BMD (p = 0.48) between these two groups. Planet engulfment has been identified as one of the mechanisms for enhancing lithium abundance in stars. However, comprehensive investigations into lithium signatures following such events remain limited. Stars born together, sharing a common origin and stellar characteristics, provide a unique opportunity to study these signatures and compare lithium abundances. We demonstrate that the distinctive signature of planet engulfment in lithium abundance is only discernible among highly similar stellar twins. We present lithium abundance measurements for 125 comoving pairs of stars, representing the largest sample to date with a single, homogeneous assessment of high-precision lithium abundance. While lithium abundance enhancements in pairs showing planet engulfment signatures are within 0.35 dex, we find that even at fixed stellar parameters (temperature and age), the intrinsic scatter in lithium abundance is typically 0.35 dex for G/F dwarfs and can be as large as 0.6 dex for older and cooler stars due to internal stellar evolution processes. Since the planet engulfment signature from lithium can be masked by stellar intrinsic scatter, our findings raise questions about relying solely on lithium as an indicator for planet engulfment or attributing lithium-richness in stars primarily to planet engulfment events.

Due to their compelling performance and appealing simplicity, metric-based meta-learning approaches are gaining increasing attention for addressing the challenges of few-shot image classification. However, many similar methods employ intricate network architectures, which can potentially lead to overfitting when trained with limited samples. To tackle this concern, we propose using mutual distillation to enhance metric-based meta-learning, effectively bolstering model generalization. Specifically, our approach involves two individual metric-based networks, such as prototypical networks and relational networks, mutually supplying each other with a regularization term. This method seamlessly integrates with any metric-based meta-learning approach. We undertake comprehensive experiments on two prevalent few-shot classification benchmarks, namely miniImageNet and Caltech-UCSD Birds-200-2011 (CUB), to demonstrate the effectiveness of our proposed algorithm. The results demonstrate that our method efficiently enhances each metric-based model through mutual distillation.

Oral squamous cell carcinoma (OSCC) contributes to more than 90% of all oral malignancies, yet the performance of traditional treatments is impeded by limited therapeutic effects and substantial side effects. In this work, we report a combinational treatment strategy based on tumor exosome-based nanoparticles co-formulating a photosensitizer (Indocyanine green) and a tyrosine kinase inhibitor (Gefitinib) (IG@EXOs) for boosting antitumor efficiency against OSCC through synergistic phototherapy-molecular targeted therapy. The IG@EXOs generate distinct photothermal/photodynamic effects through enhanced photothermal conversion efficiency and ROS generation, respectively. In vivo, the IG@EXOs efficiently accumulate in the tumor and penetrate deeply to the center of the tumor due to passive and homologous targeting. The phototherapy effects of IG@EXOs not only directly induce potent cancer cell damage but also promote the release and cytoplasmic translocation of Gefitinib for achieving significant inhibition of cell proliferation and tumor angiogenesis, eventually resulting in efficient tumor ablation and lymphatic metastasis inhibition through the synergistic phototherapy-molecular targeted therapy. We envision that the encouraging performances of IG@EXOs against cancer pave a new avenue for their future application in clinical OSCC treatment.

Abstract Background A small hyolith, with a triangular operculum and a conical-pyramidal conch with a sharp apex, originally documented as Ambrolinevitus ventricosus, is revised based on new material from the Chengjiang biota. The operculum of ‘Ambrolinevitus’ ventricosus displays strong morphological similarities with the operculum of Paramicrocornus from the Shuijingtuo Formation (Cambrian Series 2), indicating that the species should be reassigned to Paramicrocornus. Results Based on the unusual morphology of Paramicrocornus, we herein propose a new family Paramicrocornidae fam. nov. A cladistic analysis of Cambrian and Ordovician hyoliths clearly delineates hyolithids as a monophyletic group which evolved from the paraphyletic orthothecids in the early Cambrian and with Paramicrocornidae as its closest relative. Conclusions The phylogenetic analysis, together with the distribution of hyoliths from the Cambrian to the Ordovician, reveals the presumptive evolution model of both the skeleton and soft-part anatomy of hyoliths. The Family Paramicrocornidae plays an intermediate role in hyolith evolution, representing the transitional stage in the evolution from orthothecids to hyolithids.

Abstract In order to improve the sensitivity and accuracy of oil‐paper insulation partial discharge detection by the fibre‐optic Fabry–Perot (FP) ultrasonic sensor, this work studied the ultrasonic signal’s frequency characteristics of typical oil‐paper insulation partial discharges and the vibration characteristics of the FP sensor’s diaphragm in the liquid environment. Based on the works above, a multifrequency FP sensor array is proposed, consisting of several FP sensors with different resonant frequencies to detect partial discharges. The experimental results show that the liquid environment has a significant effect on the vibration characteristics of the FP sensor’s diaphragm, and the sensitivity and accuracy of partial discharge detection can be improved based on the multifrequency FP sensing array, which can also be applied in the pattern recognition and localisation of partial discharges.

Abstract Background Previous studies have revealed that low frequency repeated transcranial magnetic stimulation (rTMS) on the contralesional primary motor cortex (cM1) is less effective in severe stroke patients with poor neural structural reserve than in patients with highly reserved descending motor pathway. This may be attributed to the fact that secondary motor cortex, especially contralesional dorsal premotor cortex (cPMd), might play an important compensatory role in the motor function recovery of severely affected upper extremity. The main purpose of this study is to compare the effectiveness of low frequency rTMS on cM1 and high frequency rTMS on cPMd in subcortical chronic stroke patients with severe hemiplegia. By longitudinal analysis of multimodal neuroimaging data, we hope to elucidate the possible mechanism of brain reorganization following different treatment regimens of rTMS therapy, and to determine the cut-off of stimulation strategy selection based on the degree of neural structural reserve. Methods/design The study will be a single-blinded randomized controlled trial involving a total of 60 subcortical chronic stroke patients with severe upper limb motor impairments. All patients will receive 3 weeks of conventional rehabilitation treatment, while they will be divided into three groups and receive different rTMS treatments: cM1 low frequency rTMS (n = 20), cPMd high frequency rTMS (n = 20), and sham stimulation group (n = 20). Clinical functional assessment, multimodal functional MRI (fMRI) scanning, and electrophysiological measurement will be performed before intervention, 3 weeks after intervention, and 4 weeks after the treatment, respectively. Discussion This will be the first study to compare the effects of low-frequency rTMS of cM1 and high-frequency rTMS of cPMd. The outcome of this study will provide a theoretical basis for clarifying the bimodal balance-recovery model of stroke, and provide a strategy for individualized rTMS treatment for stroke in future studies and clinical practice. The in vivo performances of most drugs after extravascular administration are fitted well with the two-compartment pharmacokinetic (PK) model, but the estimation of absorption rate constant (ka) for these drugs becomes difficult during unavailability of intravenous PK data. Herein, we developed a novel method, called the direct method, for estimating the ka values of drugs without using intravenous PK data, by proposing a new PK parameter, namely, maximum apparent rate constant of disposition (kmax). The accuracy of the direct method in ka estimation was determined using the setting parameters (k12, k21, and k10 values at high, medium, and low levels, respectively) and clinical data. The results showed that the absolute relative error of ka estimated using the direct method was significantly lower than that obtained using both the Loo-Riegelman method and the statistical moment method for the setting parameters. Human PK studies of telmisartan, candesartan cilexetil, and tenofovir disoproxil fumarate indicated that the ka values of these drugs were accurately estimated using the direct method based on good correlations between the ka values and other PK parameters that reflected the absorption properties of drugs in vivo (Tmax, Cmax, and Cmax/AUC0-t). This novel method can be applied in situations where intravenous PK data cannot be obtained and is expected to provide valuable support for PK evaluation and in vitro-in vivo correlation establishment.

