151. New mass measurements at the neutron drip-line.

Abstract

A new SPEG mass measurement experiment has been performed to determine masses closer to the neutron drip-line in the mass region A ∼ 10-50. The precision of 37 masses has been improved and 8 masses were measured for the first time. The region covered was motivated by the study of shell structure and of shape coexistence in the region of closed shells N = 20 and N = 28. The evolution of the two neutron separation energies and the shell correction energy have been studied as a function of the neutron number. The results thus obtained provide a means of identifying, in exotic nuclei, new nuclear structure effects that are well illustrated by the changes of the conventional magic structure. Abstract

Various aspects of the application of shell model approaches to cluster transfer reactions involving exotic nuclei are discussed. A scheme to calculate Spectroscopic Amplitudes for transfer reactions is given. It is calculated for a tetraneutron in 8He as an example. Abstract: Around 270 medium-mass residual nuclei, formed in spallation reactions induced by 136Xe projectiles impinging on a liquid hydrogen target at 500 A MeV, have been unambiguously identified at GSI using the magnetic spectrometer FRS. The individual production cross sections and the longitudinal momentum distributions have been determined with high accuracy. Abstract

We investigated deep inelastic reactions in 238U + 238U collisions at 6.09, 6.49, 6.91, 7.1 and 7.35× A MeV at the VAMOS spectrometer (GANIL). A large transfer of neutrons and protons was observed at all beam energies. For a transfer of more than 10 nucleons the total kinetic energy of the detected fragments becomes independent of the beam energy and reaches values far below the Coulomb barrier for spherical fragments. This points to the formation of a di-nuclear system in the entrance channel which develops an elongated shape and a strong neck. For such reactions we expect an enhanced lifetime of the di-nuclear system which is significantly longer than the time scale for elastic and quasi-elastic reactions. Different theoretical approaches predict delay times of more than 5×10-21 s for a subset of our data. [ABSTRACT FROM AUTHOR]

Abstract

We searched for a long-living component in the collision of 238U+238U between 6.09 A and 7.35 A MeV. The experiment was performed at GANIL using the spectrometer VAMOS, tuned for observing reactions with kinematics similar to quasi-fission events. Theoretical calculations indicate that reactions with strong energy dissipation and a large number of transferred nucleons are correlated to a time delay in the decay of the giant system. We detected events of such type in the focal plane of VAMOS. These events present an excitation function increasing with bombarding energy. [ABSTRACT FROM AUTHOR]

Abstract

We have studied an effect of neutron and triton transfer reactions on the p +^6He elasticscatteringat25 MeV bymeansofcoupled - reaction - channelcalculations. Itisfoundthatwhenthetransferreactionsareexplicitlyincludedinthecalculationstheimaginarypartoftheinput$ p + $6He optical model potential has to be reduced by 52 percent while its real part enhanced by 15 percent in order to fit the elastic-scattering data. The effect of transfer channels on the real part of this potential is somewhat weaker than that of 6He breakup reported previously. However, for the imaginary part, the effect of transfer channels is dominant. It is concluded that while the breakup contribution to proton elastic scattering mainly affects the real part of the bare potential, the contribution of transfer channels affects mainly its imaginary part. Abstract

A fission-fragment-sensitive detector built for low-energy photon spectroscopy applications at the WNR “white” neutron source at Los Alamos is described. The detector consists of eight layers of thin photovoltaic cells, onto which 1 mg/cm2 of pure 238U is deposited. The detector serves as an active target to select fission events from background and other reaction channels. The fairly small thickness of the detector with respect to transmission of 20–50 keV photons permits the measurement of prompt fission-fragment X-rays. Abstract

