165. Lysosome turnover in the Retinal Pigment Epithelium in health, ageing and Age-related Macular Degeneration

Abstract

The Retinal Pigment Epithelium (RPE) phagocytoses and degrades spent Photoreceptor Outer Segments (POS) every day, placing an unparalleled burden on lysosomes in RPE cells, which require well-regulated turnover to maintain degradative capacity. With age and in Age-related Macular Degeneration (AMD), debris accumulates inside and outside RPE cells leading to the hypothesis that lysosome dysfunction could contribute to AMD pathology. This project aimed to identify the functional relevance of different subpopulations of lysosome within the heterogenous lysosomal compartment of RPE cells. Models of lysosome dysfunction were then generated in order to determine whether they recapitulate hallmarks of aging and AMD. Electron microscopy (EM) and electron tomography of primary porcine RPE (pRPE) revealed morphologically distinct subpopulations of lysosomes that form a complex network. Pulse-chase experiments showed that multilamellar Cathepsin D-rich lysosomes transition to electron dense lysosomes that stained poorly for Cathepsin D, suggesting that these subpopulations represent different stages of the lysosome cycle. Tracking of phagocytosed POS suggests that POS degradation occurs by both full fusion and kiss-and-run interactions between phagosomes and lysosomes. Correlative light and electron microscopy, and high content imaging, demonstrated that POS loading induces the gradual formation of lipofuscin-like autofluorescent granules. Loading cells with degradation- resistant UV-irradiated POS induced the formation of more and larger autofluorescent granules, thus recapitulating one of the hallmarks of aging and AMD. Treatment with the acidotropic compound chloroquine caused accumulation of undigested phagocytic, endocytic and autophagic cargo, consistent with lysosome dysfunction. This was accompanied by accumulation of lamellar lysosomes containing unprocessed lysosomal Cathepsin D, a feature 4 recapitulated by direct protease inhibition and prevented by inhibiting protein synthesis. Together, the findings in this PhD project shed new light on the identity, function and fate of lysosomes in the RPE and suggest that lysosome dysfunction could contribute to the pathology of AMD, as well as to chloroquine-induced retinopathy.

Abstract

Objective: This work evaluates a new approach for calculating the left-ventricular (LV) early filling propagation velocity (VP) from color M-Mode (CMM) echocardiograms using wavelet analysis. Unlike current approaches, the method requires no assumptions, user inputs, or heuristic conventions. Background: Current methods for measuring VP using CMM echocardiography do not account for the spatiotemporal variation of the filling wave propagation velocity. They are instead confined by empirical assumptions and user inputs that significantly hinder the accuracy of VP, subsequently limiting its clinical utility. Methods: We evaluated three methods for measuring LV early filling VP: conventional VP, the strength of propagation (VS), and VP determined from the most energetically important wave (Peak-VW), using 125 patients (Group A) with normal filling (n=50), impaired relaxation (n=25), pseudonormal filling (n=25) and restrictive filling (n=25), and in 69 patients (Group B) with normal (n=32), dilated (n=15), and hypertrophic ventricles (n=22). Results: Peak-VW most accurately distinguished normal left ventricles from diseased ones. Specifically, for Group A which spanned the stages of diastolic function, using receiver operator characteristics (ROC) and measuring their corresponding area under the curve (AUC), the AUC for Peak-VW was 0.92, versus 0.62 for conventional VP, 0.63 for VS and 0.58 for intraventricular pressure difference (IVPD). These correspond to a 50-70% improvement in classification ability. Similar improvements were found using Group B. Conclusion: A new determination of the LV early filling using wavelet analysis, Peak-VW, may provide a more accurate evaluation of diastolic function than the standard method of determining Vp and enable better diagnostic classification of patients suffering with diastolic dysfunction.

169. Geometry of joint reality: device-independent steering and operational completeness

Abstract

We look at what type of arguments can rule out the joint reality (or value definiteness) of two observables of a physical system, such as a qubit, and give several strong yet simple no-go results based on assumptions typically weaker than those considered previously. The first result uses simple geometry combined with a locality assumption to derive device-independent steering inequalities. These may also be regarded as "conditional" Bell inequalities, are simpler in principle to test than standard Bell inequalities, and for two-qubit systems are related to properties of the quantum steering ellipsoid. We also derive a Bell inequality from locality and a one-sided reality assumption, and demonstrate a close connection between device-independent steering and Bell nonlocality. Moreover, we obtain a no-go result without the use of locality or noncontextuality assumptions, based on similar geometry and an assumption that we call "operational completeness". The latter is related to, but strictly weaker than, preparation noncontextuality. All arguments are given for finite statistics, without requiring any assumption that joint relative frequencies converge to some (unobservable) joint probability distribution. Abstract

This article introduces the notion of good labellings for asymptotic lattices in order to study joint spectra of quantum integrable systems from the point of view of inverse spectral theory. As an application, we consider a new spectral quantity for a quantum integrable system, the quantum rotation number. In the case of two degrees of freedom, we obtain a constructive algorithm for the detection of appropriate labellings for joint eigenvalues, which we use to prove that, in the semiclassical limit, the quantum rotation number can be calculated on a joint spectrum in a robust way, and converges to the well-known classical rotation number. The general results are applied to the semitoric case where formulas become particularly natural.

