Gestating sows may be more susceptible to increasing dry bulb temperatures (T DB ) due to greater metabolic heat production and increased body mass, especially as gestation advances. However, there are few studies on the thermoregulatory and physiological responses of sows at differing gestation stages exposed to gradually increasing temperatures. The study objective was to determine the thermoregulatory and physiological responses of nonpregnant (n = 12; parity 3.27 ± 0.86), mid-gestation (59.7 ± 9.6 d pregnant, n = 12; parity 3.25 ± 0.83), and late-gestation (99.0 ± 4.8 d pregnant, n = 12; parity 3.33 ± 0.75) sows exposed to increasing TDB. Prior to the experiment (5.0 ± 0.7 d), jugular catheters were placed in all sows. During the experiment, the TDB was increased incrementally by 2.45 ± 0.43 °C every 60 min from 19.84 ± 2.15 to 35.54 ± 0.43 °C over 400 min, and relative humidity was recorded at 40.49 ± 18.57%. Respiration rate (RR), heart rate (HR), skin temperature, and vaginal temperature were measured, and blood samples were obtained via the jugular catheter every 20 min. Data were analyzed using PROC MIXED in SAS 9.4. RR increased at a lower TDB (P < 0.01) in late-gestation sows compared with mid-gestation and nonpregnant sows, but no differences were detected between mid-gestation and nonpregnant sows. Overall, late-gestation sows had greater RR (P < 0.01; 23 ± 2 breaths per min [ brpm ]) compared with mid-gestation (16 ± 2 brpm) and nonpregnant (15 ± 2 brpm) sows. Late-gestation sows had an overall greater HR (P < 0.01; 84 ± 5 beats per min [ bpm ]) than mid-gestation (76 ± 5 bpm) and nonpregnant (69 ± 5 bpm) sows. Late-gestation sows had overall reduced bicarbonate and total carbon dioxide levels (P = 0.02; 23.89 ± 1.97 and 25.41 ± 2.07 mmol/L, respectively) compared with mid-gestation (27.03 ± 1.97 and 28.58 ± 2.07 mmol/L, respectively) and nonpregnant (26.08 ± 1.97 and 27.58 ± 2.07 mmol/L, respectively) sows. Moreover, late-gestation sows had overall greater nitric oxide levels (P < 0.01; 248.82 ± 34.54 µM) compared with mid-gestation (110.47 ± 34.54 µM) and nonpregnant (41.55 ± 34.54 µM) sows. In summary, late-gestation sows appear to be more sensitive to increasing TDB as indicated by thermoregulatory and physiological responses when compared with mid-gestation or nonpregnant sows. Previous research demonstrates that supplementing 0.20% l -glutamine (GLN) in the diets of newly weaned and transported pigs improves growth rate to a similar extent as providing dietary antibiotics (AB). However, research comparing the effects of GLN vs. AB on intestinal physiology and the microbiome is limited. Therefore, the study objective was to compare the effects of supplementing nursery diets with GLN, AB, or no dietary antibiotics (NA) on intestinal physiology and the microbiome of pigs in a production environment following weaning and transport. Mixed-sex piglets (N = 480; 5.62 ± 0.06 kg body weight [ BW ]) were weaned (18.4 ± 0.2 d of age) and transported for 12 h in central Indiana, for two replicates, during the summer of 2016 and the spring of 2017. Pens were blocked by BW and allotted to one of the three dietary treatments (n = 10 pens/dietary treatment/replicate [8 pigs/pen]): AB (chlortetracycline [441 ppm] + tiamulin [38.6 ppm]), GLN (0.20% as-fed), or NA fed for 14 d. From day 14 to 34, pigs were fed common AB-free diets in two phases. On day 33, villus height:crypt depth tended to be increased (P = 0.07; 7.0%) in GLN and AB pigs vs. NA pigs. On day 33, glucagon-like peptide 2 (GLP-2) mRNA abundance was decreased (P = 0.01; 50.3%) in GLN and NA pigs vs. AB pigs. Crypt depth was increased overall on day 33 (P = 0.01; 16.2%) during the spring replicate compared with the summer replicate. Villus height:crypt depth was reduced (P = 0.01; 9.6%) during the spring replicate compared with the summer replicate on day 33. On day 13, tumor necrosis factor-alpha and occludin mRNA abundance was increased (P ≤ 0.04; 45.9% and 106.5%, respectively) and zonula occludens-1 mRNA abundance tended to be greater (P = 0.10; 19.2%) in the spring replicate compared with the summer replicate. In addition, AB pigs had increased (P = 0.01; 101.3%) GLP-2 mRNA abundance compared with GLN and NA pigs. Microbiome analysis indicated that on day 13, dietary treatment altered the microbiota community structure (P = 0.03). Specifically, the AB pigs tended to be distinct from both the NA and GLN pigs (P = 0.08), and Lactobacillus was increased nearly 2-fold in AB compared with NA pigs (q = 0.04) and GLN pigs (q = 0.22). In conclusion, GLN supplementation tended to improve some morphological markers of intestinal health similarly to AB pigs, while the microbiome composition in GLN pigs was more similar to NA pigs than AB pigs. All neurodegenerative diseases feature aggregates, which usually contain disease‐specific diagnostic proteins; non‐protein constituents, however, have rarely been explored. Aggregates from SY5Y‐APPSw neuroblastoma, a cell model of familial Alzheimer's disease, were crosslinked and sequences of linked peptides identified. We constructed a normalized "contactome" comprising 11 subnetworks, centered on 24 high‐connectivity hubs. Remarkably, all 24 are nucleic acid‐binding proteins. This led us to isolate and sequence RNA and DNA from Alzheimer's and control aggregates. RNA fragments were mapped to the human genome by RNA‐seq and DNA by ChIP‐seq. Nearly all aggregate RNA sequences mapped to specific genes, whereas DNA fragments were predominantly intergenic. These nucleic acid mappings are all significantly nonrandom, making an artifactual origin extremely unlikely. RNA (mostly cytoplasmic) exceeded DNA (chiefly nuclear) by twofold to fivefold. RNA fragments recovered from AD tissue were ~1.5‐to 2.5‐fold more abundant than those recovered from control tissue, similar to the increase in protein. Aggregate abundances of specific RNA sequences were strikingly differential between cultured SY5Y‐APPSw glioblastoma cells expressing APOE3 vs. APOE4, consistent with APOE4 competition for E‐box/CLEAR motifs. We identified many G‐quadruplex and viral sequences within RNA and DNA of aggregates, suggesting that sequestration of viral genomes may have driven the evolution of disordered nucleic acid‐binding proteins. After RNA‐interference knockdown of the translational‐procession factor EEF2 to suppress translation in SY5Y‐APPSw cells, the RNA content of aggregates declined by >90%, while reducing protein content by only 30% and altering DNA content by ≤10%. Heat stress (HS) during summer months can negatively affect reproductive efficiency and sow productivity. This study was designed to evaluate potential relationships between HS sensitivity and behavior with litter characteristics in replacement gilts. A total of 61 replacement gilts (108.8± 10.4 kg BW; 8 repetitions; 9d/repetition), were tested from February to May 2020 at the USDA-ARS Food Animal Behavior Laboratory in West Lafayette, IN. Pigs were housed (2/pen) in thermoneutral (TN; 22.6± 1.6°C) conditions and their behavior was recorded for 5 d. On d 6, gilts were subjected to an open field test and novel object test. Heart rate, number of escape attempts, and vocalizations were recorded during behavior testing. Vaginal temperature monitors were inserted to record body temperature (TB) every 15 min. Gilts were individually housed and exposed to cyclic HS (28.2± 0.97°C nighttime to 36.9± 1.9°C daytime; 46± 15.4% relative humidity) on d 7 to 9. Feed was provided ad libitum and feed intake was recorded daily. During the HS challenge, respiration rate (RR), skin (ear, shoulder, rump, and tail) temperature, and posture were recorded every 2 h from 0800 to 2000 h. Following the HS challenge, gilts were transported to the Purdue University swine farm, and gilts that displayed signs of estrus were