The Ediacaran–Cambrian transition is characterized by an unprecedented change in biosphere, lithosphere, and atmosphere. However, the identification of this transition and its global correlation remains debated. East Yunnan is a region of utmost importance in the study of the Ediacaran-Cambrian transition. Although strata from this region have been well studied, recognition of the base of the Cambrian continues to be controversial. This paper presents, a carbon chemostratigraphic isotope and trace element profile through the Dengying and Zhujiaqing formations in the Sujiawa section in East Yunnan. Through carbon isotope and trace element analyses of the Dengying and Zhujiaqing formations we attempt to regionally correlate the section and discuss the paleo-marine redox environment changes during this period. The Fe content of samples is low and the Mn/Sr ratios of the majority of the Sujiawa section samples is lower than 10 and shows no correlations with δ13Ccarb and δ18Ocarb values, indicating that the samples retain near-primary δ13Ccarb characteristics. The carbon isotope profile from the Sujiawa section, shows a prominent negative δ13Ccarb in the basal Zhongyicun Member (N1’, −7.3‰) that most likely represents the prominent negative excursion recorded in Ediacaran-Cambrian strata across the world. REE patterns and Y/Ho ratios were used to screen samples, to ensure that the primary seawater REE features and Ce anomalies were preserved. The samples satisfying this screening process show seawater-like REE distribution patterns in leached carbonates and may have recorded Ce anomalies of the seawater from which the carbonates precipitated. These data show that the Baiyanshao Member has Ce/Ce* values between 0.57 and 0.88, the Daibu Member between 0.56 and 0.83, and the Zhongyicun Member between 0.60 and 0.96. The Ce anomalies through the section indicates that the region experienced suboxic-oxic-anoxic conditions throughout the Ediacaran-Cambrian period. Carbon isotope and trace element analyses indicate that the strata record a negative carbon isotope excursion event and anoxic conditions during the period of deposition. This study complements the carbon chemostratigraphic information and trace element data in the region and provides valuable clues for understanding the lithofacies changes through the Ediacaran and Cambrian in different regions of South China.

Abstract Neuropathic pain is difficult to treat in clinical practice, and the underlying mechanisms are insufficiently elucidated. Previous studies have demonstrated that the neuronal Fc‐gamma‐receptor type I (FcγRI) of the dorsal root ganglion (DRG) mediates antigen‐specific pain. However, the mechanisms of neuronal FcγRI in neuropathic pain remain to be explored. Here, it is found that the activation of FcγRI‐related signals in primary neurons induces neuropathic pain in a rat model. This work first reveals that sciatic nerve injury persistently activates neuronal FcγRI‐related signaling in the DRG, and conditional knockout (CKO) of the FcγRI‐encoding gene Fcgr1 in rat DRG neurons significantly alleviates neuropathic pain after nerve injury. C‐reactive protein (CRP) is increased in the DRG after nerve injury, and CRP protein of the DRG evokes pain by activating neuronal FcγRI‐related signals. Furthermore, microinjection of naive IgG into the DRG alleviates neuropathic pain by suppressing the activation of neuronal FcγRI. These results indicate that the activation of neuronal CRP/FcγRI‐related signaling plays an important role in the development of neuropathic pain in chronic constriction injury (CCI) rats. The findings may provide novel insights into the neuroimmune responses after peripheral nerve injury and suggest potential therapeutic targets for neuropathic pain.

Dissolved organic matter (DOM) plays a major role in ecological systems, affecting the fate and transportation of iron (Fe) and phosphorus (P). To better understand the geochemical cycling of these components, soil and sediment samples were collected around a reservoir downstream of a typical temperate forest in Northeast China. The DOM fractions from these soils, river, and reservoir sediments were extracted and then characterized by spectroscopic techniques. Comparative characterization data showed that the DOM pool of the Xishan Reservoir was partly autochthonous and derived from runoff and deposition of material in terrestrial ecosystems upstream. IntroductionPlant richness is thought to improve the function of constructed wetlands (CWs), but most CWs are planted with monocultures, with only a few employed polycultures, which have drawn contradictory conclusions. We suppose functional diversity is the key to better performance of plant communities and hypothesize that CWs planted with diverse growth forms are superior in plant growth and nutrient removal.MethodsIn this study, six emergent plant species categorized into slender type (Schoenoplectus tabernaemontani, Typha orientalis), fan type (Iris sibirica, Acorus calamus) and large type (Canna indica and Thalia dealbata) were planted in monocultures, combinations (two species of the same growth form) and mixed polycultures (six species of three growth forms). We then compared how plant growth and nutrient uptake differed among treatments.ResultsIt showed that the polyculture considerably increased the removal of total nitrogen (TN) and total phosphorus (TP), but the combination did not outperform monoculture. High consistency in the patterns between underground biomass and total biomass indicated that plant roots were essential for nutrient consumption. Compared with slender and fan plants, the large plants had a greater biomass increase in polycultures, which greatly accelerated the absorption and assimilation of TN and TP.ConclusionOur study indicated that plant community with various growth forms reduced the intensity of interspecific competition, increased the functional diversity, and greatly enhanced the ability of pollutant removal. Our results also provide some suggestions for plant selection and combination designs in CWs.

ObjectiveTo perform molecular genetic analysis of a patient diagnosed with primary hypertrophic osteoarthropathy (PHO) with malnourishment, intussusception, and acro-osteolysis.Case presentationAt the age of 7 years, a boy born to a consanguineous couple was diagnosed with PHO attributed to delayed closure of the cranial suture, eczema, clubbing of fingers, and swelling of the knee and ankle. Clinical characteristics and follow-up data for 3 years were collected and analyzed. Trio whole-exome sequencing (WES) and copy number variant sequencing were used to screen for causative genetic variants. Candidate variants of the patient and his parents were confirmed by Sanger sequencing. When he was 7 years old, trio WES found that he had biallelic novel variants c.498 + 1G > A, inherited from his parents, in the HPGD gene. The patient was markedly malnourished. Ultrasonography and computed tomography showed intussusception with a gradual expansion of the duodenum, localized intestinal wall thickening, and acro-osteolysis. Cross-sectional blood tests showed that the patient had continuously decreased levels of serum 25-hydroxy vitamin D and serum ferritin at the age of 7and 10 years.ConclusionPHO due to HPGD defects is rare in pediatric patients, and finding homozygous novel c.498 + 1G > A has expanded the spectrum of causative variants of HPGD and provided a clue for genotype-phenotype correlation analysis. Similar to mouse model results, human HPGD deficiency may also cause abnormal digestive tract development, and related secondary vitamin D deficiency and acro-osteolysis should be considered in HPGD-related PHO.

