160. Efficacy and safety of biosimilar trastuzumab (CT-P6) in routine clinical practice in the Republic of Korea: a real-world post-marketing surveillance study

Abstract

ABSTRACTBackgroundThe trastuzumab biosimilar CT-P6 is approved for human epidermal growth factor receptor 2 (HER2)–positive early breast cancer (EBC), metastatic breast cancer (MBC), and metastatic gastric cancer (MGC). The objective of this post-marketing surveillance (PMS) study was to evaluate the real-world safety and effectiveness of CT-P6 in patients with HER2-positive cancers.Research design and methodsThis open-label, observational, prospective, PMS study collected data via investigator surveys from 35 centers in the Republic of Korea (5 October 2018–4 October 2022). Eligible patients with HER2-positive EBC, MBC, or MGC started CT-P6 treatment during routine clinical practice, followed by 1-year observation. Evaluations included adverse events (AEs), adverse drug reactions (ADRs), and effectiveness.ResultsSafety was analyzed in 642 patients (494 EBC, 94 MBC, 54 MGC). Overall, 325 (50.6%) patients experienced 1316 AEs, and 550 ADRs occurred in 199 (31.0%) patients. Unexpected ADRs occurred in 62 (9.7%) patients. Unexpected ADRs and ADRs of special interest did not raise any new safety signals. Among trastuzumab-naïve patients, 34/106 (32.1%) with EBC achieved pathological complete response; 30/74 (40.5%) MBC and 24/49 (49.0%) MGC patients achieved complete or partial response.ConclusionsIn a real-world setting, CT-P6 demonstrated safety and efficacy findings consistent with previous CT-P6 studies.

Abstract

Electricity generation using the piezoelectric effect is an important energy harvesting method. In this study, a solar radiation pressure‐driven crater‐shaped light pressure electric generator (LPEG) device with a Pb(Zr0.52, Ti0.48)O3 (PZT) piezoelectric layer and grained Ag layer is fabricated on GaAs(100) wafer. The electrical output of the device is improved by adsorbing Au nanoparticles (AuNPs) on the Ag layer with the surface of closely connected Ag nanoparticles (AgNPs). By controlling the size and concentration of the AuNPs, a maximum power density of 867.5 µW cm−2 is obtained under a solar simulator (AM 1.5G), which represents a 151.7% improvement over the case without AuNP adsorption. The results of Raman spectra, finite‐difference time‐domain (FDTD) simulations, and COMSOL Multiphysics demonstrate that the newly formed hotspots between AgNPs─AuNPs and AuNPs─AuNPs enhance the electric field of the incident light significantly and extend the region where the electric field is maximally amplified to 600–800 nm, resulting in increased solar radiation pressure on the PZT piezoelectric layer. Abstract

To increase mechanical strength of porous ceramics, here, an effective two‐step sintering (TSS) technique capable of producing highly porous alumina with enhanced mechanical strength is suggested. Based on the sintering theories, here, a significantly lower activation energy for densification at low temperature region allowed a beneficial temperature range for the TSS to be deduced. With a specific TSS regime (T1 = 1550°C and T2 = 1400°C), significantly higher compressive strength levels (8.00–15.24 MPa) were measured with an apparent porosity of 56.49% compared to conventional sintering (1.03–1.86 MPa) with similar apparent porosity of 57.84%. Specifically, another TSS regime (T1 = 1550°C and T2 = 1380°C) left submicron‐sized open pores within the lamellar walls, providing a hierarchical porous structure with enhanced mechanical strength. An evaluation of the mechanical stability by a finite element analysis indicated outstanding compressive strength even with small pores in the lamella walls. [ABSTRACT FROM AUTHOR]

Abstract

BackgroundA large number of genetic variations have been identified in rice. Such variations must in many cases control phenotypic differences in abiotic stress tolerance and other traits. A single feature polymorphism (SFP) is an oligonucleotide array-based polymorphism which can be used for identification of SNPs or insertion/deletions (INDELs) for high throughput genotyping and high density mapping. Here we applied SFP markers to a lingering question about the source of salt tolerance in a particular rice recombinant inbred line (RIL) derived from a salt tolerant and salt sensitive parent.ResultsExpression data obtained by hybridizing RNA to an oligonucleotide array were analyzed using a statistical method called robustified projection pursuit (RPP). By applying the RPP method, a total of 1208 SFP probes were detected between two presumed parental genotypes (Pokkali and IR29) of a RIL population segregating for salt tolerance. We focused on the Saltol region, a major salt tolerance QTL. Analysis of FL478, a salt tolerant RIL, revealed a small (< 1 Mb) region carrying alleles from the presumed salt tolerant parent, flanked by alleles matching the salt sensitive parent IR29. Sequencing of putative SFP-containing amplicons from this region and other positions in the genome yielded a validation rate more than 95%.ConclusionRecombinant inbred line FL478 contains a small (< 1 Mb) segment from the salt tolerant parent in the Saltol region. The Affymetrix rice genome array provides a satisfactory platform for high resolution mapping in rice using RNA hybridization and the RPP method of SFP analysis.
