Accumulating evidence suggests that liver injury can be induced by the over-expression of β1-adrenergic receptors (β1-ARs). High titers of autoantibodies specific to β1-adrenergic receptors (β1-AA) are detected in the sera of heart failure patients, potentially playing agonist-like roles. However, the role of β1-AA in liver function has not been characterized. In this study, we collect the sera of primary biliary cholangitis (PBC) patients, a condition which easily develops into liver fibrosis, and analyze the relationship between PBC and β1-AA. A passive immunization model is established to assess the effect of β1-AA on the liver. Subsequently, the effect of β1-AA on macrophages is investigated in vitro. Results show that PBC patients have a high titer and ratio of β1-AA, compared to controls. Liver injury and fibrosis are induced by β1-AA. In vitro experiments with ROS probe demonstrate that β1-AA induces macrophages to produce ROS and secrete TNFα. These effects can be partially reversed by metoprolol, a blocker for β1-AR. Results from the transwell and phagocytosis assays show that β1-AA promotes macrophage migration and phagocytosis. FCM tests suggest that β1-AA induces the alteration of M1 rather than M2 markers in macrophages. Finally, the Annexin V/PI assay indicates that macrophage culture supernatants stimulated by β1-AA cause hepatocyte apoptosis. Overall, these results suggest that β1-AA is involved in PBC. The β1-AA-induced activation, phagocytosis and phenotypic modification of macrophages may play an important role in the development of hepatic fibrosis and injury.

A Copula-based competing failure analysis method for multi-performance mechanism systems is studied. Some specific mechanism systems often are under two types of failure modes, the catastrophic failures leading by oversized-load and function degradation failures because of out of operation. Furthermore, these failure modes also compete with each other. Competing failure model is established by catastrophic failure probability expression and drifting Brown motion. Meanwhile Copula functions are used to describe the failure dependence and deduce joint distribution functions, in order to form the competing failure analysis method to analyse the mechanism system reliability and its evaluation law for multi-performance mechanism systems. At last, an airplane cabin door lock system is studied as a case, demonstrating the validity of proposed method.

Objective This study aimed at describing the frequency of rare variants of monogenic cerebral small vessel diseases (CSVD) in a cohort of patients with CSVD, and to explore its clinical relevance. Methods This study included CSVD patients visiting the Neurology Department of Peking Union Medical College Hospital(PUMCH) from March 2017 to January 2022, collecting their demographic and clinical information and DNA samples for whole-exome sequencing. Descriptive analysis and statistical analysis were conducted exploring the differences between monogenic CSVD-related gene mutation carriers and noncarriers. Results A total of 292 patients were included, 51.03% of whom carried one or more rare variants of monogenic CSVD-related genes. The most common rare low-frequency variants were located in the NOTCH3 gene (70 patients, 23.97%), followed by HTRA1 and COL4A1/COL4A2 (22 patients, 7.53%) respectively. Among the subgroup of patients without a family history of stroke (n=176), the frequency of rare variants was as high as 47.16%. Compared with non-carriers, the carriers were diagnosed at a younger age (58.76±13.71 vs. 63.46±13.21, P=0.003). No difference was found in phenotypes among single-SNP carriers, multiple-SNPs carriers, and noncarriers. Conclusions The frequency of rare mutation of monogenic CSVD-related genes were relatively high in Chinese CSVD cohort. The most common rare variant was within the NOTCH3, followed by HTRA1 and COL4A1/COL4A2 genes. For CSVD patients of unknown causes, genetic screening should not be neglected even if there is not a family history of the disease.

Objective To investigate anti-biofilm activity and mechanisms of emodin against methicillin-resistant Staphylococcus aureus (MRSA) in vitro. Methods Microbroth dilution was explored to determine minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC) of emodin against MRSA252 strain and clinical isolations. Inhibitory curve were draw to estimate dynamic bacteriostatic effect. Crystal violet staining was used to investigate the inhibitory effect of emodin against MRSA biofilm. The effect of emodin on the morphology of MRSA biofilm was observed by scanning electron microscopy (SEM) and laser scanning confocal microscopy (LSCM). The expression of intercellular adhesion (ica) operon was detected by fluorescence quantitative PCR. Results The MICs of emodin against different strains of MRSA ranged from 2 to 8 μg/mL, while MBCs were from 4 to 16 μg/mL. Emodin obviously inhibited the formation of biofilm in a dose-dependent manner. Besides, it destroyed biofilm structure and increased the ratio of dead bacteria under biofilm. Fluorescence quantitative PCR showed that emodin significantly down-regulated the expression of ica operon. Conclusion Emodin can inhibit MRSA biofilm in vitro, and may restrain the formation of bacterial biofilm by down-regulating the expression of ica operon.