Abstract Subarachnoid hemorrhage (SAH) is a severe subtype of stroke caused by the rupturing of blood vessels in the brain. The ability to accurately assess the degree of bleeding in an SAH model is crucial for understanding the brain‐damage mechanisms and developing therapeutic strategies. However, current methods are unable to monitor microbleeding owing to their limited sensitivities. Herein, a new bleeding assessment system using a bioprobe TTVP with aggregation‐induced emission (AIE) characteristics is demonstrated. TTVP is a water‐soluble, small‐molecule probe that specifically interacts with blood. Taking advantage of its AIE characteristics, cell membranes affinity, and albumin‐targeting ability, TTVP fluoresces in bleeding areas and detects the presence of blood with a high signal‐to‐noise (S/N) ratio. The degree of SAH bleeding in an endovascular perforation model is clearly evaluated based on the intensity of the fluorescence observed in the brain, which enables the ultrasensitive detection of mirco‐bleeding in the SAH model in a manner that outperforms the current imaging strategies. This method serves as a promising tool for the sensitive analysis of the degree of bleeding in SAHs and other hemorrhagic diseases.

Abstract Protonic ceramic electrochemical cells (PCECs) have been intensively studied as the technology that can be employed for power generation, energy storage, and sustainable chemical synthesis. Recently, there have been substantial advances in electrolyte and electrode materials for improving the performance of protonic ceramic fuel cells and protonic ceramic electrolyzers. However, the electrocatalytic materials development for synthesizing chemicals in PCECs has gained less attention, and there is a lack of systematic and fundamental understanding of the PCEC reactor design, reaction mechanisms, and electrode materials. This review comprehensively summarizes and critically evaluates the most up‐to‐date progress in employing PCECs to synthesize a wide range of chemicals, including ammonia, carbon monoxide, methane, light olefins, and aromatics. Factors that impact the conversion, selectivity, product yield, and energy efficiencies are discussed to provide new insights into designing electrochemical cells, developing electrode materials, and achieving economically viable chemical synthesis. The primary challenges associated with producing chemicals in PCECs are highlighted. Approaches to tackle these challenges are then offered, with a particular focus on deliberately designing electrode materials, aiming to achieve practically valuable product yield and energy efficiency. Finally, perspectives on the future development of PCECs for synthesizing sustainable chemicals are provided.

Achieving high loading of active sulfur yet rational regulating the shuttle effect of lithium polysulfide (LiPS) is of great significance in pursuit of high-performance lithium-sulfur (Li-S) battery. Herein, we develop a free-standing graphene-nitrogen (N), phosphorus (P) and fluorine (F) co-doped mesoporous carbon-sulfur (G-NPFMC-S) film, which was used as a binder-free cathode in Li-S battery. The developed mesoporous carbon (MC) achieved a high specific surface area of 921 m2·g–1 with a uniform pore size distribution of 15 nm. The inserted graphene network inside G-NPFMC-S cathode can effectively improve its electrical conductivity and simultaneously restrict the shuttle of LiPS. A high sulfur loading of 86% was achieved due to the excellent porous structures of graphene-NPFMC (G-NPFMC) composite. When implemented as a freestanding cathode in Li-S battery, this G-NPFMC-S achieved a high specific capacity (1,356 mAh·g–1), favorable rate capability, and long-term cycling stability up to 500 cycles with a minimum capacity fading rate of 0.025% per cycle, outperforming the corresponding performances of NPFMC-sulfur (NPFMC-S) and MC-sulfur (MC-S). These promising results can be ascribed to the featured structures that formed inside G-NPFMC-S film, as that highly porous NPFMC can provide sufficient storage space for the loading of sulfur, while, the N, P, F-doped carbonic interface and the inserted graphene network help hinder the shuttle of LiPS via chemical adsorption and physical barrier effect. This proposed unique structure can provide a bright prospect in that high mass loading of active sulfur and restriction the shuttle of LiPS can be simultaneously achieved for Li-S battery.

Abstract Background Classical understanding of allergic conjunctivitis (ACJ) suggests that ocular itch results from a mast cell-dependent inflammatory process. However, treatments that target inflammatory mediators or immune cells are often unsatisfying in relieving the stubborn itch symptom. This suggests that additional mechanisms are responsible for ocular itch in ACJ. In this study, we aim to determine the role of neuronal FcεRIa in allergic ocular itch. Methods Calcium imaging was applied to observe the effect of IgE-immune complex in trigeminal neurons. Genomic FcεRIa knockout mice and adeno-associated virus (AAV) mediated sensory neuron FcεRIa knockdown mice were used in conjunction with behavioral tests to determine ocular itch. In addition, immunohistochemistry, Western blot and quantitative RT-PCR were used for in vitro experiments. Results We found that FcεRIα was expressed in a subpopulation of conjunctiva sensory neurons. IgE-IC directly activated trigeminal neurons and evoked acute ocular itch without detectible conjunctival inflammation. These effects were attenuated in both a global FcεRIa-knockout mice and after sensory neuronal-specific FcεRIa-knockdown in the mouse trigeminal ganglion. In an ovalbumin (OVA) induced murine ACJ model, FcεRIα was found upregulated in conjunctiva-innervating CGRP+ sensory neurons. Sensory neuronal-specific knockdown of FcεRIa significantly alleviated ocular itch in the ACJ mice without affecting the immune cell infiltration and mast cell activation in conjunctiva. Although FcεRIα mRNA expression was not increased by IgE in dissociated trigeminal ganglion neurons, FcεRIα protein level was enhanced by IgE in a cycloheximide-resistance manner, with concordant enhancement of neuronal responses to IgE-IC. In addition, incremental sensitization gradually enhanced the expression of FcεRIα in small-sized trigeminal neurons and aggravated OVA induced ocular itch. Conclusions Our study demonstrates that FcεRIα in pruriceptive neurons directly mediates IgE-IC evoked itch and plays an important role in ocular itch in a mouse model of ACJ. These findings reveal another axis of neuroimmune interaction in allergic itch condition independent to the classical IgE-mast cell pathway, and might suggest novel therapeutic strategies for the treatment of pruritus in ACJ and other immune-related disorders.

Abstract Background Osteoporosis (OP) is a systemic bone disease manifested as low bone mass, destruction of bone microstructure, increased bone fragility and fracture risk. The purpose of this study was to explore the role and mechanism of PDX1 for osteogenic differentiation of adipose derived stem cells (ADSCs). Methods GSE37329 dataset was retrieved from NCBI Gene Expression Omnibus (GEO) database and performed bioinformatic analyses. ADSCs were incubated with normal medium, osteogenic induction medium (OIM) and OIM+si-PDX1. Then, alkaline phosphatase (ALP) staining and Alizarin Red Staining (ARS) were performed to assess the role of PDX1 for osteogenesis of ADSCs. PI3K inhibitor, LY294002 was then added to further explore the mechanism of PDX1 for osteogenic differentiation of ADSCs. Western blot assay was used to assess the osteogenic-related markers. Graphpad software was used to perform statistically analysis. Results A total of 285 DEGs were obtained from analysis of the dataset GSE37329, of which 145 were upregulated and 140 were downregulated genes. These differentially expressed genes mainly enriched in cell differentiation and PI3K/Akt signaling pathway. Moreover, PDX1 was decreased in osteogenic induced ADSCs. Knockdown of PDX1 significantly increased osteogenic differentiation capacity and p-PI3K and p-Akt protein levels. Administration with LY294002 could partially reversed the promotion effects of si-PDX1. Conclusion In conclusion, knockdown of PDX1 promotes osteogenic differentiation of ADSCs through the PI3K/Akt signaling pathway.

