Abstract

To explore the association between polymorphisms in microRNAs (miRNAs) and the cholinesterase (ChE) activity in omethoate-exposed workers, we recruited 180 omethoate-exposed workers and 115 controls to measure their ChE activity using acetylcholine and dithio-bis-(nitrobenzoic acid) and genotype susceptible SNPs in their miRNA by time-of-flight mass spectrometry. ChE activity in the exposure group was lower than that in the control group (P < 0.001). The analysis of covariance result showed that ChE activity was lower in the (- -/- T) genotype in miR-30a rs111456995 (1.97 ± 0.47) than in the TT genotype (2.23 ± 0.59) of the exposure group (P = 0.004). Multivariate linear regression was performed to find influencing factors on ChE activity, and variables kept in the model included omethoate exposure (b = −1.094, P < 0.001), gender (b = −0.381, P < 0.001), miR-30a rs111456995 (- -/- T)(b = −0.248, P < 0.001), and drinking (b = 0.258, P =0.019). The results suggest that individuals carrying a (- -/- T) genotype in miR-30a rs111456995 were more susceptible to damage in their cholinesterase induced by omethoate exposure. Abstract

Computer Fluid Dynamics (CFD) is a popular method for studying airflow of nasal cavities. However, the data of CFD studies has rarely been validated through experiments. To test the accuracy of CFD computation, we studied the consistency of the air pressure of nasal cavities in the CFD and the experiment. A proportional resin model of a normal human subject’s nasal cavities was created by a 3-d printer with a precision value of 0.1mm. The pressure of 63 check points in the nasal cavities in different breathing states was measured. The experimental data was compared with the data obtained by CFD simulation. At the flow rates of 180 ml s-1 and 560 ml s-1, the pressure in all check points remained highly consistent with the CFD data. At 1100 ml s-1 flow rate, there was a significant deviation in the posterior segment of the nasal cavity during exhalation. The method used in this study to measure the pressure in the nasal cavities can be used in experimental validation of CFD data. The computational methods and the boundary conditions used in this study resulted in a high agreement between the results of the CFD simulation and the experiment.Author SummaryIn the contemporary era, Computer fluid dynamics (CFD) is the mainstream method for studying air flow. Due to the complex anatomical structure of the nasal cavity, the CFD results of the nasal flow have rarely been experimentally verified. This study provides a method to verify the methods and results of nasal CFD. We printed an accurate model of a normal person’s nasal cavity with a high-precision 3D printer. In this nasal cavity model, we set 63 small holes to detect the air pressure of the places we concerned. Three different nasal flow quantity are used to represent different breathing conditions: high (1100 ml s-1), medium (560 ml s-1), and low (180 ml s-1). In medium and low nasal flow quantities, our CFD results are in good agreement with the experimental pressure values. On this basis, we analyzed the characteristics of nasal airflow in normal people. The method used in this study to measure the pressure in the nasal cavities can be used in experimental measurements of the partial resistance of the nasal cavity. With proper modification, it can be applied to the clinical practice for nasal resistance, giving more help for the design of the operation plan.

Abstract

The traditional wave motion characteristic experiment has some problems, such as the loose connection between theory and experimental teaching, limitations of the experiment due to its site and time, and poor sense of participation and interaction. Focusing on the above problems, a virtual simulation experiment system for wave motion characteristics is designed and developed. The system uses virtual reality technology, cloud platform technology, and wave simulation technology. Taking the real laboratory as the prototype, the scene structure, instrument function, and water movement are simulated with high precision. The multimode fusion teaching method has expanded the experimental content, enriched the micro‐observation function and data collection categories, enhanced the interaction of the experimental process, and created a high‐quality immersive online teaching experience for the students. Since its inception, the experiment system has served six cycles of instruction and obtained good feedback from the teachers and students. During the period of the pandemic, it provides great support to experimental teaching and serves as a good supplement to the existing teaching system. Adhering to the "student‐centered" teaching principle, the teaching team will improve the experimental system constantly, cultivate the students' independent thinking and innovation abilities, help the students with their own personalized development, and contribute to the training of excellent engineers. Abstract

The traditional medicine-Eupolyphaga sinensis walker is a rich source of functional proteins and peptides. In this study, Eupolyphaga sinensis walker were identified to include fifteen amino acids, of which hydrophobic amino acids accounted for 46.7%. Eupolyphaga sinensis walker polypeptides (EPs) were extracted by sequentially hydrolyzing with pepsin and trypsin. EPs could effectively scavenge 1,1-diphenyl-2-picrylhydrazyl radical (DPPH·), superoxide anion radical (O2−·), and hydroxyl radical (OH-·), and reduce ferric solution in vitro. EPs also enhanced the activities of Ssuperoxide dismutase (SOD), catalase (CAT) and glutathione peroxidase (GPH-Px), increased the contents of hydroxyproline (HYP), and reduced the content of Malondialdehyde (MDA) in photoaged mice skin. Histological study confirmed that EPs improved UV irradiation-induced damage of skin texture and morphology. Therefore, the extracted EPs are effective antioxidants and can serve as powerful treatment for skin photoaging.

