166. Tetramic acid-motif natural products from a marine fungus Tolypocladium cylindrosporumFB06 and their anti-Parkinson activities

Abstract

Tetramic acid-containing natural products are attracting significantly increasing attention from biologists and chemists due to their intriguing structures and biological activities. In the present study, two new tetramic acid alkaloids tolypyridone I (1) and tolypyridone J (2), together with five known ones (3–7), were isolated from cultures of a marine fungus Tolypocladium cylindrosporumFB06 isolate obtained from a marine sediment in Beaufort sea of North Alaska. Their structures were elucidated using 1D, 2D NMR, and HRESIMS. Their configurations were established on the basis of 1H coupling constants, ROESY correlations and DP4 calculations. Compound 2was isolated as mixtures of rotational isomers with C-3 to C-7 axis between 4-hydroxy-2-pyridone and 1-ethyl-3,5-dimethylcyclohexane, hindering rotation. In our unbiased screening to discover neuroprotective compounds in an in vitro Parkinson’s disease (PD) model, SH-SY5Y dopaminergic cells were treated with isolated compounds followed by treatment with 1-methyl-4-phenylpyridinium (MPP+), a parkinsonian neurotoxin. Among tested compounds, F-14329 (7) significantly protected cells from MPP+-induced cytotoxicity. MPP+-mediated cell death is known to be related to the regulation of Bcl-2 family proteins, specifically the down-regulation of anti-apoptotic Bcl-2 and the up-regulation of pro-apoptotic Bax levels. Treatment with 2 mmol/L of MPP+for 24 h significantly reduced Bcl-2 levels compared to control treated with vehicle. However, treatment with F-14329 (7) attenuated such reduction. This study demonstrates that tetramic acid-motif compounds could be potential lead compounds for treating PD.

173. An Investigation of Concentration and Health Impacts of Aldehydes Associated with Cooking in 29 Residential Buildings

Abstract

Indoor air quality can be influenced by various indoor activities. The indoor air pollutants generated by cooking activities can cause severe risks to occupants’ health in residential buildings. The present study conducted field experiments to measure indoor pollutants associated with cooking in 29 residential buildings. Due to an open plan in Korean residential buildings, the emission of indoor pollutants was measured in the kitchen and living room. Focusing on aldehydes, the indoor emission levels for various cooking methods such as grilling, frying, and boiling were analyzed. As a result, the emission of formaldehyde, acetaldehyde, and propionaldehyde was highly increased for all cooking methods. The increase rate of the emission was higher in the kitchen than that in the living room for grilling and frying. In the case of boiling, the highest concentration of aldehydes was observed. Moreover, the indoor level of aldehydes was higher in the living room than that in the kitchen. Moreover, the health risk such as cancer for occupants was assessed based on the measured data for different cooking methods. The assessment results showed that all the emissions of aldehydes for different cooking methods required instant actions to avoid cancer risk for occupants.

193. CFD simulation analysis on integrated operation of range-hood and make-up air supply for cooking-generated particulate matter

Abstract

One of the most important problems of cooking-generated particulate matter (PM) is that it rapidly disperses when the range hood is in operation during cooking. To improve the performance of the range hood and prevent the dispersion of PM, a supply of make-up air equivalent to the airflow rate of the range hood should be provided. In this regard, we place an auxiliary supply system as a make-up supply to solve such problems. The objective of this study is to evaluate the performance of the make-up air supply system and the range hood. To evaluate this system, several case studies were performed involving CFD simulations. The auxiliary supply system is optimized through three types of variables (size of diffuser, distance from the source, and flow angle). An increase in the length of the diffuser causes PM dispersion to decrease. The installation of the diffuser at a certain distance from the emission source is effective in preventing dispersion of cooking-generated PM. In the building analyzed in this study, supplying the make-up air at an angle of 10° was observed to be most effective.