Abstract Background Previous studies did not comprehensively examine the effect of adenotonsillectomy on growth and development, emotional state, quality of life, attention ability, and cognitive dysfunction in children with obstructive sleep apnea (OSA). This study aimed to explore the improvement effects of adenotonsillectomy on the growth, development, quality of life, and attention ability in children with OSA. Methods This prospective single-arm study involved children with OSA admitted at The No. 980 Hospital, Joint Logistics Support Force, PLA, China (02/2017–02/2018). The Myklebust Pupil Rating Scale (PRS), Inventory of Subjective Life Quality (ISLQ), Zung Self-rating Anxiety Scale (SAS), Conners Parent Symptom Questionnaire (PSQ), and Continuous Performance Task (CPT) were examined before and at 6 months after adenotonsillectomy. Results Forty-nine patients were enrolled. They all completed the 6-month follow-up. The body mass index increased after surgery (from 18.8 ± 4.9 to 19.3 ± 4.3 kg/m2, P = 0.008). A novel drug monitoring method is experimentally demonstrated by optically pumped atomic magnetometer. An elliptically polarized single-beam spin-exchange-relaxation-free atomic magnetometer (SERF AM) is used to realize magnetorelaxometry (MRX) measurement and thereby quantify concentrations of superparamagnetic nanoparticles in solutions. Moreover, drug-conjugated magnetic nanoparticles are monitored in mice for twelve hours to study the drug sustained-release mechanism. Our results show the great potential of optically pumped atomic magnetometer for application in biophysical, pharmaceutical and medical areas, and may promote fields such as drug delivery research, virus detection, and molecule quantification.

The integration of sensing and communication (ISAC) functionalities have recently gained significant research interest as a hardware-, power-, spectrum- and cost- efficient solution. This experimental work implements a dual-functional sensing and communication framework where a single radiation waveform, either omnidirectional or directional, can realize both sensing and communication functions. We design an orthogonal frequency division multiplexing (OFDM) based multi-user multiple input multiple output (MIMO) software-defined radio (SDR) testbed to validate the dual-functional model. We carry out over-the-air experiments to investigate the optimal trade-off factor to balance the performance for both functions. On the communication side, we obtain bit error rate (BER) results from the testbed to show the communication performance using the dual-functional waveform. On the sensing performance, we measure the output beampatterns of our transmission to examine their similarity to simulation based beampatterns. We also implement a sensing experiment to realize activity detection functions. Our experiment reveals that the dual-functional approach can achieve comparable BER performance with pure communication-based solutions while achieving fine sensing beampatterns and realistic sensing functionality simultaneously.

Qinghai-Tibet plateau with an average altitude above 4,000 m provides favorable conditions for Tibetan pigs that may have different meat quality from other pig breeds. This study was designed to compare the differences in lipid metabolism in liver and fatty acid composition in muscle between Tibetan and Duroc × Landrace × Yorkshire (DLY) pigs. Aba Tibetan pigs (n = 10), Gannan Tibetan pigs (n = 9), Nyingchi Tibetan pigs (n = 10) and DLY pigs (n = 10) were selected for the experiment. After fasting for 12 h, they were slaughtered and blood, liver and muscle samples were taken for biochemical analyses. The results showed that the intramuscular fat content was not significantly different for Tibetan pigs among the three regions (P > 0.05), which was significantly higher than that of DLY pigs (P < 0.05). However, the liver fat content of Gannan Tibetan pigs was significantly lower than those of DLY pigs and Aba and Nyingchi Tibetan pigs (P < 0.05). The hepatic lipid metabolism may be stronger in DLY pigs than in Tibetan pigs, but lipid deposition in muscle is weaker in DLY pigs than that in Tibetan pigs. Aba Tibetan pigs and Nyingchi Tibetan pigs had similar proportions of saturated, monounsaturated and polyunsaturated fatty acids, while Gannan Tibetan pigs and DLY pigs had similar types of fatty acids. The findings provide new insight into mechanisms of environmental and breed effects on pork quality.

Abstract DNA variants in or close to the human TBX15 and PAX1 genes have been repeatedly associated with facial morphology in independent genome‐wide association studies, while their functional roles in determining facial morphology remain to be understood. We generated Tbx15 knockout (Tbx15−/−) and Pax1 knockout (Pax1−/−) mice by applying the one‐step CRISPR/Cas9 method. A total of 75 adult mice were used for subsequent phenotype analysis, including 38 Tbx15 mice (10 homozygous Tbx15−/−, 18 heterozygous Tbx15+/−, 10 wild‐type Tbx15+/+ WT littermates) and 37 Pax1 mice (12 homozygous Pax1−/−, 15 heterozygous Pax1+/−, 10 Pax1+/+ WT littermates). Facial and other physical morphological phenotypes were obtained from three‐dimensional (3D) images acquired with the HandySCAN BLACK scanner. Compared to WT littermates, the Tbx15−/− mutant mice had significantly shorter faces (p = 1.08E‐8, R2 = 0.61) and their ears were in a significantly lower position (p = 3.54E‐8, R2 = 0.62) manifesting a “droopy ear” characteristic. Besides these face alternations, Tbx15−/− mutant mice displayed significantly lower weight as well as shorter body and limb length. Pax1−/− mutant mice showed significantly longer noses (p = 1.14E‐5, R2 = 0.46) relative to WT littermates, but otherwise displayed less obvious morphological alterations than Tbx15−/− mutant mice did. We provide the first direct functional evidence that two well‐known and replicated human face genes, Tbx15 and Pax1, impact facial and other body morphology in mice. The general agreement between our findings in knock‐out mice with those from previous GWASs suggests that the functional evidence we established here in mice may also be relevant in humans.

BackgroundThe immune-inflammatory response has been widely considered to be involved in the pathogenesis of post-stroke depression (PSD), but there is ambiguity about the mechanism underlying such association.MethodsAccording to Diagnostic and Statistical Manual of Mental Disorders (5th edition), depressive symptoms were assessed at 2 weeks after stroke onset. 15 single nucleotide polymorphisms (SNPs) in genes of indoleamine 2,3-dioxygenase (IDO, including IDO1 and IDO2) and its inducers (including pro-inflammatory cytokines interferon [IFN]-γ, tumor necrosis factor [TNF]-α, interleukin [IL]-1β, IL-2 and IL-6) were genotyped using SNPscan™ technology, and serum IDO1 levels were detected by double-antibody sandwich enzyme-linked immune-sorbent assay.ResultsFifty-nine patients (31.72%) were diagnosed with depression at 2 weeks after stroke onset (early-onset PSD). The IDO1 rs9657182 T/T genotype was independently associated with early-onset PSD (adjusted odds ratio [OR] = 3.008, 95% confidence interval [CI] 1.157-7.822, p = 0.024) and the frequency of rs9657182 T allele was significantly higher in patients with PSD than that in patients with non-PSD (χ2 = 4.355, p = 0.037), but these results did not reach the Bonferroni significance threshold (p > 0.003). Serum IDO1 levels were also independently linked to early-onset PSD (adjusted OR = 1.071, 95% CI 1.002-1.145, p = 0.044) and patients with PSD had higher serum IDO1 levels than patients with non-PSD in the presence of the rs9657182 T allele but not homozygous C allele (t = -2.046, p = 0.043). Stroke patients with the TNF-α rs361525 G/G genotype had higher serum IDO1 levels compared to those with the G/A genotype (Z = -2.451, p = 0.014).ConclusionsOur findings provided evidence that IDO1 gene polymorphisms and protein levels were involved in the development of early-onset PSD and TNF-α polymorphism was associated with IDO1 levels, supporting that IDO1 which underlie strongly regulation by cytokines may be a specific pathway for the involvement of immune-inflammatory mechanism in the pathophysiology of PSD.

