Atlantic salmon (Salmo salar) is the most valuable farmed fish globally and there is much interest in optimizing its genetics and rearing conditions for growth and feed efficiency. Marine feed ingredients must be replaced to meet global demand, with challenges for fish health and sustainability. Metabolic models can address this by connecting genomes to metabolism, which converts nutrients in the feed to energy and biomass, but such models are currently not available for major aquaculture species such as salmon. We present SALARECON, a model focusing on energy, amino acid, and nucleotide metabolism that links the Atlantic salmon genome to metabolic fluxes and growth. It performs well in standardized tests and captures expected metabolic (in)capabilities. We show that it can explain observed hypoxic growth in terms of metabolic fluxes and apply it to aquaculture by simulating growth with commercial feed ingredients. Predicted limiting amino acids and feed efficiencies agree with data, and the model suggests that marine feed efficiency can be achieved by supplementing a few amino acids to plant- and insect-based feeds. SALARECON is a high-quality model that makes it possible to simulate Atlantic salmon metabolism and growth. It can be used to explain Atlantic salmon physiology and address key challenges in aquaculture such as development of sustainable feeds.

Background The paper focuses on the problems of temporal functioning of obese individuals and of individuals prepared for bariatric treatment. The experience of time heavily weighs on many areas of human functioning, everyday activity, planning and achieving goals, engaging in pro-health behaviours, and in consequence on the quality of life and on physical health. Contingent on numerous factors, obesity may be related to focusing on particular aspects of time perspective. The aim of the study was to determine the specificity of particular temporal dispo-sitions in individuals prepared for bariatric surgery, and thus to devise suitable post-op psychological interven-tions. Participants and procedure The study sample comprised 28 individuals (60.7% women, mean age M = 43.82 years, SD = 10.01, mean BMI M = 44.83 kg/m 2 , SD = 6.51) awaiting bariatric surgery. The data were collected individually with the following pen-and-paper questionnaires: the Zimbardo Time Perspective Inventory, the Dark Future Scale, and the Polish adaptation of Snyder’s Adult Hope Scale, which is named the Hope for Success Questionnaire. Results The results showed the prevalence of present hedonistic time perspective in the obese. With regard to future anxiety or the hope for success, no significant deviations from the general population were observed. Conclusions The results indicate that this group is in need of temporal psychotherapy aimed at balancing the time perspec-tive. The results may also be interpreted through the lens of contextual determinants connected with task orien-tation preceding the surgery.

Nowadays, lab-on-chip (LOC) devices are attracting more and more attention since they show vast prospects for various biomedical applications. Usually, an LOC is a small device that serves a single laboratory function. LOCs show massive potential for organ-on-chip (OOC) device manufacturing since they could allow for research on the avoidance of various diseases or the avoidance of drug testing on animals or humans. However, this technology is still under development. The dominant technique for the fabrication of such devices is molding, which is very attractive and efficient for mass production, but has many drawbacks for prototyping. This article suggests a femtosecond laser microprocessing technique for the prototyping of an OOC-type device—a liver-on-chip. We demonstrate the production of liver-on-chip devices out of glass by using femtosecond laser-based selective laser etching (SLE) and laser welding techniques. The fabricated device was tested with HepG2(GS) liver cancer cells. During the test, HepG2(GS) cells proliferated in the chip, thus showing the potential of the suggested technique for further OOC development.

The aim of this article is to characterize selected problems faced by social researchers in prison facilities. The authors, referring to the literature on the subject and their own experience gained while conducting research in prisons, analyze the challenges faced by researchers who do not belong to penitentiary institutions. The first part of the article presents problems related to the organization of research in institutions of a total character, the specificity of the group of prisoners – participants of the research (including, among others, aspects of motivation, self-presentation and the cognitive and emotional functioning of prisoners) and the need for the researcher to maintain a neutral, impartial, objective attitude towards the subjects. The second part of the article is devoted to the reflection on the methodology of research on the temporal perspective, which appeared in connection with the NCN research project “Temporal aspects of the activity of men serving prison sentences – longitudinal study”. The analysis of the specificity of experiencing time by persons serving isolation penalties and the critical analysis of tools for measuring the temporal aspects of their functioning is presented. The article concludes with conclusions on factors that may increase the level of credibility of the results of research conducted in penitentiary institutions.

