Abstract Electronic fibers used to fabricate wearable triboelectric nanogenerator (TENG) for harvesting human mechanical energy have been extensively explored. However, little attention is paid to their mutual advantages of environmental friendliness, mechanical properties, and stability. Here, we report a super-strong, biodegradable, and washable cellulose-based conductive macrofibers, which is prepared by wet-stretching and wet-twisting bacterial cellulose hydrogel incorporated with carbon nanotubes and polypyrrole. The cellulose-based conductive macrofibers possess high tensile strength of 449 MPa (able to lift 2 kg weights), good electrical conductivity (~ 5.32 S cm−1), and excellent stability (Tensile strength and conductivity only decrease by 6.7% and 8.1% after immersing in water for 1 day). The degradation experiment demonstrates macrofibers can be degraded within 108 h in the cellulase solution. The designed fabric-based TENG from the cellulose-base conductive macrofibers shows a maximum open-circuit voltage of 170 V, short-circuit current of 0.8 µA, and output power at 352 μW, which is capable of powering the commercial electronics by charging the capacitors. More importantly, the fabric-based TENGs can be attached to the human body and work as self-powered sensors to effectively monitor human motions. This study suggests the potential of biodegradable, super-strong, and washable conductive cellulose-based fiber for designing eco-friendly fabric-based TENG for energy harvesting and biomechanical monitoring.

Abstract Background Cutaneous squamous cell carcinoma (cSCC) is the second most common type of skin cancer, the prognosis for patients with metastatic cSCC remains relatively poor. Thus, there is an urgent need to identify new diagnostic, prognostic, and therapeutic targets and pathways in cSCC. Results It detected a total of 37,507 lncRNA probes and 32,825 mRNA probes and found 3593 differentially expressed lncRNAs and 3236 differentially expressed mRNAs. It has been found that mRNAs ACY3, NR1D1, MZB1 has co-expression relationship with six lncRNAs, GXYLT1P3, LINC00348, LOC101928131, A-33-p3340852, A-21-p0003442 and LOC644838. Conclusions The aim of this study is to identify cSCC-specific lncRNAs and indicated that six unstudied lncRNAs may serve an important role in endoplasmic reticulum stress apoptosis, autophagy and the progression of cSCC by modulating ACY3, NR1D1 and MZB1.

Abstract Background After traumatic brain injury (TBI), an acute, robust inflammatory cascade occurs that is characterized by the activation of resident cells such as microglia, the migration and recruitment of peripheral immune cells and the release of inflammatory mediators that induce secondary cell death and impede neurological recovery. In addition, neuroinflammation can alter blood–brain barrier (BBB) permeability. Controlling inflammatory responses is considered a promising therapeutic approach for TBI. Hydroxychloroquine (HCQ) has already been used clinically for decades, and it is still widely used to treat various autoimmune diseases. However, the effects of HCQ on inflammation and the potential mechanism after TBI remain to be defined. The aim of the current study was to elucidate whether HCQ could improve the neurological recovery of mice post-TBI by inhibiting the inflammatory response via the TLR4/NF-κB signaling pathway. Methods C57BL/6 mice were subjected to controlled cortical impact (CCI) and randomly divided into groups that received intraperitoneal HCQ or vehicle daily after TBI. TAK-242 (3.0 mg/kg), an exogenous TLR4 antagonist, was injected intraperitoneally 1 h before TBI. Behavioral assessments were performed on days 1 and 3 post-TBI, and the gene expression levels of inflammatory cytokines were analyzed by qRT-PCR. The presence of infiltrated immune cells was examined by flow cytometry and immunostaining. In addition, BBB permeability, tight junction expression and brain edema were investigated. Results HCQ administration significantly ameliorated TBI-induced neurological deficits. HCQ alleviated neuroinflammation, the activation and accumulation of microglia and immune cell infiltration in the brain, attenuated BBB disruption and brain edema, and upregulated tight junction expression. Combined administration of HCQ and TAK-242 did not enhance the neuroprotective effects of HCQ. Conclusions HCQ reduced proinflammatory cytokine expression, and the underlying mechanism may involve suppressing the TLR4/NF-κB signaling pathway, suggesting that HCQ is a potential therapeutic agent for TBI treatment.

Abstract We calculate the single-particle excitation spectrum and the Landau liquid parameters for the archetypal model of solids, the three-dimensional uniform electron gas, with the variational diagrammatic Monte Carlo method, which gives numerically controlled results without systematic error. In the metallic range of density, we establish benchmark values for the wave-function renormalization factor Z, the effective mass $$m^*/m$$ m ∗ / m , and the Landau parameters $$F_0^s$$ F 0 s and $$F_0^a$$ F 0 a with unprecedented accuracy, and we resolve the long-standing puzzle of non-monotonic dependence of mass on density. We also exclude the possibility that experimentally measured large reduction of bandwidth in Na metal can originate from the charge and spin fluctuations contained in the model of the uniform electron gas.

The suppression of uncommanded motion of aircraft at high angles of attack (α) is a research topic of continuous concern in the aviation field. Aiming at the suppression of uncommanded roll-yaw motion of a canard aircraft at high angles of attack, an experimental method of virtual flight test based on reinforcement learning is proposed in this paper. In the virtual flight experiment, the agent was trained to control the jet actuators, so as to suppress the uncommanded roll-yaw motion. Force measurements were conducted to obtain the performance of the jet actuators in a low-speed wind tunnel. The results show that when the spanwise jet actuator and the reverse jet actuator were working on the same side, their control effects were suppressed by each other. Then, the stability augmentation control law was trained through virtual flight experiments based on a reinforcement learning algorithm (TD3), and the uncommanded motion was successfully suppressed. The time histories of the reinforcement learning agent’s action in tests were analyzed, showing that the agent can avoid the coupling relationship between two kinds of jet actuators during tests.

ObjectivePreventing suicide in US youth is of paramount concern, with rates increasing over 50% between 2007 and 2018. Statistical modeling using electronic health records may help identify at-risk youth before a suicide attempt. While electronic health records contain diagnostic information, which are known risk factors, they generally lack or poorly document social determinants (e.g., social support), which are also known risk factors. If statistical models are built incorporating not only diagnostic records, but also social determinants measures, additional at-risk youth may be identified before a suicide attempt.MethodsSuicide attempts were predicted in hospitalized patients, ages 10-24, from the State of Connecticut's Hospital Inpatient Discharge Database (HIDD; N = 38943). Predictors included demographic information, diagnosis codes, and using a data fusion framework, social determinants features transferred or fused from an external source of survey data, The National Longitudinal Study of Adolescent to Adult Health (Add Health). Social determinant information for each HIDD patient was generated by averaging values from their most similar Add Health individuals (e.g., top 10), based upon matching shared features between datasets (e.g., Pearson's r). Attempts were then modelled using an elastic net logistic regression with both HIDD features and fused Add Health features.ResultsThe model including fused social determinants outperformed the conventional model (AUC = 0.83 v. 0.82). Sensitivity and positive predictive values at 90 and 95% specificity were almost 10% higher when including fused features (e.g., sensitivity at 90% specificity = 0.48 v. 0.44). Among social determinants variables, the perception that their mother cares and being non-religious appeared particularly important to performance improvement.DiscussionThis proof-of-concept study showed that incorporating social determinants measures from an external survey database could improve prediction of youth suicide risk from clinical data using a data fusion framework. While social determinant data directly from patients might be ideal, estimating these characteristics via data fusion avoids the task of data collection, which is generally time-consuming, expensive, and suffers from non-compliance.