This paper attempts to summarize the actual problematic of reactor antineutrino energy spectra in the frame of fundamental and applied neutrino physics. Nuclear physics is an important ingredient of reactor antineutrino experiments. These experiments are motivated by neutrino oscillations, i.e. the measure of the θ13 mixing angle. In 2011, after a new computation of the reactor antineutrino energy spectra, based on the conversion of integral data of the beta spectra from 235U, and 239;241Pu, a deficit of reactor antineutrinos measured by short baseline experiments was pointed out. This is called the "reactor anomaly", a new puzzle in the neutrino physics area. Since then, numerous new experimental neutrino projects have emerged. In parallel, computations of the antineutrino spectra independant from the ILL data would be desirable. One possibility is the use of the summation method, summing all the contributions of the fission product beta decay branches that can be found in nuclear databases. Studies have shown that in order to obtain reliable summation antineutrino energy spectra, new nuclear physics measurements of selected fission product beta decay properties are required. In these proceedings, we will present the computation methods of reactor antineutrino energy spectra and the impact of recent beta decay measurements on summation method spectra. The link of these nuclear physics studies with short baseline line oscillation search will be drawn and new neutrino physics projects at research reactors will be briefly presented. Abstract

The aim of this work is to study the impact of the inclusion of the recently measured β decay properties of the 102,104,105,106,107Tc, 105Mo, and 101Nb nuclei in the calculation of the antineutrino (anti-ν) energy spectra arising after the fissions of the four main fissile isotopes 235,238U, and 239,241Pu in PWRs. These β feeding probabilities, measured using the Total Absorption Technique (TAS) at the JYFL facility of Jyväskylä, have been found to play a major role in the &#947 component of the decay heat for 239Pu in the 4-3000 s range. Following the fission product summation method, the calculation was performed using the MCNP Utility Reactor Evolution code (MURE) coupled to the experimental spectra built from β decay properties of the fission products taken from evaluated databases. These latest TAS data are found to have a significant effect on the Pu isotope energy spectra and on the spectrum of 238U showing the importance of their measurement for a better assessment of the reactor anti-ν energy spectrum, as well as importance for fundamental neutrino physics experiments and neutrino applied physics. Abstract

Total cross sections for proton- and deuteron-induced-fission of 208Pb and 238U have been determined in the energy range between 500 MeV and 1 GeV. The experiment has been performed in inverse kinematics at GSI Darmstadt, facilitating the counting of the projectiles and the identification of the reaction products. High precision between 5% and 7% has been achieved by individually counting the beam particles and by registering both fission fragments in coincidence with high efficiency and full Z resolution. Fission was clearly distinguished from other reaction channels. The results were found to deviate by up to 30% from Prokofiev's systematics on total fission cross sections. Abstract

The structure of the light exotic nucleus 8He was investigated using direct reactions of the 8He SPIRAL beam on a proton-rich target. The (p,p'') scattering to the state, the (p,d)7He and (p,t)6He transfer reactions, were measured at the energy . The light charged particles (p,d,t) were detected in the MUST Si-strip telescope array. The excitation spectrum of 8He was extracted from the (p,p'') reaction. Above the known excited state at 3.6 MeV, a second resonance was found around 5.4 MeV. The cross sections were analyzed within the coupled-reaction channels framework, using microscopic potentials. It is inferred that the 8He ground state has a more complex neutron-skin structure than suggested by previous models assuming a pure configuration. [Copyright &y& Elsevier]

Abstract: The recent improvements concerning the production of exotic beams in facilities such as SPIRAL at GANIL allow us to explore new areas of the nuclear chart, away from the line of stability. However, the intensities for these beams are still relatively low (few thousands of particles per second) and the cross-sections of the most of the reactions of interest are also low (of the order of milibarns, or lower). In order to increase the reaction rates with a thick target, without loss of resolution, we developed the MAYA detector, based on the active target concept. MAYA is essentially a Time-charge Projection chamber where the gas plays also the role of the target. The segmented cathode allows particle identification, three-dimensional tracking and complete reconstruction of kinematics of binary reactions with beam particles impinging on the gas inside the detector. The parameters of the chamber, such as gas type and pressure, electric field and amplification regime, are modifiable depending on the characteristics of the reaction of interest, allowing a low-energy detection threshold, and an optimum thickness. Abstract

