Abstract

Lenvatinib is a multiple receptor tyrosine kinase inhibitor targeting mainly vascular endothelial growth factor (VEGF) and fibroblast growth factor (FGF) receptors. We investigated the immunomodulatory activities of lenvatinib in the tumor microenvironment and its mechanisms of enhanced antitumor activity when combined with a programmed cell death-1 (PD-1) blockade. Antitumor activity was examined in immunodeficient and immunocompetent mouse tumor models. Single-cell analysis, flow cytometric analysis, and immunohistochemistry were used to analyze immune cell populations and their activation. Gene co-expression network analysis and pathway analysis using RNA sequencing data were used to identify lenvatinib-driven combined activity with anti-PD-1 antibody (anti-PD-1). Lenvatinib showed potent antitumor activity in the immunocompetent tumor microenvironment compared with the immunodeficient tumor microenvironment. Antitumor activity of lenvatinib plus anti-PD-1 was greater than that of either single treatment. Flow cytometric analysis revealed that lenvatinib reduced tumor-associated macrophages (TAMs) and increased the percentage of activated CD8+ T cells secreting interferon (IFN)-γ+ and granzyme B (GzmB). Combination treatment further increased the percentage of T cells, especially CD8+ T cells, among CD45+ cells and increased IFN-γ+ and GzmB+ CD8+ T cells. Transcriptome analyses of tumors resected from treated mice showed that genes specifically regulated by the combination were significantly enriched for type-I IFN signaling. Pretreatment with lenvatinib followed by anti-PD-1 treatment induced significant antitumor activity compared with anti-PD-1 treatment alone. Our findings show that lenvatinib modulates cancer immunity in the tumor microenvironment by reducing TAMs and, when combined with PD-1 blockade, shows enhanced antitumor activity via the IFN signaling pathway. Abstract

Aim: To investigate the effects of leuprorelin using a self‐administered quality‐of‐life (QOL) questionnaire in patients with recurrent gynecological cancer. Methods: Records of patients who received 3.75 mg leuprorelin every 4 weeks for the treatment of recurrent gynecological cancer were retrospectively reviewed. The physical domain of the QOL questionnaire, Care Notebook, was used to assess physical symptoms. Symptom deterioration was defined as a ≥10‐point increase in baseline score; otherwise, symptoms were defined as controlled. Radiological and serological responses were evaluated according to the 2011 Gynecological Cancer Intergroup criteria. Results: From 2007 to 2015, 25 patients were administered leuprorelin for the treatment of epithelial ovarian cancer, granulosa cell tumor, endometrial cancer, endometrial stromal sarcoma and clear cell cervical cancer (in 13, 3, 6, 2 and 1 patients, respectively). Twenty patients had received a median of three lines (range 1–12 lines) of chemotherapy. Ten patients had progressive disease during their previous round of chemotherapy. Twenty patients completed the questionnaire every 4 weeks. Following leuprorelin treatment for 8 weeks, the symptom and disease control rates were 65% (13/20) and 44% (11/25), respectively. Two patients, one each with granulosa cell tumor and endometrial cancer, had stable disease at 6 months. Among the 20 patients who completed the QOL questionnaire, symptom control and disease control at 8 weeks showed a significant correlation (P = 0.016). Conclusion: Leuprorelin had minimal anticancer activity. The physical domain of the QOL questionnaire could be used to assess effects of hormonal treatment. [ABSTRACT FROM AUTHOR]

Abstract

Heterostructures of superconducting and ferromagnetic materials are of fundamental interest because of the mutual interaction of antagonistic kinds of ordering at the S-F interface. Normally, the superconducting transition temperature Tc should be strongly suppressed at the S-F interface owing to the penetration of Cooper pairs into the ferromagnetic side. Nevertheless, constructive interactions between S and F orders have been suggested to occur via the modification of ferromagnetic order by the superconducting state. This may induce an inhomogeneous magnetic state, often called a cryptoferromagnetic state, and the relevant domain wall effect, which will lead to a local decrease of the pair-breaking parameter. However, the domain wall effect, even if it exists, is quite subtle from the experimental view point and is normally difficult to observe. Here we show that the defect-related d0 ferromagnetism in MgO and the superconductivity in MgB2 do not antagonize, but rather enhance the superconducting transition temperature Tc to any significant degree. We found in superconducting MgB2-d0 ferromagnetic MgO composites that the superconducting transition proceeds in two steps. The first at the S-F interface, between 110-120 K, then in the rest of the bulk at 39 K, which is the Tc of single phase MgB2 superconductor. Moreover, the additional transition emerges at 60 K at the S-F interface especially in the ferromagnetic side, showing a spin-glass-like magnetic state. Our findings reveal that the proximity effect in the superconductor-d0 ferromagnet heterostructures will provide the knowledge and basis to enhance the Tc value of the existing superconductors., We found that the conclusions obtained are not valid

164. GZD824 Inhibits GCN2 and Sensitizes Cancer Cells to Amino Acid Starvation Stress

Abstract

Eukaryotic initiation factor 2α(eIF2α) kinase general control nonderepressible 2 (GCN2) drives cellular adaptation to amino acid limitation by activating the integrated stress response that induces activating transcription factor 4 (ATF4). Here, we found that a multikinase inhibitor, GZD824, which we identified using a cell-based assay with ATF4 immunostaining, inhibited the GCN2 pathway in cancer cells. Indeed, GZD824 suppressed GCN2 activation, eIF2αphosphorylation, and ATF4 induction during amino acid starvation stress. However, at lower nonsuppressive concentrations, GZD824 paradoxically stimulated eIF2αphosphorylation and ATF4 expression in a GCN2-dependent manner under unstressed conditions. Such dual properties conceivably arose from a direct effect on GCN2, as also observed in a cell-free GCN2 kinase assay and shared by a selective GCN2 inhibitor. Consistent with the GCN2 pathway inhibition, GZD824 sensitized certain cancer cells to amino acid starvation stress similarly to ATF4 knockdown. These results establish GZD824 as a multikinase GCN2 inhibitor and may enhance its utility as a drug under development.SIGNIFICANCE STATEMENTGZD824, as a direct general control nonderepressible 2 (GCN2) inhibitor, suppresses activation of the integrated stress response during amino acid limitation, whereas it paradoxically stimulates this stress-signaling pathway at lower nonsuppressive concentrations. The pharmacological activity we identify herein will provide the basis for the use of GZD824 to elucidate the regulatory mechanisms of GCN2 and to evaluate the potential of the GCN2–activating transcription factor 4 pathway as a target for cancer therapy.

200. Estimation of multiple light sources from specular highlights.

Abstract

This paper proposes a method for estimating the illuminant spectral power distributions and their positional relationship of multiple light sources under a complex illumination environment. A multiband camera system is used for capturing spectral images of dielectric objects in a scene. Specular highlights are used as a clue for estimating the light source information, which are detected on curved object surfaces with the different object colors. The illuminant spectra of light sources are estimated from the camera data for each highlight areas. Then, the illuminant spectral estimates are obtained for a different set of light sources. Next, positional relationship among the light sources is predicted by classifying the detected highlights and the estimated spectra using the probabilistic relaxation labeling. The feasibility of the proposed method is examined in experiments on real scenes. [ABSTRACT FROM PUBLISHER]
