The present study aimed to research the effects of cyclic heat environment on the microbial diversity and structure of respiratory tract and cecum of chicken. A total of 360 layer-type pullets at 11 wk of age were subjected to different temperature treatments for 10 wk: constant 22°C; cyclic temperature 22°C to 24°C, 22°C to 26°C, 22°C to 28°C, 22°C to 30°C; the ambient temperature increased from 10:00, reached the set point within 1 h, and maintained until 18:00, thereafter the temperature was restored to 22°C; and the relative humidity was maintained at 60%. The result showed that feed intake of the chickens on ambient temperature 30°C group was significantly lower than that of the chickens on ambient temperature 24°C. The white blood cell, red blood cell, lymphocyte, hemoglobin, and pecked-cell volume content were highest at 24°C on 14, 16, and 18 wk. The ratio of CD3+CD4+/CD3+CD8+ T cells was lowest at 30°C. Meanwhile, the abundance of cecum bacteria in chickens at 30°C was lower than that at 24°C. Cyclic heat environment temperature treatment did not significantly affect the concentration of secretory immunoglobulin A in chicken bronchoalveolar lavage fluid (BALF) levels during 10 wk of trial. The diversity index analysis showed that the effect of 24°C on the cecum flora of chickens was optimal. Abundance of Firmicutes bacteria in the lung flora and cecum flora was lower at 30°C than at 24°C group. Similarly, the microorganism, Brevibacillus in the BALF was also significantly lower at 24°C. In conclusion, cyclic 24°C treatment was beneficial for the feed intake, blood routine indexes, microflora structure of the cecum, and respiratory tract in laying pullets.

Crystalline amino acids (AAs) exhibit high nutritional values when supplemented AA-deficient diets. However, the AAs in crystalline form in the diet are absorbed quickly than protein-bound AAs, which may take an effect on AA utilization efficiency. In this study, 2 experiments were conducted to investigate the effect of encapsulated lysine-HCl (Lys) and DL-methionine (DL-Met) on the growth performance of broiler chickens. In experiment 1, a total of 432 one-day-old male Arbor Acres broilers were subjected to 3 dietary treatments (27 pens; 16 birds per pen) for 42 d. The control group was basal diets supplemented with crystalline Lys and DL-Met, and treatment groups had basal diets supplemented with encapsulated Lys and DL-Met at the levels of 80% and 60% of control diets (80CLM, 60CLM), respectively. The growth performance, intestinal development, and transcription of AA transporters were determined. In experiment 2, 24 broiler chickens were subjected to the same treatments as in experiment 1. The plasma concentrations of free AAs were measured 0, 2, 4, and 6 h after feeding. The results showed that 80CLM treatment had no significant influence on production performance, carcass characteristics, and plasma free AAs content during the experiment compared with the control group (P > 0.05). In addition, the 80CLM group moderately enhanced gut morphology development and increased AAs' absorption capacity. However, broilers fed the 60CLM diet had lower production performance and breast muscle weight than the control group (P < 0.05), but increased villi height and B0AT mRNA expression level (P < 0.05). At h 4 after feeding, the 60CLM broilers exhibited higher concentration of Ala, Cys, and total dispensable AAs than the control group (P < 0.05). In conclusion, the result suggests that the supplemental levels of crystalline Lys and DL-Met can be effectively saved approximately for 20% by using the encapsulated form in broilers, with improvements to AAs utilization efficiency, while posing no detrimental effects on production performance. Encapsulated Lys and DL-Met would have greater potential for application when replacing crystalline AAs in broiler chickens.

Abstract Background L-citrulline (L-Cit), a non-protein amino acid, has been implicated in several physiological functions including anti-inflammatory, anti-oxidative, and hypothermic roles, however, there is a paucity of information with regards to its potential in poultry production. Methods This study was designed to investigate the effects of dietary L-Cit supplementation on the production performance, nitric oxide production, and antioxidant status of laying hens during summer period. Hy-Line Brown laying hens (n = 288, 34 weeks old) were allotted to four treatment, 6 replicates of 12 chickens each. Dietary treatments of control (basal diets), 0.25%, 0.50% and 1.00% L-Cit supplementation were fed to chickens for eight (8) weeks. Production performance, free amino acid profiles, nitric oxide production, and antioxidant properties were measured. Blood samples were collected at the 4th and 8th weeks of the experiment. Results Air temperature monitoring indicated an average daily minimum and maximum temperatures of 25.02 °C and 31.01 °C respectively. Dietary supplementation with L-Cit did not influence (P > 0.05) the production performance, and rectal temperature of laying hens. Egg shape index was increased (P

ABSTRACT: The present study was designed to investigate whether L-arginine (Arg) supplementation would influence the production performance and reproductive traits of laying hens fed low crude protein (LCP) diets. Hy-Line brown laying hens were randomly assigned to dietary treatments of control, LCP, and LCP supplemented with 0.05, 0.10, 0.15, or 0.20% Arg for 7 wk. The results showed no significant variations on the production performance, and relative organ indexes of hens. However, significant transcriptional and structural changes were evident along the HPG axis of hens. Arg supplementation linearly upregulated (P < 0.05) the gonadotropin-releasing hormone 1 (GnRH1), and gonadotropin inhibitory hormone in the hypothalamus. The pituitary growth hormone, GnRH receptor, and follicle-stimulating hormone (FSHβ) were also increased (P < 0.05). In the ovary, GnRH1, and estrogen receptor β were linearly increased by Arg, and the ovarian morphology revealed that LCP induced structural alterations which were minimally recovered by Arg supplementation. Serum insulin-like growth factor 1 (IGF-1) and nitric oxide (NO) were increased (P < 0.05) at higher levels of Arg supplementation. Therefore, supplementing high Arg (0.20%) to LCP hens influenced the ovarian morphology and modulated the gene expression of reproductive hormones in the hypothalamic-pituitary-gonadal axis of laying hens via actions that may be related to NO and IGF-1 activity.