Abstract: In two experiments performed with 20–30MeV/u highly charged heavy ions (Pb56+, U91+) channeled through thin silicon crystals, we observed the original features of superdensity, associated to the glancing collisions with atomic rows undergone by part of the incident projectiles. In particular, the very high collision rate yields a quite specific charge exchange regime, that leads to a higher ionization probability than in random conditions. X-ray measurements show that electrons captured in outer shells are prevented from being stabilized, which enhances the lifetime of the projectile inner shell vacancies. The charge state distributions and the energy loss spectra are compared to Monte-Carlo simulations. These simulations confirm, extend and illustrate the qualitative analysis of the experimental results. [Copyright &y& Elsevier]

The precision of 37 masses has been improved and 8 masses were measured for the first time. The region covered was motivated by the study of shell structure and of shape coexistence in the region of closed shells N = 20 and N = 28. The evolution of the two neutron separation energies and the shell correction energy have been studied as a function of the neutron number. The results thus obtained provide a means of identifying, in exotic nuclei, new nuclear structure effects that are well illustrated by the changes of the conventional magic structure. Abstract

Abstract: The and reactions were measured in inverse kinematics at and analyzed within the coupled reaction channels framework, the cross section being particularly large. We find that coupling to pickup has a profound effect on the elastic scattering, and that these strong coupling effects should be included in analyses of proton elastic and inelastic scattering. Through its modification of the elastic scattering wave functions this coupling will strongly affect the extraction of spectroscopic information such as the relationship between neutron and proton nuclear deformations, with important consequences for our understanding of the structure of exotic nuclei. Abstract

We present a search for Lorentz violation with 8249 candidate electron antineutrino events taken by the Double Chooz experiment in 227.9 live days of running. This analysis, featuring a search for a sidereal time dependence of the events, is the first test of Lorentz invariance using a reactor-based antineutrino source. No sidereal variation is present in the data and the disappearance results are consistent with sidereal time independent oscillations. Under the Standard-Model Extension, we set the first limits on 14 Lorentz violating coefficients associated with transitions between electron and tau flavor, and set two competitive limits associated with transitions between electron and muon flavor. Abstract

A study on cosmic muons has been performed for the two identical near and far neutrino detectors of the Double Chooz experiment, placed at $\sim$120 and $\sim$300 m.w.e. underground respectively, including the corresponding simulations using the MUSIC simulation package. This characterization has allowed to measure the muon flux reaching both detectors to be (3.64 $\pm$ 0.04) $\times$ 10$^{-4}$ cm$^{-2}$s$^{-1}$ for the near detector and (7.00 $\pm$ 0.05) $\times$ 10$^{-5}$ cm$^{-2}$s$^{-1}$ for the far one. The seasonal modulation of the signal has also been studied observing a positive correlation with the atmospheric temperature, leading to an effective temperature coefficient of $\alpha_{T}$ = 0.212 $\pm$ 0.024 and 0.355 $\pm$ 0.019 for the near and far detectors respectively. These measurements, in good agreement with expectations based on theoretical models, represent one of the first measurements of this coefficient in shallow depth installations., Comment: 20 pages, 11 figures

Abstract

Using the Double Chooz detector, designed to measure the neutrino mixing angle $\theta_{13}$, the products of $\mu^-$ capture on $^{12}$C, $^{13}$C, $^{14}$N and $^{16}$O have been measured. Over a period of 489.5 days, $2.3\times10^6$ stopping cosmic $\mu^-$ have been collected, of which $1.8\times10^5$ captured on carbon, nitrogen, or oxygen nuclei in the inner detector scintillator or acrylic vessels. The resulting isotopes were tagged using prompt neutron emission (when applicable), the subsequent beta decays, and, in some cases, $\beta$-delayed neutrons. The most precise measurement of the rate of $^{12}\mathrm C(\mu^-,\nu)^{12}\mathrm B$ to date is reported: $6.57^{+0.11}_{-0.21}\times10^{3}\,\mathrm s^{-1}$, or $(17.35^{+0.35}_{-0.59})\%$ of nuclear captures. By tagging excited states emitting gammas, the ground state transition rate to $^{12}$B has been determined to be $5.68^{+0.14}_{-0.23}\times10^3\,\mathrm s^{-1}$. The heretofore unobserved reactions $^{12}\mathrm C(\mu^-,\nu\alpha)^{8}\mathrm{Li}$, $^{13}\mathrm C(\mu^-,\nu\mathrm n\alpha)^{8}\mathrm{Li}$, and $^{13}\mathrm C(\mu^-,\nu\mathrm n)^{12}\mathrm B$ are measured. Further, a population of $\beta$n decays following stopping muons is identified with $5.5\sigma$ significance. Statistics limit our ability to identify these decays definitively. Assuming negligible production of $^{8}$He, the reaction $^{13}\mathrm C(\mu^-,\nu\alpha)^{9}\mathrm{Li}$ is found to be present at the $2.7\sigma$ level. Limits are set on a variety of other processes.

