Abstract

Ni-rich layered oxide LiNi0.8Co0.15Al0.05O2 (NCA) is one of the most promising cathode candidates for high-energy-density lithium batteries. However, the extensive application of NCA is hindered due to the notorious electrode/electrolyte interfacial issues. To address the above bottleneck, a new gel polymer electrolyte is prepared with maleic anhydride/hexafluorobutyl methacrylate/methyl methacrylate at a ratio of 1 : 1 : 1 via in situ polymerization. A high electrochemical stability window of 5.5 V vs. Li+/Li and ionic conductivity of 1.1 × 10−3 S cm−2 are achieved, which suggest the superior high-voltage stability and fast ion migration dynamics of poly(maleic anhydride–hexafluorobutyl methacrylate–methyl methacrylate) (PMHM) electrolyte that are contributed by C–F and ester/acid anhydride groups in the polymer chain. As a result, the NCA/PMHM/Li battery delivers stable cycling performance with a capacity retention of 91.8% after 200 cycles at 0.5C, and a high capacity retention of 77.7% after 500 cycles at 1C. X-ray photoelectron spectroscopy (XPS) and X-ray diffraction (XRD) results reveal the stability of interfacial chemistry and the integrity of cathode structure. This work indicates that the in situ polymerized hexafluorobutyl methacrylate-based electrolyte is imperative to address the interfacial issues and boost the development of high-energy lithium metal batteries with a Ni-rich cathode. Abstract

Hf(2mol%):Fe(0.05wt%):LiNbO3 crystals with various [Li]/[Nb] ratios of 0.94, 1.05, 1.2 and 1.38 have been grown. The photorefractive resistant ability increases with the accretion of [Li]/[Nb] ratio. When the ratio of [Li]/[Nb] is 1.20 or 1.38, the OH- absorption band shifts to about 3477cm-1. The mechanisms of the photorefractive resistant ability increase and the absorption band shift have been discussed. The exponential gain coefficient (Γ) of the crystals was measured with two-beam coupling method and the effective charge carrier concentration (Neff) was calculated. The results show that Γ and Neff increase with the accretion of [Li]/[Nb] ratio. The temperature effect of codoped Hf:Fe:LiNbO3 crystals was also studied, it was found that the exponential gain coefficient increase dramatically at about 55°C, 70°C and 110°C, this is due to the inner electric field which is resulted from structure phase change. Abstract

Limonene‐derived polycarbonate‐based alkyd resins (ARs) have been prepared by copolymerization of limonene dioxide with CO2, catalysed by a β‐diiminate zinc–bis(trimethylsilyl)amido complex, and subsequent chemical modification with soybean oil fatty acids using triphenylethylphosphonium bromide as the catalyst. This quantitative partial modification was realized via epoxy–carboxylic acid chemistry, affording ARs with higher oil lengths, lower polydispersities and higher glass transition temperatures (Tg) in comparison to a conventional polyester AR based on phthalic acid, multifunctional polyol pentaerythritol and soybean fatty acid. The novel limonene polycarbonate AR and the conventional polyester AR were evaluated as coatings and both the physical drying (without the presence of the oxidative drying accelerator Borchi® Oxy Coat) and chemical curing (with Borchi® Oxy Coat) processes of these coatings were monitored by measuring the König hardness and complex modulus development with time. A better performance was obtained for the alkyd paint containing polycarbonates modified with fatty acids (FA‐PCs), which showed a faster chemical drying, a higher König hardness and a higher Tg in coating evaluation, demonstrating that the fully renewable FA‐PCs are promising resins for alkyd paint applications. © 2019 Society of Chemical Industry [ABSTRACT FROM AUTHOR]

Abstract

Tumor cell motility is the essential step in cancer metastasis. Previously, we showed that oxytocin and epidermal growth factor (EGF) effects on cell migration in prostate cancer cells require Giα2 protein. In the current study, we investigated the interactions among G‐protein coupled receptor (GPCR), Giα2, PI3‐kinase, and Rac1 activation in the induction of migratory and invasive behavior by diverse stimuli. Knockdown and knockout of endogenous Giα2 in PC3 cells resulted in attenuation of transforming growth factor β1 (TGFβ1), oxytocin, SDF‐1α, and EGF effects on cell migration and invasion. In addition, knockdown of Giα2 in E006AA cells attenuated cell migration and overexpression of Giα2 in LNCaP cells caused significant increase in basal and EGF‐stimulated cell migration. Pretreatment of PC3 cells with Pertussis toxin resulted in attenuation of TGFβ1‐ and oxytocin‐induced migratory behavior and PI3‐kinase activation without affecting EGF‐induced PI3‐kinase activation and cell migration. Basal‐ and EGF‐induced activation of Rac1 in PC3 and DU145 cells were not affected in cells after Giα2 knockdown. On the other hand, Giα2 knockdown abolished the migratory capability of PC3 cells overexpressing constitutively active Rac1. The knockdown or knockout of Giα2 resulted in impaired formation of lamellipodia at the leading edge of the migrating cells. We conclude that Giα2 protein acts at two different levels which are both dependent and independent of GPCR signaling to induce cell migration and invasion in prostate cancer cells and its action is downstream of PI3‐kinase–AKT–Rac1 axis. 172. Auxin-inducible degron 2 system deciphers functions of CTCF domains in transcriptional regulation