ABSTRACT: Calcium and phosphorus homeostasis is crucial for the performance and bone health of laying hens. The calcium and phosphorus transporters play an important role in calcium and phosphorus absorption, reabsorption, and excretion. In the present study, Hy-Line Brown layers were sampled at brooding period (1, 4, 6 wk), growing and developing period (12, 18 wk) and laying period (20, 28, 80 wk) respectively, and the calcium transporters CaBP-D28k and PMCA1b and phosphorus transporters NPt2a and NPt2b were respectively measured in duodenum, jejunum, ileum and kidney. The result showed that serum calcium increased (P < 0.0001) and phosphorus level fluctuated (P = 0.0019), while alkaline phosphatase activity decreased with age (P < 0.0001). The mRNA and protein expressions of CaBP-D28k in small intestine elevated after maturity (P ≤ 0.0001). In contrast, the PMCA1b mRNA showed a trend to increase with age in jejunum (P = 0.0059) and ileum (P = 0.0825) whereas there was a decrease for PMCA1b protein in 12-18 wk (P ≤ 0.0009). The peak of NPt2b mRNA were observed at 28 wk in duodenum (P = 0.0001) and jejunum (P = 0.0622) and 1 wk in ileum (P < 0.0001). The NPt2b protein expression reached the top point at 4 or 6 wk and 20 wk and decreased to the lowest point around 12 wk (P ≤ 0.0850). In kidney, CaBP-D28k mRNA was not influenced by age (P = 0.4999), while PMCA1b highly expressed in 6-12 wk (P = 0.0003). The protein expressions of CaBP-D28k (P = 0.0148) and PMCA1b (P = 0.0003) decreased with age and lowly expressed in 12-18 wk and increased thereafter. In contrast, NPt2a expression increased steadily with age and decreased at 80 wk (P < 0.0001). In conclusion, the expressions of intestinal calcium and phosphorus transporters were changed by age, intestinal CaBP-D28k and renal NPt2a showed a dramatic increase after maturity, which coincide with the increased calcium and phosphorus requirement for egg production.

Nutrition during the pre-lay period takes effect on the production performance in the laying flock. This study evaluated the effects of dietary energy and protein levels in pre-lay diet on performance during the whole laying period and the egg quality, bone quality, and mRNA expression of hypothalamus-pituitary-gonadal (HPG) axis-related genes of hens at the end of the laying cycle. A total of 1,856 15-wk old Hy-Line brown pullets were randomly assigned to one of the four dietary treatments: using a 2 × 2 factorial arrangement with 2 energy levels (2,700 and 2,800 kcal/kg ME, respectively) and 2 protein levels (15 and 16.5% CP, respectively). Pullets were fed ad libitum from 15 to 20 wk and from 20 wk onward, fed with a similar laying diet till 72 wk of age. At 72 wk, the expression of genes in the hypothalamus, pituitary, ovarian, and follicles and bone quality was evaluated. At 72wk, there were no differences in production performance, BW, organ index, and ovarian parameters among the dietary treatments. High-CP diet increased the egg shape index and eggshell thickness (p < 0.05), but the eggshell breaking strength, Haugh unit, and albumen height did not differ among the treatments. Neither dietary energy nor protein level took an effect of bone quality. Low-energy diet increased the mRNA expression of gonadotropin-releasing hormone-1 (GnRH-1) in the hypothalamus (p < 0.05). The mRNA expression level of estrogen receptor-1 (ESR-1) in the hypothalamus and ovary was elevated by the 2,700 ME-15%CP diet (p < 0.05). The expression of cytochrome family 17 subfamily A polypeptide 1 (CYP17A1) in the large white follicle (LWF), small yellow follicles (SYF) and dominant follicle (DF) was decreased by the 2,800 kcal/kg diet (p < 0.05). These results indicate that the prelay diet had no influence on the production performance but had minimal effect on the eggshell characteristics and bone parameters. These results suggest that the energy and protein level of the prelay diet changes the expression of HPG axis-related genes of hens around the end of the laying cycle without changing the circulating sex hormone profile. The effect of prelay diet on the endocrinal adjustment at the end of the laying cycle needs to be investigated further.

Pulmonary fibrosis is a group of life-threatening diseases with limited therapeutic options. The involvement of cannabinoid type 1 receptors (CB1R) has been indicated in fibrotic diseases, but whether or not the activation of CB1R can be a benefit for fibrosis treatment is controversial. In this study, we investigated the effects of arachidonoylcyclopropylamide (ACPA), as a selective CB1R agonist, on bleomycin (BLM)-induced pulmonary fibrosis. We showed that ACPA treatment significantly improved the survival rate of BLM-treated mice, alleviated BLM-induced pulmonary fibrosis, and inhibited the expressions of extracellular matrix (ECM) markers, such as collagen, fibronectin, and α-SMA. The enhanced expressions of ECM markers in transforming growth factor-beta (TGF-β)-challenged primary lung fibroblasts isolated from mouse lung tissues were inhibited by ACPA treatment in a dose-dependent manner, and the fibroblast migration triggered by TGF-β was dose-dependently diminished after ACPA administration. Moreover, the increased mRNA levels of CB1R were observed in both lung fibroblasts of BLM-induced fibrotic mice in vivo and TGF-β-challenged primary lung fibroblasts in vitro. CB1R-specific agonist ACPA significantly diminished the activation of TGF-β–Smad2/3 signaling, i.e., the levels of p-Smad2 and p-Smad3, and decreased the expressions of downstream effector proteins including slug and snail, which regulate ECM production, in TGF-β-challenged primary lung fibroblasts. Collectively, these findings demonstrated that CB1R-specific agonist ACPA exhibited antifibrotic efficacy in both in vitro and in vivo models of pulmonary fibrosis, revealing a novel anti-fibrosis approach to fibroblast-selective inhibition of TGF-β-Smad2/3 signaling by targeting CB1R.

ABSTRACT: The mechanism that mediates L-citrulline (L-Cit) hypothermia is poorly understood, and the involvement of nitric oxide signaling has not been fully elucidated. Therefore, this study aimed to determine L-Cit's influence on body temperature and to ascertain the central and peripheral mechanisms associated with this response. Chicks responded to intracerebroventricular (ICV) injection of L-Cit with high and low body temperatures (P < 0.05) depending on the dose tested, for both the surface and rectal temperatures. Peripheral (i.p.) L-Cit injection did not affect body temperature responses. Nitric oxide (NO) concentration and NO synthase (NOS) were influenced with varying doses of L-Cit. Hypothalamic NO was increased at 4 µg L-Cit whereas, plasma iNOS was elevated at 2µg L-Cit treatment. However, i.p. L-Cit did not change the NO content, rather it induced higher (P < 0.05) plasma tNOS and iNOS activity, and further upregulated iNOS and nNOS gene expression in the hypothalamus. In addition, ICV L-Cit potentiated a pro- versus anti-inflammatory milieu with the induction of IL-8, IL-10, and TGFβ (P < 0.05), which may be related to the changes in body temperature. Following ICV L-Cit administration, it was observed that L-Cit caused dose variable changes in the ultrastructure of hypothalamic neurons. The lowest dose was associated with a higher number of dead or degenerating neurons, whereas the highest L-Cit dose had fewer neuronal numbers with larger sizes. Therefore, this study shows that central and peripheral L-Cit administration imposes changes in body temperature, nitric oxide production, and inflammatory responses, in a dose-dependent manner.