Abstract

A new summation method model of the reactor antineutrino energy spectrum is presented. It is updated with the most recent evaluated decay databases and with our total absorption gamma-ray spectroscopy measurements performed during the last decade. For the first time, the spectral measurements from the Daya Bay experiment are compared with the antineutrino energy spectrum computed with the updated summation method without any renormalization. The results exhibit a better agreement than is obtained with the Huber-Mueller model in the 2-5 MeV range, the region that dominates the detected flux. A systematic trend is found in which the antineutrino flux computed with the summation model decreases with the inclusion of more pandemonium-free data. The calculated flux obtained now lies only 1.9% above that detected in the Daya Bay experiment, a value that may be reduced with forthcoming new pandemonium-free data, leaving less room for a reactor anomaly. Eventually, the new predictions of individual antineutrino spectra for the 235U, 239Pu, 241Pu, and 238U are used to compute the dependence of the reactor antineutrino spectral shape on the fission fractions. Abstract

We describe a muon track reconstruction algorithm for the reactor anti-neutrino experiment Double Chooz. The Double Chooz detector consists of two optically isolated volumes of liquid scintillator viewed by PMTs, and an Outer Veto above these made of crossed scintillator strips. Muons are reconstructed by their Outer Veto hit positions along with timing information from the other two detector volumes. All muons are fit under the hypothesis that they are through-going and ultrarelativistic. If the energy depositions suggest that the muon may have stopped, the reconstruction fits also for this hypothesis and chooses between the two via the relative goodness-of-fit. In the ideal case of a through-going muon intersecting the center of the detector, the resolution is ∼40 mm in each transverse dimension. High quality muon reconstruction is an important tool for reducing the impact of the cosmogenic isotope background in Double Chooz., National Science Foundation (U.S.), United States. Abstract

The Double Chooz experiment measures the neutrino mixing angle θ13 by detecting reactor ν¯e via inverse beta decay. The positron-neutron space and time coincidence allows for a sizable background rejection, nonetheless liquid scintillator detectors would profit from a positron/electron discrimination, if feasible in large detector, to suppress the remaining background. Standard particle identification, based on particle dependent time profile of photon emission in liquid scintillator, can not be used given the identical mass of the two particles. However, the positron annihilation is sometimes delayed by the ortho-positronium (o-Ps) metastable state formation, which induces a pulse shape distortion that could be used for positron identification. In this paper we report on the first observation of positronium formation in a large liquid scintillator detector based on pulse shape analysis of single events. The o-Ps formation fraction and its lifetime were measured, finding the values of 44% ± 12% (sys.) ± 5% (stat.) and 3.68ns ± 0.17ns (sys.) ± 0.15ns (stat.) respectively, in agreement with the results obtained with a dedicated positron annihilation lifetime spectroscopy setup., National Science Foundation (U.S.), United States. Abstract