Glioma stem‐like cell (GSC) inhibition is a promising strategy for glioblastoma therapy. However, it is hampered by two main obstacles: first, delivery barriers, including the blood–brain barrier (BBB) and deep tumor‐penetrating obstacles, hinder sufficient drug accumulation in GSCs that often reside in hypoxic niches deep in glioblastoma; second, hypoxia and the immunosuppressive tumor microenvironment (TME) limit the therapeutic responses of GSC inhibition strategies. An intercellular cell adhesion molecule 1 (ICAM‐1)‐expressing M1 macrophage‐derived small extracellular vesicles loaded with a complex of bovine serum albumin, MnO2, and the small‐molecule inhibitor PTC209 (IsEV‐BMP) is constructed to achieve sequential barrier‐breaking delivery and simultaneous TME modulation to boost GSC inhibition. ICAM‐1 facilitates endothelial transcytosis‐mediated BBB crossing, microglial transcytosis‐mediated deep tumor penetration, and internalization of IsEV‐BMPs in GSCs. Hypoxia alleviation inhibits the binding of hypoxia‐inducible factor 1α (HIF‐1α) to hypoxia‐responsive elements on the gene promoters of cancer stem cell‐related markers and CD47 in GSCs, thereby improving PTC209 sensitivity and immune phagocytosis of GSCs. IsEV‐BMP achieves simultaneous microglial polarity transition and immuno‐microenvironmental modulation during microglial transcytosis‐mediated deep tumor penetration. Background: Anal fistula (AF) is an abnormal tunnel under the skin connecting the anal canal in the colon to the skin of buttocks. Fat-suppressed (FS) proton density-weighted (PDW) imaging is mainly used for the diagnosis of diseases involving bones and joints. Until now, its value in the diagnosis of anal fistula has been rarely reported. Purpose: To compare three magnetic resonance imaging (MRI) sequences – diffusion-weighted imaging (DWI), FS-PDW), and contrast-enhanced (CE) T1-weighted (T1W) imaging – for the diagnostic value of the internal opening of AF. Material and Methods: MRI scans of 132 patients suspected of having AF between December 2021 and April 2023 were retrospectively analyzed. In total, 65 patients who underwent preoperative MRI and were treated surgically were included. The lesion conspicuity and accuracy for featuring AF were calculated by evaluating the three imaging datasets DWI, FS-PDW, and CE-T1W imaging, with surgical findings serving as the reference standard for the presence of fistulas. The statistical analysis included the application of the chi-square test and Kruskal–Wallis test. Results: In 65 patients with AF, 87 internal openings of AF were confirmed. In terms of the diagnostic accuracy of the internal openings, both FS-PDW and CE-T1W imaging sequences were significantly better than DWI sequences, and the difference was statistically significant (P < 0.05). Conclusion: The FS-PDW imaging sequence showed comparable diagnostic performance of the internal opening of AF to CE-T1W imaging, which can provide an important diagnostic basis for clinical procedures. An enhanced algorithm, YOLOv5s-FCW, is put forward in this study to tackle the problems that exist in the current helmet detection (HD) methods. These issues include having too many parameters, a complex network, and large computation requirements, making it unsuitable for deployment on embedded and other devices. Additionally, existing algorithms struggle with detecting small targets and do not achieve high enough recognition accuracy. Firstly, the YOLOv5s backbone network is replaced by FasterNet for feature extraction (FE), which reduces the number of parameters and computational effort in the network. Secondly, a convolutional block attention module (CBAM) is added to the YOLOv5 model to improve the detection model's ability to detect small objects such as helmets by increasing its attention to them. Finally, to enhance model convergence, the WIoU_Loss loss function is adopted instead of the GIoU_Loss loss function. As reported by the experimental results, the YOLOv5s-FCW algorithm proposed in this study has improved accuracy by 4.6% compared to the baseline algorithm. The proposed approach not only enhances detection concerning small and obscured targets but also reduces computation for the YOLOv5s model by 20%, thereby decreasing the hardware cost while maintaining a higher average accuracy regarding detection. To investigate the effect of Co(OH)2 on the performance of the special capacitor, graphene was holed (HG) using microwave and KMnO4 and electrostatically sprayed with CNT to prepare a laminated structure bamboo woodceramics matrix (C/HG@BE). Then, Co(OH)2 was electrodeposited on the matrix to form a supercapacitor electrode (C/HG@BE-Co-x). The results showed that the Mn nanoparticles generated by KMnO4 under the action of microwave were partially embedded in the