BackgroundPrevious studies on the management of chronic subdural hematoma (cSDH) mainly focused on the risk of postoperative recurrence and measures to prevent it. In this study, we propose the use of a non-invasive postoperative treatment method, the modified Valsalva maneuver (MVM), as a means of reducing the recurrence of cSDH. This study aims to clarify the effects of MVM on functional outcomes and recurrence rates.MethodsA prospective study was conducted at the Department of Neurosurgery, Tongji Hospital, Tongji Medical College, Huazhong University of Science and Technology from November 2016 to December 2020. The study included 285 adult patients who underwent burr-hole drainage for the treatment of cSDH and received subdural drains. These patients were divided into two groups: the MVM group (n = 117) and the control group (n = 98). In the MVM group, patients received treatment with a customized MVM device for at least 10 times per hour, 12 h per day. The study's primary endpoint was the recurrence rate of SDH, while functional outcomes and morbidity 3 months after surgery were the secondary outcomes.ResultsIn the current study, 9 out of 117 patients (7.7%) in the MVM group experienced a recurrence of SDH, while 19 out of 98 patients (19.4%, p < 0.05) in the HC group experienced a recurrence of SDH. Additionally, the infection rate of diseases such as pneumonia (1.7%) was significantly lower in the MVM group compared to the HC group (9.2%, p < 0.001, odds ratio (OR = 0.1). After 3 months of the surgery, 109 out of 117 patients (93.2%) in the MVM group achieved a favorable prognosis, compared to 80 out of 98 patients (81.6%) in the HC group (p = 0.008, with an OR of 2.9). Additionally, infection rate (with an OR of 0.2) and age (with an OR of 0.9) are independent predictors of a favorable prognosis at the follow-up stage.ConclusionsThe use of MVM in the postoperative management of cSDHs has been shown to be safe and effective, resulting in reduced rates of cSDH recurrence and infection following burr-hole drainage. These findings suggest that MVM treatment may lead to a more favorable prognosis at the follow-up stage.

Peanut is an important food and feed crop, providing oil and protein nutrients. Germins and germin-like proteins (GLPs) are ubiquitously present in plants playing numerous roles in defense, growth and development, and different signaling pathways. However, the GLP members have not been comprehensively studied in peanut at the genome-wide scale. We carried out a genome-wide identification of the GLP genes in peanut genome. GLP members were identified comprehensively, and gene structure, genomic positions, motifs/domains distribution patterns, and phylogenetic history were studied in detail. Promoter Cis-elements, gene duplication, collinearity, miRNAs, protein-protein interactions, and expression were determined. A total of 84 GLPs (AhGLPs ) were found in the genome of cultivated peanut. These GLP genes were clustered into six groups. Segmental duplication events played a key role in the evolution of AhGLPs, and purifying selection pressure was underlying the duplication process. Most AhGLPs possessed a well-maintained gene structure and motif organization within the same group. The promoter regions of AhGLPs contained several key cis-elements responsive to ‘phytohormones’, ‘growth and development’, defense, and ‘light induction’. Seven microRNAs (miRNAs) from six families were found targeting 25 AhGLPs. Gene Ontology (GO) enrichment analysis showed that AhGLPs are highly enriched in nutrient reservoir activity, aleurone grain, external encapsulating structure, multicellular organismal reproductive process, and response to acid chemicals, indicating their important biological roles. AhGLP14, AhGLP38, AhGLP54, and AhGLP76 were expressed in most tissues, while AhGLP26, AhGLP29, and AhGLP62 showed abundant expression in the pericarp. AhGLP7, AhGLP20, and AhGLP21, etc., showed specifically high expression in embryo, while AhGLP12, AhGLP18, AhGLP40, AhGLP78, and AhGLP82 were highly expressed under different hormones, water, and temperature stress. The qRT-PCR results were in accordance with the transcriptome expression data. In short, these findings provided a foundation for future functional investigations on the AhGLPs for peanut breeding programs.

Summary: CCR7-triggered DC migration toward draining lymph nodes is critical for the initiation of protective immunity and maintenance of immune tolerance. How to promote CCR7-mediated DC migration to determine T cell responses under inflammatory and homeostatic conditions remains poorly understood. Here we demonstrate that the Extl1 (Exostosin like glycosyltransferase 1) promotes CCR7-triggered DC migration in a heparan sulfate proteoglycans (HSPG)-dependent manner. Mechanistically, Extl1 mediates HSPG production via its glycosyltransferase domain to inhibit C1q expression. Extl1/HSPG axis relieves C1q-mediated restriction of CCR7 surface expression and internalization, and thus enhances CCR7-dependent migratory signaling activation. Consequently, Extl1 is required for DC-mediated Th1 and Th17 responses in immune defense against bacterial infection and for Treg cell development in the prevention of autoimmunity. Our study adds mechanistic insights to the regulation of CCR7-triggered DC migration in immunity and tolerance and provides a potential target for the treatment of infectious and autoimmune diseases.

Background: Epidemiological studies have reported an association between traffic-related pollution with risk of metabolic syndrome (MetS). However, evidence from prospective studies on the association of walkability and nitrogen dioxide (NO2) with MetS is still scarce. We, therefore, aimed to evaluate the association of long-term exposure to NO2 and walkability with hazards of incident MetS. Methods: A total of 17,965 participants without MetS diagnosed within one year at baseline were included in our study from a population-based prospective cohort in Yinzhou District, Ningbo, Zhejiang Province, China. Participants were followed up by the regional Health Information System (HIS) until December 15, 2021. MetS was defined based on the criteria of Chinese Diabetes Society (CDS2004). We used walkscore tools, calculating with amenity categories and decay functions, and spatial–temporal land-use regression (LUR) models to estimate walkability and NO2 concentrations. We used Cox proportional hazards regression models to examine the association of walkability and NO2 with hazards of MetS incidence reporting with hazard ratios (HRs) and 95% confidence intervals (CIs). Results: Overall, we followed up 77,303 person-years and identified 4040 incident cases of MetS in the entire cohort. Higher walkability was inversely associated with incident MetS (HR = 0.94, 95 % CI: 0.91–0.99), whereas NO2 was positively associated with MetS incidence (HR = 1.07, 95 %CI: 1.00–1.15) per interquartile range increment in two-exposure models. Furthermore, we found a significant multiplicative interaction between walkability and NO2. Bacterial wilt disease (BWD), caused by Ralstonia solanacearum is a major challenge for peanut production in China and significantly affects global peanut field productivity. It is imperative to identify genetic loci and putative genes controlling resistance to R. solanacearum (RRS). Therefore, a sequencing-based trait mapping approach termed “QTL-seq” was applied to a recombination inbred line population of 581 individuals from the cross of Yueyou 92 (resistant) and Xinhuixiaoli (susceptible). A total of 381,642 homozygous single nucleotide polymorphisms (SNPs) and 98,918 InDels were identified through whole genome resequencing of resistant and susceptible parents for RRS. Using QTL-seq analysis, a candidate genomic region comprising of 7.2 Mb (1.8–9.0 Mb) was identified on chromosome 12 which was found to be significantly associated with RRS based on combined Euclidean Distance (ED) and SNP-index methods. This candidate genomic region had 180 nonsynonymous SNPs and 14 InDels that affected 75 and 11 putative candidate genes, respectively. Finally, eight nucleotide binding site leucine rich repeat (NBS-LRR) putative resistant genes were identified as the important candidate genes with high confidence. Two diagnostic SNP markers were validated and revealed high phenotypic variation in the different resistant and susceptible RIL lines. These findings advocate the expediency of the QTL-seq approach for precise and rapid identification of candidate genomic regions, and the development of diagnostic markers that are applicable in breeding disease-resistant peanut varieties.

