Objectives: To investigate the efficacy and safety of an improved ultrasound-guided pulsed radiofrequency (PRF) and nerve block (NB) for patients with pudendal neuralgia (PN). Methods: This retrospective analysis included 88 adults with PN treated in the Pain Department of Southwest Hospital from November 2011 to June 2021, with treatment including NB (n = 40) and PRF (n = 48). The primary outcome variable was pain severity, measured by a standardized visual analog scale (VAS). VAS values were collected at 1, 3, 7, and 14 days and 1 and 3 months after patients were treated with NB or PRF. Results: Compared with patients treated with NB (n = 40) and those treated with PRF (n = 48), no significant difference in pain reduction was observed in the short term (p = 0.739 and 0.981, at 1 and 3 days, respectively); however, in the medium and long term (1 to 3 months), there were statistically significant improvements in the PRF group over the NB group (p < 0.001). Moreover, it was noted that the average pain severity of primary PN and PN due to sacral perineurial cyst was significantly reduced with PRF therapy in the medium and long term when compared to other secondary PNs, including surgery, trauma, and diabetes. Discussion: The ultrasound-guided, improved, and innovative PRF/NB puncture path technique allows for gentler stimulation and faster identification of the pudendal nerve. The PRF technique may provide better treatments for primary PN and sacral perineurial cyst causing secondary PN in the medium and long term.

Dynamical spin-structure factor (DSF) contains fingerprint information of collective excitations in interacting quantum spin systems. In solid state experiments, DSF can be measured through neutron scatterings. However, it is in general challenging to compute the spectral properties accurately via many-body simulations. Currently, quantum simulation and computation constitute a thriving research field, which are believed to provide a very promising platform for simulating quantum many-body systems. In this paper, we establish a link between the many-body dynamics and quantum simulations by studying the nonequilibrium DSF (nDSF) measured on direct product states, which are accessible in contemporary quantum simulators with Rydberg atoms, superconducting qubits, etc. Based on the many-body calculations of transverse field Ising chains, we find the nDSF can be used to sensitively probe the multispinon continua associated with the two-spinon creation and spinon-antispinon process, etc. Moreover, we further demonstrate that the low-energy spinons can be confined—forming spinon bound states—under a finite longitudinal field. Our results pave the way of quantum simulation and manipulation of fractional excitations in highly entangled quantum many-body systems.

Objective: To establish an animal model consistent with the occurrence and development of non-alcoholic fatty liver disease (NAFLD) with which to assess the effects of a classical traditional Chinese medicine formula known as Dachaihu Decoction (DD) on NAFLD. Methods: Sixty rats were randomized into four groups: control, model, pioglitazone hydrochloride (PH) and DD in equal. NAFLD was produced via administration of a high-fat high-sugar diet for 16 weeks in all but the control group. From the 13th week, a solution of PH or DD prepared with water was delivered via intragastric administration to the PH and DD groups; the remaining two groups received an equivalent volume of distilled water. Twelve hours from the last administration, we selected eight rats from each group in random. After anesthetization, the abdominal aorta blood and liver tissues were collected. The morphological changes were observed and the levels of alanine aminotransferase (ALT), aspartate aminotransferase (AST), total cholesterol (TC), triglyceride (TG), low-density lipoprotein cholesterol (LDL-C), high-density lipoprotein cholesterol (HDL-C), fasting plasma glucose (FBG), transforming growth factor-β1 (TGF-β1), tumor necrosis factor-α (TNF-α), toll-like receptor-4 (TLR4), and nuclear factor-kappa B (NF-кB) were tested. Results: Compared with the control group, the levels of serum ALT, AST, TC, TG, LDL-C and FBG, and TGF-β1, TNF-α, TLR4, NF-кB in the model group all showed significant increases (P

A case of illegal vaccine sales in Shandong province, China, (hereinafter, the incident), which caused a lack of confidence among vaccination recipients and public panic, was uncovered in March 2016. We conducted a study comprising two cross-sectional surveys: at two months (May 2016) and seven months (October 2016) after the incident. The study aimed to evaluate the impact on immunizations; investigate the variation of the immunization coverage of the National Immunization Program Vaccines (NIPV) and the sales volume growth rate of Category II vaccines; and understand the reasons for non-vaccination and perspectives on immunization. The immunization coverage of NIPV decreased by 5.6 percentage points in the first survey, with a decline of 11.1 in the region of the incident, and decreased by 0.6 in the second survey compared to same period in 2015. The sales volume growth rate of Category II vaccines decreased by 25.8% in the study area and by 48.8% in the region of the incident in April 2016 compared to April 2015. Overall, 15.8% of respondents in the first survey and 7.0% in the second survey did not vaccinate their children according to the NIPV schedule because of the incident (χ2 = 78.463, P < 0.05). The vaccination was likely affected by the incident in varying degrees, especially in the involved region and particularly in relation to Category II vaccines. Overall, 34% of respondents avoided Category II vaccines for their children, indicating that it will take considerable time to eliminate the negative stigma associated with the incident.

Abstract Background The aim of this study was to introduce a novel intraoperative lateral ankle dislocation approach during surgical treatment for patients with unstable trimalleolar fractures involving posterior ankle comminuted fractures and compare its effects and safety with those with conventional approach. Methods From June 2006 to June 2014, 69 patients diagnosed as unstable trimalleolar fractures involving posterior ankle comminuted fracture were included in this study. The patients were divided into intraoperative dislocating ankle group (experimental group) and conventional treatment group (control group) according to surgical modalities. The following parameters including rate of primary healing, healing time, incidence of talus necrosis, incidence of post-traumatic arthritis, functional outcomes according to Baird-Jackson classification system, and any possible complications in two groups were recorded and compared. Results There were no significant differences regarding the rate of primary healing, healing time and the rate of talus necrosis in two groups (P > 0.05). The incidence of post-traumatic arthritis in experimental and control group were 0 and 24.24% (P = 0.0006), respectively. The rate of excellent and good outcomes were achieved in 91.67% in experimental group and 72.73% in control group (P = 0.038), respectively. Conclusions The findings suggest that the intraoperative ankle dislocation approach appears to be a promising surgical option for unstable trimalleolar fractures involving posterior ankle comminuted fracture because it can provide better functional outcomes and lower incidence of post-traumatic arthritis while not compromising primary healing and healing time.

As the convenient outlet to the Bo Sea and the major region of economic development in the Yellow River Basin, Shandong Province in China has undergone large changes in land use/land cover (LULC) in the past two decades with rapid urbanization and population growth. The analysis of the LULC change patterns and its driving factors in the Shandong section of the Yellow River Basin can provide a scientific basis for rational planning and ecological protection of land resources in the Shandong section of the Yellow River Basin. In this manuscript, we analyzed the spatial pattern of LULC and its spatial and temporal changes in the Shandong section of the Yellow River Basin in 2000, 2010, and 2020 by using the random forest classification algorithm with the Google Earth Engine platform and multi-temporal Landsat TM/OLI data. The driving factors of LULC changes were also quantified by the factor detector and interaction detector in the geodetector. Results show that in the past two decades, the LULC types in the study area are mainly farmland and construction land, among which the proportion of farmland area has decreased and the proportion of construction land area has increased from 19.4% to 29.7%. Based on the results of factor detector, it can be concluded that elevation, slope, and soil type are the key factors affecting LULC change in the study area. The interaction between elevation and slope, slope and soil type, and temperature and precipitation has strong explanatory power for the spatial variation of LULC change in the study area. The research results can provide data support for ecological environmental protection, sustainable, and high-quality development of the Shandong section of the Yellow River Basin, and help local governments take corresponding measures to achieve coordinated and sustainable socioeconomic and environmental development.