The Double Chooz experiment presents improved measurements of the neutrino mixing angle $\theta_{13}$ using the data collected in 467.90 live days from a detector positioned at an average distance of 1050 m from two reactor cores at the Chooz nuclear power plant. Several novel techniques have been developed to achieve significant reductions of the backgrounds and systematic uncertainties with respect to previous publications, whereas the efficiency of the $\bar\nu_{e}$ signal has increased. The value of $\theta_{13}$ is measured to be $\sin^{2}2\theta_{13} = 0.090 ^{+0.032}_{-0.029}$ from a fit to the observed energy spectrum. Deviations from the reactor $\bar\nu_{e}$ prediction observed above a prompt signal energy of 4 MeV and possible explanations are also reported. A consistent value of $\theta_{13}$ is obtained from a fit to the observed rate as a function of the reactor power independently of the spectrum shape and background estimation, demonstrating the robustness of the $\theta_{13}$ measurement despite the observed distortion., Comment: 44 pages, 25 figures. Abstract

The Double Chooz experiment presents an indication of reactor electron antineutrino disappearance consistent with neutrino oscillations. An observed-to-predicted ratio of events of 0.944±0.016(stat)±0.040(syst) was obtained in 101 days of running at the Chooz nuclear power plant in France, with two 4.25 GW(th) reactors. The results were obtained from a single 10 m(3) fiducial volume detector located 1050 m from the two reactor cores. The reactor antineutrino flux prediction used the Bugey4 flux measurement after correction for differences in core composition. The deficit can be interpreted as an indication of a nonzero value of the still unmeasured neutrino mixing parameter sin(2)2θ(13). Analyzing both the rate of the prompt positrons and their energy spectrum, we find sin(2)2θ(13)=0.086±0.041(stat)±0.030(syst), or, at 90% C.L., 0.017

Abstract

To investigate the behavior of the N = 14 neutron gap far from stability with a neutron-sensitive probe, proton elastic and 2(1)+ inelastic scattering angular distributions for the neutron-rich nucleus 22O were measured using the MUr à STrip detector array at the Grand Accélérateur National d'Ions Lourds facility. A deformation parameter beta(p,p') = 0.26 +/- 0.04 is obtained for the 2(1)+ state, much lower than in 20O, showing a weak neutron contribution to this state. A microscopic analysis was performed using matter and transition densities generated by continuum Skyrme-Hartree-Fock-Bogoliubov and quasiparticle random phase approximation calculations, respectively. The ratio of neutron to proton contributions to the 2(1)+ state is found close to the N/Z ratio, demonstrating a strong N = 14 shell closure in the vicinity of the neutron drip line.

Abstract

Background: Maps depicting binary interactions between proteins can be powerful starting points for understanding biological systems. A proven technology for generating such maps is high-throughput yeast two-hybrid screening. In the most extensive screen to date, a Gal4-based two-hybrid system was used recently to detect over 20,000 interactions among Drosophila proteins. Although these data are a valuable resource for insights into protein networks, they cover only a fraction of the expected number of interactions., Results: To complement the Gal4-based interaction data, we used the same set of Drosophila open reading frames to construct arrays for a LexA-based two-hybrid system. We screened the arrays using a novel pooled mating approach, initially focusing on proteins related to cell-cycle regulators. We detected 1,814 reproducible interactions among 488 proteins. The map includes a large number of novel interactions with potential biological significance. Informative regions of the map could be highlighted by searching for paralogous interactions and by clustering proteins on the basis of their interaction profiles. Surprisingly, only 28 interactions were found in common between the LexA- and Gal4-based screens, even though they had similar rates of true positives., Conclusions: The substantial number of new interactions discovered here supports the conclusion that previous interaction mapping studies were far from complete and that many more interactions remain to be found. Our results indicate that different two-hybrid systems and screening approaches applied to the same proteome can generate more comprehensive datasets with more cross-validated interactions. The cell-cycle map provides a guide for further defining important regulatory networks in Drosophila and other organisms.