artificially inseminated (253±29 d of age) between May and September 2020. At farrowing, litter characteristics were obtained and included total number of piglets born, total number of piglets born alive, average litter birth weight, total number of piglets weaned, and average weaning weight of the litter. Correlations between behavior, HS response, and litter characteristics were obtained using JMP 15 software. The preliminary analysis indicated no significant interactions (P < 0.05) between RR or TB with litter characteristics. Simple Summary: Exposure to thermal stress can negatively impact an animals' overall welfare, resulting in decreased body condition, slower growth rates, and in severe cases, mortality. Understanding the thermal comfort of pigs can help producers reduce thermal stress and improve the overall well-being of these animals. To understand pigs' thermal comfort, this study utilized temperature preference with weaned pigs by allowing them to select from a range of temperatures within a thermal apparatus. However, temperature preference is complicated given that a variety of factors can influence thermal comfort. Previous research has indicated that temperature preference is altered based on the number of individuals tested as this can alter their thermal comfort. Social aggregation, through huddling, results in greater heat conservation and animals find cooler temperatures more comfortable. Thus, this study looked at how social groups and different body weight could influence the temperature preference of pigs. Results showed that individual pigs preferred warmer temperatures compared to those in groups of 2 and 4, and that heavier pigs preferred cooler temperatures compared to medium- and lightweight pigs. This study demonstrates that a greater number of individuals perceive a cooler temperature as being within their comfort zone, whereas an individual does not have access to the thermal benefits of social aggregation. Housing pigs within their thermal comfort zone positively impacts productivity and performance. However, fundamental information on behavioral thermoregulatory responses of individual and group-housed pigs is meager. As a gregarious species, pigs prefer to be near one another, touching and often huddling. As pigs huddle together, they decrease their heat loss to the environment by decreasing exposed surface area and increasing mass. Additionally, pigs gain weight rapidly as they age. As an individual grows, their ability to withstand lower temperatures increases. We hypothesized that group size would alter pig thermal preference and that thermal preference would change based upon body weight. Thirty-six groups of pigs (n = 2 pigs/group) were tested in a factorial design based on group size (1, 2, or 4) and weight category (small: 5.20 ± 1.15 kg; medium: 8.79 ± 1.30 kg; and large: 13.95 ± 1.26 kg) in both sexes. Treatment groups were placed inside a chamber with a controlled thermal gradient (4.6 m × 0.9 m × 0.9 m; L × W × H) that ranged in temperature from 18 to 30 °C. Pigs habituated to the gradient for 24 h. The following 24 h testing period was continuously video recorded and each pig's location during inactivity (~70% daily budget) within the thermal apparatus was recorded every 10 min via instantaneous scan sampling. Data were analyzed using a GLM and log10 + 0.001 transformed for normality. Tukey tests and Bonferroni-corrected custom tests were used for post hoc comparisons. Peak temperature preference was determined by the maximum amount of time spent at a specific temperature. Both group size (p = 0.001) and weight category (p < 0.001) influenced the thermal location choice of pigs. Individual pigs preferred 30.31 °C, which differed from a group of 2 (20.0 °C: p = 0.003) and 4 pigs (20.0 °C: p < 0.001). The peak temperature preference of the small pigs (30.2 °C) differed from the large pigs (20.0 °C: p < 0.001) but did not differ from the medium-sized pigs (28.4 °C: p > 0.05). An intermediate-depth (1751 m) ice core was drilled at the South Pole between 2014 and 2016 using the newly designed US Intermediate Depth Drill. The South Pole ice core is the highestresolution interior East Antarctic ice core record that extends into the glacial period. The methods used at the South Pole to handle and log the drilled ice, the procedures used to safely retrograde the ice back to the National Science Foundation Ice Core Facility (NSF-ICF), and the methods used to process and sample the ice at the NSF-ICF are described. The South Pole ice core exhibited minimal brittle ice, which was likely due to site characteristics and, to a lesser extent, to drill technology and core handling procedures. [ABSTRACT FROM AUTHOR]

The Winkie Drill is an agile, commercially available rock coring system. The U.S. Ice Drilling Program has modified a Winkie Drill for subglacial rock and ice/rock interface coring, as well as drilling and coring access holes through ice. The original gasoline engine was replaced with an electric motor though the two-speed gear reducer and Unipress hand feed system were maintained. Using standard aluminum AW34 drill rod (for 33.5mm diameter core), the system has a depth capability of 120 m. The drill uses forward fluid circulation in a closed loop system. The drilling fluid is Isopar K, selected for favorable properties in polar environment. When firn or snow is present at the drill site, casing with an inflatable packer can be deployed to contain the drill fluid. The Winkie Drill will operate from sea level to high altitudes and operation results in minimal environmental impact. Significant upgrades to the Rapid Air Movement (RAM) Drill were developed and tested by the US Ice Drilling Program in 2016 through 2020 for the U.S. National Science Foundation. The design of the system leverages the existing infrastructure of the RAM Drill with the goal of greatly reducing the logistical burden of deploying the drill while maintaining the ability to drill an access hole in firn and ice to 100m in 40 min or less. In this paper, characteristics of the drill are described, along with a description of the drill performance during the testing at Raven Camp in Greenland and at WAIS Divide Camp in Antarctica. Methionine restriction (MR) dramatically extends the healthspan of several organisms. Methionine-restricted rodents have less age-related pathology and increased longevity as compared with controls, and recent studies suggest that humans might benefit similarly. Mechanistically, it is likely that the decreased IGF-1 signaling that results from MR underlies the benefits of this regimen. Thus, we hypothesized that interventions that decrease IGF-1 signaling would also produce MR-like healthspan benefits. Selenium supplementation inhibits IGF-1 signaling in rats and has been studied for its putative healthspan benefits. Indeed, we show that feeding mice a diet supplemented with sodium selenite results in an MR-like phenotype, marked by protection against diet-induced obesity, as well as altered plasma levels of IGF-1, FGF-21, adiponectin, and leptin. Selenomethionine supplementation results in a similar, albeit less robust response, and also extends budding yeast lifespan. Apoptosis has been suggested as the first step in the process of conversion of muscle into meat. While a potential role of apoptosis in postmortem proteolysis has been proposed, the underlying mechanisms by which metabolome changes in muscles would influence apoptotic and proteolytic process, leading to meat quality variation, has not been determined. Here, apoptotic and proteolytic attributes and metabolomics profiling of longissimus dorsi (LD) and psoas major (PM) muscles in pigs from two different production cycles (July–Jan vs. Apr–Sep) were evaluated. PM showed higher mitochondrial membrane permeability (MMP), concurrent