The utilization of livestock waste has attracted increasing attention in recent years. The presence of high levels of heavy metals is a major obstacle to the utilization of biogas as a fertilizer resource. In this study, the heavy metal contents in biogas residue, slurry, and discharged sewage from three representative farms of gooseries, henneries, and dairy farms in the Yangtze River Basin were investigated and assessed. The results demonstrated that heavy metals, including Cd, Mn, As, Cu, Pb, Cr, Zn, etc., could be detected in all biogas residues, with significantly different contents between farm types (p < 0.005). Specifically, biogas residues from the goosery and the dairy farms met “China’s Organic Fertilizer Standards” (COF Standards); however, Cd concentrations in biogas residues from hennery farms exceeded the limits by five times. The concentrations of Cd and Pb in biogas slurries from all of the farms exceeded the limits of the “China Farmland Irrigation Water Quality Standard” (CFIWQ Standard). In particular, the Pb concentrations in biogas slurry from the dairy farms exceeded the limits by 29 times, and the discharged sewage from all three farm types complied with the comprehensive sewage discharge standards in China; however, only that from the goosery farms was suitable for irrigation. Thus, it is recommended to increase the feed selection, biogas engineering, and biological-purification-supporting technology, and to carry out regular sampling inspections of the biogas residue, slurry, and discharged sewage for heavy metals, so that environmental and crop pollution risks can be reduced when they are used as sources of nutrients for eco-friendly agriculture.

In recent years, with the frequent global occurrence of harmful algal blooms, the use of plant allelopathy to control algal blooms has attracted special and wide attention. This study validates the possibility of turning water dropwort into a biological resource to inhibit the growth of harmful Microcystis aeruginosa blooms via allelopathy. The results revealed that there were 33 types of allelopathic compounds in the water dropwort culture water, of which 15 were phenolic acids. Regarding water dropwort itself, 18 phenolic acids were discovered in all the organs of water dropwort via a targeted metabolomics analysis; they were found to be mainly synthesized in the leaves and then transported to the roots and then ultimately released into culture water where they inhibited M. aeruginosa growth. Next, three types of phenolic acids synthesized in water dropwort, i.e., benzoic, salicylic, and ferulic acids, were selected to clarify their inhibitory effects on the growth of M. aeruginosa and their mechanism(s) of action. It was found that the inhibitory effect of phenolic acids on the growth of M. aeruginosa increased with the increase of the exposure concentration, although the algae cells were more sensitive to benzoic acid than to salicylic and ferulic acids. Further study indicated that the inhibitory effects of the three phenolic acids on the growth of M. aeruginosa were largely due to the simultaneous action of reducing the number of cells, damaging the integrity of the cell membrane, inhibiting chlorophyll a (Chl-a) synthesis, decreasing the values of F0 and Fv/Fm, and increasing the activity of the antioxidant enzymes (SOD, POD, and CAT) of M. aeruginosa. Thus, the results of this study indicate that both culture water including the rich allelochemicals in water dropwort and biological algae inhibitors made from water dropwort could be used to control the growth of noxious algae in the future.

In this paper, the problem of optimal placement of a single unmanned aerial vehicle (UAV) is studied, where the objective is to minimize the total transmit power required to provide wireless coverage for indoor users. Two cases of practical interest are considered, and efficient solutions to the formulated problem under these cases are presented. In the first case, the minimum transmit power required to cover the indoor users based on the location that has the maximum path loss inside the building is studied. We derive a functional relationship between the placement of the UAV and the dimensions of the building. In the second case, we assume that the locations of indoor users are randomly distributed inside the building. We propose to use an iterative algorithm and an exhaustive algorithm to optimize the placement of the UAV. We show that the minimum transmit power in the second case is much smaller than the transmit power in the first case. However, the equipment cost in the second case is relatively high. We validate the analysis by simulations and demonstrate the effectiveness of the proposed algorithms under different cases.

Abstract Background Inhaled nanoparticles can deposit in the deep lung where they interact with pulmonary cells. Despite numerous studies on pulmonary nanotoxicity, detailed molecular mechanisms of specific nanomaterial-induced lung injury have yet to be identified. Results Using whole-body dynamic inhalation model, we studied the interactions between aluminum oxide nanoparticles (Al2O3 NPs) and the pulmonary system in vivo. We found that seven-day-exposure to Al2O3 NPs resulted in emphysema and small airway remodeling in murine lungs, accompanied by enhanced inflammation and apoptosis. Al2O3 NPs exposure led to suppression of PTPN6 and phosphorylation of STAT3, culminating in increased expression of the apoptotic marker PDCD4. Rescue of PTPN6 expression or application of a STAT3 inhibitor, effectively protected murine lungs from inflammation and apoptosis, as well as, in part, from the induction of chronic obstructive pulmonary disease (COPD)-like effects. Conclusion In summary, our studies show that inhibition of PTPN6 plays a critical role in Al2O3 NPs-induced COPD-like lesions.

Traditional Chinese Medicine (TCM) preparations are often extracts of single or multiple herbs containing hundreds of compounds, and hence it has been difficult to study their mechanisms of action. Compound Kushen Injection (CKI) is a complex mixture of compounds extracted from two medicinal plants and has been used in Chinese hospitals to treat cancer for over twenty years. To demonstrate that a systematic analysis of molecular changes resulting from complex mixtures of bioactives from TCM can identify a core set of differentially expressed (DE) genes and a reproducible set of candidate pathways. We used in vitro cancer models to measure the effect of CKI on cell cycle phases and apoptosis, and correlated those phenotypes with CKI induced changes in gene expression. We treated two cancer cell lines with or without CKI and assessed the resulting phenotypes by employing cell viability and proliferation assays. Based on these results, we carried out high-throughput transcriptome data analysis to identify genes and candidate pathways perturbed by CKI. We integrated these differential gene expression results with previously reported results and carried out validation of selected differentially expressed genes. CKI induced cell-cycle arrest and apoptosis in the cancer cell lines tested. In these cells CKI also altered the expression of 363 core candidate genes associated with cell cycle, apoptosis, DNA replication/repair, and various cancer pathways. Of these, 7 are clinically relevant to cancer diagnosis or therapy, 14 are cell cycle regulators, and most of these 21 candidates are downregulated by CKI. Comparison of our core candidate genes to a database of plant medicinal compounds and their effects on gene expression identified one-to-one, one-to-many and many-to-many regulatory relationships between compounds in CKI and DE genes. By identifying genes and promising candidate pathways associated with CKI treatment based on our transcriptome-based analysis, we have shown that this approach is useful for the systematic analysis of molecular changes resulting from complex mixtures of bioactives.

