Abstract

Proteasomes are the multisubunit proteases thought to be involved in the generation of peptides presented by MHC class I molecules. When cells are stimulated with IFN-gamma, two MHC encoded subunits, LMP2 and LMP7, are incorporated into the proteasomal complex, presumably by displacing the housekeeping subunits, designated Y and X, respectively. These changes in the subunit composition appear to facilitate class I-mediated Ag presentation, presumably bu altering the cleavage specificities of the proteasome. Here we show that the cartilaginous fish, the most primitive class of vertebrates in which the MHC has been identified, have both LMP7 and X genes. Interestingly, nurse sharks, a member of the cartilaginous fish, appear to have two LMP7 genes, one encoding a typical LMP7 subunit and the other encoding a less typical one. In contrast, only cDNA clones with residues characteristic of X were identified in hagfishes and lampreys, the two extant members of the jawless fish in which no MHC has been identified. Pairwise amino acid sequence comparison and phylogenetic tree analysis showed that the subunits encoded by these clones were nearly equidistant from LMP7 and X, suggesting that the LMP7 gene might have emerged after the appearance of the jawless fish. Sequence comparison of the LMP7 and X/X-like subunits isolated from various vertebrate species showed that, unlike the X/X-like subunit, the LMP7 subunit displays a striking interspecies sequence variability in the vicinity of its catalytic site.

Abstract

We describe here evidence of congenital enzyme mistargeting induced not by abnormalities in the signal sequence. We examined the molecular mechanism of hereditary ornithine aminotransferase (OAT) deficiency causing gyrate atrophy of the choroid and retina (GACR). Nucleotide sequencing of OAT cDNA generated from a GACR patient's mRNA revealed a single base change from C to G at position 268, resulting in an amino acid substitution of neutral Gln(CAA) with negatively charged Glu(GAA) at position 90 (Q90E). Immunohistochemical and transient expression analyses suggested expression of a defective labile OAT in the patient's tissues. However, high-level expression and immunocytochemical analyses elucidated that Q90E OAT (the patient's OAT) was localized within the limits of cytoplasmic free ribosomes in precursor form without any mitochondrial entry, indicating that the patient's precursor OAT was synthesized and rapidly degraded because of accumulation in the cytosol. It is interesting that, although the mutation site (Q90E) in this GACR patient's OAT was within the coding sequence of the mature protein, the precursor exhibited loss of mitochondrial targeting function. These findings suggest that not only the signal sequence but a critical part of the mature sequence plays an essential role in mitochondrial entry of the OAT precursor protein.

Abstract

The amphibian Xenopus laevis is the most primitive vertebrate in which the MHC has been identified both at the biochemical and functional levels. In the present study, cDNA clones for MHC class II beta-chain genes of X. laevis were isolated using the polymerase chain reaction. In partially inbred animals with the MHC f haplotype, two distinct cDNA clones assumed to encode isotypes were identified. The deduced MHC class II beta-chains of X. laevis had structures fundamentally similar to those of their mammalian counterparts. The overall amino acid sequence identity between X. laevis and representative mammalian MHC class II beta-chains was approximately 41 to 50%. Sequence comparison of cDNA clones isolated from outbred subjects and the partially inbred frog with the MHC f haplotype showed that nucleotide substitutions are located primarily in the beta 1 domain and that non-synonymous substitutions occur more frequently than synonymous substitutions in the putative Ag-binding sites. This result indicates that, as in mammalian MHC genes, the MHC class II beta-chain genes of X. laevis are under positive Darwinian selection. Despite the fact that X. laevis is a tetraploid species, its MHC, as defined by serology, immunochemistry, and MLR, is diploidized. Family studies showed that all MHC class II beta-chain genes detectable in the genome of X. laevis segregate with serologically defined MHC haplotypes, suggesting that the second, inactivated set of MHC class II beta-chain genes was either deleted or that their sequences diverged too far to be detected by DNA hybridization. Furthermore, a nearly constant number of MHC class II beta-chain genes was detectable in most, if not all, Xenopus species with various degrees of ploidy. Thus, various polyploid species of Xenopus appear to have used a similar strategy to diploidize their MHC genes.