Ferroptosis is a recognized novel form of programmed cell death pathway, featuring abnormalities in iron metabolism, SystemXc−/glutathione axis, and lipid peroxidation regulation. A variety of ferroptosis inducers can influence glutathione peroxidase directly or indirectly via diverse pathways, leading to decreased antioxidant capacity, accumulated cellular lipid peroxides, and finally inducing ferroptosis. To date, mounting studies confirm the association of ferroptosis with various cutaneous diseases, including skin homeostasis, neoplastic diseases, infectious diseases, genetic skin disease, inflammatory skin diseases, and autoimmune diseases. There are shared characteristics regarding ferroptosis and various cutaneous diseases in terms of pathophysiological mechanisms, such as oxidative stress associated with iron metabolism disorder and accumulated lipid peroxides. Therefore, we summarize the current knowledge regarding the mechanisms involved in the regulation of ferroptosis for further discussion of its role in the pathogenesis and prognosis of skin diseases. Gaining insight into the underlying mechanisms of ferroptosis and the associated dermatological disorders could illuminate the pathogenesis and treatments of different cutaneous diseases.

Background: Some studies have reported to use some predictors before extracorporeal membrane oxygenation (ECMO) initiation to predict the acute kidney injury (AKI) risk. However, injury during the ECMO operation and the response of patients to ECMO may significantly influence the prognosis, and they are unpredictable before ECMO initiation. This study aims to develop a potential model based clinical characteristics at the 2-hour time point during ECMO for the early prediction of AKI in patients receiving ECMO. Methods: 139 patients who underwent ECMO were enrolled in this study. The clinical characteristics and the laboratory examinations at 2-hour time point during ECMO were recorded. The least absolute shrinkage and selection operator (LASSO) regression method was performed to select predictors, and logistic regression and a nomogram were used to establish the prediction model. The area under curve (AUC) of the receiver operating characteristic and calibration curve were used to analyze the discrimination and calibration of the model. K-fold cross-validation method was performed to validate the accuracy of this model. Results: Among the 139 patients receiving ECMO, 106 participants (76.26%) developed AKI. Four predictive variables including ECMO model, serum creatinine (Scr-2h), uric acid(UA-2h), and serum lactate (Lac-2h) at the 2-hour time point during ECMO were filtered from 39 clinical parameters by LASSO regression. These four predictors were incorporated to develop a model for predicting AKI risk using logistic regression. The AUC of the model was 0.905 (0.845–0.965), corresponding to 81.1% sensitivity, 90.9% specificity and 83.5% accuracy. Moreover, this model showed good consistency between observed and predicted probability based on the calibration curve (P > 0.05). The validation performed by K-fold cross-validation method showed that the accuracy was 0.874 ± 0.006 in training sets, 0.827 ± 0.053 in test sets, indicating a good capability for AKI risk prediction. Finally, a nomogram based on this model was constructed to facilitate its use in clinical practice. Conclusion: The nomogram incorporating Scr-2h,Lac-2h, UA-2h, and ECMO model may facilitate the individualized prediction of the AKI risk among patients undergoing ECMO.

Leaves are one of the vegetative organs of plants that are essential for plant growth and development. PIN-FORMED (PINs) gene is an indoleacetic acid (IAA) transporter that plays a critical role in leaf development. To determine the function of BpPIN3 in leaf polarity formation in Betula pendula, the transgenic lines with BpPIN3 overexpression (OE) and BpPIN3-reduced expression (RE) were analyzed using the Agrobacterium-mediated method. The RE lines displayed the characteristics of leaf margin adaxial upward curling, with lower expression of BpPIN3 resulting in greater rolling. Tissue localization of IAA in the auxin GUS reporter system proved that auxin in the RE was mainly distributed in the secondary veins, palisade tissues, and epidermal cells in the leaf margin area. The auxin content in the leaf margin area was significantly greater than that in the main vein tissue. The cell density of the palisade tissue and the ratio of palisade tissue to spongy tissue in the curled leaf margin of the RE lines were found to be significantly decreased. RNA-seq analysis revealed that the RE hormone-signaling pathway genes were significantly enriched compared with those of the OE and WT lines; in particular, the auxin response-related genes SAURs (i.e., SAUR23, SAUR24, SAUR28, and SAUR50) and GH3.10 were found to be significantly upregulated. qRT-PCR analysis indicated that BpPIN3 expression at the leaf margin was significantly lower than that near the main vein in the RE lines. In contrast, the expression levels of SAURs and GH3.10 were significantly higher than those near the midrib. In conclusion, BpPIN3 regulates the expression of auxin response-related genes and the polar transport of auxin to change the polar form of the proximal and distal axes of birch leaves.

With the rapid increase in vehicles, the load on the asphalt pavement increases, which aggravates the appearance of rutting disease. In order to establish an effective rutting depth prediction model, this study carried out accelerated loading tests (ALT) of asphalt pavement at three different temperature environments of 60 °C, 45 °C, and 30 °C for Ji-Qing Expressway at the end of service. The rutting depth of the test was measured using a Lr-70/700 laser rut measuring instrument, and the relationship between the rutting depth of asphalt pavement and the temperature change was examined. At the ambient temperature of 60 °C, three different loading speeds of 3.5 km/h, 4.5 km/h, and 5.5 km/h were tested to analyze the influence of load frequency on rutting depth. With the SPSS analysis software, the regression analysis of the collected test data was carried out to modify the rutting prediction model established by the indoor improved MTS test. The results show that when the loading speed is increased from 3.5 km/h to 4.5 km/h, the rutting depth can be reduced by 11%. When the loading speed increases from 4.5 km/h to 5.5 km/h, the rut depth can be reduced by 15.8%. When the loading temperature increases from 30 °C to 45 °C, the rut depth increases by 2.15 times. When the loading temperature increased from 45 °C to 60 °C, the rutting depth increased by 0.46 times.