Background: The purpose of this study was to reevaluate the efficacy of prophylactic cranial irradiation (PCI) in non-small cell lung cancer (NSCLC) with the most recent published data and to identify subgroups who may be more likely to gain benefit from PCI.Methods: We searched PubMed, Embase, and Cochrane databases for randomized trials comparing PCI with non-PCI in NSCLC patients. We pooled the data of randomized controlled trials and compared brain metastasis (BM) and overall survival (OS) between PCI group and non-PCI group.Results: Seven studies including 1,462 patients were eligible for the current meta-analysis. Compared to non-PCI group, PCI group achieved decreased BM (RR = 0.37, 95% CI: 0.26–0.52) but similar OS (HR = 1.01, 95% CI: 0.87–1.22). In subgroup analyses of BM, PCI decreased BM for subgroups by pathology (squamous cell carcinoma or non-squamous cell carcinoma) and local treatment modality (surgery or no surgery). However, PCI failed to reduce BM for patients with poor performance status (WHO 2–3). The incidence of PCI related toxicities was low and PCI was well-tolerated by the majority of NSCLC. Low grade neurocognitive function (NCF) decline was reported in NAVLT study and greater deterioration in immediate and delayed recall was reported in RTOG 0214. No significant difference in quality of life (QOL) after PCI was reported.Conclusion: PCI reduces the incidence of BM except for patients with poor performance status. However, PCI fails to prolong OS significantly for NSCLC. An individual patient data meta-analysis may identify patients that could achieve OS prolongation with PCI.

There are severe challenges for slurry pressure balance tunnel boring machine (TBM) tunnelling in sandy cobble soil of Beijing, Chengdu, and Lanzhou in China. And the problems caused by tunnelling from silty clay to sandy cobble stratum are more serious. With the change of stratum, the key parameters and surface settlement will change correspondingly. Controlling the key parameters and predicting the surface settlement accurately and efficiently is important for hazard mitigation and risk management. In this study, based on the Tsinghuayuan Tunnel project in Beijing, the key parameters and surface settlement while tunnelling from silty clay to sandy cobble stratum are studied. Firstly, the difference of key parameters while tunnelling in two different strata is analyzed. The analysis shows that immediate responses to changes in the stratum are recommended in order to ensure construction efficiency. Then, a refined 3D finite difference model is developed to simulate the slurry TBM tunnelling in different strata. For refined simulation, three key parameters obtained from measurement data were applied to the 3D models, and the simulation results were compared with the field data. Results show that the refined model has good performance in terms of the accuracy and efficiency. This study provides a good engineering practice reference for slurry TBM tunnelling in mixed strata.

Abstract Diesel exhaust particles (DEPs) are common airborne ultrafine particles (UFPs); however, few studies have examined their effects on the gastrointestinal tract. To investigate the interaction of gut microbiota and DEPs‐induced colonic injury, adult C57BL/6 mice are kept in whole‐body inhalation chambers and exposed to filtered room air (FRA) or DEPs (300 µg m−3) 1 h per day for 28 consecutive days. DEPs exposure results in colon epithelial injury with inflammatory cell infiltration and mucus depletion. Abundance of Lactobacillus in murine feces is transiently increased following 7‐day DEPs exposure and then decreased until the end of 28‐day exposure. A reduction of the colonic mucus layer thickness is observed in mice receiving gut microbiota from DEPs‐exposed mice. Mechanistically, RNA‐sequencing suggests disruption of the nitrogen metabolism pathway in DEPs‐exposed NCM460 cells. Upregulation of carbonic anhydrase 9 (CA9) expression levels is observed in epithelia following DEPs exposure both in vivo and in vitro. Oral administration of probiotics protects the mice against DEPS‐induced colon epithelial injury. The results strongly suggest the involvement of gut microbiota in response to DEPs exposure and subsequently epithelial injury in vivo. Supplementation with probiotic may be a potential way to protect against UFPs‐induced colon epithelial injury.

Herbal compatibility is the knowledge of which herbs to combine in traditional Chinese medicine (TCM) formulations. The lack of understanding of herbal compatibility is one of the key problems for the application and popularization of TCM in western society. Because of the chemical complexity of herbal medicines, it is simpler to begin to conduct compatibility research based on herbs rather than component plant secondary metabolites. We have used transcriptome analysis to explore the effects and interactions of two plant extracts (Kushen and Baituling) combined in Compound Kushen Injection (CKI). Based on shared chemical compounds and in vitro cytotoxicity comparisons, we found that both the major compounds in CKI, and the cytotoxicity effects of CKI were mainly derived from the extract of Kushen (Sophorae flavescentis). We generated and analyzed transcriptome data from MDA-MB-231 cells treated with single-herb extracts or CKI and results showed that Kushen contributed to the perturbation of the majority of cytotoxicity/cancer related pathways in CKI such as cell cycle and DNA replication. We also found that Baituling (Heterosmilax yunnanensis Gagnep) could not only enhance the cytotoxic effects of Kushen in CKI, but also activate immune-related pathways. Our analyses predicted that IL-1β gene expression was upregulated by Baituling in CKI and we confirmed that IL-1β protein expression was increased using an ELISA assay. Altogether, these findings help to explain the rationale for combining Kushen and Baituling in CKI, and show that transcriptome analysis using single herb extracts is an effective method for understanding herbal compatibility in TCM.

Abstract Particulate matter (PM) exposure has been associated with intestinal disorders. Therefore, there is an urgent need to understand the precise molecular mechanism involved and explore potential prevention strategies. In this study, inhaled PM is shown to activate inflammatory pathways in murine colon. In a panel study, it is found that ambient PM levels are significantly associated with elevated number of fecal white blood cells in healthy subjects. Acting as a promoter, PM exposure accelerates chemical carcinogenesis‐induced colonic tumor formation in a murine model. Mechanistically, RNA‐seq assays suggest activation of phosphoinositide 3‐kinase (PI3K)/AKT cascades in chronically PM‐exposed human colon mucosal epithelial cells. Ablation of up‐stream driver fibroblast growth factor receptor 4 (FGFR4) effectively inhibits inflammation and neoplasia in PM‐exposed murine colons. Notably, dietary curcumin supplement is shown to protect against PM‐induced colonic injuries in mice. Collectively, these findings identify that PM exposure accelerates colonic tumorigenesis in a PI3K/AKT‐dependent manner and suggests potential nutrient supplement for prevention.