BackgroundFew single center studies from resource-poor settings have reported about the epidemiology, clinical feature and outcome of multisystem inflammatory syndrome in children (MIS-C). However, larger data from multi-center studies on the same is lacking including from Indian setting.MethodsThis retrospective collaborative study constituted of data collected on MIS-C from five tertiary care teaching hospitals from Eastern India. Children ≤ 15 years of age with MIS-C as per the WHO criteria were included. Primary outcome was mortality.ResultsA total of 134 MIS-C cases were included (median age, 84 months; males constituted 66.7%). Fever was a universal finding. Rash was present in 40%, and conjunctivitis in 71% cases. Gastro-intestinal and respiratory symptoms were observed in 50.7% and 39.6% cases, respectively. Co-morbidity was present in 23.9% cases. Shock at admission was noted in 35%, and 27.38% required mechanical ventilation. Fifteen (11.2%) children died. The coronary abnormalities got normalized during follow-up in all except in one child. Initial choice of immunomodulation had no effect on the outcomes. Presence of underlying co-morbidity, lymphopenia, thrombocytosis, hyponatremia, increased LDH (>300 U/L), and hypoalbuminemia were the factors significantly associated an increased mortality.ConclusionsMIS-C has myriad of manifestations. Underlying co-morbidity, lymphopenia, thrombocytosis, hyponatremia, increased LDH (>300 U/L), and hypoalbuminemia were associated with an increased mortality. No difference in outcome was noted with either steroid or IVIg or both. Coronary artery abnormalities resolved in nearly all cases.

Abstract Background The ever-increasing prevalence of obesity constitutes a major health problem worldwide. A subgroup of obese individuals has been described as “metabolically healthy obese” (MHO). In contrast to metabolically unhealthy obese (MUO), the MHO phenotype has a favorable risk profile. Despite this, the MHO phenotype is still sub-optimally characterized with respect to a comprehensive risk assessment. Our aim was to increase the understanding of metabolic alterations associated with healthy and unhealthy obesity. Methods In this cross-sectional study, men and women (18–70 years) with obesity (body mass index (BMI) ≥ 30 kg/m2) or normal weight (NW) (BMI ≤ 25 kg/m2) were classified with MHO (n = 9), MUO (n = 10) or NW (n = 11) according to weight, lipid profile and glycemic regulation. We characterized individuals by comprehensive metabolic profiling using a commercial available high-throughput proton NMR metabolomics platform. Plasma fatty acid profile, including short chain fatty acids, was measured using gas chromatography. Results The concentrations of very low density lipoprotein (VLDL), intermediate density lipoprotein (IDL) and low density lipoprotein (LDL) subclasses were overall significantly higher, and high density lipoprotein (HDL) subclasses lower in MUO compared with MHO. VLDL and IDL subclasses were significantly lower and HDL subclasses were higher in NW compared with MHO. The concentration of isoleucine, leucine and valine was significantly higher in MUO compared with MHO, and the concentration phenylalanine was lower in NW subjects compared with MHO. The fatty acid profile in MHO was overall more favorable compared with MUO. Conclusions Comprehensive metabolic profiling supports that MHO subjects have intermediate-stage cardiovascular disease risk marker profile compared with NW and MUO subjects. Introduction: Serum lactate is an important parameter to assess adequacy of microcirculation in septic shock. However, prediction of outcome in pediatric septic shock with mutli organ dysfunction (MODS) based on serial lactate levels and it's clearance is not well studied. Materials and Methods: In this prospective study from a tertiary PICU in India, 43 children with septic shock and MODS aged 1month to 16 years were enrolled over one year. Our objective was to predict early ICU mortality (within 72 hours) as well as in-hospital mortality using serial serum lactate levels and it's rate of clearance. Serial serum lactate levels were measured at admission to PICU (0 hours) and 6 hourly for initial 24 hours and 12 hourly till 120 hours. Results: The median (IQR) age of enrolled children was 32 (4.5,96) months. Median (IQR) PRISM-12 score was 16.5 (12,22). Seventeen children died. Serum lactate ≥2.6 mmol/L at 24 hours predicted early ICU mortality with sensitivity and specificity of 83.3% and 85.7% (AUC: 0.873, 95% CI 0.727, 1.000). In hospital mortality could be predicted with sensitivity and specificity of 60% and 73% (AUC: 0.734, 95% CI 0.571, 0.897) when serum lactate ≥1.8 mmol/L at 24 hours. Percentage of lactate clearance (median, IQR) at 24 hours of ICU stay were 15.1 (-21.4, 24.6) and -33.2 ( -262, 0.4) (p=0.04) between survivors and non-survivors at 72 hours of ICU stay. Conclusion: Pediatric septic shock and MODS carries high mortality and serum lactate of ≥2.6 mmol/L predicted early ICU mortality with good precision.