with less extent of calpain-1 autolysis and troponin T degradation and higher abundance of HSP27 and αβ-crystallin compared to LD (P < 0.05). Apr–Sep muscles showed concurrence of extended apoptosis (indicated by higher MMP), calpain-1 autolysis and troponin T degradation, regardless of muscle effects (P < 0.05). Metabolomics profiling showed Apr–Sep muscles to increase in oxidative stress-related macronutrients, including 6-carbon sugars, some branched-chain AA, and free fatty acids. Antioxidant AA (His and Asp) and ascorbic acid were higher in July–Jan (P < 0.05). The results of the present study suggest that early postmortem apoptosis might be positively associated with pro-oxidant macronutrients and negatively associated with antioxidant metabolites, consequently affecting meat quality attributes in a muscle-specific manner. Lactating sows are among the most heat stress sensitive population in the swine herd. Exposure to high ambient temperatures causes a marked decrease in production measures and welfare in lactating sows and may have a negative downstream impact on piglet growth due to impaired sow lactogenesis. Genomic selection for improved heat stress (HS) tolerance may be a viable method to reduce the negative impacts of HS on lactating sows and their offspring. However, selection for HS tolerance is generally associated with decreased productivity. Therefore, the study objective was to evaluate the impact of genomic selection for thermotolerance on indirect measures of sow milk production under HS conditions. We hypothesized that heat tolerant sows would have a decrease in indirect measures of milk production when compared with heat sensitive sows. A total of 20 multiparous lactating sows (Large White x Landrace; parity = 4.85 ± 0.75) divergently genomically selected for heat tolerance (TOL; n = 11) or heat sensitivity (SEN; n = 9) were subjected to cyclic HS temperatures (28 to 32° C) from day 2.5 ± 1.0 post-farrowing until weaning (d 21.3 ± 1.1). On d 4, 8, 14, and 18 of lactation, indirect calorimetry was used to estimate total heat production (THP) on an individual sow basis following previously published methods by our group. In addition, sow respiration rate (RR) was measured daily at 0800, 1200, 1600, and 2000 h. All data were analyzed using PROC GLIMMIX in SAS 9.4 with individual sow included as the experimental unit. It was determined that TOL sows had an overall increase (P = 0.04; 5.68 ± 0.24 kcal·sow· kg BW0.75(-1)) in THP when compared with SEN sows (4.74 ± 0.18 kcal·sow· kg BW0.75(-1)). A treatment by day effect was observed where THP was greater (P < 0.01; 88%) for TOL sows on d 4 of lactation when compared with SEN sows. Preliminary evidence suggests that TOL sows had an overall increase (P = 0.01) in RR (78 ± 5 bpm) when compared with SEN sows (73 ± 5 bpm). Taken together, these data suggest that genomic selection methods used to increase lactating sow thermotolerance may have had a positive impact on indirect measures of lactogenesis under HS conditions. In utero heat stress (IUHS) increases the energy requirements of pigs during postnatal life, and this may compound weaning and transport stress. The study objective was to evaluate and mitigate the negative effects of IUHS following weaning and transport through the provision of a nutrient-dense (ND) nursery diet formulated to meet the greater energy requirements of IUHS pigs during the first 14 d post weaning and transport. Twenty-four pregnant gilts were exposed to thermoneutral (TN; n = 12; 17.5 ± 2.1 °C) or heat stress (HS; n = 12; cycling 26 to 36 °C) conditions for the first half of gestation (day 6 to 59) and then TN conditions (20.9 ± 2.3 °C) until farrowing. Nine TN gilts and 12 HS gilts produced litters. At weaning (16.2 ± 0.4 d), mixed sex piglets (N = 160; 4.78 ± 0.15 kg body weight [BW]) were transported (loading + transport + unloading) for 11 h 40 min. Following transport, piglets were blocked into pens (n = 4 pigs/pen) by in utero and dietary treatments: in utero thermoneutral (IUTN) + control (C) diet (n = 10 pens), IUTN + ND (n = 10 pens), IUHS + C (n = 10 pens), and IUHS + ND (n = 10 pens). Treatment diets were fed from day 1 to 14 post weaning and transport (period 1), and the C diet was fed to all pigs from day 14 to 35 post weaning and transport (period 2). Production measures were taken in 7 d intervals to calculate average daily gain (ADG), average daily feed intake (ADFI), average daily net energy intake (ADEI), gain: feed, and gain: net energy intake. Blood samples were collected prior to transport, following transport, and on days 2, 7, 14, 28, and 35 post weaning and transport to analyze cortisol, glucose, insulin, and nonesterified fatty acids. Behavior was assessed through video-recording on days 3, 5, 8, 11, and 13 post weaning and transport. In period 1, ADG was reduced (P = 0.04; 20.0 g/d) in IUHS vs. IUTN pigs. Pigs fed ND diets had reduced ADFI (P = 0.02; 9.3%) compared with C diet-fed pigs during period 1, which resulted in similar ADEI (P = 0.23; 1,115 ± 35 kcal/d). During transport, cortisol was decreased (P = 0.03; 25.8%) in IUHS vs. IUTN pigs. On day 2, glucose was decreased (P = 0.01; 13.8%) in IUHS vs. IUTN pigs. No in utero treatment-related behavior differences were observed but lying was reduced (P = 0.03; 6.5%) and standing was increased (P = 0.04; 14.1%) in ND vs. C pigs overall. Dietary antibiotic use has been limited in swine production due to concerns regarding antibiotic resistance. However, this may negatively impact the health, productivity, and welfare of pigs. Therefore, the study objective was to determine if combining dietary synbiotics and 0.20% L-glutamine would improve pig growth performance and intestinal health following weaning and transport when compared with traditionally used dietary antibiotics. Because previous research indicates that L-glutamine improves swine growth performance and synbiotics reduce enterogenic bacteria, it was hypothesized that supplementing diets with 0.20% L-glutamine (GLN) and synbiotics (SYN; 3 strains of Lactobacillus [1.2 × 10^9 cfu/g of strain/pig/d] + β-glucan [0.01 g/pig/d] + fructooligosaccharide [0.01 g/pig/d]) would have an additive effect and improve pig performance and intestinal health over that of dietary antibiotics. Mixed-sex pigs (N = 226; 5.86 ± 0.11 kg body weight [BW]) were weaned (19.4 ± 0.2 d of age) and transported for 12 h in central Indiana. Pigs were blocked by BW and allotted to one of two dietary treatments (5 to 6 pigs per pen): antibiotics (positive control [PC]; chlortetracycline [441 ppm] + tiamulin [38.5 ppm]), no antibiotics (negative control [NC]), GLN, SYN, or the NC diet with both the GLN and SYN additives (GLN + SYN) fed for 14 d. From day 14 post-weaning to the end of the grow-finish period, all pigs were provided common antibiotic-free diets. Data were analyzed using PROC GLIMMIX and PROC MIXED in SAS 9.4. Overall, haptoglobin was greater (P = 0.03; 216%) in NC pigs compared with PC pigs. On day 13, GLN and PC pigs tended to have reduced (P = 0.07; 75.2% and 67.3%, respectively) haptoglobin compared with NC pigs. On day 34, the jejunal goblet cell count per villi and per millimeter tended to be greater (P < 0.08; 71.4% and 62.9%, respectively) in SYN pigs compared with all other dietary treatments. Overall, jejunal mucosa tumor necrosis factor-alpha (TNFα) gene expression tended to be greater (P = 0.09; 40.0%) in NC pigs compared with PC pigs on day 34. On day 34, jejunal mucosa TNFα gene expression tended to be greater (P = 0.09; 33.3%, 41.2%, and 60.0%, respectively) in GLN pigs compared with SYN, GLN + SYN, and PC pigs. Although it was determined that some metrics of pig health were improved by the addition of GLN and SYN (i.e., haptoglobin and goblet cell count), overall, there were very few differences detected between