We examined the in vivo behavior of liver natural killer T cells (NKT cells) by intravital fluorescence microscopic imaging of mice in which a green fluorescent protein cDNA was used to replace the gene encoding the chemokine receptor CXCR6. NKT cells, which account for most CXCR6+ cells in liver, were found to crawl within hepatic sinusoids at 10-20 µm/min and to stop upon T cell antigen receptor activation. CXCR6-deficient mice exhibited a selective and severe reduction of CD1d-reactive NKT cells in the liver and decreased susceptibility to T-cell-dependent hepatitis. CXCL16, the cell surface ligand for CXCR6, is expressed on sinusoidal endothelial cells, and CXCR6 deficiency resulted in reduced survival, but not in altered speed or pattern of patrolling of NKT cells. Thus, NKT cells patrol liver sinusoids to provide intravascular immune surveillance, and CXCR6 contributes to liver-based immune responses by regulating their abundance.

DC-SIGN appears to bind HIV-1 in mucosal tissues and present it to T cells, leading to an infection of the T cell. DC-SIGN is a C-type lectin that binds to ICAM-3 on resting T cells.

CXCR4, a chemokine receptor, is expressed in cells of the immune and central nervous systems, and can control migration of resting leukocytes and haematopoietic progenitors. Mice lacking CXCR4 were found to show haematopoietic and cardiac defects the same as those of SDF-1-deficient mice, suggesting that it may be the only receptor for SDf-1. This is thought to be the first demonstration of G-protein-coupled chemokine receptor involvement in neuronal cell migration and patterning in the central nervous system.

Studies revealed that chemokines and their seven-transmembrane G protein-coupled receptors can allegedly block HIV replication. Such findings may help to further understand HIV activities and yield effective means to control HIV pathogenesis. Since chemokines may have important roles in the primate lentiviral replication cycle and in pathogenesis, their optimum ability to inhibit HIV replication is not yet fully determine.

The CC-CRK-5 beta-chemokine receptor functions as a co-receptor for the primary macrophage-trophic strains of the human immunodeficiency virus 1 (HIV-1) and acts along with CD4 to enable the entry of the virus into host cells. Cells co-expressing CD4 and CC-CRK-5 are highly susceptible to HIV infection while those co-expressing CD4 and other members of the CC-CRK-5 family are resistant to infection. CC-CRK-5 activates the fusion of plasma membrane with the HIV-1 envelope. Beta-chemokines are cytokines that suppress HIV infection by blocking its entry and replication in cells.

Transgenic mice have been employed to locate an intronic regulatory region which quenches only the transgene expression in CD4-CD8+ but does not affect CD4+CD8- or CD4+CD8+ T cells. The region suppresses gene expression in CD4-CD8- thymocytes also. The negative element does not depend on orientation or position.

B and T lymphocytes use a similar signal transduction mechanism to induce responses. The oligomeric receptors in these cells possess distinct recognition and signal transduction subunits. B and T cells interact with PTK cells using common sequence motifs. The balance between the actions of PTPases and receptor-regulated PTKs control the tyrosine-phosphorylating action of phospholipase C. Calcineurin and Ras are among the downstream effectors of lymphokine gene expression.

Cytoplasmic protein-tyrosine kinases (PTKs) of the Src family have important functions in signal transduction processes in several cell types. The lymphocyte-specific cytoplasmic protein-tyrosine kinase p56 (Lck) is needed for T cell growth and functioning. It is required in conjunction with the coreceptor molecules, CD4 and CD8 for the potentiation of antigen-specific signals through the T cell antigen receptor. The mechanics of Lck was studied by fusing hybrid molecules made up of extracellular transmembrane domains of CD4 to LCK or any other kinases from the Src.

Researchers are tracing the part played by CD4-p56lck association during the progress of the t-lymphocyte. The glycoproteins CD4 and CD8 interact with protein tyrosine kinase (PTK) p56lck, which is important for thymocyte maturation and activating T-cells. Although CD4 cannot relate with p56lck, it can aid the helper-cell lineage when over expressed. Several experiments have proved that CD4 and CD8 are essential for T-cell lineage determination, because of their transducing properties, and that CD4 can work without the help of p56lck in T-cell development. The PTK, with which CD4 is associated, does not seem important in picking the helper T-cell lineage.

Thymocytes differentiate into mature CD4 helper or CD8 cytotoxic cells depending on their antigen receptor specificity for MHC class II or I molecules, respectively. The CD8 differentiation and maturity pathway was analyzed using transgenic mouse systems. Results show that constitutive expression of CD4 can rescue development of mature CD8 class II reactive T cells even in mice that lack the capacity to develop CD8 cells. These results suggest a selection or stochastic mechanism of T cell differentiation.

The endocytic trafficking of CD4, a cell surface glycoprotein primarily expressed on helper Tlymphocytes, was studied to determine the mechanism by which it is constitutively internalized and recycled, but not endocytized in lymphocytes. It was discovered that an interaction between the cytoplasmic domain of CD4 andprotein tyrosine kinase p56(super lck) or PTK prevents endocytosis. It was further discovered that PTK prevents CD4 from entering coated pits by PTK, thusinhibiting endocytosis. These discoveries present a new regulatory mechanism for the internalization of a cell surface molecule.