Traditional Chinese Medicines are promising sources of new agents for controlling cancer metastasis. Compound Kushen Injection (CKI), prepared from medicinal plants Sophora flavescens and Heterosmilax chinensis, disrupts cell cycle and induces apoptosis in breast cancer; however, effects on migration and invasion remained unknown. CKI, fractionated mixtures, and isolated components were tested in migration assays with colon (HT-29, SW-480, DLD-1), brain (U87-MG, U251-MG), and breast (MDA-MB-231) cancer cell lines. Human embryonic kidney (HEK-293) and human foreskin fibroblast (HFF) served as non-cancerous controls. Wound closure, transwell invasion, and live cell imaging showed CKI reduced motility in all eight lines. Fractionation and reconstitution of CKI demonstrated combinations of compounds were required for activity. Live cell imaging confirmed CKI strongly reduced migration of HT-29 and MDA-MB-231 cells, moderately slowed brain cancer cells, and had a small effect on HEK-293. CKI uniformly blocked invasiveness through extracellular matrix. Apoptosis was increased by CKI in breast cancer but not in non-cancerous lines. Cell viability was unaffected by CKI in all cell lines. Transcriptomic analyses of MDA-MB-231indicated down-regulation of actin cytoskeletal and focal adhesion genes with CKI treatment, consistent with observed impairment of cell migration. The pharmacological complexity of CKI is important for effective blockade of cancer migration and invasion.

The cell envelope structure of Gram-negative bacteria is unique, composed of two lipid bilayer membranes and an aqueous periplasmic space sandwiched in between. The outer membrane constitutes an extra barrier to limit the exchange of molecules between the cells and the exterior environment. Donnan potential is a membrane potential across the outer membrane, resulted from the selective permeability of the membrane, which plays a pivotal role in the permeability of many antibiotics. In this review, we discussed factors that affect the intensity of the Donnan potential, including the osmotic strength and pH of the external media, the osmoregulated periplasmic glucans trapped in the periplasmic space, and the displacement of cell surface charges. The focus of our discussion is the impact of Donnan potential on the cellular permeability of selected antibiotics including fluoroquinolones, tetracyclines, β-lactams, and trimethoprim.

With the rapid development of the social economy in China, numerous Chinese cities are facing high levels of particulate matter (PM) pollution problems. In this study, high-resolution ZY-3 images and GIS techniques were used to establish the emission inventory of total suspended particle (TSP), particulate matter 10 (PM10) and particulate matter 2.5 (PM2.5) from fugitive dust sources in May 2016, and a spatial grid of 3 km × 3 km resolution was established to demonstrate the spatial distribution of PM emission. Results showed that the total emissions of TSP, PM10 and PM2.5 in Zhengzhou city were 237.5 kt·a−1, 103.7 kt·a−1 and 22.4 kt·a−1, respectively. Construction dust source was the main fugitive dust emission source in Zhengzhou city—the TSP, PM10 and PM2.5 emission of which account for 76.42%, 89.68% and 88.39%, respectively, of the total emission, followed by road dust source and soil dust source. PM emission was higher in Zhongyuan, Huiji, Jinshui and Zhengdong New District, while Zhongmou, Xingyang, Dengfeng and other remote areas had low PM emissions. Compared to other Chinese cities or regions, the PM emission from the construction dust source was at a high level in Zhengzhou city, while the PM emissions from the soil dust source and road dust source were at moderate levels.

Neoadjuvant therapy has become the standard of care for locally advanced mid-low rectal cancer. Pathological complete response (pCR) can be achieved in 12%â38% of patients. Patients with pCR have the most favorable long-term outcomes. Intensifying neoadjuvant therapy and extending the interval between termination of neoadjuvant treatment and surgery may increase the pCR rate. Growing evidence has raised the issue of whether local excision or observation rather than radical surgery is an alternative for patients who achieve a clinical complete response after neoadjuvant therapy. Herein, we highlight many of the advances and resultant controversies that are likely to dominate the research agenda for pCR of rectal cancer in the modern era. The biological mechanisms behind health effects of air pollution have not been well known. Inflammation plays an important role in occurrence and development of a wide range of diseases. In this study, we assessed the effects of short-term exposure to ambient air pollution on systemic inflammatory biomarkers among 12 508 participants who underwent routine physical examination annually at the Hebei General Hospital in Shijiazhuang, China. For each participant, white blood cell count (WBC), lymphocytes, neutrophils and eosinophils were measured for two or three times during September 2016 to December 2018. Daily concentrations of nitrogen dioxide (NO _2 ), sulfur dioxide (SO _2 ), ozone (O _3 ) and particulate matter less than 2.5 µ m in aerodynamic diameter (PM _2.5 ) were interpolated to each district, where the participants worked. The linear mixed-effects regression with a constrained distributed lag model was applied to examine the associations between air pollution and inflammatory biomarkers during lag 0–14 d. It was observed that WBC, neutrophils and eosinophils [percent change (%Δ) and 95% confidence interval (95%CI)] significantly decreased by −0.07 (−0.11, −0.04), −0.08 (−0.12, −0.03) and −0.15 (−0.25, −0.05) at lag 14 d, associated with per 10 µ g m ^−3 increase in O _3 . WBC, lymphocytes and eosinophils (%Δ and 95%CI) significantly elevated by 0.08 (0.04, 0.12), 0.16 (0.11, 0.21) and 0.22 (0.10, 0.35) at lag 0 d, associated with per 10 µ g m ^−3 increase in PM _2.5 . This study reveals short-term effects of air pollution on systemic inflammatory biomarkers in routine blood test, which is helpful for further study to explore the biological mechanisms.

Signal transducer and activator of transcription 3 (STAT3) is a transcription factor that contributes to cancer progression through multiple processes of cancer development, which makes it an attractive target for cancer therapy. The IL-6/STAT3 pathway is associated with an advanced stage in colorectal cancer patients. In this study, we identified trichothecin (TCN) as a novel STAT3 inhibitor. TCN was found to bind to the SH2 domain of STAT3 and inhibit STAT3 activation and dimerization, thereby blocking STAT3 nuclear translocation and transcriptional activity. TCN did not affect phosphorylation levels of STAT1. TCN significantly inhibited cell growth, arrested cell cycle at the G0/G1 phase, and induced apoptosis in HCT 116 cells. In addition, the capacities of colony formation, migration, and invasion of HCT 116 cells were impaired upon exposure to TCN with or without IL-6 stimulation. In addition, TCN treatment abolished the tube formation of HUVEC cells in vitro. Taken together, these results highlight that TCN inhibits various cancer-related features in colorectal cancer development in vitro by targeting STAT3, indicating that TCN is a promising STAT3 inhibitor that deserves further exploration in the future.