Primary cilia have emerged as the cellular “antenna” that can receive and transduce extracellular chemical/physical signals, thus playing an important role in regulating cellular activities. Although the electromagnetic field (EMF) is an effective treatment for bone fractures since 1978, however, the detailed mechanisms leading to such positive effects are still unclear. Primary cilia may play a central role in receiving EMF signals, translating physical signals into biochemical information, and initiating various signalingsignaling pathways to transduce signals into the nucleus. In this review, we elucidated the process of bone healing, the structure, and function of primary cilia, as well as the application and mechanism of EMF in treating fracture healing. To comprehensively understand the process of bone healing, we used bioinformatics to analyze the molecular change and associated the results with other studies. Moreover, this review summarizedsummarized some limitations in EMFs-related research and provides an outlook for ongoing studies. In conclusion, this review illustrated the primary cilia and related molecular mechanisms in the EMF-induced bone healing process, and it may shed light on future research.

Psoriasis is a chronic inflammatory immune skin disease mediated by genetic and environmental factors. As a bridge between innate and adaptive immunity, mast cells are involved in the initiation, development, and maintenance of psoriasis by interactions and communication with a variety of cells. The current review describes interactions of mast cells with T cells, Tregs, keratinocytes, adipocytes, and sensory neurons in psoriasis to emphasize the important role of mast cell-centered cell networks in psoriasis.

Investigating neural mechanisms of anesthesia process and developing efficient anesthetized state detection methods are especially on high demand for clinical consciousness monitoring. Traditional anesthesia monitoring methods are not involved with the topological changes between electrodes covering the prefrontal-parietal cortices, by investigating electrocorticography (ECoG). To fill this gap, a framework based on the two-stream graph convolutional network (GCN) was proposed, i.e., one stream for extracting topological structure features, and the other one for extracting node features. The two-stream graph convolutional network includes GCN Model 1 and GCN Model 2. For GCN Model 1, brain connectivity networks were constructed by using phase lag index (PLI), representing different structure features. A common adjacency matrix was founded through the dual-graph method, the structure features were expressed on nodes. Therefore, the traditional spectral graph convolutional network can be directly applied on the graphs with changing topological structures. On the other hand, the average of the absolute signal amplitudes was calculated as node features, then a fully connected matrix was constructed as the adjacency matrix of these node features, as the input of GCN Model 2. This method learns features of both topological structure and nodes of the graph, and uses a dual-graph approach to enhance the focus on topological structure features. Based on the ECoG signals of monkeys, results show that this method which can distinguish awake state, moderate sedation and deep sedation achieved an accuracy of 92.75% in group-level experiments and mean accuracy of 93.50% in subject-level experiments. Our work verifies the excellence of the graph convolutional network in anesthesia monitoring, the high recognition accuracy also shows that the brain network may carry neurological markers associated with anesthesia.

The Reconfigurable Intelligent Surface (RIS) constitutes one of the prominent technologies for the next generation of wireless communications. It is envisioned to enhance the signal coverage in cases when the direct link of the communication is weak. Recently, beam training based on codebook selection is proposed to obtain the optimized phase configuration of the RIS. After that, the data is transmitted and received by using the classical coherent demodulation scheme (CDS). This training approach is able to avoid the large overhead required by the channel sounding process, and it also circumvents complex optimization problems. However, the beam training still requires the transmission of some reference signals to test the different phase configurations of the codebook, and the best codeword is chosen according to the measurement of the received energy of the reference signals. Then, the overhead due to the transmission of reference signals reduces the spectral efficiency. In this paper, a zero overhead beam training for RIS is proposed, relying on data transmission and reception based on non-CDS (NCDS). At the BS, the received differential data can also be used for the determination of the best beam for the RIS. Therefore, the efficiency of the system is significantly enhanced since reference signals are fully avoided. After choosing the best codebook, NCDS is still more suitable to transmit information for high mobility scenarios as compared to the classical CDS. Analytical expressions for the Signal-to-Interference and Noise Ratio (SINR) for the non-coherent RIS-empowered system are presented. Moreover, a detailed comparison between the NCDS and CDS in terms of efficiency and complexity is also given. The extensive computer simulation results verify the accuracy of the presented analysis and showcase that the proposed system outperforms the existing solutions.

The gravity recovery and climate experiment (GRACE) provides an unprecedented opportunity to detect the spatial and temporal variation of the terrestrial water storage (TWS) for regional to continental scales. However, the GRACE system's coarse temporal resolution (∼monthly) and data discontinuity missing perplexed the TWS research during the operation. In this article, the data assimilation (DA) method was employed to integrate the vertical deformation obtained from the small baseline subset (SBAS) InSAR processing into the NASA catchment land surface model (CLSM), which improved the estimation of the TWS. First, we used a one-dimensional ensemble Kalman filter for DA research to estimate the TWS in Dali Prefecture, southwestern China. Finally, we compared the estimated TWS with the GRACE-based TWS from December 2, 2018 to January 21, 2021. The unbiased root-mean-square of the open loop (OL; without DA) method and the SBAS-InSAR DA method are 61 mm and 30 mm in Dali Prefecture, respectively. Results revealed that the numerical difference between the estimated TWS and the GRACE TWS retrievals was significantly decreased by the SBAS-InSAR DA method than the OL method. In addition, the temporal resolution of the SBAS-InSAR DA-based TWS was improved to 12 days compared with GRACE-based TWS. Furthermore, we recovered the discontinuous deletion and blank of GRACE-based TWS from 2015 to 2018 by the SBAS-InSAR DA method.

3-nitro-1,2,4-triazol-5-one (NTO) is the main component of insensitive munitions (IM) formulation because of its outstanding insensitive properties. In this paper, a series of NTO/HMX-based composite explosives were prepared and characterized. The study focuses on the effect of NTO on the performance of the formulations, especially the safety performance. The results revealed that the mechanical sensitivity of formulations was associated with NTO content, as well as the thermal conductivity, specific heat capacity and Arrhenius parameters. Then, the high amount of NTO using in formulation was proved to be helpful for NTO/HMX-based formulation to exhibit good thermal safety. Besides, by accelerating rate calorimeter (ARC) and a modified cook-off equipment, the pressure and pressure rise rate were proved as the important indicator for judging the thermal safety performance in confined spaces. Finally, the numerical simulation was used as a credible method for predicting the respond temperature of cook-off experiment.

Identification of clinically applicable molecular subtypes of pancreatic ductal adenocarcinoma (PDAC) is crucial to improving patient outcomes. However, the traditional tissue-dependent transcriptional subtyping strategies are invasive and not amenable to routine clinical evaluation. In this study, we developed a circulating extracellular vesicle (cEV) long RNA (exLR)-based PDAC subtyping method and provided exLR-derived signatures for predicting immunogenic features and clinical outcomes in PDAC. We enrolled 426 individuals, among which 227 PDACs served as an internal cohort, 118 PDACs from two other medical centers served as an independent validation cohort, and 81 healthy individuals served as the control. ExLR sequencing was performed on all plasma samples. We found that PDAC could be categorized into three subtypes based on plasma exLR profiles. Each subpopulation showed its own molecular features and was associated with patient clinical prognosis. The immunocyte-derived cEV fractions were altered among PDAC subtypes and interconnected with tumor-infiltrating lymphocytes in cancerous tissue. Additionally, we found a significant concordance of immunoregulators between tissue and blood EVs, and we harvested potential PDAC therapeutic targets. Most importantly, we constructed a nine exLR-derived, tissue-applicable signature for prognostic assessment of PDAC. The circulating exLR-based features may offer an attractive platform for personalized treatment and predicting patient outcomes in multiple types of cancer.