dietary treatments and this may be related to the stress load incurred by the pigs. Maintaining metabolic balance is a key factor in the health of dairy cattle during the transition from pregnancy to lactation. Little is known regarding the role of the circadian timing system in the regulation of physiological changes during the transition period. We hypothesized that disruption of the cow's circadian timing system by exposure to chronic light–dark phase shifts during the prepartum period would negatively affect the regulation of homeostasis and cause metabolic disturbances, leading to reduced milk production in the subsequent lactation. The objective was to determine the effect of exposure to chronic light–dark phase shift during the last 5 wk prepartum of the nonlactating dry period on core body temperature, melatonin, blood glucose, β-hydroxybutyric acid (BHB) and nonesterified fatty acid (NEFA) concentrations, and milk production. Multiparous cows were moved to tiestalls at 5 wk before expected calving and assigned to control (CTR; n = 16) or phase-shifted (PS; n = 16) treatments. Control cows were exposed to 16 h of light and 8 h of dark. Phase-shifted cows were exposed to the same photoperiod; however, the light–dark cycle was shifted 6 h every 3 d until parturition. Resting behavior and feed intake were recorded daily. Core body temperature was recorded vaginally for 48 h at 23 and 9 d before expected calving using calibrated data loggers. Blood concentrations of melatonin, glucose, BHB, and NEFA were measured during the pre- and postpartum periods. Milk yield and composition were measured through 60 DIM. Treatment did not affect feed intake or body condition. Cosine fit analysis of 24-h core body temperature and circulating melatonin indicated attenuation of circadian rhythms in the PS treatment compared with the CTR treatment. Phase-shifted cows had lower rest consolidation, as indicated by more total resting time, but shorter resting period durations. Phase-shifted cows had lower blood glucose concentration compared with CTR cows (4 mg/mL decrease), but BHB and NEFA concentrations were similar between PS and CTR cows. Milk yield and milk fat yield were greater in PS compared with CTR cows (2.8 kg/d increase). Thus, exposure to chronic light–dark phase shifts during the prepartum period attenuated circadian rhythms of core body temperature, melatonin, and rest–activity behavior and was associated with increased milk fat and milk yield in the postpartum period despite decreased blood glucose pre- and postpartum. Acoustic propagation measurements were conducted in a Thalassia testudinum meadow in the Lower Laguna Madre, a shallow bay on the Texas Gulf of Mexico coast. A piezoelectric source transmitted frequency-modulated chirps (0.1 to 100 kHz) over a 24-h period during which oceanographic probes measured environmental parameters including dissolved oxygen and solar irradiance. Compared to a nearby less vegetated area, the received level was lower by as much as 30 dB during the early morning hours. Sandia National Laboratories recently undertook a 3 year research programme to create the components of a real-world virtual power plant (VPP) that can simultaneously participate in multiple markets. The authors' research extends current state-of-the-art rolling horizon control through the application of stochastic programming with risk aversion at various time resolutions. Antibiotic use has been limited in U.S. swine production. Therefore, the objective was to determine whether supplementing l -glutamine at cost-effective levels can replace dietary antibiotics to improve piglet welfare and productivity following weaning and transport. Based on previous research, we hypothesized that withholding dietary antibiotics would negatively affect pigs while diet supplementation with 0.20% l -glutamine (GLN) would have similar effects on pig performance and health as antibiotics. Mixed sex piglets (N = 480; 5.62 ± 0.06 kg BW) were weaned (18.4 ± 0.2 d of age) and transported for 12 h in central Indiana, for 2 replicates, during the summer of 2016 and the spring of 2017. Pigs were blocked by BW and allotted to 1 of 3 dietary treatments (n = 10 pens/dietary treatment/replicate [8 pigs/pen]); antibiotics (A ; chlortetracycline [441 ppm] + tiamulin [38.6 ppm]), no antibiotics (NA), or GLN fed for 14 d. On days 15 to 34, pigs were provided common antibiotic-free diets in 2 phases. Data were analyzed using PROC MIXED in SAS 9.4. Day 14 BW and days 0 to 14 ADG were greater (P = 0.01) for A (5.6% and 18.5%, respectively) and GLN pigs (3.8% and 11.4%, respectively) compared with NA pigs, with no differences between A and GLN pigs. Days 0 to 14 ADFI increased for A (P < 0.04; 9.3%) compared with NA pigs; however, no differences were detected when comparing GLN with A and NA pigs. Once dietary treatments ceased, no differences (P > 0.05) in productivity between dietary treatments were detected. On day 13, plasma tumor necrosis factor alpha (TNF-α) was reduced (P = 0.02) in A (36.7 ± 6.9 pg/mL) and GLN pigs (40.9 ± 6.9 pg/mL) vs. NA pigs (63.2 ± 6.9 pg/mL). Aggressive behavior tended to be reduced overall (P = 0.09; 26.4%) in GLN compared with A pigs, but no differences were observed between A and GLN vs. NA pigs. Huddling, active, and eating/drinking behaviors were increased overall (P < 0.02; 179%, 37%, and 29%, respectively) in the spring replicate compared with the summer replicate. When hot carcass weight (HCW) was used as a covariate, loin depth and lean percentage were increased (P = 0.01; 4.0% and 1.1%, respectively) during the spring replicate compared with the summer replicate. In conclusion, GLN supplementation improved pig performance and health after weaning and transport similarly to A across replicates; however, the positive effects of A and GLN were diminished when dietary treatments ceased. A healthy photovoltaic (PV) array has a specific impedance between node pairs, and any ground fault changes the impedance values. Reflectometry is a well-known technique in electromagnetics, and it could be exploited to detect fault and aging-related impedance variations in a PV system. A fault detection algorithm using the spread spectrum time-domain reflectometry (SSTDR) method has been introduced in this paper. SSTDR has been successfully used for detecting and locating aircraft wiring faults. However, the wide variation in impedance throughout the entire PV system, which is caused by the use of different materials and interconnections makes PV fault detection more challenging while using reflectometry. Unlike other conventional ground-fault detection techniques specifically developed for PV arrays, SSTDR does not depend on fault-current magnitudes. Therefore, SSTDR can be used even in the absence of the solar irradiation, which makes it a very powerful fault-detection tool. The proposed PV ground-fault detection technique has been tested in a real-world PV system, and it can confidently detect PV ground faults for different configurations of PV arrays (single and double strings) and fault resistances (0.5, 5, and 10- $\Omega$). Moreover, it has been experimentally verified that our proposed algorithm works at low irradiance and can detect specific ground faults that may remain undetected using the conventional ground-fault detection and interrupter (GFDI) fuses. We prepared a series of planar titanium microfluidic (μLC) columns, each 100 mm long, with 0.15, 0.3 and 0.5 mm i.d.’s. The microfluidic columns were packed with 1.8 μm C18 sorbent and tested under isocratic and gradient conditions. The efficiency and peak capacity of these devices were monitored using a micro LC instrument with minimal extra column dispersion. Columns with serpentine channels were shown to perform worse than those with straight channels. The loss of efficiency