Abstract

Background: The aim of this project is to process psychosomatic and child and adolescent psychiatry according to a didactic concept which aims at problem-oriented, case-related, self-directed learning.Method: Docs 'n Drugs offers favourable conditions for the realisation of this concept. Accordingly, this e-learning software was extended to be able to contextually illustrate the structure of psychodiagnostic decision making.Results: The structural extension of Docs 'n Drugs has been successful; 10 psychosomatic and paediatric and adolescent psychiatric cases could be designed and consigned to the programme. Outlook: The challenge for the future is represented by the integration of these didactic elements in university teaching. Central to this is on the one hand the accessibility of the educational objectives to the learners, and on the other the guarantee of the technical conditions for smooth running. Hintergrund: Ziel dieses Projektes ist es, psychosomatische und kinder- und jugendpsychiatrische Sachverhalte gemäß eines didaktischen Konzepts, das problemorientiertes, fallbezogenes, selbstgesteuertes Lernen anstrebt, aufzubereiten.Methode: Docs 'n Drugs bietet günstige Vorrausetzungen, um dieses Konzept zu verwirklichen. Entsprechend wurde diese e-learning Software so erweitert, dass die Struktur psychodiagnostischer Entscheidungsfindung inhaltlich abgebildet werden kann. Ergebnisse: Die strukturelle Erweiterung von Docs 'n Drugs ist gelungen, 10 psychosomatische bzw. kinder- und jugendpsychiatrische Fälle konnten entworfen und im Programm hinterlegt werden.Ausblick: Die Herausforderung für die Zukunft stellt die Integration dieser didaktischen Elemente in den universitären Unterricht dar. Abstract

Medium-chain-length polyhydroxyalkanoates (mcl-PHAs) are bacterial polyesters which are produced in nature by certain Pseudomonas strains. These biopolymers are of interest because of their chirality, biodegradability and elastomeric property, while the 3-hydroxy acid monomers are a potential source of chiral synthons. Since wild-type bacteria can only be used to a limited extent for the production of tailor-made functionalized PHAs, various recombinant bacteria have been generated in order to elucidate the PHA synthesis pathway and to control and modify the metabolic carbon flux towards PHA synthesis. Production of PHAs with altered monomer composition and physical properties has already been achieved by pathway engineering. An alternative to the in vivo synthesis strategies using whole microorganisms is PHA synthesis in vitro with isolated enzymes, which is of interest for incorporation of specific monomers that are not taken up or metabolized by bacterial cells. The various synthesis strategies are discussed in the context of the possible future production of tailor-made functionalized PHAs.

Abstract

Background: The diagnosis and management of BK virus (BKV) reactivation following renal transplantation continues to be a significant clinical problem. Following reactivation of latent virus, impaired cellular immunity enables sustained viral replication to occur in urothelial cells, which potentially leads to the development of BKVassociated nephropathy (BKVAN). Current guidelines recommend regular surveillance for BKV reactivation through the detection of infected urothelial cells in urine (decoy cells) or viral nucleic acid in urine or blood. However, these methods have variable sensitivity and cannot routinely distinguish between different viral subtypes. We therefore asked whether mass spectrometry might be able to overcome these limitations and provide an additional noninvasive technique for the surveillance of BKV and identification of recipients at increased risk of BKVAN. Results: Here we describe a mass spectrometry (MS)-based method for the detection of BKV derived proteins directly isolated from clinical urine samples. Peptides detected by MS derived from Viral Protein 1 (VP1) allowed differentiation between subtypes I and IV. Using this approach, we observed an association between higher decoy cell numbers and the presence of the VP1 subtype Ib-2 in urine samples derived from a cohort of 20 renal transplant recipients, consistent with the hypothesis that certain viral subtypes may be associated with more severe BKVAN. Conclusions: This is the first study to identify BK virus proteins in clinical samples by MS and that this approach makes it possible to distinguish between different viral subtypes. Further studies are required to establish whether this information could lead to stratification of patients at risk of BKVAN, facilitate distinction between BKVAN and acute rejection (AR), and ultimately improve patient treatment and outcomes. © 2012 Konietzny et al.; licensee BioMed Central Ltd.

Abstract

International audience; In May 1992 during the interdisciplinary measurement campaign HartX (Hartheim eXperiment), several independent estimates of stand water vapor flux were compared at a 12-m high Scots pine (Pinus silvestris) plantation on a flat fluvial terrace of the Rhine close to Freiburg, Germany. Weather during the HartX period was characterized by ten consecutive clear days with exceptionally high input of available energy for this time of year and with a slowly shifting diurnal pattern in atmospheric variables like vapor pressure deficit. Methods utilized to quantify components of stand water flux included porometry measurements on understory graminoid leaves and on pine needles and three different techniques for determining individual tree xylem sap flow. Micrometeorological methods included eddy covariance and eddy covariance energy balance techniques with six independent systems on two towers separated by 40 m. Additionally, Bowen ratio energy balance estimates of water flux were conducted and measurements of the gradients in water vapor, CO2, and trace gases within and above the stand were carried out with an additional, portable 30 m high telescoping mast. Biologically-based estimates of overstory transpiration were obtained by up-scaling tree sap flow rates to stand level via cumulative sapwood area. Tree transpiration contributed between 2.2 and 2.6 mm/day to ET for a tree leaf area index (LAI) of 2.8. The pine stand had an understory dominated by sedge and grass species with overall average LAI of 1.5. Mechanistic canopy gas exchange models that quantify both water vapor and CO2 exchange were applied to both understory and tree needle ecosystem compartments. Thus, the transpiration by graminoid species was estimated at approximately 20% of total stand ET. The modelled estimates for understory contribution to stand water flux compared well with micrometeorologically-based determinations. Maximum carbon gain was estimated from the canopy models at approximately 425 mmol/(m2day) for the tree needles and at 100 mmol/(m2day) for the understory. Carbon gain was suggested by the modelling analysis to remain relatively constant during the HartX period, while water use efficiency in carbon fixation increased with decreasing vapor pressure deficit. Biologically- and micrometeorologically-based estimates of stand water flux showed good general agreement with variation of up to 20% that reflects both errors due to the inherent assumptions associated with different methods as well as natural spatial variability in fluxes. The various methods support a reliable estimate of average ET from this homogeneous canopy during HartX of about 2.6 mm/day (a maximum of about 3.1 mm/day) with an insignificant decreasing trend in correlation with decreasing vapor pressure deficit and possibly soil moisture. Findings during HartX were embedded in local scale heterogeneity with greater roughness over the forest and much higher ET over the surrounding agricultural fields which results in weak but clearly existant circulation patterns. A variety of measurements were continued after the HartX campaign. They allow us to extend our findings for six months with changing environmental conditions, including shortage of soil moisture. Hydrological estimates of soil water extractions and micrometeorological estimates of ET by the one-propeller eddy covariance (OPEC) system were in very good agreement, supporting the use of this robust eddy covariance energy balance technique for long-term monitoring.