Diabetic kidney disease (DKD) is a long-term major microvascular complication of uncontrolled hyperglycemia and one of the leading causes of end-stage renal disease (ESDR). The pathogenesis of DKD has not been fully elucidated, and effective therapy to completely halt DKD progression to ESDR is lacking. This study aimed to identify critical molecular signatures and develop novel therapeutic targets for DKD. This study enrolled 10 datasets consisting of 93 renal samples from the National Center of Biotechnology Information (NCBI) Gene Expression Omnibus (GEO). Networkanalyst, Enrichr, STRING, and Cytoscape were used to conduct the differentially expressed genes (DEGs) analysis, pathway enrichment analysis, protein-protein interaction (PPI) network construction, and hub gene screening. The shared DEGs of type 1 diabetic kidney disease (T1DKD) and type 2 diabetic kidney disease (T2DKD) datasets were performed to identify the shared vital pathways and hub genes. Strepotozocin-induced Type 1 diabetes mellitus (T1DM) rat model was prepared, followed by hematoxylin & eosin (HE) staining, and Oil Red O staining to observe the lipid-related morphological changes. The quantitative reverse transcription-polymerase chain reaction (qRT-PCR) was conducted to validate the key DEGs of interest from a meta-analysis in the T1DKD rat. Using meta-analysis, 305 shared DEGs were obtained. Among the top 5 shared DEGs, Tmem43, Mpv17l, and Slco1a1, have not been reported relevant to DKD. Ketone body metabolism ranked in the top 1 in the KEGG enrichment analysis. Coasy, Idi1, Fads2, Acsl3, Oxct1, and Bdh1, as the top 10 down-regulated hub genes, were first identified to be involved in DKD. The qRT-PCR verification results of the novel hub genes were mostly consistent with the meta-analysis. The positive Oil Red O staining showed that the steatosis appeared in tubuloepithelial cells at 6 w after DM onset. Taken together, abnormal ketone body metabolism may be the key factor in the progression of DKD. Targeting metabolic abnormalities of ketone bodies may represent a novel therapeutic strategy for DKD. These identified novel molecular signatures in DKD merit further clinical investigation.

ABSTRACT: Adiponectin (ADPN) is related to fatty acid synthesis and oxidation in mammals. In chickens, the lipid metabolism, structure and sequence of ADPN are different from that in mammals. The aim of this study was to determine the role of ADPN in broilers lipid metabolism by investigating the temporal and spatial expression profiles of ADPN and its receptors, as well as their response to feed restriction. The results showed that the abdominal fat has the highest expression level, followed by the duodenum, glandular stomach, heart, hypothalamus, liver, and skeletal muscle. Broilers have high energy mobilization during their early stage of growth, in which the fat demand in the liver and muscles is high, thus the expression of ADPN and its receptor are also increased. To study the effects of feed restriction on ADPN and lipid metabolism, broilers were fasted for 12 h and refeed for 2 h. The results showed that fasting decreased the concentration of triglyceride (TG) (P < 0.05) and total cholesterol (TCHO) (P < 0.05) in plasma. The mRNA expression of ADPN in the liver (P < 0.05), breast (P < 0.05) and thigh (P < 0.05), and the mRNA expression of ADPNR1 in the liver (P < 0.05) and duodenum (P < 0.05) were significantly increased in the Fasted group. All above phenomena were recovered after refeeding, suggesting that feed restriction may promote the utilization of fatty acids in active metabolism tissues through ADPN, to guarantee the energy homeostasis of the body. However, the AMP-activated protein kinase (AMPK) signaling pathway and hepatic lipid metabolism were not necessary to cause the above changes under this experimental condition.

ABSTRACT: In poultry production, vaccination is an effective measure to protect chickens from diseases. Vaccination, however, is a stressor that may induce stress responses that interfere with the growth and development of chickens. The interaction between the skeletal and immune systems on bone quality has gained more attention. In the present study, the influence of high frequency vaccinations on the bone development of layer pullets was investigated. Thirty 35-day-old SPF White Leghorn layer pullets were obtained and randomly subjected to the following treatments: vaccinated against Newcastle disease (ND) with LoSota vaccine once at 35-day-old (V1, control); 4 times at 35, 49, 63, and 77 d of age (V4); and 7 times at 35, 42, 49, 56, 63, 70, and 77 d of age (V7). The body weight and organ index of the spleen, thymus, and tibia were recorded. The antibody titer and serum and the tibia calcium and phosphorus concentrations were measured. The transcription levels of the IL-6, IL-17, TNF-α, receptor activator of NF-κB ligand (RANKL), and osteoprotegerin (OPG) genes were determined in spleen, thymus, and the tibia. The results showed that V7 decreased body weight and increased the ND antibody titer, compared to V1-chickens. The expression levels of IL-6, IL-17, and TNF-α were upregulated in spleen, thymus, and the tibia of V7 chickens. In the tibia, RANKL was upregulated, while OPG was downregulated by V7 treatment. The results indicate that high frequency vaccination induces immune stress and impairs bone development. The results suggest that the augmented cytokine expression in immune organs and the tibia is associated with activation of the OPG/RANKL pathway, which, in turn, enhances osteoclastogenesis. The appropriate frequency of vaccination should support optimal bone development and full immunoprotection in layer pullets.

