Abstract

Nanoprobes are advantageous over small molecular probes in sensitivity but most luminescence molecules used to construct nanoprobes often suffer from an aggregation-caused quenching effect. Herein, we rationally designed a small molecular probe Cys(StBu)-Lys(Ru(bpy)32+)-CBT (1) which “smartly” self-assembled into nanoparticles 1-NPs inside cells with non-quenched, persistent phosphorescence. Employing this property, we successfully applied 1 for long-term sensing of biothiol activity in living HepG2 cells and tumors. We envision that, by modifying the amino group with an enzyme substrate, our probe 1 could be further developed for sensing intracellular enzyme activity with non-quenched, persistent phosphorescence. 162. Tranilast directly targets NLRP3 to treat inflammasome‐driven diseases.

Abstract

Abstract: The dysregulation of NLRP3 inflammasome can cause uncontrolled inflammation and drive the development of a wide variety of human diseases, but the medications targeting NLRP3 inflammasome are not available in clinic. Here, we show that tranilast (TR), an old anti‐allergic clinical drug, is a direct NLRP3 inhibitor. TR inhibits NLRP3 inflammasome activation in macrophages, but has no effects on AIM2 or NLRC4 inflammasome activation. Mechanismly, TR directly binds to the NACHT domain of NLRP3 and suppresses the assembly of NLRP3 inflammasome by blocking NLRP3 oligomerization. In vivo experiments show that TR has remarkable preventive or therapeutic effects on the mouse models of NLRP3 inflammasome‐related human diseases, including gouty arthritis, cryopyrin‐associated autoinflammatory syndromes, and type 2 diabetes. Furthermore, TR is active ex vivo for synovial fluid mononuclear cells from patients with gout. Thus, our study identifies the old drug TR as a direct NLRP3 inhibitor and provides a potentially practical pharmacological approach for treating NLRP3‐driven diseases. [ABSTRACT FROM AUTHOR]

Abstract

d-Luciferin is the most widely used substrate for bioluminescence (BL) applications but its low chemical stability always affects its performance. Herein, we rationally designed two chemically stable precursor molecules CBT-d-cystine-CBT (d-1) and CBT-l-cystine-CBT (l-1), and subjected them to reduction-controlled condensation to form 1-oligomer and subsequent proteolysis to yield d-aminoluciferin for BL generation in cells and in vivo. Abstract

Herein, we review the development, applications and potential prospects of CBT-Cys click reaction. This click condensation reaction is based on the condensation reaction between 2-cyanobenzothiazole (CBT) and D-cysteine (D-Cys) in fireflies and has high biocompatibility and controllability in physiological solutions. Under the control of pH, reduction, or enzyme, this CBT-based click reaction has been widely applied to a wide range of biomedical fields such as protein labeling, molecular imaging (e. g., optical imaging, nuclear imaging, magnetic resonance imaging and photoacoustic imaging), nanomaterial fabrication, cancer therapy, and other potentialities.