Background: Postinfectious bronchiolitis obliterans (PIBO) is a chronic obstructive lung disease with scanty information in literature on etiology, clinical profile, treatment, and outcome. Objective: The objective of the study is to describe the clinical profile and course of children diagnosed with PIBO. Methods: A chart review of children below 18 years of age diagnosed as PIBO over the past 9 years was carried out. Details of clinical profile, laboratory investigations, imaging, treatment received, and outcome were recorded. Results: Eight children (boys 4) with PIBO were identified. Median (interquartile range [IQR]) age at the first episode of acute severe bronchiolitis such as illness and diagnosis of PIBO was 15 (6, 23.5) and 30 (16.5, 60) months, respectively, indicating a delay in diagnosis. The most common symptoms were recurrent episodes of cough (100%), fast breathing (100%), wheezing (87.5%), and fever (62.5%). Median (IQR) number of hospitalizations and episodes of antibiotic use prior to diagnosis were 2.5 (2, 5.5) and 2 (2, 4), respectively. Three (37.5%) children received mechanical ventilation during previous hospitalizations. Chest computed tomography revealed mosaic attenuation in 8 (100%), ground-glass opacities in 2 (25%), and bronchial wall thickening in 2 (25%). After diagnosis, 7 received oral steroids, 7 received hydroxychloroquine, 5 received azithromycin, and 2 received azathioprine. The median (IQR) duration of follow-up (n = 6) was 6 (1.5, 9.5) months. Median (IQR) number of pulmonary exacerbations in follow-up was 2 (1, 5). Conclusion: PIBO is still an under-recognized entity with substantial delay in diagnosis and unnecessary use of antibiotics. Clinical course with imaging findings may help to diagnose and manage this entity.

The aim of the study is to analyze the relationship between sensory processing sensitivity (SPS), attention awareness, and resilience and to determine whether attention awareness may moderate the relation between sensitivity and resilience. The sample consisted of 273 adults (239 women; Mage=24.12, SD=6.59years). The highly sensitive person scale, the resiliency assessment scale, and the mindful attention awareness scale were used in the study. The results indicate significant relationships between SPS and resilience; emotional reactivity is especially associated with lower resilience, whereas sensing the subtle is associated with higher resilience. The regression analyses revealed that SPS is a significant predictor of resilience, but diverse aspects of sensitivity explain resilience differently. Attentional awareness was found to be a significant moderator that strengthens the positive relationship between sensing the subtle and tolerance of negative emotions. The consequences of high sensitivity include high levels of distress, anxiety, and a sense of overload; therefore, searching for protective factors is important to maintain the wellbeing of highly sensitive people. As one of the characteristics of highly sensitive persons, sensing the subtle may be an important resource that allows to them to deal effectively with difficult situations. Training on attention awareness and conscious presence may be an important way to deal with negative emotions and develop personal competences. The results indicate that these strategies may be of high significance for improving wellbeing and protecting highly sensitive persons against various stress factors.