Heat stress (HS) adversely affects several physiological responses in organisms, but the underlying molecular mechanisms involved are yet to be fully understood. L-Citrulline (L-Cit) is a nutraceutical amino acid that is gaining research interest for its role in body temperature regulation and nitric oxide synthesis. This study investigated whether dietary supplementation with L-Cit (1% of basal diet) could ameliorate the effects of acute HS on thermotolerance, redox balance, and inflammatory responses of broilers. Ross 308 broilers (288 chicks) were subjected to two environments; thermoneutral at 24°C (TNZ) or HS at 35°C for 5 h, and fed two diets; control or L-Cit. The results showed that HS increased the ear, rectal (RT), and core body (CBT) temperatures of broilers, along with higher respiratory rate. The RT and CBT readings were intermittently affected with time effect, whereas, L-Cit supplementation lowered the mean CBT than the control diet. Antioxidant assays showed that superoxide dismutase was increased during HS, while, catalase was promoted by L-Cit supplementation. In addition, L-Cit induced glutathione peroxidase activity compared to the control diet during HS. Hypothalamic heat shock protein (HSP)-90 was upregulated by HS, but L-Cit downregulated heat shock factor (HSF)-1, and HSP 60 mRNA expressions. HSF 3 mRNA expression was downregulated by L-Cit under TNZ condition. More so, HS increased the plasma nitric oxide (NO) concentration but lowered the total NO synthase (tNOS) activity. In contrast, L-Cit supplementation limited NO production but increased the tNOS activity. Arginase activity was increased in the control fed group during HS but L-Cit supplementation lowered this effect. The NOS-COX pathway was significantly affected under TNZ condition, since L-Cit supplementation downregulated the mRNA expression of iNOS-COX2 in the hypothalamus, and further reduced the serum PGE2 concentration. Together, these data indicates that L-Cit influenced the antioxidant defense, heat shock response and nitric oxide regeneration both under thermoneutral and HS conditions; and that L-Cit may be directly and/or indirectly involved in the central regulation of body temperature.

ABSTRACT: Calcium (Ca) and phosphorus (P) transporters are responsible for their absorption and transport in small intestine and kidney, contributing to eggshell formation. The light-dark cycle is a primary cue in the reproduction of laying hen. In this study, we investigated the effect of different light-dark programs on eggshell quality and the expression of genes related to Ca and P transportation in laying hens. Seventy-two 56-week-old laying hens were randomly divided into two groups and reared at 16-h light and 8-h dark (control) or 9-h light and 15-h dark regime (long dark phase, LDP). The expressions of calcium transporter calbindin-D28k (CaBP-D28k), plasma membrane Ca ATPase 1b (PMCA1b), and phosphorus transporter NaPi-IIb (NPt2b) and NaPi-IIa (NPt2a) were measured in the small intestine, kidney, and eggshell gland. The results showed that feed intake (P < 0.001) and egg weight (P = 0.05) were decreased by LDP treatment. Compared with control, the eggshell hardness was increased (P = 0.011) by LDP treatment, but the eggshell thickness and the percentage of eggshell were not changed. The Ca and P contents in eggshell were increased by LDP treatment. During the scotophase, LDP-hens showed higher serum Ca (P = 0.0056) and P levels (P = 0.079) but lower alkaline phosphatase (ALP) activity than that of control hens. In the duodenum, the relative higher expression of CaBP-D28k and PMCA1b in scotophase compared to photophase was masked by LDP treatment. The expression of CaBP-D28k and osteopontin (OPN) in the eggshell gland were increased by LDP treatment, compared to control hens. In the jejunum, the protein expression levels of CaBP-D28k and PMCA1b decreased during photophase in LDP-hens. The result indicates that the increased blood Ca and P concentration during scotophase by LPD treatment is beneficial to the deposition of Ca and P in the eggshell. The result offers an alternative strategy for managing laying hens with poor eggshell quality.

Running can promote public health. However, the association between running and the built environment, especially in terms of micro street-level factors, has rarely been studied. This study explored the influence of built environments at different scales on running in Inner London. The 5Ds framework (density, diversity, design, destination accessibility, and distance to transit) was used to classify the macro-scale features, and computer vision (CV) and deep learning (DL) were used to measure the micro-scale features. We extracted the accumulated GPS running data of 40,290 sample points from Strava. The spatial autoregressive combined (SAC) model revealed the spatial autocorrelation effect. The result showed that, for macro-scale features: (1) running occurs more frequently on trunk, primary, secondary, and tertiary roads, cycleways, and footways, but runners choose tracks, paths, pedestrian streets, and service streets relatively less; (2) safety, larger open space areas, and longer street lengths promote running; (3) streets with higher accessibility might attract runners (according to a spatial syntactic analysis); and (4) higher job density, POI entropy, canopy density, and high levels of PM 2.5 might impede running. For micro-scale features: (1) wider roads (especially sidewalks), more streetlights, trees, higher sky openness, and proximity to mountains and water facilitate running; and (2) more architectural interfaces, fences, and plants with low branching points might hinder running. The results revealed the linkages between built environments (on the macro- and micro-scale) and running in Inner London, which can provide practical suggestions for creating running-friendly cities.

Background: Uric acid (UA) is a potent scavenger of oxidants in mammalian and avian species. In humans, hyperglycemia with simultaneous hyperuricemia may exert additional damage to the cardiovascular system. Chickens naturally have hyperglycemia (10.1–11.0 mmol/L) and hyperuricemia (100–900 μmol/L), which makes them an interesting model.Methods: The aim of this study was to investigate the effects of UA on the oxidative damage induced by acute exposure of high level of glucose in chicken cardiac myocytes.Results: Cell viability and the concentrations of thiobarbituric acid reactive substance (TBARS) were decreased by glucose treatment in a dose- and time-dependent manner. After acute exposure to high level of glucose (300 mM), a moderate level of UA (300 μM) increased cell viability and reduced TBARS and glutathione (GSH) content. Compared to the control or to independent high glucose (300 mM) or UA (1,200 μM) treatment, the concurrent treatment of high glucose and high UA significantly increased the TBARS, protein carbonyl contents, and ROS concentration, whereas it decreased the cell viability, superoxide dismutase (SOD) activity, and GSH content. In the presence of high glucose and UA, the nucleic protein expression of nuclear factor erythroid 2-related factor 2 (Nrf2) was decreased and the mRNA levels of the genes cat, sod1, sod2, gss, and gclc were downregulated.Conclusion: In conclusion, acute exposure of high level of glucose induced oxidative damage in the cardiac myocytes of chicken. The present result suggests that an adequate level of uric acid is helpful in alleviating the acute oxidative damage that is induced by high glucose, whereas the inhibition of the Nrf2 pathway by a high level of uric acid may render the cardiac myocytes more vulnerable to suffering from oxidative damage.