Intracranial aneurysms (IAs) are highly prevalent in the population, and their rupture poses a significant risk of death or disability. However, the treatment of aneurysms, whether through interventional embolization or craniotomy clipping surgery, is not always safe and carries a certain proportion of morbidity and mortality. Therefore, early detection and prompt intervention of IAs with a high risk of rupture is of notable clinical significance. Moreover, accurately predicting aneurysms that are likely to remain stable can help avoid the risks and costs of over-intervention, which also has considerable social significance. Recent advances in artificial intelligence (AI) technology offer promising strategies to assist clinical trials. This review will discuss the state-of-the-art AI applications for assessing the rupture risk of IAs, with a focus on achievements, challenges, and potential opportunities.

Early brain injury (EBI) subsequent to subarachnoid hemorrhage (SAH) is strongly associated with delayed cerebral ischemia and poor patient prognosis. Based on investigations into the molecular mechanisms underlying EBI, neurovascular dysfunction resulting from SAH can be attributed to a range of pathological processes, such as microvascular alterations in brain tissue, ionic imbalances, blood–brain barrier disruption, immune–inflammatory responses, oxidative stress, and activation of cell death pathways. Research progress presents a variety of promising therapeutic approaches for the preservation of neurological function following SAH, including calcium channel antagonists, endothelin-1 receptor blockers, antiplatelet agents, anti-inflammatory agents, and anti-oxidative stress agents. EBI can be mitigated following SAH through neuroprotective measures. To enhance our comprehension of the relevant molecular pathways involved in brain injury, including brain ischemia–hypoxic injury, neuroimmune inflammation activation, and the activation of various cell-signaling pathways, following SAH, it is essential to investigate the evolution of these multifaceted pathophysiological processes. Facilitating neural repair following a brain injury is critical for improving patient survival rates and quality of life.

After aging, the adhesiveness of asphalt deteriorates, leading to a reduction in the durability of asphalt mixtures and affecting the service life of asphalt pavements. To enhance the anti-aging performance of asphalt, this study employed the method of melt blending to prepare three types of modified asphalt: graphene/SBS modified asphalt (G/SBSMA), crumb rubber/SBS modified asphalt (CR/SBSMA), and petroleum resin/SBS modified asphalt (PR/SBSMA). Different dosages of the three types of modified asphalt were tested for changes in conventional performance indicators. The optimal dosages of graphene, crumb rubber, and C9 petroleum resin were determined to be 2%, 15%, and 5%, respectively. Based on the theory of surface free energy, the effects of anti-aging agents on the microscopic properties of SBS modified asphalt before and after aging were analyzed using the three-liquid method. The mechanisms of strength attenuation at the asphalt–aggregate interface under water exposure and aging were revealed. The results showed that with the increase of aging gradient, the asphalt-aggregate biphasic system became more active. The cohesive energy and peel energy of SBS modified asphalt increased continuously, while the adhesive energy decreased continuously, leading to a decrease in the energy ratio parameter. Resin-based anti-aging agents exhibited the most significant improvement in asphalt adhesion performance, while graphene demonstrated a more stable enhancement in asphalt’s water stability during the aging stage.

At the dawn of the next-generation wireless systems and networks, massive multiple-input multiple-output (MIMO) in combination with leading-edge technologies, methodologies, and architectures are poised to be a cornerstone technology. Capitalizing on its successful integration and scalability within 5G and beyond, massive MIMO has proven its merits and adaptability. Notably, a series of evolutionary advancements and revolutionary trends have begun to materialize in recent years, envisioned to redefine the landscape of future 6G wireless systems and networks. In particular, the capabilities and performance of future massive MIMO systems will be amplified through the incorporation of cutting-edge technologies, structures, and strategies. These include intelligent omni-surfaces (IOSs)/intelligent reflecting surfaces (IRSs), artificial intelligence (AI), Terahertz (THz) communications, and cell-free architectures. In addition, an array of diverse applications built on the foundation of massive MIMO will continue to proliferate and thrive. These encompass wireless localization and sensing, vehicular communications, non-terrestrial communications, remote sensing, and inter-planetary communications, among others.

A novel monocrystalline AlN interfacial layer formation method is proposed to improve the device performance of the fully recessed-gate Al2O3/AlN/GaN Metal-Insulator-Semiconductor High Electron Mobility Transistors (MIS-HEMTs), which is achieved by plasma-enhanced atomic layer deposition (PEALD) and in situ N2 plasma annealing (NPA). Compared with the traditional RTA method, the NPA process not only avoids the device damage caused by high temperatures but also obtains a high-quality AlN monocrystalline film that avoids natural oxidation by in situ growth. As a contrast with the conventional PELAD amorphous AlN, C-V results indicated a significantly lower interface density of states (Dit) in a MIS C-V characterization, which could be attributed to the polarization effect induced by the AlN crystal from the X-ray Diffraction (XRD) and Transmission Electron Microscope (TEM) characterizations. The proposed method could reduce the subthreshold swing, and the Al2O3/AlN/GaN MIS-HEMTs were significantly enhanced with ~38% lower on-resistance at Vg = 10 V. What is more, in situ NPA provides a more stable threshold voltage (Vth) after a long gate stress time, and ΔVth is inhibited by about 40 mV under Vg,stress = 10 V for 1000 s, showing great potential for improving Al2O3/AlN/GaN MIS-HEMT gate reliability.

Large-slenderness-ratio (LSR) aircraft exhibit more severe lateral–directional coupling compared with other aircraft, which poses a significant challenge to their flight safety, especially during high-speed maneuvers. Reliable attitude decoupling control is, therefore, essential for LSR aircraft. In this study, a novel control framework that combines incremental nonlinear dynamic inversion (INDI) and extended state observer (ESO) is proposed for supersonic roll maneuver control of LSR aircraft. The ESO is used to estimate the angular acceleration on the basis of an onboard mathematical model. The acceleration estimator based on ESO achieves superior noise reduction compared with the complementary filter (CF) and reduces the onboard model requirement without significantly sacrificing estimation accuracy. Monte Carlo (MC) simulations and frequency–domain analysis demonstrate the effectiveness and robustness of the proposed controller. The sensitivity of parameter uncertainties is also investigated, revealing that the natural frequency of the actuator is the most critical parameter affecting robustness. Finally, flight tests validate the effectiveness of the proposed control structure.