Abstract Although the Co‐based Heusler compounds are predicted to be half‐metals, their sub‐picosecond demagnetization dynamics upon laser excitation show a transition‐metal‐like behavior. Any possible role of ultrafast nonlocal spin transport on the ultrafast demagnetization of half‐metallic Heusler compounds has only been inferred indirectly by time‐resolved magneto‐optical Kerr effect. Here, an ultrafast optically driven spin current traveling from Co2FeSi half metal into an adjacent Pt layer by using terahertz (THz) emission time‐domain spectroscopy (TDS) is demonstrated. By varying the magnetic field and excitation symmetry, the sizable THz generation from Co2FeSi/Pt stack arises from optically induced spin transport across the Co2FeSi/Pt interface in conjunction with spin‐to‐charge current conversion, via the inverse spin Hall effect (ISHE). The chemical ordering, interface roughness and magnetization of the samples become vital to engineer the THz emission properties. The amplitude of THz emission is comparable to that of CoFeB/Pt stack, thereby indicating Co2FeSi as an efficient ultrafast light‐driven spin current injector. Finally, by varying the pumping energy, the contributions are distinguished to the ultrafast spin current from thermalization and optical excitation of majority spins in Heusler alloy. This work illustrates THz emission TDS as a powerful tool to investigate the ultrafast spintronic properties of Heusler alloy/normal‐metal bilayers.

Objective The purpose of this study was to establish a nomogram predictive model of clinical risk factors for post-stroke depression (PSD). Patients and Methods We used the data of 202 stroke patients collected from Xuanwu Hospital from October 2018 to September 2020 as training data to develop a predictive model. Nineteen clinical factors were selected to evaluate their risk. Minimum absolute contraction and selection operator (LASSO, least absolute shrinkage and selection operator) regression were used to select the best patient attributes, and seven predictive factors with predictive ability were selected, and then multi-factor logistic regression analysis was carried out to determine six predictive factors and establish a nomogram prediction model. The C-index, calibration chart, and decision curve analyses were used to evaluate the predictive ability, accuracy, and clinical practicability of the prediction model. We then used the data of 156 stroke patients collected by Xiangya Hospital from June 2019 to September 2020 for external verification. Results The selected predictors including work style, number of children, time from onset to hospitalization, history of hyperlipidemia, stroke area, and the National Institutes of Health Stroke Scale (NIHSS) score. The model showed good prediction ability and a C index of 0.773 (95% confidence interval: [0.696–0.850]). It reached a high C-index value of 0.71 in bootstrap verification, and its C index was observed to be as high as 0.702 (95% confidence interval: [0.616–0.788]) in external verification. Decision curve analyses further showed that the nomogram of post-stroke depression has high clinical usefulness when the threshold probability was 6%. Conclusion This novel nomogram, which combines patients’ work style, number of children, time from onset to hospitalization, history of hyperlipidemia, stroke area, and NIHSS score, can help clinicians to assess the risk of depression in patients with acute stroke much earlier in the timeline of the disease, and to implement early intervention treatment so as to reduce the incidence of PSD.

Maternal folate deficiency increases the risk of neural tube defects (NTDs), but the mechanism remains unclear. Here, we established a mouse model of NTDs via low folate diets combined with MTX-induced conditions. We found that a significant increase in butyrate acid was observed in mouse NTDs brains. In addition, aberrant key crotonyl-CoA-producing enzymes acyl-CoA synthetase short-chain family member 2 (ACSS2) levels and lysine crotonylation (Kcr) were elevated high in corresponding low folate content maternal serum samples from mouse NTD model. Next, proteomic analysis revealed that folate deficiency led to global proteomic modulation, especially in key crotonyl-CoA-producing enzymes, and dramatic ultrastructural changes in mouse embryonic stem cells (mESCs). Furthermore, we determined that folate deficiency induced ACSS2 and Kcr in mESCs. Surprisingly, folic acid supplementation restored level of ACSS2 and Kcr. We also investigated overall protein post-translational Kcr under folate deficiency, revealing the key regulation of Kcr in glycolysis/gluconeogenesis, and the citric acid cycle. Our findings suggest folate deficiency leads to the occurrence of NTDs by altering ACSS2. Protein crotonylation may be the molecular basis for NTDs remodeling by folate deficiency.

BackgroundAcupuncture may become a treatment for postpartum depression (PPD). Currently, little is known about the use of acupuncture in the treatment of PPD from the point of view of practitioners. The aim of this study was to explore practitioners' perspectives on the treatment of PPD with acupuncture and provide suggestions for future improvement.MethodsThis study employed a qualitative descriptive method. Semistructured, open-ended interviews were conducted with 14 acupuncture practitioners from 7 hospitals via face-to-face or telephone interviews. The data were collected using interview outline from March to May 2022 and analysed using qualitative content analysis.ResultsIn general, the use of acupuncture for treating PPD was positively regarded by practitioners. They claimed that acupuncture is both safe and helpful for breastfeeding women who are experiencing emotional discomfort and that it can alleviate a variety of somatic symptoms. The following three themes were extracted: (a) patient acceptance and compliance; (b) acupuncture as a treatment for PPD; and (c) the advantages and drawbacks of acupuncture treatment.ConclusionPractitioners' optimistic outlooks demonstrated that acupuncture is a promising treatment option for PPD. However, the time cost was the most significant barrier to compliance. Future development will focus mostly on improving acupuncture equipment and the style of service.

Objective. Research evidence suggests a link between periodontitis (PD) and atrial fibrillation, but the nature of this link is unclear. This study aimed to systematically review and evaluate the association between PD, other oral diseases, and atrial fibrillation and the role of oral hygiene in preventing atrial fibrillation. Methods. We searched the Medline, Embase, Cochrane Library, and Web of Science databases for the clinical study of oral health and atrial fibrillation from inception to November 2022. Oral health conditions included PD and other oral inflammatory diseases, regular oral hygiene, and tooth brushing. The primary outcomes were the risk of new-onset atrial fibrillation in patients with oral disease, the effect of regular oral care on preventing atrial fibrillation, the effect of frequent tooth brushing on preventing atrial fibrillation, and the incidence of atrial fibrillation in PD patients. Results. Eight clinical trials with a total of 4,328,355 patients were included. The result of the research showed that PD and other impaired oral health may be associated with new-onset atrial fibrillation, and its severity was dose-responsive to the risk of atrial fibrillation. The incidence of atrial fibrillation in patients with severe PD was about 16.3%. Moreover, PD may increase the risk of long-term arrhythmia in patients with atrial fibrillation. Regular oral care and frequent tooth brushing can reduce the incidence of atrial fibrillation. Conclusion. Regular and moderate oral hygiene, frequent tooth brushing, and prevention of PD and other oral inflammatory diseases could reduce the occurrence of atrial fibrillation. It is recommended to strengthen the popularization of oral health knowledge in the publicity related to atrial fibrillation.