BackgroundSerotonin (5-HT), a monoaminergic neurotransmitter, involves in the regulation of many physiological functions. In the present study, the effects of 5-hydroxytryptophan (5-HTP), the precursor of 5-HT, on lipid metabolism and intestinal immune function in broiler chickens were investigated in chickens.MethodsTwo hundred broilers were divided randomly into two groups and fed separately with a corn-soybean basal diet (CD) or the basal diet supplemented with 0.2% 5-HTP.ResultsThe results showed that 5-HTP reduced (P < 0.05) feed intake and the abdominal fat pad weight. 5-HTP treatment tended to upregulate the mRNA level of adiponectin receptor 1 (ADP1R) and ADP2R in abdominal fat but had no significant influence on their protein levels (P > 0.05). In 5-HTP-chickens, lipopolysaccharide exposure decreased secretory immunoglobulin A (sIgA) concentrations in serum and the duodenal contents. Expression of mRNA encoding interleukin (IL), tumor necrosis factor-α (TNF-α), and transforming growth factor-β (TGF-β) decreased after 5-HTP treatment; however, LPS increased expression significantly in 5-HTP-treated chickens compared with CD chickens. In 5-HTP-chickens, the phosphorylation of mitogen-activated protein kinase (MAPK) and nuclear factor-kappa B (NF-κB) were reduced, but the phosphorylation of ribosomal p70S6 kinase (p70S6K) was increased in the duodenum.ConclusionIn summary, the result suggests that dietary 5-HTP supplementation reduces accumulation of abdominal fat and is beneficial to intestinal immune function.

Objective: Glucocorticoids (GCs) can induce oxidative damage in skeletal muscles. The purpose of this study was to demonstrate a high caloric (HC) diet rich in soy oil would change the oxidative stress induced by a GC. Methods: The effect of dexamethasone (DEX) and HC diet on oxidative stress in plasma, skeletal muscles (M. pectoralis major, PM; M. biceps femoris, BF), and mitochondria were determined. The biomarkers of oxidative damage and antioxidative enzyme activity were determined. The fatty acid profile of muscles and the activities of complex I and II in mitochondria were measured. Results: The results showed that DEX increased the concentrations of oxidative damage markers in plasma, muscles, and mitochondria. The activity of complex I was significantly suppressed by DEX. DEX-chickens had higher proportions of polyunsaturated fatty acids and lower proportions of monounsaturated fatty acids in the PM. A HC diet decreased the levels of oxidative damage biomarkers in plasma, muscles, and mitochondria. The interaction between DEX and diet suppressed the activities of complex I and II in HC-chickens. Discussion: Oxidative damage in skeletal muscles and mitochondria was the result of GC-induced suppression of the activity of mitochondrial complex I. A HC diet improved the antioxidative capacity and reduced the oxidative damage induced by the GC.