The aim of the study is to analyze the relationship between sensory processing sensitivity and occupational burnout. This analysis makes it possible to explore the problem of sensitivity as a predictor of burnout syndrome and to discuss adequate coping strategies in the work environment to improve employee well-being. The sample consists of 516 employees (236 women; Mage = 28.97, SD = 6.62 years). The Highly Sensitive Person Scale (HSPS; 27 items) was used to measure sensory processing sensitivity; the Oldenburg Burnout Inventory (OLBI; 16 items) was used to measure exhaustion and disengagement from work. The analysis of sensitivity is based on the ‘DOES’ model of a highly sensitive person, which includes depth of processing (D), overstimulation (O), emotional reactivity and empathy (E), and sensing the subtle (S). Burnout due to exhaustion and disengagement is analyzed. The factor analysis of the HSPS confirms its three-factor structure. The results indicate significant relationships between sensory processing sensitivity and burnout symptoms. However, the influence of specific aspects of sensitivity on the burnout problem varies: the results indicate that higher emotional reactivity is linked to worse burnout symptoms, but sensing the subtle shows the opposite effect and may be a protective factor against exhaustion. Significant gender differences were observed [F(511,1) = 136.63, p < 0.001; η2 = 0.21]; women revealed a significantly higher level of general sensitivity (Mwomen = 4.66, SD = 0.62) as compared to men (Mmen = 4.01; SD = 0.64). Awareness of one’s high sensory-processing sensitivity and its potential outcomes in the work environment may be essential in order to implement appropriate regulatory strategies. Proactive strategies aimed at reducing prolonged emotional overload may be critical for highly sensitive employees. Recognizing high sensitivity may reduce burnout symptoms and improve well-being at work.

The Salmon gill poxvirus (SGPV) has emerged in recent years as the cause of an acute respiratory disease that can lead to high mortality in farmed Atlantic salmon presmolts, known as Salmon gill poxvirus disease. SGPV was first identified in Norway in the 1990s, and its large DNA genome, consisting of over 206 predicted protein-coding genes, was characterized in 2015. This review summarizes current knowledge relating to disease manifestation and its effects on the host immune system and describes dissemination of the virus. It also demonstrates how newly established molecular tools can help us to understand SGPV and its pathogenesis. Finally, we conclude and ask some burning questions that should be addressed in future research.

Conjugate-vaccine immunogens require three components: a carrier protein, an antigen, and a crosslinker, capable of coupling antigen to carrier protein, while preserving both T-cell responses from carrier protein and B-cell responses from antigen. We previously showed that the N-terminal eight residues of the HIV-1 fusion peptide (FP8) as an antigen could prime for broad cross-clade neutralizing responses, that recombinant heavy chain of tetanus toxin (rTTHC) as a carrier protein provided optimal responses, and that choice of crosslinker could impact both antigenicity and immunogenicity. Here, we delve more deeply into the impact of varying the linker between FP8 and rTTHC. In specific, we assessed the physical properties, the antigenicity, and the immunogenicity of conjugates for crosslinkers ranging in spacer-arm length from 1.5 to 95.2 Å, with varying hydrophobicity and crosslinking-functional groups. Conjugates coupled with different degrees of multimerization and peptide-to-rTTHC stoichiometry, but all were well recognized by HIV-fusion-peptide-directed antibodies VRC34.01, VRC34.05, PGT151, and ACS202 except for the conjugate with the longest linker (24-PEGylated SMCC; SM(PEG)24), which had lower affinity for ACS202, as did the conjugate with the shortest linker (succinimidyl iodoacetate; SIA), which also had the lowest peptide-to-rTTHC stoichiometry. Murine immunizations testing seven FP8-rTTHC conjugates elicited fusion-peptide-directed antibody responses, with SIA- and SM(PEG)24-linked conjugates eliciting lower responses than the other five conjugates. After boosting with prefusion-closed envelope trimers from strains BG505 clade A and consensus clade C, trimer-directed antibody-binding responses were lower for the SIA-linked conjugate; elicited neutralizing responses were similar, however, though statistically lower for the SM(PEG)24-linked conjugate, when tested against a strain especially sensitive to fusion-peptide-directed responses. Overall, correlation analyses revealed the immunogenicity of FP8-rTTHC conjugates to be negatively impacted by hydrophilicity and extremes of length or low peptide-carrier stoichiometry, but robust to other linker parameters, with several commonly used crosslinkers yielding statistically indistinguishable serological results.