and peak capacity was more prominent for wider i.d. columns, presumably due to on-column band broadening imparted by the so-called “race-track” effect. The loss of chromatographic performance was partially mitigated by tapering the turns (reduction in i.d. through the curved region). While good performance was obtained for 0.15 mm i.d. devices even without turn tapering, the performance of 0.3 mm i.d. columns could be brought on par with capillary LC devices by tapering down to 2/3 of the nominal channel width in the turn regions. The loss of performance was not fully compensated for in 0.5 mm devices even when tapering was employed; 30% loss in efficiency and 10% loss in peak capacity was observed. The experimental data for various devices were compared using the expected theoretical relationship between peak capacity P c and efficiency N ; ( P c −1) =  N 0.5  × const. While straight μLC columns showed the expected behavior, the devices with serpentine channels did not adhere to the plot. In utero heat stress (IUHS) has several postnatal consequences in pigs that compromise health, increase stress response, and reduce performance. These phenotypes may be caused by epigenetic modifications such as DNA methylation, which are heritable molecular modifications that impact gene expression and phenotypic outcomes without changing the DNA sequence. Therefore, we aimed to compare the DNA methylation profiles between in-utero thermoneutral (IUTN) and IUHS pigs to identify differentially methylated regions. Twentyfour pregnant gilts were evenly assigned to either a thermoneutral (17.5 ± 2.1°C) or heat stress (cycling 26 to 36°C) chamber from d 0 to 59 of gestation, followed by thermoneutral conditions (20.9 ± 2.3°C) for the rest of gestation and until the piglets were weaned. At 105 d of age, 10 IUTN and 10 IUHS piglets were euthanized and Longissimus dorsi muscle samples were collected and used to perform whole-genome bisulfite sequencing (WGBS). Purified genomic DNA was fragmented and bisulfite conversion was performed. Illumina platforms were used to sequence WGBS libraries. All pigs had similar proportions of methylation at CpG sites. Twohundred-sixty-eight genomic regions were differentially methylated between IUTN and IUHS pigs. These identified regions are located across all pig chromosomes and ranged from 2 (SSC18) to 40 (SSC10). Eighty-five unique differentially-methylated genes were identified. These genes have been reported to be involved in key biological processes such as transcriptional repressor activity and tRNA processing (e.g., SKOR2, TRMT6, TSEN2), cellular response to heat stress (e.g., CCAR2), placental vascularization (e.g., FZD5), central nervous system (e.g.,VEPH1), cholesterol biosynthesis (e.g., CYB5R1), insulin receptor substrate (e.g.,IRS2), synaptic transmission (e.g.,RIMBP2), neurotrophic factor receptor activity (e.g.,LIFR), immune response (e.g., CD84), DNA repair (e.g., CHD1L), and cell proliferation and endocrine signaling (e.g., SSTR1, CYB5R1). In utero heat stress negatively impacts fetal development and alters postnatal phenotypes in swine. However, the effects of heat stress (HS) on fetal growth during early gestation is relatively unknown. The study objective was to evaluate the effects of gestational HS on measures of fetal growth in early gestation. Twentythree gestating gilts were exposed to HS (n = 12; cyclic 26-38°C) or thermoneutral (TN; n = 11; 18.5 ± 1.2°C) conditions from gestation d 6 ± 0.9 to 33 ± 1.7. Vaginal temperature was recorded in 15 min intervals using data loggers. Respiration rates (RR) were recorded daily at 0800, 1200, 1600, and 2000 h. All gilts were individually-housed, and limit-fed 1.82 kg/d. Gilt body weights (BW) were measured weekly and average daily gain and gain:feed were calculated. At gestation d 33 ± 1.7, gilts were harvested and fetal weights and counts were recorded. Data were analyzed using PROC GLIMMIX. Vaginal temperatures and RR were greater (P < 0.01) in HS (38.5 ± 0.1°C and 59 ± 2 bpm, respectively) versus TN gilts (38.0 ± 0.1°C and 29 ± 2 bpm, respectively). Although no initial BW differences were observed (P = 0.39; 148.8 ± 4.1 kg), final BW tended to be greater (P < 0.07) for HS (166.5 ± 3.8 kg) versus TN (156.8 ± 4.0 kg) gilts. Average daily gain was increased (P < 0.01) for HS (0.5 ± 0.1 kg/d) versus TN (0.3 ± 0.1 kg/d) gilts. Gain:feed was greater (P < 0.01) for HS (0.27 ± 0.03) versus TN (0.16 ± 0.03) gilts. No fetal weight (P = 0.23; 4.3 ± 0.2 g/fetus) or count (P = 0.15; 15.2 ± 0.9 fetuses/gilt) differences were detected. Proper insulin homeostasis appears critical for adapting to and surviving a heat load. Further, heat stress ( HS) induces phenotypic changes in livestock that suggest an increase in insulin action. The current study objective was to evaluate the effects of HS on whole-body insulin sensitivity. Female pigs (57 ± 4 kg body weight) were subjected to two experimental periods. During period 1, all pigs remained in thermoneutral conditions ( TN; 21°C) and were fed ad libitum. During period 2, pigs were exposed to: (i) constant HS conditions (32°C) and fed ad libitum ( n = 6), or (ii) TN conditions and pair-fed ( PFTN; n = 6) to eliminate the confounding effects of dissimilar feed intake. A hyperinsulinemic euglycemic clamp ( HEC) was conducted on d3 of both periods; and skeletal muscle and adipose tissue biopsies were collected prior to and after an insulin tolerance test ( ITT) on d5 of period 2. During the HEC, insulin infusion increased circulating insulin and decreased plasma C-peptide and nonesterified fatty acids, similarly between treatments. From period 1 to 2, the rate of glucose infusion in response to the HEC remained similar in HS pigs while it decreased (36%) in PFTN controls. Prior to the ITT, HS increased (41%) skeletal muscle insulin receptor substrate-1 protein abundance, but did not affect protein kinase B or their phosphorylated forms. Sample introduction in microfluidic liquid chromatography often generates wide zones rather than peaks, especially when a large sample volume (relative to column volume) is injected. Formation of wide injection zones can be further amplified when the sample is dissolved in a strong eluent. In some cases sample breakthrough may occur, especially when the injection is performed into short trapping columns. To investigate the band formation and subsequent zone focusing under gradient elution in situations such as these, we developed the Repetitive Injection Method (RIM), based on the temporally resolved introduction of two discrete peaks to a column, mimicking both the leading and trailing edges of a larger, singly injected sample zone. Using titanium microfluidic 0.32 mm I.D. columns, the results of RIM experiments were practically identical to injection of a correspondingly larger single zone volume. It was also experimentally shown that zone width (spacing between two injected peaks) decreases during gradient elution. We utilized RIM experiments to investigate wide sample zones created by strong sample solvent, and subsequent gradient zone focusing for a series of compounds. This experimental work was compared with computationally simulated chromatograms. The success of sample focusing during injection and gradient elution depends not only on an analyte's absolute retention, but also on how rapidly the analyte's retention changes during the mobile phase gradient. The drilling of a deep borehole in ice is an undertaking that spans several seasons. Over recent decades such drilling has become widespread in both polar regions. Owing to the remoteness of the drill sites, considerable cost is associated with the drilling of a deep borehole of several thousand meters. The replicate coring system allows re-drilling of ice core at select depths within an existing parent borehole. This effectively