Digital depth model (DDM) is the use of limited and discrete depth points to realize the digital expression of the undulations of the seabed terrain surface. According to the different organization methods of depth point data, it is divided into Regular Grid DDM (GRID-DDM) and Triangulated Irregular Network DDM (TIN-DDM). Compared with GRIDDDM, TIN-DDM has not undergone any data interpolation processing, and directly uses the measured depth as its model sampling point. Therefore, TIN-DDM has relatively prominent advantages in reflecting topographical changes. Based on TIN-DDM, the conclusions of seabed topography analysis are more accurate. Different from the traditional TIN-DDM that serves the needs of navigation applications, TIN-DDM (non-navigational TIN-DDM) that serves the analysis and expression of authentic terrain needs to focus on the accurate expression of the seabed terrain, that is to ensure the accuracy of the expression of seabed topographic features and the adequacy of the expression of seabed topography in the process of multi-scale expression (synthesis) of TIN-DDM. Constrained by the traditional navigational TIN-DDM serving the safety and security thinking of ships, there are few researches on automatic synthesis algorithms specifically for non-navigational TIN-DDM application requirements. Inspired by the idea of the terrestrial TIN-DEM multi-scale expression algorithm, most of the existing non-nautical TINDDM automatic synthesis algorithms are improved by the terrestrial TIN-DEM multi-scale expression algorithm. However, it needs to be pointed out that unlike terrestrial TIN-DEM, which already contains complete land topographic control information (generally composed of typical topographic feature points) during data collection, the seabed topography measurement cannot directly obtain the seabed topographic feature points and the related seabed topography range, due to the particularity of the measurement methods. The existing automatic synthesis algorithm is difficult to realize the full mining of the deep-level terrain feature information in the non-navigational TIN-DDM, and the exploration of the correlation between the spatial scale and the terrain shape judgment conclusion is not deep enough. Therefore, this paper proposes a non-navigational TIN-DDM automatic synthesis algorithm that takes into account the accurate determination of terrain features and full expression of topography. First, taking the local Delaunay influence domain as the range constraint of the sampling point terrain feature, analyzing the numerical change law of the critical rolling ball radius in the positive (negative) direction, establishing the TIN-DDM sampling point terrain type judgment criterion for micro-topography, and determining the topographic unevenness of sampling points within the range of local topographic features; Secondly, using the overall TIN-DDM as a reference for the range of topographic features of the sampling point, analyzing the numerical change law of the contact degree between the rolling ball and the TIN-DDM sampling point, establishing the criteria for determining the terrain type of the sampling point for macro-topography, and determining the skeleton and detailed terrain sampling points within the overall terrain feature range; On this basis, with the critical rolling ball radius as the associated link, based on the terrain unevenness of the TIN-DDM sampling points in the local area, analyzing the changing laws of topographic features caused by continuous longitudinal changes in the location of the sampling points, demonstrating the numerical correlation between the skeleton topographic sampling points and the critical rolling ball radius, and exploring the continuous state change trend of the topographic features of the sampling points from micro to macro, constructing TINDDM sampling point terrain feature quantitative evaluation index that takes into account the expression of seabed topography, and designing TIN-DDM automatic synthesis algorithm flow based on TIN-DDM sampling evaluation index; Finally, through experiments, the effectiveness of the algorithm in this paper is proved. The data used in the experiment is the TIN-DDM depth data of a certain sea area in China, which contains 12774 sampling points in total, and the topography is complex and the degree of undulation is large Based on Surfer software to generate the terrain surface of the original seabed terrain and the isobath with a depth of 5 meters, as shown in Figure 1. Eliminating the invalid data and locating the doubtful data are realized by using the algorithm of density clustering by referring to the image features of outliers in the rearview from the perspective of manual interactive processing. The MultiBeam EchoSounder (MBES) bathymetric datasets are automatically divided into three types: credible data, doubtful data, and invalid data by the clustering algorithm. The reliable data are retained, invalid data are automatically eliminated, and the doubtful data are judged manually. This kind of automatic outlier cleaning algorithm with partial manual intervention can solve the contradiction between the low reliability of automatic processing algorithms and the low efficiency of manual interactive processing. An example shows that the algorithm improves the reliability of the results obtained from the automatic processing algorithm to a certain extent, and also, the algorithm is of great significance for the realization of high reliability and high-efficiency cleaning of outliers in MBES bathymetric data. As the world continues to explore the ocean, the demand for efficient access to high precision underwater topography is increasing, the accuracy and efficiency of underwater terrain acquisition nowadays will not be able to meet the needs of technology development. As one of the most efficient and precise equipment, the MBES system is widely used worldwide in underwater topography surveys. In recent years, the original MBES data proliferating. However, for a long time, the cleaning of outliers in MBES data with high reliability mainly relies on manual operation, which has an extended processing cycle and dramatically limits data processing efficiency. The automatic processing algorithms that have been proposed improve processing efficiency, but the credibility can not reach that of human processing; it is challenging to apply it in multibeam surveying, especially with nautical charting. Therefore, there is a crucial problem in MBES data processing:how to improve the credibility of the results processed with the automatic algorithm. After years of research, many automatic data cleaning algorithms have been proposed both at home and abroad. The cleaning mechanism can be summarized as follows:Setting up several simple thresholds; Filtering by conditions based on the statistical results;establishing local topography model. The boundary between the recognition and validation of outliers is blurred by most of the existing automatic algorithms, and the MBES soundings are directly divided into two categories according to the specific conditions: credible and ineffective. But in surveying practice, there are usually a few data belonging to the third category, which we call doubtful data. The third category of data has to be verified by professionals, who need to determine with the experience combined with the investigation data of the survey area and instrument characteristics. For example, the causes of a small cluster of points isolated from the central cluster may be suspended solids in water or angular objects, such as stones; the former should be marked as invalid data and eliminated, while the latter should be marked as trusted data to be retained. Most of the existing automatic cleaning algorithms mark the doubtful data as credible data or invalid data across the board, which is the key to the low reliability of the current automatically processing algorithms. Although dubious data accounted for a small percentage, it is crucial because it often represents the characteristics area where the underwater topography changes dramatically or targets unnatural topographic relief. Once been misjudged, it would have a significant influence on the measuring result. Currently, there is no appropriate unartificial solution to validate the doubtful data. Although the doubtful data that need manual validation is rare, manually Screening for doubtful data ping by ping would also take much time. Therefore, it is a feasible program that could improve the credibility and efficiency of data processing, that the doubtful data are located automatically and validated manually. With the matures of the machine learning algorithm, more and more scholars apply it to the cleaning of multibeam outliers, due to its unique advantages in target recognition. Yang Fanlin et al. introduced an erosion and expansion clustering algorithm to realize the automatic cleaning of MBES outliers in the deepwater area(Yang et al., 2007). Xiaolong Chen et al. used the K-Means clustering algorithm to detect outliers based on single pulse-sounding data. In this algorithm, a ping was divided into multiple clustering windows according to the topographic change. With beams in the center of a ping as initial clustering samples, a cluster on behalf of the actual underwater topography was clustered from the central beam to the edge; and the outliers are weed out automatically. This method solves the problem of recognition accuracy, but the K-Means algorithm is sensitive to the noise. As a result, the algorithm may fail when there are large areas of continuous abnormal or missing data. Although the methods above can classify MBES soundings appropriately, they still directly divide the data into two categories: credible and invalid, and the validity of the doubtful data is not isolated; the processing results have the problem of credibility. For this reason, an automatic data cleaning algorithm with partially manual intervention was proposed, Based on the Density Clustering algorithm DBSCAN (Density-based Spatial Clustering of Applications with Noise). DBCSAN algorithm is used to divide the bathymetric data into three categories: credible, doubtful, and invalid, retains the credible data and eliminates the invalid data, and the dubious data submitted to manual validation. The method could effectively improve the credibility of the automatic data cleaning algorithm. Simultaneously, compared with manual processing, the method in this paper still dramatically enhances efficiency. A contradictory problem has been solved, that the automatic processing method has high efficiency but low credibility, while manual processing method has high reliability but low efficiency of this contradiction The main steps of the algorithm include two parts: identification of outliers based on DBSCAN and validation with manual intervention. The first step is to divide the original data into three categories: credible data, invalid data, and doubtful data through the clustering algorithm. The trusted data is retained, the invalid data is eliminated, and the doubtful data is submitted to the manual validation. Finally, the doubtful data will be classified as credible data or invalid data. The concept of 'doubtful data' is introduced into the automatic cleaning of outliers in multibeam bathymetric data, which provides a new idea for the automatic cleaning of outliers. A new automatic cleaning method in multibeam bathymetric data is established, which uses the DBSCAN algorithm to classify bathymetric data and manually validate the doubtful data. This automatic processing algorithm effectively divides raw data into three types: credible data, doubtful data, and invalid data. Retain the credible data and eliminate invalid data. The validation of the dubious data is carried with manual intervention. An experiment was conducted to investigate the effect of peripheral insulin treatment on appetite in chicks. Six-d-age chicks with ad libitum feeding or fasting for 3 h before injection received a subcutaneous injection of 0, 1, 3, 5, 10, or 20 IU of insulin or vehicle (saline). The results showed peripheral insulin treatment (1 to 20 IU) did not alter significantly the feed intake in chicks under either ad libitum feeding or fasting conditions within 4 h (p>0.05). Compared with the control, plasma glucose concentration was significantly decreased after insulin treatment of 3, 5, 10, and 20 IU for 4 h in chicks with ad libitum feeding (p0.05). All results suggest peripheral administration of insulin has no effect on appetite in chicks.

A nonlinear dynamic model is developed to analyze the stability of a pilot-operated valve-control hydraulic system. The dynamic model includes motion of the valve spool and fluid dynamics in the system. Characteristics such as pressure flow across the valve port and orifices, pressure, and flow rate in valve chambers are taken into consideration. Bifurcation analysis is proposed and examined by numerical simulation results when the feedback orifice diameter changes. The effects of different system parameters such as pilot-operating pressure, spring stiffness, and overlap of inlet port on the stability border of the system are studied by two-dimensional bifurcation analyses. The study identifies that bifurcation can occur in the system and lead to sustained self-excited vibration with parameters in certain region of the parameter space. It suggests that the vibration can be effectively predicted and prevented by selecting system parameters from the asymptotic stable parameter region.