In order to investigate the influence of steel fiber content and concrete strength on the flexural toughness of ultra-high performance concrete with coarse aggregate (CA-UHPC), unnotched and notched beams were prepared for flexural tests. Three groups with water-binder ratios of 0.15，0.17, 0.19 were investigated, and the fiber contents of each group were 1.5%, 2.0%, 2.5%, and 3.0%, respectively. The JG/T-472–2015 and CECS13 methods were used to evaluate the test results. The results showed that when the fiber content is from 1.5% to 2%, the toughness efficiency is the highest. Pearson correlation coefficients was an effect way to evaluate the relationships between compressive strength and equivalent flexural strength, peak load and peak deflection of specimens. The fracture energy of the specimens increased linearly with the steel fiber contents. With the increasing of flexural strength, the fracture energy showed a trend of increase before decrease，and the fracture energy was the largest when the flexural strength was 24.1 MPa. Furthermore, according to the experimental fact, a new approach was proposed to modify the CECS13 method. By comparison, the modified method has better predictive validity.

Haematopoietic necrosis in Carassius auratus is one of the most serious diseases in recent years caused by CyHV-2 (cyprinid herpesvirus 2) which has caused huge economic losses in aquaculture. Currently, the isolation and propagation of CyHV-2 in vitro is difficult due to the lack of sensitive cell lines, which hinder further studies on the pathogenesis and gene functions of the virus. In this study, Carassius carassius gill cell line (CCG) was constructed and had been passaged up to 90 passages at 25 ℃ in Medium199 (M199) with 15–20% Foetal bovine serum (FBS). Cellular karyotype analysis showed that the chromosome number was 46 at passage 50. CCG could be successfully transfected with GFP reporter gene with an efficiency of more than 60% suggesting that CCG could be used for exogenous gene expression in vitro. CCG infected with CyHV-2 displayed typical cytopathic effects (CPE), and viral replication was confirmed by PCR, indirect immunofluorescence assay (IFA) and transmission electron microscopy (TEM) assay, respectively. In conclusion, the CCG cell line can be used as a useful tool to study the propagation and pathogenesis of CyHV-2.

ObjectiveThis systematic review and meta-analysis compares the outcome between MMBC and unifocal breast cancer (UFBC), in order to provide a theoretical basis for the design of an appropriate clinical therapeutic strategy of MMBC patients.MethodsPubMed, Embase, The Cochrane Library, Web of science, CNKI, WanFang Data, CBM and VIP database were searched from inception to July 2021, and observational studies reporting the outcome of patients with MMBC and UFBC were included. We extracted or calculated the mortality rates of MMBC and UFBC patients; and obtained the hazard ratios; odds ratios; relative risks; and the corresponding 95% confidence intervals from the eligible studies. All the meta-analyses were conducted by using the Stata 15.0 software.Results31 eligible studies comprising a total of 15,703 individuals were included. The meta-analysis revealed that MMBC did not have a significant association with poor overall survival (HR=1.04, 95% CI=0.96-1.12), disease-free survival (HR= 1.07, 95% CI= 0.84-1.36), breast cancer-specific survival (HR=1.42, 95% CI= 0.89-2.27), recurrence-free survival (HR= 0.878, 95% CI= 0.652-1.182), local recurrence-free survival (HR= 0.90, 95% CI= 0.57-1.42), and contralateral breast cancer risk (RR= 0.908, 95% CI= 0.667-1.234). However, MMBC appeared to have a correlation with a slightly higher risk of death (OR=1.31, 95% CI=1.18-1.45).ConclusionPatients with MMBC appeared to have a higher risk of death, however, it may not be independently associated with poorer outcomes. Considering the inter-study heterogeneity and other limitations, our results need to be validated by further multicenter prospective studies with a large sample size in the future.

BackgroundHomocysteine (Hcy) has been indicated to be involved in pathophysiology of post stroke depression (PSD). There is a lack of research to study the relationship between Hcy metabolism genes and PSD. Our study aims to investigate the relationship among Hcy metabolism genes, Hcy, and early-onset PSD.Materials and methodsWe recruited 212 patients with stroke and collected their peripheral blood sample, clinical data, and laboratory test on admission. 12 single nucleotide polymorphisms (SNPs) in methylenetetrahydrofolate reductase (MTHFR), methionine synthase reductase (MTRR), and methionine synthase (MTR) genes were genotyped by high-resolution melt analysis. PSD was diagnosed by DSM-V at 2 weeks after stroke. Binary logistic regression and haplotype analysis were used to examine the association between Hcy metabolism genes and PSD. Mediation analysis was performed to clarify whether the SNPs exerted their effect on PSD by affecting the Hcy level.Results81 patients were diagnosed with PSD, and the incidence rate was 38.2%. Hcy level in PSD group was significantly higher than it in non-PSD group (p = 0.019). MTHFR rs1801133 AA genotype an A allele were associated with an elevated risk of PSD after adjustment for some confounding factors (OR = 4.021, 95% CI: 1.459∼11.080, p = 0.007 for AA genotype; OR = 1.808, 95% CI: 1.172∼2.788, p = 0.007 for A allele). Furthermore, the effect of MTHFR rs1801133 AA genotype on PSD was mediated by Hcy (OR = 1.569, 95% CI: 0.013∼3.350, p < 0.05).ConclusionMTHFR rs1801133 and Hcy were associated with PSD, and MTHFR rs1801133 may exert an effect on PSD via mediating Hcy level. This offers a new perspective for treating PSD and understanding the mechanism of PSD.

This paper is dedicated to a thorough analysis of how the transition to automatic driving in train operations influences the real-time rescheduling model. Based on the distinctive impact of the ATO system on real-time rescheduling, we have proposed a mixed-integer linear programming model that combines train re-timing, reordering, and the minimization of passenger delays. To validate the effectiveness of our model, we present several experiments conducted using data from the Beijing–Shanghai high-speed railway line. The results unequivocally demonstrate that our ATO-based model significantly mitigates train delay time, demonstrating its practical value in optimizing high-speed railway operations.

Ammonium toxicity in macrophytes reduces growth and development due to a disrupted metabolism and high carbon requirements for internal ammonium detoxification. To provide more molecular support for ammonium detoxification in the above-ground and below-ground parts of Myriophyllum spicatum, we separated (using hermetic bags) the aqueous medium surrounding the below-ground from that surrounding the above-ground and explored the genes in these two regions. The results showed an upregulation of asparagine synthetase genes under high ammonium concentrations. Furthermore, the transcriptional down and/or upregulation of other genes involved in nitrogen metabolism, including glutamate dehydrogenase, ammonium transporter, and aspartate aminotransferase in above-ground and below-ground parts were crucial for ammonium homeostasis under high ammonium concentrations. The results suggest that, apart from the primary pathway and alternative pathway, the asparagine metabolic pathway plays a crucial role in ammonium detoxification in macrophytes. Therefore, the complex genetic regulatory network in M. spicatum contributes to its ammonium tolerance, and the above-ground part is the most important in ammonium detoxification. Nevertheless, there is a need to incorporate an open-field experimental setup for a conclusive picture of nitrogen dynamics, toxicity, and the molecular response of M. spicatum in the natural environment.