Abstract

Drosophila melanogaster is a proven model system for many aspects of human biology. Here we present a two-hybrid-based protein-interaction map of the fly proteome. A total of 10,623 predicted transcripts were isolated and screened against standard and normalized complementary DNA libraries to produce a draft map of 7048 proteins and 20,405 interactions. A computational method of rating two-hybrid interaction confidence was developed to refine this draft map to a higher confidence map of 4679 proteins and 4780 interactions. Statistical modeling of the network showed two levels of organization: a short-range organization, presumably corresponding to multiprotein complexes, and a more global organization, presumably corresponding to intercomplex connections. The network recapitulated known pathways, extended pathways, and uncovered previously unknown pathway components. This map serves as a starting point for a systems biology modeling of multicellular organisms, including humans.

Abstract

Background: Chediak-Higashi syndrome (CHS) is an inherited immunodeficiency disease characterized by giant lysosomes and impaired leukocyte degranulation. CHS results from mutations in the lysosomal trafficking regulator (LYST) gene, which encodes a 425-kD cytoplasmic protein of unknown function. The goal of this study was to identify proteins that interact with LYST as a first step in understanding how LYST modulates lysosomal exocytosis., Materials and Methods: Fourteen cDNA fragments, covering the entire coding domain of LYST, were used as baits to screen five human cDNA libraries by a yeast two-hybrid method, modified to allow screening in the activation and the binding domain, three selectable markers, and more stringent confirmation procedures. Five of the interactions were confirmed by an in vitro binding assay., Results: Twenty-one proteins that interact with LYST were identified in yeast two-hybrid screens. Four interactions, confirmed directly, were with proteins important in vesicular transport and signal transduction (the SNARE-complex protein HRS, 14-3-3, and casein kinase II)., Conclusions: On the basis of protein interactions, LYST appears to function as an adapter protein that may juxtapose proteins that mediate intracellular membrane fusion reactions. The pathologic manifestations observed in CHS patients and in mice with the homologous mutation beige suggest that understanding the role of LYST may be relevant to the treatment of not only CHS but also of diseases such as asthma, urticaria, and lupus, as well as to the molecular dissection of the CHS-associated cancer predisposition.

Abstract

Two large-scale yeast two-hybrid screens were undertaken to identify protein-protein interactions between full-length open reading frames predicted from the Saccharomyces cerevisiae genome sequence. In one approach, we constructed a protein array of about 6,000 yeast transformants, with each transformant expressing one of the open reading frames as a fusion to an activation domain. This array was screened by a simple and automated procedure for 192 yeast proteins, with positive responses identified by their positions in the array. In a second approach, we pooled cells expressing one of about 6,000 activation domain fusions to generate a library. We used a high-throughput screening procedure to screen nearly all of the 6,000 predicted yeast proteins, expressed as Gal4 DNA-binding domain fusion proteins, against the library, and characterized positives by sequence analysis. These approaches resulted in the detection of 957 putative interactions involving 1,004 S. cerevisiae proteins. These data reveal interactions that place functionally unclassified proteins in a biological context, interactions between proteins involved in the same biological function, and interactions that link biological functions together into larger cellular processes. The results of these screens are shown here.

Abstract

Mating pheromones stimulate Saccharomyces cerevisiae yeast cells to form a pointed projection that becomes the site of cell fusion during conjugation. To investigate the role of mating projections, we screened for mutations that enhanced the weak mating defect of MATa ste2-T326 cells that are defective in forming pointed projections. These cells are also 10-fold more sensitive to alpha-factor pheromone because ste2-T326 encodes truncated alpha-factor receptors that are not regulated properly. Mutations in AXL1, STE6 and FUS3 were identified in the screen. AXL1 was studied further because it is required for efficient a-factor pheromone production and for selecting the site for bud morphogenesis. Mutation of AXL1 did not enhance the morphogenesis or pheromone sensitivity defects of ste2-T326. Instead, the synergistic mating defect was apparently due to decreased a-factor production because the axl1Delta ste2-T326 cells mated well with a sst2 alpha mating partner that is supersensitive to a-factor. When combined with a wild-type mating partner, the ste2-T326 axl1Delta cells failed to mate because they did not lock cell walls, one of the earliest steps in conjugation. Analysis of axl1Delta in combination with other mutations that cause defects in morphogenesis or pheromone sensitivity (e.g. bar1, sst2, afr1) indicated that both phenotypes of ste2-T326 cells, supersensitivity to alpha-factor and the defect in forming pointed projections, contributed to the synergistic mating defect. We suggest a model that the synergistic mating defect is caused by the combined effects of ste2-T326 and axl1Delta on the presentation of a-factor to partner cells. Altogether, these results demonstrate an important linkage between the incoming and outgoing pheromone signals during the intercellular communication that promotes yeast mating., (Copyright 1999 John Wiley & Sons, Ltd.)