increases the yield of the existing borehole and permits re-sampling of ice in areas of high scientific value. The replicate coring system achieves this through the combination of actuators, motors, sensors and a computerized control system. After the NEEM (Greenland) deep ice-core drilling was declared terminated with respect to developing stratigraphic climate reconstructions, efforts were turned toward collecting basal icesheet debris and, if possible, drilling into the bedrock itself. In 2010, several meters of banded debrisrich ice were obtained under normal ice-drilling operations with the NEEM version of the Hans Tausen (HT) drill, but further penetration was obstructed by a rock in the path of the drill head at 2537.36 m. During short campaigns in 2011 and 2012, attempts were made to penetrate further using various reinforced ice cutters mounted on the HT drill head, tailored to cut through rock. These had some success in penetrating coarse material, but produced severely damaged cutters. Additionally a 51 mm diameter diamond cutting tipped rock drill was adapted to fit the NEEM drill. With this device, several additional meters of core containing subglacial sediments, rocks and rock fragments were collected. With these tools 1.39 m of additional material were obtained during the 2011 field season, and 7.1 m during 2012. Subglacial water refreezing into the newly formed borehole hindered further penetration, and the bedrock interface was not reached before final closure of the NEEM Camp. [ABSTRACT FROM AUTHOR]

On 1 December 2011 the West Antarctic Ice Sheet (WAIS) Divide ice-core project reached its final depth of 3405 m. The WAIS Divide ice core is not only the longest US ice core to date, but is also the highest-quality deep ice core, including ice from the brittle ice zone, that the US has ever recovered. The methods used at WAIS Divide to handle and log the drilled ice, the procedures used to safely retrograde the ice back to the US National Ice Core Laboratory (NICL) and the methods used to process and sample the ice at the NICL are described and discussed. A balanced omega-6/omega-3 polyunsaturated fatty acid (PUFA) ratio has been linked to health benefits and the prevention of many chronic diseases. Current dietary intervention studies with different sources of omega-3 fatty acids (omega-3) lack appropriate control diets and carry many other confounding factors derived from genetic and environmental variability. In our study, we used the fat-1 transgenic mouse model as a proxy for long-term omega-3 supplementation to determine, in a well-controlled manner, the molecular phenotype associated with a balanced omega-6/omega-3 ratio. The fat-1 mouse can convert omega-6 to omega-3 PUFAs, which protect against a wide variety of diseases including chronic inflammatory diseases and cancer. Both wild-type (WT) and fat-1 mice were subjected to an identical diet containing 10% corn oil, which has a high omega-6 content similar to that of the Western diet, for a six-month duration. We used a multi-platform lipidomic approach to compare the plasma lipidome between fat-1 and WT mice. In fat-1 mice, an unbiased profiling showed a significant increase in the levels of unesterified eicosapentaenoic acid (EPA), EPA-containing cholesteryl ester, and omega-3 lysophosphospholipids. The increase in omega-3 lipids is accompanied by a significant reduction in omega-6 unesterified docosapentaenoic acid (omega-6 DPA) and DPA-containing cholesteryl ester as well as omega-6 phospholipids and triacylglycerides. Targeted lipidomics profiling highlighted a remarkable increase in EPA-derived diols and epoxides formed via the cytochrome P450 (CYP450) pathway in the plasma of fat-1 mice compared with WT mice. Integration of the results of untargeted and targeted analyses has identified a lipidomic biosignature that may underlie the healthful phenotype associated with a balanced omega-6/omega-3 ratio, and can potentially be used as a circulating biomarker for monitoring the health status and the efficacy of omega-3 intervention in humans. A sample of 406 subjects completed a questionnaire testing the influence of personal spirituality, education and several other factors on sexual tension in cross-sex friendships (CSF Tension). The subjects included 143 students from a Christian university (CU), 137 from a secular junior college (JC), and 127 non-students (NS). The primary criterion variable was the amount of CSF Tension experienced: Each subject rated to what extent sexual tension made friendship difficult due to different characteristics (e.g., he/she is physically attractive, dressed seductively) of 16 hypothetical friendships. A measure of sexuality—a composite of 12 questions adapted from the Self-Assessment Survey—assessed sexual activity, values, and ideation. Other predictors included neuroticism, openness to experience, extraversion, and demographics. JC students rated significantly more liberal than CU students on all 12 of the sexuality questions but no CSF-Tension differences were found for 15 of the 16 hypothetical CSF comparisons. JC students also rated two standard deviations lower on spirituality than CU students. Also, NS differed from students by being older, more spiritual, more educated, less involved in sexual activities, and experienced less CSF Tension. For the entire sample, spirituality was associated with sharply lower levels of sexual activities and ideation (r = −.601) and significantly lower levels of CSF Tension. ABSTRACT A multistaged geophysical methodology was recently used at site 15Ck10, a state-protected Middle Woodland period Adena (500 bce to ce 250) conical burial mound in Central Kentucky, USA. Data from this research are used to develop anthropological interpretations on the nature of burial mound construction and regional interaction between separate mortuary traditions in Kentucky. Methods utilized in this research include magnetic gradiometry, electromagnetic induction, ground-penetrating radar, electrical resistivity tomography and downhole magnetic susceptibility. Together, these methods helped identify a circular ditch-and-embankment earthwork enclosing the burial mound, as well as possible off-mound activity areas commonly reported in the Adena literature. Within the mound, multiple anomalies were discovered that are consistent with hypothesized geophysical responses of mound stages and funerary features. A review of burial mounds excavated by the University of Kentucky during the depression-era work relief programmes (i.e. the Works Progress Administration - WPA) of the 1930s and early 1940s provide a vehicle with which to interpret these features. Comparisons of our data with length and width dimensions of known mortuary facilities documented in WPA excavations suggest that two distinct burial types were used at 15Ck10. These data are used to build a comprehensive site interpretation and create a relative dating context for the site. A novel method for the detection of nitrate was developed using simplified nitrate reductase (SNaR) that was produced by genetic recombination techniques. The SNaR consists of the fragments of the Mo-molybdopterin (MO- MPT) binding site and nitrate reduction active site and has high activity for nitrate reduction. The method is based on a unique combination of the enzyme-catalyzed reduction of nitrate to nitrite by thin-layer coulometry followed by spectroscopic measurement of the colored product generated from the reaction of nitrite with Griess reagents. Coulometric reduction of nitrate to nitrite used methyl viologen (MV2+) as the electron transfer mediator for SNaR and controlled potential coulometry in an indium tin oxide (ITO) thin-layer electrochemical cell. Absorbance at 540 nm was proportional to the concentration of nitrate in the sample with a linear range