graphene sheet mesh and formed holes in the graphene sheet after removal. The matrix had a clear laminated structure and good electrical conductivity. The electrical resistivity was approximately 0.163 Ω cm at 1000 ℃ sintering temperature and remained low after the electrode was modified Co(OH)2. Meanwhile, the Co(OH)2 was uniformly deposited on the surface and within pores of the wall of the matrix and anchored by PPy to reduce fall. The electrode of C/HG@BE-Co-1500 modified with Co(OH)2 exhibited a good pseudocapacitance effect. At the current density of 0.1 A/g, the specific capacitance was approximately 255.12 F/g, and the retention of specific capacitance was maintained at 83.75% after 10000 cycles. When the energy density was 40.78 Wh/kg, the power density was 480 W/kg, indicating excellent electrochemical performance, which contributed of the synergistic action of electronic double layer capacitor and pseudocapacitance. Melon damping off, which has a negative impact on melon quality and yield, can be safely and effectively managed with Trichoderma. Melon cultivar 'Longtian No. 1' was evaluated at both the adult and seedling stages in a pot experiment. The Rs and PD liquids were utilized as CK1 and CK2, respectively. Trichoderma viride Tv286 treatments T1B, T2B, T3B, and T4B were used based on Rs at concentrations of 104, 105, 106, and 107 CFU·g−1, respectively. The impact of several treatments on the antioxidant system and seedling quality of melon were assessed at 15, 25, and 35 days after sowing. We examined the effects of several treatments on melon quality, yield attributes, and physiological and biochemical markers during the adult stage at 10, 20, and 30 days after pollination. The effects of several treatments on melon damping off were also studied. Applying T. viride Tv286 at different rates effectively increased the activities of enzymes, including catalase (CAT), peroxidase (POD), superoxide dismutase (SOD), ascorbate peroxidase (APX), and polyphenol oxidase (PPO), in the leaves of melon seedlings, significantly reduced the malondialdehyde (MDA) content, and improved the root–shoot ratio and seedling strength index. In terms of its influence on promoting the effect of antioxidant system indicators, T3B performed well. Melon seedlings treated with T3B showed higher CAT, POD, SOD, APX, and PPO activities in their leaves 35 days after sowing compared to CK1 (189.74, 169.61, 175.36, 224.20, and 477.39%, respectively). The strong seedling index and root–shoot ratio showed improvements of 130.43 and 79.71%, respectively, and the MDA content dropped by 35.66% at 35 days after sowing compared to CK1. Varying the rates at which T. viride Tv286 was applied increased the nitrate reductase (NR) activity and nitrate nitrogen, proline (Pro), chlorophyll, soluble sugar, and soluble protein contents in mature melon leaves, increasing melon quality and yield. T3B is the most effective marketing campaign. Compared to CK1, mature T3B leaves had higher NR activity, nitrate nitrogen content, chlorophyll content, soluble sugar content, soluble protein content, and Pro content 30 days after melon pollination (100.40, 135.17, 68.59, 93.65, 158.13, and 238.67%, respectively). The soluble solids, soluble protein, soluble sugar, vitamin C contents, and yield of melon fruit increased by 50.07, 126.82, 60.62, 70.79, and 61.45%, respectively, at 30 days after melon pollination compared to CK1. Optimal management of melon damping off can be accomplished with the application of T. viride Tv286 at different concentrations, with T3B exhibiting the best effect. The control effects reached 90.48 and 72.99% at the seedling and adult stages, respectively. Overall, T. viride Tv286 improved seedling quality, damping off control efficacy, melon yield and quality, and the antioxidant system during the seedling stage and enhanced physiological and biochemical characteristics during the adult stage. Trifluoromethyl (CF3) and other fluoroalkyl groups are of great significance in the fields of pharmaceutical chemistry and agricultural chemicals. Fluoroalkyl acids, especially trifluoroacetic acid (TFA) is considered the most ideal fluoroalkylation reagent due to its low cost and easy availability. However, the extremely high oxidation potential requirement of TFA limits its wide application. In recent years, since visible-light-induced fluoroalkylation through the ligand-to-metal charge transfer (LMCT) process can overcome the above limitations, it has become an effective synthetic tool for the construction of fluorinated compounds with complex molecules and structures. In this review, according to the classification of different metal catalysts, we summarize the trifluoromethylation and fluoroalkylation of olefins, heteroaromatics, and terminal alkynes in different