Abstract

Site-directed spin labeling (SDSL) of large RNAs for electron paramagnetic resonance (EPR) spectroscopy has remained challenging to date., Site-directed spin labeling (SDSL) of large RNAs for electron paramagnetic resonance (EPR) spectroscopy has remained challenging to date. We here demonstrate an efficient and generally applicable posttranscriptional SDSL method for large RNAs using an expanded genetic alphabet containing the NaM-TPT3 unnatural base pair (UBP). An alkyne-modified TPT3 ribonucleotide triphosphate (rTPT3COTP) is synthesized and site-specifically incorporated into large RNAs by in vitro transcription, which allows attachment of the azide-containing nitroxide through click chemistry. We validate this strategy by SDSL of a 419-nucleotide ribonuclease P (RNase P) RNA from Bacillus stearothermophilus under non-denaturing conditions. The effects of site-directed UBP incorporation and subsequent spin labeling on the global structure and function of RNase P are marginal as evaluated by Circular Dichroism spectroscopy, Small Angle X-ray Scattering, Sedimentation Velocity Analytical Ultracentrifugation and enzymatic assay. Continuous-Wave EPR analyses reveal that the labeling reaction is efficient and specific, and Pulsed Electron–Electron Double Resonance measurements yield an inter-spin distance distribution that agrees with the crystal structure. The labeling strategy as presented overcomes the size constraint of RNA labeling, opening new avenues of spin labeling and EPR spectroscopy for investigating the structure and dynamics of large RNAs.

Abstract

The stay-green trait is recognised as a key drought adaptation mechanism in cereals worldwide. Stay-green sorghum plants exhibit delayed senescence of leaves and stems, leading to prolonged growth, a reduced risk of lodging and higher grain yield under end-of-season drought stress. More than 45 quantitative trait loci (QTL) associated with stay-green have been identified, including two major QTL (Stg1 and Stg2). However, none of the contributing genes that regulate functional stay-green are known. Here we show that the PIN FORMED family of auxin efflux carriers induce some of the causal mechanisms driving the stay-green phenotype in sorghum, with SbPIN4 and SbPIN2 located in Stg1 and Stg2, respectively. We found that 9 of 11 sorghum PIN genes aligned with known stay-green QTL. In transgenic studies, we demonstrated that PIN genes located within the Stg1 (SbPIN4), Stg2 (SbPIN2) and Stg3b (SbPIN1) QTL regions acted pleiotropically to modulate canopy development, root architecture and panicle growth in sorghum, with SbPIN1, SbPIN2 and SbPIN4 differentially expressed in various organs relative to the non stay-green control. The emergent consequence of such modifications in canopy and root architecture is a stay-green phenotype. Crop simulation modelling shows that the SbPIN2 phenotype can increase grain yield under drought.

Abstract

An electrochemical nitrogen reduction reaction (NRR) under mild conditions offers a promising alternative to the traditional Haber-Bosch process in converting abundant nitrogen (N2) to high value-added ammonia (NH3). In this work, iron phthalocyanine (FePc) was homogeneously immobilized on pyridine-functionalized carbon nanotubes to form a well-tuned electrocatalyst with an FeN5 active center (FePc-Py-CNT). Synchrotron X-ray absorption and Fourier transform infrared spectroscopy proved the presence of an Fe-N coordination bond between FePc and surface-bound pyridine. The resulting hybrid exhibited notably enhanced electrocatalytic NRR performance compared to FePc immobilized on CNTs based on pi-pi stacking interactions (FePc-CNT), resulting in doubled NH3 yield (21.7 mu g 1 h mgcat-1h-1) and Faradaic efficiency (22.2%). Theoretical calculations revealed that the axial coordination on FePc resulted in partial electron transfer from iron to pyridine, which efficiently suppresses the adsorption of H+ and improves the chemisorption of N2 at Fe sites. Meanwhile, the interfacial electron transfer was facilitated by pyridine as an electron transfer relay between FePc and CNTs. Abstract

Online product reviews are valuable resources to collect customer preferences for product improvement. To retrieve consumer preferences, it is important to automatically extract product features from online reviews. However, product feature extraction from Chinese online reviews is challenging due to the particularity of the Chinese language. This research focuses on how to accurately extract and prioritize product features and how to establish product improvement strategies based on the extracted product features. First, an ensemble deep learning based model (EDLM) is proposed to extract and classify product features from Chinese online reviews. Second, conjoint analysis is conducted to calculate the corresponding weight of each product feature and a weight-based Kano model (WKM) is proposed to classify and prioritize product features. Various comparative experiments show that the EDLM model achieves impressive results in product feature extraction and outperforms existing state-of-the-art models used for Chinese online reviews.