It is crucial for agricultural production to identify the trigger that switches plants from vegetative to reproductive growth. Agricultural sustainability in semiarid regions is challenged by nitrogen (N) fertilizer overuse, inadequate soil water, and heavy carbon emissions. Previous studies focused on the short-term effects of a single application of N and water but have not investigated the long-term effects of different irrigation and N fertilizer regimens on crop yields and yield components. N application is routinely coupled with water availability, and crop yields can be maximized by optimizing both. We examined the growth of western wheatgrass [Pascopyrum smithii (Rydb.) Á. Löve], a temperate-region forage and turf grass, using multiple different combinations of N fertilizer [(NH4)2·CO3] and irrigation levels over 3 years to determine optimal field management. We conducted multifactorial, orthogonally designed field experiments with large sample sizes, and measured fertile tillers m-2 (Y1), spikelets/fertile tillers (Y2), florets/spikelet (Y3), seed numbers/spikelet (Y4), seed weight (Y5), and seed yield (Z) to study factors associated with the switch between vegetative and reproductive growth. Fertilization had a greater effect on seed yield and yield components than irrigation. Y1 had the strongest positive effect on Z, whereas Y5 had a negative effect on Z. Irrigation and fertilization affected Z, Y1, and Y5. Fertilizer concentrations were positively correlated with Z, Y1, and Y5, whereas irrigation levels were negatively correlated. Germination of seeds during the transportation or after prolonged storage naturally and inevitably decreases because of ageing, but germination potential can be partially restored with seed priming treatments. A novel attempt was made to investigate the effects of combined treatments and to optimize the conditions for naturally aged seeds of tall fescue (Festuca arundinacea Schreb.), orchardgrass (Dactylis glomerata L.) and Russian wild rye (Psathyrostachys juncea (Fisch.) Nevski) using an orthogonal activity level experimental design [factor A: Azospirillum brasilense concentration, factor B: three seed priming treatments (H2O, MgSO4 and H2O2) and factor C: different priming times]. Multivariate regression model analysis was applied to determine the interactive effects of pairwise factors (A and C) and to optimize experimental conditions. The results showed that the mixed treatments positively affected seed germination and seedling growth. The three seed priming treatments were the dominant factors for germination promotion, whereas the bacterial concentration had the largest effect on seedling growth and the activity of superoxide dismutase (SOD), especially root elongation. The malondialdehyde content was reduced, the activities of SOD, peroxidase and catalase were triggered, and ascorbate peroxidase activity was also affected by the co-treatment. The combined results of all determined attributes showed that A. brasilense bio-priming with H2O2 priming constituted the optimal combination. The optimal bacterial concentration of A. brasilense and the time of seed priming were 52.3 × 106 colony forming units (CFU) mL-1 and 17.0 h, respectively.

Diabetic neuropathic pain (DNP) is a common and serious complication of diabetic patients. The pathogenesis of DNP is largely unclear. The proinflammation proteins, CXCR4, and TNF-α play critical roles in the development of pain, while their relative roles in the development of DNP and especially its progression is unknown. We proposed that establishment of diabetic pain models in rodents and evaluating the stability of behavioral tests are necessary approaches to better understand the mechanism of DNP. In this study, Von Frey and Hargreaves Apparatus was used to analyze the behavioral changes of mechanical allodynia and heat hyperalgesia in streptozotocin-induced diabetic rats at different phases of diabetes. Moreover, CXCR4 and TNF-α of spinal cord dorsal and dorsal root ganglia (DRG) were detected by western blotting and immunostaining over time. The values of paw withdrawal threshold (PWT) and paw withdrawal latencies (PWL) were reduced as early as 1 week in diabetic rats and persistently maintained at lower levels during the progression of diabetes as compared to control rats that were concomitant with significant increases of both CXCR4 and TNF-α protein expressions in the DRG at 2 weeks and 5 weeks (the end of the experiments) of diabetes. By contrast, CXCR4 and TNF-α in the spinal cord dorsal horn did not significantly increase at 2 weeks of diabetes while both were significantly upregulated at 5 weeks of diabetes. The results indicate that central sensitization of spinal cord dorsal may result from persistent peripheral sensitization and suggest a potential reference for further treatment of DNP.

Soil strength isotropy and coaxiality of the principal axes of stress and plastic strain rate tensors are often assumed in modelling the soils around tunnels, in the conventional plastic theory. However, the complexity of the soil response during the excavation of tunnelling cannot be well exhibited. In this paper, a recently developed two-dimensional (2D) elastoplastic constitutive model incorporating both soil strength anisotropy and non-coaxiality is firstly reviewed and then implemented in the finite element platform ABAQUS through the user-defined material subroutine (UMAT). This model is then numerically applied to analyze tunnel excavation problems. Two case studies, i.e., centrifuge testing and field testing, are carried out and compared with the numerical simulations to investigate the influences of soil anisotropy and non-coaxiality on the stress paths of soils around the tunnel crown and the subsurface settlement induced by tunnel excavations. The results show that the representative soil elements around tunnel crown experience severe principal stress orientations. The predictions of normalized subsurface settlement troughs can be improved by considering initial soil strength anisotropy compared to the conventional Mohr–Coulomb model. A larger value of the non-coaxial coefficient results in a larger magnitude of the maximum vertical displacement. The normalized subsurface settlement troughs are better improved in the case of centrifuge testing than that of the field testing.

The application range of the earth pressure balance (EPB) shield has been expanded due to advances in optimization methods, one of which is the application of foam conditioning. This method is widely used in EPB tunnelling owing to its strong applicability in different hydrological and geological conditions. When applying the foam conditioning method under different circumstances, it is necessary to optimize the conditioning parameters. Therefore, it is necessary to propose a test procedure for evaluating foam properties and the conditioning effect. This paper proposes a procedure to assess foaming agents by the mixing test and microscopic observation of the foam and a procedure that combines the slump test, compression test, and shear test to assess the foam-conditioned soil and determine the optimal parameters of conditioning. The test method is introduced and performed on fully weathered granite from Guangzhou Metro Line 21. The test results demonstrate that the foam injection ratio and pressure and type of the foaming agent all influence the performance of the conditioned soil. Moreover, the suggested conditioning scheme is proposed, and the application of the scheme can improve the tunnelling efficiency.

Background/Aims: Recurrent gene mutation has been identified by the analysis of exonic DNA from lung adenocarcinoma, but its progression has not been extensively profiled. The investigation of the mutational landscape of tumors provides new insights into cancer genome evolution and further discovers the interplay of somatic mutation, adaptation of clones to their environment and natural selection. Cancer development involves cycles of genomic damage, epigenetic deregulation, and increased cellular proliferation that eventually culminate in the carcinoma phenotype. Methods: Comparative whole exome sequencing of both primary and metastatic tumor tissues from four patients of stage IV lung adenocarcinoma patients with chest wall metastasis was performed. Both primary and metastatic tumors were diagnosed through biopsy followed by surgical resection. All tumor specimens were cut into several pieces to assess potential heterogenic clones within the tumor tissue. Adjacent normal lung tissue was also obtained to provide germline mutation background. Results: By modeling and analyzing progression of the cancer metastasis based on non-synonymous variants, we defined the extent of heterogeneity of cancer genomes and identified similar cancer evolution pattern in the four patients: metastasis was an early event occurring right after the primary cancer formation and evolution in the metastatic tumor was continuously and simultaneously in progression with that in the primary tumor. By characterizing the clonal hierarchy of genetic lesions, we further charted a pathway of oncogenic events along which genes may drive lung adenocarcinoma metastasis, such as TAS2R31 and UMODL1, involving in G-protein coupled receptor protein signaling pathway. Conclusion: The candidate genes identified in this study may become targets for the treatment of lung adenocarcinoma metastasis.