metal catalytic systems and their corresponding reaction mechanisms. The photocatalytic fluoroalkylation via LMCT is believed to expedite the development of fluoro-containing drugs, and more novel fluoroalkylation methologies using this strategy will be disclosed. Simple Summary: Through gesture recognition and detection of sika deer, farmers can observe the gestures of sika deer without physical contact, providing data and technical support for the intelligent and welfare-oriented breeding of sika deer. This study is based on the YOLOv8 network model. By optimizing the convolution module, incorporating the attention mechanism, and enhancing the detection head module, a new method for detecting sika deer poses was developed. The method was assessed using four behavioral datasets, which included standing, lying, eating, and attacking. The pose-recognition accuracy of sika deer significantly improved to an average of 91.6%, laying a foundation for the health assessment and information management of sika deer. As the sika deer breeding industry flourishes on a large scale, accurately assessing the health of these animals is of paramount importance. Implementing posture recognition through target detection serves as a vital method for monitoring the well-being of sika deer. This approach allows for a more nuanced understanding of their physical condition, ensuring the industry can maintain high standards of animal welfare and productivity. In order to achieve remote monitoring of sika deer without interfering with the natural behavior of the animals, and to enhance animal welfare, this paper proposes a sika deer individual posture recognition detection algorithm GFI-YOLOv8 based on YOLOv8. Firstly, this paper proposes to add the iAFF iterative attention feature fusion module to the C2f of the backbone network module, replace the original SPPF module with AIFI module, and use the attention mechanism to adjust the feature channel adaptively. This aims to enhance granularity, improve the model's recognition, and enhance understanding of sika deer behavior in complex scenes. Secondly, a novel convolutional neural network module is introduced to improve the efficiency and accuracy of feature extraction, while preserving the model's depth and diversity. In addition, a new attention mechanism module is proposed to expand the receptive field and simplify the model. Furthermore, a new pyramid network and an optimized detection head module are presented to improve the recognition and interpretation of sika deer postures in intricate environments. The experimental results demonstrate that the model achieves 91.6% accuracy in recognizing the posture of sika deer, with a 6% improvement in accuracy and a 4.6% increase in mAP50 compared to YOLOv8n. Compared to other models in the YOLO series, such as YOLOv5n, YOLOv7-tiny, YOLOv8n, YOLOv8s, YOLOv9, and YOLOv10, this model exhibits higher accuracy, and improved mAP50 and mAP50-95 values. The overall performance is commendable, meeting the requirements for accurate and rapid identification of the posture of sika deer. In this study, a quantitative method based on fluorescein isothiocyanate (FITC)-labelled Hizikia fusiforme polyphenol–polysaccharide complex (HPC) and its purified fractions (PC1, PC4) was used, and its pharmacokinetics and tissue distribution were investigated in mice. The results showed that the FITC-labelled method had good linearity (R2 > 0.99), intra-day and inter-day precision (RSD, %) consistently lower than 15%, recovery (93.19–106.54%), and stability (RSD < 15%), which met the basic criteria for pharmacokinetic studies. The pharmacokinetic and tissue distribution results in mice after administration showed that all three sample groups could enter the blood circulation. and HPC-FITC had a longer half-life (T1/2: 26.92 ± 0.76 h) and mean retention time (MRT0–∞: 36.48 h) due to its larger molecular weight. The three groups of samples could be absorbed by the organism in a short time (0.5 h) mainly in the stomach and intestine; the samples could be detected in the urine after 2 h of administration indicating strong renal uptake, and faecal excretion reached its maximum at 12 h. The samples were also detected in the urine after 2 h of administration. This study provides some theoretical basis for the tissue distribution pattern of polyphenol–polysaccharide complex. All inorganic perovskite CsPbX3 (X = Br, I, Cl) materials emerge as one of the popular materials in the field of optoelectronic devices due to their outstanding photoelectric properties. Their photoelectric detection performance and stability could be further improved by combining mixed halide perovskites with other semiconductor materials, achieving high-performance variable wavelength photodetectors. In this work, high-quality