The transport of calcium and phosphorus is mainly relied on their corresponding transporters. The aim of this study was to determine the effect of dietary phosphorus level on the expression of the relevant calcium and phosphorus transporters in laying hens, which has a large amount of calcium and phosphorus input from intestine and output from kidney and eggshell gland. Thirty-six 25-week-old Hy-line Brown hens were fed diets with different available phosphorus level (AP, 0.15, 0.41, and 0.82%), respectively. The expression of phosphorus transporters type IIa and type IIb Na/Pi co-transporter (NPt2a, NPt2b), calcium transporter calbindin-D28k (CaBP-D28k), and plasma membrane Ca ATPase 1b (PMCA1b) were measured in small intestine, kidney, and eggshell gland by RT-PCR and western blot. The results showed that serum calcitriol and PTH concentrations were not affected (P > 0.05) by dietary AP levels. Duodenum had the highest mRNA and protein expression level of NPt2b than jejunum and ileum (P < 0.05). The protein expression abundance of CaBP-D28k and PMCA1b were higher in duodenum than that in jejunum and ileum (P < 0.05). 0.15%-AP diet upregulated the ileal mRNA expression level of NPt2b and renal mRNA expression level of NPt2a (P < 0.05), while downregulated the protein abundance of NPt2b and CaBP-D28k mRNA expression in shell gland (P < 0.05). In conclusion, both the Ca and P transporters were highly expressed in duodenum. Low AP diet decreased protein expression abundance of NPt2b in duodenum while upregulated the mRNA expression level of NPt2a in kidney. The result suggests that both the phosphorus absorption in proximal intestine and its reabsorption in kidney are involved in the adaption to low AP diet.

Reticuloendotheliosis virus (REV), a gammaretrovirus in the Retroviridae family, causes an immunosuppressive, oncogenic, and runting–stunting syndrome in multiple avian hosts. Allicin, the main effective component of garlic, has a broad spectrum of pharmacological properties. The hypothesis that allicin could relieve REV-induced immune dysfunction was investigated in vivo and in vitro in the present study. The results showed that dietary allicin supplementation ameliorated REV-induced dysplasia and immune dysfunction in REV-infected chickens. Compared with the control groups, REV infection promoted the expression of inflammatory cytokines including interleukin (IL)-1β, IL-6, IL-10, interferon (IFN)-γ, and tumor necrosis factor-α (TNF-α), whereas, allicin reversed these changes induced by REV infection. The decreased levels of IFN-α, IFN-β, and IL-2 were observed in REV-infected chickens, which were significantly improved by allicin. Allicin suppressed the REV-induced high expression of toll-like receptors (TLRs) as well as melanoma differentiation-associated gene 5 (MDA5) and the activation of mitogen-activated protein kinase (MAPK) and the nuclear factor kappa B p65. REV stimulated the phosphorylation of JNK, ERK, and p38, the downstream key signaling molecules of MAPK pathway, while allicin retarded the augmented phosphorylation level induced by REV infection. The decreased phosphorylation level of ERK was associated with REV replication, suggesting that ERK signaling is involved in REV replication, and allicin can alleviate the REV-induced immune dysfunction by inhibiting the activation of ERK. In addition, REV infection induced oxidative damage in thymus and spleen, whereas allicin treatment significantly decreased the oxidative stress induced by REV infection, suggesting that the antioxidant effect of allicin should be at least partially responsible for the harmful effect of REV infection. In conclusion, the findings suggest that allicin alleviates the inflammation and oxidative damage caused by REV infection and exerts the potential anti-REV effect by blocking the ERK/MAPK pathway.

Acute bacterial meningitis is still considered one of the most dangerous infectious diseases in children. To investigate the prevalence and antibiotic resistance profiles of cerebrospinal fluid (CSF) pathogens in children with acute bacterial meningitis in Southwest China, CSF samples from 179 meningitis patients (3 days to 12 years old) with positive culture results were collected from 2012 to 2015. Isolated pathogens were identified using the Vitek-32 system. Gram stain results were used to guide subcultures and susceptibility testing. The antimicrobial susceptibility of isolates was determined using the disc diffusion method. Of the isolates, 50.8% were Gram-positive bacteria, and 49.2% were Gram-negative bacteria. The most prevalent pathogens were E. coli (28.5%), Streptococcus pneumoniae (17.8%), Staphylococcus epidermidis (10.0%), Haemophilus influenzae type b (9.5%), and group B streptococcus (7.2%). In young infants aged ≤3 months, E. coli was the organism most frequently isolated from CSF (39/76; 51.3%), followed by group B streptococcus (13/76; 17.1%) and Streptococcus pneumoniae (8/76; 10.5%). However, in young infants aged >3 months, the most frequently isolated organism was Streptococcus pneumoniae (24/103; 23.3%), followed by Staphylococcus epidermidis (18/103; 17.5%) and Haemophilus influenzae type b (16/103; 15.5%). Antimicrobial susceptibility tests indicated that for E. coli isolates, the susceptibility rates to aminoglycosides ranged from 56.8% to 100.0%, among them, amikacin was identified as the most effective against E. coli. As for cephalosporins, the susceptibility rates ranged from 29.4% to 78.4%, and cefoxitin was identified as the most effective cephalosporin. In addition, the susceptibility rates of piperacillin/tazobactam and imipenem against E. coli were 86.3% and 100%. Meanwhile, the susceptibility rates of Streptococcus pneumoniae isolates to penicillin G, erythromycin, chloramphenicol, ceftriaxone and tetracycline were 68.8%, 0.0%, 87.5%, 81.3% and 0.0%, respectively. Gentamycin, ofloxacin, linezolid and vancomycin were identified as the most effective antibiotics for Streptococcus pneumoniae, each with susceptibility rates of 100%. It was notable that other emerging pathogens, such as Listeria monocytogenes and group D streptococcus, cannot be underestimated in meningitis.

Heat transfer performances and flow structures of laminar impinging slot jets with power-law non-Newtonian fluids and corresponding typical industrial fluids (Carboxyl Methyl Cellulose (CMC) solutions and Xanthangum (XG) solutions) have been studied in this work. Investigations are performed for Reynolds number Re less than 200, power-law index n ranging from 0.5 to 1.5 and consistency index K varying from 0.001 to 0.5 to explore heat transfer and flow structure of shear-thinning fluid and shear-thickening fluid. Results indicate that with the increase of n, K for a given Re, wall Nusselt number increases mainly attributing to the increase of inlet velocity U. For a given inlet velocity, wall Nusselt number decreases with the increase of n and K, which mainly attributes to the increase of apparent viscosity and the reduction of momentum diffusion. For the same Re, U and Pr, wall Nusselt number decreases with the increase of n. Among the study of industrial power-law shear-thinning fluid, CMC solution with 100 ppm shows the best heat transfer performance at a given velocity. Moreover, new correlation of Nusselt number about industrial fluid is proposed. In general, for the heat transfer of laminar confined impinging jet, it is best to use the working fluid with low viscosity.