Abstract

Background: CTCF is a well-established chromatin architectural protein that also plays various roles in transcriptional regulation. While CTCF biology has been extensively studied, how the domains of CTCF function to regulate transcription remains unknown. Additionally, the original auxin-inducible degron 1 (AID1) system has limitations in investigating the function of CTCF. Results: We employ an improved auxin-inducible degron technology, AID2, to facilitate the study of acute depletion of CTCF while overcoming the limitations of the previous AID system. As previously observed through the AID1 system and steady-state RNA analysis, the new AID2 system combined with SLAM-seq confirms that CTCF depletion leads to modest nascent and steady-state transcript changes. A CTCF domain sgRNA library screening identifies the zinc finger (ZF) domain as the region within CTCF with the most functional relevance, including ZFs 1 and 10. Removal of ZFs 1 and 10 reveals genomic regions that independently require these ZFs for DNA binding and transcriptional regulation. Notably, loci regulated by either ZF1 or ZF10 exhibit unique CTCF binding motifs specific to each ZF. Conclusions: By extensively comparing the AID1 and AID2 systems for CTCF degradation in SEM cells, we confirm that AID2 degradation is superior for achieving miniAID-tagged protein degradation without the limitations of the AID1 system. The model we create that combines AID2 depletion of CTCF with exogenous overexpression of CTCF mutants allows us to demonstrate how peripheral ZFs intricately orchestrate transcriptional regulation in a cellular context for the first time.

MLL-rearranged (MLL-r) leukemias count for more than 80% of infant leukemia, ~5-10% of B-cell acute lymphoblastic leukemia (B-ALL), and ~10% of acute myeloid leukemia (AML) cases, where they confer a particularly poor outcome. Despite treatment with intensive multi-agent chemotherapy, most MLL-r patients ultimately relapsed after an initial remission. So far, the molecular mechanism by which MLL-r leukemia maintains progression and prevents differentiation remains largely unclear.

Abstract

High glass transition temperature ( T g ) thiol-ene networks (TENs) based on poly(limonene carbonate)s (PLCs), derived from orange oils and of potential degradability are described here. PLCs with moderate molecular weight were prepared by copolymerization of limonene oxide with CO 2 and subsequent breakdown reactions. These PLCs were cured with multifunctional thiol monomers in the presence of thermal initiators via thiol-ene chemistry to generate poly(thioether-co-carbonate) networks. The thermal curing experiments were optimized by a kinetic study using real-time ATR-FTIR, in which a delayed gelation was observed. For the first time, an interesting “cage” effect was observed during the network formation initiated by DCP, in which the addition reactions of pendant isopropenyls on high molecular weight PLC chains were significantly enhanced by thiol-ene crosslinking at 160 °C. The resulting homogeneous TENs with high T g s (>100 °C) and a wide range of thermomechanical properties, including rubbery moduli from 2.9 to 28.2 MPa, were obtained. Abstract

Multidrug-resistant Acinetobacter baumannii, a common pathogen responsible for nosocomial infections, is the main cause for outbreaks of infectious diseases, such as pneumonia, meningitis, and bacteremia, especially among critically ill patients. Epidemic A. baumanniiis a growing public health concern as it is resistant to all existing antimicrobial agents, thereby necessitating the development of new therapeutic approaches to mount an effective immune response against this bacterial pathogen. In this study, we identified a critical role for type I interferon (IFN) in epigenetic regulation during A. baumanniiinfection and established a central role for it in multiple cell death pathways. A. baumanniiinfection induced mixed cell death constituted of apoptosis, pyroptosis, and necroptosis. Mechanically, A. baumanniitriggered TRIF-dependent type I IFN production, which in turn induced the expression of genes Zbp1, Mlkl, caspase-11, and Gsdmdvia KAT2B-mediated and P300-mediated H3K27ac modification, leading to NLRP3 inflammasome activation, and potentially contributed to GSDMD-mediated pyroptosis and MLKL-dependent necroptosis. Our study offers novel insights into the mechanisms of type I IFN and provides potential therapeutic targets for infectious and inflammatory diseases.

Abstract

Fully biobased poly(limonene carbonate)s (PLCs) have been prepared by copolymerization of limonene oxide with CO 2 , using a β-diiminate zinc-bis(trimethylsilyl)amido complex as the polymerization catalyst, and subsequent transcarbonation reactions using 1,5,7-triazabicyclo-[4.4.0]dec-5-ene (TBD) as the catalyst, combined with 1,10-decanediol as the transcarbonation agent. Quantitative partial post-modification of these polycarbonates was fulfilled via thiol-ene chemistry using two mercaptoalcohols with different chain lengths, viz . 2-mercaptoethanol and 6-mercaptohexanol. The thermal properties and hydroxyl values (OHVs) of the resulting hydroxyl PLCs were modulated by controlling the type and amount of incorporated thioether species. The curing kinetics of these PLCs with blocked/non-blocked multifunctional isocyanates was studied by ATR-FTIR followed by the solvent casting and curing under the optimal conditions. The good acetone resistance and high transparency and hardness of the coatings demonstrated that the fully bio-based PLCs with adequate molecular weights and OHVs are promising resins for coating applications. Abstract

The most aggressive of four medulloblastoma (MB) subgroups are cMyc-driven group 3 (G3) tumors, some of which overexpress EZH2, the histone H3K27 mono-, di-, and trimethylase of polycomb-repressive complex 2. Ezh2 has a context-dependent role in different cancers as an oncogene or tumor suppressor and retards tumor progression in a mouse model of G3 MB. Engineered deletions of Ezh2in G3 MBs by gene editing nucleases accelerated tumorigenesis, whereas Ezh2 re-expression reversed attendant histone modifications and slowed tumor progression. Candidate oncogenic drivers suppressed by Ezh2 included Gfi1, a proto-oncogene frequently activated in human G3 MBs. Gfi1disruption antagonized the tumor-promoting effects of Ezh2loss; conversely, Gfi1 overexpression collaborated with Myc to bypass effects of Trp53 inactivation in driving MB progression in primary cerebellar neuronal progenitors. Although negative regulation of Gfi1by Ezh2 may restrain MB development, Gfi1 activation can bypass these effects.