The RNA-binding glycine-rich proteins (RBGs) of the glycine-rich protein family play vital roles in regulating gene expression both at the transcriptional and post-transcriptional levels. However, the members and functions in response to abiotic stresses of the RBG gene family remain unclear in Brassica oleracea. In this study, a total of 19 BoiRBG genes were identified through genome-wide analysis in broccoli. The characteristics of BoiRBG sequences and their evolution were examined. An analysis of synteny indicated that the expansion of the BoiRBG gene family was primarily driven by whole-genome duplication and tandem duplication events. The BoiRBG expression patterns revealed that these genes are involved in reaction to diverse abiotic stress conditions (i.e., simulated drought, salinity, heat, cold, and abscisic acid) and different organs. In the present research, the up-regulation of BoiRBGA13 expression was observed when subjected to both NaCl-induced and cold stress conditions in broccoli. Moreover, the overexpression of BoiRBGA13 resulted in a noteworthy reduction in taproot lengths under NaCl stress, as well as the inhibition of seed germination under cold stress in broccoli, indicating that RBGs play different roles under various stresses. This study provides insights into the evolution and functions of BoiRBG genes in Brassica oleracea and other Brassicaceae family plants.

With the development of deepfake technology, deepfake detection has received widespread attention. Although some deepfake forensics techniques have been proposed, they are still very difficult to implement in real-world scenarios. This is due to the differences in different deepfake technologies and the compression or editing of videos during the propagation process. Considering the issue of sample imbalance with few-shot scenarios in deepfake detection, we propose a multi-feature channel domain-weighted framework based on meta-learning (MCW). In order to obtain outstanding detection performance of a cross-database, the proposed framework improves a meta-learning network in two ways: it enhances the model’s feature extraction ability for detecting targets by combining the RGB domain and frequency domain information of the image and enhances the model’s generalization ability for detecting targets by assigning meta weights to channels on the feature map. The proposed MCW framework solves the problems of poor detection performance and insufficient data compression resistance of the algorithm for samples generated by unknown algorithms. The experiment was set in a zero-shot scenario and few-shot scenario, simulating the deepfake detection environment in real situations. We selected nine detection algorithms as comparative algorithms. The experimental results show that the MCW framework outperforms other algorithms in cross-algorithm detection and cross-dataset detection. The MCW framework demonstrates its ability to generalize and resist compression with low-quality training images and across different generation algorithm scenarios, and it has better fine-tuning potential in few-shot learning scenarios.

Optimizing Cropping patterns is important for the improvement of regional agricultural economic efficiency and sustainable development. However, there are few studies on the sustainability of cropping patterns in hilly areas. Here, we studied four new three-maturing cropping patterns in a typical ecological site in the hilly areas of southwest China. An analytical method combining economic efficiency evaluation and energy value analysis was used to evaluate and compare the economic efficiency and sustainability of the new cropping model and the traditional cropping model. We explored the construction of a new three-crop cropping model suitable for the southwest hilly area to improve the economic benefits of agricultural production and improve the sustainability of agricultural production. To solve the above problems, we constructed eight cropping patterns and classified them as follows: The Traditional Double Cropping System: T1, oilseed rape-summer soybean; T2, oilseed rape-summer maize; T3, wheat-summer maize. Traditional Triple Cropping System: T4, wheat/spring maize/summer soybean. Novel Triple Cropping System: T5, forage oilseed rape-spring maize/summer soybean; T6, forage oilseed rape-spring maize/peanut; T7, potato-spring maize/peanut; T8, potato-spring maize/summer soybean. The results of the study showed that compared with the Traditional Double Cropping System and the Traditional Triple Cropping System, the Novel Triple Cropping System increased the economic yield by an average of 100.39% and 49.18%, the economic production capacity by 71.32% and 36.48%, the biological yield by 12.53% and 4.90%, and the biological production capacity by 13.59% and 5.80%. The economic benefits of the Novel Triple Cropping System were significantly improved, with economic profits increased by CNY 9068 ·hm−2 and CNY 7533 ·hm−2 compared with the Traditional Double Cropping System and the Traditional Triple Cropping System. The energy value analysis further revealed the characteristics of the Novel Triple Cropping System as a high input and high output model. The Novel Triple Cropping System increased energy value inputs by 6.56% and 4.25%, and energy value outputs by 13.69% and 4.27% compared with the Traditional Double Cropping System and the Traditional Triple Cropping System, respectively. This high level of inputs stems mainly from a significant increase in labor inputs. Meanwhile, the energy-value indicator analysis of the Novel Triple Cropping System shows its lower dependence on natural resources, greater production intensification, and increased system stability. As a result, the Novel Triple Cropping System showed higher sustainable production capacity. In summary, the results of this study can provide a theoretical basis for optimizing cropping patterns and promoting high-yield and the sustainable development of agriculture.

Asphalt mixtures will inevitably be affected by rainwater and the effect of the wet temperature cycle during a pavement’s life span. Especially in coastal areas such as Guangdong and Hainan in China, asphalt pavement is particularly susceptible to the sizeable wet temperature cycle formed by the high temperature and sudden temperature drop of rainstorms in summer. For this study, we used a homemade sizeable wet temperature cycle environment simulation device to analyze the decay characteristics and the mechanical properties of asphalt pavements in this environment; modified bending and tensile strength and shear strength tests were used to study the decay patterns of shear strength, bending, and tensile strength, and the stiffness modulus of asphalt mixtures with different air voids and different pavement depths under the action of a sizeable wet temperature cycle. In addition, the Grey correlation method was used to analyze the significance of each influencing factor on the decay of mechanical properties, and mathematical fitting was used to establish the prediction equation of the mechanical properties of asphalt mixtures. The results show that with the increase in the number of sizeable wet temperature cycles, the asphalt mixture’s shear strength, flexural tensile strength, and flexural tensile modulus decrease, the degree of decay increases, and the rate of decay gradually slows down. In the case of the same number of sizeable wet temperature cycles, the degree of decay of the asphalt mixtures gradually decreases with increasing depth or decreasing void ratio. After 100 sizeable wet temperature cycles, the maximum values of the decay of shear strength, modulus of strength, and flexural tensile strength were 22.30%, 23.29%, and 32.01%. The importance of the influence of each factor on the decay of the mechanical properties is as follows: the number of sizeable wet temperature cycles > void rate > depth. The prediction equations of the established mechanical properties have a good prediction effect, and the correlation between predicted values and actual values can be up to 0.925. The prediction equations can effectively predict the mechanical properties of asphalt mixtures with different air voids and depths under the action of sizeable wet temperature cycles.

