ACI 318-19 and ASCE 7-22 specify code-compliant and emulative seismic force-resisting systems (SFRS) including intermediate precast shear wall systems. For the seismic design of a precast shear wall system, its successful performance heavily depends on the capacity design of inevitable discrete connections between precast wall panels, thus playing a critical role in controlling yielding to occur within steel reinforcements at the vicinity of intended locations or components. This is imperative for achieving not only ductility but also adequate energy dissipation, ensuring the system can effectively withstand seismic forces. To this end, the overstrength is essentially required to prevent any brittle failure, and the connections designed not to yield can be protected by addressing the 1.5Sy condition as specified in ACI 318-19 and ASCE 7-22. However, there can be a blind spot for the precast connections designed to yield. To this end, this study showed some cases based on practical examples from which a reasonable magnitude of overstrength (Sy) was tentatively recommended for better safety in the seismic design of the intermediate precast shear wall system with precast connections designed to be yielded. Iron is crucial for the metabolism and growth of most prokaryotic cells. The ferric uptake regulator (Fur) protein plays a central role in regulating iron homeostasis and metabolic processes in bacteria. It ensures the proper utilization of iron and the maintenance of cellular functions in response to environmental cues. Fur proteins are composed of an N-terminal DNA-binding domain (DBD) and a C-terminal dimerization domain (DD), typically existing as dimers in solution. Fur proteins have conserved metal-binding sites named S1, S2, and S3. Among them, site S2 serves as a regulatory site, and metal binding at S2 results in conformational changes. Additionally, as a transcriptional regulator, Fur specifically binds to a consensus DNA sequence called the Fur box. To elucidate the structural and functional properties of Fur proteins, various structures of metal- or DNA-bound Fur proteins or apo-Fur proteins have been determined. In this review, we focus on the structural properties of Fur proteins according to their ligand-bound state and the drug development strategies targeting Fur proteins. The current study longitudinally investigated the relationships between Korean tertiary-level students' English names adopted and used at their teacher's request, and native speakerism, identity, and emotions. Thirteen graduate students enrolled in an English-medium course and their Korean teacher participated in the study. The research instruments included classroom observations, reflective journal writing, and in-depth, semi-structured interviews. The results indicated that the English names bore, in complicated and changing ways, on the identities and emotions of the students and teacher. Native speakerism was found to be constantly supported by all the students, regardless of their attitudes and perceptions toward English names, and the teacher. [ABSTRACT FROM AUTHOR]

Droplet single-cell RNA-sequencing (dscRNA-seq) has enabled rapid, massively parallel profiling of transcriptomes. However, assessing differential expression across multiple individuals has been hampered by inefficient sample processing and technical batch effects. Here we describe a computational tool, demuxlet, that harnesses natural genetic variation to determine the sample identity of each droplet containing a single cell (singlet) and detect droplets containing two cells (doublets). These capabilities enable multiplexed dscRNA-seq experiments in which cells from unrelated individuals are pooled and captured at higher throughput than in standard workflows. Using simulated data, we show that 50 single-nucleotide polymorphisms (SNPs) per cell are sufficient to assign 97% of singlets and identify 92% of doublets in pools of up to 64 individuals. Given genotyping data for each of eight pooled samples, demuxlet correctly recovers the sample identity of >99% of singlets and identifies doublets at rates consistent with previous estimates. We apply demuxlet to assess cell-type-specific changes in gene expression in 8 pooled lupus patient samples treated with interferon (IFN)-β and perform eQTL analysis on 23 pooled samples.

Increased consumption of Western-type diets and environmental insults lead to wide-spread increases in the plasma levels of saturated fatty acids and lipoprotein oxidation. The aim of this study is to examine whether palmitate and minimally modified low-density lipoprotein (mmLDL) exert an additive effect on macrophage activation. We found that CXCL2 and TNF-α secretion as well as ERK and p38 phosphorylation were additively increased by co-treatment of J774 macrophages with palmitate and mmLDL in the presence of lipopolysaccharide (LPS). Furthermore, the analysis of differentially expressed genes using the KEGG database revealed that several pathways, including cytokine-cytokine receptor interaction, and genes were significantly altered. These results were validated with real-time PCR, showing upregulation of Il-6, Csf3, Il-1β, and Clec4d. The present study demonstrated that palmitate and mmLDL additively potentiate the LPS-induced activation of macrophages. These results suggest the existence of synergistic mechanisms by which saturated fatty acids and oxidized lipoproteins activate immune cells.