Abstract

To what extent are research results influenced by subjective decisions that scientists make as they design studies? Fifteen research teams independently designed studies to answer five original research questions related to moral judgments, negotiations, and implicit cognition. Participants from 2 separate large samples (total N > 15,000) were then randomly assigned to complete 1 version of each study. Effect sizes varied dramatically across different sets of materials designed to test the same hypothesis: Materials from different teams rendered statistically significant effects in opposite directions for 4 of 5 hypotheses, with the narrowest range in estimates being d = -0.37 to + 0.26. Meta-analysis and a Bayesian perspective on the results revealed overall support for 2 hypotheses and a lack of support for 3 hypotheses. Overall, practically none of the variability in effect sizes was attributable to the skill of the research team in designing materials, whereas considerable variability was attributable to the hypothesis being tested. In a forecasting survey, predictions of other scientists were significantly correlated with study results, both across and within hypotheses. Abstract

Harmonization of acquisition and analysis protocols is an important step in the validation of BOLD MRI as a renal biomarker. This harmonization initiative provides technical recommendations based on a consensus report with the aim to move towards standardized protocols that facilitate clinical translation and comparison of data across sites. We used a recently published systematic review paper, which included a detailed summary of renal BOLD MRI technical parameters and areas of investigation in its supplementary material, as the starting point in developing the survey questionnaires for seeking consensus. Survey data were collected via the Delphi consensus process from 24 researchers on renal BOLD MRI exam preparation, data acquisition, data analysis, and interpretation. Consensus was defined as ≥ 75% unanimity in response. Among 31 survey questions, 14 achieved consensus resolution, 12 showed clear respondent preference (65-74% agreement), and 5 showed equal (50/50%) split in opinion among respondents. Recommendations for subject preparation, data acquisition, processing and reporting are given based on the survey results and review of the literature. These technical recommendations are aimed towards increased inter-site harmonization, a first step towards standardization of renal BOLD MRI protocols across sites. We expect this to be an iterative process updated dynamically based on progress in the field.

Abstract

We experimentally test the recently predicted anisotropic invariance properties of pure three-qubit states, via generation and measurement of polarisation-path entangled three-qubit states. These properties do not require aligned reference frames and can be determined from measurements on any two of the qubits. They have several applications, such as a universal ordering of pairwise quantum correlations, strong monogamy relations for Bell inequalities and quantum steering, and a complementarity relation for Bell nonlocality versus 3-tangle, some of which we also test. The results indicate that anisotropic invariance, together with the three qubit Bloch vector lengths, can provide a robust and complete set of invariants for such states under local unitary transformations., Comment: 6 pages, 1 figure

Abstract

Hypertension is the major attributable risk factor for cardiovascular diseases (CVD) including heart failure (HF). As blood pressure increases, the risk of HF increases concomitantly. The incidence of HF continues to increase as people are living longer with comorbid diseases, and HF is a leading cause of morbidity and mortality in the United States. Black adults are disproportionately affected by hypertension and HF and develop CVD at younger ages compared to White adults. There are several factors which may contribute to these disparities including metabolic factors and sociodemographic factors. Treatment of hypertension can reduce the risk for HF. Lifestyle modification can be effective at reducing the risk of both hypertension and subsequent HF. Anti-hypertensive medications are available which are effective, safe, and cost-effective. Despite wide recognition that hypertension is a strong risk factor for HF, effective treatments for hypertension, and guideline recommendations for better blood pressure control, hypertension control rates remain poor in the United States. In this paper, we discuss the relationships between hypertension and HF, the impact of racial disparities on hypertension and HF, and prevention and treatment strategies. 191. Struggles for the soul of higher education : a genealogy of graduate employability

Abstract

Graduate employability is a central part of the contemporary debate over the purpose and value of higher education. This thesis undertakes a Foucauldian genealogy to interrogate the discursive formation of graduate employability in higher education in the United Kingdom. To the author’s knowledge this is the first attempt to do so. The thesis constructs a genealogy in two ways. Firstly, it identifies in the contemporary literature five distinct discourses of graduate employability. This establishes the discursive character of graduate employability. Secondly, it interrogates five higher education policy documents published since the Second World War to propose how different discourses might emerge through distinctive historical conditions. Each represents a critical moment of emergence of a distinctive configuration of graduate employability. Thus, the illusion of graduate employability as a singular, neutral concept is dissolved. It is shown to be contingent upon the social, economic and political conditions of the times. Graduate employability is thus a product of power relations and not merely a technical concept. In unmasking graduate employability as contingent it reveals forgotten attempts to mould and shape higher education through policy. Graduate employability thus becomes part of the apparatus of the disciplining of higher education. Furthermore, the research points to the futility of addressing graduate employability as something externally imposed upon the ‘true’ mission of higher education. Instead, higher education is inescapably bound up in this repeated contest for its own soul and is thus empowered to creatively resist. It therefore affirms the agency of higher education in shaping the very idea of graduate employability. The major contributions to knowledge are, therefore: the demonstration of graduate employability as discursive; the reconstruction of those historical conditions that have enabled the emergence of different discourses; and the application of Foucault’s genealogical toolkit to a novel aspect of higher education policy.