of 1−160 μM and a sensitivity of 8000 AU M-1. The method required less than 60 μL of sample. Detection of nitrate could also be performed by measuring the charge associated with coulometry. However, the spectroscopic procedure gave superior performance because of interference from the large background charge associated with coulometry. Results for the determination of nitrate concentration in several natural water samples using this device with spectroscopic detection are in good agreement with analysis done with a standard method. Metal catalyst free carbon nanotube (MCFCNT) whiskers were first used as an electrode modification material on a gold electrode surface for zinc voltammetric measurements. A composite film of Nafion and MCFCNT whiskers was applied to a gold electrode surface to form a mechanically stable sensor. The sensor was then used for zinc detection in both acetate buffer solution and extracted bovine serum solution. A limit of detection of 53 nM was achieved for a 120 s deposition time. The zinc in bovine serum was extracted via a double extraction procedure using dithizone in chloroform as a zinc chelating ligand. Excessive heat exposure reduces intestinal integrity and post-absorptive energetics that can inhibit wellbeing and be fatal. Therefore, our objectives were to examine how acute heat stress (HS) alters intestinal integrity and metabolism in growing pigs. Animals were exposed to either thermal neutral (TN, 21°C; 35–50% humidity; n = 8) or HS conditions (35°C; 24–43% humidity; n = 8) for 24 h. Compared to TN, rectal temperatures in HS pigs increased by 1.6°C and respiration rates by 2-fold (P<0.05). As expected, HS decreased feed intake by 53% (P<0.05) and body weight (P<0.05) compared to TN pigs. Ileum heat shock protein 70 expression increased (P<0.05), while intestinal integrity was compromised in the HS pigs (ileum and colon TER decreased; P<0.05). Furthermore, HS increased serum endotoxin concentrations (P = 0.05). Intestinal permeability was accompanied by an increase in protein expression of myosin light chain kinase (P<0.05) and casein kinase II-α (P = 0.06). Protein expression of tight junction (TJ) proteins in the ileum revealed claudin 3 and occludin expression to be increased overall due to HS (P<0.05), while there were no differences in claudin 1 expression. Intestinal glucose transport and blood glucose were elevated due to HS (P<0.05). This was supported by increased ileum Na+/K+ ATPase activity in HS pigs. SGLT-1 protein expression was unaltered; however, HS increased ileal GLUT-2 protein expression (P = 0.06). Linear mode conversion of Langmuir/z waves to electromagnetic radiation near the plasma and upper hybrid frequency in the presence of density gradients is potentially relevant to type II and III solar radio bursts, ionospheric radar experiments, pulsars, and continuum radiation for planetary magnetospheres. Here, we study mode conversion in warm, magnetized plasmas using a numerical electron fluid simulation code when the density gradient has a wide range of angle, δ, to the ambient magnetic field, B0, for a range of incident Langmuir/z wavevectors. Our results include: (1) Left-handed polarized ordinary (oL) and right-handed polarized extraordinary (xR) mode waves are produced in various ranges of δ for Ω0 = (ωL/c)1/3(ωce/ω) < 1.5, where ωce is the (angular) electron cyclotron frequency, ω is the angular wave frequency, L is the length scale of the (linear) density gradient, and c is the speed of light; (2) the xR mode is produced most strongly in the range, 40° < δ < 60°, for intermediately magnetized plasmas with Ω0 = 1.0 and 1.5, while it is produced over a wider range, 0° ≤ δ ≤ 90°, for weakly magnetized plasmas with Ω0 = 0.1 and 0.7; (3) the maximum total conversion efficiencies for wave power from the Langmuir/z mode to radiation are of order 50%-99% and the corresponding energy conversion efficiencies are 5%-14% (depending on the adiabatic index γ and β = Te/mec2, where Te is the electron temperature and me is the electron) for various Ω0; (4) the mode conversion window becomes wider as Ω0 and δ increase. Hence, the results in this paper confirm that linear mode conversion under these conditions can explain the weak total circular polarization of interplanetary type II and III solar radio bursts because a strong xR mode can be generated via linear mode conversion near δ ∼ 45°. Lactating sows are especially sensitive to heat stress (HS) due to high metabolic demands resulting from milk output. Therefore, there is a need to develop effective cooling technologies that reduce the impact of HS on lactating sows. The study objective was to determine whether electronically controlled cooling pads (ECP) would allow sows to remain euthermic under HS conditions. Twelve multiparous (2.69 ± 0.85) lactating sows (265.4 ± 26.1 kg) and litters (11.4 ± 0.7 piglets/litter) were assigned to either a non-functional ECP (NECP; n = 6) or an ECP (n = 6), housed in farrowing crates, and tested over two repetitions. Sows were provided feed and water ad libitum and all sows were exposed to HS (28.27 ± 1.42°C nighttime to 35.14 ± 0.70°C daytime). Body temperature (TB), was measured hourly using vaginal implants, and respiration rate (RR) was measured in 30 min intervals from 0600 to 2100 hrs over 2 d representing mid- and late lactation. Mixed model periodic regression equations were fitted to the TB and RR to time. The final model for TB included the effect of replicate, random effect of sow, day of lactation as a covariate, single-phase sine and cosine variables for each treatment and 2-phase sine and cosine periodic regression variables. The final model for RR included the random effect of sow, effect of treatment, replicate, day of lactation, sleeping, and single-phase sine and cosine functions for each treatment. An interaction (P < 0.05) between treatment and sine or cosine was observed for TB. For RR there was no interaction for treatment and cosine (P = 0.07), but an interaction (P < 0.05; -9.94) was observed for sine. Heat stress (HS) decreases lactation output in sows due to an attempt to reduce metabolic heat production; however, this negatively affects litter growth performance. Therefore, the study objective was to determine whether electronically controlled cooling pads (ECP) would improve indirect measures of lactation output (e.g., metabolic heat production) and litter growth performance in HS exposed sows. Over two repetitions, 12 multiparous (2.69 ± 0.85) lactating sows (265.4 ± 26.1 kg) and litters were assigned to either an ECP (n = 6) or a non-functional ECP (NECP; n = 6) and placed into farrowing crates within indirect calorimeters from d 3.7 ± 0.5 to d 18.7 ± 0.5 of lactation. Litters were standardized across all sows (11.4 ± 0.7 piglets/litter), and sows were provided ad libitum feed and water. All sows were exposed to cyclical HS (28.27 ± 1.42°C nighttime to 35.14 ± 0.70°C daytime). On d 4, 8, 14, and 18 of lactation, indirect calorimetry was performed on each individual sow and litter to determine total heat production (THP). Body temperature (TB), was measured hourly using vaginal implants, and respiration rate (RR) was measured daily at 0700, 1100, 1300, 1500, and 1900 h. Litter weights were obtained at birth and weaning. An overall decrease (P < 0.01; 25 bpm) in RR and maximum daily TB (P = 0.02; 0.40°C) was observed in ECP versus NECP sows. An increase in THP (P < 0.01; 20.4%) and THP/kg0.75 (P < 0.01; 23.1%) was observed for ECP when compared to NECP sows and litters. Litter growth rate was increased (P < 0.01; 20.8%) in ECP versus NECP sows. In summary, the use of ECP improves litter growth, thermoregulatory measures, and bioenergetic parameters associated with greater milk production in lactating sows exposed to cyclical heat stress. Urine collection is a useful