OBJECTIVES:To examine the factors associated with the routine immunization status of children aged 2-3 years in China for gaining a better understanding of the Expanded Program on Immunization and to provide evidence for formulating specific strategies to guide the allocation of health resources. METHODS:We analyzed data from 45095 children aged 2-3 years in the 2013 National Immunization Coverage Survey to identify the sociodemographic and provider-associated factors affecting the full immunization status of children. Univariate and multiple logistic regression analyses were performed. RESULTS:The immunization rate for children aged 2-3 years ranged from 95.9% (diphtheria and tetanus toxoid with pertussis vaccine, 4th dose) to 99.5% (Japanese encephalitis vaccine, 1st dose) and was 93.1% for full immunization. In terms of sociodemographic factors, male children [adjusted OR (AOR): 1.115; 95% confidence interval(CI):1.016-1.222], minority children (AOR: 1.632; 95% CI: 1.457-1.828), children of fathers with less than high school education (AOR: 1.577; 95% CI: 1.195-2.081), those born at home (AOR: 4.655; 95% CI: 3.771-5.746), those who immigrated from an adjacent county (AOR: 2.006; 95% CI: 1.581-2.546), and those living in urban-rural fringe areas (AOR: 1.807; 95% CI: 1.475-2.214) or mountainous areas (AOR: 1.615; 95% CI: 1.437-1.814) had significantly increased odds of not being fully immunized. In terms of provider-associated factors, administration of vaccines at home (AOR: 2.311; 95% CI: 1.316-4.059), household reminders (AOR: 2.292; 95% CI: 1.884-2.789), and travel time to vaccination providers of >40 minutes (AOR: 1.622; 95% CI: 1.309-2.010) were negatively associated with immunization rates. In addition, compared to 3-year-old years, 2-year-old children (AOR: 1.201; 95% CI: 1.094-1.318) were less likely to be fully immunized. CONCLUSIONS:All included factors except maternal education level and distance from home to vaccination providers significantly affected immunization rates. Appropriate reminders and accessibility of immunization services played key roles in improving the immunization status. More attention to high-risk groups identified in this study may reduce the disparities in routine childhood immunization in China.

miR-362 is a recently discovered member of the microRNA family, and it modulates a variety of physical activities and plays an important role in the occurrence and development of many tumors. However, the biological functions of hsa-miR-362-5p in non-small-cell lung carcinoma (NSCLC) are unknown. Transwell assay and colony formation were used to determine the migration, invasion, and proliferation of NSCLC cells in vitro. A subcutaneous tumor model in nude mice was established to detect NSCLC tumor growth in vivo. The direct binding of miR-362 to the 3′UTR of Semaphorin 3A (Sema3A) was confirmed by luciferase reporter assay. In this study, we found that the level of miR-362 was higher in NSCLC tissues than in adjacent normal tissues and that the level of miR-362 expression was also elevated in five NSCLC cell lines (A549, 95-D, H1299, H292, and H460) relative to a human normal lung epithelial cell line (BEAS2B). Furthermore, miR-362 promoted NSCLC cell invasion, migration, and colony formation in vitro and tumor formation in vivo. Next, we identified the miR-362 target gene Sema3A, which is significantly correlated with metastasis. Sema3A expression was increased in normal tissues relative to NSCLC tissues. This result is consistent with the fact that miR-362 expression is negatively correlated with Sema3A expression in clinical tissue samples and indicated that miR-362 can regulate Sema3A expression in NSCLC cells and consequently affect NSCLC invasion, migration, and colony formation. Taken together, these findings on the newly identified miR-362/Sema3A axis elucidate the molecular mechanism of NSCLC invasion and migration and could lead to a potential therapeutic target in NSCLC treatment.

Major water-soluble ions were analyzed for two-year precipitation samples in Wanzhou, a typical urban site of the hinterland of Chinese Three Gorges Reservoir. The pH values of the precipitation were in the range of 4.0 to 8.3, and the volume-weighted mean (VWM) value was 5.0. The concentration order of anions and cations was as follows: SO42->NO3->Cl->F- and NH4+>Ca2+>Na+>K+>Mg2+, respectively. Good correlations were found between SO42- and NH4+, SO42- and Ca2+, NO3- and NH4+, and NO3- and Ca2+, implying their co-occurrence in the precipitation, most likely as (NH4)2SO4, (NH4)HSO4, NH4NO3, CaSO4, and Ca(NO3)2. The sum of all measured ions was 416.4 μeq L−1, indicating serious air pollution in Wanzhou. NH4+ and Ca2+ were the most important ions neutralizing the acidic compounds in the precipitation; their major sources included agricultural activity and crustal dust. Local anthropogenic activities, for example, coal burning and traffic related sources, contributed most of SO42- and NO3-. The equivalent concentration ratio of SO42-/NO3- was 4.5, indicating that excessive emission of sulfur was the main reason leading to the precipitation acidity in Wanzhou. However, this ratio was lower than the ratio (5.9) in 2000s in Wanzhou, indicating that the contribution of nitric acid to the acidity of precipitation was strengthening.

Ferrous shape memory alloy, Fe41Ni28Co17Al11.5Ta2.5B0.05, has shown large superelastic strain and strength in previous study. In the fabrication of this alloy, aging process is crucial for the formation of shape memory effect/superelasticity. However, its phase evolution on aging time is not clearly known. In this study, we systematically studied the phase diagram of this alloy on aging time. It is found that the unaged alloy shows a strain glass transition. With the aging time proceeding, the martensitic transformation gradually emerges. The phase diagram can be explained by the formation of coherent precipitates induced by aging. The heterogeneous strain between coherent precipitates and matrix is the driving force responsible for the emerging martensitic transformation. The generic explanation is supposed to be useful in martensitic transformation engineering for developing novel shape memory alloys from non-transforming materials.

Based on the dynamics of diagonal entropy (DE), we provide a nonequilibrium method to study the properties of many-body localization (MBL) transition including the critical point and the universality class. By systematically studying the dynamical behaviors of DE in the fully explored Heisenberg spin chain with quasiperiodic field, we demonstrate the DE method can efficiently detect the transition point W_{c} between the thermal and MBL phase. We further use the method to study the MBL transition in the isotropic XX-ladder model, showing W_{c}∼8.05. We also widely explore the XX-ladder model with various parameters. Our results indicate that the MBL transition in the Heisenberg model and the XX-ladder model belong to distinct universality classes according to the obvious difference between the scaling exponents. These results can be tested in ongoing quantum simulation experiments with larger qubit numbers, since the diagonal elements of the density matrix directly yielding the DE can be easily obtained by repeatedly running single-shot measurements.