196. On the dynamics of initially correlated open quantum systems: theory and applications

Abstract

We show that the dynamics of any open quantum system that is initially correlated with its environment can be described by a set of (or less) completely positive maps, where d is the dimension of the system. Only one such map is required for the special case of no initial correlations. The same maps describe the dynamics of any system-environment state obtained from the initial state by a local operation on the system. The reduction of the system dynamics to a set of completely positive maps allows known numerical and analytic tools for uncorrelated initial states to be applied to the general case of initially correlated states, which we exemplify by solving the qubit dephasing model for such states, and provides a natural approach to quantum Markovianity for this case. We show that this set of completely positive maps can be experimentally characterised using only local operations on the system, via a generalisation of noise spectroscopy protocols. As further applications, we first consider the problem of retrodicting the dynamics of an open quantum system which is in an arbitrary state when it becomes accessible to the experimenter, and explore the conditions under which retrodiction is possible. We also introduce a related one-sided or limited-access tomography protocol for determining an arbitrary bipartite state, evolving under a sufficiently rich Hamiltonian, via local operations and measurements on just one component. We simulate this protocol for a physical model of particular relevance to nitrogen-vacancy centres, and in particular show how to reconstruct the density matrix of a set of three qubits, interacting via dipolar coupling and in the presence of local magnetic fields, by measuring and controlling only one of them., Comment: 19 pages. Abstract

The set of all qubit states that can be steered to by measurements on a correlated qubit is predicted to form an ellipsoid---called the quantum steering ellipsoid---in the Bloch ball. This ellipsoid provides a simple visual characterisation of the initial 2-qubit state, and various aspects of entanglement are reflected in its geometric properties. We experimentally verify these properties via measurements on many different polarisation-entangled photonic qubit states. Moreover, for pure 3-qubit states, the volumes of the two quantum steering ellipsoids generated by measurements on the first qubit are predicted to satisfy a tight monogamy relation, which is strictly stronger than the well-known monogamy of entanglement for concurrence. We experimentally verify these predictions, using polarisation and path entanglement. We also show experimentally that this monogamy relation can be violated by a mixed entangled state, which nevertheless satisfies a weaker monogamy relation.

We consider the coupling of quantum fields to classical gravity in the formalism of ensembles on configuration space, a model that allows a consistent formulation of interacting classical and quantum systems. Explicit calculations show that there are solutions for which two quantum fields are in an entangled state, even though their interaction occurs solely via a common classical gravitational field, and that such entangled solutions can evolve from initially unentangled ones. Abstract

Bell's inequality was originally derived under the assumption that experimenters are free to select detector settings independently of any local "hidden variables" that might affect the outcomes of measurements on entangled particles. This assumption has come to be known as "measurement independence" (also referred to as "freedom of choice" or "settings independence"). For a two-setting, two-outcome Bell test, we derive modified Bell inequalities that relax measurement independence, for either or both observers, while remaining locally causal. We describe the loss of measurement independence for each observer using the parameters $M_1$ and $M_2$, as defined by Hall in 2010, and also by a more complete description that adds two new parameters, which we call $\hat{M}_1$ and $\hat{M}_2$, deriving a modified Bell inequality for each description. These "relaxed" inequalities subsume those considered in previous work as special cases, and quantify how much the assumption of measurement independence needs to be relaxed in order for a locally causal model to produce a given violation of the standard Bell-Clauser-Horne-Shimony-Holt (Bell-CHSH) inequality. We show that both relaxed Bell inequalities are tight bounds on the CHSH parameter by constructing locally causal models that saturate them. For any given Bell inequality violation, the new two-parameter and four-parameter models each require significantly less mutual information between the hidden variables and measurement settings than previous models. We conjecture that the new models, with optimal parameters, require the minimum possible mutual information for a given Bell violation. Abstract

'Locality' is a fraught word, even within the restricted context of Bell's theorem. As one of us has argued elsewhere, that is partly because Bell himself used the word with different meanings at different stages in his career. The original, weaker, meaning for locality was in his 1964 theorem: that the choice of setting by one party could never affect the outcome of a measurement performed by a distant second party. The epitome of a quantum theory violating this weak notion of locality (and hence exhibiting a strong form of nonlocality) is Bohmian mechanics. Recently, a new approach to quantum mechanics, inspired by Bohmian mechanics, has been proposed: Many Interacting Worlds. While it is conceptually clear how the interaction between worlds can enable this strong nonlocality, technical problems in the theory have thus far prevented a proof by simulation. Here we report significant progress in tackling one of the most basic difficulties that needs to be overcome: correctly modelling wavefunctions with nodes.