Abstract With millions of years of evolution, owls have developed many excellent characteristics in terms of their flight. The speed of an owl in flight is similar to the relative speed of the blade of a small wind turbine with respect to air. Therefore, the owl wing airfoil is selected as the design airfoil of the wind turbine blade to reduce the flow separation under low Reynolds number. In this study, we analyze an owl wing‐section airfoil and the non‐smooth leading‐edge shape of an owl's wing, and implement an orthogonal optimum design to optimize the wavelength and amplitude of the non‐smooth leading edge. We extract the cross‐sectional features of the airfoil and the non‐smooth leading‐edge shape of the wing. Based on the orthogonal optimum design results, we determine the optimal combination of the wavelength, amplitude, and airfoil, and then design a horizontal‐axis wind turbine blade through bionic coupling. The flow field at different tip speed ratios (TSRs) is simulated using the k‐ωSST turbulence model at the rated wind speed. The results show that the power coefficient (CP) of the bionic wind turbine at a high tip speed ratio is 17.7% higher than that of the standard type. Furthermore, we analyze the operation of the turbine at TSRs of 2 and 5. At a high TSR, the leading edge bulge of the bionic wind turbine blade can change the flow direction distribution of the airflow on the blade surface, make the airflow to adhere to the suction surface, and then reduce the stall area on the suction surface of the blade. Thus, the wind turbine produces higher torque, thereby generating higher power.

Abstract Background Numerous studies have suggested that fasting plasma glucose (FPG) was associated with the risk of mortality. However, relationship on longitudinal changes of FPG with the risk of mortality remained inconsistent. Methods We examined the association of FPG at baseline and its longitudinal changes with risk of mortality based on a cohort study in Yinzhou, China, during 2010–2018. Cox regression models and competing risk models were separately used to examine the association of FPG levels and long‐term fluctuation with risk of total and cause‐specific mortality. Results Subjects who had an impaired fasting glucose or diabetes suffered a higher risk of total mortality than subjects who had a normal fasting glucose (HRs and 95% CIs: 1.17 [1.01–1.35], 1.30 [1.10–1.53], respectively). The HR for total mortality was 1.54 (95% CI: 1.29–1.84) and for cancer mortality was 1.41 (95% CI: 1.04–1.92) in the highest quartile of coefficient of variation of FPG. Trajectory analysis indicated that subjects with a significantly changed FPG suffered a higher risk of total mortality. Conclusion According to this cohort study, we found that long‐term fluctuation of FPG was significantly associated with the risk of total and cancer mortality. Our findings suggest that long‐term fluctuation of FPG could be used as an efficient indicator for predicting the subsequent risk of mortality.

Background: There are increasing concerns towards the transmission of extended spectrum-β-lactamases (ESBL)-producing Enterobacteriaceae in hospital intensive care units (ICUs) at obstetrics & gynaecology departments. The aim of this study was to determine the clinical characteristics and prevalence of ESBL-producing Escherichia coli (E. coli) and Enterobacter cloacae (E. cloacae) isolates collected from ICUs at obstetrics & gynaecology departments in a tertiary care hospital, China. This study also explored the treatment options for E. coli and E. cloacae infections. Methods: E. coli and E. cloacae isolates from ICU samples were identified by using the Vitek 2 Compact System with the GN and ASTGN13 cards. Antimicrobial susceptibility profiles were determined by using the broth microdilution method. Double-disk synergy test (DDST) was performed to screen for ESBLs and combined with the EDTA-disc synergy to detect the production of carbapenemase. Enterobacterial repetitive intergenic consensus (ERIC)-PCR was applied to investigate the clonality of the isolates. Results: A total of 223 strains isolated from 283 hospitalized patients in the ICU with nosocomial infections between 2017 and 2019 were analyzed. Of these, 104 isolates were classified as E. coli and 103 isolates as E. cloacae by the VITEK GNI system. Of the 207 isolates, 131 (63.3%) were separated from sputum or tracheal secretions. ESBL-screen positive was 45.2% (47/104) for E. coli, and 44.7% (46/103) for E. cloacae. Resistance rates of ESBL-producing E. coli and E. cloacae isolates were 95.5% and 91.3% for ampicillin, respectively; 80.6% and 76.1% for ampicillin/tazobactam; 88.1% and 28.3% for ciprofloxacin; 89.6% and 15.2% for levofloxacin; 34.3% and 45.7% for netilmicin; 82.1% and 41.3% for compound sulfamethoxazole; 20.9% and 43.5% for amikacin; 58.2% and 37.0% for gentamicin; 20.9% and 69.6% for piperacillin/tazobactam. Additionally, all ESBL-producing isolates were fully resistant to cefazolin, cefuroxime, ceftazidime, ceftriaxone, cefepime, and aztreonam. On the other hand, isolates were fully susceptible to imipenem and meropenem. Results of ERIC-PCR in all of ESBL-producing E. coli isolates exhibited 11 distinct patterns with a similarity coefficient of 0.8. Only one distinct ERIC pattern was observed amongst the 46 strains of ESBL-producing E. cloacae. Analysis of ERIC patterns demonstrated that there was an outbreak of nosocomial infection of ESBL-producing E. coli and E. cloacae in obstetrics & gynecology ICU of this hospital. Discussion: Our data indicate that the ESBL-producing E. coli and E. cloacae are circulating in the ICU and constitute a major source infection spread. It is necessary to increase surveillance of infections in the ICU and develop adequate infection prevention strategies.

The differential diagnosis of renal tumors and abscesses is crucial owing to their different treatments. Although antibacterial administration and radiological examination are excellent means for distinction, misdiagnosis is common and may lead to severe consequences, such as the need for nephrectomy. Here, we report a case involving a 52-year-old Asian woman with a renal mass for which a differential diagnosis was challenging. The mass persisted after administration of intravenous antibiotic therapy for 1 month. A computed tomography scan indicated an inflammatory lesion, whereas magnetic resonance imaging suggested a diagnosis of a tumor. Despite these indications, a right renal abscess was suspected during robot-assisted laparoscopic surgery, and nephron-sparing surgery was performed, which allowed confirmation of the final pathological result by biopsy specimen. Postoperatively, the mass gradually decreased in size after antibiotic therapy for a further month. This case, in which a renal abscess mimicked a tumor and the patient almost underwent a nephrectomy, highlights the need for caution in establishing therapeutic schedules for patients with inaccurate diagnoses. The management strategies for such patients must be reviewed and improved.

In the adult skeleton, the bone remodeling process involves a dynamic coordination between osteoblasts and osteoclasts, which is disrupted in diseases with high bone turnover rates and dysregulated transforming growth factor beta 1 (TGF-β1). However, little is known about how TGF-β1 signaling mediates bone resorption. Here, we described a pedigree with a heterozygous variant in TGF-β1 (R218C) that resulted in aberrant activation of TGF-β1 through an activating mechanism that caused Camurati-Engelmann disease (CED). We showed that CED patients have high levels of active Rho GTPases and the migration-related proteins Integrin β1 and Integrin β3 in their peripheral blood. HEK293T cells transfected with a plasmid encoding this mutant expressed high levels of TGF-β1 and active Rho GTPases. Furthermore, activation of Rho by TGF-β1 increased osteoclast formation and bone resorption, with increased migration of pre-osteoclasts, as well as cytoskeletal remodeling of pre-osteoclasts and mature osteoclasts. Importantly, pharmacological inhibition of Rho GTPases effectively rescued hyperactive TGF-β1-induced osteoclastogenesis in vitro. Overall, we propose that Rho GTPases mediate TGF-β1-induced osteoclastogenesis and suggest that Rho-TGF-β1 crosstalk is associated with high bone turnover in CED.