The liver is the primary organ regulating glucose metabolism. In our recent study, cinnamon improved liver function in diabetic mice. However, it is not clear whether cinnamon can reduce the glycemia of diabetic animals by regulating liver glucose metabolism. The purpose of this study was to investigate the hypoglycemic mechanism of cinnamon powder (CP) from the perspective of regulating liver glucose metabolism. To achieve this, different doses of CP (200, 400, or 800 mg/kg body weight) were given to diabetic mice by gavage once per day for 8 weeks. These mice were compared with healthy controls, untreated diabetic mice, and diabetic mice treated with metformin (the main first-line drug for type 2 diabetes). CP treatment effectively reduced fasting blood glucose levels and food intake, improved glucose tolerance and fasting serum insulin levels, and decreased glycated serum protein levels in diabetic mice. Furthermore, treatment with CP increased liver glycogen content and reduced the level of the gluconeogenesis precursor pyruvate in the liver. Data obtained by qPCR and western blotting suggested that CP improved glucose metabolism disorders by regulating AMPKα/PGC1α-mediated hepatic gluconeogenesis and PI3K/AKT-mediated hepatic glycogen synthesis. CP exhibits good hypoglycemic effects by improving hepatic glycogen synthesis and controlling hepatic gluconeogenesis. Therefore, CP may be applied as a functional food to decrease blood glucose.

Mycoplasma bovis is a global concern for the cattle industry owing to its high rates of infection and resulting morbidity, but its pathogenesis remains poorly understood. Metabolic pathways and characteristics of M. bovis clinical strain were elucidated by comparing the differential expression of metabolites between M. bovis clinical strain NX114 and M. bovis international reference strain PG45. Metabolites of M. bovis in the logarithmic stage were analyzed based on the non-targeted metabolomic technology of ultra-high performance liquid chromatography-mass spectrometry (UHPLC-MS). We found 596 metabolites with variable expression, of which, 190 had substantial differences. Differential metabolite analysis of M. bovis NX114 showed organic acids and their derivatives, nucleosides, and nucleotide analogs as important components. We found O-Phospho-L-serine (SEP) as a potential signature metabolite and indicator of pathogenicity. The difference in nucleic acid metabolites reflects the difference in growth phenotypes between both strains of M. bovis. According to KEGG enrichment analysis, the ABC transporter synthesis route had the most differential metabolites of the first 15 differential enrichment pathways. This study reflects the species-specific differences between two strains of M. bovis and further enriches our understanding of its metabolism, paving the way for further research into its pathogenesis.

Aging is a biological process that leads to the progressive deterioration and loss of physiological functions in the human body and results in an increase in morbidity and mortality, and aging-related disease is a major global problem that poses a serious threat to public health. Polysaccharides have been shown to delay aging by reducing oxidative damage, suppressing inflammatory responses, and modulating intestinal microbiota. Our previous studies have shown that polysaccharide CPP-1 extracted from the root of Codonopsis pilosula possesses noticeable anti-oxidant activity in vitro. Thus, in our study, we tested the anti-aging effect of CPP-1 in naturally aging mice (in vivo). Eighteen C57/BL mice (48-week-old, male) were divided into a control group, high-dose CPP-1 group (20 mg/mL), and low-dose CPP-1 group (10 mg/mL). We discovered that CPP-1 can exert a reparative effect on aging stress in the intestine and liver, including alleviating inflammation and oxidative damage. We revealed that CPP-1 supplementation improved the intestinal microbiota composition and repaired the intestinal barrier in the gut. Furthermore, CPP-1 was proved to modulate lipid metabolism and repair hepatocyte injury in the liver by influencing the enterohepatic axis associated with the intestinal microbiota. Therefore, we concluded that CPP-1 prevents and alleviates oxidative stress and inflammatory responses in the intestine and liver of aging mice by modulating the intestinal microbiota-related gut–liver axis to delay aging.

To solve the problems of long experiment period and difficult measurement in core imbibition experiments, fracture—matrix microfluidic chips of different sizes, boundary conditions and wettability regulated by surface property modification were designed to research the imbibition mechanisms of oil-water, oil-surfactant solution and oil-Winsor III type surfactant solution. In the oil-water, and oil-wettability modification system imbibition process, oil was replaced from the matrix through Haines jump, the capillary back pressure was the main resistance blocking the flow of oil, the reduction of interfacial tension caused the weakening of Haines jump, reduction of oil discharge rate, and increase of oil recovery. The imbibition of oil-water or oil-surfactant solution with low interfacial tension was a counter- current imbibition process dominated by capillary force, in which all boundaries had similar contribution to imbibition, and the recovery data obtained from this experiment fit well with the classic imbibition scaling equation. The imbibition of oil and Winsor III type surfactant solution was a co-current imbibition process dominated by gravity under super-low interfacial tension, and is essentially the formation and re-balance of neutral microemulsion. The imbibition dynamics obtained from this experiment fit well with the modified imbibition scaling equation.

Pear (Pyrus spp.) is the third-largest economic crop in China after apples (Malus pumila Mill.) and citrus (Citrus reticulata Blanco), and it is mainly cultivated by smallholders. Currently, the yield of Chinese pear ranks midlevel globally, with only 17.9 t⋅ha−1⋅year−1, which is lower than that of the United States (36.0 t⋅ha−1⋅year−1). However, the factors limiting pear production dominated by smallholders are unclear. We interviewed 75 smallholders about 18 yield-related indicators for pear-typical planting areas. The boundary line model was used to analyze the contribution of internal factors and dominant external factors affecting yield and to simulate strategies for increasing yield through the scenario analysis. The results showed that the average gap between the average and highest attainable yields for smallholders was 10.5 t⋅ha−1⋅year−1 in Luniao County. Among individual yield-limiting factors, chemical fertilizer nitrogen (N) input (13.3%) was the most significant, followed by the soil-available N content (12.0%) and leaf magnesium content (12.0%). Overall, the contribution of all soil factors (42.7%) was the largest compared with the other factor categories. However, the contribution of internal factors could not be ignored and accounted for 25.3% of the total. A scenario analysis showed that comprehensive strategies considering soil, management, and internal factors achieved the largest yield improvement (14%), as did reducing the fertilizer application rate (66%) compared with only using soil or leaf diagnosis methods. Therefore, integrated methods should be considered when developing pear orchard management measures and include soil, management, and internal factors.

As patients are dissatisfied with the excessive weight of Co-based knee femoral components, it is an urgent need to reduce their weight without compromising the excellent mechanical properties. In the present study, femoral components of knee with Ti6Al4V alloy (86 g) or Co–Cr alloy (192 g) were printed by selective laser melting (SLM). In order to improve the mechanical properties and biocompatibility of 3D-printed Ti6Al4V, TiN and TiCrN coating layers were deposited on the surface of SLM Ti6Al4V using arc ion plating (AIP) system. The surface characterizations were analyzed by scanning electron microscopy (SEM) equipped with energy dispersive spectrometer (EDS), X-ray diffraction (XRD), contact angle tensiometer, and contact profilometer. The wear property, hardness value and corrosion resistance in phosphate buffered saline (PBS) were also investigated through vickers hardness, sliding wear test, electrochemical impedance spectroscopy and potentiodynamic polarization. The antibacterial capability and biocompatibility were assessed with Staphylococcus aureus and mesenchymal stem cells (MSCs) in vitro. Results illustrated that, after coating SLM Ti6Al4V with thin TiN and TiCrN layers, the hardness, anti-wear performance and corrosion resistance of SLM Ti6Al4V were significantly improved and became comparable to SLM CoCr. The TiN and TiCrN coated SLM Ti6Al4V also showed moderate antibacterial ability, and excellent biocompatibility in comparation with SLM Ti6Al4V and SLM CoCr.