Background: Hepatopulmonary syndrome (HPS) is a serious complication of advanced liver disease that is characterised by intrapulmonary vascular dilatation (IPVD) and arterial hypoxemia. Pulmonary vascular remodelling (PVR) is an important pathological feature of HPS, but the potential mechanisms underlying PVR remain undefined. Recent findings have established the essential role of changes in Annexin A2 (ANXA2) in controlling the phenotypic modulation of pulmonary artery smooth muscle cells (PASMCs) in PVR associated with HPS. However, the mechanism by which upstream signalling regulates ANXA2 is unclear. Methods: In the present study, computational analysis was used to predict which miRNA might target the 3´-untranslated region (3´-UTR) of the ANXA2 mRNA. Real-time PCR and western blotting were performed to study the level of correlation between ANXA2 and the differentiation marker with the predicted miRNAs in PASMCs stimulated with serum from normal rats or those with HPS. Functional analysis of the miRNA and a luciferase reporter assay were performed to demonstrate that the predicted miRNA suppressed ANXA2 expression by directly targeting the predicted 3´-UTR site of the ANXA2 mRNA. Results: Computational analysis predicted that miR-206 would target the 3´-UTR of ANXA2 mRNA. In HPS rat serum-stimulated PASMCs, the expression of miR-206 displayed an inverse correlation with ANXA2, while a positive correlation was observed with the phenotypic marker smooth muscle α-actin (SM α-actin). The miRNA functional analysis and luciferase reporter assay demonstrated that miR-206 effectively downregulated the expression of ANXA2 by binding to the 3´-UTR of the ANXA2 mRNA. Consistently, miR-206 effectively inhibited the HPS rat serum-induced phenotypic modulation and proliferation, while these effects were reversed in ANXA2-overexpressing PASMCs. Conclusion: This study demonstrates that miR-206 inhibits the HPS rat serum-induced phenotypic modulation and proliferation in PASMCs by down-regulating ANXA2 gene expression.

For Wireless Sensor Networks, energy efficiency is always a key consideration in system design. Compressed sensing is a new theory which has promising prospects in WSNs. However, how to construct a sparse projection matrix is a problem. In this paper, based on a Bayesian compressed sensing framework, a new adaptive algorithm which can integrate routing and data collection is proposed. By introducing new target node selection metrics, embedding the routing structure and maximizing the differential entropy for each collection round, an adaptive projection vector is constructed. Simulations show that compared to reference algorithms, the proposed algorithm can decrease computation complexity and improve energy efficiency.

This paper presents an agricultural management service system that aims to meet the needs of Internet of Things (IoT) information upgrades in China’s family farms. The proposed agricultural management service system consists of Near Field Communication (NFC) tags, in-field service nodes, and smartphones. NFC tags are used as the core identifier of various agricultural management elements. The in-field service node, which is based on a programmable system-on-chip with intellectual property cores (IP core), supports distributed agriculture device management and smartphone operations. Smartphones in the proposed system include the management assistant application (app) and management service app, which are designed for agricultural management support functions and agricultural management application requirements. Through this system, the needs of diverse agricultural management practices can be effectively satisfied by a unified system structure. The practical results show that the design can be used to construct diversified agricultural IoT information application service systems simply and effectively, and it is especially suitable for Chinese family farm operators who are implementing IoT information upgrades for smart agriculture.

Bias thermal sensitivity is a significant performance parameter of a silicon resonant accelerometer (SRA) and is normally used to evaluate the degree of engineering practicability. Theoretical analysis demonstrates that temperature-induced stress is the dominant factor that determines the bias temperature drift of the custom-designed SRA. To solve this issue, this paper presents an SRA embedded in an isolation frame with stress insensitive anchor that prevents the resonant beams suffering from the thermal stress along the sense axis and thus improving the bias stability. Moreover, a high sensitivity device is achieved by integrating the vibrating beams with the comb fingers without conventional additional mass design. The experimental results show that the nominal resonant frequency of the SRA is around 93 kHz with the sensitivity and nonlinearity of 223.7 Hz/g and 5.1‱. The thermal sensitivities of the two resonant beams are −27.6 ppm/°C and −28.8 ppm/°C, respectively, which can be considered as the results owing to temperature change of the Young’s modulus without the thermal stress effect. The bias thermal sensitivity and the stability (1σ) after compensation are tested to be approximately 0.7 mg/°C and 1 mg over the temperature range from −40 °C to 60 °C with ±80 g measurement range.

In this paper, an ocean wave measurement technique and a newly developed short-range K-band radar are tested. In previous work, the technique and its feasibility were studied based on numerical simulations and wave tank experiments, while its performance at sea was still unknown. Surface current, Stokes drift, and wave breaking can greatly complicate interpreting radar backscatters. The feasibility of the technique needed to be further investigated with sea experiments. Experiments were carried out at a stationary site and from a moving platform. The short-range K-band radar transmitted continuous wave and received backscatters at low-grazing angles. The Bragg-scattering from the radar’s effective footprint dominated the backscatters. The Doppler shift frequency of the Bragg-scattering was attributed to the phase velocity of Bragg waves and modulated by the surface motions induced by current, Stokes drift, platform, and gravity waves. These sources of the Doppler shift frequency were analyzed, and the components induced by wind waves were successfully retrieved and converted into wave spectra that were consistent with the measurements of wave rider buoy. The experimental investigation further validated the feasibility of using short-range K-band radar to measure ocean waves.

Ginkgolides are the major active component of Ginkgo biloba for inhibition of platelet activating factor receptor. An azide-alkyne Huisgen cycloaddition reaction was used to introduce a triazole nucleus into the target ginkgolide molecules. A series of ginkgolide-1,2,3-triazole conjugates with varied functional groups including benzyl, phenyl and heterocycle moieties was thus synthesized. Many of the designed derivatives showed potent antiplatelet aggregation activities with IC50 values of 5~21 nM.

This study explores the relationship between vicarious experiences of nature, environmental behavioral beliefs, environmental attitudes and adolescents' environmental behavior. Based on a sample of 1476 adolescents from five high schools in Jiangsu Province, China, we proposed a model that was supported by structural equation modelling analysis. The model suggests that vicarious experiences of nature are positively associated with adolescents' environmental behavior. Environmental attitudes and environmental behavioral beliefs play partial mediator roles in the relationship of vicarious experiences of nature and environmental behavior. In addition to the significant direct impact of vicarious experiences of nature on environmental behavior, indirect effects are also achieved through behavioral beliefs, environmental attitudes and the serial multiple mediation effect of the two variables. These research results imply that we must pay attention to the important role of vicarious experiences of nature in cultivating adolescents' environmental behavior. In addition, the theoretical and practical implications of this research are discussed, as well as the limitations and potential for future research.

Problems concerning moving target tracking are being proposed and applied widely currently along with the maturity of computer technology and the fast advancement of computer vision. As a very important area in computer vision, moving target tracking is widely used in real life, for example, interactivity between human and computer, and so on. In this paper, Kalman filter, as a highly efficient recursive filter, is the common video method employed to track moving object in video. The moving state of moving object can be estimated in accordance with the location of the moving target. The tracking video is decomposed into frames in the initialization period and every frame is made up of a piece of picture; then, the targets in each frame are identified by means of color recognition; next, position the moving target, identify the center coordinates; at last, the coordinate of the previous and current frames is inputted and the location of the moving target in the present frame is estimated by Kalman filter.