Myostatin, a member of the TGF-β superfamily of secreted proteins, is expressed primarily in skeletal muscle. It negatively regulates muscle mass and is associated with glucocorticoid-induced muscle atrophy. However, it remains unclear whether myostatin is involved in glucocorticoid-induced muscle protein turnover. The aim of the present study was to investigate the role of myostatin in protein metabolism during dexamethasone (DEX) treatment. Protein synthesis rates and the expression of the genes for myostatin, ubiquitin-proteasome atrogin-1, MuRF1, FoxO1/3a and mTOR/p70S6K were determined. The results show that DEX decreased (P0.05). The phosphorylation levels of mTOR and p70S6K were decreased by DEX treatment (P 0.05). In conclusion, the present study suggests that the myostatin signalling pathway is associated with glucocorticoid-induced muscle protein catabolism at the beginning of exposure. Myostatin is not a main pathway associated with the suppression of muscle protein synthesis by glucocorticoids.

An experiment was conducted to investigate the effects of dietary energy level on the performance and immune function of stressed broiler chickens (Gallus gallus domesticus). A total of 96 three-day-old male broiler chickens (Ross × Ross) were divided into two groups. One group received a high energy (HE) diet and the other group received a low energy (LE) diet for 7 days. At 5 days of age, the chickens from each group were further divided into two sub-groups and received one of the following two treatments for 3 days: (1) subcutaneous injection of corticosterone, twice per day (CORT group; 2 mg of CORT/kg BW in corn oil) and (2) subcutaneous injection of corn oil, twice per day (Control/Sham treatment group). At 10 days of age, samples of blood, duodenum, jejunum, and ileum were obtained. Compared with the other three groups, the LE group treated with CORT had the lowest average daily gain (ADG) and the poorest feed conversion ratio (FCR, P < 0.05). Furthermore, CORT treatment decreased the relative weight (RW) of the bursa independent of the dietary energy level, but it decreased the RW of the thymus only in the chickens fed the LE diet. By contrast, CORT administration decreased the RW of the spleen only in the chickens fed the HE diet (P < 0.05). The plasma total protein, albumin, tumor necrosis factor alpha, interleukin 2 and immunoglobulin G (IgG) levels were affected by the CORT treatment (P < 0.05); however, these factors were not significantly affected by the dietary energy level. Toll-like receptor-5 mRNA level was down-regulated by CORT injection in the duodenum and ileum (P < 0.05) and showed a trend of down-regulation in the jejunum (P=0.0846). The present study showed that CORT treatment induced immunosuppressive effects on the innate immune system of broiler chickens, which were ameliorated by consumption of higher dietary energy.

The chicken immune system is immature at the time of hatching. The development of the respiratory immune system after hatching is vital to young chicks. The aim of this study was to investigate the effect of dietary vitamin A supplement levels on respiratory mucin and IgA production in chicks. In this study, 120 one-day-old broiler chicks were randomly divided into 4 groups consisting of three replicates of 10 broilers and subjected to dietary vitamin A supplement levels of 0, 1,500, 6,000, or 12,000 IU/kg for seven days. Compared with control birds, vitamin A supplementation significantly increased the mucin and IgA levels in the bronchoalveolar lavage fluid (BALF) as well as the IgA level in serum. In the lungs, vitamin A supplementation downregulated TNF-α and EGFR mRNA expression. The TGF-β and MUC5AC mRNA expression levels were upregulated by vitamin A supplementation at a dose of 6,000 IU/kg, and the IL-13 mRNA expression level was increased at the 12,000 IU/kg supplement level. Vitamin A deficiency (control) significantly decreased the mRNA expression levels of MUC2, IgA, EGFR, IL-13 and TGF-β in trachea tissue. Histological section analysis revealed that the number of goblet cells in the tracheal epithelium was less in the 0 and 12,000 IU/kg vitamin A supplement groups than in the other groups. In conclusion, vitamin A deficiency suppressed the immunity of the airway by decreasing the IgA and mucin concentrations in neonatal chicks. This study suggested that a suitable level of vitamin A is essential for the secretion of IgA and mucin in the respiratory tract by regulating the gene expression of cytokines and epithelial growth factors.

Tin oxide SnO2 films were prepared by RF magnetron sputtering. The effects of oxygen partial pressure percentage on the SnO2 property have been investigated to obtain relatively high-resistivity SnO2 films which could be used as buffer layers to optimize the performance of CdTe/CdS solar cells. The oxygen partial pressure percentage varied in the range of 1%~10%. The results show that the introduction of oxygen would suppress the deposition and growth of SnO2 films. Electrical measurement suggests that the film resistivity decreases with the increase of oxygen pressure. The SnO2 films with resistivity of 232 Ω cm were obtained in pure Ar atmosphere. All SnO2 films fabricated with different oxygen partial pressure percentage have almost the same optical band gap.

The loss caused by the failure of production equipment, especially power machinery equipment, has increased greatly, and the maintenance cost has also increased accordingly. When the low-speed and heavy-duty equipment fails, its fault signal generally shows the characteristics of non-stationary and non-periodic. The loss caused by the failure of production equipment, especially power machinery equipment, has increased greatly, and the maintenance cost has also increased accordingly. Some large-scale machinery has become the most basic equipment in modern large-scale production equipment. In the event of failure of these devices, local or even entire production lines will be discontinued. The earlier the fault is discovered, the more helpful the safe and reliable operation of the mechanical equipment. In order to ensure the normal production order, it is urgent to carry out research work on the micro-shock feature extraction of low-speed heavy-duty equipment failure. Based on the wavelet packet analysis method, this paper analyzes the types and characteristics of low-speed heavy-duty equipment faults. The fault signal data acquisition and analysis of low-speed heavy-duty equipment can realize the analysis of the time domain and frequency domain of the signal. Confocal laser scanning microscopy has been extended to the field of optical microscopy, including confocal fluorescence microscopy, confocal scanning fiber microscopy, color confocal laser microscopy, confocal grating optical microscopy and so on. It will soon become an important tool for semiconductor material testing and biomedical diagnosis. Scanning near field optical microscopy (SNOM) and surface plasmon resonance (SPR) are used to study the surface plasmon resonance (SPR) of gold films. A novel Kretschmann type surface plasmon resonance coupling device with unique structure was designed and built.