Abstract

G-protein-coupled receptors (GPCRs) transduce the signals for a wide range of hormonal and sensory stimuli by activating a heterotrimeric guanine nucleotide-binding protein (G protein). The analysis of loss-of-function and constitutively active receptor mutants has helped to reveal the functional properties of GPCRs and their role in human diseases. Here we describe the identification of a new class of mutants, dominant-negative mutants, for the yeast G-protein-coupled alpha-factor receptor (Ste2p). Sixteen dominant-negative receptor mutants were isolated based on their ability to inhibit the response to mating pheromone in cells that also express wild-type receptors. Detailed analysis of two of the strongest mutant receptors showed that, unlike other GPCR interfering mutants, they were properly localized at the plasma membrane and did not alter the stability or localization of wild-type receptors. Furthermore, their dominant-negative effect was inversely proportional to the relative amount of wild-type receptors and was reversed by overexpressing the G-protein subunits, suggesting that these mutants compete with the wild-type receptors for the G protein. Interestingly, the dominant-negative mutations are all located at the extracellular ends of the transmembrane segments, defining a novel region of the receptor that is important for receptor signaling. Altogether, our results identify residues of the alpha-factor receptor specifically involved in ligand binding and receptor activation and define a new mechanism by which GPCRs can be inactivated that has important implications for the evaluation of receptor mutations in other G-protein-coupled receptors.

Abstract

The POL3 encoded catalytic subunit of DNA polymerase delta possesses a highly conserved C-terminal cysteine-rich domain in Saccharomyces cerevisiae. Mutations in some of its cysteine codons display a lethal phenotype, which demonstrates an essential function of this domain. The thermosensitive mutant pol3-13, in which a serine replaces a cysteine of this domain, exhibits a range of defects in DNA repair, such as hypersensitivity to different DNA-damaging agents and deficiency for induced mutagenesis and for recombination. These phenotypes are observed at 24 degrees, a temperature at which DNA replication is almost normal; this differentiates the functions of POL3 in DNA repair and DNA replication. Since spontaneous mutagenesis and spontaneous recombination are efficient in pol3-13, we propose that POL3 plays an important role in DNA repair after irradiation, particularly in the error-prone and recombinational pathways. Extragenic suppressors of pol3-13 are allelic to sdp5-1, previously identified as an extragenic suppressor of pol3-11. SDP5, which is identical to HYS2, encodes a protein homologous to the p50 subunit of bovine and human DNA polymerase delta. SDP5 is most probably the p55 subunit of Pol delta of S. cerevisiae and seems to be associated with the catalytic subunit for both DNA replication and DNA repair.

Abstract

DNA polymerases (Pol) alpha, delta and epsilon are necessary for replication of nuclear DNA. Pol delta interacts permanently or transiently with numerous accessory proteins whose identification may shed light on the function(s) of Pol delta. In vitro mutagenesis was used to induce thermosensitive (ts) mutations in the DNA polymerase delta gene (POL3). We have attempted to clone two recessive extragenic suppressors of such ts mutants (sdp1 for mutation pol3-14 and sdp5-1 for mutation pol3-11) by transforming thermoresistant haploid strains pol3-14 sdp1 and pol3-11 sdp5-1 with wild-type genomic libraries in singlecopy or multicopy vectors. None of the thermosensitive transformants so obtained was identified as being sdp1 or sdp5-1. Instead, three genes were cloned whose products interfere with the activity of suppressors. One of them is the type 1 protein phosphatase gene, DIS2. Another is a novel gene, ASM4, whose gene product is rich in asparagine and glutamine residues.