tool to analyze intestinal permeability in cattle for research and diagnostic purposes. However, urine sampling techniques often rely on total waste collection, which reduces the ability to perform more frequent sampling and obtain accurate and sterile urine volumes. A potential alternative is urethral catheters, which have been used in cows and weaned heifers. However, urethral catheters have not been thoroughly tested in pre-weaned dairy heifer calves. The study objective was to develop a urethral catheter placement procedure in pre-weaned heifer calves for continuous and accurate urine collection. Fifteen Holstein heifer calves had catheters placed at 8 ± 2 days (37.5 ± 3.38 kg BW) and 40 ± 2 days (59.3 ± 5.38 kg BW) of age. During the procedure, calves were individually housed (1.87 m2/calf) and restrained. The vulva was cleaned using betadine and 70% ethanol and then a sterile, lubricated 8.9 cm speculum was inserted into the vagina. A sterile guidewire (145 cm x 0.89 cm) was inserted into a lubricated sterile 10 FR catheter. The catheter was inserted into the urethral opening (~5-7 cm into vagina), guided into the bladder, and the catheter balloon was filled with water (10 mL). The guide wire was removed, and urine flow confirmed correct placement before a 4 L urinary drainage bag was attached to the catheter. Individual calf health observations were made twice over a 24-hour period and included vaginal discharge, bleeding, redness or inflammation, and tissue discharge in the urine. Occurrence rate was determined using PROC FREQ in SAS 9.4. Regardless of catheterization timing, bleeding and tissue discharge occurred at a rate of 3.33% ± 0.18, and vaginal discharge and inflammation occurred at a rate of 6.70% ± 0.25. As more countries seek solutions to their de-carbonization targets using renewable energy (RE) technologies, interconnection standards and national grid codes for distributed energy resources (DER) are being updated to support higher penetrations of RE and improve grid stability. Common grid-code revisions mandate DER devices, such as solar inverters and energy storage systems, ride-through (RT) voltage and frequency disturbances. This is necessary because as the percentage of generation from DER increases, there is a greater risk power system faults will cause many or all DER to trip, triggering a substantial load-generation imbalance and possible cascading blackout. This paper demonstrates for the first time a methodology to verify commercial DER devices are compliant to new voltage, frequency, and rate of change of frequency (ROCOF) RT requirements established in IEEE Std. 1547-2018. The methodology incorporates a software automation tool, called the SunSpec System Validation Platform (SVP), in combination with a hardware-in-the-loop (HIL) system to execute the IEEE Std. 1547.1-2020 RT test protocols. In this paper, the approach is validated with two commercial photovoltaic inverters, the test results are analyzed for compliance, and improvements to the test procedure are suggested. A novel catalyst free carbon nanotube (CNT) electrode was fabricated by a Carbo Thermo Carbide Conversion (CTCC) method. The morphology and nanostructure of the CNT electrodes were characterized by transmission electron microscopy (TEM) and Raman spectroscopy. Electrochemical characterization using potassium ferricyanide, ferroin and catechol shows these CNTs have higher current capacity, faster electron transfer rate and larger microscopic surface area than glassy carbon (GC) electrode. Anodic stripping voltammetry (ASV) and cathodic stripping voltammetry (CSV) were used to determine Mn concentration using metal catalyst free carbon nanotube (MCFCNT) electrodes and square wave stripping voltammetry (SWSV). The MCFCNTs are synthesized using a Carbo Thermal Carbide Conversion method which results in a material that does not contain residual transition metals. Detection limits of 120 nM and 93 nM were achieved for ASV and CSV, respectively, with a deposition time of 60 s. CSV was found to be better than ASV in Mn detection in many aspects, such as limit of detection and sensitivity. The CSV method was used in pond water matrix addition measurements. A microfabricated electrochemical cell has been developed as a disposable detector for flow injection analysis (FIA) and high-performance liquid chromatography (HPLC). The simplicity of the fabrication process allows this detector to be used as a low-cost, disposable device that can be replaced easily if its performance degrades rather than disassembling the detector and polishing the electrode surface, which is the usual procedure. The detector consists of thin film-metal electrodes-platinum working electrode, platinum auxiliary electrode, and silver/silver chloride coated on Pt reference electrode-deposited on a polyimide substrate with a locking layer of chromium in between. A microfluidic cover made of polyimide directs the solution flow across the electrodes. The detector was evaluated with FIA of ferrocyanide and HPLC of ascorbic acid and acetaminophen and a mixture of two pharmaceutical compounds-dextrorphan and levallorphan-with acetaminophen internal standard. The HPLC calibration curves showed good linearity (R² > 0.99). Limits of detection were 1 nM for acetaminophen, 1 nM for ascorbic acid, 50 nM for dextrorphan, and 80 nM for levallorphan. When detected with a commercial detector dextrorphan and levallorphan had lower limits of detection, 3 and 5 nM, respectively. Chromatograms of the mixture were comparable to those obtained with a commercial detector. The detector could be used continuously for about 48 h with FIA and about 10-20 h with HPLC after which performance gradually degraded as the AgCl on the reference electrode dissolved causing loss of potential control. Although cognitive deficits are common in persons with multiple sclerosis (MS), the relationship between subjective complaints and objective impairment is sometimes obscured. To elaborate this issue, the present study examined the relationship between subjective complaints of dysexecutive syndrome, neuropsychological performance, and self-reported activities of daily living in 42 people with MS and 13 control participants. Regression analyses revealed that subjective complaints of impairment, measured by the Frontal Systems Behavior Scale (FrSBe), emerged as a significant predictor of neuropsychological deficit and poor adaptive function. Accordingly, subjective complaints of dysexecutive function in MS may serve as a potent indicator of cognitive and functional impairment. The coastal plain of the Arctic National Wildlife Refuge hosts seven species of migratory shorebirds listed as highly imperiled or high priority by the U.S. Shorebird Conservation Plan and five species listed as Birds of Conservation Concern by the U.S. Fish and Wildlife Service. During the first comprehensive shorebird survey of the 674 000 ha ‘1002 Area’ on the coastal plain, we recorded 14 species of breeding shorebirds at 197 rapidly surveyed plots during June 2002 and 2004. We also estimated detection ratios with a double counting technique, using data collected at 37 intensively studied plots located on the North Slope of Alaska and northern Canada. We stratified the study area by major habitat types, including wetlands, moist areas, uplands, and riparian areas, using previously classified Landsat imagery. We developed population estimates with confidence limits by species, and estimated the total number of shorebirds in the study area to be 230 000 (95% CI: 104 000-363 000), which exceeds the biological criterion for classification as both a Western Hemisphere Shorebird Reserve Network Site of International Importance (100 000 birds) and a Ramsar Wetland of International Importance (20 000 birds), even when conservatively estimated.