Osteoarthritis, as a common joint disease among middle-aged and elderly people, has many problems, such as diverse pathogenesis, poor prognosis and high recurrence rate, which seriously affects patients’ physical and mental health and reduces their quality of life. At present, the pathogenesis of osteoarthritis is not completely clear, and the treatment plan is mainly to relieve symptoms and ensure basic quality of life. Therefore, it is particularly urgent to explore the pathogenesis of osteoarthritis. Protein, as organic macromolecule which plays a major role in life activities, plays an important role in the development of disease. Through protein omics, this study found that with the increase of age, excessive sulfur oxidation occurred in endoplasmic reticulum of chondrocytes, which then drove the occurrence of inflammatory reaction, and provided a direction for the follow-up molecular targeted.

Introduction This study aimed to examine the trends in adherence to Physical Activity Guidelines for Americans (PAG) as well as the association between them and cardiometabolic risk factors among US adults with pre-diabetes.Research design and methods This study included 6734 participants who were diagnosed with pre-diabetes from the National Health and Nutrition Examination Survey 2007–2008 to 2017–2018. The logistic regression model and linear regression model were used to test the trends in adherence to PAG. The multivariable linear regression model was used to examine the association between adherence to PAG and cardiometabolic risk factors.Results The rate of adherence to the PAG for aerobic physical activity was not significantly changed (64.1% in 2007–2008 to 66.4% in 2017–2018, p=0.599). CRISPR/Cas technology originated from the immune mechanism of archaea and bacteria and was awarded the Nobel Prize in Chemistry in 2020 for its success in gene editing. Molecular diagnostics is highly valued globally for its development as a new generation of diagnostic technology. An increasing number of studies have shown that CRISPR/Cas technology can be integrated with biosensors and bioassays for molecular diagnostics. CRISPR-based detection has attracted much attention as highly specific and sensitive sensors with easily programmable and device-independent capabilities. The nucleic acid-based detection approach is one of the most sensitive and specific diagnostic methods. With further research, it holds promise for detecting other biomarkers such as small molecules and proteins. Therefore, it is worthwhile to explore the prospects of CRISPR technology in biosensing and summarize its application strategies in molecular diagnostics. This review provides a synopsis of CRISPR biosensing strategies and recent advances from nucleic acids to other non-nucleic small molecules or analytes such as proteins and presents the challenges and perspectives of CRISPR biosensors and bioassays.

Ascorbate peroxidase (APX), an important antioxidant enzyme, plays a significant role in ROS scavenging by catalyzing the decrease of hydrogen peroxide under various environmental stresses. Nevertheless, information about the APX gene family and their evolutionary and functional attributes in peanut (Arachis hypogea L.) was not reported. Therefore, a comprehensive genome-wide study was performed to discover the APX genes in cultivated peanut genome. This study identified 166 AhAPX genes in the peanut genome, classified into 11 main groups. The gene duplication analysis showed that AhAPX genes had experienced segmental duplications and purifying selection pressure. Gene structure and motif investigation indicated that most of the AhAPX genes exhibited a comparatively well-preserved exon-intron pattern and motif configuration contained by the identical group. We discovered five phytohormones-, six abiotic stress-, and five growth and development-related cis-elements in the promoter regions of AhAPX. Fourteen putative ah-miRNAs from 12 families were identified, targeting 33 AhAPX genes. Furthermore, we identified 3,257 transcription factors from 38 families (including AP2, ARF, B3, bHLH, bZIP, ERF, MYB, NAC, WRKY, etc.) in 162 AhAPX genes. Gene ontology and KEGG enrichment analysis confirm the role of AhAPX genes in oxidoreductase activity, catalytic activity, cell junction, cellular response to stimulus and detoxification, biosynthesis of metabolites, and phenylpropanoid metabolism. Based on transcriptome datasets, some genes such as AhAPX4/7/17/77/82/86/130/133 and AhAPX160 showed significantly higher expression in diverse tissues/organs, i.e., flower, leaf, stem, roots, peg, testa, and cotyledon. Likewise, only a few genes, including AhAPX4/17/19/55/59/82/101/102/137 and AhAPX140, were significantly upregulated under abiotic (drought and cold), and phytohormones (ethylene, abscisic acid, paclobutrazol, brassinolide, and salicylic acid) treatments. qRT-PCR-based expression profiling presented the parallel expression trends as generated from transcriptome datasets. Our discoveries gave new visions into the evolution of APX genes and provided a base for further functional examinations of the AhAPX genes in peanut breeding programs.

BackgroundKeloids are a fibroproliferative disease characterized by unsatisfactory therapeutic effects and a high recurrence rate.ObjectiveThis study aimed to investigate keloid-related circulating metabolic signatures.MethodsUntargeted metabolomic analysis was performed to compare the metabolic features of 15 keloid patients with those of paired healthy volunteers in the discovery cohort. The circulating metabolic signatures were selected using the least absolute shrinkage. Furthermore, the selection operators were quantified using multiple reaction monitoring-based target metabolite detection methods in the training and test cohorts.ResultsMore than ten thousand metabolic features were consistently observed in all the plasma samples from the discovery cohort, and 30 significantly different metabolites were identified. Four differentially expressed metabolites including palmitoylcarnitine, sphingosine, phosphocholine, and phenylalanylisoleucine, were discovered to be related to keloid risk in the training and test cohorts. In addition, using linear and logistic regression models, the respective risk scores for keloids based on a 4-metabolite fingerprint classifier were established to distinguish keloids from healthy volunteers.ConclusionsIn summary, our findings show that the characteristics of circulating metabolic fingerprinting manifest phenotypic variation in keloid onset.

Possessed of easy access to development and fair economic benefits, medium-high maturity continental shale oil and gas have become the focus of shale oil and gas study in the future. Shale oil and gas mainly occur in pores, but studies on the pore characteristics of shale oil and gas occurrence are by no means sufficient. Focused on shale from the Middle Jurassic Lianggaoshan Formation in Well TYX, Fuling block, southeast of Sichuan Basin where a breakthrough in shale oil and gas exploration was recently achieved, this study selects core samples and conducts a series of analyses, including vitrinite reflectance analysis, kerogen microscopic examination experiment, total organic carbon (TOC) content analysis, mineral composition analysis, gas content measurement, isothermal adsorption experiment, S1 content analysis, and others. The analyses are to identify the pore characteristics of the continental medium and high maturity shale oil and gas by virtue of scanning electron microscope (SEM) with Ar-ion milling and the image processing software ImageJ. The conclusions are drawn as follows: in terms of lithofacies, medium-high maturity continental shale oil and gas mainly occur in organic-rich clay shale and organic-rich mixed shale; with regard to material composition, shale oil and gas mainly occur in organic matter, illite-smectite mixed layers and illite. Shale adsorbed gas content accounts for at most 40% of the total shale gas content and shale free gas content takes up at least 60% of the total shale gas content. Pores of solid bitumen, solid bitumen-clay mineral complex mass, clay minerals, structured vitrinite, and funginite are mostly developed in shale. Among them, the first three types of pores are the main reservoir space in shale considering their large number, good roundness, medium pore diameter, fairly good roundness of pore edges, and the complex shapes which altogether contribute to the large surface porosity.