CsPbBr3-xClx microcrystals were successfully grown on silicon (Si) substrates by chemical vapor deposition (CVD) method. By varying the Br/Cl ratio, the prepared CsPbBr3-xClx heterojunction photodetectors exhibit excellent response time in the scale of microsecond and superior narrowband detection capability in a wide wavelength ranges and excellent environmental stability, achieving the maximum responsivity of 102.6 A/W and detectivity of 5.20 × 1012 Jones under 7.8µW /cm2 light illumination intensity. In the realm of urban planning and environmental evaluation, the delineation and categorization of land types are pivotal. This study introduces a convolutional neural network-based image semantic segmentation approach to delineate parcel data in remote sensing imagery. The initial phase involved a comparative analysis of various CNN architectures. ResNet and VGG serve as the foundational networks for training, followed by a comparative assessment of the experimental outcomes. Subsequently, the VGG+U-Net model, which demonstrated superior efficacy, was chosen as the primary network. Enhancements to this model were made by integrating attention mechanisms. Specifically, three distinct attention mechanisms—spatial, SE, and channel—were incorporated into the VGG+U-Net framework, and various loss functions were evaluated and selected. The impact of these attention mechanisms, in conjunction with different loss functions, was scrutinized. This study proposes a novel network model, designated VGG+U-Net+Channel, that leverages the VGG architecture as the backbone network in conjunction with the U-Net structure and augments it with the channel attention mechanism to refine the model's performance. This refinement resulted in a 1.14% enhancement in the network's overall precision and marked improvements in MPA and MioU. A comparative analysis of the detection capabilities between the enhanced and original models was conducted, including a pixel count for each category to ascertain the extent of various semantic information. Marine sand, in addition to oil and gas resources, is the second-largest marine mineral resource. The rational development and utilization of marine sand resources are conducive to the growth of the marine economy. In the process of marketing marine sand in China, local authorities are required to delineate auctioned sand mining areas after a general survey, commonly referred to as preliminary exploration. Marine sand can be categorized into surface marine sand and buried marine sand. Buried marine sand deposits are buried beneath the sea floor, making it challenging to locate them due to their thin thickness. Consequently, there exist numerous technical difficulties associated with marine sand exploration. We conducted the preliminary research work in the waters off Guangdong Province of the South China Sea, employing a reduced drilling and identifying a potentially extensive deposit of marine sand ore. In this study, various geophysical methods such as sub-bottom profile survey, single-channel seismic survey, and drilling engineering were employed in the northern offshore waters of the South China Sea. As a result, two distinct marine sand bodies were delineated within the study area. Additionally, five reflective interfaces (R1, R2, R3, R4, and R5) were identified from top to bottom. These interfaces can be divided into five seismic sequences: A1, B1, C1, D1, and E1, respectively. Three sets of strata were recognized: the Holocene Marine facies sediment layer (Q4m), the Pleistocene alluvial and pluvial facies sediment layer (Q3al+pl), as well as the Pleistocene Marine facies sedimentary layer (Q3m). In total, two placers containing marine sand have been discovered during this study. We estimated the volume of marine sand and achieved highly favorable results of the concept that we are proposing a geologic exploration approach that does not involve any previous outcropping analogue study. Raman spectroscopy is very sensitive to the U‐O bonding strength changes under high pressure, and it could reveal important information about the bonding and stoichiometry of the U‐O systems. The pressure and temperature (P–T) phase diagram of U3O8 was investigated by Raman spectroscopy via four independent P–T experimental paths. The phase boundaries between different phases have been determined by the changes in Raman spectra. A new phase (α′‐U3O8) was observed below 233 K at 0.6 GPa or 198 K at zero pressure accompanied by the discontinuities in Raman wavenumber and the new shoulder peak near the B2(6) Raman modes. Upon compressing at low temperature (123 K), the high‐pressure irreversible phase transition from orthorhombic to cubic structure (α′‐δ) was observed at around 11.3 GPa, evidenced by the merging of B2(6) and B′2(6) Raman modes.