Abstract

• We determined the profile of differently expressed genes between endometrial cancer (EC) and normal tissues using TCGA RNA-seq data. • We performed comprehensive bioinformatical analysis and identified the most enriched GO terms, KEGG pathways, hub genes in the protein-protein interaction network and their association with the mortality of EC patients. • AURKA, CENPA and KIF2C were found as potential biomarkers for endometrial cancer with significant prognostic effect. The molecular basis of the mechanism and the potential biomarkers of endometrial cancer (EC) remain to be studied. In the present study, we hypothesized that the comprehensive characterization of transcriptional changes in EC could help achieve this aim. By taking advantage of RNA-seq data from The Cancer Genome Atlas, we determined the profile of differently expressed genes (DEGs) between EC tumor tissues and normal samples. On this basis, we performed Gene Ontology and Kyoto Encyclopedia of Genes and Genomes pathways enrichment analyses. The interacting partners for each of the DEGs were explored and a protein-protein interaction network was constructed. Consequently, 10 hub genes were identified and their association with mortality in EC patients was investigated. The genes, AURKA, CENPA , and KIF2C , were found to be potential biomarkers for EC with a significant prognostic effect. Abstract

Landslide dams often exhibit non-uniform structures due to the rapid and dynamic accumulation of various types of landslide materials. These variations in dam composition significantly impact breach progression, causing significant errors in current model tests and numerical simulations with uniform structure assumptions This adversely affects peak flow predictions and decisions related to risk prevention and control. This study conducted a series of flume tests to examine the influence of non-uniform structures on the failure mechanisms of landslide dams. A 3D dam breach model was used to replicate these flume experiments and obtain more detailed information. Large Eddy Simulation (LES) captures the hydraulic features of dam-break flow, while equilibrium equations model morphological changes in landslide dams amid complex terrains using suspended and bed load transport. The findings indicate that in a uniform dam, the presence of coarse particles shifts erosion characteristics from layered erosion to step-pool development. Furthermore, as the content of coarse particles increases, the number of steps also rises. In the vertically non-uniform structure, the upper layer affects the formation of the breach, the middle layer affects the dam break and the breach flow, and the bottom layer affects the remaining dam height. The presence of an armor layer results in a higher water level and accelerates breach formation with a larger breach flow. In the horizontally non-uniform structure, the upper part of the overflow side is the core area of the breach, the lower part of the overflow side and the upper part of the opposite side affect the downcutting and widening rates of the breach, respectively. Preferentially, the breach tends to develop towards the relatively weaker of the two sides. • The failure modes of vertically and horizontally non-uniform structural dams were systematically examined. • The pivotal impact of coarse particles on the erosion characteristics of landslide dams was investigated. • Explored the role of different regions during the breaking process and breach flow, discussing their combined effects. • The effectiveness of the new numerical model in simulating the effects of non-uniform structures were validated. Abstract

Aims: Multi-elemental stoichiometry can represent the biogeochemical niches of species, which can further guide community assemblage. Mycorrhizae play a key role in plant elemental acquisition. Whether and how mycorrhizal type influences the plant multi-elemental stoichiometries and elemental coordination in above- and belowground organs, however, remain unclear. Methods: We determined the concentrations of C, N, P, K, Ca, Mg, Fe, Mn, Cu, Zn, Na, Pb and Sr and some elemental ratios in the leaves and roots of arbuscular mycorrhizal (AM) trees (Phoebe bournei, Schima superba, Michelia maudiae and Cunninghamia lanceolata) and ectomycorrhizal (ECM) trees (Cyclobalanopsis glauca, Pinus massoniana and P. elliottii) in subtropical China. The effects of root traits and soil abiotic factors on elemental concentrations and ratios were reported. Results: The elemental concentrations and ratios differed substantially between the AM and ECM trees, indicating the their biogeochemical niches' differentiation. The C, P, Mg and Mn concentrations and the K:Mg andCa:Mg ratios for AM and the N, Mn and Na concentrations and the Fe:Mn ratio for the ECM species were strongly correlated between leaves and roots, but the correlations were positive or negative depending on elemental composition and mycorrhizal type. The root branching ratio, branching intensity and length played important roles in the plant elemental stoichiometries. Conclusions: The biogeochemical niche's differentiation between the AM and ECM species emphasizes the important role of mycorrhizal type in the assemblage of co-occurring species. This study offers a new opinion for species choosing in mixed forests from the view of the biogeochemical niche's separation. [ABSTRACT FROM AUTHOR]