Background and objective It has been proved that cyclooxygenase-2 (COX-2) is a key factor in lung cancer oncogenesis. COX-2 can be induced by a number of cytokines and growth factors and can be regulated by the JAK/STAT signaling pathway. Inhibiting the expression of COX-2 can prevent the development of lung cancer. The aim fo this study is to investigate whether the epidermal growth factor (EGF) can stimulate the signal transducers and activators of transcription 5 (STAT5) as well as to discover the effects of the STAT5 signaling pathway on the COX-2 in human lung adenocarcinoma A549 cells. Methods The phenomenon of STAT5 activation stimulated by the EGF was assayed through immunofluorescence and Western blot. The adenovirus containing the wild-type (WT)-STAT5 (AdWT-STAT5) plasmid, dominant-negative (DN)-STAT5 (Ad-CMV5Stat5aΔ740) plasmid, and STAT5 siRNA were transfected into A549 cells. The latter two groups were stimulated using EGF. Reverse transcriptase polymerase chain reaction was used to detect the mRNA expression of COX-2. Results STAT5 was not activated in A549 cells in vitro. EGF stimulation significantly increased the level of the p-STAT5 protein and induces the shuttling of p-STAT5 from the cytoplasm into the nucleus. STAT5 activation was crucial for the COX-2 expression induced by the EGF. STAT5 was required for COX-2 expression, but can mediated the effects of the COX-2 expression through pathways that were independent of transcriptional activation. Conclusion COX-2 expression is dependent on STAT5 phosphorylation. A second pathway does not require STAT5 phosphorylation.

Genetic predisposition increases the risk of familial breast cancer. Recent studies indicate that genetic predisposition for familial breast cancer can be ethnic-specific. However, current knowledge of genetic predisposition for the disease is predominantly derived from Western populations. Using this existing information as the sole reference to judge the predisposition in non-Western populations is not adequate and can potentially lead to misdiagnosis. Efforts are required to collect genetic predisposition from non-Western populations. The Egyptian population has high genetic variations in reflecting its divergent ethnic origins, and incident rate of familial breast cancer in Egypt is also higher than the rate in many other populations. Using whole exome sequencing, we investigated genetic predisposition in five Egyptian familial breast cancer families. No pathogenic variants in BRCA1, BRCA2 and other classical breast cancer-predisposition genes were present in these five families. Comparison of the genetic variants with those in Caucasian familial breast cancer showed that variants in the Egyptian families were more variable and heterogeneous than the variants in Caucasian families. Multiple damaging variants in genes of different functional categories were identified either in a single family or shared between families. Our study demonstrates that genetic predisposition in Egyptian breast cancer families may differ from those in other disease populations, and supports a comprehensive screening of local disease families to determine the genetic predisposition in Egyptian familial breast cancer.

We describe a technique to measure ocean wave period, height and direction. The technique is based on the characteristics of transmission and backscattering of short-range K-band narrow beam continuous wave radar at the sea surface. The short-range K-band radar transmits and receives continuous signals close to the sea surface at a low-grazing angle. By sensing the motions of a dominant facet at the sea surface that strongly scatters signals back and is located directly in front of the radar, the wave orbital velocity can be measured from the Doppler shift of the received radar signal. The period, height and direction of ocean wave are determined from the relationships among wave orbital velocity, ocean wave characteristics and the Doppler shift. Numerical simulations were performed to validate that the dominant facet exists and ocean waves are measured by sensing its motion. Validation experiments were conducted in a wave tank to verify the feasibility of the proposed ocean wave measurement method. The results of simulations and experiments demonstrate the effectiveness of the short-range K-band narrow beam continuous wave radar for the measurement of ocean waves.

Atrial myxoma is the most frequent tumor arising mainly in atrial septum and its origin remains uncertain. It has been reported that a subpopulation of stem-like cells are present in benign tumors and responsible for tumor initiation and maintenance. In this study, we investigated whether stem-like cells could contribute to the atrial cardiac myxoma. Immunohistology data confirmed that a population of cells bearing the surface markers CD19, CD45, and CD44 resided in a mucopolysaccharide-rich matrix of myxoma. Moreover, we isolated myxoma cells with phase-bright culture method and confirmed that myxoma derived cells express robust level of CD19, CD45, and CD44. Furthermore, the pluripotency of this population of cells also was validated by cardiomyocytes and smooth muscle cells differentiation in vitro. Our results indicate that primary cardiac myxoma may arise from mesenchymal stem cells with the ability to generate tumors with multilineage differentiation. In conclusion, this study for the first time verified that stem-like cells are present in atrial myxoma and this population of cells may have the capacity for myxoma initiation and progression.

A novel approach to the synthesis of 6-amino-7-hydroxyflavone (1) is described. Reaction in acetone of 2′,4′-dihydroxy-5′-nitroacetophenone and benzoyl chloride in the presence of potassium carbonate affords 3-benzoyl-7-hydroxy-6-nitroflavone, which is cleaved in 5% ethanolic potassium hydroxide to give 1-(2,4-dihydroxy-5-nitrophenyl)-3- phenyl-1,3-propanedione. The 1,3-diketone thus formed is then transformed into 7-hydroxy- 6-nitroflavone, followed by reduction to afford the title compound.

Toll-like receptors (TLRs) play a crucial role in the early defence against invading pathogens, yet our understanding of TLRs in marsupial immunity is limited. Here, we describe the characterisation of nine TLRs from a koala immune tissue transcriptome and one TLR from a draft sequence of the koala genome and the subsequent development of an assay to study genetic diversity in these genes. We surveyed genetic diversity in 20 koalas from New South Wales, Australia and showed that one gene, TLR10 is monomorphic, while the other nine TLR genes have between two and 12 alleles. 40 SNPs (16 non-synonymous) were identified across the ten TLR genes. These markers provide a springboard to future studies on innate immunity in the koala, a species under threat from two major infectious diseases.

Prostate cancer (PCa) is the most frequently diagnosed cancer for men in the developed world. Androgen receptor signaling pathway plays an important role in prostate cancer progression. Recent studies show that microRNA miR-124 exerts a tumor suppressive function in prostate cancer. However, the relationship between AR and miR-124 is unclear. In the present study, we found a negative feedback loop between AR and miR-124 expression. On one hand, miR-124 was a positively regulated target gene of the AR, on the other hand, overexpression of miR-124 inhibited the expression of AR. In addition, we found that miR-124-2 and miR-124-3 promoters were hypermethylated in AR-negative PCa cells. Furthermore, overexpression of miR-124 inhibited proliferation rates and invasiveness capacity of PCa cells in vitro, and suppressed xenograft tumor growth in vivo. Taken together, our results support a negative feedback loop between AR and miR-124 expression. Methylation of miR-124-2 and miR-124-3 may serve as a biomarker for AR-negative PCa cells, and overexpression of miR-124 might be of potential therapeutic value for the treatment of PCa.