Soil microorganisms play crucial roles in nutrient cycling and determining soil quality and fertility; thus, they are important for agricultural production. However, the impacts of long-term fertilization on soil microbial community remain ambiguous due to inconsistent results from different studies. The objective of this study was to characterize changes in bacterial and fungal diversity and community structures after 12 years of different fertilization in aeolian sandy soil by analyzing 16S rRNA and ITS rRNA gene sequences and the soil properties to discover the driving factors. Eight different fertilizer treatments have been set up since 2009: no fertilizer (CK), chemical N fertilizer (N), chemical N and P fertilizer (NP), chemical N, P and K fertilizer (NPK), pig manure only (M), pig manure plus chemical N fertilizer (MN), pig manure plus chemical N and P fertilizer (MNP), pig manure plus chemical N, P, and K fertilizer (MNPK). The results indicated that the long-term application of chemical fertilizer reduced soil pH, whereas the addition of pig manure alleviated a decrease in soil pH value. Chemical fertilizer plus pig manure significantly improved soil available nutrients and soil organic carbon. Long-term MNPK fertilization resulted in changes in bacterial diversity due to effects on specific bacterial species; by contrast, all fertilization treatments resulted in changes in fungal diversity due to changes in soil properties. Principal component analysis indicated that fertilization had a significant effect on soil microbial community structure, and the effect of chemical fertilizer combined with pig manure was greater than that of chemical fertilizer alone. Soil available phosphorus, total phosphorus, and pH were the most important factors that influenced bacterial taxa, whereas soil pH, total phosphorus, organic carbon, ammonium nitrogen and nitrate nitrogen were the most important factors influencing fungal taxa after 12 years of fertilization in aeolian sandy soil.

Due to its high genetic diversity and broad host range, Ralstonia solanacearum, the causative phytopathogen of the bacterial wilt (BW) disease, is considered a “species complex”. The R. solanacearum strain FJ1003 belonged to phylotype I, and was isolated from the Fuzhou City in Fujian Province of China. The pathogen show host specificity and infects tobacco, especially in the tropical and subtropical regions. To elucidate the pathogenic mechanisms of FJ1003 infecting tobacco, a complete genome sequencing of FJ1003 using single-molecule real-time (SMRT) sequencing technology was performed. The full genome size of FJ1003 was 5.90 Mb (GC%, 67%), containing the chromosome (3.7 Mb), megaplasmid (2.0 Mb), and small plasmid (0.2 Mb). A total of 5133 coding genes (3446 and 1687 genes for chromosome and megaplasmid, respectively) were predicted. A comparative genomic analysis with other strains having the same and different hosts showed that the FJ1003 strain had 90 specific genes, possibly related to the host range of R. solanacearum. Horizontal gene transfer (HGT) was widespread in the genome. A type Ⅲ effector protein (Rs_T3E_Hyp14) was present on both the prophage and genetic island (GI), suggesting that this gene might have been acquired from other bacteria via HGT. The Rs_T3E_Hyp14 was proved to be a virulence factor in the pathogenic process of R. solanacearum through gene knockout strategy, which affects the pathogenicity and colonization ability of R. solanacearum in the host. Therefore, this study will improve our understanding of the virulence of R. solanacearum and provide a theoretical basis for tobacco disease resistance breeding.

The upstream Fujiang River are famous for their high mountains and narrow canyons, which results in a conflict of interest between local residents and land use. To make good use of precious land resources is a meaningful work for local people and government. Therefore, field surveys and theoretical analysis were carried out in the upstream Fujiang River to explore the land-use evaluation and utilization advice. Firstly, we analyzed the factors which affect the land use of debris flow depositional area. Secondly, we chose the factors of soil fertility quality, soil particle conservation, and the geological engineering condition to set up the land-use evaluation method. Thirdly, through index classification, we give the planting type, land-use type, construction type respectively, according to grading and assignment value for each factor, we built the debris flow disaster-prone land utilization mode R by using an analytical hierarchy process method. Lastly, taking Huangjiaba village as an example, based on the debris flow disaster land-use evaluation method and debris flow hazard evaluation, we identified four land-use scenarios. The results show that the value of the four scenarios is between 1.00 and 4.00, the four scenarios utilization advices are given based on each R-value and Huangjiaba natural condition. It also obtained that the developmental tendencies of the debris flow disaster areas appear to deposit recently, the frequency of debris-flow activity is evolving from high to stable, on making good use of debris flow deposit fan’s usage is necessary to explore the disaster prone land and avoid disaster loss. Further studies need to systematically consider more factors to evaluate debris flow and disaster-prone land utilization, which can provide a reference for the rural space planning and revitalization for the local government.

Trans-catheter arterial embolization (TAE) plays an important role in treating various diseases. The available embolic agents lack X-ray visibility and do not prevent the reflux phenomenon, thus hindering their application for TAE therapy. Herein, we aim to develop a multifunctional embolic agent that combines the X-ray radiopacity with local procoagulant activity. The barium sulfate nanoparticles (BaSO4 NPs) were synthesized and loaded into the polyvinyl alcohol/chitosan (PVA/CS) to prepare the radiopaque BaSO4/PVA/CS microspheres (MS). Thereafter, thrombin was immobilized onto the BaSO4/PVA/CS MS to obtain the thrombin@BaSO4/PVA/CS MS. The prepared BaSO4/PVA/CS MS were highly spherical with diameters ranging from 100 to 300 μm. In vitro CT imaging showed increased X-ray visibility of BaSO4/PVA/CS MS with the increased content of BaSO4 NPs in the PVA/CS MS. The biocompatibility assessments demonstrated that the MS were non-cytotoxic and possessed permissible hemolysis rate. The biofunctionalized thrombin@BaSO4/PVA/CS MS showed improved hemostatic capacity and facilitated hemostasis in vitro. Additionally, in vivo study performed on a rabbit ear embolization model confirmed the excellent X-ray radiopaque stability of the BaSO4/PVA/CS MS. Moreover, both the BaSO4/PVA/CS and thrombin@BaSO4/PVA/CS MS achieved superior embolization effects with progressive ischemic necrosis on the ear tissue and induced prominent ultrastructural changes in the endothelial cells. The findings of this study suggest that the developed MS could act as a radiopaque and hemostatic embolic agent to improve the embolization efficiency.
